Localized synthesis of molecular chaperones sustains neuronal proteostasis.

Neurons are challenged to maintain proteostasis in neuronal projections, particularly with the physiological stress at synapses to support intercellular communication underlying important functions such as memory and movement control. Proteostasis is maintained through regulated protein synthesis and degradation and chaperone-assisted protein folding. Using high-resolution fluorescent microscopy, we discovered that neurons localize a subset of chaperone mRNAs to their dendrites, particularly more proximal regions, and increase this asymmetric localization following proteotoxic stress through microtubule-based transport from the soma. The most abundant chaperone mRNA in dendrites encodes the constitutive heat shock protein 70, HSPA8. Proteotoxic stress in cultured neurons, induced by inhibiting proteasome activity or inducing oxidative stress, enhanced transport of Hspa8 mRNAs to dendrites and the percentage of mRNAs engaged in translation on mono and polyribosomes. Knocking down the ALS-related protein Fused in Sarcoma (FUS) and a dominant mutation in the heterogenous nuclear ribonucleoprotein A2/B1 (HNRNPA2B1) impaired stress-mediated localization of Hspa8 mRNA to dendrites in cultured murine motor neurons and human iPSC-derived neurons, respectively, revealing the importance of these RNA-binding proteins in maintaining proteostasis. These results reveal the increased dendritic localization and translation of the constitutive HSP70 Hspa8 mRNA as a crucial neuronal stress response to uphold proteostasis and prevent neurodegeneration.

Elevation of major constitutive heat shock proteins is heat shock factor independent and essential for establishment and growth of Lgl loss and Yorkie gain-mediated tumors in Drosophila.

Cancer cells generally overexpress heat shock proteins (Hsps), the major components of cellular stress response, to overcome and survive the diverse stresses. However, the specific roles of Hsps in initiation and establishment of cancers remain unclear. Using loss of Lgl-mediated epithelial tumorigenesis in Drosophila, we induced tumorigenic somatic clones of different genetic backgrounds to examine the temporal and spatial expression and roles of major heat shock proteins in tumor growth. The constitutively expressed Hsp83, Hsc70 (heat shock cognate), Hsp60 and Hsp27 show elevated levels in all cells of the tumorigenic clone since early stages, which persists till their transformation. However, the stress-inducible Hsp70 is expressd only in a few cells at later stage of established tumorous clones that show high F-actin aggregation. Intriguingly, levels of heat shock factor (HSF), the master regulator of Hsps, remain unaltered in these tumorous cells and its down-regulation does not affect tumorigenic growth of lgl- clones overexpressing Yorkie, although down-regulation of Hsp83 prevents their survival and growth. Interestingly, overexpression of HSF or Hsp83 in lgl- cells makes them competitively successful in establishing tumorous clones. These results show that the major constitutively expressed Hsps, but not the stress-inducible Hsp70, are involved in early as well as late stages of epithelial tumors and their elevated expression in lgl- clones co-overexpressing Yorkie is independent of HSF.

De novo transcriptome assemblies of red king crab (Paralithodes camtschaticus) and snow crab (Chionoecetes opilio) molting gland and eyestalk ganglia - Temperature effects on expression of molting and growth regulatory genes in adult red king crab.

Red king crab (Paralithodes camtschaticus) and snow crab (Chionoecetes opilio) are deep-sea crustaceans widely distributed in the North Pacific and Northwest Atlantic Oceans. These giant predators have invaded the Barents Sea over the past decades, and climate-driven temperature changes may influence their distribution and abundance in the sub-Arctic region. Molting and growth in crustaceans are strongly affected by temperature, but the underlying molecular mechanisms are little known, particularly in cold-water species. Here, we describe multiple regulatory factors in the two high-latitude crabs by developing de novo transcriptomes from the molting gland (Y-organ or YO) and eye stalk ganglia (ESG), in addition to the hepatopancreas and claw muscle of red king crab. The Halloween genes encoding the ecdysteroidogenic enzymes were expressed in YO, and the ESG contained multiple neuropeptides, including molt-inhibiting hormone (MIH), crustacean hyperglycemic hormone (CHH), and ion-transport peptide (ITP). Both crabs expressed a diversity of growth-related factors, such as mTOR, AKT, Rheb and AMPKα, and stress-responsive factors, including multiple heat shock proteins (HSPs). Temperature effects on the expression of key regulatory genes were quantified by qPCR in adult red king crab males kept at 4°C or 10°C for two weeks during intermolt. The Halloween genes tended to be upregulated in YO at high temperature, while the ecdysteroid receptor and several growth regulators showed tissue-specific responses to elevated temperature. Constitutive and heat-inducible HSPs were expressed in an inverse temperature-dependent manner, suggesting that adult red king crabs can acclimate to increased water temperatures.

Biological effects of chronic exposure of Blaptica dubia (Blattodea: Blaberidae) nymphs to static and extremely low frequency magnetic fields.

In this paper, we analyzed the effects of chronic exposure (5 months) to static magnetic field (110 mT; SMF) and extremely low frequency magnetic field (ELF MF; 10 mT, 50 Hz) on Blaptica dubia nymphs. We have examined acetylcholinesterase (AChE) activity and heat shock protein 70 (HSP70) level, two sensitive biomarkers of stress in terrestrial insects. Relative growth rate (RGR), as a life history trait, was estimated. AChE activity was determined spectrophotometrically and HSP70 levels were quantified using indirect non-competitive ELISA and Western blotting. Calculated RGR was significantly changed upon exposure to both types of ambiental MFs. The effects of chronic exposure of B. dubia nymphs to SMF and ELF MF (50 Hz) were observed as decreased activity of AChE. The increased level of HSP70 was present only after exposure to SMF. The strength of ELF MF was most likely below the energy level needed to induce the expression of this stress protein. Different patterns of the expression of two HSP70 isoforms, where isoform 2 was sensitive only to SMF, are most likely a possibly switch - off in the expression of constitutive and/or inducible HSP70 isoforms.

Dorsomorphin induces cancer cell apoptosis and sensitizes cancer cells to HSP90 and proteasome inhibitors by reducing nuclear heat shock factor 1 levels.

ObjectiveHeat shock factor 1 (HSF1), a transcriptional regulator of heat shock proteins (HSPs), is an attractive therapeutic target for cancer. However, only a few HSF1 inhibitors have been identified so far.MethodsThe mRNA and protein levels of HSF1, HSPs, cleaved PARP, and phosphorylated HSF1 were examined by real-time PCR and Western blot. Forced expression, RNA interference, and immunofluorescence assay were used for mechanistic studies. Cell viability and apoptosis were measured by WST-8 assay and flow cytometry, respectively. Xenograft studies were performed in nude mice to evaluate the effect of dorsomorphin and an HSP90 inhibitor on tumor growth.ResultsDorsomorphin suppressed multiple stimuli-induced and constitutive HSPs expression in cancer cells. Mechanistic studies revealed that dorsomorphin reduced heat-induced HSP expression independent of adenosine monophosphate activated protein kinase. Dorsomorphin reduced heat-stimulated HSF1 Ser320 phosphorylation and nuclear translocation, as well as resting nuclear HSF1 levels in cancer cells. Dorsomorphin induced cancer cell apoptosis by inhibiting HSF1 expression. A structure-activity study revealed that the 4-pyridyl at the 3-site of the pyrazolo [1, 5-a]pyrimidine ring is critical for the anti-HSF1 activities of dorsomorphin. Dorsomorphin sensitized cancer cells to HSP90 and proteasome inhibitors and inhibited HSP70 expression induced by these inhibitors in vitro. In tumor-bearing nude mice, dorsomorphin enhanced HSP90 inhibitor-induced cancer cell apoptosis, tumor growth inhibition, and HSP70 expression. ConclusionsDorsomorphin is an HSF1 inhibitor. It induces cancer cell apoptosis, sensitizes cancer cells to both HSP90 and proteasome inhibitors, and suppresses HSP upregulation by these drugs, which may prevent the development of drug resistance. Hence, dorsomorphin and its derivates may serve as potential precursors for developing drugs against cancer.

Ocean acidification increases the sensitivity of and variability in physiological responses of an intertidal limpet to thermal stress

Abstract. Understanding physiological responses of organisms to warming and ocean acidification is the first step towards predicting the potential population- and community-level ecological impacts of these stressors. Increasingly, physiological plasticity is being recognized as important for organisms to adapt to the changing microclimates. Here, we evaluate the importance of physiological plasticity for coping with ocean acidification and elevated temperature, and its variability among individuals, of the intertidal limpet Cellana toreuma from the same population in Xiamen. Limpets were collected from shaded mid-intertidal rock surfaces. They were acclimated under combinations of different pCO2 concentrations (400 and 1000 ppm, corresponding to a pH of 8.1 and 7.8) and temperatures (20 and 24 ∘C) in a short-term period (7 days), with the control conditions (20 ∘C and 400 ppm) representing the average annual temperature and present-day pCO2 level at the collection site. Heart rates (as a proxy for metabolic performance) and expression of genes encoding inducible and constitutive heat-shock proteins (hsp70 and hsc70) at different heat-shock temperatures (26, 30, 34, and 38 ∘C) were measured. Hsp70 and Hsc70 play important roles in protecting cells from heat stresses, but have different expression patterns, with Hsp70 significantly increased in expression during stress and Hsc70 constitutively expressed and only mildly induced during stress. Analysis of heart rate showed significantly higher temperature coefficients (Q10 rates) for limpets at 20 ∘C than at 24 ∘C and post-acclimation thermal sensitivity of limpets at 400 ppm was lower than at 1000 ppm. Expression of hsp70 linearly increased with the increasing heat-shock temperatures, with the largest slope occurring in limpets acclimated under a future scenario (24 ∘C and 1000 ppm pCO2). These results suggested that limpets showed increased sensitivity and stress response under future conditions. Furthermore, the increased variation in physiological response under the future scenario indicated that some individuals have higher physiological plasticity to cope with these conditions. While short-term acclimation to reduced pH seawater decreases the ability of partial individuals against thermal stress, physiological plasticity and variability seem to be crucial in allowing some intertidal animals to survive in a rapidly changing environment.

Heat shock proteins expressed in the marsupial Tasmanian devil are potential antigenic candidates in a vaccine against devil facial tumour disease.

The Tasmanian devil (Sarcophilus harrisii), the largest extant carnivorous marsupial and endemic to Tasmania, is at the verge of extinction due to the emergence of a transmissible cancer known as devil facial tumour disease (DFTD). DFTD has spread over the distribution range of the species and has been responsible for a severe decline in the global devil population. To protect the Tasmanian devil from extinction in the wild, our group has focused on the development of a prophylactic vaccine. Although this work has shown that vaccine preparations using whole DFTD tumour cells supplemented with adjuvants can induce anti-DFTD immune responses, alternative strategies that induce stronger and more specific immune responses are required. In humans, heat shock proteins (HSPs) derived from tumour cells have been used instead of whole-tumour cell preparations as a source of antigens for cancer immunotherapy. As HSPs have not been studied in the Tasmanian devil, this study presents the first characterisation of HSPs in this marsupial and evaluates the suitability of these proteins as antigenic components for the enhancement of a DFTD vaccine. We show that tissues and cancer cells from the Tasmanian devil express constitutive and inducible HSP. Additionally, this study suggests that HSP derived from DFTD cancer cells are immunogenic supporting the future development of a HSP-based vaccine against DFTD.

Protective role of TAT-HSP70 after myocardial I/R injury

Myocardial ischemia reperfusion injury I/R adversely affects cardiac function. Heat shock proteins (HSPs) are a highly conserved family of proteins with diverse functions expressed by all cells exposed to environmental stress including myocardila injury. We investigated release of small constitutive heat shock proteins (HSPs) from mouse myocardium and the effects of TAT-HSP70 after myocardial I/R via occluding the left coronary artery (LAD). The results support the hypothesis that elevated HSPs in myocardium after ischemia and reperfusion and contributes to the inflammatory mechanism of myocardial functional injury. Further investigation of the significance of HSPs accumulation to the evolution of myocardial injury.

Expression of HSP70 genes in skin of zebu (Tharparkar) and crossbred (Karan Fries) cattle during different seasons under tropical climatic conditions

Skin is most important environmental interface providing a protective envelope to animals. It's always under the influence of both internal and external stressors. Heat shock proteins (HSP) are highly conserved stress proteins which play crucial roles in environmental stress tolerance and thermal adaptation. Present study was planned to observe the relative mRNA expression of inducible (HSP70.1 and HSP70.2) and constitutive (HSP70.8) HSP in skin of zebu (Tharparkar) and crossbred (Karan Fries) cattle during different seasons. Skin biopsies were collected from rump region of each animal, aseptically during winter, spring and summer season. Quantitative real time polymerase chain reaction was performed to examine the gene expression of constitutive (HSP70.8) and inducible (HSP70.1 and HSP70.2) HSP in skin of both the breeds during different seasons. Present study observed higher expression of both constitutive and inducible HSP genes in both the breeds during summer and winter than spring season, but magnitude of increase was higher during summer than winter. During summer season, expression pattern of HSPs in skin showed breed differences, where constitutive HSP expression was higher in Tharparkar than Karan Fries and that of inducible HSP was higher in Karan Fries than Tharparkar. Hence, present study suggested that HSP may be conveniently used as biomarkers for assessing protective response of skin against heat stress in zebu and crossbred cattle. Variation in expression between breeds is associated with their heat tolerance and thermal adaptability. In summary, skin of zebu cattle (Tharparkar) is more resistant to summer stress than crossbred (Karan Fries), providing greater protection against heat stress during summer season. Superior skin protective mechanism of zebu (Tharparkar) than crossbred (Karan-Fries) cattle against heat stress may contribute to superior adaptability of zebu cattle to tropical climatic conditions than crossbreed.

Heat stress induces formation of cytoplasmic granules containing HSC70 protein.

Using indirect immunofluorescence, in this study we showed that the constitutive heat shock protein HSC70 forms granule-like structures in the cytoplasm of human cells several days after the exposure to heat stress. It was shown that this effect is not the result of HSC70 overexpression under heat stress and is not due to the formation of hyperthermia-induced translational stress granules in the cytoplasm.

Materials & Methods: Five week old male mice, [non-transgenic (NTG; n=19) and Hsp70 transgenic littermates (TG; n=14; villin promoter driven epithelial cell Hsp70 expression)] were randomly divided into low fat (LF; 10 kcal% fat) or high fat (HF; 60 kcal% fat) diet groups (NTG/LF, NTG/HF, TG/LF, TG/HF). All mice continued on the protocol for 12 weeks. At week 13, colonic scrapings were collected and homogenized for determining Hsp70 levels (ELISA, R&D Systems) and tight junction protein expression (Western blot). Western blots probed for occludin (Life Technologies) and constitutive heat shock protein 70 (HSC70, Santa Cruz Biotech) were run using pooled samples of each group (n= 6-10 per group). Densitometry readings were completed using a ChemiDoc MP Imager (Bio-Rad) and data normalized using HSC70 as a loading control with NTG/LF as the reference sample. Hsp70 data was analyzed by 2-way ANOVA and presented as a mean ± SEM. Results: Measurement of colonic Hsp70 levels showed a significant (p < 0.05, Fig. 1), 100 fold, increase in the TG group compared to its NTG littermates. Western blot for occludin using pooled samples representative of each group suggested TG mice expressed an increased amount of this protein compared to their NTG littermates regardless of diet (Fig. 2). Discussion: Hsp70 TG mice demonstrated increased levels of occludin, suggesting that it may enhance gut barrier function by increasing tight junction proteins independent of diet. Further examination of Hsp70's affect on colonic tight junction proteins may provide a possible target for the prevention and treatment of obesity

Abstract 3625: Tumor-specific human monoclonal antibodies isolated from cancer patients

Abstract Introduction: New cancer-specific antigens are needed to fully realize the potential of antibody-based cancer therapies. Cancer cells express novel antigens that are characteristic of the transformed state and may engender an antibody response in patients. We hypothesize that some of these antibodies detect neo-antigens shared by diverse tumor types. We optimized a hybridoma method for cloning human IgG monoclonal antibodies (mAbs) by screening for binding to heterologous cell lines. Here, we describe four mAbs, isolated from cancer patients, that bind to cancer cells. Methods: We developed immunofluorescence binding assays with the A431, MCF-7, and MDA-MB-231 cell lines, detecting cell-reactive human IgG with the Operetta (Perkin Elmer). The assays were adapted for extracellular binding as well as internalization using specific culture conditions. We tested sera from 59 lung cancer patients with diverse clinical features. 24 of the samples contained IgG reactivity with A431 cells at &amp;gt; 1:100 titer, and 17 had evidence of IgG internalization. We generated hybridomas using peripheral blood B-cells isolated from two of these lung cancer patients, as well as B-cells from a lymph node of a stage I, ER+PR+ breast cancer patient. We characterized the binding of four mAbs using immunohistochemistry (IHC), microscopy, flow cytometry and immunoblotting. One mAb was used to identify its cognate antigen by immunoprecipitation and mass spectrometry. Results: In a panel of 59 lung cancer patient sera, 24 had IgG that bound the A431 cell line, and 17 of these had evidence of internalization. We isolated three human mAbs from two lung cancer patients and one mAb from the breast cancer lymph node. We performed IHC with three of the mAbs in formalin fixed, paraffin embedded normal and tumor tissues and found primarily tumor cell binding to a variety of tumor types. The 5A6 (lung) bound 3 of 4 non-small cell lung, 3 of 4 colon, 2 of 4 ovarian and 1 of 4 small cell lung cancers. 19F10 (lung) bound 1 of 4 colon and 2 of 4 small cell lung. 2G4 (breast) bound 4 of 4 non-small cell lung, 3 of 4 prostate, 3 of 4 ovarian, and 2 of 4 colon cancers. Some normal tissue binding was observed, such as small intestine and distal renal tubule (2G4 and 19F10), pancreatic islet (19F10), and prostate (2G4). The 6A10 mAb (lung) bound to the constitutive heat shock protein, HSC70, which was localized in the plasma membrane of cancer cell lines. Conclusion: We have cloned 4 human mAbs from cancer patients. This supports the hypothesis that patients with active malignancies make antibodies capable of binding cancer-specific antigens. We identified HSC70, a protein that has an altered cell localization in cancer cells, as one such antigen. The three mAbs tested by IHC bound to diverse tumor types that often differed from the tumors of the patients where the B-cells originated. The human anti-cancer antibody repertoire holds promise as a tool to identify new targets for mAb-based cancer diagnostics and therapeutics. Correlation between depression, anxiety, and polymorphonuclear cells' resilience in ulcerative colitis: the mediating role of heat shock protein 70.

BackgroundTo investigate whether anxiety and depression levels are associated with Heat Shock Protein 70 (HSP70) induction in the colon of patients with ulcerative colitis (UC).MethodsThe design was cross-sectional. Clinical activity was assessed by the Rachmilewitz Index (CAI). Three psychometric questionnaires were used: Zung Depression Rating Scale (ZDRS), Spielberg State-Trait Anxiety Inventory (STAI), Hospital Anxiety and Depression Scale (HADS). Colon biopsies were obtained from each affected anatomical site. Severity of inflammation was assessed by eosin/hematoxylin. Constitutive (HSP70c) and inducible (HSP70i) HSP70 expression were immunohistochemically studied.Results29 UC patients were enrolled (69% men). Mean age was 46.5 years (SD: 19.5). Inflammation severity was moderate in 17 patients, severe in 6, and mild in 6. The mean number of years since diagnosis was 7.9 (SD: 6.5). The mean CAI was 6.4 (SD: 3.1). In active UC, there was downregulation of HSP70c in inflamed epithelium, without significant HSP70 induction. In 22/29 cases of active cryptitis, polymorphonuclear cells (PMN) clearly expressed HSP70i, with weak, focal positivity in the other 7 cases. Except for the hospital anxiety scale, scores in all psychometric tools were higher in patients with strong HSP70i immunoreactivity in the PMN. Logistic regression showed a strong positive relationship between HSP70i immunoreactivity in the PMN cells and scores in the trait anxiety, ZDRS, and hospital depression scales, (Odds ratios 1.3, 1.3, and 1.5; P = 0.018, 0.023, and 0.038; Wald test, 5.6, 5.2, and 4.3 respectively) and a weaker but significant positive correlation with the CAI (Odds ratio 1.654; P = 0.049; Wald test 3.858).ConclusionHSP70 is induced in PMN cells of UC patients and its induction correlates with depression and anxiety levels.

Intraspecific Variation in Thermal Tolerance and Acclimation Capacity in Brook Trout (Salvelinus fontinalis): Physiological Implications for Climate Change

Cold-water fishes are becoming increasingly vulnerable as changing thermal conditions threaten their future sustainability. Thermal stress and habitat loss from increasing water temperatures are expected to impact population viability, particularly for inland populations with limited adaptive resources. Although the long-term persistence of cold-adapted species will depend on their ability to cope with and adapt to changing thermal conditions, very little is known about the scope and variation of thermal tolerance within and among conspecific populations and evolutionary lineages. We studied the upper thermal tolerance and capacity for acclimation in three captive populations of brook trout (Salvelinus fontinalis) from different ancestral thermal environments. Populations differed in their upper thermal tolerance and capacity for acclimation, consistent with their ancestry: the northernmost strain (Lake Nipigon) had the lowest thermal tolerance, while the strain with the most southern ancestry (Hill's Lake) had the highest thermal tolerance. Standard metabolic rate increased following acclimation to warm temperatures, but the response to acclimation varied among strains, suggesting that climatic warming may have differential effects across populations. Swimming performance varied among strains and among acclimation temperatures, but strains responded in a similar way to temperature acclimation. To explore potential physiological mechanisms underlying intraspecific differences in thermal tolerance, we quantified inducible and constitutive heat shock proteins (HSP70 and HSC70, respectively). HSPs were associated with variation in thermal tolerance among strains and acclimation temperatures; HSP70 in cardiac and white muscle tissues exhibited similar patterns, whereas expression in hepatic tissue varied among acclimation temperatures but not strains. Taken together, these results suggest that populations of brook trout will vary in their ability to cope with a changing climate.

Human myocardium releases heat shock protein 27 (HSP27) after global ischemia: the proinflammatory effect of extracellular HSP27 through toll-like receptor (TLR)-2 and TLR4.

The myocardial inflammatory response contributes to cardiac functional injury associated with heart surgery obligating global ischemia/reperfusion (I/R). Toll-like receptors (TLRs) play an important role in the mechanism underlying myocardial I/R injury. The aim of this study was to examine the release of small constitutive heat shock proteins (HSPs) from human and mouse myocardium after global ischemia and examine the role of extracellular small HSP in myocardial injury. HSP27 release was assessed by enzyme-linked immunosorbent assay. Anti-HSP27 was applied to evaluate the role of extracellular HSP27 in the postischemic inflammatory response and functional injury in mouse hearts. Isolated hearts and cultured coronary vascular endothelial cells were exposed to recombinant HSP27 to determine its effect on proinflammatory signaling and production of proinflammatory mediators. HSP27 levels were markedly elevated in coronary sinus blood of patients and in coronary effluent of mouse hearts after global ischemia. Neutralizing extracellular HSP27 suppressed myocardial nuclear factor (NF)-κB activation and interleukin (IL)-6 production and improved cardiac function in mouse hearts. Perfusion of HSP27 to mouse hearts induced NF-κB activation and IL-6 production and depressed contractility. Further, recombinant HSP27 induced NF-κB phosphorylation and upregulated monocyte chemoattractant protein (MCP)-1 and intercellular adhesion molecule (ICAM)-1 production in both human and mouse coronary vascular endothelial cells. TLR2 knockout (KO) or TLR4 mutation abolished NF-κB phosphorylation and reduced MCP-1 and ICAM-1 production induced by extracellular HSP27 in endothelial cells. In conclusion, these results show that the myocardium releases HSP27 after global ischemia and that extracellular HSP27 is proinflammatory and contributes to the inflammatory mechanism of myocardial functional injury. Both TLR2 and TLR4 are involved in mediating the proinflammatory effect of extracellular HSP27.

Temporal changes in stress and tissue-specific metabolic responses to municipal wastewater effluent exposure in rainbow trout

Sub-chronic exposure to municipal wastewater effluent (MWWE) in situ was recently shown to impact the acute response to a secondary stressor in rainbow trout (Oncorhynchus mykiss). However, little is known about whether MWWE exposure in itself is stressful to the animal. To address this, we carried out a laboratory study to examine the organismal and cellular stress responses and tissue-specific metabolic capacity in trout exposed to MWWE. Juvenile rainbow trout were exposed to 0, 20 and 90% MWWE (from a tertiary wastewater treatment plant), that was replenished every 2d, for 14d. Fish were sampled 2, 8 or 14d post-exposure. Plasma cortisol, glucose and lactate levels were measured as indicators of organismal stress response, while inducible heat shock protein 70 (hsp70), constitutive heat shock protein 70 (hsc70) and hsp90 expression in the liver were used as markers of cellular stress response. Impact of MWWE on cortisol signaling was ascertained by determining glucocorticoid receptor protein (GR) expression in the liver, brain and, heart, and metabolic capacity was evaluated by measuring liver glycogen content and tissue-specific activities of key enzymes in intermediary metabolism. Plasma glucose and lactate levels were unaffected by exposure to MWWEs, whereas cortisol showed a transient increase in the 20% group at 8d. Liver hsc70 and hsp90, but not hsp70 expression, were higher in the 90% MWWE group after 8d. There was a temporal change in GR expression in the liver and heart, but not brain of trout exposed to MWWE. Liver glycogen content and activities liver gluconeogenic enzyme phosphoenolpyruvate carboxykinase (PEPCK), and alanine aminotransferase (AlaAT) were significantly affected by MWWE exposure. The glycolytic enzymes pyruvate kinase (PK) and hexokinase (HK) activities were significantly higher temporally by MWWE exposure in the gill and heart, but not in the liver and brain. Overall, a 14d exposure to MWWE evokes a cellular stress response and perturbs the cortisol stress response in rainbow trout. The tissue-specific temporal changes in the metabolic capacity suggest enhanced energy demand in fish exposed to MWWE, which may eventually lead to reduced fitness.

Temperature as a cellular stress of Chinook salmon embryo cells

In this study, we used Chinook salmon embryo cells (CHSE‐214) to assess the relationship between the timing of the expression of heat shock mRNAs and the timing of heat‐induced cell death. We first examined the timing of cell death [lactate dehydrogenase (LDH) release] after a 24hr exposure to 22, 30, 37, 45, and 55°C. As expected, cells did not release LDH after 24 hrs of exposure to their acclimation temperature (22°C). At 30°C LDH release was detected beginning at 6, and all of the cells were dead by 24hrs. Exposure to 37°C, LDH released at 3hrs; all cells were dead by 6hrs. At 45°C the cells released LDH within the first hour, and were all dead by 3hrs of exposure. At 55°C the cells began to release LDH within the first 15mins, and were all dead within 1hr of exposure. mRNAs for heat shock proteins were assessed at 37°C and it was found that the constitutive heat shock protein (Hsc70) was present at all time points, and there was no significant difference in the expression of the message for the inducible form of the heat shock protein (Hsp70) over a 6 hour period. These data will help us to further understand the molecular meaning of the critical thermal max of fish. Protein deprivation attenuates Hsp expression in fat tissue

For ruminants, dietary protein is the first limiting component to the utilization of low-quality forage. Throughout gestation, low-protein intake may result in prenatal programming that causes various metabolic disturbances and physiological modulations to dams and their developing embryos. We studied the effect of long-term low-protein diet (LPD) on physiological, biochemical, and molecular parameters of the energy status in gestating beef cows. LPD resulted in significant reductions in feed intake and heart rate and promoted a negative retained energy status already after 3weeks. Elevated levels of plasma creatinine and non-esterified fatty acids indicate endogenous degradation of fat and protein as a response to the demands in energy and nitrogen. Increasing levels of β-hydroxybutyrate confirmed the negative energy status obtained by the physiological measurements. At the molecular level, subcutaneous fat, Hsp90, Hsp70, and proteasome subunits decreased significantly after 3months on LPD, in parallel with an increase of adipocyte fatty acid-binding protein. These results may indicate a decrease in turn-over of proteins, at the cost of induced lipolysis, and suggest that the response to protein deprivation, when examined in an energy-storing tissue, includes downregulation of the constitutive heat shock proteins involved in the protein degradation pathway of energy production and upregulation of tissue-specific genes such as those involved in energy production from fat degradation.

Constitutive heat shock protein 70 interacts with α-enolase and protects cardiomyocytes against oxidative stress

Constitutive heat shock protein 70 (Hsc70) is a molecular chaperone that has been shown to protect cardiomyocytes against oxidative stress. However, the molecular mechanism responsible for this protection remains uncertain. To understand the mechanism associated with the myocardial protective role of Hsc70, we have embarked upon a systematic search for Hsc70-interacting proteins. Using adenosine diphosphate (ADP) affinity chromatography and mass spectrometry, we have identified α-enolase, a rate-limiting enzyme in glycolysis, as a novel Hsc70-interacting protein in the myocardium of both sham and myocardial ischemia-reperfused Sprague–Dawley rat hearts. This interaction was confirmed by co-immunoprecipitation (IP) assays in the myocardial tissues and H9c2 cardiomyocytes and protein overlay assay (POA). It was further shown that Hsc70-overexpression alleviated the H2O2-induced decrease of α-enolase activity and cell damage, and Hsc70 deficiency aggravated the decrease of α-enolase activity and cell damage in H2O2 treated H9c2 cells. Our research suggests that the protective effect of Hsc70 on the cardiomyocytes against oxidative stress is partly associated with its interaction with α-enolase.

Molecular characterization and expression of a heat-shock cognate 70 (Hsc70) and a heat-shock protein 70 (Hsp70) cDNAs in Rana (Pelophylax) lessonae embryos

Heat-Shock Protein 70 (HSP70) is a class of highly conserved proteins which is involved in essential functions as molecular chaperones and in the acquired tolerance processes. In this work, two cDNAs encoding a constitutive Hsc70 and an inducible Hsp70 from the water frog Rana (Pelophylax) lessonae, belonging to the Rana (P.) esculenta complex of central Italy, have been isolated and characterized. The two cDNA clones, named Rl-Hsc70 and Rl-Hsp70, encode 646 and 640 amino acid proteins respectively, which present extremely conserved functional domains characteristic of cytosolic members of the HSP70 family. Comparative studies of the amino acid sequences showed that Rl-Hsc70 and Rl-Hsp70 had the highest homology with constitutive and inducible HSP70 members of other amphibian species. The phylogenetic analysis also demonstrated a separate clustering of the Rl-Hsc70 and Rl-Hsp70 with constitutive and inducible members from other vertebrate species. Heat-inducibility assays performed during embryogenesis showed that the two isolated mRNAs displayed different expression profiles. Rl-Hsp70 was induced only in heat shock-treated embryos, whereas Rl-Hsc70 transcript levels, which were constitutively modulated in non-stressed embryos, did not increase following the heat treatment. In situ hybridization experiments demonstrated that both transcripts showed a tissue-specific enrichment in the central nervous system and in the somites.
