Pre-Treatment, Extraction Solvent, and Color Stability of Anthocyanins from Purple Sweetpotato.

Purple sweetpotato anthocyanins (PSPA) exhibit significant potential as food colorants with associated health benefits. However, challenges related to browning and instability have hindered the application of PSPA. In this study, various pre-treatments and solvents for PSPA extraction were evaluated based on color, anthocyanin yields, antioxidant capabilities, and brown index. Browning markedly influenced the color and reduced the antioxidant capacity. Optimal results were obtained with the pre-treatment of "steaming of unpeeled whole sweetpotato" and the solvent "1% citric acid-ddH2O". Furthermore, the color stability of purified PSPA solutions was evaluated under pH levels from 1 to 13 at 25 °C and 65 °C. The PSPA solutions showed a color spectrum from magenta, blue/green, and then to yellow across the pH range. The blue/green hues at pH 10-12 rapidly degraded, while the magenta hue at lower pH showed higher color stability. Elevated temperatures significantly accelerated the PSPA degradation. However, PSPA solutions at pH 1-2 exhibited remarkable color stability, with no spectral decay at either 65 °C for 12 h or 25 °C for 32 days. These results provide valid guidance for the extraction, preservation, and application of PSPA in the food industry.

The structure of anthocyanins and the copigmentation by common micromolecular copigments: A review

Under natural physiological conditions, anthocyanins can keep bright and stable color for a long time due to the relatively stable acid-base environment of plant vacuoles and the copigmentation from various copigment substances, such as polyphenols, nucleotides, metallic ions and other substances. Therefore, the copigmentation caused by copigments is considered an effective way to stabilize anthocyanins against adverse environmental conditions. This is attributed to the covalent and noncovalent interactions between colored forms of anthocyanins (flavylium ions and quinoidal bases) and colorless or pale yellow organic molecules (copigments). These interactions are usually manifested in both hyperchromic effect and bathochromic shifts. In addition to making anthocyanins more stable, the copigmentation also could make an important contribution to the diversification of their tone. Based on the molecular structure of anthocyanins, this review focuses on the interaction mode of auxochrome groups or copigments with anthocyanins and their effects on the chemical and color stability of anthocyanins.

The Stability of Anthocyanins and Their Derivatives through Clay Minerals: Revising the Current Literature

In recent years, anthocyanins, natural dyes, have promoted great scientific and technological interest. Their intrinsic antioxidant properties and health benefits make them ideal representatives of natural dyes as replacements for synthetic dyes. However, their instability can limit their use. A promising strategy to increase the color stability of anthocyanins is their interaction with clay minerals structures. Clay minerals have characteristics favorable to the stabilization of natural dyes, such as adsorption capacity, good surface chemistry, ion exchange capacity, abundance, non-toxicity, and environmental compatibility. This review summarizes relevant works that present different clays or clay minerals as robust inorganic matrices for incorporation, improved stability, and increased resistance against the thermal- and photodegradation of anthocyanins. In addition, several authors propose different applications for the formed anthocyanin-based hybrids.

Novel color stability and colorimetry-enhanced intelligent CO2 indicators by metal complexation of anthocyanins for monitoring chicken freshness

This study aims to improve the color stability of anthocyanins and develop a CO2-sensitive indicator based on black goji anthocyanin (BGA) extract. Although the BGA extracts showed distinct color changes, such as red–purple–blue, their intrinsic color diminished after 24 h. A metal complexation method was used for the high color stability of BGA. BGA extracts were chelated with various concentrations of Al3+ [0 – 20% (w/w)]. It showed high color stability and strong intensity in a dose-dependent manner. A CO2-sensitive indicator sachet was developed using hydroxypropyl methylcellulose hydrogel, based on 5% (w/w) Al3+-BGA complexes. The indicator was applied to the chicken breast and detected its spoilage after 3 days with its changing color to greyish blue, due to the microbial growth to 7.00 log CFU/g. These results demonstrated the possibility of chelated anthocyanin complexes as indicating dyes and the ability to monitor the food quality changes through noticeable color changes.

Recovery of phenolic compounds from purple onion peel using bio‐based solvents: Thermal degradation kinetics and color stability of anthocyanins

The aim of this study was to evaluate the influence of bio-based solvent (ethanol, water, and acetic acid) on the extraction of phenolic (TPC), anthocyanins (TAC), and flavonoids (TFC) from purple onion peel and also analyze the effects of storage temperature on the kinetics of anthocyanin degradation and color stability of the phenolic-rich extract obtained. The purple onion extract obtained with 60% ethanol (E60E) showed, respectively, TPC, TAC, and TFC about 3.4, 2.5, and 4.45 times higher than the other solvents used. E60E showed higher antioxidant and antibacterial potential compared to other solvents. E60E was stored at 4, 25, and 38°C, a half-life of 285, 48.81, and 21.80 days was obtained, respectively. The degradation of anthocyanins in E60E occurred by an endothermic and non-spontaneous reaction with activation energy of 54.65 kJ mol−1, being strongly related to the thermal sensitivity of the phenolic compound. Novelty impact statement Purple onion peel is a food waste rich in bioactive compounds, being a low-cost alternative for obtaining extracts with coloring and functional potential. The results of this study indicate that the purple onion peel is an excellent source for obtaining a phenolic-rich extract with high antioxidant and antimicrobial activity. The anthocyanin, color, and stability index indicates that the extract can applied in several biosectors (e.g., pharmaceutical, cosmetic, and food industries).

Improved Stability of Blue Colour of Anthocyanins from Lycium ruthenicum Murr. Based on Copigmentation.

Natural blue food colourant is rare. The aim of this work was to screen compounds from the common copigments that could improve the blue tones of anthocyanins (ACNs) and to investigate the effect of different copigments on the colour stability of anthocyanins in neutral species. International Commission on Illumination (CIE) colour space, UV, IR, NMR, atomic force microscopy (AFM) and computational chemistry methods were utilised to evaluate ACNs from Lycium ruthenicum Murr. (LR), which is complexed with food additives and biological agents. The results indicate that Pro−Xylane (PX), Ectoin (ECT) and dipotassium glycyrrhizinate (DG) enhance the blue colour of the ACNs. ACNs−PX presents a colour close to Oxford Blue and has a surface height of 2.13 ± 0.14 nm and slightly improved stability. The half−life of ACNs−DG is improved 24.5−fold and had the highest complexation energy (−50.63/49.15) kcal/mol, indicating hydrogen bonds and π−π stacking forces enhance stability. These findings offer a new perspective for anthocyanin utilisation as a blue colourant and contribute to the large−scale application of LR.

Computational design of a chitosan derivative for improving the color stability of anthocyanins: Theoretical calculation and experimental verification

The objective of this study was to design a chitosan (CS) derivative with good protective effect on the color stability of anthocyanins (ACNs) under accelerated storage. The binding affinities and interactions of 12 organic acids with cyanidin-3-O-glucoside (C3G) were evaluated using quantum mechanics method. Sinapic acid (SinA) showing the strongest interaction with C3G was selected for the synthesis of SinA-grafted-CS (SinA-g-CS), which was further characterized by FTIR and 1H NMR. Under accelerated storage conditions (40 °C), SinA-g-CS significantly improved the color stability of black rice anthocyanins (BRA) in the presence of l-ascorbic acid (pH 3.0), and showed a better protective effect than that of CS. Moreover, molecular dynamics simulation analysis showed SinA-g-CS formed more hydrogen bonds with C3G than CS. Our study demonstrated that SinA-g-CS designed by computational methods can effectively protect ACNs from degradation, and has the potential to be used in ACN-rich beverages as a replacement for CS.

Protective Effect and Mechanism of Soybean Insoluble Dietary Fiber on the Color Stability of Malvidin-3-O-glucoside.

Anthocyanins have great health benefits, especially malvidin. Vitis amurensis Rupr are rich in malvidin, and malvidin-3-O-glucoside (Mv3G) monomer is the most abundant. However, natural anthocyanins are unstable, which limits their wide application in the food field. Soybean insoluble dietary fiber (SIDF) has high stability, and it can be used as an inert substrate to construct a stable system, which may improve the stability of anthocyanins. The optimal condition to construct a stable system of SIDF and Mv3G at pH 3.0 was determined by an orthogonal experiment. The results indicated that SIDF effectively improved the stability of Mv3G under different pH values (1.0~7.0), high temperature (100 °C for 100 min), and sunlight (20 ± 2 °C for 30 d) conditions. The absorption peak intensity of the UV–VIS spectrum of SIDF-Mv3G was enhanced, which indicated that there was interaction between SIDF and Mv3G. Fourier transform infrared spectroscopy analyses revealed that the -OH stretching vibration peak of SIDF-Mv3G was changed, which indicated that the interaction between SIDF and Mv3G was due to hydrogen bonding. X-ray diffraction analysis showed that the crystalline morphology of SIDF was opened, which was combined with Mv3G, and SIDF made Mv3G change to a more stable state. Scanning electron microscope analysis showed that SIDF and Mv3G were closely combined to form an inclusion complex. Overall, this study provides valuable information for enhancing the color stability of anthocyanins, which will further expand the application of anthocyanins in the food field.

Analyzing the Interaction between Anthocyanins and Native or Heat-Treated Whey Proteins Using Infrared Spectroscopy.

The color stability of anthocyanins (ACN) has been shown to be improved by interaction with whey proteins (WP). In this study, we explore the ACN–WP interaction using Fourier transform infrared spectroscopy (IR). ACN from purple corn, grape, and black carrot (50 μM) were evaluated. IR spectra (4000–700 cm−1) were collected for native and preheated (40–80 °C) WP (5 mg/mL) and ACN–WP mixtures at pH 7.4. Soft independent modeling of class analogy was used to analyze the IR data. The WP secondary structure changed after heat treatments and after interaction with ACN. As expected, the WP α-helices decreased, and β-sheet increased after heat treatment. The intensities of the WP amide I and II bands decreased after ACN addition, revealing a decrease in the WP α-helix content. Higher preheating temperatures (70–80 °C) resulted in a more disordered WP structure that favored stronger WP–ACN interactions related to amide III changes. Addition of ACN stabilized WP structure due to heat denaturation, but different ACN structures had different binding affinities with WP. WP structure had less change after interaction with ACN with simpler structures. These results increase our understanding of ACN–WP interactions, providing a potential strategy to extend anthocyanin color stability by WP addition.

Improved color stability of anthocyanins in the presence of ascorbic acid with the combination of rosmarinic acid and xanthan gum

This study investigated the protective effect and mechanism of action of combined use of rosmarinic acid (RA) and xanthan gum (XG) on the stability of anthocyanins (ACNs) in the presence of l-ascorbic acid (pH 3.0). The addition of RA and XG, alone and in combination, significantly enhanced the color stability of ACNs, and the combined use of RA and XG showed the best effect. FTIR, 1H NMR, AFM and computational molecular simulation analyses revealed that the improvement in ACN stability following the combined addition of RA and XG was due to intermolecular interactions such as hydrogen bonding and van der Waals forces. In the ACN-RA-XG ternary complexes, XG had stronger binding interactions with ACNs than RA. Our findings provide a valuable potential to enhance the stability of ACNs in the presence of ascorbic acid with the combined use of RA and XG.

Monitoring the Interaction between Thermally Induced Whey Protein and Anthocyanin by Fluorescence Quenching Spectroscopy.

The color stability of anthocyanins was shown to improve with addition of whey proteins (WP). The goal of this study was to investigate the binding mechanisms of purple corn, grape and black carrot anthocyanin extracts to native and preheated WP (40–80 °C, 3.6 μM) at a pH of 3 using fluorescence quenching spectroscopy. The fluorescence spectra were collected with an excitation wavelength of 280 nm at 25 °C, 35 °C and 45 °C. The quenching data were analyzed by using the Stern–Volmer equation. The fluorescence intensity of WP decreased (up to 73%) and its λmax increased (by ~5 nm) with increasing anthocyanin concentration (0–100 μM). The quenching data showed that the interaction between anthocyanin extracts and WP was a static quenching process. Thermodynamic analysis showed their binding was mainly through hydrophobic interactions. Their binding affinity was higher for preheated WP than native WP and decreased gradually with increasing preheating temperature. Black carrot anthocyanin extract had the lowest binding affinity with WP, likely due to the larger molecular structure. These results help better understand the protection mechanism of native and preheated WP on anthocyanin color stability, expanding the application of anthocyanins as food colorants that better withstand processing and storage.

Color Stability of Anthocyanins Copigmentation from Red Rice (Oryza sativa L.) Bran by Spectrophotometry UV-Vis

Red Rice (Oryza sativa L.) Bran is a waste that is often found in our environment. Pigment rice contains polar pigment anthocyanins (ACNs). Anthocyanins are a natural pigment that has low color stability and are easily degraded by environmental influences. In the present study, we have tried to increase the color stability of anthocyanins by copigmentation. Anthocyanins extraction was carried out using maceration techniques in 95% ethanol solvents. The extract of anthocyanins was characterized by spectrophotometer UV-Vis and Fourier transform infrared (FTIR). The results show that the spectrum UV-Vis and IR were obtained a characteristics peak at 470, 420, 340 nm and a hydroxyl group at 3400-3500 cm-1, respectively. The various of Fe3+ and Al3+ as copigment and composition of anthocyanins to Fe3+ and Al3+ have been investigated. Anthocyanins stability was followed by measuring the loss of color by spectrophotometer UV-Vis, and the results show that Addition of Fe3+ and Al3+ could improve the anthocyanins stability.

Stabilization of Black Rice (Oryza Sativa, L. Indica) Anthocyanins Using Plant Extracts for Copigmentation and Maltodextrin for Encapsulation.

Black rice (Oryza sativa L. indica) is one of the cereal commodities considered to be a source of anthocyanin. One of the obstacles of using anthocyanins as natural food colorants is their low stability. The objective of this study was to determine the effect of natural plants extracts on anthocyanin copigmentation and to study the stability of the encapsulated copigmented anthocyanin during storage under different temperatures and light exposure. Dried bilimbi leaves (BL) and sow thistle leaves (STL) were extracted using ethanol to obtain the crude flavonoid extract that was subsequently purified using solid phase extraction. Both crude (CBL, CSTL) and purified (PBL, PSTL) extract were used as copigment agents with the molar ratio of 1:0.1 to 1:10 at pH 1 and pH 3. The copigmentation effect was evaluated by measuring the wavelength and absorbance shifts at wavelengths of 500 to 600 nm. The stability of the dried anthocyanin powder was evaluated under different temperatures and light exposure for up to 4 weeks. Anthocyanin was successfully copigmented with BL and STL extracts showing a wavelength shift and absorbance increase at pH 1 and pH 3. Purification of BL enhanced the hyperchromic effect more than that of the crude form, in contrast to the STL. Meanwhile, the retention of absorbance (%) for encapsulated copigmented anthocyanin samples was within 74% to 83% at the end of the storage period. PRACTICAL APPLICATION: Natural food pigment such as anthocyanin obtained from plant extract usually shows very limited stability under various processing conditions. This study shows that color stability of anthocyanin extracted from black rice can be improved by copigmentation with flavonoids extracted from particular plants. Encapsulation of the copigmented anthocyanin could further enhance its color stability. The availability of encapsulated and copigmented anthocyanins with improved color stability is expected to provide more alternatives for especially the beverage industry to choose suitable natural colorant for their products.

Anthocyanin pigment stability of Cornus mas-Macrocarpa under treatment with pH and some organic acids.

The use of colors in food industry is essential for the creation of new products or their improvement. As an important pigment group, anthocyanin could be used as a natural coloring pigment in foods. This study aims at exploring strategies that result in color stability of anthocyanin in pear‐shaped variety of blueberry (Cornus mas–Macrocarpa). In this study, the effects of different pH values (1, 2, 3, 4) as well as various concentrations (0, 120, 240, 480, 960 mg/L) of five copigments, including tannic, caffeic, benzoic, and coumaric acids, on anthocyanin copigment complexes (ratio 1:1) were investigated. The studied copigments were tannic, caffeic, benzoic, and coumaric acids. Anthocyanin was influenced by the highest concentration of 960 mg/L copigment in the presence of different pHs. Five groups were considered, one of which contained anthocyanin without copigment and the rest consisted of copigments. To evaluate the response of copigmentation through spectrophotometer, absorbance from samples was measured after 30 min of adding copigment to anthocyanin in the range of 450–600 nm wavelengths. The results showed that caffeic acid possessed the greatest anthocyanin stability compared to other copigments and it was the best copigment. An increase in the concentrations of copigments led to a higher level of anthocyanin stability and changes in hyperchromic and bathochromic. Moreover, the results revealed that the strongest hyperchromic effect for all organic acids was observed in pH 2, and the strongest bathochromic changes were observed in pH 4.

Anthocyanin condensed forms do not affect color or chemical stability of purple corn pericarp extracts stored under different pHs

Purple corn is rich in anthocyanins, some of which are condensed with flavanols. The aim was to determine the impact of anthocyanin condensed forms extracted from purple corn pericarp on color and chemical stability at different pHs compared with the complete extract, and an extract without condensed forms. Extracts were dissolved at pH values ranging from 2.0 to 6.0 and stored for 12weeks at 22°C. Color stability of anthocyanins decreased as the pH increased. Slight color differences were observed throughout time at pH 2 (ΔE from 0.2 to 3.6). After 12weeks, pH 6 caused substantial changes in color (ΔE=17.7 to 47.5); and reduced the predicted half-life of total anthocyanins (ranging from 1.8 to 3weeks), compared to pH 2 (44.6 to 60.7weeks). Condensed forms had degradation kinetics similar to monomeric anthocyanins. Purple corn pericarp pigments can be used in acid beverages with an acceptable shelf-life.

The Effect of pH and Color Stability of Anthocyanin on Food Colorant

Anthocyanins are naturally occurring pigments of red and purple. Red anthocyanin pigments provide a strong and sharp and widely applied in various industries such as food coloring or drink. Anthocyanins isolated by maceration, extraction and thin layer chromatography (TLC). The extract has been obtained from the initial stages of maceration then separated into several fractions by chromatography to isolate fractions colored dark red. Identification of chemical compounds with TLC (Thin Layer Chromatography) is able to distinguish the fraction of anthocyanin produced. FTIR (Fourier Transform Infrared Spectroscopy) used to identification of the functional group of a compound. The UV-Vis absorption spectra have to produce maximum absorbance values that describe the intensity of anthocyanin spectra in different colors for different pH. Anthocyanins are more stable at low pH (acidic conditions) which gives a red pigment. Meanwhile, the higher the pH value of anthocyanin will provide color fading of the color blue. So as a food colorant, anthocyanin with a low pH or height pH has a significant effect on the food colorant.

Stability improvement of natural food colors: Impact of amino acid and peptide addition on anthocyanin stability in model beverages

Anthocyanins are prone to chemical degradation and color fading in the presence of vitamin C. The potential of three amino acids (l-phenylalanine, l-tyrosine, l-tryptophan) and a polypeptide (ε-poly-l-lysine) in prolonging the color stability of purple carrot anthocyanins (0.025%) in model beverages (0.05% l-ascorbic acid, citric acid, pH 3.0) stored at elevated temperature (40°C/7 days) was examined. In the absence of amino acids or peptides, anthocyanin degraded at first-order reaction rate. Addition of amino acids or peptide (0.1%) increased the color stability of anthocyanins, with the most significant improvement observed for l-tryptophan. The average half-life of anthocyanin color increased from 2 days to 6 days with l-tryptophan addition. Fluorescence quenching measurements revealed that the l-tryptophan interacted with anthocyanins mainly through hydrogen bonding, although some hydrophobic interaction may also have been involved. Overall, this study suggests that amino acid or peptide addition may prolong the color stability of anthocyanin in beverage products.

Stabilization of Black Soybean Anthocyanin by Chitosan Nanoencapsulation and Copigmentation

To stabilize the color and the antioxidant activity of black soybean anthocyanin, the potential of nanoencapsulation and copigmentation was investigated. Anthocyanin was copigmented with sinapic acid and nanoencapsulated by the ionic gelation method. The stability of anthocyanin in terms of color and antioxidant activity was significantly improved by both nanoencapsulation and copigmentation (P < 0.05). The nanoencapsulation was more efficient in improving the antioxidant activity and the color stability of Anthocyanin at high temperature. In addition, the highest stability of anthocyanin was observed in the copigmented anthocyanin-loaded nanoparticles, in which half-lives and retention were more than 2.6 and 7.4-fold greater than those of nonencapsulated anthocyanin. These results suggested that a combination of copigmentation and nanoencapsulation could be an effective technique for improving the color and antioxidant stability of anthocyanin. Practical Applications Although anthocyanin of black soybean has diverse health-promoting properties, such as antioxidant, anti-obesity, and hypolipidemic effects, few studies have been conducted with black soybean anthocyanin. In this manuscript, to stabilize the color and the antioxidant activity of black soybean anthocyanin, the potential of nanoencapsulation and copigmentation was investigated. The result suggests that a combination of copigmentation and nanoencapsulation could be useful for improving anthocyanin stability in terms of color and antioxidant activity. Therefore, this strategy for stability of black soybean anthocyanin could be practically employed in the food and nutrition industry because it can improve not only visual color but also biological activity of anthocyanin-related products.

THE INFLUENCE OF VARIOUS CONCENTRATION OF RED ROSES (ROSA DAMASCENA MILL) FLOWER EXTRACT TO ANTHOCYANIN COLOR STABILITY JELLY AS BIOLOGY LEARNING SOURCE

Red roses flower (Rosa damascena Mill) is plant which can be cultivated in Indonesia. Red roses flower have a natural pigment contain that anthocyanin which can be used as food coloring. The crown of roses flower contain an anthocyanin pigment that include to flavonoid and kind of sianidin anthocyanin which have a function as antioxidant or free radical preventing. Pigment as bioactive is important compound that useful for human health. The aim of this research is to know various concentration influence of red roses flower extract and to know effective concentration that the most influence to color stability of anthocyanin jelly. Research types with treatment, repetition, and control. The design of this research using completely randomized design (RAL), with 6 treatment (0%, 1%, 2%, 3%, 4%, and 5%) and repeated 4 times. Population of this research is jelly. Sample of this research is jelly which added by red roses flower extract. If the data result is have normality, that the data is analysed by Anava one-way test and continue with Duncan’s 5%. Result of this research shown that there was influence of various concentration of red roses flower extract to color stability of anthocyanin jelly. Result test for color stability, the most influence concentration for brightness is 4%, red color is 5%, and yellow color is 0%.
