Aqua drugs and chemicals used in commercial aquaculture in Mymensingh, Bangladesh

Mymensingh is a division in north-central Bangladesh renowned for its lush landscapes and agricultural productivity, with a strong focus on aquaculture as a key part of its economy. The study was performed to investigate the aqua drugs companies and their products used in commercial aquaculture farms in Mymensingh division. Questionnaire based data were collected from representatives of various aqua drug companies, district and upazila fisheries offices, chemical vendors, farmers, and hatchery owners of the Mymensingh division. The present study found aquaculture practices in Mymensingh rely heavily on various antibiotics, probiotics, disinfectants, and feed additives to prevent the spread of diseases, maintain water quality, and increase growth of culture fishes. Drugs from various companies are widely utilized across the Mymensingh division. Fishtech BD contributing the highest percentage (12%), followed by Eon Pharmaceutical Ltd. (11%) and ACI Animal Health (10%), while AVON Animal Health made the lowest contribution (2%). Among the various drugs from different companies, Renamycin (antibiotic), Timsen and Emsen (disinfectants), Pond Care (probiotics), Zeolite Gold, Gasonex, and Mega Zeo Plus (zeolite) are commonly preferred by the farmers. The study also found that many farmers lack knowledge about chemical ingredients, proper dosages, and appropriate application methods. Therefore, adequate training and implementation of government rules and regulations are necessary to ensure responsible use.

Comparative genomics of dusky kob (Argyrosomus japonicus, Sciaenidae) conspecifics: Evidence for speciation and the genetic mechanisms underlying traits.

Dusky kob (Argyrosomus japonicus) is a commercially important finfish, indigenous to South Africa, Australia, and China. Previous studies highlighted differences in genetic composition, life history, and morphology of the species across geographic regions. A draft genome sequence of 0.742 Gb (N50 = 5.49 Mb; BUSCO completeness = 97.8%) and 22,438 predicted protein-coding genes was generated for the South African (SA) conspecific. A comparison with the Chinese (CN) conspecific revealed a core set of 32,068 orthologous protein clusters across both genomes. The SA genome exhibited 440 unique clusters compared to 1928 unique clusters in the CN genome. Transportation and immune response processes were overrepresented among the SA accessory genome, whereas the CN accessory genome was enriched for immune response, DNA transposition, and sensory detection (FDR-adjusted p < 0.01). These unique clusters may represent an adaptive component of the species' pangenome that could explain population divergence due to differential environmental specialisation. Furthermore, 700 single-copy orthologues (SCOs) displayed evidence of positive selection between the SA and CN genomes, and globally these genomes shared only 92% similarity, suggesting they might be distinct species. These genes primarily play roles in metabolism and digestion, illustrating the evolutionary pathways that differentiate the species. Understanding these genomic mechanisms underlying adaptation and evolution within and between species provides valuable insights into growth and maturation of kob, traits that are particularly relevant to commercial aquaculture.

Harnessing Hue: Advances and Applications of Fish Skin Pigmentation Genetics in Aquaculture

Fish exhibit a broad spectrum of colors and patterns facilitated by specialized cells known as chromatophores. The vibrant coloration of fish, controlled by complex genetic and environmental interactions, serves critical roles in ecological functions such as mating, predation, and camouflage. This diversity not only makes fish an invaluable model for exploring the molecular mechanisms of pigmentation but also significantly impacts their economic value within the aquaculture industry, where color traits can drive marketability and breeding choices. This review delves into the sophisticated biological processes governing fish pigmentation and discusses their applications in enhancing aquaculture practices. By exploring the intersection of genetic regulation, environmental influences, and advanced breeding techniques, this review highlights both the scientific understanding and practical applications of fish coloration, providing a bridge between basic biological research and its application in commercial aquaculture.

Genomic architecture and sex chromosome systems of commercially important fish species in Asia – Current status, knowledge gaps and future prospects

AbstractAquaculture is one of the fastest growing agro‐food industries in the world and Asia dominates the world's fish production (70% of total) with a high potential to ensure global food security. The leading aquaculture fish species in Asia show sexual dimorphism for traits such as growth, age at sexual maturity etc. in which one sex outperforms the other, increasing the need for controlling sex‐ratio in the farming system. Despite the huge contribution of Asian aquaculture to global fish production, the Asian aquaculture is facing a lot of challenges, for example, maintaining the genetic purity of the fish species and balancing the sex‐ratio in the culture system. Many of these difficulties arise due to a lack of deep understanding of fish genetics, or to the duplicated genome of cultured species, or to the unavailability of high‐quality reference genomes and information on sex determination mechanisms (crucial for monosex production and establishing appropriate breeding programme). This review has described the existing knowledge of the sex determination mechanisms in fish and genome architecture of four commercially important fish groups in Asia (carps, tilapias, catfishes and snakeheads). We also have outlined possible strategies for identifying sex‐determining gene/s using modern genome sequencing and computational technologies. Finally, we have highlighted the existing challenges and future prospects of Asian aquaculture. We anticipate that developments in genome sequencing technologies will help to address the biological complexities and challenges that exist in commercial aquaculture and will be of value for the future growth of the aquaculture industry in Asia.

Microplastics weaken the digestion and absorption functions in the golden pompano (Trachinotus blochii) by affecting the intestinal structure, bacteria and metabolites

Microplastics are difficult to degrade and widespread environmental pollutants. Coastal areas are hardest hit of microplastic pollution as they receive significant amounts of microplastics discharged from inland sources. Golden pompano (Trachinotus blochii) is a high commercial valuable marine aquaculture fish species, most of the golden pompano are raised in coastal areas, which means they are at significant risk of exposure to microplastics. Therefore, we exposed golden pompano to 10 μg/L, 100 μg/L and 1000 μg/L of 5 μm spherical polystyrene microplastics and conducted a 14-day stress experiment. Histopathology results showed the intestinal villi shrank. The 16s sequencing analysis revealed that microplastics significantly impacted the abundance and community structure of intestinal microorganisms, which may affect the metabolic function of the gastrointestinal tract. Metabolomics sequencing of the intestinal contents showed that microplastics caused disruptions in lipid, glucose, and amino acid metabolism, thus compromising the normal digestion and absorption functions in the intestinal system. In addition, the activation of various pathways, including the intestinal endocrine system, proline metabolism, and signal transduction, which can lead to the occurrence of several diseases. This study combined various methods to investigate the adverse effects of microplastics on intestinal digestion and absorption, and provided new insights into the toxic mechanisms of microplastics.

Isolation and evaluation of local microalgal isolates as feed for larval rearing of Crassostrea tulipa towards optimisation of mangrove oyster seed production in Ghana

AbstractThis study explored the potential of three local microalgal isolates as feed for oyster larvae in laboratory‐rearing experiments, towards the optimisation of seed production of Crassostrea tulipa to support its large‐scale farming along the West African coast. Three species of local microalgae—Rhodomonas sp., Nannochloropsis sp. and Pseudanabaena sp.—were isolated from waters off the coast of Ghana, West Africa, following a serial dilution technique. The growth performance of the isolates was assessed in the laboratory through daily estimation of cell density until the stationary phase was observed. Characterisation of the microalgal isolates was carried out by estimation of their biovolume, carbon content and energy content. Biovolumes of the microalgae were calculated from the estimated equivalent spherical diameters using proposed geometric shapes and formulae. Carbon weight and carbon energy content were subsequently calculated using derived conversions. The three microalgal isolates showed potential for large‐scale cultivation in the laboratory with marked differences in daily increases in cell densities. Nannochloropsis sp. and Rhodomonas sp. recorded the highest and the lowest peak densities of 2.4 × 105 and 1.5 × 105 cell mL−1, respectively, from an initial inoculating cell density of 1.05 × 105 cell mL−1. The estimated mean biovolumes of Rhodomonas sp., Nannochloropsis sp. and Pseudanabaena sp. were 238.9, 8.182 and 42.42 µm3, respectively, and the corresponding derived carbon energy contents were 1.7 × 10−6, 7.13 × 10−8 and 1.05 × 10−7 J, respectively. Results from a laboratory rearing experiment indicated that the individual microalgal isolates supported the growth and survival of oyster larvae at different scales, but a mixed diet of the three promoted superior growth and survival of C. tulipa larvae. The three local microalgal isolates‐ Rhodomonas sp., Nannochloropsis sp., and Pseudanabaena sp.‐ were well adapted to laboratory culture conditions, and the observed differences in growth and survival of the oyster larvae fed on these algal diets could be due to the differences in diet properties and biochemical compositions of the different species. A combination of the three algal diets, however, provided complementaery nutrients for the optimal growth and survival of C. tulipa larvae. The outcome of this study shows that local microalgal isolates have the potential to support hatchery rearing of C. tulipa, which is essential for the development of commercial mangrove oyster aquaculture in West Africa.

Genome-wide association analysis of the resistance to infectious hematopoietic necrosis virus in two rainbow trout aquaculture lines confirms oligogenic architecture with several moderate effect quantitative trait loci

Infectious hematopoietic necrosis (IHN) is a disease of salmonid fish that is caused by the IHN virus (IHNV), which can cause substantial mortality and economic losses in rainbow trout aquaculture and fisheries enhancement hatchery programs. In a previous study on a commercial rainbow trout breeding line that has undergone selection, we found that genetic resistance to IHNV is controlled by the oligogenic inheritance of several moderate and many small effect quantitative trait loci (QTL). Here we used genome wide association analyses in two different commercial aquaculture lines that were naïve to previous exposure to IHNV to determine whether QTL were shared across lines, and to investigate whether there were major effect loci that were still segregating in the naïve lines. A total of 1,859 and 1,768 offspring from two commercial aquaculture strains were phenotyped for resistance to IHNV and genotyped with the rainbow trout Axiom 57K SNP array. Moderate heritability values (0.15–0.25) were estimated. Two statistical methods were used for genome wide association analyses in the two populations. No major QTL were detected despite the naïve status of the two lines. Further, our analyses confirmed an oligogenic architecture for genetic resistance to IHNV in rainbow trout. Overall, 17 QTL with notable effect (≥1.9% of the additive genetic variance) were detected in at least one of the two rainbow trout lines with at least one of the two statistical methods. Five of those QTL were mapped to overlapping or adjacent chromosomal regions in both lines, suggesting that some loci may be shared across commercial lines. Although some of the loci detected in this GWAS merit further investigation to better understand the biological basis of IHNV disease resistance across populations, the overall genetic architecture of IHNV resistance in the two rainbow trout lines suggests that genomic selection may be a more effective strategy for genetic improvement in this trait.

Enhancement of southern flounder (Paralichthys lethostigma) sperm velocity and fertility by direct activation of sperm epidermal growth factor and adenylyl cyclase signaling pathways

Development of southern flounder commercial aquaculture is limited by poor sperm motility and fertility, but the underlying causes and mechanisms regulating sperm motility are unknown. In vitro treatment of southern flounder sperm with progestin hormones rapidly increases velocity (swimming speed), a major component of sperm motility, and also fertility through membrane progestin receptor alpha (mPRα) and increases in Acy (adenylyl cyclase)/cAMP signaling. The progestin-induced increase of Atlantic croaker sperm velocity is also mediated through epidermal growth factor receptor (Egfr) transactivation resulting in extracellular-regulated kinase 1/2 (Erk1/2) signaling, but its role in regulating southern flounder sperm velocity is unknown. The hypotheses that Egfr transactivation and Erk1/2 signaling mediates progestin-induced increases in sperm velocity and calcium levels in southern flounder, and that direct stimulation of sperm Egfr/Erk1/2 and Acy/cAMP pathways increases sperm fertility in strip spawning trials were tested in the present study. Golf, Egfr, Erk1/2, and protein kinase A were detected on southern flounder sperm by immunocytochemistry. Involvement of Egfr/Erk1/2 signaling in increasing sperm velocity and calcium levels was investigated by preincubating sperm with two EGFR inhibitors, AG1478, and AG825, a matrix metalloproteinase inhibitor, Ilomastat, and two ERK1/2 inhibitors, PD98059, and U0126, for 30 min prior to 1 min treatment with progestins. Sperm swimming speed was observed under a microscope after activation with a hyperosmotic medium and recorded for 1 min. Whereas treatment with the inhibitors did not alter basal sperm velocity and calcium levels, all of them significantly attenuated velocity and calcium increases in response to progestins. Treatment in vitro for 1 min with the Egfr agonist, recombinant human EGF (0.1 nM and 100 nM), mimicked the stimulatory effects of progestins on sperm velocity, the increase in calcium, and also fertility in a strip spawning trials with ovulated southern flounder eggs. The results suggest Egfr/Erk1/2 signaling is involved in sperm velocity and fertility responses to progestins in southern flounder. Direct in vitro treatment with the Acy activator, forskolin (10 μM) for 1 min mimicked the stimulatory action of progestins on sperm calcium levels, velocity, and fertility, suggesting that Acy/cAMP signaling also mediates increased sperm swimming speed and fertility. Importantly, both EGF and forskolin also directly stimulated southern flounder sperm velocity and fertility at the end of the reproductive season when sperm were no longer responsive to progestins. The results suggest that EGF and forskolin could potentially be used as pharmacological agents to enhance reproductive performance of male southern flounder broodstock.

Development and evaluation of the inactivated vaccine against Aeromonas schubertii in hybrid snakehead

Aeromonas schubertii is a global pathogen of fish that causes internal nodular diseases and death in severe cases. In the present study, we isolated nine pathogenic strains from diseased hybrid snakehead (Channa argus♂ × Channa maculata♀) which were formally identified as A. schubertii by molecular identification, including 16S rRNA and gyrB gene sequencing. Subsequently, we determined the extracellular enzyme activity of six isolates and compared pathogenicity to investigate potential relationships with geographic characteristics. Two highly pathogenic strains, AS-14 and AS-16, were finally identified as inactivated vaccine candidates. Subsequently, we evaluated different concentrations of the antigen for each vaccine by performing experiments in hybrid snakehead; fish were inoculated with inactivated vaccine via the intraperitoneal route and a booster injection was administered 14 days later. Relative percent survival (RPS) for fish immunized with high doses of the two vaccines (1 × 108 CFU/fish) were 54.4% and 57.4%, respectively; these values were higher than other groups of fish receiving different doses (1 × 106 and 1 × 107 CFU/fish). The bacterial load of tissues harvested from the fish immunized with high doses of the vaccine groups were significantly lower than those of fish immunized with low doses. Titers of specific antibodies against A. schubertii in fish immunized with a high dose of AS-16 vaccine were maintained at high levels when tested six weeks after the booster injection. Furthermore, immune-related genes in the spleen, including MHC I, MHC II, CD4, CD8α and IgM, all exhibited high levels of expression, thus suggesting that hybrid snakehead underwent an adaptive immune response post-vaccination. Collectively, our findings enrich our understanding of the specific causative agents underlying nodular diseases in hybrid snakehead and provide scientific reference guidelines for the development of vaccines against A. schubertii. The development of such vaccines will be conducive to the sustainable development of hybrid snakehead in commercial aquaculture.

Assessing selection potential for Pacific oyster (Crassostrea gigas) to a North American OsHV-1 μvar: Comparing two experimental assay methods

Ostreid Herpesvirus - 1 is a global threat to Pacific oyster aquaculture as catastrophic farm losses have occurred where OsHV-1 microvariants (μvars) become endemic. Genetic selection has proven effective for increasing survival to OsHV-1 exposure in multiple Pacific oyster breeding programs, but a novel μvar detected in San Diego Bay California has the potential to severely impact commercial aquaculture along the North American Pacific coast as no OsHV-1 μvar selected population is available commercially. Using a Molluscan Broodstock Program population descending from imports originating in the Miyagi prefecture (Japan), we evaluated the potential for genetic selection to improve survival against the San Diego Bay OsHV-1 μvar using a laboratory trial. Two experimental methods were evaluated, a common garden method where all 15 full-sibling families were pooled, and a petri dish method, where each family was confounded within replicate. No striking differences were found between experimental methods, and both experiments displayed high heritability (0.69 and 0.64) of the tested trait, a broad range of estimated breeding values, and a moderate prediction accuracy for the survival trait. A significant genotype-by-environment interaction occurred, highlighting variability in laboratory-based disease challenges, but the two experiments yielded similar results with minor to moderate re-ranking of mean family survival. Overall, the Miyagi population of Pacific oysters curated by the Molluscan Broodstock Program is a promising population to use for artificial selection to increase survival against San Diego Bay OsHV-1 μvar in attempts to obviate farm losses if the virus expands its currently isolated range.

Rapid on-site authentication of three cockle species using real-time loop-mediated isothermal amplification (LAMP) assay

Cockles, crucial for commercial fishing and aquaculture, emerge as highly marketable shellfish. However, the morphological similarities among various cockle species are frequently exploited for shellfish fraud. In this study, we developed LAMP assays (end-point LAMP and real-time LAMP) to detect three commercially important cockle species: Tegillarca granosa, Anadara broughtonii, and Anadara kagoshimensis. The primers were designed based on mitochondrial DNA and used to develop LAMP assays. Exclusivity was assessed using 17 shellfish species, and no cross-reactivity was confirmed. The estimate of the detection limit was determined to be 0.0001 ng. The established assays were further refined to create an on-site real-time LAMP assay by combining fast DNA isolation with a real-time fluorometer for field application. The effectiveness of the on-site real-time LAMP assay was successfully verified using 38 commercial samples, with analysis accuracy confirmed using a DNA extraction kit. Consequently, the on-site real-time LAMP assay can complete the whole procedure, including DNA extraction, real-time LAMP reaction, and data analysis within a 45 min timeframe, maintaining the accuracy of detecting three cockle species from processed cockle samples in the field. Our method proves valuable for detecting shellfish fraud by identifying cockle species using samples of both raw and processed products in the field.

Sampling time for different matrices in stress assessment of farmed Atlantic salmon post-smolt

The sustainability of commercial aquaculture production depends critically on prioritizing fish welfare management. Besides monitoring welfare parameters such as fish behaviour and water quality, fish stress level can also provide a reliable measure of the welfare status of farmed fish. Cortisol and 5 of its metabolites (5β-THF, cortisone, 5β-DHE, 5β-THE, β-cortolone) were previously identified by the authors as suitable stress biomarkers of farmed Atlantic salmon. Based on this knowledge, the present study aimed to investigate the time-related dynamics of these metabolites in plasma, skin mucus, bile and faeces over a 72 h- period. The objective was to determine the optimal sampling time for each matrix and to understand the clearance pathway of these metabolites following stress. An experiment was carried out using a total of 90 Atlantic salmon with an average weight of 438 (±132) g. The average sea temperature was 6.9 °C during the experimental period. A control group of 10 fish was first collected before the remaining 80 fish were submitted to a stress of netting and subsequent relocation into two separate cages. From each of these two stress groups, 10 fish were sampled at 1 h, 2 h, 4 h, 6 h and 12 h, 24 h, 48 h, 72 h after the stress event respectively. The concentrations of cortisol and its metabolites were measured at each of the sampling timepoint. The results demonstrated that plasma cortisol metabolites reached the highest concentration 4 h after stress and remained elevated despite the slight decrease for the remaining timepoints. The peak level was observed at 12 h post-stress in skin mucus and 24 h in bile and faeces. The findings suggest that these timepoints are the optimal for sampling Atlantic salmon post-smolt following stressful events in acute stress studies. Furthermore, the results reveal that analysing cortisol and its metabolites, both in free and conjugated forms, rather than free cortisol provides greater flexibility as their concentrations are less affected by sampling procedure. This study confirms the appropriateness of skin mucus and faeces as less-invasive sample matrices for fish stress evaluation and provides a basis for further developing low invasive tools for monitoring the welfare of farmed salmonid.

Assessing methods for detecting Alexandrium catenella (Dinophyceae) and paralytic shellfish toxins in Southeast Alaska.

Blooms of Alexandrium catenella threaten to disrupt subsistence, recreational, and commercial shellfish harvest in Alaska, as the paralytic shellfish toxins (PSTs) produced pose a serious public health risk and can lead to costly shutdowns for shellfish farmers. Current methods of PST detection in the region range from monitoring programs utilizing net tows to detect A. catenella to direct shellfish tissue testing via mouse bioassay (MBA) for commercial aquaculture harvest, as well as various optional testing methods for subsistence and recreational harvesters. The efficacy and feasibility of these methods vary, and they have not been directly compared in Southeast Alaska. In this study, we sought to assess and compare A. catenella and PST early detection methods to determine which can provide the most effective and accurate warning of A. catenella blooms or PST events. We found microscope counts to be variable and prone to missing lower numbers of A. catenella, which may be indicative of bloom formation. However, quantitative polymerase chain reaction (qPCR) significantly correlated with microscope counts and was able to effectively detect even low numbers of A. catenella on all sampling days. Paralytic shellfish toxin concentrations measured by enzyme-linked immunosorbent assay and MBA significantly correlated with each other, qPCR, and some microscope counts. These results show that qPCR is an effective tool for both monitoring A. catenella and serving as a proxy for PSTs. Further work is needed to refine qPCR protocols in this system to provide bloom warnings on an actionable timescale for the aquaculture industry and other shellfish harvesters. Sensory profiles and market potential of burbot Lota lota maculosa- An emerging aquaculture species

The aquaculture industry needs to increase the diversity of fish species to meet consumers' escalating demands. Burbot (Lota maculosa) has emerged as a promising candidate for such diversification. Nonetheless, a dearth of knowledge exists concerning their sensory characteristics and overall consumer acceptance. This study sought to investigate these crucial aspects, which are instrumental in assessing the market potential of this fish species. A consumer survey was conducted in Idaho from March through July 2017 to initially evaluate burbot's consumer acceptance and market potential. Results showed that over 96% of consumers liked the texture of burbot, 92% liked the flavor of the burbot, and 90% would try burbot again. Significant reasons for consumers' willingness to eat burbot included cooking preparation, tasty, mild flavor, excellent texture, and excellent flavor. Results from this indicate that burbot is a promising food fish candidate for commercialization. Based on these results, a follow-up panelists survey and sensory analysis were conducted in April 2019 to further investigate consumer acceptance and sensory properties of burbot when compared to other prominent aquaculture species (trout and tilapia). The three species exhibited different sensory profiles; however, burbot was ranked significantly higher for aroma, flavor, and taste. Conversely, panelists found tilapia and trout to be significantly (P < 0.05) firmer than burbot for firmness. The results from the sensory testing showed that 87% and 81% of panelists preferred burbot to tilapia and trout, respectively, and 79% would be willing to try this as a new species on the market. The distinct profiles of burbot indicate that this fish species would be highly marketable and if available would contribute to the diversification of U.S. commercial aquaculture by offering a delicate white fillet option to salmonids.

Screening of temperature-responsive signalling molecules during sex differentiation in Asian yellow pond turtle (Mauremys mutica)

BackgroundThe Asian yellow pond turtle (Mauremys mutica) is an important commercial freshwater aquaculture species in China. This species is a highly sexually dimorphic species, with males growing at a faster rate than females and exhibits temperature-dependent sex determination (TSD), in which the incubation temperature during embryonic development determines the sexual fate. However, the mechanisms of the sex determination or sex differentiation in the Asian yellow pond turtle are remain a mystery.ResultsTemperature-specific gonadal transcriptomics of the Asian yellow pond turtle were performed during the thermosensitive period (stage 15) using RNA-seq technology to identify candidate genes that initiate gonadal differentiation. We uncovered candidates that were the first to respond to temperature. These candidates were sexually dimorphic in expression, reflecting differences in gonadal (Cirbp, Runx1) and germline differentiation (Vasa, Nanos1, Piwil2), gametogenesis (Hmgb3, Zar1, Ovoinhibitor-like, Kif4), steroid hormone biosynthesis (Hsd17b5, Hsd17b6), heat shock (Dnajb6, Hsp90b1, Hsp90aa1) and transient receptor potential channel genes (Trpm1, Trpm4, Trpm6, Trpv1).ConclusionsOur work will provide important genetic information to elucidate the mechanisms of sex control in the Asian yellow pond turtles, and will contribute important genetic resources for further studies of temperature-dependent sex determination in turtles.

Effects of manipulating Kingfish (Seriola lalandi) routine oxygen demand and supply on ventricular and skeletal muscle mitochondrial function

SummaryYellowtail kingfish (Seriola lalandi) are a highly active marine species, with increasing value in commercial aquaculture. While practices which manipulate oxygen (O2) dynamics through metabolic demand and environmental O2 supply have been shown to impact juvenile growth rate in fish development, there has been no work exploring the effects of O2 at the subcellular level. In addition, manipulation of O2 supply or demand via changes in environmental factors such as O2 levels or salinity, may impact whole animal performance. Still, there has been little work exploring the physiological effects at a subcellular level. Mitochondria (mt) consume O2 and are pivotal in central metabolism, directly impacting growth, and dictating whole animal performance. Here we explore the effects of manipulating O2 demand and supply on mt in yellowtail kingfish ventricle and skeletal muscle by exposure to chronic hyperoxia (200% [O2], 35 ppt) and to hypoosmotic conditions (100% [O2], 17.5 ppt). Chronic exposure to hypoosmotic conditions resulted in greater overall biomass, while it lowered relative ventricular mass compared to control fish. Mitochondrial respiratory flux for most measures remained consistent with control fish, with the exception of cytochrome c oxidase (COX) respiration. Chronic hyperoxia had no impact on overall morphology, and no significant effect on ventricle mt respiration, yet increased COX activity in skeletal muscle. These data indicate that lowering routine mt O2 demands in S. lalandi with hypoosmotic conditions has more impact than increasing O2 availability, and that this may enhance biomass output while maintaining mt integrity.

Supply Chain-Based Coral Conservation: The Case of Mozuku Seaweed Farming in Onna Village, Okinawa

Blue foods, including seaweed, have been overlooked in food systems analysis and policy-making due to a lack of available data. However, seaweed cultivation is gaining attention as a restorative aquaculture that could contribute to ocean health by serving as blue carbon or nurturing seagrass beds. Commercial restorative aquaculture may provide market-based solutions for improving ocean health. The Onna Village Fisheries Cooperative producers have been restoring corals, knowing empirically that when the coral weakens, the yield of Mozuku seaweed drops. Furthermore, measures taken to reduce red soil run-off prevent the reduction in Mozuku quality and quantity and protect seagrass beds, since Mozuku cultivators have continued to use them as an important nursery. The fishery cooperative and the seaweed processing company, Igeta Takeuchi Co., Ltd., have jointly developed high-quality Mozuku that is resistant to climate change and extreme weather conditions through strain selection. Based on this case, this paper examines the following questions: (1) What quality assurance technique is necessary to continue restorative aquaculture as a market-based solution over the long term? (2) What social platforms and communication channels are available to stakeholders to maintain restorative aquaculture in the long run? To answer these questions, Japanese consumer cooperatives that established “the Mozuku Fund” are also examined. This case study of Mozuku highlights how the sustainability and quality of marine products are maintained throughout the whole supply chain, focusing on the power of the Japanese consumers’ cooperative.

Phenotypic plasticity and carryover effects in an ecologically important bivalve in response to changing environments

Phenotypic plasticity can improve an organism’s fitness when exposed to novel environmental conditions or stress associated with climate change. Our study analyzed spatiotemporal differences in phenotypic plasticity and offspring performance in Olympia oysters Ostrea lurida. This species is an ecosystem engineer and is of great interest for commercial and restoration aquaculture. We used a multidisciplinary approach to examine acute and long-term physiological differences in O. lurida in response to in situ oceanographic conditions in a dynamic inland sea. We outplanted oysters to different areas in Puget Sound, Washington, affixing cages to anchor lines of oceanographic monitoring buoys. This allowed us to couple high-resolution oceanographic data with organism’s phenotypic response. To assess spatiotemporal differences in oyster physiological performance, we collected oysters after six-months and one year of acclimatization at four field sites. During each collection period we evaluated changes in shell properties, diet, metabolism, and reproduction. Adult growth, δ13C and δ15N isotopic signatures, and gametogenesis were affected by both seasonal and environmental conditions. In the winter, oysters from all sites had higher respiration rates when exposed to acute thermal stress, and lower respiration response to acute pH stress. Lipid content, sex ratio and shell strength were unchanged across locations. Offspring growth rates between sites at experimental temperature 20°C closely reflected parental growth rate patterns. Offspring survival was not correlated with growth rates suggesting different energetic trade-offs in oyster offspring. The metabolic response (respiration) of larvae reached its highest point at 20°C but sharply decreased at 25°C. This indicates that larvae are more sensitive to temperature stress, as adults did not exhibit a reduction in metabolic response at 25°C. By deploying genetically similar oysters into distinct environments and employing a wide range of physiological methodologies to examine performance and fitness, our results indicate that Olympia oysters exhibit a high degree of phenotypic plasticity and show evidence of parental carryover.

Early ontogenesis from embryo to juvenile in Senegalese sole Solea senegalensis under laboratory conditions.

Senegalese sole, Solea senegalensis, is a flatfish of high commercial value in the world. It has been identified as an interesting and promising species for marine commercial aquaculture diversification in Europe for at least four decades and was introduced to China in 2003. Early ontogenesis from embryo to juvenile stages in S. senegalensis was analysed under controlled laboratory conditions to provide morphological information for aquaculture. From 0 to 59 days post hatching (dph), 10-20 larvae were sampled and measured each day (0-17 dph) or every 2-6 days (17-59 dph). Morphological characteristics from the egg to the juvenile stage were described. The eggs were separate and spherical with multiple oil globules. After 3 dph, the yolk sac was completely absorbed, mouth and anus were open, a swim bladder appeared, and larvae began feeding on rotifers (Brachionus plicatilis). The larvae began metamorphosis as the notochord flexed upward and the left eye migrated upward after 10 dph. The left eye migrated to the dorsal midline at 15 dph. At 19 dph, the left eye was translocated to the right-ocular side, and the juveniles adopted a benthic lifestyle. The swim bladder degenerated, and the juveniles completed metamorphosis at 23 dph. The growth patterns of some parameters (TL, SL, BH, BW) during larval and juvenile development stages were identified. The inflection points, which are slopes of growth changes, were calculated in growth curves. Three inflection points occurring in the growth curves of larvae and juveniles were found to be associated with metamorphosis, weaning, and transitions in feeding habits. The basic information of embryo development and ontogenesis in this study represents a valuable contribution to the S. senegalensis industry, especially in artificial breeding and rearing techniques.

Examination of the effects of impaired spermatogenesis on sterility in triploid oysters (Crassostrea gigas), and implications for commercial aquaculture

Sterility caused by triploidization holds promising commercial implications in aquaculture. In this study, we conducted a thorough investigation into the spermatogenesis of triploid male oysters (Crassostrea gigas) to evaluate their fertility. Histological analysis and immunostaining revealed the presence of numerous immature spermatocytes and a markedly low abundance of mature spermatozoa in triploid males, indicating a robust blockade in spermatogenesis. Ultrastructural analysis and apoptosis analysis demonstrated that triploid males underwent normal spermatogonial mitosis during early spermatogenesis, like diploids. Analysis of phosphorylated histone H2AX (γ-H2AX) displayed heightened γ-H2AX staining in developed triploid male gonads, with a significantly higher number of apoptotic cells confirmed by the TUNEL assay compared to diploid counterparts. These findings suggest a potential arrest at the zygotene stage, preventing the completion of the zygotene-pachytene transition and ultimately leading to cell apoptosis. Flow cytometry results confirmed aneuploidy in spermatozoa from triploid oysters, containing approximately 1.5 times the DNA content of spermatozoa from diploid oysters. Notably, the sperm head size and flagella length in triploid oysters surpassed those in their diploid counterparts. Occasional observations of two-tailed spermatozoa in triploid males were noted. This study provides novel insights into the blocked spermatogenesis in triploid oysters, emphasizing the sterility of triploid male oysters and its relevance in aquaculture practices.
