Efficacy of TurmiZn, a Metallic Complex of Curcuminoids-Tetrahydrocurcumin and Zinc on Bioavailability, Antioxidant, and Cytokine Modulation Capability

Complexes of curcumin with metals have shown much-improved stability, solubility, antioxidant capability, and efficacy when compared to curcumin. The present research investigates the relative bioavailability, antioxidant, and ability to inhibit inflammatory cytokine production of a curcuminoid metal chelation complex of tetrahydrocurcumin-zinc-curcuminoid termed TurmiZn. In vitro uptake assay using pig intestinal epithelial cells showed that TurmiZn has an ~3-fold increase (p ≤ 0.01) in uptake compared to curcumin and a ~2-fold increase (p ≤ 0.01) over tetrahydrocurcumin (THC). In a chicken model, an oral 1-g dose of TurmiZn showed a ~2.5-fold increase of a specific metabolite peak compared to curcumin (p = 0.004) and a ~3-fold increase compared to THC (p = 0.001). Oral doses (5 g/Kg) of TurmiZn in rats also showed the presence of curcumin and THC metabolites in plasma, indicating bioavailability across cell membranes in animals. Determination of the antioxidant activity by a 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) radical scavenging assay indicated that TurmiZn was about 13x better (p ≤ 0.0001) than curcumin and about 4X better (p ≤ 0.0001) than THC, in reducing free radicals. In vitro experiments further showed significant (p ≤ 0.01) reductions of lipopolysaccharide (LPS)-induced proinflammatory cytokines such as interleukin (IL) IL-6, IL-8, IL-15, IL-18, and tumor necrosis factor (TNF)-alpha, while showing a significant (p ≤ 0.01) increase of granulocyte-macrophage colony-stimulating factor (GM-CSF) in dog kidney cells. In vivo cytokine modulations were also observed when TurmiZn was fed for 6 weeks to newborn chickens. TurmiZn reduced IL-1 and IL-6, but significantly reduced (p ≤ 0.01) IL-10 levels while there was a concurrent significant (p = 0.02) increase in interferon gamma compared to controls. Overall, these results indicate that TurmiZn has better bioavailability and antioxidant capability than curcumin or THC and has the ability to significantly modulate cytokine levels. Thus, TurmiZn could be an excellent candidate for a novel ingredient that can be incorporated into food and supplements to help overall health during the aging process.

The impact of COVID-19 pandemic course in the number and severity of hospitalizations for other natural causes in a large urban center in Brazil.

The COVID-19 pandemic may indirectly impact hospitalizations for other natural causes. Belo Horizonte is a city with 2.5 million inhabitants in Brazil, one of the most hardly-hit countries by the pandemic, where local authorities monitored hospitalizations daily to guide regulatory measures. In an ecological, time-series study, we investigated how the pandemic impacted the number and severity of public hospitalizations by other natural causes in the city, during 2020. We assessed the number and proportion of intensive care unit (ICU) admissions and in-hospital deaths for all-natural causes, COVID-19, non-COVID-19 natural causes, and four disease groups: infectious, respiratory, cardiovascular, and neoplasms. Observed data from epidemiological week (EW) 9 (first diagnosis of COVID-19) to EW 48, 2020, was compared to the mean for the same EW of 2015-2019 and differences were tested by Wilcoxon rank-sum test. The five-week moving averages of the studied variables in 2020 were compared to that of 2015-2019 to describe the influence of regulatory measures on the indicators. During the studied period, there was 54,722 hospitalizations by non-COVID-19 natural causes, representing a 28% decline compared to the previous five years (p<0.001). There was a concurrent significant increase in the proportion of ICU admissions and deaths. The greater reductions were simultaneous to the first social distancing decree or occurred in the peak of COVID-19 hospitalizations, suggesting different drivers. Hospitalizations by specific causes decreased significantly, with greater increase in ICU admissions and deaths for infectious, cardiovascular, and respiratory diseases than for neoplasms. While the first reduction may have resulted from avoidance of contact with healthcare facilities, the second reduction may represent competing causes for hospital beds with COVID-19 after reopening of activities. Health policies must include protocols to address hospitalizations by other causes during this or future pandemics, and a plan to face the rebound effect for elective deferred procedures.

The Effect of COVID-19 on Adult Cardiac Surgery in the United States in 717,103 Patients

Background: COVID-19 has changed the world as we know it, and the United States continues to accumulate the largest number of COVID-related deaths worldwide at the time of publication. There exists a paucity of data regarding the effect of COVID-19 on adult cardiac surgery trends and outcomes on a national level.Methods: The STS Adult Cardiac Surgery Database was queried and analyzed from January 1, 2018 to June 30, 2020. The Johns Hopkins COVID-19 database was queried from February 1, 2020 to January 1, 2021. Surgical and COVID-19 volumes, trends, and outcomes were analyzed on a national and regional level. Observed-to-expected ratios were used to analyze mortality for all risk-adjustable cardiac procedures.Findings: A total of 717,103 adult cardiac surgery patients and more than 20 million COVID-19 patients were included for analysis. The South Atlantic (21.1%) and Great Lakes (17.4%) regions contributed the most patients during the study period. Nationally, there was a 52.7% reduction in overall adult cardiac surgery volume, and a 65.5% reduction in elective cases. The Mid-Atlantic region was most affected by the first COVID-19 surge, with a 69.7% reduction in overall case volume and 80.0% reduction in elective cases. In the Mid-Atlantic and New England regions, the observed-to-expected mortality for isolated coronary bypass increased as much as 1.48 times (148% increase) pre-COVID rates. After the first COVID-19 surge, nationwide cardiac surgical case volumes increased but did not return to baseline, indicating a COVID-associated deficit of cardiac surgery patients.Interpretation: This study is the largest analysis of COVID-19 related impact on adult cardiac surgery volume, trends, and outcomes. During the COVID-19 pandemic, cardiac surgery volume suffered dramatically, particularly in the Mid-Atlantic and New England regions during the first surge of COVID-19 cases, with a concurrent significant increase in observed-to-expected 30-day mortality. The acute effect of resistance exercise on limb blood flow.

What is the central question of this study? How does resistance exercise affect peripheral haemodynamics in the active and inactive limb? What is the main finding and its importance? Preliminary data indicate that resistance exercise increases flow and shear rate in the active limb transiently. The same exercise has minimal, short-lasting influence on peripheral haemodynamics in the inactive limb, but further research is required to elaborate on resistance exercise-mediated changes in vascular function in active and inactive limbs. Current evidence indicates that to achieve maximum health benefits, regular resistance exercise should be a key component of structured physical activity. Several studies have revealed that regular resistance exercise may be associated with impaired vascular function, although this finding is inconsistent. Proposed explanations for impairment include substantial increases in blood pressure and increased retrograde blood flow in active limbs promoted by resistance exercise. However, few studies have examined the acute haemodynamics of resistance exercise in active - and even fewer in inactive - limbs. The purpose of this study was to characterise the haemodynamic responses in peripheral arteries in active and inactive limbs in response to resistance exercise using upper and lower limbs. Ten participants (five male, five female) familiar with resistance training performed three sets of 10 isotonic repetitions of right-sided bicep curls or knee extensions on separate days. Blood flow, shear rate and muscle oxygenation in the active and inactive limb, and blood pressure were measured before and for 3min after each set. Blood flow increased in response to resistance exercise in the active limb (∼8-fold and ∼6-fold for the upper and lower limb respectively), with concurrent significant increases in mean and antegrade shear rate. In the inactive limb, blood flow more than doubled for both upper and lower limb exercise, transiently, with no significant change in retrograde shear rate. These acute blood flow profiles following resistance exercise are not indicative of long-term vessel impairment based on current understanding of blood flow and shear stress patterns.

Blood Orange Juice Consumption Increases Flow-Mediated Dilation in Adults with Overweight and Obesity: A Randomized Controlled Trial

Epidemiological studies have indicated an inverse association between citrus fruit consumption and cardiovascular disease (CVD) risk. There is, however, a paucity of data concerning effects of blood orange juice (BOJ) intake on endothelial function and cardiovascular risk biomarkers. We examined short-term effects of BOJ on endothelial function, blood pressure, lipid profile, and inflammatory markers in healthy participants of European origin who were overweight or obese. In a randomized, controlled, single-blind, crossover trial, 15 men and women (age: 28.7±6.5 y; BMI: 28.3±3.1 kg/m2) consumed BOJ or a sugar-matched control drink (CD) (200 mL twice daily) for 2 wk with a washout period of 1 wk. Endothelial function, measured as flow-mediated dilation (FMD) (primary outcome), and the secondary outcomes blood pressure, anthropometric measures, lipid profile, inflammatory markers, markers of vasodilation and vasoconstriction, and urinary flavanone metabolites were evaluated prior to and at the end of each treatment period following an overnight fast. Changes between treatments over time were assessed using repeated-measures ANOVA. The results demonstrate a significant increase in FMD following BOJ consumption (pre: 8.15%±2.92%; post: 10.2%±3.31%; P=0.002) compared with CD (pre: 8.11%±2.52%; post: 7.77%±2.43%; time × treatment interaction: P=0.001). Concurrent significant increases in urinary hesperetin-3'-glucuronide and hesperetin-7-glucuronide were observed following BOJ supplementation only (time × treatment interaction: P≤0.01). Baseline blood pressure, lipid profile, high-sensitivity C-reactive protein, and endothelin-1 were generally within healthy ranges and unaffected by the intervention. A 2-wk consumption of BOJ exerted favorable effects on endothelial function in healthy women and men who were overweight or obese, which is likely mediated by the combined actions of anthocyanin and flavanone metabolites on mechanisms that contribute to enhancing NO bioavailability. 2-Methoxyestradiol inhibits high fat diet-induced obesity in rats through modulation of adipose tissue macrophage infiltration and immunophenotype

Recently, experimental studies demonstrated that 2-methoxyestradiol (2ME2) ameliorates high fat diet (HFD)-induced obesity and restores insulin sensitivity. However, the mechanisms underlying these effects are unveiled yet. The current study was undertaken to test the hypothesis that 2ME2 exerts its effects by modulating adipose tissue macrophages (ATMs) accumulation, polarization and immunophenotypes. The experiment was carried out in males Wistar rats (n = 28) for 13 consecutive weeks. In HFD-fed group; body weight, glucose intolerance, serum insulin, HOMA-IR, lipid profile and adipose tissue (AT) weight were significantly higher compared to normal standard diet (NSD)- fed rats. However, treatment of HFD-fed rats with 2ME2 (200 μg/kg/day; i.p. from the beginning of the 9th week) resulted in significant enhancements in all these parameters as compared to HFD-fed rats. Treatment with 2ME2 was associated with a significant reduction in macrophage infiltration in the AT, shifting macrophage polarization towards M2 phenotype as indicated by significant decrease in the expression of pro-inflammatory M1 macrophages markers (IL-6, IL-1β, CD11c and iNOS) and concurrent significant increase in the M2 anti-inflammatory macrophage markers (Arginase 1 and IL-10). 2ME2 ameliorates HFD-induced obesity and glucose intolerance through inhibition of ATM infiltration in AT and shifting macrophage polarization from pro-inflammatory M1 to M2 anti-inflammatory phenotypes.

The MODULATORY EFFECTS OF BONE MARROW-DERIVED MESENCHMAL STEM CELLS ON CYCLOPHOSPHAMIDE INDUCED HEPATOTOXICITY IN CARCINOMA MICE

Objective: Cyclophosphamide (CPA) is a chemotherapeutic agent, induces hepatotoxicity as one of its side effects. Therefore, the present work was designed to investigate the protective role of bone marrow-derived mesenchymal stem cells (BM-MSCs) on CPA–induced hepatotoxicity in Ehrlich Ascites Carcinoma bearing-mice (EAC) and to test whether BM-MSCs influences the antitumor properties of the CPA.&#x0D; Methods: The hepatoprotective effects of BM-MSCs (single dose of 100 µl of a cell suspension containing allogenic BM-MSCs, i. v.) was evaluated in a model of hepatotoxicity by CPA (10 mg/kg/d i. p.) in EAC-female mice for one month. The anti-tumor activities of CPA and BM-MSCs were assessed by measuring mean tumor weight, mean survival time and the increase in life span. Moreover, ALT, AST, GGT, MDA, GSH, SOD, IL-6, IL10, caspase-3 and Bcl2 were measured.&#x0D; Results: The i. p. administration of CPA and BM-MSCs resulted in significant reductions in tumor size and mean tumor weight as well as caused concurrent significant increases in the life span as compared to the EAC mice. Furthermore, BM-MSCs ameliorated the liver enzyme markers namely ALT, AST, GGT, and hepatic oxidative stress through inhibition of MDA level that correlated with significant improvement in antioxidant status via increasing GSH and SOD levels as compared to both EAC and EAC+CPA groups. Moreover, BM-MSCs treatment significantly reduced the inflammatory marker level IL-6 as well as increment the level of IL-10 with subsequent decreases apoptosis via a depletion in the caspase-3 associated with an enhancement in the level of Bcl2 as compared to EAC group and EAC+CPA group. Minor histological lesions were observed in the liver tissue sections of mice treated with CPA and BM-MSCs as compared to the high histological lesions observed in the liver of the EAC group and CPA treated group.&#x0D; Conclusion: These results concluded that the combination treatment of BM-MSCs with CPA exhibited promising potential antitumor efficacy with greater safety than CPA treatment alone in mice via its antioxidant, anti-inflammatory and antiapoptotic effects.

Azathioprine Has a Deleterious Effect on the Bone Health of Mice with DSS-Induced Inflammatory Bowel Disease.

Patients with inflammatory bowel disease (IBD) often present poor bone health and are 40% more at risk of bone fracture. Studies have implicated autophagy in IBD pathology and drugs used to treat IBD stimulate autophagy in varying degrees, however, their effect on the skeleton is currently unknown. Here, we have utilised the dextran sulphate sodium (DSS) model of colitis in mice to examine the effects of the thiopurine drug azathioprine on the skeleton. Ten-week-old male mice (n = 6/group) received 3.0% DSS in their drinking water for four days, followed by a 14-day recovery period. Mice were treated with 10 mg/kg/day azathioprine or vehicle control. Histopathological analysis of the colon from DSS mice revealed significant increases in scores for inflammation severity, extent, and crypt damage (p < 0.05). Azathioprine provided partial protection to the colon, as reflected by a lack of significant difference in crypt damage and tissue regeneration with DSS treatment. MicroCT of vehicle-treated DSS mice revealed azathioprine treatment had a significant detrimental effect on the trabecular bone microarchitecture, independent of DSS treatment. Specifically, significant decreases were observed in bone volume/tissue volume (p < 0.01), and trabecular number (p < 0.05), with a concurrent significant increase in trabecular pattern factor (p < 0.01). Immunohistochemical labelling for LC3 revealed azathioprine to induce autophagy in the bone marrow. Together these data suggest that azathioprine treatment may have a deleterious effect on IBD patients who may already be at increased risk of osteoporotic bone fractures and thus will inform on future treatment strategies for patient stratification.

Growth retardation and apoptotic death of tumor cells by Artemisia herba-alba oral administration in Ehrlich solid carcinoma bearing mice

At present, there is a rapidly growing interest in studying the cytotoxic effects of Artemisia herba alba Asso, Asteraceae, in various cancer cell lines. However, its antitumor effectiveness has not been investigated. Therefore, the current study was conducted to study the effect of A. herba alba extract on the proliferation and growth of solid tumor cells in Ehrlich Solid Carcinoma bearing mice. Oral administration of A. herba alba extract resulted in significant reductions in tumor size, tumor weight and mice body weight, as well as caused concurrent significant increases in the DNA breakages and apoptotic DNA damage induction in a time-dependent manner. A. herba alba extract also raised the expression level of p53 gene and reduced of K-ras expression in a time-dependent manner. Minor histological lesions were observed in the liver and kidney tissues sections of mice administered A. herba alba extract compared with the high histological lesions observed in the liver and kidney tissues of artesunate and cisplatin treated groups. Thus, we concluded that A. herba alba extract exhibited promising potential antitumor efficacy with greater safety than artesunate and the commercially used anticancer drug cisplatin in mice.

Evaluation of Temporal Changes in Urine-based Metabolomic and Kidney Injury Markers to Detect Compound Induced Acute Kidney Tubular Toxicity in Beagle Dogs.

Urinary protein biomarkers and metabolomic markers have been leveraged to detect acute Drug Induced Kidney Injury (DIKI) in rats; however, the utility of these indicators to enable early detection of DIKI in canine models has not been well documented. Therefore, we evaluated temporal changes in biomarkers and metabolites in urine from male and female beagle dogs. Gentamicin- induced kidney lesions in male dogs were characterized by moderate to severe tubular epithelial cell degeneration/necrosis, epithelial cell regeneration and dilation; and a unique urinebased metabolomic fingerprint. These metabolite changes included time and treatment-dependent increases in lactate, taurine, glucose, lactate, alanine, and citrate as well as 9 other known metabolites. As early as 3 days post dose, gentamicin induced increases in urinary albumin, clusterin, neutrophil gelatinase associated protein (NGAL) and total protein concentrations. Urinary albumin, clusterin, and NGAL showed earlier and more robust elevations than traditional kidney safety biomarkers, blood urea nitrogen and serum creatinine. Elevations in urinary kidney injury molecule 1 (KIM-1) were less reliable for detection of gentamicin nephrotoxicity in dogs based on values generated utilizing multiple first-generation, canine-specific KIM-1 immunoassays. The metabolic fingerprint was further evaluated in male and female dogs that received Compound A which induced slightly reversible renal tubular alterations characterized as degeneration/necrosis and concurrent significant increases in urinary taurine amongst other markers. These data support further investigations to demonstrate the value of urinary metabolites, albumin, clusterin, NGAL and taurine as promising markers to enable early detection of DIKI in dogs.

Three cycles of AC chemotherapy regimen increased oxidative stress in breast cancer patients: A clinical hint.

Background:Recent studies have suggested the importance of oxidant/antioxidant status in initiation and progression of breast cancer. The aim of this study was to evaluate oxidative stress markers in breast cancer patients before and after 3 cycles of chemotherapy with adriamycin and cytoxan (AC). Also, in this study the effect of age and the stage of disease on oxidative stress markers were compared and evaluated.Methods:This study included 60 women with newly diagnosed stage II-III breast cancer who underwent chemotherapy with AC as the therapy-first strategy after surgery. Serum samples were obtained before treatment and after the third chemotherapy. Then, serum total antioxidant status (TAS) and malondialdehyde (MDA) as lipid peroxidation marker were analyzed. Moreover, the effects of the subject’s age and clinical disease stage were investigated.Results:A concurrent significant increase in MDA (p<0.001) and a significant decrease in TAS (p<0.001) were also observed after 3 cycles of AC chemotherapy. In addition, some changes were found in the status of oxidative stress markers which were associated with age and clinical disease stage.Conclusion:Our data indicated that chemotherapy with AC increase the oxidative stress in breast cancer patients. The present study indicated that higher stages of the breast cancer are associated with significant increases of oxidative stress markers.

Trends in Route of Hysterectomy With the Implementation of a Comprehensive Robotics Training Program [21B

INTRODUCTION: To evaluate trends in mode of hysterectomy performed at a large teaching hospital and a small community hospital over a 5 year period following the implementation of a comprehensive robotics training program. METHODS: A review of hysterectomies done at a large teaching hospital and a smaller, community hospital in the same health system over a 5 year period was performed to determine how many hysterectomies were performed using an open, laparoscopic, or vaginal approach. The study included 5175 women, and was evaluated after the implementation of a comprehensive teaching, training, mentoring and proctoring program at each hospital. Laparoscopic procedures included TLH and LSCH, as well as RATLH and RASCH. Vaginal Hysterectomies included TVH and LAVH cases. RESULTS: We found a significant decrease in the percent of cases performed through an open approach over the course of the study, with a concurrent significant increase in the percent of robotic procedures performed. There was also a significant decrease in the percent of case performed vaginally over this period, but this was only observed at the smaller community hospital site. CONCLUSION: At the larger, teaching hospital, the implementation of a comprehensive robotics training program was associated with a significant conversion of cases performed through an open approach to a robotic assisted laparoscopic approach. This was not associated with a decline in the percent of vaginal cases performed. At the smaller, community hospital, more dramatic increases were observed with respect to robotic hysterectomies: however, a decline in the percent of cases performed vaginally was observed.

Frozen ground temperature trends associated with climate change in the Tibetan Plateau Three River Source Region from 1980 to 2014

Long-term (1981�2014) trends in soil temperatures at depths ranging from 0�320 cm were used to examine relationships between regional climate change and soil temperatures in the Three River Source Region (TRSR) of the Tibetan Plateau (TP). Air temperature, precipitation, maximum depth of frozen ground and maximum snow depth were also analysed for trends and for correlations with soil temperatures. A significant warming trend was evident in the TRSR during the 35 yr analysed. Significant soil warming was detected, at rates of 0.706°C decade �1 for surface soils (0 cm), 0.477°C decade �1 for shallow layer soils (5�20 cm) and 0.417°C decade �1 for deep layer soils (40�320 cm). There was a clear effect of air temperature on soil temperature, as seen from the concurrent significant increases in air and soil temperature trends and the strong cor - relation between them. The relationship between precipitation and soil temperatures was com - plicated. Higher levels of precipitation on the ground also resulted in higher soil temperatures during the summer in the frozen soil, while the process of freezing and thawing had an inhibitory effect on the soil temperature increase. The warming trends in soil temperature are expected to continue with the degradation of frozen soil in the TRSR.

THIAFENTANIL-AZAPERONE-XYLAZINE AND CARFENTANIL-XYLAZINE IMMOBILIZATIONS OF FREE-RANGING CARIBOU (RANGIFER TARANDUS GRANTI) IN ALASKA, USA.

Carfentanil-xylazine (CX) has been the primary drug combination used for immobilizing free-ranging ungulates in Alaska, US since 1986. We investigated the efficacy of a potential new drug of choice, thiafentanil (Investigational New Animal Drug A-3080). Captive trials indicated that thiafentanil-azaperone-medetomidine could provide good levels of immobilization. However, field trials conducted in October 2013 on free-ranging caribou ( Rangifer tarandus granti) calves showed the combination too potent, causing three respiratory arrests and one mortality. The protocol was revised to thiafentanil-azaperone-xylazine (TAX), with good results. The induction time was not significantly different between the two combinations. However, the recovery time was significantly shorter for the TAX group than for the CX group. A physiologic evaluation was performed on 12 animals immobilized on CX and 15 animals on TAX. Arterial blood was collected after induction and again after 10 min of intranasal oxygen supplements (1 L/min). Both groups had significant increases in partial pressure of arterial oxygen after oxygen treatment. There was a concurrent significant increase in partial pressure of arterial carbon dioxide in both groups. Rectal temperature increased significantly in both groups during the downtime, which is consistent with other studies of potent opioids in ungulates. On the basis of our results, we found TAX to be a potential alternative for the current CX protocol for immobilizing free-ranging caribou calves via helicopter darting.

Protective Effects of Ginger (Zingiber officinale) Extract against Diabetes-Induced Heart Abnormality in Rats.

BackgroundDiabetic cardiomyopathy is an important causal factor in morbidity and mortality among diabetic patients, and currently, no effective means are available to reverse its pathological progress. The purpose of the present study was to investigate the effect of ginger extract on apolipoproteins (apo) A and B, hyperhomocysteinemia, cathepsin G and leptin changes, as well as cardiac fibrosis and heart muscle cell proliferation under hyperglycemic conditions in vivo.MethodsTwenty-four male Wistar rats were divided into three groups, namely: control, non-treated diabetic, and ginger extract-treated diabetic groups. The ginger extract-treated diabetic group received a 50 mg daily dose of ginger extract intragastrically for 6 weeks.ResultsThe results revealed concurrent significant increases in plasma C-reactive protein (CRP), homocysteine (Hcy), cathepsin G and apoB levels and decreases in apoA and leptin levels in the non-treated diabetic group compared to the control group. Moreover, heart structural changes, including fibrosis and heart muscle cell proliferation, were observed in non-treated diabetic rats compared to the control rats. Significant amelioration of changes in the heart structure together with restoration of the elevated levels of Hcy and CRP, leptin, cathepsin G, and apoA and B were found in the ginger extract-treated diabetic group compared to the non-treated diabetic group.ConclusionThe findings indicated that ginger extract significantly reduces heart structural abnormalities in diabetic rats and that these effects might be associated with improvements in serum apo, leptin, cathepsin G, and Hcy levels and with the antioxidant properties of ginger extract.

The content and structure of electronic health records in physiotherapy

Participants wore an accelerometer for 7 days and completed the Nordic Musculoskeletal Questionnaire before the intervention period and again following the 12 week intervention.Accelerometer determined sedentary time, sustained sedentary time, breaks in sedentary time, light activity and MVPA on work days and during work hours was analysed using repeated measures t-tests to evaluate the overall effect of any intervention across all participants. Linear regression models were used to test effects between the organisations and between intervention groups. McNemar’s test was used to assess changes inmusculoskeletal pain following the intervention period. Results: For all participants, therewas a significant reduction in sedentary time on work days (−1.6%, p= 0.006) and during work hours (−1.7%, p= 0.014) and a concurrent significant increase in light activity during work hours (1.5%, p= 0.036) and MVPA on work days (0.6%, p= 0.012). In addition, there was a significant increase in the number of breaks/sedentary hour work days (0.64, p= 0.005) and during work hours (0.72, p= 0.015). Further, following the intervention period, there was no significant difference in the proportion of participants reporting musculoskeletal pain in the different body regions nor was there a significant difference in the reported number of body regions experiencing musculoskeletal pain. Conclusion(s):Participatoryworkplace interventions that modify work practice to reduce occupational sedentary behaviour can reduce sedentary time, increase the frequency of breaks in sedentary time and improve light intensity activity andMVPA of office workers. Small changes in occupational sedentary time did not impact on musculoskeletal symptoms of office workers. Implications: Currently, it is still inconclusive whether changes in occupational sedentary behaviour can change musculoskeletal symptoms of office workers. Further research is needed to establish dose response relationships between occupational sedentary behaviour and various health outcomes, including work-related musculoskeletal symptoms.

Assessment of Hypolipidaemic Effect of Leptadenia Hastata Leaves in Albino Rats

This study was designed to determine the effect of Leptadenia hastata leaves on serum lipid profile and glucose in experimentally-induced hyperlipidemic Wistar rats. Sixteen adult Wistar rats were randomly assigned into four groups of four rats each. Group 1 was the hyperlipidemic group (positive control), Group 2 was the hyperlipidemic supplemented with L. hastata leaves group, Group 3 was the normolipidemic supplemented with L. hastata leaves group and Group 4 normal group fed on normal diet (negative control). The changes in the serum lipid profile and glucose were monitored after three weeks. The result of the serum lipid analysis showed a significant (p<0.05) reduction in the serum total cholesterol, low-density lipoprotein cholesterol with concurrent significant (p<0.05) increase in serum high density lipoprotein cholesterol in hyperlipidemic group treated with L. hastata leaves (group 2) compared with the hyperlipidemic group (group 1). Serum glucose level was also significantly higher in hyperlipidemic group (p<0.05) compared with other three groups. The result of this study suggests possible hypolipidemic effect of L. hastata leaves.

Augmentation of antioxidant system: Contribution to antimalarial activity of Clerodendrum violaceum leaf extract

Reactive oxygen species are known to mediate various pathological conditions associated with malaria. In this study, the antioxidant potential of Clerodendrum violaceum leaf extracts, an indigenous antimalarial remedy, was evaluated. Total phenol, flavonoid, selenium, vitamins C and E contents of Clerodendrum violaceum leaf extracts were determined. The free radical scavenging activities of the extracts against DPPH, superoxide anion and hydrogen peroxide coupled with their reducing power were also evaluated in vitro. Moreover, responses of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR) in a rodent malaria model to a 4-day administration of Clerodendrum violaceum leaf extracts were also evaluated. The methanolic extract was found to contain the highest amounts of antioxidant compounds/element and also demonstrated the highest free radical scavenging activity in vitro. The results showed a significant decrease (p < 0.05) in SOD and CAT activities with a concurrent significant (p < 0.05) increase in GPx and GR activities in both erythrocytes and liver of untreated Plasmodium berghei NK65-infected animals compared to the uninfected animals. The extracts were able to significantly increase (p < 0.05) SOD and CAT activities and significantly reduce (p < 0.05) GPx and GR activities in both the liver and erythrocytes compared to those observed in the untreated infected animals. The results suggest the augmentation of the antioxidant system as one of the possible mechanisms by which Clerodendrum violaceum extract ameliorates secondary effects of malaria infection, alongside its antiplasmodial effect in subjects.

The influence of channel deepening on estuarine turbidity levels and dynamics, as exemplified by the Ems estuary

Active deepening of tidal channels usually results in the alteration of the vertical and the horizontal tide. This may lead to concurrent significant increases in mean suspended matter concentrations (SPM) in coastal plain estuaries, the turbidity maximum (ETM) included. This is exemplified by an extensive analysis of the Ems estuary, a prototypical heavily stressed estuary in the Dutch-German border area. Measurements show that the SPM concentrations in the lower reaches of the estuary have increased an average of 2- to 3-fold between 1954 and 2005, with a 10-fold increase observed in the upper estuary (tidal river). Longitudinal profiles of surface SPM demonstrate that the ETM has moved upstream by up to 25 km and has broadened into a zone 30 km in length which extends into the freshwater tidal river. On an annual scale, variations in freshwater discharge significantly influence the formation and breakdown of the ETM: during low river discharge the ETM approaches equilibrium over 2–3 months, whilst elevated river discharges relocate the ETM downstream over several weeks.An exploratory, semi-analytical model is calibrated to simulate the equilibrium SPM distribution in the upper estuary during five time periods from 1965 to 2005, using archival bathymetric and tidal data. Results suggest that the deepening of tidal channels and a reduction in hydraulic drag have most likely resulted in a landward shift of the SPM trapping location. The measured increase in SPM concentrations and the development of fluid mud around the 1990s likely contributed to reduced mixing and bottom drag, creating a feedback loop that further altered tidal and SPM dynamics. It is argued that the removal of some non-erodible (consolidated) layers in the lower reaches of the estuary has created new internal sediment sources that may be responsible for feeding the observed high SPM concentrations, rather than increased sediment input from the boundaries. All findings are based on and supported by measured short-term seasonal fluctuations, as well as long-term developments of yearly averaged concentrations in the longitudinal SPM distribution.

Long-term ethanol consumption initiates atherosclerosis in rat aorta through inflammatory stress and endothelial dysfunction

Controversy exists on whether alcohol has a direct cardioprotective effect or it provokes atherosclerosis, so the present study sought to assess the effect of chronic consumption of ethanol on the markers of endothelial function, vessel rigidity, and atherosclerosis in the aorta of rat. Male Wistar rats were selected randomly and exposed to ethanol (4.5g⁄kg of 20% w/v solution in saline) once per day for 6weeks. Blood pressure, hemodynamic parameters, foam cell formation, intercellular adhesion molecule-1, vascular cell adhesion molecule-1, endothelial-leukocyte adhesion molecule-1, and high-sensitivity C-reactive protein (CRP) were assessed in ethanol treated rats and compared with either sham or control rats. The results revealed a concurrent significant increase of adhesion molecules, CRP levels, systolic, diastolic, pulse, and dicrotic pressures as well as enhanced formation of foam cell in ethanol-treated rats. These findings implicate that long-term ethanol exposure provokes atherogenic and hemodynamic changes via significant induction of proinflammatory response, augmenting of cell adhesion molecules, stiffness in rat aorta wall and induction of foam cell formation.
