A nationwide survey on the endosulfan residues in Chinese cotton field soil: Occurrence, trend, and ecological risk

The nationwide occurrence of endosulfan residues in cotton fields has not yet been investigated. Therefore, in this study, 202 surface soil samples from 27 cities were collected from cotton fields in 8 major cotton-planting provinces of China, covering more than 97% of the national cotton sown area. The results showed that endosulfan residues were detected in cotton fields throughout the country. The main type of residue found was endosulfan sulfate (ES-sulfate), followed by β-endosulfan and α-endosulfan, with average concentrations of 0.475, 0.129, and 0.048 μg/kg, respectively. Significant spatial variations in the endosulfan residues was noted, and the highest concentration of endosulfan residues was observed in the northwest inland cotton-growing area, followed by that in the Yellow River basin and Yangtze River basin cotton-growing areas. The endosulfan residues showed significant positive correlations with soil organic matter and soil clay contents. The α/β endosulfan ratio was determined to be in the range of 0.02–1.20, indicating that endosulfan residues originated from the endosulfan application performed in historical cotton cultivation efforts. Together with the literature data, the concentrations of α-endosulfan and β-endosulfan residues peaked in 2015 and 2017, respectively, and showed an overall decreasing trend from 2002 to 2021. The results of the ecological risk assessment suggested that Folsomia candida was most sensitive to endosulfan residues, with 20.8% of the sites presenting a high risk. However, in general, the soil ecological risk of cotton fields across the country was low. Our study demonstrated that China has achieved promising results in controlling the use and pollution of endosulfan, especially after 2014.

Determination of Pesticide Residue in Brinjal Sample Using HPTLC and Developing a Cost-Effective Method Alternative to HPLC

Two analytical techniques HPLC (high performance liquid chromatography) and HPTLC (high performance thin layer chromatography) were validated to reveal the quality and quantity of pesticide residues (organophosphorus, organochlorine, and pyrethroids) in brinjal samples collected from a local market of Faisalabad. The HPTLC methods showed linear behavior for standard samples and residue was in the range of 1–130 ng. The organochlorine (α-endosulfan) contaminates the samples at 4, 5, 9, and 10 weeks, and detected quantity was less than MRL (minimum residue level) of the FAO (Food and Agriculture Organization of United Nations), i.e., 0.5 mg·kg−1. The organophosphorus pesticide (chlorpyriphos, methamidophos, monocrotophos, dichlorvos, carbosulfan, profenophos, and dimethoate) residue contaminated the samples and violated the MRL limit. Pyrethroids (deltamethrin, β-cyhalothrin, and cypermethrin) were present at appreciable levels, in samples of 1, 3, 4, 6, 8, and 9 weeks. The concentration of β-cyhalothrin (0.25 mg·kg−1) and cypermethrin (0.205 mg·kg−1) was significantly higher than that of all detected pesticides. The carbosulfan and deltamethrin contaminated all 10-week samples. The HPLC analysis of samples was carried out to confirm the efficiency of HPTLC as cost-effective method. The concentration of α-endosulfan, chlorpyriphos, dimethoate, monocrotophos, profenophos, deltamethrin, and cypermethrin in brinjal samples through the HPTLC method showed similar residual concentration with HPLC analysis.

ORGANOCHLORINE PESTICIDE RESIDUES IN AGRICULTURAL SOILS FROM RÉGION DES PLATEAUX IN TOGO

Region des Plateaux in Togo is the highest producer of cash crops (coffee, cocoa, cotton) as well as vegetables. This region is the area with the supreme use of chemical pesticides to enhance agricultural production in Togo (west Africa). As effective as these chemicals may be in achieving this goal, the incidence of their residues on the environment, together with the associated adverse effects, have been of serious concerns. The objective of this study is to assess soils contamination by organochlorine pesticides in vegetable production areas within this region. Fifty-four soil samples were randomly collected from selected farmers’ plots within six villages. The analytical methods included solvent extraction of pesticide residues and their subsequent quantification using gas chromatography – electron capture detector (GC-ECD). Twenty one different organochlorine pesticides were analyzed. The analytical results showed that only aldrin, endosulfan (α-, β- and sulfate), and hexachlorocyclohexanes (HCH) isomers, including α-HCH, β-HCH, δ-HCH and γ-HCH were detected in various concentrations. The residuals of aldrin varied from detection limit (DL) (< 0.001 μg/kg) to 0.93 ± 0.03 μg/kg, the concentrations of α-endosulfan from DL to 1.88 ± 0.41 μg/kg, β-endosulfan from DL to 0.4 ± 0.01 μg/kg and endosulfan sulfate from DL to 3.87 ± 0.19 μg/kg. On the other hand the concentration of each isomer of HCH ranged from DL to 0.89 ± 0.07 μg/kg (α-HCH), 14.6 ± 0.12 μg/kg (β-HCH), 4.27 ± 0.61 μg/kg (γ-HCH) and 2.92 ± 0.05 μg/kg (δ-HCH). The pesticide residue concentrations found in the studied soils were highly below maximum residue limits varying from 100 to 8000 μg/kg set by Food and Agriculture Organization of the United Nations (FAO). The detected pesticide residues reflected the type of pesticides that have been used in the study areas. The differences in the concentrations of these pesticide residues in the soil samples are likely the result of substantial inter-farm variation in intensive use of pesticide as well as the variation in their degradation dynamics. The findings of this study provide information on residue levels of organochlorine pesticides in soils from “Region des Plateaux” in Togo. Routine monitoring of pesticide residues in these study areas is necessary for the prevention, control and reduction of environmental pollution, leading to minimize health risks.

Distribution of OCPs and PCBs in Mussels (Mytilus galloprovincialis) from the Marmara Sea Coastal Sites

Mussel samples were collected monthly between October-2010 and October-2011 from four stations (Bosphorus, Bandırma, Gelibolu, Tekirdağ) in the Marmara Sea. Two consecutive months' samples were homogenized and combined as a single group for analysis. Mussel samples were analyzed for Organochlorine pesticides (OCPs); (total-DDT, total-HCH, Endrin, α-Endosulfan, β-Endosulfan, Heptachlor) and Polychlorinated biphenyls (PCBs); (PCB 28, PCB 52, PCB 138, PCB 153, and PCB 180). All analyses were done according to Eurofins house method in ERGO Laboratory in Germany. Concentrations of α-endosulfan and heptachlor in mussel tissues were below method detection limits. The annual average OCPs concentrations among the stations ranged between 0.02 and 1.45ng/g (wet weight), 1.9-99.75ng/g (lipid weight) whereas the annual average PCBs concentrations among the stations ranged between 0.03 and 0.40ng/g (wet weight), 1.71-26.48ng/g (lipid weight), respectively. There was no relation between fat content of mussels and residues of the contaminants. PCB 138 and PCB 153 were the most predominant PCBs, while total-DDT and total-HCH were the most predominant OCPs in the mussels. Total-DDT concentrations were higher compared to total-HCH and PCBs isomers. Measured levels were below the national and international committees' and institutions' limits for human consumption and protection of aquatic biota.

Effects of vitamin C on pathology and caspase-3 activity of kidneys with subacute endosulfan toxicity

Endosulfan is an insecticide that is composed of two stereoisomers: α- and β- endosulfan in an approximate ratio of 70:30. Owing to its widespread use, poisoning of both humans and animals is possible. We examined the toxic effects of endosulfan on New Zealand white rabbit kidneys. Rabbit kidneys were examined histopathologically and caspase-3 activity was detected using immunohistochemistry. Animals were divided into four groups: Group 1 was given a sublethal dose of endosulfan in corn oil by oral gavage daily for 6 weeks, Group 2 was given endosulfan + vitamin C during the same period, Group 3 was given corn oil daily and vitamin C on alternate days, Group 4 was given only corn oil daily throughout the experiment. By the end of experimental period, the concentration of α-endosulfan was greater than the β-endosulfan concentration in the kidneys of both of endosulfan treated groups (Groups 1 and 2). Decreased accumulation of α- and β-endosulfan was observed in Group 2, possibly because of the antioxidant effect of the vitamin C. Histopathological examination revealed hemorrhages, tubule cell necrosis, glomerular infiltration, glomerulosclerosis and proteinaceous material in the tubules, and Bowman spaces in the kidneys of Group 1. Caspase-3 reaction was stronger in Group 1 than in the other groups. Apoptotic activity was most frequent in proximal tubule cells. Endosulfan is toxic to rabbit kidneys. Vitamin C treatment reduced the accumulation of endosulfan in kidneys and reduced its toxicity.

Determination of hormonally active chlorinated chemicals in waters at sub µg/L level using stir bar sorptive extraction-liquid desorption followed by negative chemical ionization-gas chromatography triple quadrupole mass spectrometry

A new analytical method has been developed for the screening, evaluation and determination of sub µg/L concentrations of chlorinated chemicals in environmental waters. Ten different compounds included in the European water policy have been considered and determined by stir bar sorptive extraction (SBSE) followed by liquid desorption and negative chemical ionization gas chromatography coupled to triple quadrupole mass spectrometry (SBSE-LD-NCI-GC-QqQ-MS). Analyte extraction was performed using SBSE with 100 mL sample volume, 5% MeOH, 10% NaCl and an extraction time of 14 h of mechanized stirring, which provided best analytical recoveries. Desorption of analytes was carried out using 5 mL acetonitrile (ACN) and 5 min of two successive sonication steps which resulted best performance without any carry over. The method detection limits (MDL) ranged 3–22 ng/L and 7–29 ng/L for river waters and wastewaters respectively. The experimental recoveries for all compounds were >60% with <20% deviation in the reproducibility. Pentachloroanisole alone reported a low maximum recovery of 54% under given conditions. Matrix influenced signal enhancement was observed more in case of triclosan, which was managed by using matrix matched calibration standards apart from using double helix injection liner. This method was applied to the analysis of different river waters and wastewater effluent samples collected from Alcala de Henares, Spain. Four compounds namely chloropyriphos-ethyl, triclosan, α-endosulfan and endosulfan sulphate were positively identified in river waters and three compounds viz., chloropyriphos-ethyl, α-endosulfan and endosulfan sulphate were detected in wastewater effluents. The concentrations of α-endosulfan ranged between 11.3–22.9 ng/L and 16.3–33.8 ng/L in river waters and wastewaters respectively. Almost 40% of the water samples analysed showed much higher concentrations of α-endosulfan than prescribed European environmental quality standard limit for surface waters of 5 ng/L.

The concentration and distribution of organochlorine pesticides in the air from the karst cave, South China

Organochlorine pesticides (OCPs) in the air of Dayan Cave in Guilin were analyzed, to investigate the source and contamination levels. Generally, air outside of the cave had much higher concentrations than inside air, and both outside and inside air showed much lower contamination levels than those observed in other regions in China and abroad. The ratios of α-/γ-HCHs and the percentages of β-(or δ-)HCH implied a residue of historical local technical HCH contamination. The DDT concentrations, o,p'-DDT/p,p'-DDT, and p,p'-DDE/p,p'-DDT suggested fresh DDT-containing products acted as the primary source of DDT for the air in Dayan Cave. An intermediate level concentration of α-endosulfan (156.00pg/m(3)) was observed in the outside air, implying local source was dominant. Based on the similar sources of HCH and DDT and the relationships between outside and inside air, it appeared that outside air pollutants affected on inside HCH and DDT's distribution. For other OCPs, outside air might be major contributor to inside air contamination under conditions of high concentrations in outside air compared with low levels in inside air for semi-closed karstic caves.

Effect of vitamin C in reducing the toxicity of endosulfan in liver in rabbits

In this study, the effect of endosulfan, an organochlorine pesticide, and the ameliorating effect of vitamin C on the livers of New Zealand white rabbits were studied. Livers of the rabbits were examined grossly and histopathologically, and caspase-3 activity was detected by immunohistochemical methods. A total of twenty-four rabbits were divided into four groups ( n=6). Rabbits in Group I (END) were daily given a sublethal dose of endosulfan (1 mg/kg bw) in corn oil by oral gavage for 6 weeks. Group II (END+C) received the same dose of endosulfan and additionally Vit C (20 mg/kg bw) every other day during this period. Group III (OIL+C) received corn oil daily by oral gavage and vitamin C every other day for 6 weeks. Group IV (OIL), the control group, received only corn oil daily, by oral gavage throughout the experiment. The concentration of α-endosulfan in the END group was higher in livers (0.102±0.012 ppb) than the β-endosulfan (0.072±0.001 ppb). Decreased accumulation of α and β endosulfan was observed in the END+C group (0.025±0.003 and 0.016±0.002 ppb, respectively) ( p<0.0001). The most prominent gross findings at the necropsy were seen in the END group, in which swollen and pale livers were commonly observed. Hemorrhages, degenerations, necrosis, and in some rabbits bile duct hyperplasia were the marked histopathological findings of the END group. Caspase-3 positive reaction was more severe in this group than in the others. An ameliorating effect of Vit C on gross, histopathological and immunohistochemical findings was observed in the END+C group. The results revealed that endosulfan is highly toxic for rabbit livers. However, toxicity was decreased by Vit C treatment, which reduced the accumulation of endosulfan in livers four-fold.

Disappearance of endosulfan residues from seawater and sediment under laboratory conditions

The dissipation rate of endosulfan isomers (α and β) in seawater and sediment was studied. The disappearance rate of both isomers from seawater and pure water was compared, and the same measurements were made in both sterile and unsterile marine sediment. Flasks of water and sediment, fortified with a dispersion of a commercial endosulfan 350 g litre−1 EC, Protodan 35®, were incubated under laboratory light at room temperature for 82 days. A micro on-line extraction method and GC-ECD was used to determine the pesticide and its metabolites. The dissipation of endosulfan (in two phases of first-order kinetics) occurred more rapidly in seawater than in pure water. At the end of the experiment, the concentration of α-endosulfan in sterile sediment was four times greater than in unsterile sediment, while the dissipation rate of β-endosulfan in unsterile sediment was approximately double that observed in sterile sediment. The dissipation of both forms in sediment occurred in a single stage. Endosulfan β-isomer was more persistent than α-isomer in both sterile and unsterile sediment. Dissipation of endosulfan sum of α- and β-isomers in sediment at the end of the experiment ranged from 80% (sterile) to 95% (unsterile). Endosulfan sulfate was detected in water and sediment as the main metabolite. © 2000 Society of Chemical Industry
