Effect of alternating total mixed ration and pasture feeding on the fatty acid content and health indices of Jersey and Fleckvieh x Jersey milk

Certain fatty acids (FAs), such as omega-3 and conjugated linoleic acid (CLA) are considered essential FAs with beneficial health effects for humans. Milk is considered a relatively inexpensive and readily available source of these FAs and is part of a recommended healthy daily diet. The aim of the study was to determine the effect of diet changes on the FA composition of Jersey (J) and Fleckvieh x Jersey (FxJ) milk. Cows were alternately put on kikuyu-ryegrass pasture, followed by a feedlot system, then returned to kikuyu-ryegrass pasture for four weeks each. The same concentrate mixture was fed to cows regardless of feeding system. The feedlot system consisted of a partial total mixed ration (pTMR). Milk samples were collected two and four weeks after diet changes and stored at −20 °C until laboratory analysis by gas chromatography. The FA concentration of milk was not affected by breed, although it was affected significantly by diet changes. Most notably, the total omega-3 and -6 FAs decreased when cows were fed pTMR while increasing when the diet was changed back to pasture feeding. The CLA content of milk was similarly affected. That is, concentrations decreased significantly when the cows were on the pTMR diet and increased when cows were put back on the pasture-based diet. The results suggested that the health benefits of milk fat are affected negatively when cows are fed pTMR compared with being fed in a pasture-based system. The health benefits of milk are reduced owing to decreased levels of the CLA content of milk fat. Therefore, feeding additional hay in a pasture-based production system should be reconsidered when aiming to produce milk that provides health beneficial qualities. An overview of conjugated linoleic acid: microbial production and application.

Conjugated linoleic acid (CLA) has attracted considerable attention in health due to its important physiological properties proved in several in vivo experiments. Many bacteria, especially some probiotics, are able to produce CLA from the linoleic acid (LA) present in milk. In this review, CLA production by microorganisms is described. Then factors on the influencing the microbial production and the initial CLA content in milk fat are introduced. After a glimpse on the content of CLA in dairy products and human body, health benefits of CLA including anti-cancer, anti-diabetic, antiathrosclerosis and anti-osteoporosis properties, as well as prevention of body fat increase and function as stimulator of the immunity system are explained.

Enrichment of Milk with Conjugated Linoleic Acid by Supplementing Diets with Fish and Sunflower Oil

There is an increase interesting in enrichment of milk with Conjugated Linoleic Acid (CLA) due to its anti-oxidative and anti-carcinogenic properties. The objective of this study was to investigate the effect of supplementing diets fed to lactating goats with sunflower, fish oil and its blend. Eight lactating Nubian goats were fed a base diet (T1), diet supplemented with 2% sunflower oil (on dry matter (DM) basis) (T2), diet supplemented with 2% fish oil (T3) and diet supplemented with 2% sunflower and fish oil (T4) for 84 day. Milk composition milk fat, protein (%) decreased in T2, T3 and T4 compared with control (T1) while there was no significant differences between treatments in milk lactose content. CLA content in milk fat was higher in response to fish oil or sunflower and fish oil blend compared with control (T1). The results indicated that supplementing diets fed to lactating goats with sunflower, fish oil increased CLA contents in the milk 2-4 times than control.

Short communication. Effect of forage source (grazing vs. silage) on conjugated linoleic acid content in milk fat of Holstein-Friesian dairy cows from Galicia (NW Spain)

The aim of this study was to investigate the effect of different feeding proportions of forage ―grazing vs. silage― on milk fatty acids (FA) profile and conjugated linoleic acid (CLA) content of autumn calving Holstein-Friesian cows (n = 61) at CIAM (Galicia, NW Spain). Three treatments (S, 100% silage; G/S, 50% grazing + 50% silage; G, 100% grazing) were set and milk FA profile of dairy cows was determined by gas chromatography-mass spectrometry. The G group showed a decrease in short (p < 0.05) and medium chain FA (p < 0.001), with an increase in long chain FA (p < 0.001) in comparison to the G/S and S groups, which showed the lowest levels (p < 0.001) of mono- and polyunsaturated FA. The CLA content in milk fat increased (p < 0.001) linearly in relation to the increased proportion of fresh grass in the diet of dairy cows from 0.49 and 0.82 to 1.14 g/100 g FA for the treatments S, G/S and G, respectively. During spring and summer, the levels of CLA were three times higher (p < 0.001, +0.76 g/100 g FA) in milk from dairy cows at the G group than in cows at the S group and twice higher (p < 0.001, +0.40 g/100 g FA) than in cows at the G/S group. High proportion of grass in the diet of cows increased CLA content, with the highest levels of unsaturated FA and the lowest levels of saturated FA, increasing the added value of milk on grazing systems using available farm resources.

A comparative study of fatty acid composition and CLA concentration in commercial cheeses

A total of 52 Italian and French commercial cheeses were analysed for fatty acid composition and conjugated linoleic acid (CLA) concentration. The cheeses were first grouped according to the ruminant species of the milk used in manufacturing, then according to the following criteria: (1) cheeses obtained from milk of the same ruminant species but using different production technologies; (2) cheeses produced from milk of different ruminant species but through similar cheesemaking processes. Comparative studies of fatty acid composition and CLA levels in the different cheese classes were carried out. In our study, the arrangement of the ruminant species according to the increasing CLA concentration in cheese was: goat = cow < sheep. Sheep cheeses had the highest levels of CLA (9.86 mg/g fat), α-linolenic acid (0.75%) and trans-vaccenic acid (1.63%), and the lowest contents of linoleic acid (1.80%) and oleic acid (16.83%). The comparison of cheeses obtained from milk of the same ruminant species but through different production technologies reported statistically significant differences in the fatty acid profiles that could be due to a different degree of lipolysis in the cheeses compared. Nevertheless, statistically significant differences in CLA concentration were not found; this suggests that the factors involved in the cheesemaking process generally do not affect the CLA content in milk fat.

Influence of Supplementing Dairy Cows Grazing on Pasture with Feeds Rich in Linoleic Acid on Milk Fat Conjugated Linoleic Acid (CLA) Content

Three experiments were conducted to investigate the hypothesis that cows grazing on pasture produce the highest proportion of c-9 t-11 CLA in milk fat and no further increase can be achieved through supplementation of diets rich in linoleic acid, such as full-fat extruded soybeans or soybean oil. In experiment 1, 18 lactating Holstein cows were used in a randomized complete block design with measurements made from wk 4 to 6 of the experiment. In experiment 2, three cannulated lactating Holstein cows were used in a 3x3 Latin square design. Each period was 4 wk with measurements made in the final wk of each period. Cows in both experiments were assigned at random to treatments: a, conventional total mixed ration (TMR); b, pasture (PS); or c, PS supplemented with 2.5 kg/cow per day of full-fat extruded soybeans (PES). In both experiments, feed intake, milk yield, milk composition, and fatty acid profile of milk and blood serum were measured, along with fatty acid composition of bacteria harvested from rumen digesta in experiment 2. In experiment 3, 10 cows which had continuously grazed a pasture for six weeks were assigned to two groups, with one group (n = 5) on pasture diet alone (PS) and the other group (n = 5) supplemented with 452 g of soy oil/cow per day for 7 d (OIL). In experiment 1, cows in PS treatment produced 350% more c-9, t-11 CLA compared with cows in TMR treatment (1.70 vs. 0.5% of fat), with no further increase for cows in PES treatment (1.50% of fat). Serum c-9, t-11 CLA increased by 233% in PS treatment compared with TMR treatment (0.21 vs. 0.09% of fat) with no further increase for cows in PES treatment (0.18% of fat). In experiment 2, cows in PS treatment produced 300% more c-9 t-11 CLA in their milk fat compared with cows in TMR treatment (1.77 vs. 0.59% of fat), but no further increase for cows in PES treatment (1.84% of fat) was observed. Serum c-9, t-11 CLA increased by 250% for cows in PS treatment compared with cows in TMR treatment (0.27 vs. 0.11% of fat), with no further increase for cows in PES treatment (0.31% of fat). The c-9, t-11 CLA content of ruminal bacteria for cows in PS treatment was 200% or more of TMR treatment, but no further increase in bacterial c-9, (-11 CLA for cows in PES treatment was observed. Supplementation of soy oil in experiment 3 also did not increase the c-9 t-11 CLA content of milk fat compared with cows fed a full pasture diet (1.60 vs. 1.54% of fat). Based on these findings, it was concluded that supplementing with feeds rich in linoleic acid, such as full-fat extruded soybeans or an equivalent amount of soy oil, to cows grazing perennial ryegrass pasture may not increase milk fat c-9 t-11 CLA contents.

Changes in fatty acid composition of milk from lactating dairy cows during transition to and from pasture

Five Holstein cows producing an average of 25.4 ± 6.4 kg/d and 347 ± 111 days in milk were used to study the changes in fatty acid composition in milk from lactating dairy cows during transition to and from pasture. The 45-d experiment was divided into 3 periods. During the first 2 d of the experiment (Period I), cows were fed a TMR diet containing 50% conserved forage and 50% grain. On day 3, cows were turned out to pasture and remained on all-pasture diet for 29 d (Period II). On day 32, cows were withdrawn from pasture and offered a TMR diet until day 45 (Period III). Milk yield was recorded daily, and milk fat content and fatty acid (FA) composition were determined daily for composite samples collected from a.m. and p.m. milkings. Data were analyzed using spline regression ( H 0 = zero slope) to determine whether there was any change in the concentration of a particular fatty acid after a specific time or whether it had stabilized. Cows produced an average of 25.2 ± 5.5, 13.7 ± 5.6, and 12.1 ± 4.8 kg/d of milk with 3.6, 4.2, and 3.6% fat during pre-pasture, pasture, and post-pasture diets, respectively. The conjugated linoleic acid (CLA) content was 0.45% of total fat during pre-pasture, reached to a maximum of 2.53% on day 23 into pasture and plateaued thereafter for the period. Milk fat C 18:1 t-11 content was 2.89% of total fat during pre-pasture, reached a maximum of 7.95% after day 22 in pasture, and plateaued afterwards. The C 18:2 content declined gradually from pre-pasture to pasture diet with no further decrease observed after day 22, while C 18:3 content increased until day 7 on pasture. The post-pasture CLA content in milk fat reached a value similar to pre-pasture within 4 days after the cows were withdrawn from pasture. No change in other fatty acids was observed after day 7 once the cows were switched to post-pasture diet. In the present study, it took 23 days to establish the highest level of CLA in milk fat after turning cows out to pasture, whereas only 4 days were needed to bring it back to the original level once the cows were withdrawn from pasture. Other milk fatty acids were stabilized around day 23 after turning cows out to pasture and by day 7 after being withdrawn from pasture and put back on a TMR diet inside the barn.

Effects of fatty acid sources on conjugated linoleic acid (CLA) and other fatty acids in dairy milk

Conjugated linoleic acid (CLA) is aniticarcinogenic, antiatherogenic and antidiabetogenic actives. Research has therefore focused on methods of increasing CLA content in milk fat. Amount of CLA in milk fat was highly related to biohydrogenation of unsaturated fatty acid of rumen microbes. (Bauman et al., 1999). Linoleic acid (C18:2) were the precursors of CLA synthesis. The CLA was also synthesized in the mammary gland of lactating ruminants, using oleic acid (C18:1) as a precursor and activity of delta 9-desaturase (Griinari and Bauman, 1999). Linoleic acid is high in soybean oil (SO) (54.4%) and tuna oil (TO) (20.3%) while oleic acid is high in pork oil (PO) (43.5%) and groundnut oil (GO) (40.7%). Therefore, the objective of this experiment was to compare the increasing of CLA and fatty acid composition in milk fat form cows fed dietary oils obtained from either animal or plant sources.

Effects of Dietary Sources of Vegetable Oils on Performance of High-Yielding Lactating Cows and Conjugated Linoleic Acids in Milk

This study was conducted to examine the effects of dietary supplementation with vegetable oils on performance of high-yielding lactating cows and conjugated linoleic acid (CLA) content in milk fat. Twelve lactating Holstein cows in early lactation (30 to 45 d postpartum) were used in a triple 4×4 Latin square design. In each period, the cows in each group were fed the same basal diet and received one of the following treatments: 1) control (without oil), 2) 500g of cottonseed oil, 3) 500g of soybean oil, and 4) 500g of corn oil. Each experimental period lasted for 3 wk, with the first 2 wk used for adaptation to the diet. Supplementation with vegetable oils tended to increase milk yield, with the highest milk yield in the cottonseed oil group (35.0 kg/d), compared with the control (34.4 kg/d). Milk fat percentage was decreased, but there were few effects on percentage and yield of milk protein as well as milk fat yield. The cows fed added soybean oil produced milk with the highest content of trans-11C18:1 (23.8 mg/g of fat), which was twice that of the control (12.6 mg/g of fat). Content of cis-9, trans-11 CLA in milk fat increased from 3.5 mg/g in the control to 6.0, 7.1, and 10.3 mg/g for the cows fed oils from cottonseed, corn, and soybean, respectively. A significant linear relationship existed between trans-11C18:1 and cis-9, trans-11 CLA. Supplementation with oils doubled the content of total fatty acids in blood plasma, with little difference between different vegetable oil sources. Octadecenoic acid content was significantly higher in blood plasma of animals fed added oils from cottonseed and soybean than those fed with corn oil and control. The plasma trans-11C18:1 content was significantly higher in the oil-added animals than in control. Supplementation of vegetable oils tended to improve milk production of lactating cows, and the CLA content in milk fat was significantly increased. Soybean oil seemed to be the optimal source to increase CLA production.

Dose Response of Milk Fat to Intravenous Administration of the trans-10, cis-12 Isomer of Conjugated Linoleic Acid

Intravenous infusion of conjugated linoleic acid (CLA) was evaluated as a simpler method than abomasal infusion and the feeding of calcium salts to examine milk fat depression. The objectives were to determine the dose-dependent response of milk fat and plasma metabolites to intravenous administration of the trans-10, cis-12 isomer of CLA, an isomer identified to possess an inhibitory effect on milk fat synthesis. Four multiparous Holstein cows averaging 123±30 d in milk were randomly assigned to treatments in a 4×4 Latin square design. Catheters were inserted into the jugular vein for infusions and blood sampling. Treatments consisted of intravenous infusions of 0, 2, 4, and 6 g/d CLA (>95% trans-10, cis-12 CLA). Infusates contained 72 g/d of a parenteral solution, saline, and CLA to 90ml. Periods were of 5 d duration with a 7 d wash out. Milk was sampled at each milking and analyzed for fat, protein, and fatty acids. Blood samples were obtained on the last day of each period. Dry matter intake (22.4±2.4 kg/d), milk yield (28.5±3.3 kg/d), and protein percent (3.26±0.08%) of cows were not affected by treatment. However, milk fat percentage was reduced linearly with increasing doses of CLA. Milk fat percentage was 4.17, 3.53, 3.29, and 2.92% on d 5 for treatments 0, 2, 4, and 6 g/d CLA, respectively. Concentrations (4.2 mg/g of fat) of cis-9, trans-11 CLA in milk fat were not affected by treatment. However, an increase in the trans-10, cis-12 CLA content of milk fat was observed. Milk fat contained 0.00, 0.02, 0.06, and 0.10mg of trans-10, cis-12 CLA per g of fat (SEM = 0.065) for treatments 0, 2, 4, and 6 g/d CLA, respectively. Plasma NEFA concentration increased linearly with the dose of the trans-10, cis-12 CLA. Intravenous infusion of the trans-10, cis-12 isomer of CLA depressed milk fat in a linear manner over the range of infusion studied and, therefore, is an alternative to abomasal infusion.

Influence of pasture and concentrates in the diet of grazing dairy cows on the fatty acid composition of milk.

In five short-term experiments conducted in Victoria in 1997 and 1998, grazing dairy cows were given either pasture alone or pasture supplemented with high-energy concentrates, and the fatty acid profiles of milk fat were measured. We established the effects of these feeds on some aspects of milk fat of importance for human nutrition, but we specifically focused on the hypothesis that conjugated linoleic acid (CLA) concentrations in milk fat increase as pasture intake increases, and decrease as more concentrates are fed. In agreement with previous research, feeding fresh pasture alone resulted in high concentrations (1.0-1.8 g/100 g milk fat) of CLA. When the effect of level of pasture consumption on CLA content was examined, a significant positive relationship (r2 = 0.35; P < 0.05) was obtained. When cereal grain concentrates were used to supplement pasture intake, the CLA content of milk fat generally declined (P < 0.05), except where the amount of concentrates given led to a marked reduction in total milk fat concentration. The use of cereal grain concentrates also generally resulted in significant (P < 0.05) increases in medium-chain saturated fatty acids, but always reduced the contribution of butyric acid to milk fat, from 4.5 to 3.9 g/100 g milk fat, on average.

Trans-10, cis-12 Conjugated Linoleic Acid Decreases Lipogenic Rates and Expression of Genes Involved in Milk Lipid Synthesis in Dairy Cows

Feeding conjugated linoleic acid (CLA) reduces milk fat synthesis in lactating dairy cows, and the effect has been shown to be specific for the trans-10, cis-12 CLA isomer. Our objectives were to examine potential mechanisms by which trans-10, cis-12 CLA inhibits milk fat synthesis. Multiparous Holstein cows (n = 4) in late lactation were used in a balanced 2×2 crossover design. Treatments consisted of a 5 d abomasal infusion of either skim milk (control) or purified trans-10, cis-12 CLA (13.6 g/d) emulsified in skim milk. On d 5 of infusion, mammary gland biopsies were performed and a portion of the tissue analyzed for mRNA expression of acetyl CoA carboxylase, fatty acid synthetase, Δ9-desaturase, lipoprotein lipase, fatty acid binding protein, glycerol phosphate acyltransferase and acylglycerol phosphate acyltransferase. Lipogenic capacity was evaluated with another portion of the tissue. Infusion of trans-10, cis-12 CLA decreased milk fat content and yield 42 and 48%, respectively and increased the trans-10, cis-12 CLA content in milk fat from <0.1 to 4.9 mg/g. Reductions in milk fat content of C4 to C16 fatty acids contributed 63% to the total decrease in milk fat yield (molar basis). Analysis of the ratios of specific fatty acid pairs indicated trans-10, cis-12 CLA also shifted fatty acid composition in a manner consistent with a reduction in Δ9-desaturase. Mammary explant incubations with radiolabeled acetate established that lipogenic capacity was decreased 82% and acetate oxidation to CO2 was reduced 61% when cows received trans-10, cis-12 CLA. Infusing trans-10, cis-12 CLA also decreased the mRNA expression of all measured enzymes by 39 to 54%. Overall, data demonstrated the mechanism by which trans-10, cis-12 CLA inhibits milk fat synthesis includes decreasing expression of genes that encode for enzyme involved in circulating fatty acid uptake and transport, de novo fatty acid synthesis, desaturation of fatty acids and triglyceride synthesis.

Analysis of Variation in cis-9, trans-11 Conjugated Linoleic Acid (CLA) in Milk Fat of Dairy Cows

Conjugated linoleic acid (CLA) is a fatty acid with numerous putative health benefits and is a natural component of ruminant-derived food products. An intermediate in rumen biohydrogenation is cis-9, trans-11 CLA, the major CLA isomer in milk fat. However, the major source of cis-9, trans-11 CLA in milk is endogenous synthesis by Δ9-desaturase conversion of trans-11C18:1, another rumen biohydrogenation intermediate. The desaturase indices serve as a proxy for Δ9-desaturase activity and are calculated from the ratios of fatty acid pairs that represent product/substrate for this enzyme. This study analyzed individual animal variation in milk fat content of cis-9, trans-11 CLA and in desaturase indices in milk fat. Thirty lactating Holstein cows were allocated to one of three treatment groups: one received a standard total mixed ration, one received a diet that produced an elevated milk fat content of CLA, and a third treatment group was alternated between these diets at 3-wk intervals over the 12-wk study. There was a two- to threefold variation among individuals on the same diet for both milk fat content of CLA and desaturase indices in milk fat. This hierarchy was maintained to a large extent over the 12-wk study even in the variable treatment group that alternated between the two diets. Within the variable diet treatment, some animals consistently had a substantial response in milk fat content of CLA to dietary shifts, whereas other cows had little or no response. We conclude that while diet is a major determinant of the CLA content in milk fat, individual animal differences also have a substantial effect. The variation among individuals includes differences related to both rumen biohydrogenation and Δ9-desaturase activity in the mammary gland.

Independent effects of dietary linoleic and linolenic fatty acids on the conjugated linoleic acid content of cows’ milk

AbstractIt may be desirable to increase the level of conjugated linoleic acid (CLA) in milk as a health benefit in human nutrition. The purpose of this work was to separate the effects of linoleic and linolenic acids on CLA production in dairy cows and to determine to what extent endogenous synthesis contributes to cis-9, trans-11 CLA concentration in milk fat. Eight lactating cows and four non-lactating duodenal fistulated cows were used in a 4 ✕ 4 Latin-square design. All cows received a basal diet of grass silage that was supplemented with one of four concentrates, which were designed to differ in their linoleic and linolenic acid contents. The oil components of the concentrates were produced from mixtures of olive, linseed, rape, soya and sunflower oils to produce the four treatments: low linoleic/ low linolenic acid (LL), low linoleic/high linolenic acid (LH), high linoleic/low linolenic acid (HL) and high linoleic/ high linolenic acid (HH). Milk cis-9, trans-11 CLA contents were 0·8, 0·9, 0·9 and 1·1 g/100 g fatty acid methyl esters (P &lt; 0·05) and yields were 5, 7, 7 and 8 g/day (P &lt; 0·05) for the LL, LH, HL and HH treatments, respectively. The yields of trans-C18:1fatty acids in milk were 19, 22, 21 and 23 g/day (P &lt; 0·05), respectively. Taking the data for the cis-9, trans-11 CLA content and flow of duodenal fluid from the fistulated cows and representing this in terms of dietary intake by the lactating animals, the amounts of cis-9, trans-11 CLA produced in the rumen were calculated to be 0·8, 0·9, 1·2 and 1·1 g/day (P &lt; 0·05) and for trans-C18:1fatty acids 58, 58, 66 and 69 g/day (P &lt; 0·05). Increasing linoleic and/or linolenic acids in the diet can increase the cis-9, trans-11 CLA content of cows’ milk. Only diets high in linoleic acid increased cis-9, trans-11 CLA production in the rumen. On all four diets, more than 80% of cis-9, trans-11 CLA in milk was produced endogenously by Δ9-desaturase from trans-11 C18:1in the mammary gland. Cows on the same diet have different milk fat cis-9, trans-11 CLA concentrations that may be partially explained by differences in Δ9-desaturase activity between cows. Increasing the activity of Δ9-desaturase in the mammary gland may offer greater potential for enhancing the cis-9, trans-11 CLA content of milk fat than increasing cis-9, trans-11 CLA production in the rumen.

The Effect of Nonstructural Carbohydrate and Addition of Full Fat Extruded Soybeans on the Concentration of Conjugated Linoleic Acid in the Milk Fat of Dairy Cows

Conjugated linoleic acid (CLA), a naturally occurring anticarcinogen found in dairy products, is a byproduct of incomplete ruminal biohydrogenation of unsaturated fatty acids. Our objective was to determine the effect of nonstructural carbohydrate sources, addition of full fat extruded soybeans as a source of unsaturated fatty acids, and possible interactions on the milk fat content of CLA. Cows (n = 20) were assigned to a 4 × 4 Latin square involving two sources of nonstructural carbohydrate, high starch (corn) or high pectin (citrus pulp), with or without addition of extruded soybeans. Milk yield was not affected by nonstructural carbohydrate source, but milk production was increased by 7.8 to 10.5% with dietary additions of extruded soybeans. Milk fat content did not differ between treatments, but fatty acid composition was affected. Cows fed extruded soybean diets had reduced concentrations of C8 to C16 fatty acids and increased concentrations of octadecenoic acids. Diets with extruded soybeans also resulted in more than a doubling in milk fat concentration and yield of CLA. Nonstructural carbohydrate source had only minor effects on CLA, and there was no interaction with extruded soybeans. Milk fat content of trans-C18:1 and CLA were closely related (r2 = 0.77). However, variation among cows was about threefold for each of the diets and rank order of individual cows differed among diets. Overall, we demonstrated that diet modification can be used to alter CLA content of milk fat, but there was substantial individual cow variation for all diets.

Exogenous Conjugated Linoleic Acid Isomers Reduce Bovine Milk Fat Concentration and Yield by Inhibiting De Novo Fatty Acid Synthesis

Conjugated linoleic acid (CLA) is a potent anticarcinogen secreted in milk of ruminants, but it inhibits de novo fatty acid synthesis and desaturation in mammary cell cultures. The potential for increasing CLA content of milk fat and the effects of elevated CLA availability on milk fat secretion were investigated. Four Holstein cows were used in a single crossover design with repeated measures to determine milk fatty acid concentration in response to a 24-h infusion of 200 g linoleic acid (LA) or a mixture of 100 g LA plus 100 g CLA (LCLA). Milk and blood samples were obtained 12 h before infusion and at 12-h intervals from 0 to 72 h. Compared with LA infusion, total CLA concentration in blood plasma at 24 h in response to LCLA was elevated fivefold, whereas CLA content of plasma triglycerides was increased 10-fold. Milk fat yield from 24 to 72 h was ∼34% lower in response to LCLA compared with LA, due primarily to reduced yield of fatty acids with six to 16 carbons. Amount of CLA in milk increased from 0.5 g/100 g total fatty acids at 0 h to 3.3 g/100 g at 36 h in response to LCLA. Concentration of stearic acid in milk fat at 36 h in response to LCLA was nearly double the stearic acid concentration in response to LA. Oleic and arachidonic acid concentrations in milk declined as stearic acid increased in response to LCLA. Results indicated CLA content of milk fat reflects the amount available for absorption from the small intestine, and CLA appeared to be a potent inhibitor of de novo fatty acid synthesis and desaturation in the mammary gland.
