Sensitivity of Eucalyptus globulus to red and blue light with different combinations and their influence on its efficacy for contaminated soil phytoremediation

The influence of combined red and blue light on the capacity of Eucalyptus globulus to phytoremediate a metal-polluted soil was evaluated in this study. Five combinations of blue and red light (0%, 10%, 25%, 50% and 100% blue) at the same intensity were used to treat E. globulus, and its biomass generation, metal uptake and water absorption in phytoremediation under different light treatments were assessed. The plant produced significantly more biomass under blue light, regardless of the ratio, than under single red or white light. The highest biomass was generated under the light ratio of B10R90. In addition, light combination influenced the metal concentrations in different plant tissues. The highest concentrations of Cd and Cu in roots appeared under the light ratio of B0. All metals in plant shoots achieved their highest concentrations under the light ratio of B100, except Pb. Comparing with control, red and blue light combined in varying proportions increased the efficiency remove Cd, Pb and Cu by 50.6–65.6, 71.1–88.7 and 28.9–70.6%, respectively,. The leachate volume under blue and red light combinations was 46.7–66.0% less than control with the combination of B10R90 mitigating the most metal loss. Light sources with different spectra combinations can enhance the phytoremediation efficiency of Eucalyptus globulus and alleviate leaching risk at the same time.

Responses of wheat to zinc sulfate fertilizer and plant growth‐promoting rhizobacteria under cadmium stress in soil

AbstractCadmium (Cd) is a major pollutant in soils as a result of extensive use of fertilizers, mining and industrial discharges. Zinc (Zn) and certain bacterial species have been known to alleviate Cd toxicity in plants. In this study, the individual and combined effects of the application of Zn and Pseudomonas species with the aim of reducing Cd stress in wheat cultivars were investigated. Plants (durum wheat and bread wheat) were exposed to different concentrations of Cd and Zn, and either P. putida or P. fluorescens in a growth chamber. Concentrations of Zn, Cd, chlorophyll (Chl), carotenoid, hydrogen peroxide (H2O2), and malondialdehyde (MDA), as well as antioxidant enzyme activities were assayed. The addition of Zn in soils reduced the toxicity of Cd in durum wheat more than in bread wheat even though there was more uptake of Zn in bread wheat. Analysis of variance showed that by using Zn fertilizer and Pseudomonas species the amounts of peroxidase (POD), polyphenoloxidase (PPO), MDA, and H2O2 were reduced at three growth stages. Surprisingly, with increasing Zn concentration, Cd concentration in plant tissue was slightly increased, which suggests that adding Zn to soil could facilitate Cd desorption from soil particles. Application of Pseudomonas and Zn could be a promising solution to reduce detrimental effects of Cd, especially in durum wheat.

Soil Enzyme Activity in Soils Subjected to Flooding and the Effect on Nitrogen and Phosphorus Uptake by Oilseed Rape.

Waterlogging presents one of the greatest constraints for agricultural crops. In order to elucidate the influences of waterlogging stress on the growth of oilseed rape, a pot experiment was performed investigating the impact of waterlogging on nitrogen (N) and phosphorus (P) accumulation in oilseed rape, and mineral N and available P profiles and enzyme activities of soils. The experiment included waterlogging treatments lasting 3 (I), 6 (II), and 9 (III) days, and a control treatment without waterlogging (CK). Results showed that waterlogging lasting 3 or more days significantly depressed the growth of oilseed rape, and prolonged the recovery time of plant growth with the period of flooding. Waterlogging notably influenced the N and P concentrations in plant tissues, and also affected mineral N, available P profiles, and activities of enzymes (including urease, phosphatase, invertase, and catalase) in the soils. With the exception of catalase, flooding suppressed the activity of urease, phosphatase, and invertase to varying degrees, and the longer the flooding time, the greater the suppression. The effect of waterlogging on mineral N and P profiles resulted from the altered proportions of NH4+-N and NO3--N, and the decreased available P concentrations in these soils, respectively. The effect on P was more significant than on N in both soil nutrient profile and plant utilization.

Selenium induces changes of rhizosphere bacterial characteristics and enzyme activities affecting chromium/selenium uptake by pak choi (Brassica campestris L. ssp. Chinensis Makino) in chromium contaminated soil

Understanding the chemical response and characteristics of bacterial communities in soil is critical to evaluate the effects of selenium (Se) supplement on plant growth and chromium (Cr)/Se uptake in Cr contaminated soil. The rhizosphere soil characteristics of pak choi (Brassica campestris L. ssp. Chinensis Makino) were investigated in soil contaminated with different levels and forms of Cr when supplemented with Se. Although inhibition of plant growth caused by Cr stress was not completely alleviated by Se, Cr content in plant tissues decreased in Cr(VI)120Se5 treatment (Cr(VI): 120 mg kg−1 soil; Se: 5 mg kg−1 soil) and its bioavailability in soil decreased in Cr(III)200Se5 (Cr(III): 200 mg kg−1 soil; Se: 5 mg kg−1 soil) treatment. Moreover, antagonism of Cr and Se on soil enzyme activities and bacterial communities were revealed. Notably, results of Cr(VI) reduction and Se metabolism functional profiles confirmed that bacterial communities play a critical role in regulating Cr/Se bioavailability. Additionally, the increases of Se bioavailability in Cr contaminated soil were ascribed to oxidation of Cr(VI) and reduction of Se reductases proportions, as well as the enhancing of pH in soil. These findings reveal that Se has the potential capacity to sustain the stability of microdomain in Cr contaminated soil.

Biochar alleviates fluoride toxicity and oxidative stress in safflower (Carthamus tinctorius L.) seedlings

An original research was laid out as factorial to evaluate the possible effects of biochar (0, 25 and 50 g kg−1 soil) on mitigating fluoride toxicity (non-contamination, 100, 200, 400 and 800 mg NaF kg−1 soil) in safflower seedlings. Increasing fluoride toxicity up to 200 mg NaF kg−1 soil did not decrease the safflower growth. However, the growth of plants under 400 and 800 mg NaF kg−1 was reduced by enhancing soluble fluoride concentration in the soil. This growth reduction was the consequence of an increase in fluoride content of plant tissues, generation of super oxide radicals and hydrogen peroxide, lipid peroxidation, misbalancing potassium and calcium ions, and a decrease in synthesis of photosynthetic pigments including chlorophylls, carotenoids, anthocyanin, flavonoids and xanthophyll's and photochemical efficiency of photosystem II. Application of biochar decreased the fluoride solubility, fluoride content of plant tissues, oxidative stress and antioxidant enzymes activities, leading to an increase in cation exchange capacity of soil and the pH, calcium and potassium uptakes, maximum efficiency of photosystem II, synthesis of photosynthetic pigments, and plant growth. In general, addition of 50 g biochar to 1 kg soil was the best treatment for alleviation of the fluoride toxicity in safflower plants.

Trace elements removal ability and antioxidant activity of Phragmites australis (from Algeria)

In this study, roots, stems and leaves of the worldwide distributed macrophyte Phragmites australis (common reed) were tested as potential removal and biomonitors of trace elements contamination in sediment. In particular, the concentrations (100, 200, and 500 mg/kg) of the following elements were analyzed: Zn, Cu, Pb, and Fe. Results showed that the amount of concentrations in plant tissues is significantly (p ≤ 0.01) dependent on the kind of organ and element. Trace element concentrations decreased according to the pattern of Fea (Rootsa > Stemsb > Leavesb) > Znb (Roota > Leavesb > Stemsc) > Cuc (Rootsa > Leavesb > Stemsc) > Pbc (Rootsa > Stemsb > Leavesc), as well as the roots acted as the main centers of bioaccumulation for all elements studied, and stems as the transit organs for translocation from roots to leaves. The major mechanisms employed by the plant were probably phytostabilization on the basis of the calculated Biological Concentration Factor (BCF – metal concentration ratio of plant root to soil); and Translocation Factor (TF – metal concentration ratio of plants roots to above ground part). Finally, due to the low scavenger effect of the radical DPPH, we excluded the hypothesis of the use of antioxidant mechanism in the tolerance of metals.

A distributed experiment demonstrates widespread sodium limitation in grassland food webs.

Sodium (Na) has a unique role in food webs as a nutrient primarily limiting for plant consumers, but not other trophic levels. Environmental Na levels vary with proximity to coasts, local geomorphology, climate, and with anthropogenic inputs (e.g., road salt). We tested two key predictions across 54 grasslands in North America: Na shortfall commonly limits herbivore abundance, and the magnitude of this limitation varies inversely with environmental Na supplies. We tested them with a distributed pulse experiment and evaluated the relative importance of Na limitation to other classic drivers of climate, macronutrient levels, and plant productivity. Herbivore abundance increased by 45% with Na addition. Moreover, the magnitude of increase on Na addition plots decreased with increasing levels of plant Na, indicating Na satiation at sites with high Na concentrations in plant tissue. Our results demonstrate that invertebrate primary consumers are often Na limited and track local Na availability, with implications for the geography of invertebrate abundance and herbivory.

Alleviation of arsenic toxicity by phosphate is associated with its regulation of detoxification, defense, and transport gene expression in barley

Arsenic (As) contamination in soils has posed a severe threat to safe crop production. The previous studies showed the antagonism between phosphorus (P) and As in plant growth and As uptake, while the mechanisms of alleviating As toxicity by P is not completely clear. Due to the limiting P condition, it is imperative to understand how low P addition can be used to suppress arsenate As (V) uptake and the subsequent mechanisms involved. Thus in this study we investigated the effect of P addition on As uptake, anti-oxidative enzyme activity, and anti-oxidant content, and the relative expression of transport, defense, and detoxification genes using two barley genotypes differing in As toxicity tolerance. P addition significantly reduced As concentration in plant tissues, and caused the great changes in activities of catalase and superoxide dismutase, glutathione content, and the relative expression of examined genes when the plants of the two barley genotypes were exposed to 100 μmol L−1 As, with ZDB160 (As-tolerant) being much more affected than ZDB475 (As-sensitive). The current results show that P addition can alleviate As toxicity by regulating the expression of As transport, defense, and detoxification genes to a greater extent in As tolerance of barley, suggesting the possibility of controlling As uptake and toxicity by applying low amount of P fertilizers in the As-contaminated soils.

Weekly Survivorship Curves of Soybean Aphid Biotypes 1 and 4 on Insecticidal Seed-Treated Soybean.

Thiamethoxam, an insecticide used in soybean seed treatments, effectively suppresses soybean aphids (Aphis glycines) Matsumura (Hemiptera: Aphididae) for a short time after planting. However, exactly when and how quickly soybean aphid populations could increase is unknown. Likewise, we lack data on virulent soybean aphid biotypes (that can overcome soybean resistance) when fed on seed-treated soybean. Determining the survival of soybean aphids over time on insecticidal seed-treated soybean is critical for improving soybean aphid management and may provide insights to manage aphid virulence to aphid resistant-soybean. In greenhouse and field experiments, aphid-susceptible soybean plants (with and without an insecticidal seed treatment) were infested at 7, 14, 21, 28, 35, and 42 days after planting (DAP). We compared aphid survival among biotypes 1 (avirulent) and 4 (virulent) and insecticide treatment 72 h after infestation. We also measured thiamethoxam concentrations in plant tissue using liquid chromatography-tandem mass spectrometry. As expected, soybean aphid survival was significantly lower on seed-treated soybean up to 35 DAP for both biotypes, which correlates with the decrease of thiamethoxam in the plant over time. Moreover, we found no significant difference between avirulent and virulent biotype survivorship on insecticidal seed-treated soybean plants, although we did find significantly greater survival for the virulent biotype compared with the avirulent biotype on untreated soybean in the field. In conclusion, our study further characterized the relative short duration of seed treatment effectiveness on soybean aphid and showed that survivorship of virulent aphids on seed-treated soybean is similar to avirulent aphids.

Growth, Seed Yield, Mineral Nutrients and Soil Properties of Sesame (Sesamum indicum L.) as Influenced by Biochar Addition on Upland Field Converted from Paddy

Sesame is an important oilseed crop cultivated worldwide. However, research has focused on biochar effects on grain crops and vegetable and there is still a scarcity of information of biochar addition on sesame. This study was to assess the effect of biochar addition on sesame performance, with a specific emphasis on growth, yield, leaf nutrient concentration, seed mineral nutrients, and soil physicochemical properties. A field experiment was conducted on an upland field converted from paddy at Tottori Prefecture, Japan. Rice husk biochar was added to sesame cropping at rates of 0 (F), 20 (F+20B), 50 (F+50B) and 100 (F+100B) t ha−1 and combined with NPK fertilization in a first cropping and a second cropping field in 2017. Biochar addition increased plant height, yield and the total number of seeds per plant more in the first cropping than in the second cropping. The F+50B significantly increased seed yield by 35.0% in the first cropping whereas the F+20B non-significantly increased seed yield by 25.1% in the second cropping. At increasing biochar rates, plant K significantly increased while decreasing Mg whereas N and crude protein, P and Ca were non-significantly higher compared to the control. Soil porosity and bulk density improved with biochar addition while pH, exchangeable K, total N, C/N ratio and CEC significantly increased with biochar, but the effect faded in the second cropping. Conversely exchangeable Mg and its plant tissue concentration decreased due to competitive ion effect of high K from the biochar. Biochar addition is effective for increasing nutrient availability especially K for sesame while improving soil physicochemical properties to increase seed yield, growth and seed mineral quality.

Effects of phosphorus in the wastewater from intensive catfish farming ponds on the growth and phosphorus uptake of <i> Hymenachne acutigluma </i> (Stued.)

Hymenachne aquatic grass, Hymenachne acutigluma was planted in the wastewater from intensive striped catfish (Pangasianodon hypophthalmus) cultivating ponds containing 2.1 mg N/L, which was enriched with a serious of inorganic phosphorus concentrations. The experiment was arranged in a completely randomized design with three replications in the net house for 42 days. The results showed that P concentrations did not significantly affect the growth of Hymenachne. The presence of high P concentrations resulted in the increase of P content in plant tissues leading to higher phosphorus absorption at the P levels of 8 and 10 mg P/L. H. acutigluma removed 12.1–27.6% P from 88.3–95.9% P in the wastewater of striped catfish pond. This result indicated the low concentrations of N (2.1 mg N/L) and of 1–10 mg P/L were not optimal for the growth of H. acutigluma.Hymenachne acutigluma, intensive striped catfish, wastewater, phosphorus, biomass, nutrient uptake.

Selenium Distribution and Translocation in Rice (Oryza sativa L.) under Different Naturally Seleniferous Soils

Selenium (Se) accumulation in plant foods may be providing dietary Se to minimize the health problems related to Se deficiency. In this study, rice plants were cultivated in different naturally seleniferous soils (0.5–1.5 mg Se kg−1). Se concentration in rice plant tissues was analysed, and the distribution and translocation of Se in rice were also studied. The effect of exogenous Se on yield and Se concentration in rice grain was also investigated by spraying Na2SeO3 (15 mg L−1, 15 g ha−1). Results show that Se concentration in root, straw and grain of rice was increased with increased concentrations of Se in seleniferous soils. The root accumulated higher Se than straw and grain under the same naturally seleniferous soil. Spraying Se significantly increased Se concentration in grain, hull, brown rice and polished rice compared with spraying water. Se concentration in the grain fractions was in the following order: Bran > brown rice > whole grain > polished rice > hull. About 13.7% Se in wholegrain was discarded by milling process if about 6.9% of it was polished as bran. Se-enriched rice could be produced in naturally seleniferous soils with Se concentration from 0.5 to 1.0 mg kg−1, and this polished rice would provide enough Se (60–80 μg day−1) to satisfy the human requirement. Therefore, naturally seleniferous soils may be an effective way to produce Se-enriched rice without spraying Se fertilizer, which will be more economically feasible and environmentally friendly for without exogenous Se added to the soils or plants. However, the polished rice and brown rice, produced by spraying Na2SeO3 (15 g ha−1) or grown in soil with total Se upto 1.5 mg kg−1 was not suitable for daily human consumption, unless diluted with Se-deficient rice to meet the standard (≤0.3 mg Se kg−1). This study imparted a better understanding of the utilization of seleniferous soils and Se-enriched rice for human health and food safety.

Phytoremediation of soil contaminated with PCBs using different plants and their associated microbial communities

In this work, we evaluate the abilities of the plants Brassica juncea, Avena sativa, Brachiaria decumbens, and Medicago sativa to uptake polychlorinated biphenyls (PCBs) and induce degradation of soil microorganisms from contaminated soil. Removal of PCBs 44, 66, 118, 153, 170, and 180 was evaluated in both rhizospheric and nonrhizospheric soils. Microbial and bphA1 gene quantifications were performed by real-time PCR. The PCB concentrations in plant tissues and soil were determined, and a fluorescein diacetate (FDA) hydrolysis assay was used to measure microbial activity in soil. The removal percentages for all PCB congeners in planted soil versus unplanted control soil were statistically significant and varied between 45% and 63%. PCBs 118, 153, 138, and 170 were detected in Brachiaria decumbens roots at different concentrations. In planted soil, an increase in the concentration of bacteria was observed compared to the initial concentration and the concentration in unplanted control soil; however, no significant differences were identified between plants. The number of copies of the bphA1 gene was higher in rhizospheric versus non- rhizospheric soil for all plants at the end of the experiment. However, alfalfa and oat rhizospheric soil showed significant differences in the copy number of the bphA1 gene. In general, the concentration of fluorescein in the rhizospheric soil was greater than that in the nonrhizospheric soil. Although the plants had a positive effect on PCB removal, this effect varied depending on the type of PCB, the plant, and the soil.

Rapid detection of cadmium and its distribution in Miscanthus sacchariflorus based on visible and near-infrared hyperspectral imaging

Monitoring the effectiveness of Miscanthus sacchariflorus to meet the basic requirements for environmental remediation projects is an important step in determining its use as a productive bioenergy crop for phytoremediation. Conventional chemical methods for the determination of cadmium (Cd) contents involve time-consuming, monotonous and destructive procedures and are not suitable for high-throughput screening. In the present study, visible and near-infrared hyperspectral imaging technology combined with chemometric methods was used to assess the Cd concentrations in M. sacchariflorus. The total Cd concentrations in different plant tissues were measured using an inductively coupled plasma-mass spectrometer. Partial least-squares regression and least-squares support vector machine were implemented to estimate Cd contents from spectral reflectance. Successive projections algorithm and competitive adaptive reweighted sampling (CARS) methodology were used for selecting optimal wavelength. The CARS-partial least-squares regression model resulted in the most accurate predictions of Cd contents in M. sacchariflorus leaves, with a determination coefficient (R2) of 0.87 and a root mean square error (RMSE) value of 97.78 for the calibration set, and an R2 value of 0.91 and a RMSE value of 75.95 for the prediction set. The CARS-least-squares support vector machine model resulted in the most satisfactory predictions of Cd contents in roots, with R2 values of 0.95 (RMSE, 0.92 × 103) for the calibration set and 0.90 (RMSE, 1.64 × 103) for the prediction set. Finally, the Cd concentrations in different plant tissues were visualized on the prediction maps by predicted spectral features on each hyperspectral image pixel. Thus, visible and near-infrared imaging combined with chemometric methods produces a promising technique to evaluate M. sacchariflorus' Cd phytoremediation capability in high-throughput metal-contaminated field applications.

Cadmium in plants: uptake, toxicity, and its interactions with selenium fertilizers.

Cd is the third major contaminant of greatest hazard to the environment after mercury and lead and is considered as the only metal that poses health risks to both humans and animals at plant tissue concentrations that are generally not phytotoxic. Cd accumulation in plant shoots depends on Cd entry through the roots, sequestration within root vacuoles, translocation in the xylem and phloem, and Cd dilution within the plant shoot throughout its growth. Several metal transporters, processes, and channels are involved from the first step of Cd reaching the root cells and until its final accumulation in the edible parts of the plant. It is hard to demonstrate one step as the pivotal factor to decide the Cd tolerance or accumulation ability of plants since the role of a specific transporter/process varies among plant species and even cultivars. In this review, we discuss the sources of Cd pollutants, Cd toxicity to plants, and mechanisms of Cd uptake and redistribution in plant tissues. The metal transporters involved in Cd transport within plant tissues are also discussed and how their manipulation can control Cd uptake and/or translocation. Finally, we discuss the beneficial effects of Se on plants under Cd stress, and how it can minimize or mitigate Cd toxicity in plants.

Metabolic engineering strategies for caffeic acid production in Escherichia coli

Caffeic acid (CA; 3,4-dihydroxycinnamic acid) is an aromatic compound obtained by the phenylpropanoid pathway. This natural product has antioxidant, antitumor, antiviral, and anti-inflammatory activities. It is also a precursor of CA phenethyl ester (CAPE), a compound with potential as an antidiabetic and liver-protective agent. CA can be found at low concentrations in plant tissues, and hence, its purification is difficult and expensive. Knowledge regarding the pathways, enzymes, and genes involved in CA biosynthesis has paved the way for enabling the design and construction of microbial strains with the capacity of synthesizing this metabolite. In this review, metabolic engineering strategies for the generation of Escherichia coli strains for the biotechnological production of CA are presented and discussed.

Connection between the disease resistance of sour cherry genotypes and the carbohydrate content of the leaf and phloem tissues

The objective of the present study was to establish a possible connection between disease resistance and the carbohydrate content of plant tissues by examining sour cherry genotypes with different tolerance levels in homeostasis. Research on the sour cherry – Monilinia laxa interaction involved the comparison of two Hungarian cultivars (‘Érdi bőtermő’ and ‘Csengődi’) and their offsprings (8) by measuring the quantity of homeostatic carbohydrate fractions in their leaves and phloem tissues. The results demonstrated that the glucose quantity and the ratio of glucose and fructose to sucrose were correlated with the disease resistance of sour cherry cultivars and their hybrids. The glucose content was higher in susceptible genotypes and lower in tolerant genotypes. The hexose:sucrose ratios of susceptible genotypes were significantly higher than those of tolerant genotypes.

Variations in cadmium accumulation and distribution among different oilseed rape cultivars in Chengdu Plain in China.

Cadmium (Cd) is a widespread toxic heavy metal trace pollutant worldwide. The ability of Cd absorption and accumulation highly varies among different species and varieties. In order to screen oilseed rape cultivars which are appropriate for cultivation and application in Cd-contaminated soils, we conducted the field trial of 32 oilseed rape varieties in Shifang County of Chengdu Plain. The various biomass, Cd accumulation, and distribution patterns were investigated via determining the Cd concentration in different plant tissues. Moreover, the food safety risks of rapeseeds were finally assessed. The results indicated diverse responses to Cd stress appeared in various tested varieties, including plant biomass, seed yield, Cd concentration, and proportion in different tissues. And most Cd were concentrated in non-edible parts. Through cluster analysis, we found that Nanchongjie, Pengzhoubai, and J-25 belong to high-biomass and high-Cd-accumulated groups in experimental cultivars, which indicated that they could possess more biomass and gather higher Cd content in overground part, so they could be great materials for phytoremediation in Cd-polluted area. Besides, combined with the risk assessment of food safety in rapeseeds, cultivars 72A and 47 with the traits of high yield, low-Cd concentration, and low food safety risk can be considered as suitable materials to widely plant as cash crop. These results provide valuable reference for practical planting and application of oilseed rape in Cd-polluted areas.

Transcriptomic changes under stress conditions with special reference to glutathione contents

Plants are sessile organisms. They have to endure the environmental catastrophe throughout their life cycle. To survive in these hostile surroundings, plants posses an efficient and fine-tuned defense mechanism. This is a well established fact that various defense molecules viz. salicylic acid (SA), jasmonic acid, abscisic acid (ABA), ethylene (ET) and so on, are working in a synergistic as well as antagonistic fashion to establish and activate the effective defense mechanism. Interestingly, glutathione is gaining a gradual importance in this complex scenario. This multifunctional biomolecule exists in two forms. The reduced form, viz. GSH, is primarily present at millimolar concentrations in various plant tissues as compared to its oxidized form, glutathione disulfide, GSSG. Proteo-genomics analysis confirmed that GSH plays a vital role in plant resistance against biotic and abiotic stresses by stimulating various defense genes and proteins. In recent times, it has been reported that modulation of GSH contents transmits information through diverse signaling mechanisms. GSH also modulates various stresses and defense related genes by interacting with ABA and ET in response to abiotic stress conditions. However, there are still many unanswered questions about the intricate molecular mechanism of GSH’s contribution in plant defense. With these backgrounds, presently we primarily discussed the transcriptomic changes under stress conditions in Arabidopsis thaliana at altered GSH contents. Transcriptomic profiling of phytoalexin-deficient mutant (pad2.1), a GSH depleted A. thaliana mutant, in response to combined cold and osmotic stress treatment, was compared to that of A. thaliana ecotype Col-0, the wild type, with a view to identify the genes altered under changed GSH conditions to combat stress. It was evident from these datasets that the transcript level responses of pad2.1 to this treatment were massive. Again, analysis of combined cold and osmotic stress treated other mutants of Arabidopsis transcriptome was performed to elucidate the crosstalk between the ABA, ET and GSH. Results revealed the differential regulation of about 2313 and 4131 transcripts in A. thaliana mutants viz. ethylene insensitive (ein2) and ABA deficient 1(aba1.6) respectively. Together, present findings elucidate an active interplay of GSH with SA, ET, and ABA to combat environmental stress conditions in planta.

Mixotrophic acidophiles increase cadmium soluble fraction and phytoextraction efficiency from cadmium contaminated soils

A profound concern in developing microbially-assisted phytoextraction is that introduced microbes not only remove heavy metals from contaminated soils but also enhance metal uptake into plant tissues from the treated soils. Cadmium (Cd) removal efficiencies were compared after leaching with deionized water (CK), acidified basal salts medium (acid control), cell-free spent medium (spent bioleaching) and mixotrophic acidophiles (two-step bioleaching). Two-step bioleaching using the mixotrophic acidophiles removed 34% of total Cd and 87% of available Cd, significantly more than CK (3% and 4%), acid control (12% and 51%) and spent bioleaching (26% and 75%). Pot experiments of water spinach growing in four treated soils were conducted to evaluate the Cd uptake performance in plants. Notably, the mixotrophic acidophiles increased Cd concentration in plant tissues by 78% compared to the CK group. More interestingly, the mixotrophic acidophiles were not colonized in soils but caused the compositional increase of indigenous microbes such as the genera of Alicyclobacillus, Clostridium sensu strict and Streptacidiphilus. Meanwhile, two-step bioleaching had little effects on soil structure and physicochemical properties determined by the spectroscopy characteristics analysis. These results implied that the mixotrophic acidophiles facilitated the development of microbially-assisted phytoextraction technology.
