Design and optimization of a colorimetric biosensor using functionalized gold nanoparticles and bi-functional linker for agriculture and food industry

This study aims to develop a highly sensitive colorimetric detection system utilizing functionalized gold nanoparticles (f-AuNPs) and bifunctional linkers (BLs) for the detection of pathogens and contaminants in food matrices. The primary objective was to explore and optimize key parameters, including reaction time, system volume and the concentration of bifunctional linkers, in order to enhance the system’s detection capabilities. The core mechanism of the bi-functional linker-based detection system is based on the aggregation of streptavidin-functionalized AuNPs (stAuNPs), which varies in accordance with the concentration of the bifunctional linkers (BLs) and reflects the quantity of effective linkers (EALs) available in the system. The process comprises the binding of the targets and BLs, a reduction in EAL concentration, an increase in the BLs concentration required for the aggregation of the stAuNPs, and a shift in the range exhibiting a visible color change (REVC). In systems without a target, all BLs induce aggregation. In contrast, in systems with a target present, some BLs are bound by the target, reducing EAL concentration and altering the optimal BLs concentration for stAuNP aggregation. The system was shown to be capable of detecting protein in PBS at concentrations as low as 2 nM, as well as Salmonella at 101 CFU/mL. In whole milk, the detection limits were 20 nM for protein and 102 CFU/mL for Salmonella. The principal conclusion is that the BLs-based system provides a robust and adaptable platform for pathogen detection, even in complex food matrices, obviating the need for preprocessing. The findings demonstrate the system’s potential for practical applications in the fields of food safety and agricultural contaminant detection, offering clear visual signals and high sensitivity.

Detection of food contaminants: A review of established rapid analytical techniques and their applications and limitations

AbstractFood contaminants have consistently posed a serious threat to human health and have caused various food safety problems. The most common techniques currently used for the detection of food contaminants, include instrumental and immunoassay methods; however, these traditional methods have shown some limitations when applied to the analysis of food contaminants. Therefore, in recent years, more and more researchers have been devoted to the development of rapid, reliable, sensitive, and suitable detection techniques for the detection of food contaminants in food matrices. In this review, we first classify the analytical methods proposed in recent years into colorimetric methods, fluorescence methods, photoelectrochemical methods, electrochemical methods, nucleic acid amplification methods, and other methods, and illustrate their detection principles. In addition, the paper highlights the strategies and recent research advances for each method through typical application examples. Finally, the future perspectives of these analytical methods are presented. We hope that this review will provide researchers with valuable insights to guide the sensitive detection of food contaminants.

Threat or treat: Exposure assessment and risk characterisation of chemical contaminants in soft drinks and chocolate bars in various Polish population age groups.

In the frame of the European Food Risk Assessment (EU-FORA) fellowship programme, two studies on chemical contaminants in food matrices were carried out in Warsaw, Poland, at the Department of Food Safety and Chemical Analysis, Institute of Agricultural and Food Biotechnology. The first study addressed health concerns about the dietary exposure to bisphenol A (BPA) contamination due to consumption of soft drink by Polish population. BPA is an organic additive used in the production of epoxy resins and polycarbonate plastics and because of this it is used in the internal coating of cans and in plastic bottle production. Depending on several factors, BPA can migrate from these materials to the soft drink and so, it can be ingested by consumers causing hormonal and reproductive disorders. To estimate the Polish population exposure to BPA, several soft drinks belonging to different brands were purchased from a supermarket in the city of Warsaw and analysed. The result of the analysis highlight that mean BPA exposure in the Polish population exceeds the tolerable daily intake proposed by the EFSA scientific opinion, raising health concerns. On the other hand, the second study, focused on cadmium exposure due to chocolate consumption by Polish population, did not raise any health concern. Cadmium is a heavy metal that naturally occurs in its inorganic form in the environment and its presence in chocolate derives only from the cocoa beans and not from contamination during processing. Its accumulation in the human body can create several adverse effects, including renal dysfunction and failure. To estimate the Polish population exposure to cadmium, several chocolate bars were purchased from a supermarket in the city of Warsaw and analysed. The results of the analysis show that cadmium exposure in the Polish population does not exceed the tolerable weekly intake proposed by the EFSA scientific opinion.

Recent advances in rare earth ion-doped upconversion nanomaterials: From design to their applications in food safety analysis.

The misuse of chemicals in agricultural systems and food production leads to an increase in contaminants in food, which ultimately has adverse effects on human health. This situation has prompted a demand for sophisticated detection technologies with rapid and sensitive features, as concerns over food safety and quality have grown around the globe. The rare earth ion-doped upconversion nanoparticle (UCNP)-based sensor has emerged as an innovative and promising approach for detecting and analyzing food contaminants due to its superior photophysical properties, including low autofluorescence background, deep penetration of light, low toxicity, and minimal photodamage to the biological samples. The aim of this review was to discuss an outline of the applications of UCNPs to detect contaminants in food matrices, with particular attention on the determination of heavy metals, pesticides, pathogenic bacteria, mycotoxins, and antibiotics. The review briefly discusses the mechanism of upconversion (UC) luminescence, the synthesis, modification, functionality of UCNPs, as well as the detection principles for the design of UC biosensors. Furthermore, because current UCNP research on food safety detection is still at an early stage, this review identifies several bottlenecks that must be overcome in UCNPs and discusses the future prospects for its application in the field of food analysis.

A review on recent advances in mass spectrometry analysis of harmful contaminants in food.

Food safety is a widespread global concern with the emergence of foodborne diseases. Thus, establishing accurate and sensitive detection methods of harmful contaminants in different food matrices is essential to address and prevent the associated health risks. Among various analytical tools, mass spectrometry (MS) can quantify multiple impurities simultaneously due to high resolution and accuracy and can achieve non-target profiling of unknown pollutants in food. Therefore, MS has been widely used for determination of hazardous contaminants [e.g., mycotoxin, pesticide and veterinary drug residues, polychlorinated biphenyls (PCBs), dioxins, acrylamide, perfluorinated compounds (PFCs) and p-Phenylenediamine compounds (PPDs) in food samples]. This work summarizes MS applications in detecting harmful contaminants in food matrices, discusses advantages of MS for food safety study, and provides a perspective on future directions of MS development in food research. With the persistent occurrence of novel contaminants, MS will play a more and more critical role in food analysis.

Miniaturized liquid chromatography applied to the analysis of residues and contaminants in food: A review.

The humankind is pretty dependent on food to control several biological processes into the organism. As the world population increases, the demand for foodstuffs follows the same trend claiming for a high food production situation. For this reason, a substantial amount of chemicals is used in agriculture and livestock husbandries every year, enhancing the likelihood of contaminated foodstuffs being commercialized. This outlook becomes a public health concern; thus, the governmental regulatory agencies impose laws to control the residues and contaminants in food matrices. Currently, one of the most important analytical techniques to perform it is LC. Despite its already recognized effectiveness, it is often time consuming and requires significant volumes of reagents, which are transformed into toxic waste. In this context, miniaturized LC modes emerge as a greener and more effective analytical technique. They have remarkable advantages, including higher sensitivity, lower sample amount, solvent and stationary phase requirements, and more natural coupling to MS. In this review, most of the critical characteristics of them are discussed, focusing on the benchtop instruments and their related analytical columns. Additionally, a discussion regarding the last 10 years of publications reporting miniaturized LC application for the analysis of natural and industrial food samples is categorized. The main chemical classes as applied in the crops are highlighted, including pesticides, veterinary drugs, and mycotoxins.

Effects of Ions on Core-Shell Bimetallic Au@Ag NPs for Rapid Detection of Phosalone Residues in Peach by SERS

The sensitivity of surface-enhanced Raman spectroscopy (SERS) extremely relies on experimental parameters including pH and aggregating agents. Using Au@Ag nanoparticles (NPs) with 26 nm Au core size and 6 nm Ag shell thickness as the substrate, the effect of cationic (Ca2+, K+, Na+) and anionic (Cl−, Br−) aggregating agents was investigated on the SERS detection of phosalone. The optimum concentrations of the aggregating agents in relation to the maximum SERS intensity differed broadly from 1 × 10−2 mol/L for CaBr2 to 1 × 101 mol/L for KCl. Both anions and cations greatly affected the SERS enhancement. With employing Br− as the anion, Ca2+ and K+ showed the maximum SERS intensities for phosalone, while using Ca2+ as the cation, Br− produced the maximum SERS enhancement. Among these aggregating agent combinations, the maximum SERS enhancement was achieved by employing 1 × 10−2 mol/L CaBr2 at pH 3.0. The detection limits of phosalone in standard solution and peach were 0.02 mg/L and 0.2 mg/kg, respectively. This study demonstrated that the Au@Ag NP-based SERS approach could be used as a sensitive, rapid, and simple method for detecting trace contaminants in food matrices.

Nontargeted Detection Methods for Food Safety and Integrity.

Nontargeted workflows for chemical hazard analyses are highly desirable in the food safety and integrity fields to ensure human health. Two different analytical strategies, nontargeted metabolomics and chemical database filtering, can be used to screen unknown contaminants in food matrices. Sufficient mass and chromatographic resolutions are necessary for the detection of compounds and subsequent componentization and interpretation of candidate ions. Analytical chemistry-based technologies, including gas chromatography-mass spectrometry (GC-MS), liquid chromatography-mass spectrometry (LC-MS), nuclear magnetic resonance (NMR), and capillary electrophoresis-mass spectrometry (CE-MS), combined with chemometrics analysis are being used to generate molecular formulas of compounds of interest. The construction of a chemical database plays a crucial role in nontargeted detection. This review provides an overview of the current sample preparation, analytical chemistry-based techniques, and data analysis as well as the limitations and challenges of nontargeted detection methods for analyzing complex food matrices. Improvements in sample preparation and analytical platforms may enhance the relevance of food authenticity, quality, and safety.

Untargeted food chemical safety assessment: A proof-of-concept on two analytical platforms and contamination scenarios of tea

This study aims at assessing the capability of comparing and combining different instrumental platforms in an untargeted approach with a view of detecting chemical contaminants in food matrices at low levels. A strategy based on liquid chromatography-high resolution mass spectrometry (LC-HRMS) and chemometrics has been applied on two different complex food contamination scenarios, with tea as study product. The first scenario aimed at mimic the presence of a dozen of contaminants at levels just above regulatory limits (i.e. 10 and 30 μg/kg); the second scenario, more complex, aimed at simulate the presence of several different contaminations at levels close to regulatory limits (10 μg/kg) in different samples. This work was carried on two LC-HRMS platforms (with respectively ToF and Orbitrap mass analyzer technologies), and a highly automated data treatment workflow was implemented to deal with data acquired on both platforms. The untargeted approach performed well on all scenarios (even the most complex) and analytical platforms. Performance comparison between LC-HRMS technologies was made possible thanks to a vendor-neutral data treatment process.

Fabrication of gold nanorods for SERS detection of thiabendazole in apple

Thiabendazole (TBZ) is a kind of pesticide that is widely used in agriculture, and its residue may pose a threat to human health. In order to measure TBZ residues in food samples, a surface-enhanced Raman spectroscopy (SERS) method combined with a homogeneous and reusable gold nanorods (GNR) array substrate was proposed. GNR with a high uniformity was synthesized and then applied to the self-assembly of a GNR vertically aligned array. The relative standard deviation (RSD) of the array for SERS could reach 15.4%, and the array could be reused for more than seven times through the treatment of plasma etching. A logarithmic correlation between TBZ concentration and Raman intensity was obtained, with the best determination coefficient (R2) and the corresponding limit of detection (LOD) of 0.991 and 0.037 mg/L in methanol solution, and 0.980 and 0.06 ppm in apple samples, respectively. The recoveries of TBZ in apple samples ranged from 76% to 107%. This study provided a rapid and sensitive approach for detecting TBZ in apples based on SERS coupled with GNR array substrate, showing great potential for analyzing other trace contaminants in food matrices.

Recent approaches for on-line analysis of residues and contaminants in food matrices: A review.

This review highlights recent developments for on-line determination of residues and contaminants in complex matrices such as food samples. This involves the on-line coupling of a sample preparation technique (as the first "dimension") with a chromatographic system (second "dimension"), usually followed by mass spectrometry. Although frequently treated as quite distinct techniques, the role of all devices utilized as the first dimension in this approach aims to decrease the sample complexity while eliminating as much as possible the matrix contaminants to facilitate the qualitative and quantitative determination of the compounds of interest. This review will focus on the following techniques as the first dimension: (i) on-line solid-phase extraction; (ii) in-tube solid-phase microextraction; (iii) matrix solid-phase dispersion; and (iv) turbulent flow chromatography. The second dimension is usually performed using a chromatographic column to isolate the analyte(s) of interest for further mass spectrometry determination. A description of the basis of this on-line approach and its distinct set up possibilities is presented, which is followed by a critical review of the literature covering this subject in the last ten years (focusing on the last five years) with emphasis on the analysis of residue and contaminants in food samples.

Short communication: Monitoring the presence of perfluoroalkyl substances in Italian cow milk

Perfluoroalkyl substances (PFAS) are fully fluorinated compounds widely used during the last 60 yr in the production of multiple industrial and consumer applications, such as food packaging, nonstick cookware, cleaning agents, and many more. These emerging contaminants have recently become of concern for human health because of their potential negative effects. The risk of exposure to PFAS for humans is mainly related to diet, and the increasing interest in food safety has led the European Commission to call Member States to monitor these contaminants in food matrices. The purpose of the present work was to perform the first monitoring on the presence of perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA), the 2 main and most widely investigated molecules of this family, in cow milk commercially available in Italy. We used an analytical protocol consisting of liquid-liquid extraction followed by 2 purification steps through solid-phase extraction cartridges and injection on an ultra-performance liquid chromatography-tandem mass spectroscopy system. The analysis of 67 samples of different types of cow milk from Italy demonstrated that contamination by PFOS was often present, although at relatively low concentrations (up to 97ng/L), whereas PFOA was rarely found. On the basis of these results and data reported in the literature on this matrix, milk does not seem to be a major source of PFAS compared with other food categories such as fish and seafood. However, variability among different types of milk must be taken into account, and surveys of milk-derived products would be helpful to better define the risk for consumers.
