Fetal and Maternal Thyroid Hormone Responses to Ethanol Exposure During the Third Trimester Equivalent of Gestation in Sheep

Abnormal thyroid hormone system function in the mother or fetus during pregnancy can result in brain defects, some of which resemble those found in children with fetal alcohol syndrome. It has been hypothesized that ethanol may act to mediate alcohol-related birth defects in part by altering thyroid hormone system function. We investigated whether a binge pattern of maternal ethanol consumption over the last trimester equivalent of gestation in sheep results in an alteration in fetal or maternal thyroid function. Pregnant ewes received saline or ethanol beginning on day 109 of gestation (term, 145 days) for three consecutive days per week followed by 4 days without exposure. The fetuses were surgically instrumented on day 113, and experiments were performed on days 118 or 132. Fetal and maternal blood samples were collected, and plasma tri-iodothyronine (T3), thyroxine (T4), and free T4 concentrations were measured. Fetal T3 and T4 on day 118, fetal T3 on day 132, and maternal T3 on day 132 were lower in response to ethanol. Fetal and maternal free T4 and maternal T4 did not change in response to ethanol. Fetal thymus and adrenal weights were reduced in response to ethanol. We conclude that, in sheep, maternal ethanol exposure during the third trimester equivalent resulting in blood ethanol concentrations that are commonly achieved by ethanol abusers decreases circulating thyroid hormone concentrations in the mother and fetus and fetal thyroid and thymus mass.

Limitations in energy intake affect the ability of young turkeys to cope with low ambient temperatures

Young turkeys exposed to low ambient temperature ( T a) showed significantly reduced body weight, which coincided with a reduction in energy intake and with changes in the circulatory system to accommodate higher oxygen demand. These changes included a significant increase in hematocrit, hemoglobin concentration, plasma triiodothyronine (T 3) concentration, blood volume, and blood oxygen capacity. At the relatively high T a, changes to accommodate heat dissipation included significant increases in plasma volume and panting rate. These compensations were sufficient to control body temperature ( T b). However, the higher energy expenditure for maintenance followed by significantly higher plasma triiodothyronine (T 3) concentration, but with lower energy intake at low T a, suggest a physical limitation in the ability to further increase energy intake as T a declines.

The Effect of Early Feeding on Growth and Small Intestinal Development in The Posthatch Poult

Poults with early access to feed in the hatchery or turkey house grew more than those reared under standard commercial practice. During 48 h posthatch, fed poults utilized yolk and exogenous feed to increase BW by 11 g. The small intestine increased from 3.8% of BW at hatch to 8.9% after 48 h. In contrast, BW in feed-deprived poults decreased by 10 g, whereas the small intestine increased slightly in weight and composed 4.5% of BW after 48 h. The number of cells per villus and the villus surface area increased dramatically posthatch in the duodenum but more slowly in the jejunum and ileum. Enterocyte width changed little, but length increased more than twofold in the duodenum and by approximately 50% in the jejunum and ileum by 6 d posthatch. Lack of access to feed depressed the rate of growth of villi and enterocyte length in all intestinal segments until 6 d posthatch.All intestinal epithelial cells were proliferating at hatch, which changed rapidly within 48 h posthatch, with proliferating cells becoming located mainly in the intestinal crypts where about half of the cells were proliferating. In feed-deprived poults, the decrease in the proportion of proliferating cells in the crypt was greater than that of fed poults; after refeeding, an increase in the rate of proliferation was observed in feed-deprived poults.Plasma concentrations of Na, glucose, triglycerides, and phospholipids were not affected by feed deprivation; however, nonesterified fatty acid concentrations were enhanced in feed-deprived poults, indicating a greater use of fatty acids for energy. Plasma triiodothyronine (T3) concentrations, which may mediate some of the intestinal effects of feed deprivation, were depressed in poults without access to feed.

Epidemiology and diagnosis of subclinical iodine deficiency in crossbred cattle of Punjab

To determine the prevalence of iodine deficiency in crossbred cows (taurus x indicus) of Punjab and to assess the diagnostic value of the plasma concentrations of thyroid hormones, total cholesterol and free fatty acids in iodine deficiency. The efficacy of ethiodised oil, as a long-term supplement of iodine, was also evaluated. Base-line survey of iodine status was conducted by measuring plasma inorganic iodine (PII) concentration in 48 crossbred cows from 31 dairy units. Concentrations of plasma triiodothyronine (T3), thyroxine (T4), total cholesterol and free fatty acids were compared between iodine deficient and normal cows. Circulating T4 and T3 concentrations were also determined before and after injection of ethiodised oil in cows with low PII levels. The mean prevalence of iodine deficiency was 35.9% and showed considerable geographical variation from 0 to 86% within Punjab. Cardinal clinical signs of iodine deficiency were absent and basal plasma T3 and T4 concentrations and their ratio did not differ between deficient and control cows. Activity of circulating T4 and T4:T3 ratio increased in response to ethiodised oil injection and persisted beyond 70 days after injection. Asymptomatic iodine deficiency is prevalent in crossbred cows of Punjab and response to iodine supplementation is the most reliable index of thyroid dysfunction associated with iodine deprivation. The results of this study also support the view that injection of 1 mL of 78% ethiodised oil can prevent iodine deficiency for more than 70 days.

The effects of dietary L-carnitine supplementation on the performance, organ weights and circulating hormone and metabolite concentrations of broiler chickens reared under a normal or low temperature schedule

1. The present study examined the effects of the addition of 100 mg/kg L-carnitine to the basal starter (containing 17.8 mg/kg L-carnitine) and finisher (containing 22.9 mg/kg L-carnitine) diets on performance, organ weights and plasma hormone and metabolite concentrations of male and female broiler chickens. The broiler chickens were reared either in a room with a normal temperature (NT) program or with a low temperature (LT) program (rapid decrease from 28°C to 20°C at 14 d of age). 2. Broiler chickens reared under the LT schedule consumed more food and attained a greater body weight at 42 d of age than their counterparts reared under NT schedule, without any difference in food efficiency or abdominal fat content. Dietary L-carnitine supplementation had no significant effects on any of these production parameters, except for a reduction in the abdominal fat content of female NT chickens. However, the LT schedule and dietary L-carnitine supplementation greatly increased absolute and proportional heart weights. The elevated heart weights were not due to right ventricle hypertrophy. 3. Both the LT program and L-carnitine supplementation increased circulating plasma triiodothyronine concentrations. There were also some transient effects of both experimental variables on plasma growth hormone, glucose and triglyceride concentrations. 4. L-carnitine did not improve broiler performance. However, this result does not mean that L-carnitine supplementation cannot have beneficial effects in other circumstances. In view of the elevated proportional heart weights, it can also be argued that L-carnitine is a potential agent for reducing the incidence of metabolic diseases in broiler chickens.

Persistent hormonal effects of stress are not due to reduced food intake or exposure to stressed rats.

Exposure to inescapable stress elicits persistent effects on the physiology and behavior of rats. Elevated basal plasma corticosterone concentrations have been observed for several days after cessation of stress. In this study, we measured hormonal concentrations in multiple axes at multiple levels, 24 h after one or three consecutive exposures to the same stress paradigm. The data indicated persistent activation of plasma corticosterone and prolactin concentrations, whereas plasma triiodothyronine, thyroxine, luteinizing hormone, and growth hormone concentrations were inhibited after either one or three stress sessions. In addition, we isolated the effects of restraint/tail shock per se from the effects of being moved and exposed to other stressed rats, and from the effects of reduced feeding produced by our stress protocol. The data clearly indicated that the stress paradigm, rather than exposure to stressed rats or decreased nutrient intake, is necessary to induce the persistent physiologic changes we observe after stressor exposures.

Influence of thyrotropin-releasing hormone administration at birth on thermoregulation in lambs delivered by cesarean

Objective: We examined the hypothesis that exogenous stimulation with thyrotropin-releasing hormone immediately before umbilical cord clamping can improve thermoregulation in near-term lambs delivered by cesarean. Study Design: Twin lambs were injected with either saline solution alone (control, n = 12) or thyrotropin-releasing hormone in saline solution (n = 16) and were immediately placed in a warm (30°C; n = 14) or cool (15°C; n = 14) ambient temperature. In vivo measurements of temperature control (colonic temperature, oxygen consumption, and incidence of shivering) were then performed during the first 6 hours after birth, in conjunction with plasma thyroid hormone measurements. Brown adipose tissue was then sampled for measurement of uncoupling protein 1 abundance. Results: Plasma triiodothyronine concentrations were significantly higher in lambs treated with thyrotropin-releasing hormone than in control lambs between 3 and 6 hours after birth, as were plasma thyroxine concentrations 1 and 5 hours after birth. Delivery temperature had no effect on plasma thyroid hormone concentrations. At 6 hours after birth the abundance of uncoupling protein 1 was higher in lambs treated with thyrotropin-releasing hormone than in control lambs. Lambs treated with thyrotropin-releasing hormone exhibited a lower incidence of shivering than did control lambs between 5 and 6 hours after birth, and an effect of ambient temperature on the incidence of shivering was observed only in the control group. From 3 to 6 hours after birth colonic temperature was significantly higher in cool-delivered lambs treated with thyrotropin-releasing hormone than in the control group. Oxygen consumption was higher in cool-delivered lambs than warm-delivered lambs, but this was not influenced by thyrotropin-releasing hormone. Irrespective of delivery temperature, lambs treated with thyrotropin-releasing hormone possessed more pericardial adipose tissue and hepatic glycogen than did control lambs. Conclusion: Thyrotropin-releasing hormone treatment stimulated thyroid hormone secretion in the neonatal lamb and improved thermoregulation during the first 6 hours after birth in near-term lambs delivered by cesarean under cool conditions. Skeletal and heart muscle protein turnover during long-term exposure to high environmental temperatures in young rats

A study was undertaken to determine the long-term effects of a hot environment on protein turnover in skeletal and cardiac muscles of young homeothermic animals. Three groups of 36 male 28 day old rats were housed at 35 degrees C (hot group), 25 degrees C (control group), or 25 degrees C but pair-fed to the intake of the hot group (pair-fed group). Rates of protein synthesis and degradation were measured in vivo on days 5, 10, 15, and 20. By day 20, soleus and gastrocnemius (skeletal muscle) protein masses were 7 and 14% lower in the hot group and 31 and 21% lower in the pair-fed group compared with the control group (P < 0.05). The fractional rate of protein synthesis (k(syn)) was on average 11% lower (P < 0.05) in the hot group compared with control rats and was not different from pair-fed rats. The fractional rate of skeletal muscle protein degradation (k(deg)) in hot rats was slightly lower than in control rats; k(deg) was on average 18% higher (P < 0.05) in the pair-fed group compared with the hot group and this difference appeared to be most prominent on day 5. In heart, by day 20, protein mass was 30% lower in the hot group and 40% lower in the pair-fed group compared with control rats (P < 0.05). k(syn) was on average 19% lower (P < 0.05) in the hot group compared with the control group, but not different from pair-fed rats. In the heart there were no differences in k(deg) among treatments. Plasma triiodothyronine (T3) concentration was lower in the hot group, but not in the pair-fed group, compared with controls. In conclusion, chronic exposure to hot environments was associated with lower skeletal and cardiac muscle mass and protein turnover; lower protein mass in this tissue was due to decreased k(syn); this is consistent with lower plasma T3 concentrations. In pair-fed rats, k(syn) was also reduced, but interestingly k(deg) was not, resulting in a greater loss of skeletal muscle mass. These results suggest that heat exposure invokes physiological adaptations to preserve skeletal muscle mass despite decreased food intake. In the heart, loss of protein was a result of decreased k(syn), which can be primarily ascribed to lower food intake.

Glucose-insulin relationships and thyroid status of cockerels selected for high or low residual food consumption.

The plasma glucose-insulin relationships and thyroid status were investigated in two lines of adult cockerels divergently selected for high (R+) or low (R-) residual food consumption (RFC). For a given body weight, R+ birds had a 74% higher food intake than R- birds. Plasma glucose concentrations were significantly lower in the R+ line compared with the R- when fasted, whereas R+ birds exhibited a significantly lower plasma insulin concentration than R- birds either in fed or fasted state. After an overnight fast, R+ birds also exhibited a higher sensitivity to exogenous insulin in view of its more pronounced hypoglycaemic effect. After an oral glucose load, the glucose disposal of R+ cockerels was faster despite lower glucose-induced plasma insulin concentration. Whilst plasma triacylglycerol concentrations were lower in the R+ line when fed, plasma non-esterified fatty acid concentrations were higher in fasted R+ than R- cockerels (684 v. 522 mumol/l). Higher plasma triiodothyronine concentrations were observed in fed R+ compared with R- birds (3.0 v. 2.1 nmol/l respectively). The higher plasma concentrations of triiodothyronine associated with lower concentrations of insulin could account for the leanness and the elevated diet-induced thermogenesis previously observed in the R+ line.

Birth weight responses to shearing ewes in early to mid gestation

AbstractThis study compared the effect on foetal growth of shearing ewes in very early pregnancy (day 50) with the effect of shearing later in pregnancy (day 70 or 100). One hundred and sixty ewes were allocated at day 49 of pregnancy to four ‘time of shearing’ treatments — pregnancy day 50 (P50), P70, P100 (no. = 45 ewes per treatment) and unshorn (shearing of this group occurred 43 days after weaning) (no. = 25); two methods of shearing (by standard comb and cover comb); and two levels (1 or 2) of pregnancy/rearing rank. All ewes had been mated over a 28-day period and grazed pasture throughout the trial. The mid point of lambing was 27 August 1996. Live weights of ewes (corrected for fleece weight) were not influenced by shearing time at any stage except at day 45 of lactation when ewes shorn at P50, P70 or P100 were about 3⋅0 kg lighter than unshorn ewes. Shearing method had no effect on ewe live weight. Time of shearing and shearing method had no effect on ewe greasy fleece production or wool tensile strength. Shearing significantly (P &lt; 0⋅05) influenced the birth weight of single- (by 0⋅7 to 0⋅8 kg) but not twin-born lambs. Maternal plasma triiodothyronine concentration increased proportionately by 0⋅47 to 0⋅85 (P &lt; 0⋅05) and lasted for at least 20 days in response to shearing irrespective of time of shearing. No difference in plasma insulin concentration was detected between shearing groups but plasma glucose concentration proportionately increased by 0⋅06 to 0⋅16 (P &lt; 0⋅05) for 10 to 20 days after shearing. These results indicate that shearing between day 50 and 100 of pregnancy can increase lamb birth weights and that the response is associated with an increase in thyroid hormone concentrations in the maternal circulation.

Skeletal and cardiac muscle protein turnover during cold acclimation in young rats.

The effect of long-term cold exposure on skeletal and cardiac muscle protein turnover was investigated in young growing animals. Two groups of 36 male 28-day-old rats were maintained at either 5 degrees C (cold) or 25 degrees C (control). Rates of protein synthesis and degradation were measured in vivo on days 5, 10, 15, and 20. Protein mass by day 20 was approximately 28% lower in skeletal muscle (gastrocnemius and soleus) and approximately 24% higher in heart in cold compared with control rats (P < 0.05). In skeletal muscle, the fractional rates of protein synthesis (k(syn)) and degradation (k(deg)) were not significantly different between cold and control rats, although k(syn) was lower (approximately -26%) in cold rats on day 5; consequent to the lower protein mass, the absolute rates of protein synthesis (approximately -21%; P < 0. 05) and degradation (approximately -13%; P < 0.1) were lower in cold compared with control rats. In heart, overall, k(syn) (approximately +12%; P < 0.1) and k(deg) (approximately +22%; P < 0.05) were higher in cold compared with control rats; consequently, the absolute rates of synthesis (approximately +44%) and degradation (approximately +54%) were higher in cold compared with control rats (P < 0.05). Plasma triiodothyronine concentration was higher (P < 0.05) in cold compared with control rats. These data indicate that long-term cold acclimation in skeletal muscle is associated with the establishment of a new homeostasis in protein turnover with decreased protein mass and normal fractional rates of protein turnover. In heart, unlike skeletal muscle, rates of protein turnover did not appear to immediately return to normal as increased rates of protein turnover were observed beyond day 5. These data also indicate that increased rates of protein turnover in skeletal muscle are unlikely to contribute to increased metabolic heat production during cold acclimation.

The Influence of Acute Hypoxia and Carotid Body Denervation on Thermoregulation During Non‐Rapid Eye Movement Sleep in the Developing Lamb

We investigated the influence of ambient temperature on the thermoregulatory response to hypoxia in developing lambs before (at 4 and 14 days of age) and after (17 and 30 days of age) carotid body denervation (CBD). Lambs were studied during non-rapid eye movement sleep at thermoneutral (23-15 C) and cool (10-5 C) ambient temperatures, during normoxia and acute hypoxia (inspired oxygen content of 13 %). Measurements of oxygen consumption, arterial partial pressures of O2 and CO2, colonic temperature, incidence of shivering and plasma concentrations of thyroid hormones, cortisol, insulin and glucose were made under each condition. Oxygen consumption was higher at cool compared with thermoneutral ambient temperatures and decreased during hypoxia during cooling at all stages. At 4 days of age, only one lamb shivered during cooling in normoxia, but 4 out of 12 lambs shivered during hypoxia and colonic temperature fell, significantly, by 0.2 C. At 14 days, 8 out of 12 lambs shivered during cooling, of which 6 continued to shiver during hypoxia but colonic temperature did not change significantly. Plasma triiodothyronine concentrations increased on cooling at 4 and 14 days, an affect that was inhibited by hypoxia at 4, but not 14 days of age. At 17 days of age, i.e. post-CBD, plasma thyroid hormone concentrations and oxygen consumption were lower during cold exposure compared with intact lambs at 14 days of age. In CBD lambs, imposing further hypoxia resulted in colonic temperature falling 0. 6 C during cooling, with only 2 out of 10 lambs shivering. Plasma glucose and insulin, but not cortisol, concentrations decreased during hypoxia, irrespective of age or CBD. It is concluded that hypoxia has an important influence on metabolism and thermoregulation, which is modulated by age and environmental conditions. Compromised carotid body function, in lambs older than 2 weeks of age, can result in severe hypoxia and thermoregulatory dysfunction even with modest environmental cooling.

Effect of early-age thermal conditioning and food restriction on performance and thermotolerance of male broiler chickens

1.The effects of early-age thermal conditioning and food restriction on performance and thermotolerance were studied in male broiler chickens, in 2 trials. 2. Chickens were exposed to 36degree+/-1degreeC and 70% to 80% relative humidity (RH) for 24 h at the age of 5 d (thermal conditioning, TC), or to food restriction (FR) at the age of 7 to 14 d, or to both treatments (TC+FR), while a control group was reared under standard conditions. At the age of 42 d, chickens were thermally challenged by a heat stress of 35degree+/-1degreeC and 20% to 30% RH for 6 h. 3. In both experiments, weight gain of the TC chickens between the ages of 7 and 42 d was significantly higher than those of other treatments and was associated with higher food intake. 4. Early-age TC significantly increased body temperature (T ). Thermal challenge at the age of 42 d markedly increased T in all groups but that of the TC groups was the lowest. b 5. Mortality during thermal challenge was significantly lower in the treated chickens, except for the FR group in trial 2. 6. Plasma triiodothyronine (Tb) concentration was greatly depressed in all treatment groups during the 3 thermal challenge. However, the lowest concentration was observed in the TC group, suggesting that these chickens exhibit the lowest rate of heat production under such conditions. 7. Thermal conditioning reduced the increase of haematocrit with age, whereas food restriction resulted in an increase in haematocrit immediately after FR. Thermal challenge resulted in a haematocrit decline in all groups, with the lowest values in the TC and TC+FR chickens. 8. It can be concluded that, because the TC treatment improved thermotolerance (possibly by reducing heat production) and performance, it has advantages over the FR and TC+FR treatments.

Mink Dam Weight Changes During the Lactation Period: II. Energy Consumption and Plasma Concentrations of Thyroid Hormones and Insulin

Consumption of metabolizable energy (ME) and plasma concentrations of triiodothyronine (T3) and thyroxine (T4) were studied in lactating scanblack mink in the period from 1991 to 1994. In 1994, the plasma concentrations of insulin were also investigated. The objectives of the study were to describe the daily ME consumption and the plasma concentrations of T3, T4 and insulin during lactation and to evaluate the factors that influence ME consumption, and the concentrations of T3, T4 and insulin. The factors included were dam age, dam body weight, litter size and litter weight. The daily consumption was recorded from 72 dams in 85 lactations. Blood samples were collected at weekly intervals. Dam body weights were recorded before blood sampling and at days 14, 28, and 42 post partum. Kit body weights were recorded at parturition, and at days 14, 28, and 42 post partum. Between the first and the fourth weeks post partum, there was a steady increase in daily ME consumption from 1095 kJ to 1796 kJ. During the fifth week of lactation, the concentrations of T3 and T4 and insulin were low. In adult dams, ME consumption and concentration of T3 were lower than in yearlings. Litter weight had a greater influence on dam ME consumption than the number of kits. The results imply that in the lactation period a heavy demand is put on the mink dam. Despite the increase in ME consumption during lactation, the body weight loss of the dams indicates that they mobilize body reserves, especially in the final part of lactation. During the fourth week of lactation, the dams reach an upper limit for feed consumption. Insufficient feed consumption, and the increased milk production were associated with low concentrations of T3, T4 and insulin.

Effect of propylthiouracil-induced hypothyroidism on thyroid hormone profiles and tissue deiodinase activity in cashmere goats

The relationships between dose of propylthiouracil (PTU), circulating plasma thyroid hormone concentrations and 5′-monodeiodinase enzyme activity were investigated to determine the dose and duration of PTU treatment required to suppress plasma thyroid hormone levels in cashmere goats. Six groups of four cashmere goats were orally dosed for 2 months with between 1.1 and 35 mg/kg/bw per day of PTU. Mean thyroxine (T4) concentrations in goats treated with 1.1 mg/kg of PTU did not differ significantly from the control animals at any time during the study. However, in goats treated with 2.2 to 35 mg/kg/day of PTU, T4 concentrations declined in a dose dependent manner and plateaued after approximately 1 month of treatment. Mean plasma triiodothyronine (T3) concentrations were not significantly affected by any of the PTU doses. Type I 5′-monodeiodinase (type I 5′-MDI) activity in liver and kidney in the 1.1 mg/kg group was suppressed to 25 and 37%, respectively, of values recorded in control animals (24.3 vs. 98.4 for liver, and 15.7 vs. 42.4 pmol iodine released/h/mg protein for kidney), and was below assay sensitivity at doses of PTU>4 mg/kg. Type II 5′-monodeiodinase activity in goat skin was detected at approximately 11.5 fmol iodine released/h/mg/protein and was not apparently affected by PTU treatment. It is concluded that a dose≤1 mg/kg/bw PTU can be used in long-term studies in goats, to inhibit extrathyroidal conversion of T4 to T3 via type I 5′-MDI without influencing circulating thyroid hormone profiles.

Control of hepatic insulin-like growth factor II gene expression by thyroid hormones in fetal sheep near term.

The effects of thyroid hormones on hepatic insulin-like growth factor (IGF) II gene expression and their interaction with cortisol in the ontogenic control of this gene were investigated in fetal sheep during late gestation (term 145 +/- 2 days) and after experimental manipulation of fetal plasma hormone concentrations. In intact fetuses, a significant decrease in hepatic IGF-II mRNA abundance was observed between 127-130 and 142-145 days of gestation, which coincided with the normal prepartum rise in plasma cortisol and triiodothyronine (T3) concentrations. This ontogenic decline in hepatic IGF-II gene expression was abolished in fetuses in which the prepartum rise in plasma T3, but not cortisol, was prevented by fetal thyroidectomy. At 127-130 days, downregulation of hepatic IGF-II mRNA abundance was induced prematurely in intact fetuses by an infusion of cortisol for 5 days (2-3 mg . kg-1 . day-1 iv). Plasma concentrations of cortisol and T3 in the cortisol-infused intact fetuses were increased to values seen close to term. Similar findings were observed in thyroidectomized fetuses, in which, despite thyroidectomy, cortisol infusion significantly increased plasma T3 concentrations and caused a premature decrease in hepatic IGF-II mRNA levels. However, in intact fetuses at 127-130 days, the increasing of T3 concentrations alone by exogenous T3 infusion (8-12 microg . kg-1 . day-1 iv for 5 days) had no effect on hepatic IGF-II mRNA levels. Overall, a decrease in hepatic IGF-II mRNA abundance was only observed in fetuses in which there were concurrent increases in plasma cortisol and T3 concentrations. When observations from all fetuses were considered, irrespective of gestational age or treatment, hepatic IGF-II mRNA levels were negatively correlated with plasma cortisol and T3 but not thyroxine concentrations. Partial correlation analysis of hepatic IGF-II, cortisol, and T3 values showed that the plasma concentration of cortisol in the fetus had the predominant effect on hepatic IGF-II mRNA abundance. These findings show that T3 may mediate, in part, the maturational effects of cortisol on hepatic IGF-II gene expression but that it is ineffective without a concomitant rise in fetal plasma cortisol. Hence, increased concentrations of both cortisol and T3 appear necessary to induce downregulation of hepatic IGF-II mRNA abundance in fetal sheep close to term.

Lowering of plasma triiodothyronine level and sympathetic activity does not alter hypoalbuminaemia in rats fed on a diet of low protein concentration.

Previous studies have described high plasma triiodothyronine (T3) concentrations and sympathetic activity in rats fed on low-protein diets. The present investigation examined how the nutritional status of rats fed on a low-protein diet was affected when these hormonal changes were reduced by drug administration. The low-protein diet (LP group) prevented growth, reduced plasma albumin levels, elevated plasma T3 concentration, and increased both the weight of the interscapular brown adipose tissue (BAT) and the activity of BAT cytochrome c oxidase (EC 1.9.3.1). Lowering the plasma T3 concentration (with carbimazole; CA group) elevated the plasma insulin concentration, promoted a small increase in the plasma albumin concentration and caused weight gain in comparison with the LP group. Reduction of sympathetic activity (with alpha-methyl-p-tyrosine; MT group) promoted a small elevation in plasma albumin concentration accompanied by a diminished T3 concentration, BAT weight, and an increase in fat deposition in relation to LP rats. In a second experiment, simultaneous lowering of the plasma T3 concentration and sympathetic activity (CA/MT group) resulted in weight gain associated with elevated plasma insulin concentration and fat deposition and a marked reduction in BAT cytochrome c oxidase activity. However no change in the hypoalbuminaemia was observed. The results of the present study suggest that in spite of the previously described increase in metabolic rate in fed on a diet with low-protein concentration when compared with controls, the mechanisms involved in the control of BAT activity and fat deposition seem to be independent of those which cause liver protein depletion and hypoalbuminaemia.

Maternal nutrition in early-mid gestation and placental size in sheep.

We investigated the influence of restricted maternal nutrition between 30 and 80 d gestation on placental growth. Singleton-bearing ewes were fed on either 0.6 (i.e. nutrient restricted) times their energy requirements of 2.25 times this amount (i.e. controls) up to 80 d gestation, when their placentas and fetuses were sampled and analysed. Nutrient-restricted ewes lost body condition score but not body weight and had lower plasma thyroid hormone concentrations than controls, but there were no differences in plasma glucose, non-esterified fatty acids or 3-hydroxybutyrate concentrations between groups. There was no effect of maternal nutrient restriction on fetal weight, conformation or organ weight with the exception of brain weight which was lower nutrient-restricted ewes. Nutrient restriction had no effect on total placental weight, or proportion of inverted placentomes, but was associated with an increased abundance of small placentomes and decreased weight of the fetal but not maternal components of the placenta. Fetal cotyledons form nutrient-restricted ewes also had a lower DNA but higher haemoglobin concentration than those sampled from controls. The plasma concentration of triiodothyronine in umbilical cord plasma was also increased in fetuses from nutrient-restricted ewes. In conclusion, maternal nutrient restriction during early-mid gestation is associated with a smaller placenta.

Energy balance of laying hens selected on residual food consumption

1. Energy balance of adult hens divergently selected for high (R + ) or low (R- ) residual food consumption was investigated using indirect calorimetry. Three experiments were conducted: feeding behaviour of individual hens, hens having free access to food or fasting and hens tube-fed at 70, 100 or 130% of the control intake of both lines. 2. R + hens ate significantly more than R- (+ 48%). This difference was maximum at the onset of the light (+ 120%) and not significant during the rest of the light period. Although both lines spent the same total time eating, R + hens exhibited more frequent but shorter meals than R- ones, suggesting a higher feeding activity in R + hens. 3. True metabolisable energy (TME) intake was 28% greater in R+ than in R- birds. Basal heat production did not differ significantly between genotypes. Heat increment of feeding (HI) or diet-induced thermogenesis was significantly enhanced in R + birds: + 23.9 vs 13.7 kJ/100 kJ TME intake in R- . 4. When tube-fed and placed in darkness, restricted-fed R + hens had the same HI as R- birds. When fed at 100% and 130% of control intake, R + hens demonstrated a regulatory thermogenesis, which allowed them to dissipate the excess of energy ingested compared to R- hens. 5. No significant modification was observed in plasma triglyceride, phospholipid, uric acid, glucose or insulin concentrations between lines, suggestive of the use of similar thermogenic pathways. While plasma triiodothyronine (T3) concentrations did not differ between genotypes, plasma thyroxine (T4) appeared higher in unfed R + birds and equal in fed conditions in both lines. 6. A regulatory part of HI allowed R + hens to get rid of their excessive energy intake. An increased turnover of lipids could be involved.
