Identifying Effective Biosecurity Measures for Preventing the Introduction of Classical Swine Fever in Pig Farms in Japan: Under the Condition of Absence/Presence of Observable Infected Wild Boar

The outbreak of infectious diseases in swine, such as classical swine fever (CSF), has become a significant concern in the pig‐farming industry. In Japan, after the re‐emergence of CSF in 2018, farms are now exposed to the risk of transmission from infected wild boar and CSF‐contaminated farms. This study aimed to identify biosecurity measures that were effective for the prevention of CSF introduction into farms during the period from the beginning of the CSF epidemic to the implementation of a vaccination campaign for domestic pigs at risk. The probability of virus introduction was assumed to be increased by the elevated risk from CSF‐infected wild boar and infected farms around the farm. The risk from infected wild boar was represented by the prevalence of CSF in wild boar or the occupancy of 1‐km grid cells with infected wild boar within 10‐km radii from a pig farm and the occurrence of CSF outbreaks on neighboring farms. Conversely, the probability of virus introduction was assumed to decrease in response to on‐farm biosecurity measures being implemented on each farm. The implementation of biosecurity measures on the farms and farm attributes were obtained through a questionnaire survey. Analyses were performed on each farm under the weekly situations where infected wild boar were both absent and present in the vicinity using a binomial generalized linear model. On farms where infected wild boar were not present around farms, daily washing and disinfecting of work clothing in pig houses was identified as the main measure to reduce the risk of CSF introduction into farms. On farms with infected wild boar in the vicinity, the absence of public roads on the farm and preventing wildlife intrusion into the areas where pig carcasses were stored were demonstrated to be effective in preventing CSF introduction. Based on the assumption that strict and comprehensive biosecurity measures are required to prevent CSF introduction, the implementation of these potentially effective measures is worth being prioritized.

Measuring impact of vaccination among wildlife: The case of bait vaccine campaigns for classical swine fever epidemic among wild boar in Japan.

Understanding the impact of vaccination in a host population is essential to control infectious diseases. However, the impact of bait vaccination against wildlife diseases is difficult to evaluate. The vaccination history of host animals is generally not observable in wildlife, and it is difficult to distinguish immunity by vaccination from that caused by disease infection. For these reasons, the impact of bait vaccination against classical swine fever (CSF) in wild boar inhabiting Japan has not been evaluated accurately. In this study, we aimed to estimate the impact of the bait vaccination campaign by modelling the dynamics of CSF and the vaccination process among a Japanese wild boar population. The model was designed to estimate the impact of bait vaccination despite lack of data regarding the demography and movement of wild boar. Using our model, we solved the theoretical relationship between the impact of vaccination, the time-series change in the proportion of infected wild boar, and that of immunised wild boar. Using this derived relationship, the increase in antibody prevalence against CSF because of vaccine campaigns in 2019 was estimated to be 12.1 percentage points (95% confidence interval: 7.8–16.5). Referring to previous reports on the basic reproduction number (R0) of CSF in wild boar living outside Japan, the amount of vaccine distribution required for CSF elimination by reducing the effective reproduction number under unity was also estimated. An approximate 1.6 (when R0 = 1.5, target vaccination coverage is 33.3% of total population) to 2.9 (when R0 = 2.5, target vaccination coverage is 60.0% of total population) times larger amount of vaccine distribution would be required than the total amount of vaccine distribution in four vaccination campaigns in 2019.

Estimation of infection risk on pig farms in infected wild boar areas—Epidemiological analysis for the reemergence of classical swine fever in Japan in 2018

In September 2018, classical swine fever (CSF) reemerged in Japan after 26 years’ absence. The first case was detected at a pig farm in Gifu Prefecture, in the center of Japan, and the disease spread to both domestic pigs and wild boar (Sus scrofa). The spread of CSF in wild boar is extremely difficult to control and is thus a great threat to domestic pig farms, and understanding the transmission risk from wild boar to domestic pigs is essential to implement effective control measures that will prevent domestic pig infection. Therefore, this study elucidates the transmission risk from wild boar to domestic pigs by introducing a transmission kernel that is dependent on the distance between infected wild boar and pig farms, and then estimating the risk area of infection from wild boar by describing the transmission probability. The study used epidemiological data from Gifu Prefecture in the period from September 2018 to March 2019, including a total of 171 1-km grid cells where an infected wild boar was detected and pig farm data from 13 infected and 34 uninfected farms. The estimated infection risk area within 28 days matched well with the observed data. The risk area widened gradually during the epidemic, and at the end of March, the risk area extended over a range of approximately 75 km from east to west and 40 km from north to south (almost 3000 km2). Ten out of the 13 infected farms and four out of the 34 uninfected farms were located within the high-risk area (>60 % infection probability). In contrast, one infected farm and 18 uninfected farms were located within the low-risk area (<5 % infection probability). When several infected grid cells were detected within 5 km of a pig farm, the risk of infection from wild boar within 28 days was more than 5 %. This analysis provides an estimate of the potential spatial range over which CSF virus can spread between wild boar and domestic pig farms, and can be used to inform the early detection of CSF-suspected pigs and the strengthening of biosecurity measures that will effectively prevent and control the disease based on the infection risk level.

A Simulation Model to Determine Sensitivity and Timeliness of Surveillance Strategies.

Animal surveillance systems need regular evaluation. We developed an easily applicable simulation model of the German wild boar population to investigate two evaluation attributes: the sensitivity and timeliness (i.e. the ability to detect a disease outbreak rapidly) of a surveillance system. Classical swine fever (CSF) was used as an example for the model. CSF is an infectious disease that may lead to massive economic losses. It can affect wild boar as well as domestic pigs, and CSF outbreaks in domestic pigs have been linked to infections in wild boar. Awareness of the CSF status in wild boar is therefore vital. Our non-epidemic simulation model is based on real data and evaluates the currently implemented German surveillance system for CSF in wild boar. The results show that active surveillance for CSF fulfils the requirements of detecting an outbreak with 95% confidence within one year after the introduction of CSF into the wild boar population. Nevertheless, there is room for improved performance and efficiency by more homogeneous (active and passive) sampling of wild boar over the year. Passive surveillance alone is not sufficient to meet the requirements for detecting the infection. Although CSF was used as example to develop the model, it may also be applied to the evaluation of other surveillance systems for viral diseases in wild boar. It is also possible to compare sensitivity and timeliness across hypothetical alternative or risk-based surveillance strategies.

The control of classical swine fever in wild boar.

Classical swine fever (CSF) is a viral disease with severe economic consequences for domestic pigs. Natural hosts for the CSF virus (CSFV) are members of the family Suidae, i.e., Eurasian wild boar (sus scrofa) are also susceptible. CSF in wild boar poses a serious threat to domestic pigs. CSFV is an enveloped RNA virus belonging to the pestivirus genus of the Flaviviridae family. Transmission of the infection is usually by direct contact or by feeding of contaminated meat products. In recent decades CSF has been successfully eradicated from Australia, North America, and the European Union. In areas with dense wild boar populations CSF tends to become endemic whereas it is often self-limiting in small, less dense populations. In recent decades eradication strategies of CSF in wild boar have been improved considerably. The reduction of the number of susceptible animals to a threshold level where the basic reproductive number is R0 < 1 is the major goal of all control efforts. Depending on the epidemiological situation, hunting measures combined with strict hygiene may be effective in areas with a relatively low density of wild boar. Oral immunization was shown to be highly effective in endemic situations in areas with a high density of wild boar.

Controlling of CSFV in European wild boar using oral vaccination: a review.

Classical swine fever (CSF) is among the most detrimental diseases for the swine industry worldwide. Infected wild boar populations can play a crucial role in CSF epidemiology and controlling wild reservoirs is of utmost importance for preventing domestic outbreaks. Oral mass vaccination (OMV) has been implemented to control CSF in wild boars and limit the spill over to domestic pigs. This retrospective overview of vaccination experiences illustrates the potential for that option. The C-strain live vaccine was confirmed to be highly efficacious and palatable baits were developed for oral delivery in free ranging wild boars. The first field trials were performed in Germany in the 1990’s and allowed deploying oral baits at a large scale. The delivery process was further improved during the 2000’s among different European countries. Optimal deployment has to be early regarding disease emergence and correctly designed regarding the landscape structure and the natural food sources that can compete with oral baits. OMV deployment is also highly dependent on a local veterinary support working closely with hunters, wildlife and forestry agencies. Vaccination has been the most efficient strategy for CSF control in free ranging wild boar when vaccination is wide spread and lasting for a sufficient period of time. Alternative disease control strategies such as intensified hunting or creating physical boundaries such as fences have been, in contrast, seldom satisfactory and reliable. However, monitoring outbreaks has been challenging during and after vaccination deployment since OMV results in a low probability to detect virus-positive animals and the live-vaccine currently available does not allow serological differentiation of infected from vaccinated animals. The development of a new marker vaccine and companion test is thus a promising option for better monitoring outbreaks during OMV deployment as well as help to better determine when to stop vaccination efforts. After rabies in red fox, the use of OMV against CSF in European wild boar can be considered as a second example of successful disease control in wildlife. The 30 years of disease control experience included in this review may provide options for improving future disease management within wild populations.

Descriptive summary of the classical swine fever control in wild boar in Germany since 2005

Classical swine fever (CSF) in wild boar repeatedly appeared in different federal states of the Federal Republic of Germany since 1995, from which it has been successfully eradicated sometimes fast, sometimes in a more time taking way using oral immunization as a main element of control. Since 2005 the cases focused solely on North Rhine-Westphalia and Rhineland-Palatinate. In the present study, therefore, the situation of CSF in wild boar has been closely investigated concerning the period 2005 to 2012 in these two regions. It is noteworthy that in this period two different variants of the virus subtype 2.3 occurred in two regionally defined areas of the "Eifel" and "Westerwald" as well as in the "Pfalz". The two Federal States have undertaken extensive oral vaccination campaigns and surveillance activities, which enabled an assessment of the existing virus prevalence and serological prevalence in the different regions. After an initial high serological prevalence, caused probably by interaction of infection and vaccination, the serological levels stabilized seasonally adjusted in a range from 50 to 60% in almost all areas. The vaccination campaigns have been maintained by both Federal States over a period of at least 2.5 years after virus has been detected for the last time. In consequence Germany as a whole has been recognized for the first time to be officially free from CSF in wild boar. By genotyping of virus isolates it has been demonstrated that the virus changed over time and played a role in the outbreak area "Westerwald".

Escape of classical swine fever C-strain vaccine virus from detection by C-strain specific real-time RT-PCR caused by a point mutation in the primer-binding site

Classical swine fever (CSF) is one of the most important diseases of pigs. Vaccination in the European Union is limited to emergency situations. Currently, vaccination for the purpose of disease control is carried out in wild boar populations. Wild boar are in most cases vaccinated using an oral bait vaccine based on the live modified vaccine virus C-strain “Riems”. A real-time reverse transcription polymerase chain reaction (RT-PCR) protocol for differentiation of C-strain “Riems” vaccine virus from CSF virus (CSFV) field isolates was published previously. In this real-time RT-PCR system differentiation is based on two nucleotide difference one at the 3′ end of each of the primer-binding sites in the E RNS encoding genome region. During extensive diagnostic use of this protocol in an outbreak of CSF in wild boar in Germany, some C-strain positive field samples were found to give negative results in the C-strain “Riems” specific real-time RT-PCR, but positive results in a pan-CSFV real-time RT-PCR system. Moreover, sequencing of C-strain “Riems” vaccine batches for intramuscular use revealed differences in the E RNS encoding region. This led to the assumption that mutations in the corresponding primer-binding site of the C-strain specific system had appeared in the field, and possibly also during manufacturing of different vaccine batches. To test this hypothesis and restore sensitivity, a new primer set for detection of the possible C-strain virus quasi species was designed and tested.

The purpose of this paper is to define diagnostic procedures for wild boar after the completion of oral immunisation against classical swine fever (CSF). Epidemiological analysis of CSF in wild boar in Germany demonstrated that it is vital to carry out virological investigations on all animals found dead, sick or involved in traffic accidents. In principle, this should ensure an effective and prompt diagnosis of CSF. In addition, a defined number of wild boar, especially young animals < or = 6 months old, should also be tested for CSF virus to guarantee a high confidence level in the virological monitoring. Which animals should be examined serologically depends on the age class investigated, the season in which vaccination was stopped and the period of time since completion of vaccination. Therefore, different serological procedures have been defined for different situations during the first three years after completion of oral immunisation.

Incidence and control of CSF in wild boar in Europe

The possibility that Classical swine fever (CSF) persists in a wild boar population has been shown in some areas of France, Germany and Italy in recent years. Availability of susceptible animals and frequency of infectious contacts are major variables influencing disease evolution. In the European Union (EU) measures to control CSF in the wild boar are enforced in the framework of Council Directive 80/217/EEC, including eradication plans that must be enforced taking account of the local epidemiological situation. Oral vaccination experiments are being carried out in Germany, which have not led to final conclusions concerning the efficacy of vaccination in eradicating disease.
