In Vitro Inhibitory Effect of Nigella sativa L. Extracts on SARS-COV-2 Spike Protein-ACE2 Interaction

Ethnopharmacological relevanceA catastrophic outbreak of severe acute respiratory syndrome (SARS-CoV-2; Coronavirus disease-19; COVID-19) was first detected in Wuhan, Hubei Province, China, in December 2019. Many experimental and clinical studies have focused on the effectiveness of medicinal plants such as Nigella sativa (NS) in combating SARS-CoV-2. In this study, we aimed to evaluate the in vitro effect of NS seed extract on SARS-CoV-2 spike protein (S1)-angiotensin-converting enzyme 2 (S-ACE2) interaction. Materials and methodsNS seed extracts used for the assay were prepared in chloroform, ethanol, and water by Soxhlet extraction and recovered by rotary evaporation. The inhibition percentage of S1-ACE2 interaction was analyzed using ELISA-SARS-CoV-2 S1 Protein-ACE2 Binding Inhibitor Screening Kit. Chemical finger-printing of the extracts was done using RP-HPLC. ResultsSignificant concentration-dependent inhibition of the S1-ACE2 interaction was observed with chloroform, ethanol, and water extracts, ranging from 0.01 to 10 mg/ml. The P-values for the extracts were as follows: 0.0055, 0.0937, 0.0013, and 0.0003 for chloroform extract; 0.0876, 0.0703, 0.0183, and 0.0071 for ethanol extract; and 0.0915, 0.0312, 0.0006, and 0.0006 for water extract. The 50% inhibitory concentration (IC50) was determined to be 0.132 mg/ml, 0.288 mg/ml, and 4.06 mg/ml for chloroform, ethanol, and water extracts, respectively. ConclusionThe in vitro analysis utilizing SARS-CoV2 spike (S1) and ACE2 proteins proved that NS seed extracts have the potential to inhibit the S-ACE2 interaction, which warrants further studies that could lead to potential drug discovery for SARS-CoV2 infection.

Influence of fecal fermentation on the anthelmintic activity of proanthocyanidins and ellagitannins against human intestinal nematodes and Caenorhabditis elegans.

Some tannin-rich plants such as Combretum mucronatum and Phyllanthus urinaria are widely used in Africa for the control of parasitic nematodes in both humans and livestock. Tannins have been recognized as an alternative source of anthelmintic therapies, and hence, recent studies have focused on both the hydrolyzable and condensed tannins. These groups of compounds, however, have poor oral bioavailability and are metabolized by gut microbiota into lower molecular weight compounds. The role of these metabolites in the anthelmintic activities of tannins has not been explored yet. This study investigated the effects of fecal metabolism on the anthelmintic potential of procyanidin C1 (PC1) and geraniin and the tannin-enriched extracts of C. mucronatum (CML) and P. urinaria (PUH), which contain these compounds, respectively. Metabolites were formed by anaerobic fermentation of the test compounds and extracts in a fresh human fecal suspension for 0 h, 4h, and 24h. Lyophilized samples were tested in vitro against hookworm larvae and whipworm (Trichuris trichiura) larvae obtained from naturally infected human populations in Pru West District, Bono East Region, Ghana, and against the wildtype strain of Caenorhabditis elegans (L4). Both extracts and compounds in the undegraded state exhibited concentration-dependent inhibition of the three nematodes. Their activity, however, significantly decreased upon fecal metabolism. Without fermentation, the proanthocyanidin-rich CML extract was lethal against hookworm L3 (LC50 = 343.5μg/mL, 95% confidence interval (CI) = 267.5-445.4), T. trichiura L1 (LC50 = 230.1μg/mL, CI = 198.9-271.2), and C. elegans (LC50 = 1468.1μg/mL, CI = 990.3-1946.5). PUH, from which the ellagitannin geraniin was isolated, exhibited anthelmintic effects in the unfermented form with LC50 of 300.8μg/mL (CI = 245.1-374.8) against hookworm L3 and LC50 of 331.6μg/mL (CI = 290.3-382.5) against T. trichiura L1, but it showed no significant activity against C. elegans L4 larvae at the tested concentrations. Similarly, both compounds, procyanidin C1 and geraniin, lost their activity when metabolized in fecal matter. The activity of geraniin at a concentration of 170μg/mL against C. elegans significantly declined from 30.4% ± 1.8% to 14.5% ± 1.5% when metabolized for 4h, whereas that of PC1 decreased from 32.4% ± 2.3% to 8.9% ± 0.9% with similar treatment. There was no significant difference between the anthelmintic actions of metabolites from the structurally different tannin groups. The outcome of this study revealed that the intact bulky structure of tannins (hydrolyzable or condensed) may be required for their anthelmintic action. The fermented products from the gut may not directly contribute toward the inhibition of the larvae of soil-transmitted helminths.

Phytochemical screening, antioxidant, and cytotoxic activities of sequentially extracted Euclea natalensis leaf extracts

Oxidative stress plays a major role in the pathogenesis of many diseases like diabetes, cancer, cardiovascular diseases, and neurodegenerative diseases. Oxidative stress has become a potential for therapeutic target in the prevention and management of such diseases. This has spiked the interest in the use of natural antioxidants for disease prevention. The present study was aimed at assessing the antioxidant and cytotoxic properties of sequentially extracted Euclea natalensis leaf extracts. The total phenolics and flavonoids were estimated, and antioxidant activity was determined using 2,2-diphenyl-1-picryl hydrazyl (DPPH), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS), total antioxidant capacity, and the ferric reducing/antioxidant power (FRAP) assays. The cytotoxic capacity against HeLa cells was evaluated using WST-1 (4-[3-(4-Iodophenyl)-2-(4-nitro-phenyl)-2H-5-tetrazolio]-1,3-benzene sulfonate) colorimetric assay. Qualitative phytochemical screening showed the presence of phytochemicals such as phenols, flavonoids, tannins, terpenoids and glycosides except in the hexane extract. The total phenolic and flavonoid content ranged from 0.58–118.7 mg GAE/g DW and 9.8–20.1 mg GAE/g DW respectively. All extracts displayed a concentration-dependent inhibition of DPPH and ABTS, with IC50 values in the range 0.04–0.122 mg/mL and 0.079–0.205 mg/mL respectively. The aqueous extract had the highest total antioxidant capacity and ferric reducing power of 40.4 mg AAE/g DW and 0.98 mg Vitamin C equivalence/g DW. The IC50 value for cytotoxicity was in the range 155.3–211.3 mg/mL with the dichloromethane-methanol extracts being the least cytotoxic. It was concluded that Euclea natalensis leaf extracts contain bioactive phytochemicals with antioxidant potential and less cytotoxic capacity.

Isolation of Alpha-Glucosidase Inhibitors from the Panamanian Mangrove Plant Mora oleifera (Triana ex Hemsl.) Ducke.

Panama boasts an expansive mangrove area and stands as one of the most biodiverse countries in America. While mangrove plants have long been utilized in traditional medicine, there are still unstudied species whose potential medicinal applications remain unknown. This study aimed to extract bioactive compounds from Mora oleifera (Triana ex Hemsl.) Ducke, an understudied mangrove species. Through bioassay-guided fractionation of the crude extract, we isolated seven active compounds identified as lupenone (1), lupeol (2), α-amyrin (3), β-amyrin (4), palmitic acid (5), sitosterol (6), and stigmasterol (7). Compound structures were determined using spectroscopic analyses, including APCI-HR-MS and NMR. Compounds 1-7 displayed concentration-dependent inhibition of the alpha-glucosidase enzyme, with IC50 values of 0.72, 1.05, 2.13, 1.22, 240.20, 18.70, and 163.10 µM, respectively. Their inhibitory activity surpassed acarbose, the positive control (IC50 241.6 µM). Kinetic analysis revealed that all compounds acted as competitive inhibitors. Docking analysis predicted that all triterpenes bonded to the same site as acarbose in human intestinal alpha-glucosidase (PDB: 3TOP). A complementary metabolomic analysis of M. oleifera active fractions revealed the presence of 64 compounds, shedding new light on the plant's chemical composition. These findings suggest that M. oleifera holds promise as a valuable botanical source for developing compounds for managing blood sugar levels in individuals with diabetes.

Modulation of PETase active site flexibility and activity on morphologically distinct polyethylene terephthalate substrates by surface charge engineering

Enzymatic hydrolysis of polyethylene terephthalate (PET) waste is a compelling strategy for environmentally friendly recycling of a major pollutant. Here, we investigate the effects of surface charge point mutations both proximal and distal to the active site of the mesophilic PET-degrading enzyme IsPETase and the thermostable V3 variant with superior activity. The vicinal K95A mutation significantly inhibited IsPETase activity on mechanically processed PET powder. Conversely, this mutation significantly increased hydrolysis of PET powder in the V3 PETase. Activity of both enzymes on PET film was inhibited by the K95A mutation, highlighting complex interplay between mutation context and substrate morphology. Further installing the distal R132N and R280A surface charge mutations potentiated activity of V3 on all substrates tested. This variant afforded 100 % degradation of pre-processed bottle-grade PET powder in 3 days at 40°C reaction temperature, a 3-fold improvement over IsPETase. Whilst reduction of positive charge on the PETase surface is known to reduce interaction with PET, molecular dynamics simulations suggest this can be offset by context-dependent modulation of active site flexibility, which differentially impacts both hydrolysis of morphologically distinct PET substrates and the concentration-dependent inhibition phenomenon observed for PETase.

Identification of Matrine as a Kirsten rats Arcomaviral oncogene homolog inhibitor alleviating chemotherapy-induced neuropathic pain

BackgroundChemotherapy-induced peripheral neuropathy (CIPN) represents a prevailing and severe clinical concern, characterized by limited availability of clinically effective treatment strategies. Current evidence endorses matrine's potential as a neuroprotective and analgesic agent for CIPN. Nevertheless, the precise targets and mechanisms of action of matrine remain insufficiently explored, impeding comprehensive pharmacological investigation and clinical application. ObjectiveThis study endeavors to elucidate the analgesic and neuroprotective effects of matrine in mice with vincristine-induced neuropathic pain. A focal point is the identification of matrine's specific target and the underlying molecular mechanisms governing its analgesic and neuroprotective actions. MethodsTo discern matrine's analgesic effects in CIPN mice, we conducted behavioral experiments encompassing the Von Frey filament test and Hargreaves Test. Furthermore, we conducted electrophysiological and histopathological assessments involving HE staining, Nissl staining, and Fluoro-Jade B staining to evaluate matrine's effects on neuroprotection within dorsal root ganglia and the spinal cord of CIPN mice. Sequentially, thermal shift assay, GTP hydrolysis assay, and nucleotide exchange assay were executed to validate matrine's inhibitory effects on KRAS. Molecular docking and site-directed mutagenesis experiments were implemented to identify the precise binding pocket of matrine on KRAS. Lastly, matrine's inhibitory effects on downstream signaling pathways of KRAS were confirmed through experiments conducted at animal model. ResultsMatrine exhibited a notable increase in mechanical withdrawal threshold and thermal withdrawal latency in vincristine-treated mice. This compound substantially ameliorated the neurofunctional blockade associated with sensory and motor functions induced by vincristine. Moreover, matrine mitigated pathological damage within DRG and the L4-L5 spinal cord regions. The study's MST experiments indicated matrine's substantial elevation of KRAS's melting temperature. The GTP hydrolysis and nucleotide exchange assays revealed concentration-dependent inhibition of KRAS activity by matrine. Molecular docking provided insight into the binding mode of matrine with KRAS, while site-directed mutagenesis verified the specific binding site of matrine on KRAS. Lastly, matrine's inhibition of downstream Raf/Erk1/2 and PI3K/Akt/mTOR signaling pathways of KRAS was confirmed in VCR mice. ConclusionCompared to previous studies, our research has identified matrine as a natural inhibitor of the elusive protein KRAS, often considered "undruggable." Furthermore, this study has revealed that matrine exerts its therapeutic effects on chemotherapy-induced peripheral neuropathy (CIPN) by inhibiting KRAS activation, subsequently suppressing downstream signaling pathways such as Raf/Erk1/2 and PI3K/Akt/mTOR. This investigation signifies the discovery of a novel target for matrine, thus expanding the potential scope of its involvement in KRAS-related biological functions and diseases. These findings hold the promise of providing a crucial experimental foundation for forthcoming drug development initiatives centered around matrine, thereby advancing the field of pharmaceutical research.

The Topical Novel Formulations of Interferon α-2в Effectively Inhibit HSV-1 Keratitis in the Rabbit Eye Model and HSV-2 Genital Herpes in Mice.

Herpes simplex viruses type 1 (HSV-1) and type 2 (HSV-2) are widespread human pathogens that establish chronic latent infections leading to recurrent episodes. Current treatments are limited, necessitating the development of novel antiviral strategies. This study aimed to assess the antiviral efficacy of novel topical formulations containing interferon alpha-2b (IFN α-2b) against HSV-1 and HSV-2. The formulations, Oftalmoferon® forte (eye drops) and Interferon Vaginal Tablets, demonstrated potent antiviral effects against HSV-1 and HSV-2 in Vero cells, respectively, with concentration-dependent inhibition of viral replication. Subsequently, their efficacy was tested in animal models: HSV-1 keratitis in the rabbit eye model and HSV-2 genital herpes in mice. Oftalmoferon® forte effectively treated HSV-1 keratitis, reducing clinical symptoms and ulcerations compared to virus control. Interferon Vaginal Tablets showed promising results in controlling HSV-2 genital herpes in mice, improving survival rates, reducing clinical signs, weight loss and viral replication. The novel IFN α-2b formulations exhibited significant antiviral activity against HSV infections in cell culture and animal models. These findings suggest the potential of these formulations as alternative treatments for HSV infections, particularly in cases resistant to current therapies. Further studies are warranted to optimize treatment regimens and assess clinical efficacy in humans.

Evaluation of the synergistic effects of curcumin-resveratrol co-loaded biogenic silica on colorectal cancer cells.

Colorectal cancer (CRC) remains a significant global health concern, being the third most diagnosed cancer in men and the second most diagnosed cancer in women, with alarming mortality rates. Natural phytochemicals have gained prominence among various therapeutic avenues explored due to their diverse biological properties. Curcumin, extracted from turmeric, and resveratrol, a polyphenol found in several plants, have exhibited remarkable anticancer activities. However, their limited solubility and bioavailability hinder their therapeutic efficacy. To enhance the bioavailability of these compounds, nanomaterials work as effective carriers with biogenic silica (BS) attracting major attention owing to their exceptional biocompatibility and high specific surface area. In this study, we developed Curcumin-resveratrol-loaded BS (Cur-Res-BS) and investigated their effects on colorectal cancer cell lines (HCT-116 and Caco-2). Our results demonstrated significant concentration-dependent inhibition of cell viability in HCT-116 cells and revealed a complex interplay of crucial proto-onco or tumor suppressor genes, such as TP53, Bax, Wnt-1, and CTNNB1, which are commonly dysregulated in colorectal cancer. Notably, Cur-Res-BS exhibited a synergistic impact on key signaling pathways related to colorectal carcinogenesis. While these findings are promising, further investigations are essential to comprehensively understand the mechanisms and optimize the therapeutic strategy. Moreover, rigorous safety assessments and in vitro studies mimicking the in vivo environment are imperative before advancing to in vivo experiments, ensuring the potential of Cur-Res-BS as an efficient treatment for CRC.

Wedelolactone suppresses breast cancer growth and metastasis via regulating TGF-β1/Smad signaling pathway.

Breast cancer is a malignant tumor with high invasion and metastasis. TGF-β1-induced epithelial-mesenchymal transition (EMT) is crucially involved in the growth and metastasis of breast cancer. Wedelolactone (Wed) is extracted from herbal medicine Ecliptae Herba, which is reported to have antineoplastic activity. Here, we aimed to elucidate the efficacy and mechanism of Wed against breast cancer. The effects of Wed on migration and invasion of 4T1 were detected. The expression of EMT-related markers was detected by Western blot and qPCR. The 4T1 orthotopic murine breast cancer model was established to evaluate the therapeutic effect of Wed on the growth and metastasis of breast cancer through TGF-β1/Smad pathway. Wed inhibited the proliferation, migration and invasion of 4T1. It exhibited concentration-dependent inhibition of p-Smad2/3. Wed also reversed the expression of EMT-markers induced by TGF-β1. In addition, Wed suppressed the growth and metastasis of breast cancer in mice. It also affected p-Smad3 expression as well as EMT-related genes, suggesting that its anti-breast cancer effect may be related to the TGF-β1/Smad pathway. Wed reverses EMT by regulating TGF-β1/Smad pathway, potentially serving as a therapeutic agent for breast cancer. Wed is expected to be a potential drug to inhibit TGF-β1/Smad pathway-related diseases.

Evaluation of In vitro Anticancer Activity (Ovarian Cancer Cells-pa1) and Zebrafish Embryo Toxicity of Parmelia perlata Ethanolic Extract

The Phytochemical composition and bioactivity of an ethanolic extract of Parmelia perlata were analyzed in this study using established procedures. The extract revealed a rich array of phytochemicals, showcasing its diverse composition, and notably exhibited strong antioxidant activity according to the 1, 1-diphenyl-2- picrylhydrazyl (DPPH) test. Furthermore, the research explored the extract’s potential in preventing Bovine Serum Albumin (BSA) denaturation, comparing it with the well-known medication Diclofenac. The concentration-dependent inhibition of BSA denaturation by P. perlata ethanolic extract was comparable to the effects observed with diclofenac on BSA. The ethanolic extract demonstrated concentration-dependent antibacterial activity against gram-negative and gram-positive bacterial isolates. While gentamicin served as a positive control with larger zones of inhibition, P. perlata extract displayed significant antibacterial efficacy against all six tested organisms. Moreover, in vitro testing against human ovarian (PA-1) cell lines demonstrated the extract’s anticancer potential. A concentration-dependent reduction in cell viability, culminating in the lowest percentage at 1000 µg/ml, was observed. The IC50 value of 31.2 µg/mL showed that the P. perlata extract strongly inhibited the PA-1 cell line. This was confirmed by the observed changes in the cellular morphology when exposed to higher concentrations of the extract. Further, the ethanolic extract of P. perlata was subjected to various concentrations to evaluate its embryotoxicity on zebrafish embryos. The concentration of extract was determined to have a safety limit of less than 50 µg/L. In summary, P. perlata ethanolic extract has emerged as a promising natural resource, exhibiting a spectrum of bioactivities, including potent antioxidant, anti-inflammatory, antibacterial, anticancer properties, and embryonic toxicology study. These findings suggest that its potential utility in pharmaceutical applications is warranted, and further exploration and development are being suggested.

Insights into the antioxidant, anti-inflammatory and anti-microbial potential of Nigella sativa essential oil against oral pathogens

Oral disorders can exert systemic ramifications beyond their localized effects on dental tissues, implicating a wide array of physiological conditions. The utilization of essential oils (EOs) for protection of oral health represents a longstanding practice. Consequently, in this investigation, essential oil derived from Nigella sativa seeds (NSEO) underwent isolation via the hydro-distillation process, followed by a comprehensive evaluation of its antioxidant, anti-inflammatory, anti-fungal, antibacterial activities, and cytocompatibility. The isolated NSEO manifested as a pale-yellow substance and was found to harbor a diverse spectrum of bioactive constituents, including steroids, triterpenoids, flavonoids, phenols, proteins, alkaloids, tannin, sesquiterpenoid hydrocarbons, monoterpenoid alcohol, and monoterpenoid ketone (thymoquinone). Notably, the total phenolic content (TPC) and total flavonoid content (TFC) of NSEO were quantified at 641.23 μg GAE/gm and 442.25 μg QE/g, respectively. Furthermore, NSEO exhibited concentration-dependent inhibition of protein denaturation, HRBC membrane stabilization, and hemolysis inhibition. Comparative analysis revealed that NSEO and chlorhexidine (CHX) 0.2% displayed substantial inhibition of hemolysis compared to aspirin. While NSEO and CHX 0.2% demonstrated analogous antibacterial activity against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa, NSEO showcased heightened efficacy against Lactobacillus acidophilus and Candida albicans. Additionally, NSEO exhibited pronounced effects against periodontal pathogens such as Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis, Tannerella forsythia, and Prevotella intermedia. Importantly, no cytotoxicity was observed on human gingival fibroblast cell lines. These findings underscore the potential of NSEO as a potent antibacterial and antifungal agent in the management of oral microbial pathogens, thereby offering avenues for the development of innovative therapies targeting diverse oral inflammatory conditions. Nevertheless, further investigations are imperative to unlock its full therapeutic repertoire.

Nanoparticle-based delivery of harmine: A comprehensive study on synthesis, characterization, anticancer activity, angiogenesis and toxicity Evaluation

The effective treatment of cancer presents numerous challenges, including drug resistance and the risk of detrimental effects on normal tissues. Harmine, a beta-carboline alkaloid, has demonstrated diverse biological properties. This study aimed to synthesize and characterize harmine encapsulated in polylactic-co-glycolic acid (PLGA) nanoparticles (Ha-PLGA-NPs) to investigate their potential as agents against cancer and angiogenesis. The synthesized Ha-PLGA-NPs were thoroughly characterized, exhibiting a connected rod-shaped crystal which some retaining the spherical shape of nanoparticles with an average size of 302.96 nm. Furthermore, the nanoparticles demonstrated a dispersion index of 0.23 and a surface charge of −16.51 mV. In vitro cytotoxicity assays conducted on the breast cancer cell line (MCF-7) revealed that Ha-PLGA-NPs possessed significant cytotoxic properties, with an observed IC50 value of 87.74 μg/mL. Notably, no substantial cytotoxicity was observed in human foreskin fibroblasts, indicating a favorable selectivity towards cancer cells. Evaluation of the anti-angiogenic activity of Ha-PLGA-NPs demonstrated a concentration-dependent inhibition of angiogenesis. Mechanistic investigations indicated that the observed inhibition was mediated through the regulation of key genes involved in angiogenesis, including caspase 3, caspase 9, VEGF, and VEGF-R. In vivo studies involving dietary administration of Ha-PLGA-NPs in mice revealed improvements in weight gain, feed intake, liver enzyme levels, and redox potential. These findings underscore the potential of Ha-PLGA-NPs as a promising therapeutic agent for cancer treatment. The observed effects are attributed to their ability to induce programmed cell death and inhibit angiogenesis, thus offering a multifaceted approach to combat cancer.

N-Acetyl-L-cysteine attenuates titanium dioxide nanoparticle (TiO2 NP)-induced autophagy in male germ cells

Titanium dioxide nanoparticles (TiO2 NPs) are widely used in consumer products, raising concerns about their impact on human health. This study investigates the effects of TiO2 NPs on male germ cells while focusing on cell proliferation inhibition and underlying mechanisms. This was done by utilizing mouse GC-1 spermatogonia cells, an immortalized spermatogonia cell line. TiO2 NPs induced a concentration-dependent proliferation inhibition with increased reactive oxygen species (ROS) generation. Notably, TiO2 NPs induced autophagy and decreased ERK phosphorylation. Treatment with the ROS inhibitor N-Acetyl-l-cysteine (NAC) alleviated TiO2 NPs-induced autophagy, restored ERK phosphorylation, and promoted cell proliferation. These findings call attention to the reproductive risks posed by TiO2 NPs while also highlighting NAC as a possible protective agent against reproductive toxins.

Antibacterial activity of pimaradienoic acid from Eleutherococcus trifoliatus (L.) S. Y. Hu.

Purpose: To investigate the antibacterial activity of pimaradienoic acid, a natural product isolated from Eleutherococcus trifoliatus. Methods: Pimaradienoic acid was purified from E. trifoliatus and tested against three Gram-positive bacteria. Minimum inhibitory concentrations (MICs) were determined, and bacterial growth curves were measured. Scanning electron microscopy (SEM) was used to observe morphological changes in bacteria after drug treatment. Results: Pimaradienoic acid exhibited concentration-dependent inhibition of the growth of the three bacteria tested. The MIC and bacterial growth dynamics results indicated that pimaradienoic acid had potent antibacterial activity. SEM revealed that pimaradienoic acid disrupted the bacterial membrane, leading to cell death. Conclusions: Pimaradienoic acid has significant antibacterial activity against Gram-positive bacteria, suggesting its potential as a novel antimicrobial agent.

Silibinin-loaded chitosan-capped silver nanoparticles exhibit potent antimicrobial, antibiofilm, and anti-inflammatory activity against drug-resistant nosocomial pathogens

Nanoparticles capped with natural products can be a cost-effective alternative to treat drug-resistant nosocomial infections. Therefore, silibinin-loaded chitosan-capped silver nanoparticles (S-C@AgNPs) were synthesized to evaluate their antimicrobial and anti-inflammatory potential. The S-C@AgNPs plasmon peak was found at 430 nm and had a particle size distribution of about 130 nm with an average hydrodynamic diameter of 101.37 nm. The Scanning Electron Microscopy images showed the presence of sphere-shaped homogeneous nanoparticles. The Fourier Transform Infrared Spectroscopy analysis confirmed the loading of silibinin and chitosan on the AgNPs surface. The minimum inhibitory concentration of the S-C@AgNPs was reported between 3.12 μg/ml to 12.5 μg/ml and a minimum bactericidal concentration between 6.25 μg/ml to 25 μg/ml against drug-resistant nosocomial pathogens. Moreover, concentration-dependent significant inhibition of the biofilm formation was reported against P. aeruginosa (70.21%) and K. pneumoniae (71.02%) at 30 μg/ml, and the highest destruction of preformed biofilm was observed at 100 μg/ml against P. aeruginosa (89.74%) and K. pneumoniae (77.65%) as compared to individual bacterial control. Additionally, the fluorescence live/dead assay for bacterial biofilm confirmed that 100 µg/ml effectively inhibits the biofilm formed by these pathogens. S-C@AgNPs also showed anti-inflammatory activity, which is evident by the significant decrease in the proinflammatory cytokines and chemokines level in THP1 cells treated with LPS. This study concluded that S-C@AgNPs have potent antimicrobial, antibiofilm, and anti-inflammatory properties and could be a potential option for treating drug resistant nosocomial infections.

Effects of Chlorogenic Acid on Lowering IgE-Binding Capacity of Soybean 7S: Comparison between Covalent and Noncovalent Interaction.

Allergenicity of soybean 7S protein (7S) troubles many people around the world. However, many processing methods for lowering allergenicity is invalid. Interaction of 7S with phenolic acids, such as chlorogenic acid (CHA), to structurally modify 7S may lower the allergenicity. Hence, the effects of covalent (C-I, periodate oxidation method) and noncovalent interactions (NC-I) of 7S with CHA in different concentrations (0.3, 0.5, and 1.0 mM) on lowering 7S allergenicity were investigated in this study. The results demonstrated that C-I led to higher binding efficiency (C-0.3:28.51 ± 2.13%) than NC-I (N-0.3:22.66 ± 1.75%). The C-I decreased the α-helix content (C-1:21.06%), while the NC-I increased the random coil content (N-1:24.39%). The covalent 7S-CHA complexes of different concentrations had lower IgE binding capacity (C-0.3:37.38 ± 0.61; C-0.5:34.89 ± 0.80; C-1:35.69 ± 0.61%) compared with that of natural 7S (100%), while the noncovalent 7S-CHA complexes showed concentration-dependent inhibition of IgE binding capacity (N-0.3:57.89 ± 1.23; N-0.5:46.91 ± 1.57; N-1:40.79 ± 0.22%). Both interactions produced binding to known linear epitopes. This study provides the theoretical basis for the CHA application in soybean products to lower soybean allergenicity.

Phenol content, potential antioxidant and anti-urolithic effects of fruit and leaf extracts from Algerian Capparis spinosa L.

Background: Ethnobotanical studies have historically documented the traditional medicinal use of Caper (Capparis spinosa L.) in treating kidney stones and other chronic renal ailments. Aims: This study aimed to evaluate the phenolic content, antioxidant capacity, and potential inhibitory effects of Capparis spinosa L. fruit and leaf extracts on calcium oxalate nucleation and aggregation. Material and Methods: Plant material was collected from the Tessala Mountains in Algeria. Total phenolic content was determined using the Folin-Cioclateu method, while in vitro antioxidant activity was assessed via the DPPH assay. In vitro anti-urolithic testing involved the use of turbidimetry in kinetic experiments to evaluate calcium oxalate nucleation and aggregation. Results: The IC50 values for antioxidant activity against DPPH were 8.67 mg/mL for fruit extracts and 0.57 mg/mL for leaf extracts. Furthermore, the investigated extracts exhibited a concentration-dependent inhibition of calcium oxalate crystal nucleation and aggregation. At the highest concentrations of leaf and fruit extracts, the corresponding inhibition rates were 74.47% and 66.06% for the nucleation test and 67.75% and 54.19% for the aggregation assay, respectively. Conclusions: These findings substantiate the traditional usage of Capparis spinosa L. in managing urinary lithiasis, suggesting its potential application as either a preventive measure or a therapeutic intervention for urinary stone formation. Unveiling the molecular mechanisms of size-dependent effect of polystyrene micro/nano-plastics on Chlamydomonas reinhardtii through proteomic profiling

Microplastics have become a prevalent environmental pollutant due to widespread release and production. Algae, as primary producers, play a crucial role in maintaining the ecological balance of freshwater environments. Despite reports on the inhibition of microalgae by microplastics, the size-dependent effects on microalgae and associated molecular mechanism remain poorly understood. This study investigates the impacts of three polystyrene micro/nano-plastics (PS-MNPs) with different sizes (100 nm, 350 nm, and 6 μm) and concentrations (25–200 mg/L) on Chlamydomonas reinhardtii (C. reinhardtii) throughout its growth period. Results reveal size- and concentration-dependent growth inhibition and induction of oxidative stress by PS-MNPs, with microalgae exhibiting increased vulnerability to smaller-sized and higher-concentration PS-MNPs. Proteomics analysis elucidates the size-dependent suppression of proteins involved in the photosynthesis process by PS-MNPs. Photosynthetic activity assays demonstrate that smaller PS-MNPs more significantly reduce chlorophyll content and the maximal photochemical efficiency of photosystem II. Finally, electron microscope and Western blot assays collectively confirm the size effect of PS-MNPs on microalgae growth is attributable to suppressed protein expression rather than shading effects. This study contributes to advancing our understanding of the intricate interactions between micro/nano-plastics and algae at the molecular level, emphasizing the efficacy of proteomics in dissecting the mechanistic aspects of microplastics-induced biological effects on environmental indicator organisms.

Comparison of the efficiency of traditional MICP and two-step MICP method for immobilizing heavy metals in aquatic environments

The application of the microbially induced carbonate precipitation (MICP) method for remediating heavy metals (i.e., HMs) has recently garnered significant attention. Nevertheless, the inhibition of urease activity by toxic Cd2+, Pb2+, Zn2+, and Cu2+ poses a challenge for MICP-based remediation of HMs contamination. This study: (1) first performed the traditional MICP tests (in which the bacterial solution, urea solution, and HMs were mixed simultaneously), and investigated the toxic effect of HMs on the urease activity and the immobilization efficiency, (2) analyzed the toxicity and immobilization mechanism during the MICP process by combining the simulation and XRD tests, (3) conducted the two-step MICP tests (which initially mixed the bacterial solution and urea solution to promote urea hydrolysis, then added the HMs solutions for HMs precipitation) to improve the immobilization efficiency. The tube experiments and simulations were investigated in the HMs concentration range from 1 to 10 mmol/L. Indicators including ammonium concentration, HMs concentrations, and pH were measured/recorded during the tests. The results show that soluble HMs exhibit a concentration-dependent inhibition of urea hydrolysis during the traditional MICP process, resulting in a decreasing immobilization efficiency. The two-step MICP method can effectively immobilize almost the Cd2+ and Zn2+ when the initial urea hydrolysis period exceeds 1–2 h. In addition, a high immobilization rate of over 90% can be achieved for Cu-contaminated solutions at the optimal first-step reaction time. Compared with the traditional MICP procedure, the effective two-step MICP method exhibits more promising application prospects for the immobilization of soluble HMs in aquatic environments.

Mechanistic insight into the anti-alternaria activity of bimetallic zinc oxide and silver/zinc oxide nanoparticles

Alternaria alternata is an opportunistic phytopathogen that negatively impact the growth and production of a wide variety of host plants. In this study, we evaluated the antifungal potential of biogenic ZnO, and bimetallic silver and zinc oxide (Ag/ZnO) nanoparticles synthesized using seed extract of Abrus precatorious and characterized using different analytical tools. In vitro antifungal potentials of ZnO and Ag/ZnO nanoparticles were carried out using the food poison technique. Morphological and ultrastructure of the A. alternata treated with the nanoparticles were carried out using high resolution scanning and transmission electron microscopy (HRSEM and HRTEM). In addition, changes in polysaccharide production, chitin content and enzymatic (cellulase and lipase) activities of A. alternata were assayed. Double peak signifying a UVmax of 353.88 and 417.25 nm representing Ag and ZnO respectively was formed in the bimetallic nanoparticles. HRSEM and HRTEM results shows agglomerated nanoparticles with particle and crystallite size of 23.94 and 16.84 nm for ZnO nanoparticles, 35.12 and 28.99 nm for Ag/ZnO nanoparticles respectively. In vitro antifungal assay shows a significant concentration-dependent inhibition (p < 0.05) of A. alternata mycelia with highest percentage inhibition of 73.93 % (ZnO nanoparticles) and 68.26 % (Ag/ZnO nanoparticles) at 200 ppm. HRSEM and HRTEM micrographs of the treated A. alternata mycelia shows alteration of the cellular structure, clearance of the cytoplasmic organelles and localization of the nanoparticles within the cell. A. alternata treated with 200 ppm nanoparticles show a significant decrease (p < 0.05) in the polysaccharides and chitin contents, cellulase and lipase activities. The results suggests that ZnO and Ag/ZnO nanoparticles mode of action may be via alteration of the fungal cell wall through the inhibition of polysaccharides, chitin, cellulases and lipases synthesis. ZnO and Ag/ZnO nanoparticles may be a promising tool for the management and control of disease causing fungal phytopathogens.
