Spatial distribution and clustering of contagious caprine pleuropneumonia in goats of Amhara region, Ethiopia

A cross-sectional study with a multistage cluster sampling technique was undertaken from January to July 2019 in Amhara region to identify spatial clusters and distributions of contagious caprine pleuropneumonia (CCPP) hotspots. In the study, a total of 2080 goats from 258 flocks across 60 villages in 12 districts were tested for CCPP serostatus using Competitive Enzyme-Linked Immunosorbent Assay (C-ELISA). Villages were taken as an aggregate unit to detect spatial distribution and clustering. Spatial autocorrelation, interpolation, and spatial scan statistics analyses were employed to analyze the spatial patterns and clusters of CCPP serostatus. The overall seroprevalence of CCPP at the animal level was 5.1 % (95 % CI: 3.8–6.6). The spatial distribution of CCPP seropositivity was non-random at the village level (Moran's I: 0.400791, P-value <0.01). Two statistically significant spatial clusters of CCPP seropositive were found: one in West Gojjam and the other encompassing parts of North Wollo, Oromiya Liyu zone, and South Wollo zones. The CCPP seropositivity showed spatial variation across the Amhara region. High CCPP seropositive spots were found in the southwestern and northeastern parts of the Amhara region. Generally, the findings of this study provide valuable insights in developing disease control strategies, prioritizing target intervention areas and measures, and designating CCPP disease-free zones. Applying movement restrictions and vaccinating goats will be important intervention measures to prevent and control CCPP in the region.

Seroprevalence and associated risk factors of contagious caprine pleuropneumonia in selected districts of South Wollo Zone Northeast Ethiopia

Contagious caprine pleuropneumonia (CCPP) is a severe and devastating respiratory disease of goats, which is characterized by severe serofibrinous pleuropneumonia accompanied by high morbidity and mortality. A cross-sectional study was conducted from July 2022 to January 2023 to determine the seroprevalence of CCPP and identify risk factors associated with the occurrence of CCPP in goats in five selected districts of the South Wollo Zone of the Eastern Amhara region. A total of 384 sera samples were collected from goats and examined for antibodies specific to Mycoplasma capricolum subspecies capripneumoniae (Mccp) using Competitive Enzyme-Linked ImmunoSorbent Assay (cELISA) test. Out of the total examined sera, 26 samples were positive for CCPP, giving an overall seroprevalence of 6.7% (95% CI = 6.64–9.77). A seroprevalence of 5.05%, 4.65%, 2.78%, 12.90%, and 10.77% were recorded in Ambasel, Tehuledere, Kalu, Dessie Zuria and Kutaber districts, respectively. However, there was no statistically significant difference among these five districts (p > 0.05). The seroprevalence of CCPP varies significantly between age groups and agroecology (p < 0.05). However, the seroprevalence did not vary with sex, body condition score (BCS), and flock size (p > 0.05). Old-aged goats (OR = 4.10) and goats found in the lowlands (OR = 5.09) were at higher risk of infection with CCPP than young-aged goats and goats found in the highlands, respectively. In conclusion, the present seroprevalence investigation indicated the occurrence of CCPP in those selected study districts of the South Wollo Zone. Therefore, appropriate control measures, including avoiding the mixing of flocks and vaccination should be designed and implemented especially in the lowland areas and older goats to reduce the further spread and magnitude of the disease.

A molecular epidemiological investigation of contagious caprine pleuropneumonia in goats and captive Arabian sand gazelle (Gazella marica) in Oman

BackgroundContagious caprine pleuropneumonia (CCPP) is a fatal WOAH-listed, respiratory disease in small ruminants with goats as primary hosts that is caused by Mycoplasma capricolum subspecies capripneumoniae (Mccp). Twelve CCPP outbreaks were investigated in 11 goat herds and a herd of captive Arabian sand gazelle (Gazella marica) in four Omani governorates by clinical pathological and molecular analysis to compare disease manifestation and Mccp genetic profiles in goats and wild ungulates.ResultsThe CCPP forms in diseased and necropsied goats varied from peracute (5.8%), acute (79.2%) and chronic (4.5%) while all of the five necropsied gazelles showed the acute form based on the clinical picture, gross and histopathological evaluation. Colonies of Mccp were recovered from cultured pleural fluid, but not from lung tissue samples of one gazelle and nine goats and all the isolates were confirmed by Mccp-specific real time PCR. Whole genome-single nucleotide polymorphism (SNP) analysis was performed on the ten isolates sequenced in this study and twenty sequences retrieved from the Genbank database. The Mccp strains from Oman clustered all in phylogroup A together with strains from East Africa and one strain from Qatar. A low variability of around 125 SNPs was seen in the investigated Omani isolates from both goats and gazelles indicating mutual transmission of the pathogen between wildlife and goats.ConclusionRecent outbreaks of CCPP in Northern Oman are caused by Mccp strains of the East African Phylogroup A which can infect goats and captive gazelles likewise. Therefore, wild and captive ungulates should be considered as reservoirs and included in CCPP surveillance measures.

Development and validation of an indirect ELISA for the serodiagnosis of contagious caprine pleuropneumonia in goats

Mycoplasma capricolum subsp. capripneumoniae (Mccp) is a dreadful pathogen of contagious caprine pleuropneumonia (CCPP). Mccp local strains have recently been isolated from naturally infected goats in northern areas of Pakistan. In this study, we established an in-house indirect ELISA (iELISA) based on whole-cell proteins (WCP) of local Mccp strains for the detection of anti-Mccp antibodies. Serum samples from naturally (n=500) and experimentally infected (n=20) and non-infected animals (n=100) were used for the establishment of iELISA. The specificity and sensitivity of the iELISA were determined using a commercially available competitive ELISA (cELISA) kit as a reference standard. The cut-off value (OD450) was found 0.53 by ROC analysis. As indicated by a kappa value of 0.88 and 96.66% agreement, both assays showed perfect agreement. An iELISA based on WCP of local Mccp detected seroconversion in experimental goats 7 days post-infection (dpi). The iELISA revealed a sensitivity of 90.83% (95% CI: 83.77%, 95.55%) and a specificity of 92.31% (95% CI: 84.79%, 96.85%) and has no cross-reaction with related mycoplasma epidemic diseases. For both inter-assay and intra-assay precision, the iELISA percentage coefficient of variation (%CV) was 4.8% and 1.74%, respectively. In conclusion, this in-house iELISA based on local Mccp-WCP was determined as a sensitive and specific assay for the diagnosis of Mccp infection.

Seroprevalence of Contagious Caprine Pleuropneumonia (CCPP) in Bharatpur, Chitwan, Nepal

A study was conducted to determine the seroprevalence of Contagious Caprine Pleuropneumonia (CCPP) in 106 goats of Bharatpur Municipality of Chitwan, Nepal from July to October 2021. A questionnaire was used to determine the potential risk factors. Collected samples were subjected to a competitive enzyme-linked immunosorbent assay (cELISA) for the detection of antibodies. The seroprevalence of Contagious Caprine Pleuropneumonia in goats was 4.71% (5/106). Although seroprevalence was high among females and goats below or equal to 5 years, there was no statistically significant. This study was carried out in the Chitwan district for the first time, applying the (Mccp) Antibody test technique, considering the need for control of this economically important disease for poor livestock farmers in Chitwan and it can play important role in the epidemiological study of CCPP in Nepal.

Risk factors associated with contagious caprine pleuropneumonia in goats of Amhara region, Ethiopia

Contagious caprine pleuropneumonia (CCPP) is a serious contagious disease of goats, sheep and wild ruminants caused by Mycoplasma capricolum subspecies capripneumoniae. The disease is known for its high mortality, morbidity and economic losses. A cross-sectional study using multistage cluster sampling technique was conducted in Amhara region from January 2019 to July 2019 to estimate seroprevalence and identify risk factors of CCPP occurrence in the region. A total of 2080 goats from 61 villages and 12 districts of the region were tested for CCPP serostatus using Competitive Enzyme-Linked Immunosorbent Assay (C-ELISA). A multilevel mixed-effect logistic regression model was used to identify risk factors of CCPP seropositivity at animal and flock-level. The serum sample results revealed an overall animal level seroprevalence of 5.1% (95% CI: 3.8–6.6) and flock-level prevalence of 26.0% (95% CI: 19.7–33.4). At individual animal level, presence of other health problems (OR = 45.9 (95% CI: 25.3–83.4)), age (adult age (OR = 6.2 (95% CI:3.4–11.4)) and old age (OR = 13.1 (95% CI: 6.2–27.8))), and breed type (Afar (OR= 32.3 (95% CI: 2.9–366.1)), Central highland (OR=13.7 (95% CI: 1.3–140.6)), and western highland (OR=16.2 (95% CI: 1.4–185.7))) were identified as risk factors for CCPP seropositivity. In contrast, contact with other flocks (OR = 59.9 (95% CI: 6.1–585.6)), presence of trade route (OR = 3.1 (95% CI: 1.0–9.1)) and presence of sheep (OR = 2.6 (95% CI: 1.2–5.7)) were flock-level risk factors for CCPP seropositivity. Generally, CCPP appears to be common among goats of Amhara region. Goat flocks dominated with older age animals; breeds of Afar, central highland, and western highland; raise with sheep; have contact with other flocks; and kept along trade routes are more at risk for CCPP. Hence, awareness creation to the producers, movement control, and regular prophylactic vaccination should be considered to control CCPP in Amhara region.

Seroprevalence of Peste des Petits Ruminants and Contagious Caprine Pleuropneumonia Coinfections in Goats in Kwale County, Kenya.

Goats are among the most important small ruminants affected by Peste des Petits ruminants (PPR) and contagious caprine pleuropneumonia (CCPP) diseases, two of the most significant constraints worldwide to the production of small ruminant species. Herein, the competitive enzyme-linked immunosorbent assay (cELISA) and the latex agglutination test (LAT) were used to determine the coinfections of PPR and CCPP in goats in Kwale County on Kenya's South Coast. A total of 368 serum samples were collected from goats of various ages and sexes exhibiting respiratory distress in the four subcounties of Kwale County (Kinango, Lunga Lunga, Matuga, and Msambweni) and screened for PPR and CCPP antibodies. Of the 368 goats sampled, 259 (70.4%) were females and 109 (29.6%) were males, and 126 (34.2%), 71 (19.3%), 108 (29.3%), and 63 (17.1%) samples were collected from Kinango, Matuga, Lunga Lunga, and Msambweni, respectively. The overall PPR seropositivity rate was 48.6% (179/368); rates in Kinango, Lunga Lunga, Matuga, and Msambweni were 70.6%, 29.6%, 49.3%, and 36.5%, respectively. The overall CCPP seropositivity rate was 45.4% (167/368), while rates in Kinango, Lunga Lunga, Matuga, and Msambweni were 51.6%, 49.1%, 36.6%, and 36.5%, respectively. Notably, the seropositivity of PPR was higher in male (53.3%) than in female (46.72%) goats, though not statistically significant. In addition, the CCPP seropositivity rates were not significantly different between male (44.0%) and female (45.9%) goats. Regarding age, the PPR seropositivity rates were 45.9%, 55.8%, and 52.3% in adults, kids, and weaners, respectively. For CCPP, the seropositivity rates were 48.3%, 40.4%, and 42.3% in adults, kids, and weaners, respectively. The coinfection rate of PPR and CCPP was 22.3% (82/368). Despite the high coinfection, univariate analysis revealed no relationship between PPR and CCPP infections. However, given the high PPR and CCPP infection rates, as a result of separate or coinfection, there is a need to upscale or intensify vaccination in the county.

Investigation of acute-phase proteins and cytokines response in goats with contagious caprine pleuropneumonia with special reference to their diagnostic accuracy.

Acute-phase proteins (APPs) have always had valued diagnostic potentialities in response to infection. This study aimed to evaluate the diagnostic accuracy of selected APPs and proinflammatory cytokines (PIC) in goats with contagious caprine pleuropneumonia (CCPP) under field conditions. Moreover, to highlight the role of tested biomarkers in CCPP pathogenesis. Fifty-eight goats (38 confirmed cases with CCPP and 20 healthy controls) were involved in this investigation. C-reactive protein (CRP), procalcitonin (PCT), haptoglobin (HP), fibrinogen (Fb), serum amyloid A (SAA), selected PIC (IL1-α, IL1-β, IL-6, interferon-gamma (IFN-γ) and tumor necrosis factor-alpha (TNF-α)) levels were investigated in serum samples from all goats under investigation. Latex agglutination test was used for diagnosis of goats with CCPP. For microbiological investigations, nasopharyngeal swabs (from all goats), lung tissues and pleural fluids (from only necropsied goats) were collected. This study revealed that all tested parameters have a high to moderate degree of diagnostic performance for CCPP. Magnitudes of increase in levels of APPs (CRP, HP and SAA) were stronger than PIC, IFN-γ, Fb and PCT. All tested parameters showed high diagnostic accuracy (AUROC >90%), except HP (AUROC = 87.3%) and IFN-γ (AUROC = 78.8%) showed moderate accuracy in differentiation of goats with and without CCPP infection. For detecting goats with and without CCPP infection, HP had the lowest sensitivity (Se = 81.6%) and Fb had the lowest specificity (Sp = 85.0%) among the APPs parameters tested. However, PCT showed the highest Se (100%) and Sp (95.0%) to detect goats with and without CCPP infection among tested parameters. Conclusively, this study endorses the significance of selected APPs and PIC as additional screening diagnostic parameters for naturally occurring CCPP in goats. However, it does not replace traditional methods for diagnosis of CCPP in goats. Furthermore, APPs and PIC have an important role in disease pathogenesis in goats.

Sero-Prevalence of Contagious Caprine Pleuropneumonia in Goats in Afgoye District Lower Shabelle Region, Somalia

Background: Contagious caprine pleuropneumonia (CCPP), caused by Mycoplasma capricolum subsp. Capripneumoniae (Mccp), is one of the most important goat diseases causing high morbidity and mortality, these results in heavy economic losses to countries like Somalia, which is a country affected the civil war of the 1990s. Objective: Generally, the disease presence has been reported in the country. However, there is no quantitative data on the prevalence and distribution of the disease has been scientifically reported in the country. Methods: A cross-sectional study was conducted from September 2019 to June 2020 in three villages of Afgoye district of Somalia to assess the seroprevalence and identify the associated risk factors for the occurrence of the CCPP. The risk factors considered included sex, age, and locations. Results: A total of 100 blood samples were collected and evaluated by Capri-Latex Agglutination Test (Capri-LAT). A total of 49/100 (49%, CI 95%: (38.86%-59.2%) goats were seropositive for anti-CCPP antibody. The epidemiological risk factors considered, sex, age and locations, were not significantly associated with the CCPP sero prevalence (p&gt;0.05). However, the sero positivity was slightly higher in female (51%, 95% CI: 39.8-62.6), &gt;3 years old goats (86.3%, 95% CI: 48.9-87.4) and Markazka village (56%, 95% CI: 41.3-70.0) compared to male (40%, 95% CI: 19.1-63.9), 1-3 years old (57.5%, 95% CI: 40.9- 73.0), and Shareeco (43.8%, 95% CI: 19.8-70.1) and Siinka (41.2%, 95% CI: 24.7-59.3) villages, respectively. Conclusion: The present study indicates the prevalence of CCPP in the studies areas and this requires practicable measures to be put in place towards the control of the disease.

The first identification of contagious caprine pleuropneumonia (CCPP) in sheep and goats in Egypt: molecular and pathological characterization.

Contagious caprine pleuropneumonia (CCPP) is one of the most fatal and contagious diseases of goats. To date, the occurrence of CCPP in Egypt has not been reported. During the period from 2017 to 2018, 200 goats and 400 sheep from Matrouh Governorate (Al Alamein and El Hammam cities) were suspected to have CCPP; animals were examined to confirm the presence of CCPP infection as well as the epidemiological status, clinical features, and molecular and histopathologic characteristics of lung tissues. Additionally, a treatment trial was performed to assess the efficacy of anti-mycoplasma therapy in the treatment of clinical cases of this disease. The occurrence of CCPP was 32.5% and 5% in goats and sheep, respectively, while case fatality was 30% and 8% in goats and sheep, respectively. The clinical forms of CCPP in both sheep and goats varied from per-acute to acute or chronic cases. Histopathological analysis of lung tissues from dead cases (either sheep or goats) revealed different stages of broncho- and pleuropneumonia ranging from per-acute to acute or chronic stages. Lung tissues showed severe congestion of interalveolar capillaries, flooding of alveoli and bronchi with a fibrinous exudate, a high degree of pleural thickening, and multifocal areas of necrosis that were sometimes sequestered in the fibrous capsule. Isolation of Mycoplasma capricolum subspecies capripneumoniae (Mccp) was confirmed in all dead cases by agar and broth culture methods and polymerase chain reaction. The treatment trial revealed that the marbofloxacin and spiramycin groups had a higher cure rate (70%) than the oxytetracycline group (40%) and a lower fatality rate (30%) than the oxytetracycline group (60%). Conclusively, infection with CCPP in goats and sheep is considered to be novel for Mccp in Egypt, where this species is considered to be the main pathogen in goats, not in sheep. Additionally, it could be concluded that treatment may be effective only if given early. Further comprehensive surveys are required to investigate the risk of CCPP in goats and sheep in all Egyptian governorates.

Risk factors associated with contagious caprine pleuro-pneumonia in goats in pastoral areas in the Rift Valley region of Kenya

A cross-sectional study to determine risk factors associated with sero-prevalence of contagious caprine pleuro-pneumonia (CCPP) in goats was carried out between the months of March, 2014 and March, 2015 in Pokot East, Turkana West and Kajiado Central Sub-counties. A semi-structured questionnaire focusing on risk factors for CCPP was completed for each flock whose serum samples were collected. A logistic regression model was developed to assess the association between the risk factors and CCPP sero-positivity. Of the 54 flocks, 49 (90.7%) presented at least one sero-positive animal. Two hundred and four of the 432 goats tested sero-positive at monoclonal antibody based competitive Enzyme-linked immuno-sorbent assay (c-ELISA), hence a sero-prevalence of 47.2% (95% CI=42.5– 51.9). Previous exposure of flocks to CCPP (p<0.001, OR=52.8; CI=6.45, 432), distant sources of veterinary drugs (p<0.001, OR=6.17; CI=3.41, 11.1), movement of goats to dry season feeding areas (p<0.001, OR=4.31; CI=2.39, 7.75) and markets as a source of new introductions to the flock (p=0.033, OR=1.86; CI=1.05, 3.27) were identified as risk factors significantly associated with CCPP sero-prevalence. The findings provide further evidence supporting the high prevalence and endemic state of the disease in pastoral flocks and hence there is need for adequate measures to be put in place to control the disease effectively.

Seroprevalence Study on Goat Contagious Caprine Pleuropneumonia in Jabalpur, Madhya Pradesh

The present study highlights the seroprevalence of contagious caprine pleuropneumonia (CCPP) in goats using slide agglutination test. A total of 1427 serum samples from goats belonging to Jabalpur district of Mahakaushal region of Madhya Pradesh were screened over a period of one year (i.e. April 2014 to March 2015). All the samples were screened for CCPP antibodies by slide agglutination test (SAT) using colored CCPP antigen. The overall seroprevalence of contagious caprine pleuropneumonia in goats was 10.65 per cent (152 out of 1427 goats). Seroprevalence of CCPP in organised goatry was higher (i.e. 13.54%) than the unorganised sector of goatry (i.e. 9.01%). Sex wise seroprevalence was marginally higher in female (10.67%) than the male (10.61%). SAT for CCPP detection using colored antigen was found to be quick, simple and low cost with ease of application in field condition without the need of any specialized training and equipments.

Sero-prevalence and associated risk factors of peste des petits ruminants and contagious caprine pleuro-pneumonia in goats and sheep in the Southern Zone of Tanzania

A retrospective Sero-prevalence analysis was conducted in 2012 in order to find out whether contagious caprine pleuro-pneumonia (CCPP) and peste des petits ruminants (PPR) had already been introduced in Mtwara and Lindi regions of Southern Tanzania by 2007 and 2009. A total of 477 randomly selected sera from a bank of 3500 small ruminant samples that were collected as part of Rift Valley Fever surveillance of 2007 in Mtwara and Lindi regions were used in this study. Seroconversion was also evaluated in the 504 sera that were collected in 2009 as part of disease outbreak investigations in Tandahimba and Newala districts of Mtwara region. Seroconversions to CCPP and PPR were tested using competitive ELISA. In addition, information on different variables available in the existing surveillance forms gathered during sampling was used in the analysis of risk factors associated with seropositivity to the two diseases. The overall seroprevalence of CCPP for the sera of 2007 and 2009 in goats was 52.1% (n=447) and 35.5% (n=434) respectively; while in sheep the seroprevalence was 36.7% (n=30) and 22.9% (n=70) respectively. Seroconversion to PPR in goats and sheep was 28.7% (n=434) and 35.7% (n=70) respectively based on the sera of 2009. However, no antibodies were detected in the 2007 sera. Mixed infections were detected in 7.4% (n=434) of the goat and 12.9% (n=70) of sheep samples. Significant risk factors associated with seropositivity to CCPP in 2007 included introduction of new animals in flocks (OR=3.94; 95% CI 1.86–8.36; p<0.001) and raising animals in government farms (OR=4.92; 95% CI 1.57–15.76; p=0.02); whereas, seropositivity to CCPP in 2009 increased with introduction of new animals in flocks (OR=18.82; 95% CI 8.06–43.96; p<0.001), raising animals in government farms (OR=4.04; 95% CI 2.69–6.42; p<0.001) and raising animals in Newala district (OR=2.35; 95% CI 1.53–3.62; p<0.001). On the other hand, predictors for seropositivity to PPR in 2009 were introduction of new animals in flocks (OR=2.83; 95% CI 1.73–4.62; p<0.001) and communal grazing of animals (OR=7.60; 95% CI 1.77–32.58; p=0.01). Therefore, these results show that CCPP was already circulating in goats in the southern zone by 2007 and that PPR was probably introduced thereafter. Their presence in this emerging animal keeping area in Tanzania calls for improved surveillance and control systems.

Efficacy of marbofloxacin for naturally occurring contagious caprine pleuropneumonia

The aim of the present study is to determine the efficacy of marbofloxacin used in goats with naturally occurring contagious caprine pleuropneumonia (CCPP). The study was performed in two groups (consisting of 15 animals in each group) with two different doses of 10% aqueous solution of marbofloxacin injected intramuscularly into the semitendinous muscle. 2 mg/kg BW for 3 days (total dose administered: 6 mg/kg BW) was injected to the first group (group 1) and 3 mg/kg for 2 times every other day (total dose administered: 6 mg/kg BW) was injected to the second group (group 2). Microbiological analyses revealed that the causative agent of the disease was Mycoplasma capricolum subsp. capripneumoniae. Cure rates for groups 1 and 2 were determined as 100% (15/15 goats) and 93% (14/15 goats), respectively. The results of this field trial suggest that marbofloxacin could be an effective drug against CCPP in goats.

Effect of danofloxacin (Advocin A180) on goats affected with contagious caprine pleuropneumonia

The efficacy of danofloxacin (Advocin A180) was evaluated for the treatment of contagious caprine pleuropneumonia (CCPP) caused by Mycoplasma capricolum subsp. capripneumoniae. Ten healthy Angora goats, confirmed free of CCPP, were exposed to clinically affected animals from a natural outbreak in Thrace, Turkey. After 14 days exposure, 8 goats showed pyrexia ( > or = 41 degrees C). Shortly after, the Angora goats were divided randomly into two groups. Five of these were injected with danofloxacin (6 mg/kg subcutaneously), which was repeated after 48 h; the five remaining animals received saline. Goats were monitored clinically and blood samples were collected for serology. Animals with severe disease were withdrawn from the trial. Goats completing the study were euthanized at day 42. Lung tissue and bronchial fluid were collected for mycoplasma isolation. All danofloxacin-treated goats showed resolution of clinical disease by the end of the trial. Two saline-treated goats failed to complete the study owing to CCPP. Danofloxacin-treated goats showed fewer lung lesions and had significantly lower combined clinical scores than saline controls (p < 0.001). Danofloxacin was found to be highly effective in the treatment of CCPP in goats.
