A clinical case of catarrhal mastitis in a cow

Food security includes not only the provision of basic foodstuffs to the population but also the guarantee of their quality and safety for the consumer. These criteria are essential for milk as the primary food product. The pathological processes, which are developed in the breast, harm the quality of milk, and the medical products used in their treatment, may affect the safety of this product. The article has represented the results of the bioregulatory approach in treating catarrhal mastitis in cows using medical products containing ultra-low doses of biologically active substances. These products do not have a withdrawal period regarding milk. The use of this approach makes it possible to increase the safety of treatment, which in turn will lead to economic benefits. Clinical symptoms of mastitis were determined by clinical signs (increasing local temperature, soreness, thickening of the udder, changing in color and consistency of milk, presence of flakes and clots). A сow No. 3095 was selected with characteristics of clinical symptoms of catarrhal mastitis of the rear left lobe of the udder. The following scheme was used for therapy: intramuscular injection of Traumeel 10 ml once a day, intracisternal injection of Lachesis Сompositum 10 ml into the affected part once a day, Traumeel(gel) locally after milking. The choice of bioregulatory preparations was based on the components and their action. On the third day, the cow's udder swelling decreased, local temperature normalized. The color and consistency of milk during control milking and its examination with the Avita diagnostic test showed an improvement from (+++) to (++). Complete clinical and laboratory recovery was diagnosed on the sixth day. A cow was returned to the general herd. Based on the treatment results, it can be concluded that this treatment scheme for catarrhal mastitis is effective. The absence of a withdrawal period regarding milk and the acceleration of recovery rates make it economically profitable.

The Indicators of Clinical and Subclinical Mastitis in Equine Milk.

Simple SummaryMastitis, the inflammation of the mammary gland, is a major problem in a mare’s perinatal period, negatively affecting both the health of the mare and newborn foal and the quality of milk produced on dairy equine farms. The detection of mastitis is therefore one of the important goals in the equine breeding and dairy industry. This study aimed to determine the somatic cell count (SCC), the percentage of the immune cells, the electrical conductivity (ECM), and bacteriological index (BII) in milk collected from mares with (CM) and without (NCM) clinical symptoms of mastitis. The increase in examined indicators is suspected to be a subclinical mastitis indicator, therefore the study aimed to separate two subgroups, mares with (SM) and without (NSM) subclinical symptoms of mastitis. In milk from NCM mares the values of SCC, immune cells, and ECM increased immediately after birth and weaning, whereas during the rest of the lactation period their values were at a low level. Similarly, in milk from CM mares, the values of examined indicators were high and comparable to the weaning time. An increase in the level of examined indicators may become an early indication of subclinical mastitis.The somatic cell count in milk (SCC) and electrical conductivity of milk (ECM) are indicators of the health status of the mammary gland. Among somatic cells, mainly polymorphonuclear neutrophils (PMN), macrophages (MAC), and lymphocytes (LYM) are rated. This study aimed to determine the SCC, PMN, MAC, LYM, ECM, and bacteriological index (BII) in milk collected from mares with (CM) and without (NCM) clinical symptoms of mastitis concerning mares with (SM) and without (NSM) subclinical mastitis. Milk samples were collected from 27 mares divided into NCM (n = 12) and CM (n = 15) groups. In samples, SCC quantification, cytological examinations, ECM measurement, and bacteriological examination were performed. In NCM mares, the values of SCC, PMN, MAC, LYM, and ECM were higher in initial than in consecutive examined days after birth until weaning. After weaning the proportion of SCC, PMN, MAC, LYM, ECM, and BII increased and did not differ with the average values in CM mares. These equine milk indicators may reflect an early symptom of subclinical mastitis and in the future may be used in the early detection of mastitis or as a tool of assessment of the health status of the mammary gland in the dairy equine farm.

​Studies on Cefquinome Sulfate Intramammary Infusion Composited with Varied Excipients for the Prevention of Dairy Cows Mastitis in the Dry Cow Period

Background: The prevention and treatment of dairy cow mastitis during the dry period are crucial for the dairy industry. In this work, 200 dairy cows without mastitis and normal entering dry period were selected and randomly divided into 4 groups, for the first time, to explore the prevention effects using the different formulations of Cefquinome Sulfate Breast Injection with varied excipients. Methods: Three formulations with various excipients were further evaluated based on the drug residue analysis, somatic cell reduction in the milk and clinical symptoms of mastitis. In the control group, cloxacillin benzathine was utilized for comparisons. Result: All three Cefquinome test groups showed a better effect on prevention than the benzathine cloxacillin group in terms of drug release time, post-partum drug residue, reducing somatic cell count and preventing mastitis in the dry period. Various excipients showed slightly different impacts on the drug residues, the changes in somatic cell counts of dairy cows before and after medication. The prevention effects on mastitis from different excipients were similar after 30 d.

Rumen microbiome structure and metabolites activity in dairy cows with clinical and subclinical mastitis

BackgroundDue to the high prevalence and complex etiology, bovine mastitis (BM) is one of the most important diseases to compromise dairy cow health and milk quality. The shift in milk compositions has been widely investigated during mastitis, but recent studies suggested that gastrointestinal microorganism also has a crucial effect on the inflammation of other peripheral tissues and organs, including the mammary gland. However, research focused on the variation of rumen inner-environment during mastitis is still limited. Therefore, the ruminal microbial profiles, metabolites, and milk compositions in cows with different udder health conditions were compared in the present study. Furthermore, the correlations between udder health status and ruminal conditions were investigated. Based on the somatic cell counts (SCC), California mastitis test (CMT) parameters and clinical symptoms of mastitis, 60 lactating Holstein dairy cows with similar body conditions (excepted for the udder health condition) were randomly divided into 3 groups (n = 20 per group) including the healthy (H) group, the subclinical mastitis (SM) group and the clinical mastitis (CM) group. Lactation performance and rumen fermentation parameters were recorded. And rumen microbiota and metabolites were also analyzed via 16S rRNA amplicon sequencing and untargeted metabolomics, respectively.ResultsAs the degree of mastitis increased, rumen lactic acid (LA) (P < 0.01), acetate, propionate, butyrate, valerate (P < 0.001), and total volatile fatty acids (TVFAs) (P < 0.01) concentrations were significantly decreased. In the rumen of CM cows, the significantly increased bacteria related to intestinal and oral inflammation, such as Lachnospiraceae (FDR-adjusted P = 0.039), Moraxella (FDR-adjusted P = 0.011) and Neisseriaceae (FDR-adjusted P = 0.036), etc., were accompanied by a significant increase in 12-oxo-20-dihydroxy-leukotriene B4 (FDR-adjusted P = 5.97 × 10− 9) and 10beta-hydroxy-6beta-isobutyrylfuranoeremophilane (FDR-adjusted P = 3.88 × 10− 10). Meanwhile, in the rumen of SM cows, the Ruminiclostridium_9 (FDR-adjusted P = 0.042) and Enterorhabdus (FDR-adjusted P = 0.043) were increased along with increasing methenamine (FDR-adjusted P = 6.95 × 10− 6), 5-hydroxymethyl-2-furancarboxaldehyde (5-HMF) (FDR-adjusted P = 2.02 × 10− 6) and 6-methoxymellein (FDR-adjusted P = 2.57 × 10− 5). The short-chain fatty acids (SCFAs)-producing bacteria and probiotics in rumen, including Prevoterotoella_1 (FDR-adjusted P = 0.045) and Bifidobacterium (FDR-adjusted P = 0.035), etc., were significantly reduced, with decreasing 2-phenylbutyric acid (2-PBA) (FDR-adjusted P = 4.37 × 10− 6).ConclusionThe results indicated that there was a significant shift in the ruminal microflora and metabolites associated with inflammation and immune responses during CM. Moreover, in the rumen of cows affected by SM, the relative abundance of several opportunistic pathogens and the level of metabolites which could produce antibacterial compounds or had a competitive inhibitory effect were all increased.

Rumen and Hindgut Bacteria Are Potential Indicators for Mastitis of Mid-Lactating Holstein Dairy Cows.

Mastitis is one of the major problems for the productivity of dairy cows and its classifications have usually been based on milk somatic cell counts (SCCs). In this study, we investigated the differences in milk production, rumen fermentation parameters, and diversity and composition of rumen and hindgut bacteria in cows with similar SCCs with the aim to identify whether they can be potential microbial biomarkers to improve the diagnostics of mastitis. A total of 20 dairy cows with SCCs over 500 × 103 cells/mL in milk but without clinical symptoms of mastitis were selected in this study. Random forest modeling revealed that Erysipelotrichaceae UCG 004 and the [Eubacterium] xylanophilum group in the rumen, as well as the Family XIII AD3011 group and Bacteroides in the hindgut, were the most influential candidates as key bacterial markers for differentiating “true” mastitis from cows with high SCCs. Mastitis statuses of 334 dairy cows were evaluated, and 96 in 101 cows with high SCCs were defined as healthy rather than mastitis according to the rumen bacteria. Our findings suggested that bacteria in the rumen and hindgut can be a new approach and provide an opportunity to reduce common errors in the detection of mastitis.

Identification of Emerging Human Mastitis Pathogens by MALDI-TOF and Assessment of Their Antibiotic Resistance Patterns.

Lactational mastitis constitutes one of the main causes of undesired weaning, depriving the mother–infant pair from the benefits of breastfeeding; therefore, this condition should be considered a relevant public health issue. The role of specific microorganisms remains unclear since human milk cultures and antibiotic susceptibility testing (AST) are not routinely performed, despite the fact that this would be key to ensure an early and effective diagnosis and treatment. The objective of this study was to describe the culturable microbial diversity in 647 milk samples from breastfeeding women with clinical symptoms of mastitis by Matrix-Assisted Laser Desorption Ionization Time-of-Flight Mass Spectrometry (MALDI-TOF) VITEK MS technology and to analyze the antimicrobial susceptibility profiles of a collection of isolates from these samples by the VITEK 2 AST system. Staphylococcus epidermidis was the most common species isolated from mastitis samples (87.6%), while Staphylococcus aureus was detected in 22.1%. Streptococci constituted the second (68.6%) most prevalent bacterial group, with Streptococcus mitis/oralis, Streptococcus salivarius, and Streptococcus parasanguinis detected with frequencies of 40.8, 36.8, and 14.4%, respectively. The antibiotic susceptibility profiles of 642 staphylococcal isolates indicated a remarkable resistance to benzylpenicillin (88.3%) and erythromycin (67.3%) with differences between species. A high percentage of Staphylococcus isolates were resistant to at least one antibiotic (Staphylococcus hominis, 100%; S. epidermidis, 98.2%; S. aureus, 92.9%; Staphylococcus lugdunensis, 90.5%) and the percentage of multidrug-resistance (MDR) isolates was noticeable (S. hominis, 81%; S. epidermidis, 64.4%; S. aureus, 11.5%; S. lugdunensis, 10.5%). In relation to streptococcal isolates (n = 524), AST revealed high or moderate percentages of resistance to erythromycin (68.7%), benzylpenicillin (63.7%), ampicillin (51.5%), and tetracycline (30.8%). Antibiotic resistance to at least one antibiotic was detected in 97.6% of S. parasanguinis, 92.6% of S. salivarius, 83.3% of S. mitis/oralis, and 72.4% of Streptococcus vestibularis isolates. A significant number of MDR streptococcal isolates was also found (S. parasanguinis, 51.2%; S. salivarius, 39.3%; S. mitis/oralis, 34.6%; and S. vestibularis, 19%). The results highlight the important role of coagulase-negative staphylococci and streptococci as human mastitis-causing agents. Moreover, the high rates of antimicrobial resistance among these microorganisms must be contemplated as an issue of clinical relevance in relation to treatment options.

Detection virulence factors of Klebsiella pneumonia from cattle by using PCR tec.

Identification of K. pneumoniae was avaluated using conventional microbiological characteristics and molecular assays .Milk specimens were collected from cattle suffered from clinical symptoms of mastitis from different farms of AL-Qadisyiah province.Out of 45 samples obtained, 20 isolates ( 44.4 %) were detected as K. pneumonia according to morphology of colonies and biochemical features.Molecular detection of Klebsiella pneumoniae based 16S rRNA gene for determination two virulence factor genes(rmpA,magA) by using specific primer.These genes potent the pathogenesis of Klebisella pneumonia. The primers were made in this study by using NCBI-GenBank and primer3 plus design online. The primer is made by company in Korea (Bioneer).Moleculer detection of isolates which give away specific PCR products of of 312bp for magA gene and 835bp for rmpA gene .The magA and rmpA genes were amplified in six (30%)and five(25%) isolates out of 20 isolates of Klebsiella pneumonia.

Coagulase-negative staphylococci (CNS) as an aetiological factor of mastitis in cows

The aim of the present study was to determine the proportions of individual coagulase-negative Staphylococcus species in clinical and subclinical mastitis. The material consisted of 100 CNS isolates obtained from 223 milk samples collected from cows with clinical and subclinical mastitis. Coagulase-negative staphylococci constituted 44.8% of all isolated microorganisms. CNS were isolated from the mammary gland secretions of 86 cows from farms in the Lublin region (Poland). Clinical mastitis was found in 20 whereas subclinical mastitis in 66 study cows (23.3% and 76.7%, respectively). The symptoms of clinical mastitis were mild. The clinical forms of mastitis concerned mainly the first or second lactation. Subclinical mastitis was most commonly observed during the second lactation. Four CNS species (S. xylosus, S. chromogenes, S. haemolyticus and S. sciuri) were isolated from clinical and subclinical mastitis. S. xylosus was the commonest CNS species isolated from cows with clinical mastitis whereas S. chromogenes was the most prevalent one in subclinical mastitis cases. The three CNS species (S. warneri, S. hominis and S. saprophyticus) caused only subclinical mastitis.

Observations on experimental mycotic mastitis in goats.

Summary This study deals with the experimental production of mastitis in healthy goats by infusing Cryptococcus neoformans, Candida albicans, C. stellatoidea, C. krusei, C. tropi-calis, C. pseudotropicalis, C. parapsilosis and C. guilliermondii strains through the teat canal. The clinical symptoms of mastitis appeared in all the goats injected with the fungal strains. Inoculation of penicillin intramammarily prior to giving fungal infection appeared to increase the pathogenicity of fungi.

Cytomegalovirus and Epstein–Barr virus in breast milk are associated with HIV-1 shedding but not with mastitis

Breast milk HIV-1 load is associated with clinical and subclinical mastitis, and both milk viral load and mastitis are associated with increased mother-to-child-transmission of HIV-1 through breastfeeding. Bacterial infections may cause clinical mastitis, but whether other copathogens common in HIV-1 infection are associated with subclinical mastitis or HIV-1 shedding is unknown. A cross-sectional study of HIV-1-infected breastfeeding women in Zimbabwe was performed to examine the relationship between a wide range of breast coinfections, mastitis, and HIV-1 shedding. Breast milk was cultured for bacteria and fungi and tested by PCR for mycobacteria, mycoplasmas, human herpesvirus (HHV)-6, HHV-7, HHV-8, cytomegalovirus, Epstein-Barr virus, and HIV-1 RNA and DNA. Symptoms of clinical mastitis were documented and subclinical mastitis was identified by breast milk sodium concentration (Na) and leukocyte counts. Coinfections of milk were not associated with clinical or subclinical mastitis in the 217 women studied. Detection of HIV-1 RNA, but not DNA, in breast milk was associated with cytomegalovirus concentration (odds ratio = 1.8, P = 0.002) and detection of Epstein-Barr virus (odds ratio = 3.8, P = 0.0003) but not other coinfections in multivariate analysis. Coinfection of breast milk with bacteria, fungi, or herpes viruses was not associated with mastitis. The associations between shedding of cytomegalovirus and Epstein-Barr virus with HIV-1 in milk suggest a local interaction between herpes virus infection and HIV-1 independent of mastitis. Cytomegalovirus and Epstein-Barr virus infections may impact HIV-1 shedding in breast milk and the risk of MTCT.

Preliminary Findings on the Characterization of Bacteria Causing Mastitis by Gas Chromatography

SUMMARY Metabolic products produced by Corynebacterium ulcerans, Enterobacter aerogenes, Escherichia coli, Staphylococcus aureus, Streptococcus agalactiae, Strep, dysgalactiae and Strep, uberis isolated from cows with clinical symptoms of mastitis, were determined by direct injection of culture fluids into a gas chromatograph. The products formed in the presence and absence of glucose after growth in screw-cap bottles and shake cultures showed a marked difference and can be used to characterize these organisms.

The udder as a possible source of coliform organisms in milk

1. It seems to be the general opinion of veterinary contributors on the subject that infection of the udder with coliform organisms is rapidly followed by clinical symptoms of mastitis, and marked changes in the appearance of the secretion. There would therefore appear to be little danger of contamination of bulk milk from such sources except during the few hours between infection and the appearance of the clinical symptoms.2. Injury to the teats or quarters may result in infection of milk with coliform organisms owing to the difficulty of thorough cleaning of the malformed tissue.3. Several cases of infection of quarters in the entire absence of clinical or other symptoms of disease have been cited. There is no evidence that such cases can be classed as subclinical forms of the disease as is the case in streptococcic mastitis. The evidence in two cases cited indicates that milk drawn from a quarter can remain infected with coliform organisms for a considerable period with no apparent clinical or other symptoms of disease.4. There is sufficient evidence to warrant the examination of quarter samples in cases where infection of bulk milk with coliform organisms from other more common sources appears to be unlikely.
