Comparative toxicity of fipronil, malathion, and thiamethoxam on the stingless bee Tetragonisca fiebrigi (Schwarz, 1938)

Stingless bees are important pollinators for various plant crops. We investigated the susceptibility of Tetragonisca fiebrigi to sublethal concentrations of insecticides fipronil, malathion, and thiamethoxam (administered through contact and ingestion) by determining the LC50 values after 24 hours of exposure and analyzing changes in the activity of esterase isoenzymes and the chromatin in brain cells. The LC50 values showed that all three insecticides were highly toxic through contact and ingestion. Electrophoretic analysis revealed that the relative EST-4 (carboxylesterase) activity in T. fiebrigi was partially inhibited by malathion and fipronil ingestion. Moreover, the EST-4 band intensity was increased following high-concentration thiamethoxam (contact) exposure, indicating the increased relative activity of this isoenzyme to detoxify the compound. In the cytochemical analysis of brain cells, the critical electrolyte concentration (CEC) points for the control stingless bees and malathion ingestion-exposed and thiamethoxam-exposed (contact and ingestion) stingless bees were in the range of 0.20-0.30 M MgCl2, whereas that for malathion contact-exposed bees was 0.15 M MgCl2, indicating chromatin relaxation and suggesting an increase in gene expression. In conclusion, T. fiebrigi stingless bees are susceptible to the insecticides tested, and the parameters analyzed may be used as biomarkers to detect the presence of these compounds.

Acute toxic effects of thiamethoxam on Chinese mitten crab Eriocheir sinensis.

The information about toxic effects of thiamethoxam on non-target aquatic organisms is still incomplete. The semi-static toxicity test method was used to investigate the acute toxic effects of thiamethoxam on Eriocheir sinensis. The results showed that the median lethal concentration (LC50) of thiamethoxam to E. sinensis at 96h was 510μg/L, and the safety concentration (SC) was 51μg/L. After 96h exposure to thiamethoxam, the survival rates of crabs at concentrations of 0, 151.11, 226.67, 340, and 510μg/L were 100%, 76.19%, 64.29%, 61.91%, and 46.43%, respectively. A significant (P < 0.05) decrease of the number of hemocytes was observed in thiamethoxam groups. With the increase of thiamethoxam concentration, the phagocytic activity of hemocytes, the activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px), total antioxidant capacity (T-AOC), and the activities of protease, amylase, and lipase of crabs increased firstly and then decreased, and the above indexes reached the maximum in 151.11μg/L thiamethoxam group. Collectively, a high concentration of thiamethoxam (510μg/L) had a great effect on the gene expression of immune metabolism-related factors in hepatopancreas and gill of crabs. These findings indicated that thiamethoxam exposure had the ability to impair immune and metabolic systems and resulted in the reduction of survival rate of crabs.

Single and joint toxicity assessment of acetamiprid and thiamethoxam neonicotinoids pesticides on biochemical indices and antioxidant enzyme activities of a freshwater fish Catla catla

Neonicotinoids pesticides are extensively used in many countries due to their high insect selectivity. Acetamiprid and thiamethoxam are the neonicotinoids most commonly detected in the aquatic environment. This work examined the single and joint toxicity of acetamiprid and thiamethoxam in a freshwater fish Catla catla. Fish were exposed to acetamiprid (0.5 mg/L and 1 mg/L), thiamethoxam (0.01 mg/L and 0.5 mg/L) and their binary mixtures (0.5 mg/L of acetamiprid and 0.01 mg/L of thiamethoxam) for 96 h. The stress biomarkers such as glucose, protein, electrolytes, Na+/K+ -ATPase and oxidative stress were evaluated. Among the biochemical parameters, plasma protein, electrolytes (sodium, potassium and chloride) and gill ATPase activity were decreased in response to individual and binary mixtures treatments. In contrast, blood glucose level showed significant increase in all the treatments. Exposure to various concentrations of acetamiprid and thiamethoxam resulted in significant decrease in superoxide dismutase (SOD) activity in the gill tissue. However, SOD activity was significantly elevated during binary mixtures treatment. Glutathione peroxidase (GPx), catalase (CAT), glutathione-S-transferase (GST) and reduced glutathione (GSH) levels in gills were decreased significantly after individual and binary mixtures treatments. Fish exposed at individual and binary mixtures significantly elevated the level of LPO in gill tissue. Our findings suggest that multi-biomarker approach can be effectively used to assess the effects of joint toxicity of pesticides and to monitor the neonicotinoids pesticides in the aquatic environment.

Multibiomarker responses after exposure to a sublethal concentration of thiamethoxam in the African honeybee (Apis mellifera intermissa)

Thiamethoxam is an insecticide mainly used in agriculture to control insect pests. However, non-target insect species, such as honeybees, may also be impacted. In this study, adults of Apis mellifera intermissa were orally exposed under laboratory conditions to a sublethal concentration of thiamethoxam (CL25= 0.17 ng/μl) for 9 days and the effects were evaluated at the biochemical level, by monitoring specific oxidative stress and neuronal biomarkers. Results showed an increase in the antioxidant enzymes, glutatione-S-transferase (GST), catalase (CAT) and glutathione peroxidase (GPx) and in content of malondialdehyde (MDA).The activity of acetylcholinesterase (AChE) was downregulated as evidence of a neurotoxic action and no significant change was observed in glutathione (GSH). Exposure to the insecticide thiamethoxam induced oxidative stress and defense mechanisms affecting honeybee physiology.

Neonicotinoid contamination in wildflowers collected from citrus orchards in a northwestern Mediterranean Region (Spain) after tree foliar treatments

Ground-cover vegetation attracts and harbors beneficial insects to the agrosystem, playing an important role in conservation biological control. Integrated pest management (IPM) program guidelines recommend the implantation of sowed or resident wild covers in perennial crops. Given the high-quality fruit requirements, even in IPM programs, insecticides can be required in citrus crops. This study presents, over a year, the levels of neonicotinoids (thiamethoxam and imidacloprid) in not-target ground-cover wildflowers growing spontaneously in citrus orchards after foliar treatment of citrus trees. The presence and persistence of these neonicotinoids in different wildflower species were studied. Concentrations of thiamethoxam and imidacloprid in whole wildflowers ranged from < method quantification limit (MQL) to 52.9 ng g−1 and from < MQL to 98.6 ng g−1, respectively. Thiamethoxam was more frequently detected than imidacloprid. Thiamethoxam and imidacloprid were detected up to 336 and 230 days after treatment, respectively. The highest detection frequencies (100%) and highest thiamethoxam and imidacloprid mean concentrations (26.0 ± 7.3 ng g−1 and 11.0 ± 10.6 ng g−1, respectively) occurred in wildflowers collected 9 days after the treatments. Since application, a clear decrease in the concentration of both compounds and differences in the accumulation depending on wildflower species were observed. Cross contamination was detected, indicating a transport from adjacent treated plots. Maintaining a cover crop in citrus orchards may lead to detrimental effects on non-target arthropods if these neonicotinoid compounds are used for pest control since they can entail a chronic exposure during at least 230 days for imidacloprid and 336 days for thiamethoxam.

Uptake, translocation, and metabolism of thiamethoxam in soil by leek plants

Thiamethoxam (TMX) is commonly applied on leek plants by root irrigation. It might be taken up by leek plants and thus has lasting dietary risk. In this study, the uptake, translocation, and metabolism of TMX in leek plants were investigated. The results obtained from both the hydroponic and soil experiments indicated that TMX could be easily translocated upward and accumulated in leek shoots after being absorbed by roots. The total absorbed TMX amount (Mtotal) in leek plants from the tested soils varied greatly with its adsorption governed by soil characteristics. Interestingly, Mtotal was closely correlated with the concentration of TMX in in situ pore water, indicating that TMX in in situ pore water could be a useful approach to predict uptake of this chemical by leek plants from various soils. Profoundly, clothianidin (CLO) was detected with concentration of 0.07–1.54 mg/kg in roots and 0.27–4.12 mg/kg in shoots at 14 d, respectively, suggesting that TMX is easily converted into CLO in leek plants. The results showed that TMX used in soil is easily absorbed by leek and accumulated in edible parts accompanying with formation of CLO.

Embryonic Exposure to Thiamethoxam Reduces Survival and Alters Neurobehavior of Fathead Minnows.

Thiamethoxam is a commonly used neonicotinoid insecticide that acts as a nicotinic acetylcholine receptor (nAChR) agonist. Although vertebrates are less sensitive to neonicotinoid insecticides than invertebrates, some neonicotinoids have been shown to cause neurobehavioral changes in larval fishes. In the present study, we examine the neurobehavioral toxicity of acute and chronic exposure to environmentally relevant concentrations of thiamethoxam in fathead minnows at two different life stages. Whereas acute exposure of embryos to thiamethoxam does not appear to stimulate spontaneous contractions within 1 min, chronic exposure of embryos to 1.57 µg or more thiamethoxam/L caused increased mortality as well as a subtle increase in spontaneous contraction frequency (SCF), which was negatively correlated with early hatching success. Chronic exposure of embryos to 155 µg thiamethoxam/L impaired predator escape response, and chronic exposure to 0.02-14.61 µg thiamethoxam/L impaired foraging efficiency of some fish. Fathead minnows exposed to thiamethoxam beginning post hatch did not experience changes to measured health or neurobehavioral indicators. Taken together, our findings indicate that embryonic life stages are more sensitive to thiamethoxam exposure than later larval life stages. Because early exposure to thiamethoxam can cause deficits in predatory escape behaviors and may impair foraging success, further study of the potential direct and nondirect impacts of thiamethoxam on wild fish populations is warranted.Environ Toxicol Chem 2022;41:1276-1285. © 2022 SETAC.

Influence of biochar on the sorption and leaching of thiamethoxan in soil

This study evaluated the long-term effect of biochar application on the sorption and desorption of thiamethoxam in a Haplic Plinthosol. The experiment was conducted in a randomized block design with combination of doses of mineral fertilizer NPK (0 and 300 kg·ha−1 formula 05-25-15) and biochar (0, 16 and 32 Mg ha−1). Deformed soil samples were collected in the field from the 0-0.10 m layer of all plots to determine the sorption and desorption of the thiamethoxam insecticide in the soil; fulvic acid (FA), humic acid (AH) and humin (HUM) carbon contents; and total organic carbon. The Batch slurry method was used to evaluate sorption and desorption. The Freundlich isotherm adequately described thiamethoxam sorption in all treatments. The application of biochar increased the sorption (Kfs) and decreased the desorption (Kfd) of thiamethoxam. The sorption intensity (1/n) showed reduction characteristics as the soil concentration of thiamethoxam increased. Biochar has a sorption effect on the soil through covalent bonds and H-bonds with the insecticide molecules, thereby indirectly increasing the sorption potential in the chemical fractions of the organic matter of soil. The application of 32 Mg ha−1 of biochar significantly reduced thiamethoxam in the soil solution. Altogether, the present study reveals that biochar application in soil is a promising tool for mitigating the contaminant potential of thiamethoxam in subsurface waters.

Thiamethoxam affects the developmental stages of banded gourami (Trichogaster fasciata)

Thiamethoxam (THM), a type III systemic neonicotinoid insecticide, is widely used in agriculture in many countries, including Bangladesh. THM can enter the aquatic systems through the runoff, spray-drift and groundwater leaching and can affect the aquatic organisms, including fish. Current environmental levels of THM in Bangladesh waters are variable. However, the presence of this chemical in the aquatic environment and its possible effects on the fish inhabiting those water bodies is concerning. To understand the effects of environmental THM on the development of embryo and larvae, the present study used banded gourami (Trichogaster fasciata), a freshwater fish species distributed in different Asian countries, including Bangladesh. In laboratory setting, fertilized eggs (n = 100) and one-day-old banded gourami larvae (n = 100) were exposed to six concentrations of THM (0, 0.02, 0.2, 2, 20, 200 mg/L) in three replicates, in which three concentrations were within the environmentally relevant levels. Hatching rate, incubation period, mortality, and malformations of embryo and larvae were observed. The hatching success and survival of embryo and larvae significantly decreased with increasing THM concentrations. The 24-h LC50 of THM for the embryo was 4.24 mg/L. The 24-h, 48-h, 72-h, and 96-h LC50 values of THM for one-day-old larvae were 12.20, 3.80, 0.78, and 0.27 mg/L, respectively. Overall developmental malformations included lordosis, notochord abnormality, yolk-sac edema, dark brown yolk sac, hemorrhage, and irregular caudal fin. These abnormalities in embryos were common across all the concentrations of THM applied. The results of the present study suggest that environmentally relevant concentrations of THM can induce developmental defects in the embryo and larvae of banded gourami.

Thiamethoxam as an inadvertent anti-aphrodisiac in male bees

Sexual reproduction is common to almost all multi-cellular organisms and can be compromised by environmental pollution, thereby affecting entire populations. Even though there is consensus that neonicotinoid insecticides can impact non-target animal fertility, their possible impact on male mating success is currently unknown in bees. Here, we show that sublethal exposure to a neonicotinoid significantly reduces both mating success and sperm traits of male bumblebees. Sexually mature male Bombus terrestris exposed to a field-realistic concentration of thiamethoxam (20 ng g−1) or not (controls) were mated with virgin gynes in the laboratory. The results confirm sublethal negative effects of thiamethoxam on sperm quantity and viability. While the latency to mate was reduced, mating success was significantly impaired in thiamethoxam-exposed males by 32% probably due to female choice. Gynes mated by exposed males revealed impaired sperm traits compared to their respective controls, which may lead to severe constraints for colony fitness. Our laboratory findings demonstrate for the first time that neonicotinoid insecticides can negatively affect male mating success in bees. Given that holds true for the field, this provides a plausible mechanism contributing to declines of wild bee populations globally. The widespread prophylactic use of neonicotinoids may therefore have previously overlooked inadvertent anti-aphrodisiac effects on non-target animals, thereby limiting conservation efforts.

Acute and Chronic Effects of Clothianidin, Thiamethoxam and Methomyl on Chironomus dilutus

Organism tolerance thresholds for emerging contaminants are vital to the development of water quality criteria. Acute (96-h) and chronic (10-day) effects thresholds for neonicotinoid pesticides clothianidin and thiamethoxam, and the carbamate pesticide methomyl were developed for the midge Chironomus dilutus to support criteria development using the UC Davis Method. Median lethal concentrations (LC50s) were calculated for acute and chronic exposures, and the 25% inhibition concentrations (IC25) were calculated for the chronic exposures based on confirmed chemical concentrations. Clothianidin effect concentrations were 4.89 µg/L, 2.11 µg/L and 1.15 µg/L for 96-h LC50, 10-day LC50 and 10-day IC25, respectively. Similarly, thiamethoxam concentrations were 56.4 µg/L, 32.3 µg/L and 19.6 µg/L, and methomyl concentrations were 244 µg/L, 266 µg/L and 92.1 µg/L. Neonicotinoid effect concentrations compared favorably to previously published 96-h and 14-day LC50 concentrations, and methomyl effect concentrations were within the acute survival range reported for Chironomus species and other organisms.

Exposure assessment of neonicotinoid insecticides and their metabolites in Chinese women during pregnancy: A longitudinal study

Neonicotinoid insecticides (NNIs) are the most widely used insecticides globally and ubiquitous in the environment, which has led to widespread human exposure. However, studies on internal exposure levels of NNIs and their metabolites in pregnant women are scarce. In this study, we measured nine parent NNIs and ten main metabolites in 1224 urine samples donated by 408 pregnant women at three trimesters. In the urine samples, the unadjusted vs. specific gravity (SG) adjusted median concentrations and detection frequencies (DFs) of desmethyl-acetamiprid (DM-ACE; 1.01 vs. 1.08 ng/mL; DF: 99.7%), 5-hydroxy-imidacloprid (5-hydroxy-IMI; 0.54 vs. 0.56 ng/mL; 98.5%), imidacloprid-olefin (IMI-olefin; 0.41 vs. 0.44 ng/mL; 99.3%), and desnitro-imidacloprid (DN-IMI; 0.12 vs. 0.12 ng/mL; 90.4%) were higher than their corresponding parent NNIs, acetamiprid (ACE; <0.01 vs. <0.01 ng/mL; 26.4%) and imidacloprid (IMI; 0.04 vs. 0.04 ng/mL; 69.9%). The unadjusted and SG-adjusted median concentrations of clothianidin (CLO), thiamethoxam (THM), and desmethyl-clothianidin (DM-CLO) were 0.05 vs. 0.07, 0.05 vs. 0.06, and 0.04 vs. 0.05 ng/mL, with the DFs of 61.0%, 57.5%, and 75.7%, respectively. The cumulative exposure level, imidacloprid-equivalent total NNIs (IMIeq), was generated by the relative potency factor approach considering the toxic effects of NNIs and their metabolites. The unadjusted IMIeq varied from 0.17 ng/mL (SG-adjusted: 0.20) to 1969 ng/mL (SG-adjusted: 1817) with a median of 14.1 ng/mL (SG-adjusted: 14.1). A decreased trend was observed in urinary NNIs and their metabolites throughout the three trimesters. Maternal age, educational level, and household income were related to the concentrations of NNIs and their metabolites. DM-ACE, 5-hydroxy-IMI, and IMI-olefin were significantly lower in winter than in autumn; DN-IMI, THM, CLO, and DM-CLO were significantly higher in both summer and autumn than in winter. The maximum estimated daily intake of IMIeq [34.8 μg/kg-body weight (bw)/d] was lower than the chronic reference dose of IMI (57 μg/kg-bw/d) currently recommended by the United States Environmental Protection Agency. Human health risk of exposure to NNIs and their main metabolites warranted further studies.

Urinary neonicotinoid insecticides in children from South China: Concentrations, profiles and influencing factors

Neonicotinoid insecticides can selectively interact with the unique nicotinic acetylcholine receptor subtypes in insects and are considered to be low toxic to mammals. However, there is still insufficient knowledge on human exposure to neonicotinoid insecticides, especially for children. This study aimed to investigate urinary concentrations and profiles of neonicotinoid insecticides in South China children and to analyze potential influencing factors. Six neonicotinoid insecticides, including imidacloprid (IMI), thiamethoxam (THM), acetamiprid (ACE), clothianidin (CLO), thiacloprid (THD) and dinotefuran (DIN), exhibited high detection frequencies (>90%) in urine samples collected from 305 children, suggesting broad exposure in South China children. The median concentrations were determined to be 0.13, 0.21, 0.01, 0.19, 0.002 and 1.64 μg/L, respectively. Among the target neonicotinoids, urinary concentrations of CLO and THM exhibited a significant and positive correlation between each other (p < 0.05), suggesting similar sources of these two chemicals.

Quantifying neonicotinoid insecticide residues in milkweed and other forbs sampled from prairie strips established in maize and soybean fields

Adding habitat within agricultural ecosystems is necessary to reverse declines in pollinator abundance and diversity. Understanding pesticide exposure to pollinator habitat near crop fields is necessary to support risk assessments. Neonicotinoids applied to maize and soybean seeds can be transported from crop fields to adjacent habitat through dust drift during planting and/or through overland runoff or subsurface flow following planting. Pollinators, especially bees (e.g. Apidae) and monarch butterflies (Danaus plexippus), could be exposed to neonicotinoids through ingestion of contaminated plant material (e.g. milkweed leaves). Neonicotinoids have been detected in pollinator-attractive habitats near crop fields; however, the magnitude and seasonal variation of the concentration of these insecticides has not been determined. We quantified concentrations of clothianidin, thiamethoxam, and imidacloprid in soil and forb leaf tissue, including milkweed (Asclepias spp.) located within reconstructed prairies (3–4 years post-establishment) within or adjacent to maize or soybean fields. Samples taken from April through August in 2017 and 2018 were analyzed with liquid chromatography-tandem mass spectrometry; 100% of soil, 80% of vegetation from blooming forbs, and 80% of milkweed leaf tissue samples had at least one neonicotinoid present above the method detection limit (0.07–0.9 ng g-1). The maximum concentrations detected in 2017 or 2018 of clothianidin, thiamethoxam, and imidacloprid in milkweed leaf tissue samples were 6.6, 12.9, and 2.8 ng g-1, respectively. These values are 10 to 130-fold lower than the chronic LC10 values for monarch larvae, indicating it is unlikely that this route of neonicotinoid exposure will cause adverse effects to monarch larvae.

A food-ingested sublethal concentration of thiamethoxam has harmful effects on the stingless bee Melipona scutellaris

In recent years, the importance of bee's biodiversity in the Neotropical region has been evidencing the relevance of including native bees in risk assessments. Therefore, the sublethal effects of the insecticide thiamethoxam on the survival and morphological parameters of the stingless bee Melipona scutellaris were investigated in the present study. Cells from both non-target organs (Malpighian tubules and midgut) and target organs (brain) were analyzed for morphological alterations using light microscopy and transmission electron microscopy. The findings showed that when M. scutellaris foragers were exposed to a sublethal concentration of thiamethoxam (LC50/100 = 0.000543 ng a. i./μL), longevity was not reduced but brain function was affected, even with the non-target organs attempting to detoxify. The cellular damage in all the organs was mostly reflected in irregular nuclei shape and condensed chromatin, indicating cell death. The most frequent impairments in the Malpighian tubules were loss of microvilli, disorganization of the basal labyrinth, and cytoplasmic loss. These characteristics are related to an attempt by the cells to increase the excretion process, probably because of the high number of toxic molecules that reach the Malpighian tubules and need to be secreted. In general, damages that compromise the absorption of nutrients, excretion, memory, and learning processes, which are essential for the survival of M. scutellaris, were found. The present results also fill in gaps on how these bees respond to thiamethoxam exposure and will be useful in future risk assessments for the conservation of bee biodiversity.

Juvenile hormone and transcriptional changes in honey bee worker larvae when exposed to sublethal concentrations of thiamethoxam

Thiamethoxam, an insecticide with high usage and large amounts of environmental residues, has been reported to affect the pupation and survival of honey bee larvae at sublethal concentrations. The molecular mechanisms are not fully understood. In this study, we measured the response of juvenile hormone (JH) to environmental concentrations of thiamethoxam using liquid chromatography-tandem mass spectrometry (LC-MS/MS), monitored the dynamic changes in the transcription of genes encoding major JH metabolic enzymes (CYP15A1, FAMET, JHAMT and JHE) using RT-qPCR, and analysed the transcriptome changes in worker larvae under thiamethoxam stress using RNA-seq. Thiamethoxam significantly increased the levels of JH3 in honey bee larvae, but no significant changes in the transcript levels of the four major metabolic enzymes were observed. Thiamethoxam exposure resulted in 140 differentially expressed genes (DEGs). P450 CYP6AS5 was upregulated, and some ion-related, odourant-related and gustatory receptors for sugar taste genes were altered significantly. The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed that amino acid metabolism and protein digestion and absorption were influenced by thiamethoxam. These changes may do harm to honey bee caste differentiation, foraging behaviour related to sensory perception and nutrient levels of bee colonies. These results represent the first assessment of the effects of thiamethoxam on JH in honey bee larvae and provides a new perspective and molecular basis for the study of JH regulation and thiamethoxam toxicity to honey bees.

Enzymatic responses in the head and midgut of Africanized Apis mellifera contaminated with a sublethal concentration of thiamethoxam

The increasing use of insecticides, promoted by the intensification of agriculture, has raised concerns about their influence on the decline of bee colonies, which play a fundamental role in pollination. Thus, it is fundamental to elucidate the effects of insecticides on bees. This study investigated the damage caused by a sublethal concentration of thiamethoxam - TMX (0.0227 ng/μL of feed) in the head and midgut of Africanized Apis mellifera, by analyzing the enzymatic biomarkers, oxidative stress, and occurrence of lipid peroxidation. The data showed that the insecticide increased acetylcholinesterase activity (AChE) and glutathione-S-transferase (GST), whereas carboxylesterase (CaE3) activity decreased in the heads. Our results indicate that the antioxidant enzymes were less active in the head because only glutathione peroxidase (GPX) showed alterations. In the midgut, there were no alkaline phosphatase (ALP) or superoxide dismutase (SOD) responses and a decrease in the activity of CaE was observed. Otherwise, there was an increase in GPX, and the TBARS (thiobarbituric acid reactive substances) assay also showed differences in the midgut. The TBARS (thiobarbituric acid reactive substances) assay also showed differences in the midgut. The results showed enzymes such as CaE3, GST, AChE, ALP, SOD, and GPX, as well as the TBARS assay, are useful biomarkers on bees. They may be used in combination as a promising tool for characterizing bee exposure to insecticides.

Histopathological alterations in the gills of banded gourami (Trichogaster fasciata) exposed to thiamethoxam

Thiamethoxam (THM) is a widely used agro-pesticide in Bangladesh, though European Union has banned this neonicotinoid insecticide due to its toxic effects on non-target organisms. A 60-day experiment was conducted to study the effect of sub-lethal concentrations of THM on gills of Banded Gourami Trichogaster fasciata. The experiment was conducted in 15 PVC tanks each containing 100L water. The 96-h LC50 value of THM was estimated as 161.06mg/L for Banded Gourami. Based on estimated 96-h LC50 value, the experiment was carried out using a control (0 mg/L of THM) and five sub-lethal concentrations (9.37, 18.75, 37.5, 75 and 150 mg/L of THM in water) with three replications. For collection of gills, fishes were sampled on day-30 and day-60 of exposure to THM. Several histopathological changes were observed in the gill tissues of treated fishes viz. clubbing and reduction of gill filaments, telangiectasis of gill lamellae, haemorrhage and damage of gill raker. Water quality parameters (e.g. DO, pH and temperature) were recorded every 15 days interval. The DO levels were found to decrease significantly with increasing THM concentrations and time of exposure, whereas no noticeable changes were observed for pH and temperature. Therefore, neonicotinoid crop insecticide like THM is capable of damaging gill tissues of an small indigenous fish Banded Gourami.

Efficacy of Two Neonicotinoid Insecticides against Invasive Wood Borer Aromia bungii Larvae in Dietary Toxicity Test.

Simple SummaryThis study investigated the effects of two neonicotinoid insecticides (thiamethoxam and dinotefuran) on alien wood borer Aromia bungii, which invaded Japan recently. Small neonates and large larvae were fed artificial diet with different insecticide concentrations and then reared for 3 (neonates) or 12 (large larvae) weeks in the laboratory. Diet excavation immediately dropped in larvae exposed to high concentrations of both insecticides (≥1 ppm in neonates and ≥10 ppm in large larvae). Their growth was significantly suppressed, and the survival rate gradually declined over time (≥87% decline over 12 weeks in large larvae). These effects were similar between neonates and large larvae, but neonates were affected more by lower insecticide concentrations than large larvae. The two insecticides gradually debilitate A. bungii larvae. In practical use, rapid suppression of A. bungii wood boring damage can be expected by injecting these insecticides into infested trees. However, a relatively long–term retention of the insecticides may be required to kill the larvae in the trees, especially large larvae. Neonates may be controlled with less insecticides and shorter exposure than large larvae.In recent years, insecticide trunk injection was put into practical use for controlling wood boring pests. However, few studies have investigated the dose–response relationships between insecticides and wood–boring pests in detail. This study used two commercial formulations of the neonicotinoid insecticides thiamethoxam and dinotefuran and investigated their dose–response relationships with invasive wood borer Aromia bungii (Coleoptera: Cerambycidae) larvae. Neonates and late instar larvae were reared with an artificial diet containing different insecticide concentrations (0.01–100 ppm) in the laboratory, and their diet excavation activity, survival rate, and weight change were recorded. Diet excavation immediately dropped in larvae exposed to high concentrations of thiamethoxam or dinotefuran (≥1 ppm in neonates and ≥10 ppm in late instar larvae). The weight and survival rate gradually declined over 12 weeks in late instar larvae. These results suggest that the two neonicotinoids intoxicate and debilitate A. bungii larvae gradually to death. In practical use, rapid suppression of A. bungii wood boring damage can be expected by trunk injection of neonicotinoid insecticides. However, a relatively long-term retention of the insecticides may be required to kill large larvae. Neonates may be controlled with lower insecticide dosage and shorter exposure than larger larvae.

Temporal Profile of Neonicotinoid Concentrations in Cotton, Corn, and Soybean Resulting from Insecticidal Seed Treatments

Neonicotinoids have been implicated as a contributing factor to the observed decreases in honey bee populations. It has been suggested that honey bees can be exposed to seed-treated neonicotinoids through pollen and nectar from treated plants. To investigate the uptake and persistence of neonicotinoids in plant tissue and soil, we conducted seed treatment trials with corn, cotton, and soybean planted in Mississippi, Arkansas, and Tennessee during the 2013 and 2014 growing seasons. Leaf tissue was collected and analyzed beginning shortly after emergence until plants began to flower to better understand how neonicotinoid concentrations change in plant tissues over time. The youngest leaf in the terminal of the plant was sampled as an indicator of the neonicotinoid concentrations within the plant. Soil samples were also collected and analyzed for neonicotinoid concentrations at the first and last sampling dates. The mean clothianidin concentrations in corn treated with Poncho® 250, 500, and 1250 seed treatments declined by 99.3, 99.3, and 97.8 percent, respectively, as the plants developed from seedlings to reproductive plants. The mean concentration of imidacloprid detected in Aeris®-treated cotton decreased by 99.6 percent during the sampling period. For cotton seed treated with Avicta® Duo, the mean concentrations of thiamethoxam and clothianidin in leaf tissue declined by 99.9 and 100 percent, respectively, by the time flowering occurred. There was a 99.9 percent reduction in the mean concentration of thiamethoxam by the time of flowering in leaf tissue from soybean treated with a CruiserMaxx® seed treatment. Mean clothianidin concentrations completely diminished (&lt;1 ng/g) in CruiserMaxx®- and Poncho®/VOTiVO®-treated soybean plants by the time plants reached reproductive growth. The data for neonicotinoid concentrations in the soil were more variable than leaf tissue samples, and the reduction in neonicotinoid concentrations in leaf tissues did not closely correlate with concentrations in the soil. Our results suggest that neonicotinoid insecticides, when used as seed treatments in these crops, decline rapidly throughout vegetative growth stages. However, the biological impact on target or non-target arthropods was not examined.
