Influence of toys on behavioural patterns of pigs and their association with the concentration of serotonin in blood plasma

Improvement of welfare of piglets is one of the key factors which underlie the effectiveness of producing pork and the profitability of pig farming. This article offers a solution to the main industrial problem – creation of comfortable maintenance conditions for pigs using toys which satisfy their ethological needs. Piglets used in the experiment were kept in the swine complex of agricultural production of Agrofirm Mig-Service-Agro Cooperative in Novoodesky district of Mykolaiv Oblast (Ukraine). The objective of the study was assessment of the effect of toys on the behaviour of piglets, level of concentration of hormone serotonin in blood plasma and their energy of growth. The experiment was performed on 120 weaned piglets aged 28 days, which were divided according to the principle of analogues with 60 individuals in each group: I – the control group, the animals were kept with no toys; II – the experimental group, where the animals had toys. During the experiment chronometric monitoring of the piglets’ behavioural acts was carried out using video observation, we extracted blood samples for determination of the content of the hormone serotonin using the method of highly-efficient fluid chromatography and assessed the growth of the piglets. After the end of the experiment, we determined that the presence of toys reduced the intragroup aggression, which was accompanied by decrease in the duration of fights and almost three-fold increase in playing behaviour. Among piglets which had access to toys, the number of individuals observed to bite tails and ears decreased from 11 to 3. In the period of the study, the concentration of serotonin in the blood plasma of piglets who had free access to toys was 10 times higher compared with animals of the control group. Change in the behaviour patterns among the animals of the experimental group led to significant differences regarding the parameters of growth, where probable increase in the live weight at the age of 90 days was observed, which was associated with significant increase in their mean daily weight gain compared with the animals of the control group. This study confirms that use of toys for piglets during growth completion can prevent features of aggression and abnormal forms of stereopathy within groups, increase the live weight and mean daily weight gain and improve emotional condition, which affects the welfare of pigs.

Influence of temperature on prevalence of health and welfare conditions in pigs: time-series analysis of pig abattoir inspection data in England and Wales.

The prevalence of many diseases in pigs displays seasonal distributions. Despite growing concerns about the impacts of climate change, we do not yet have a good understanding of the role that weather factors play in explaining such seasonal patterns. In this study, national and county-level aggregated abattoir inspection data were assessed for England and Wales during 2010-2015. Seasonally-adjusted relationships were characterised between weekly ambient maximum temperature and the prevalence of both respiratory conditions and tail biting detected at slaughter. The prevalence of respiratory conditions showed cyclical annual patterns with peaks in the summer months and troughs in the winter months each year. However, there were no obvious associations with either high or low temperatures. The prevalence of tail biting generally increased as temperatures decreased, but associations were not supported by statistical evidence: across all counties there was a relative risk of 1.028 (95% CI 0.776-1.363) for every 1 °C fall in temperature. Whilst the seasonal patterns observed in this study are similar to those reported in previous studies, the lack of statistical evidence for an explicit association with ambient temperature may possibly be explained by the lack of information on date of disease onset. There is also the possibility that other time-varying factors not investigated here may be driving some of the seasonal patterns.

227 Leveraging on high-throughput phenotyping technologies to optimize livestock genetic improvement and husbandry

Abstract The advent of fully automated data recording technologies and high-throughput phenotyping (HTP) systems has opened up a myriad of opportunities to advance breeding programs and livestock husbandry. Such technologies allow scoring large number of animals for novel phenotypes and indicator traits to boost genetic improvement, as well as for real-time monitoring of animal behavior and development for optimized management decisions. HTP tools include, for example, image analysis and computer vision, sensor technology for motion, sound and chemical composition, and spectroscopy. Applications span from health surveillance, precision nutrition, and control of meat and milk composition and quality. However, the application of HTP requires sophisticated statistical and computational approaches for efficient data management and appropriate data mining, as it involves large datasets with many covariates and complex relationships. In this talk we will discuss some of the challenges and potentials of HTP in livestock. Some examples to be presented include the utilization of automated feeders to record feed intake and to monitor feeding behavior in broilers, milk-spectra information to predict dairy cattle feed intake, and image analysis and computer vision to monitor growth and body condition in pigs and cattle. HTP and big data will become an essential component of modern livestock operations in the context of precision animal agriculture, boosting animal welfare, environmental footprint, and overall sustainability of animal production.

Relationships between body temperatures and inflammation indicators under physiological and pathophysiological conditions in pigs exposed to systemic lipopolysaccharide and dietary deoxynivalenol.

We studied the constancy of the relationship between rectal and intraabdominal temperature as well as their linkage to inflammatory markers (leucocyte counts, kynurenine-to-tryptophan ratio (Kyn-Trp ratio), tumour necrosis factor alpha (TNF-α) in healthy and in pigs exposed to lipopolysaccharide (LPS) and/or deoxynivalenol (DON). Barrows (n=44) were fed 4weeks either a DON-contaminated(4.59mg DON/kg feed)or a control (CON) diet andequipped with anintraabdominaltemperature logger and a multicatheter system (V.portaehepatis, V.lienalis, Vv.jugulares) facilitating infusion of 0.9% NaCl (CON) or LPS(7.5μg/kgBW) and simultaneous blood sampling. Body temperatures were measured and blood samples taken every 15min for leucocyte counts, TNF-α and Kyn-Trp ratio. Combination of diet and infusion created six groups: CON_CONjug .-CONpor. , CON_CONjug. -LPSpor. , CON_LPSjug. -CONpor. , DON_CONjug. -CONpor. , DON_CONjug. -LPSpor. , DON_LPSjug. -CONpor. . The relationship between both temperatures was not uniform for all conditions. Linear regression revealed that an intraabdominal increase per 1°C increase in rectal temperature was ~25% higher in all LPS-infused pigs compared to NaCl-infusion, albeit diet and site of LPS infusion modified the magnitude of this difference. Inflammatory markers were only strongly present under LPS influence and showed a significant relationship with body temperatures. For example, leucocyte counts in clinically inconspicuous animals were only significantly correlated to core temperature in DON-fed pigs, but in all LPS-infused groups, irrespective of diet and temperature method. In conclusion, the gradient between body core and rectal temperature is constant in clinically inconspicuous pigs, but not under various pathophysiological conditions. In the latter, measurement of inflammatory markers seems to be a useful completion.

The effect of social isolation, gender and familiarity with the experimental procedure on tests of porcine nociceptive thresholds

ObjectiveTo investigate the effects of habituation and isolation on mechanical nociceptive thresholds in pigs at the pelvic limbs and at the tail. Study designProspective randomized multifactorial study. AnimalsThirty-two healthy castrated male (experiment 1), and 12 castrated male and 12 female (experiment 2) Danish Landrace × Yorkshire pigs, weighing 63.5 ± 0.8 kg and 55.4 ± 0.6 kg (the mean ± SD, experiment 1 and 2, respectively). MethodsMechanical nociceptive thresholds were quantified with a von Frey anesthesiometer applied to two distinct anatomical regions (tail and pelvic limbs). Pigs receiving the mechanical challenge in the pelvic limbs were tested inside a cage, whereas pigs exposed to stimuli at the tail region were tested in an open arena. For both experiments, the effect of familiarity to the procedure was evaluated by comparing thresholds of nociception in habituated versus naïve pigs. The presence of a companion animal was also evaluated in pigs receiving stimuli at the pelvic limbs. ResultsPigs tested inside the cage were affected by the habituation to the procedure as indicated by the increase in willingness and time spent by the animals in the test cage. This effect was reflected in the lower mechanical nociceptive thresholds (medians with 25–75 percentiles) recorded for familiar pigs compared with naïve animals [495 g (302–675) versus 745 g (479–1000), respectively; p = 0.026]. Mechanical nociceptive thresholds measured at the tail of the pigs in the open arena were not affected by the familiarity of the animals with the experimental procedure. Conclusions and clinical relevanceThe current results reiterate the value of habituation in research involving animal behaviour. Further characterization of the methodology is needed to allow its application in the evaluation of clinical conditions in pigs.

FIRST REPORTED CASE OF DOG ASSOCIATED PIG RABIES IN GHANA

Background: Pig rabies is uncommon and there is paucity of information on rabies in pigs in West African countries other than Nigeria. This communication presents a case of dog associated pig rabies in Adidome, Ghana. Materials and Methods: Case history, Dog assessment in adjoining communities, human exposure, clinical presentations and mortality in affected pigs were evaluated using standard techniques. Laboratory screening of brain samples collected was by Fluorescent antibody technique. Results: Stray dog bitten affected pigs showed anorexia, constipation, hyper excitation, twitching of head, roaring and foaming with resultant category 1 and 11 human exposure, 17% mortality and 75% case fatality rate. Laboratory examination of swine brain specimens tested positive for Rabies virus using Fluorescent Agglutination Test. Conclusion: This report described the first documented case of dog associated pig rabies in Ghana and emphasis was laid on continued advocacy and prompt reporting of suspected neurological conditions in pigs in Ghana

Effects of habituation to the testing facility on mechanical nociceptive thresholds of pigs

Abstract Aims This study aimed to gain methodological knowledge about quantification of mechanical nociceptive thresholds in pigs stimulated on the hind legs. Methods Thirty-two castrated male pigs were used, weighing 50–60kg, 16 weeks of age and housed in standard pens (4.40m×4.40m) with ad libitum access to water and feed. Mechanical nociceptive testing (4 stimulations/pig) was done by an electronic von Frey anesthesiometer (IITC Life Science Inc., CA) with a rigid, hollow plastic tip (cut-off: 1000 gf) and an area of 0.3 mm2. Stimulations targeted caudal aspects of the metatarsus while the pigs were restrained in a cage measuring 35cm×40cm (length×width). During the last 3d before nociceptive testing, half of the pigs were habituated to the experimental set-up twice daily. Results Across the three days, the habituation procedure led to a reduction in time required to approach and release the pigs from the home pen (P<0.05) and to increased acceptance of the confinement (P<0.01). The habituated animals had a lower mechanical nociceptive threshold (median: 495gf (302–675) vs 745gf (479–1000); P<0.05), and a decreased occurrence of censored observations (0 vs 25%; P<0.05) compared to animals tested without habituation. Conclusions The present experiment focussed on methodological aspects of hand-held tests of mechanical nociceptive thresholds in pigs. The results show that the pigs responded to a 3-day habituation regime by increased ease of handling and tolerance of being confined in a test cage. In addition, habituation to the test environment and handling led to reduced mechanical nociceptive thresholds as well as a lowered proportion of censored observations. The current results reiterate the value of habituation in research involving animal behaviour. Further characterization of the methodology is needed to allow its application in the evaluation of clinical conditions in pigs.

Feed intake, digestibility, growth performance, and blood profile of pigs fed mixtures of dried and ground fig (Ficus sur) fruits and graded levels of maize.

This study was conducted to examine the effect of feeding a mixture of different proportions of Ficus sur fruits (FSF) and ground maize grain (MG) on intake, digestibility, growth, and blood profile on Yorkshire pigs. Dietary treatments comprised 100% FSF and 0% MG (100FSF), 67% FSF and 33% MG (67FSF), 33% FSF and 67% MG (33FSF), and 0% FSF and 100% MG (0FSF). Noug cake and soybean meal were included to meet nutrient requirement of the animals at isonitrogenous (18% crude protein (CP)) level with graded levels of energy. Twenty pigs with an average initial weight of 27.75 ± 1.4kg were grouped according to their body weight, and animals from each group were randomly assigned to four dietary treatments. There was no difference (P > 0.05) in dry matter, crude protein, and metabolizable energy intakes among the treatments. Organic matter (P < 0.05), ether extract (P < 0.01), and nitrogen-free extract (NFE) intakes were higher (P < 0.0001) for pigs fed with 0FSF than 100FSF. The digestibility of dry matter, crude protein, ether extract, and NFE did not differ (P > 0.05) among treatments. However, organic matter digestibility (P < 0.05) was highest for pigs in 0FSF than 100FSF. Conversely, crude fiber digestibility was in the reverse trend. No significant differences were observed in body weight gain among treatments (P > 0.05). The blood counts and sera metabolites of pigs across the treatments fall within the optimum range. Packed cell volume (PCV) and hemoglobin (Hgb) were higher, but creatinine and cholesterol concentrations were lower in 100FSF than 0FSF indicating better health conditions in pigs fed with FSF. It is, therefore, concluded that feeding FSF has comparable effects with MG on the metabolic performance, growth, and blood profile of pigs.

Gestational heat stress alters postnatal offspring body composition indices and metabolic parameters in pigs.

The study objectives were to test the hypothesis that heat stress (HS) during gestational development alters postnatal growth, body composition, and biological response to HS conditions in pigs. To investigate this, 14 first parity crossbred gilts were exposed to one of four environmental treatments (TNTN, TNHS, HSTN, or HSHS) during gestation. TNTN and HSHS dams were exposed to thermal neutral (TN, cyclical 18–22°C) or HS conditions (cyclical 28–34°C) during the entire gestation, respectively. Dams assigned to HSTN and TNHS treatments were heat-stressed for the first or second half of gestation, respectively. Postnatal offspring were exposed to one of two thermal environments for an acute (24 h) or chronic (five weeks) duration in either constant TN (21°C) or HS (35°C) environment. Exposure to chronic HS during their growth phase resulted in decreased longissimus dorsi cross-sectional area (LDA) in offspring from HSHS and HSTN treated dams whereas LDA was larger in offspring from dams in TNTN and TNHS conditions. Irrespective of HS during prepubertal postnatal growth, pigs from dams that experienced HS during the first half of gestation (HSHS and HSTN) had increased (13.9%) subcutaneous fat thickness compared to pigs from dams exposed to TN conditions during the first half of gestation. This metabolic repartitioning towards increased fat deposition in pigs from dams heat-stressed during the first half of gestation was accompanied by elevated blood insulin concentrations (33%; P = 0.01). Together, these results demonstrate HS during the first half of gestation altered metabolic and body composition parameters during future development and in biological responses to a subsequent HS challenge.

The proliferation and differentiation of primary pig preadipocytes is suppressed when cultures are incubated at 37°Celsius compared to euthermic conditions in pigs

Given similarities in metabolic parameters and cardiovascular physiology, the pig is well positioned as a biomedical model for metabolic disease and obesity in humans. Better understanding molecular mechanisms governing porcine adipocyte hyperplasia may provide insight into the regulation of adipose tissue development that is useful both when considering the pig as a commodity and when extrapolating porcine data to human disease. Primary cultures of pig stromal-vascular cells have served as a useful tool for investigating factors that regulate preadipocyte proliferation and differentiation. However, such cultures have generally been maintained at 37°C in vitro despite euthermia being 39°C in pigs. To address potential concerns about the physiological relevance of culturing primary pig preadipocytes under what would be hypothermic conditions in vivo, the objective of this study was to investigate the effect of culture temperature on the proliferation and differentiation of pig preadipocytes in primary culture. Culturing primary preadipocytes at 37 rather than 39°C decreases their proliferation rates based upon cleavage of the tetrazolium salt, MTT (P < 0.001), reduction of resazurin (P < 0.001), and daily cell counts (P < 0.001). Likewise, culturing primary porcine preadipocytes at 37°C suppressed their adipogenic potential based upon monitoring adipogenesis morphologically, biochemically, and via the expression of mRNA encoding adipogenic marker genes. Collectively, these data indicate the proliferation and differentiation of primary pig preadipocytes is suppressed when cultures are incubated at 37°C compared to normal body temperature of pigs. This may confound investigation of factors that impact adipocyte hyperplasia in the pig.

Influence of intense neuromuscular blockade on surgical conditions during laparotomy: a pig model.

Intense neuromuscular block may improve surgical conditions in ileus laparotomies; however, it is difficult to evaluate. The aim of this study was to investigate if neuromuscular block improved surgical conditions in pigs with artificial ileus laparotomy. Six pigs were endotracheally intubated, mechanically ventilated, anesthetized with propofol and fentanyl, and randomized into two groups in a cross-over assessor-blinded design. Neuromuscular block was established with rocuronium. Artificial laparotomy for ileus was performed. We investigated the influence of intense neuromuscular block on surgical conditions with a subjective rating scale, force needed to close the fascia, incidences of abdominal contractions while suctioning the lungs, width of the wound diastase and operating time as outcome parameters. In all six pigs no abdominal contractions occurred while suctioning the lungs at intense neuromuscular block. Without neuromuscular block we detected abdominal contractions seen as hiccups and bucking. In all six pigs during intense neuromuscular block we found no visible electromyographic (EMG) activity in the abdominal muscles while suctioning the lungs. Without neuromuscular block suctioning the lungs elicited brief periods of abdominal EMG activity. No difference was found in the force needed to close the fascia when comparing no neuromuscular block with intense neuromuscular block. Furthermore, no significant differences were found in the width of the diastase, operating time and subjective ratings using a four-point rating scale when comparing no neuromuscular block with intense neuromuscular block. However, these outcomes were related to the order of the suturing round. Intense neuromuscular block prevented abdominal muscle contractions but did not influence the force needed to close the fascia.

Simultaneous dual-isotope SPECT/CT with 99mTc- and 111In-labelled albumin microspheres in treatment planning for SIRT

To investigate simultaneous dual-isotope SPECT/CT with two differently radioisotope-labelled albumin-microsphere fractions for treatment planning of hepatic radioembolisation. In addition to (99m)Technetium-labelled albumin microspheres (commercially available), we performed labelling with (111)Indium. Binding stability of (111)Indium-labelled microspheres was tested in vitro and in vivo in mice. Simultaneous dual-isotope SPECT/CT imaging was validated in an anthropomorphic torso phantom; subsequently, dual-isotope SPECT/CT was performed under in-vivo conditions in pigs (n = 3) that underwent transarterial injection of (99m)Technetium- and (111)Indium-labelled microspheres in the liver (right and left hepatic artery, respectively), in both kidneys and in the gluteal musculature. In total, n = 18 transarterial injections were performed. In-vitro testing and in-vivo studies in mice documented high binding stability for both (99m)Technetium-labelled and (111)Indium-labelled microsphere fractions. In phantom studies, simultaneous dual-isotope SPECT/CT enabled reliable separation of both isotopes. In pigs, the identified deposition of both isotopes could be accurately matched with intended injection targets (100 %, 18/18 intended injection sites). Furthermore, an incidental deposition of (99m)Technetium-labelled microspheres in the stomach could be correlated to the test injection into a right hepatic artery. Simultaneous dual-isotope SPECT/CT after transarterial injection with (99m)Technetium- and (111)Indium-labelled microspheres is feasible. Thus, it may offer additional, valuable information compared to single (99m)Technetium-labelled albumin examinations. • Simultaneous dual-isotope SPECT/CT with (111) In- and (99m) Tc-labelled albumin microspheres is feasible. • Differentiation of two microsphere fractions after transarterial injection is possible. • The origin of an extra-hepatic microsphere deposition can be correlated to the corresponding artery. • This technique could reduce the setup time for selective internal radiation treatment.

Evaluation of suitable reference genes for gene expression studies in porcine PBMCs in response to LPS and LTA

BackgroundAs an in vitro model porcine peripheral blood mononuclear cells (PBMCs) is frequently used as for immunogenetic research with the stimulation of bacterial antigens. To investigate the immunocompetence of PBMCs for recognition of Gram-positive and Gram-negative bacteria and in order to dissect the pathogenesis of diseases, gene expression assay is most commonly used. The gene expressions are required to normalize for reference genes which have tremendous effect on the results of expression study. The reference genes should be stably expressed between different cells under a variety of experimental conditions, but recent influx of data showed that expression stability of reference genes are varied under different experimental conditions. But data regarding the expression stability of reference genes in porcine PBMCs are limited. Therefore, this study was aimed to know whether the expression stability of commonly used reference genes in PBMCs is affected by various bacterial antigens under different experimental conditions in pigs.ResultsThe mRNA expression stability of nine commonly used reference genes (B2M, BLM, GAPDH, HPRT1, PPIA, RPL4, SDHA, TBP and YWHAZ) was determined by RT-qPCR in PBMCs that were stimulated by LPS and LTA in vitro as well as cells un-stimulated control and non-cultured were also consider for this experiment. mRNA expression levels of all genes were found to be affected by the type of stimulation and duration of the stimulation (P < 0.05). geNorm software revealed that in case of irrespective of stimulation (without considering the type of stimulation), RPL4, PPIA and B2M were the most stable reference genes in PBMCs; in case of the control group, PPIA, BLM and GAPDH were the most stable reference genes. PPIA, B2M and RPL4 were the most stable reference genes in LPS stimulated PBMCs; and YWHAZ, RPL4 and PPIA were the most stably expressed reference genes in the case of LTA stimulated PBMCs. When LPS was used combined with LTA for the stimulation, YWHAZ, B2M and SDHA remained the most stable genes. PPIA, BLM and GAPDH were found to be most stably expressed reference genes when PBMCs were not cultured. NormFinder revealed different sets of stably expressed reference genes in PBMCs under different experimental conditions. Moreover, geNorm software suggested that the geometric mean of the three most stable genes would be the suitable combination for accurate normalization of gene expression study.ConclusionThere was discrepancy in the ranking order of reference genes obtained by different analysing algorithms (geNorm and NormFinder). In conclusion, the geometric mean of the RPL4, B2M and PPIA seemed to be the most appropriate combination of reference genes for accurate normalization of gene expression data in porcine PBMCs without knowing the type of bacterial pathogenic status of the animals and in the case of mixed infection with Gram-negative and Gram-positive bacteria. In case of PBMCs without any stimulation, PPIA, BLM and GAPDH could be suggested as suitable reference genes.

Health status and bio-security plans on pig farms

Preservation of necessary level of swine herd health status is the most important aspect of bio-security, farm production and successful welfare protection. It involves a list of bio-security measures which must be essential part of production technology, including good rearing conditions and other prophylactic measures appliance. According to previously performed investigations, a list of elements required to establish standards for bio-security for particular pig farm was created. The list includes elements related to assure spatial., microclimate and hygienic conditions for pigs rearing, care, nutrition and watering, surveillance, regrouping and transport of swine, disease outbreak control, barns equipment and production process organization which are performed by veterinarians and zootechnicians. Bio-security standards must concern necessary level of swine herd health status and organization, size and type of operation and anticipate all compulsory measures for health control and disease spreading prevention, particularly those whose outbreak must be reported. In bio-security plan structure present epidemiological situation, potential herd health and production threats, as well as possible solutions must be concerned.

Fear assessment in pigs exposed to a novel object test

The experiment aimed to study approach and locomotive behaviour as indicators of fear in a novel object test carried out in pigs. Thirty post-weaning (30 kg) and 30 finishing (90 kg) pigs were exposed to visual, auditory and olfactory novel stimuli during 2 different experiments. The facilities consisted of a test pen in which a trough was located. The trough contained chopped apples. Once the animals were trained to enter the test pen individually they were subjected to 3 different fear stimuli. These stimuli were applied in the test pen and next to the trough. The variables studied were feeding behaviour, approach behaviour (the distance and position of the animal with respect to the trough) and locomotive behaviour (general activity, reluctance to move, turning back and retreat attempts). Two groups were studied: saline and midazolam treated group. Twenty minutes before the start of the sessions, 15 post-weaning and finishing pigs received an intramuscular injection of 0.20 and 0.15 mg/kg, respectively, midazolam (Dormicum ®). The saline pigs (15 animals per group) were injected with saline. The administration of midazolam increased the feeding behaviour and approaching behaviour, and reduced the locomotive behaviour. In front of the visual and olfactory stimuli post-weaning pigs showed a higher general activity than finishing pigs, but the contrary was found when the auditory stimulus was applied. The olfactory stimulus was more related to the turning back behaviour, whereas the visual stimulus was more related to retreat attempts. Although it could be concluded that reluctant to move was the most common response to the different fear stimuli applied in our study regardless of the age of animals, the combination of reluctant to move and turning back would be a good criterion to assess fear in domestic pigs. The use of midazolam as anxiolytic for studies of fear in commercial conditions in pigs is recommended.

Influence of housing systems on stillbirth and mortality rate in preweaning pigs farrowed by different gilt breeds

The aim of this paper was to investigate the influence of housing systems on stillbirth and mortality rate in preweaning pigs farrowed by different gilt breeds. The investigation included first farrowings of 225 Large White (LW) gilts, 297 Swedish Landrace (SL) gilts and 260 crossbreed gilts (LWxSL) in the period of five years. Despite of positive influence of straw through decreased number of stillborn pigs, especially in LW gilts, in further process of production until weaning, less appropriate microclimatic conditions developed in farrowing houses with straw, especially if accompanied with poor ventilation, can result with higher number of perished pigs and lower number of weaned pigs. Most sensible to unsuitable microclimatic conditions in straw bedded farrowing houses, according to the number of perished pigs before weaning, were the pigs from SL gilts, while the pigs from crossbreed gilts LWxSL were the most resistant. These results suggest that adding straw requires special attention on influence of such production on microclimate conditions in pigs’ biozone in order to maintain its initial benefits in the time of farrowing through the whole preweaning period, especially in pure breeds of gilts.

Difference in Sensitivity to Culture Condition Between In Vitro Fertilized and Somatic Cell Nuclear Transfer Embryos in Pigs

We evaluated the developmental competence of somatic cell nuclear transfer (SCNT) embryos using in vitro embryo culture systems. Embryos were cultured in NCSU-23, NCSU-23 supplemented with essential and non-essential amino acids (NCSU-23aa) or modified PZM-5 supplemented with BSA instead of PVA (mPZM-5). The rates of blastocyst formation were significantly higher in the mPZM-5 group than in the other groups, regardless of the method of embryo production (38.0 vs. 25.3 or 29.1% for IVF, 18.2 vs. 8.7 or 9.4% for SCNT, respectively). The mean cell numbers of IVF and SCNT blastocysts were also significantly higher in mPZM-5 than in the other groups (62.0 vs. 42.3 or 43.0 for IVF, 46.5 vs. 29.4 or 31.3 for SCNT, respectively). Next, the embryos were cultured in mPZM-5 from days 0 to 4 and then in mPZM-5 (P/P), NCSU-23 (P/N) or NCSU-23aa (P/Naa) until day 6. The rates of blastocyst formation were similar among the 3 two-step culture systems in both embryo groups (36.2, 34.2, and 33.6% for IVF, 20.8, 14.1, and 17.2% for SCNT, respectively). The mean cell number in the IVF and SCNT blastocysts was significantly lower in P/N than in P/P and P/Naa (46.5 vs. 63.5 and 68.7 for IVF, 29.3 vs. 45.5 and 39.7 for SCNT, respectively). Next, we examined the effect of media on apoptosis in IVF and SCNT blastocysts. The apoptosis indices in the blastocysts derived from either NCSU-23 or mPZM-5 were analyzed by TUNEL assay. The apoptosis index of the SCNT blastocysts was significantly lower in mPZM-5 than in NCSU-23 (8.8 vs. 13.6%), whereas no such difference was observed between groups in the IVF embryos (5.1 vs. 4.4%). These data suggested that SCNT embryos were more easily affected by culture environment compared with IVF embryos, offering the possibility to further enhance the developmental competence of SCNT embryos by developing more appropriate culture conditions in pigs.

Endotoxemic pulmonary hypertension is largely mediated by endothelin-induced venous constriction

To analyze the effect of endothelin-1 on pulmonary arterial and venous contractile force in vitro and on up- and downstream pulmonary vascular resistance in vivo under sham and endotoxemic conditions in pigs. In vitro: paired preparations of pulmonary arteries and veins were mounted in a myograph (n=13) for measurements of contractile responses to increasing concentrations of phenylephrine, endothelin-1, and sarafotoxin (endothelin receptor type B agonist). In vivo: 20 pigs were anesthetized, mechanically ventilated, and subjected to phenylephrine (reference substance), endothelin-1, sarafotoxin, endotoxin, and tezosentan (dual endothelin receptor antagonist). Hemodynamic and gas-exchange variables were monitored. Pulmonary capillary pressure, used for calculation of upstream and downstream vascular resistance, was assessed by the pulmonary vascular occlusion technique. Pulmonary veins were more sensitive than arteries to endothelin-1 both in vitro and in vivo. This difference was more pronounced with sarafotoxin, where almost no arterial effects were noted. In vivo and in vitro exposure to phenylephrine resulted in selective arterial constriction. Endotoxin infusion resulted in pulmonary hypertension with a clear downstream dominance. The latter changes including the increase in capillary pressure were totally abolished by intervention with the dual endothelin receptor antagonist tezosentan. The endothelin system is extensively involved in endotoxemic pulmonary venous hypertension, an effect possibly mediated by the endothelin B receptor.

Risk assessment of DFD meat due to pre-slaughter conditions in pigs

A polychotomous logistic regression model was used to identify and assess the risk factors for pork becoming dark, firm and dry meat (DFD). A total of 116 deliveries, comprising 3075 commercial pigs delivered from different farms to five commercial Spanish pig abattoirs were surveyed. The DFD condition was described as an ordinal response variable (normal, moderate and serious) based on measurements of pH 24 in the Semimembranosus muscle. The abattoir, the floor of the lorry, the season, the gender, and the stocking density during transportation influenced the risk of DFD, as well as on-farm fasting time, lairage time and estimated carcass lean content. No effect of the RYR1 gene in the risk of DFD was found. Abattoirs should be especially careful with females slaughtered in winter, where the risk of serious DFD is 4.6% higher than with males slaughtered in summer. The risk of DFD increased with high stocking density and lairage time, and with on-farm fasting times longer than 22 h. Our results revealed that lowering the stocking density from 0.37 to 0.50 m 2 per 100 kg pig during transport would increase the risk of DFD pork by 11%.
