Herbal remedies through cactus nutrients

The main constituent of Opuntiaficus-indica cladodes is water (80-95%), followed by small amounts of carbohydrates (3-7%), fiber (1-2%), and protein (0. 5-1%); other compounds are only partly known and have not been quantitatively determined. The sugar moiety includes mucilaginous components containing polymers, such as chains of (1-4)-linked β-D-galacturonic acid and R (1-2)-linked L-rhamnose residues. Beneficial effects of herbal remedies can be obtained from active constituents present in the whole plant, parts of the plant (e. g., flowers, fruits, roots or leaves), or plant materials or combinations thereof, whether in crude or processed state. Cactus (Opuntiaficus-indica) commonly known as prickly pear belongs to the family Cactaceae. Opuntiaficus-indica produces sweet, nutritionally rich edible fruits; its tender cladodes are used as fresh green vegetable and salad. In South Africa, Mediterranean areas and South American this species is also cultivated for its edible fruit (prickly pear), although in some countries different parts of the plant are utilized in the food and cosmetic industry. Opuntiaficus-indica, fruits and stems, have been traditionally used in folk medicine in several countries for several medicinal purposes. Cactus dietary fiber is composed of several chemical components that are resistant to digestive enzymes such as cellulose, hemicelluloses, pectin, lignin, gums, etc. The benefits associated with fiber content are well known, especially for the prevention of illnesses such as diabetes, treatment of gastrointestinal disorders, illnesses associated with low dietary fiber intake, reduction of glucose values in the blood, anti-hyperlipidemic and anti-hypercholesterolemic effects. Throughout history, the benefits of consuming dietary fiber have been recognized. Soluble fibers, including pectins, gums, and mucilages, increase the viscosity of food in the gut, slowing or reducing sugar absorption. The effect of soluble fiber in reducing serum glucose concentrations is a proposed mechanism of action for the herbal hypoglycemic cactus. It has been reported that Opuntiaficus-indica contain phenolic compounds like ferulic acid, feruloyl-sucrose and sinapoyl-diglucoside, fatty acids like palmitic acid, stearic acid, oleic acid, vaccenic acid and linoleic acid (seeds, peel and juicy pulp). It has been observed that Opuntiaficus-indica oil extract with these components’ present antioxidant and anti-free radical activity, having a potential as anticancer, anti-inflammatory, hypoglycemic, hypolipidemic and hypocholesterolemic activities. Cactus fiber tablet contains 500 mg standardized cactus fiber; as well as common tableting excipients were used. Cactus fiber showed an increased fecal fat excretion compared with placebo. No adverse events were reported throughout the study period. The fruit peel contains large amounts of isorhamnetin. Isorhamnetin (3'-methoxy-3,4',5,7-tetrahydroxyflavone) exerts anticancer action by inhibition of epidermal growth factor (EGF)-induced neoplastic cell transformation through a direct lowering of MAP (mitogen-activated protein)/ERK (extracellular signal regulated kinase) kinase1 and phosphoinositol 3-kinase signaling pathways. Isorhamnetin exhibits cardio protective effects by improving viability of neonatal rat ventricular myocytes under invitro ischemia/reperfusion (I/R) via inhibition of lactate dehydrogenase (LDH) activity and prevention of apoptosis. Isorhamnetin improves skin barrier function through activation of Peroxisome Proliferator-Activated Receptor (PPAR)-αand suppression of inflammatory cytokines production. It also inhibits a dipocyte differentiation of murine 3T3 fibroblasts via a decrease of adiponectine expression and secretion, and down regulation of mRNAs of PPAR-γand C/EBP-α, the major adipogenic nuclear receptors. In contrast, isorhamnetin significantly increases the expression of PPAR-γin tumor tissues obtained from xenograft model of gastric cancer cells and, in combination with chemotherapeutic drugs, causes strong antiproliferative effects and cytotoxicity. The linoleic acid content in cactus cladode (34. 87%) is thus close to the percentage (29% to 40. 41%) found in argan oil. It is however lower than in extracts from barely (51. 26%) and soybean (53. 0%), respectively. Cactus pear (O. boldinghii) cladodes having high fiber content and potential health benefits bakery products have been produced and evaluated from composite flours of wheat (WF) and cactus pear stems (SF). Orange-yellow cactus pear fruit pulp has been used to produce a dehydrated cactus pulp sheet and pasteurized cactus pear fruit juices.

Extraction, characterization, and rheology of Opuntia ficus indica cladode polysaccharides

In this study, hot water extraction was applied to obtain water-soluble pectin of Opuntia ficus indica (OFI) cladode. The yield was found 0.18–0.06% in fresh-weight with the degree of esterification at 48 ± 1.25% expressing that extracted polysaccharide has low-methoxyl (LM) pectin features. Some properties of OFI cladode pectin and commercial LM pectin were investigated. Infrared spectrums of the samples were found mostly similar by having major pectin constituents. 0.25, 0.5, 1, 2 and 3% w/v solutions were prepared for rheological properties at 25, 30, 35 and 40°C. Solutions prepared both using commercial pectin (CP) and OCP represented Newtonian behavior at applied shear rate region. More than 1% w/v concentration, CP showed higher thickening property compared to OCP solutions at various temperatures where CP samples were heavier compared to OCP. The activation energy of OCP solutions was higher than CP meaning that CP had stronger interaction between molecules by representing better stability. Novelty impact statement Opuntia ficus indica (OFI) is well known as the fruit part, however, the cladode part has a great potential for food use as a polysaccharide source. Cladode is rich in water-soluble pectic polysaccharides and there is no commercial cladode pectin available on the market. OFI cladode can be used as a cheap source of low-methoxyl pectin with some modifications due to the application area.

Comparative Analysis of the Chemical Composition and Physicochemical Properties of the Mucilage Extracted from Fresh and Dehydrated Opuntia ficus indica Cladodes.

The development of sustainable extraction methods to obtain natural products constitutes a challenge for the food industry. The aim of this work was to compare yield, separation efficiency, chemical composition, and physicochemical properties of the mucilage extracted from fresh cladodes (FNM) and mucilage extracted from dehydrated cladodes (DNM) of O. ficus indica. Suspensions of fresh and dehydrated cladodes (4% w/w) were prepared for mucilage extraction by using a mechanical separation process. Subsequently, the separated mucilage was precipitated with ethyl alcohol (1:2 v/v) then, yield and separation efficiency were determined. The mucilage was characterized by measuring Z potential, viscosity, color, and texture attributes. Additionally, chemical proximate analysis, scanning electron microscopy, and thermogravimetric analysis (TGA) were conducted. No significant differences (p < 0.05) were detected in the yield and separation efficiencies between samples. Nevertheless, the dehydration process of cladodes prior to mucilage extraction increased protein, ashes, nitrogen free extract, and calcium content. The viscosity was higher in DNM than in FNM. The TGA revealed a different thermal behavior between samples. In addition, the DNM showed lower L (darkness/lightness), cohesiveness, adhesiveness, and springiness values than those of FNM. These results support that differences found between the chemical and physicochemical properties of DNM and those of FNM will determine the applications of the mucilage obtained from the O. ficus indica cladodes in the food, pharmaceutical, and cosmetic industries.

Opuntia ficus indica waste as a cost effective carbon source for lactic acid production by Lactobacillus plantarum

Opuntia ficus indica (OFI) waste was evaluated as a fermentation feedstock for lactic acid production using Lactobacillus plantarum.Dilute acid pretreatment of the OFI cladodes (OFIC) was performed for extracting maximum fermentable sugars by optimizing process parameters using statistical optimization method. The best results were obtained with HCl 1% (v/v), temperature 120 °C, residence time 40 min, granulation 350 µm and substrate loading 5% (w/v), the sugar concentration reached 24 g/L with low concentration of hydroxymethylfurfural. The feasibility of producing lactic acid from OFI fruit peel (OFIFP) as a source of carbon was also investigated. Lactobacillus plantarum was shown to have a capacity for lactic acid production from OFIC350 (granulation 350 µm) hydrolysate and OFIFP extract without detoxification. The highest lactic acid yields of 0.46 and 0.78 g/g were obtained from enzymatic hydrolysate of pretreated OFIC350 and OFIFP extract, respectively.

Characterisation of Zinc-bearing sulphate phases formed during the synthesis of phosphoric acid and Zinc removal by the ligands of Opuntia ficus-indica

ABSTRACT Phosphogypsum (PG) is a solid waste generated from phosphate fertilisers industries. It represents a serious threat to the aquatic and terrestrial environment because of its acidity and its high content in heavy metals and radionuclides. The aim of this work is to describe the formation of PG during the synthesis of the phosphoric acid, the entrapment of Zinc (Zn) in PG and its lixiviation in presence of a natural organic matter extracted as powder from the cladodes of Opuntia ficus-indica (OFI) using physical and chemical characterisation techniques such as FTIR, XRD, SEM-EDX, laser diffraction, and AAS. The formation of PG mainly occurs in the pH range between 4.6 and 3 and it accompanies the transformation of H2(PO4)− into phosphoric acid H3PO4. The maximal Zn incorporation within the PG was reached at pH 6 and decreased progressively with pH. Zinc was found to have a great tendency to migrate from PG particles to OFI’s suspensions since a maximum Zn removal percentage of 93% was achieved.

Extraction and Physicochemical Characterization of Dried Powder Mucilage from Opuntia ficus-indica Cladodes and Aloe Vera Leaves: A Comparative Study.

Cactaceae and Asphodelaceae are native desert plants known for their high mucilage content, which is a polysaccharide of growing interest in the food, cosmetic, and pharmaceutical industries. In this study, powdered mucilage was obtained from cladodes of Opuntia ficus-indica (OFI) and aloe vera (AV) leaves, and their molecular, morphological, and thermal properties were investigated and compared. Additionally, their dietary fiber content was determined. Three-dimensional molecular models were calculated for both mucilages using ab initio methods. Vibrational spectra (FTIR and Raman) revealed intramolecular interactions and functional groups that were specified with the help of theoretical ab initio and semi-empirical calculations. SEM micrographs measured at magnifications of 500× and 2000× demonstrated significantly different superficial and internal morphologies between these two mucilages. Thermal analysis using DSC/TGA demonstrated superior thermal stability for the OFI mucilage. The dietary fiber content in OFI mucilage was more than double that of AV mucilage. Our results show that both dehydrated mucilages present adequate thermal and nutritional properties to be used as functional ingredients in industrial formulations; however, OFI mucilage exhibited better physicochemical and functional characteristics than AV mucilage as a raw material.

Characteristics of cellulose fibers from Opuntia ficus indica cladodes and its use as reinforcement for PET based composites

ABSTRACT With the aim of valorizing the unexplored Moroccan resources, Opuntia ficus indica (OFI) cladodes were proposed as a renewable source for the production of cellulose. In this work, cellulose microfibers were extracted from OFI cladodes using one of the most suitable methods for the extraction of cellulose based on the alkaline delignification. The morphological (SEM), thermal (TGA/DTG), and structural (XRD, FTIR, and 13C NMR) analysis of the isolated cellulose has been identified. Afterward, the reinforcing capability of cellulose fibers was examined using polyethylene terephthalate (PET) as a matrix by combining extrusion and compression molding processes. The effects of fibers loading on physical properties of the composites, such as tensile strength and Young’s Modulus before and after water absorption, were studied. The stability of the optimal composite was also examined based on the dependence of rheological parameters (G’ and G”) with strain and time. Composites containing 15 wt% of fibers exhibited the best mechanical properties in dry and wet cases, reaching 88% increase from neat PET in Young’s modulus and 36% increase in the tensile strength. However, excellent structural stability of the optimal composite was found with a predominant elastic behavior resulting from the good interface adhesion between cellulose fibers and PET matrix. The present work showed new routes for the valorization of OFI cladodes in the development of composite materials.

Thermal degradation kinetics of Opuntia Ficus Indica flour and talc-filled poly (lactic acid) hybrid biocomposites by TGA analysis

This paper reports the effect of lignocellulosic flour and talc powder on the thermal degradation behavior of poly (lactic acid) (PLA) by thermogravimetric analysis (TGA). Lignocellulosic flour was obtained by grinding Opuntia Ficus Indica cladodes. PLA/talc/ Opuntia Ficus Indica flour (OFI-F) biocomposites were prepared by melt processing and characterized using Wide-angle X-ray scattering (WAXS) and Scanning Electron Microscope (SEM). The thermal degradation of neat PLA and its biocomposites can be identified quantitatively by solid-state kinetics models. Thermal degradation results on biocomposites compared to neat PLA show that talc particles at 10 wt % into the PLA matrix have a minor impact on the thermal stability of biocomposites. Loading OFI-F and Talc/OFI-F mixture into the PLA matrix results in a decrease in the maximum degradation temperature, which means that the biocomposites have lower thermal stability. The activation energies (Ea) calculated by the Flynn Wall Ozawa (FWO) and Kissinger Akahira Sunose (KAS) model-free approaches and by model-fitting (Kissinger method and Coats-Redfern method) are in good agreement with one another. In addition, in this work, the degradation mechanism of biocomposites is proposed using Coats-Redfern and Criado methods.

Bio‐based sustainable films from the Algerian Opuntia ficus‐indica cladodes powder: Effect of plasticizer content

AbstractThe present study investigated the fabrication and characterization of bio‐based sustainable films composed of a terrestrial plant raw material, namely Opuntia ficus‐indica (OFI) cladodes powder (CP) and a marine seaweed derivative, namely agar (A). The effect of glycerol concentration on the properties of the casted films was evaluated at four different contents, namely 30, 40, 50 and 60 wt%. The films present UV‐blocking properties, as well as moderate mechanical performance, thermal stability, and water vapor transmission rate (WVTR). The results point to an increase in thickness, elongation at break, moisture content, water solubility, and WVTR with increasing glycerol content. On the contrary, Young's modulus, tensile strength, and water contact angle decreased as glycerol concentration increased. The best combination is obtained for the film with 30% glycerol, based on an intermediate compromise between physical, mechanical, thermal, and barrier properties. All these outcomes express the potentiality of the powder obtained from grinding the OFI cladodes as raw material to produce low‐cost films for the development of sustainable packaging materials.

Antimicrobial activities of crude solvent extract and synthesized silver nanoparticles of Opuntia ficus-indica L. Cladodes

Opuntia ficus-indica cladodes was examined for their antimicrobial activities by using crude extract and synthesized silver nanoparticles against human pathogenic microbial strains, namely Staphylococcus aureus, Klebsiella pneumoniae, Klebsiella oxytoca, Proteus mirabilis and Candida albicans. All extracts demonstrated moderate activity against the tested microbes in a range between 0.53 and 2.33 cm. The maximum inhibition activity was found to be against K. pneumoniae while the lowest against S. aureus. Synthesized silver nanoparticles (Ag-NPs) showed inhibition activities between 1.82 and 2.03 cm. The maximum activity was recorded against K. oxytoca, while the lowest activities against P. mirabilis. In context of antimicrobial activity, there was no significant difference either solvent crude extract or synthesized silver nanoparticles against tested microbes. Therefore, crude extracts of O. ficus-indica cladodes or Ag-NPs could be used as an alternative natural drug.

Effect of Opuntia ficus indica extract on methotrexate-induced testicular injury: a biochemical, docking and histological study

Methotrexate (MTX) is a chemotherapeutic medicine used in the treatment of several types of cancer and inflammatory diseases. It exhibits several drawbacks especially on highly dividing and developing cells. This study aimed to assess the role of Opuntia ficus indica ethanolic extract on testicular damage induced by MTX in rat. MTX was administrated for 10 days (20 mg/kg). Extract of cactus cladodes (Opuntia ficus indica) was given to MTX-treated rats (0.4 g/kg). Spermatozoa were collected from cauda epididymis and analyzed for sperm count and motility. Testis samples were used for histopathological and oxidative stress studies (assessment of malondialdehyde (MDA) levels, protein carbonyls (PCs), catalase (CAT) glutathione peroxidase (GPx) and superoxide dismutase (SOD) activities). Moreover, levels of testosterone were measured in serum by radioimmunoassay. Our results showed that MTX had destructive effects on sperm count and motility associated with significant decrease in testosterone levels in MTX group. This effect was then confirmed by docking results. Testis of MTX group showed increased oxidative stress status. In fact, PCs and MDA were increased and CAT, GPx and SOD were decreased suggesting increased reactive oxygen species and deficiency in enzymatic antioxidant. These findings were associated with disrupted testicular morphology as assessed by histological study. Cladodes extract had protective effects on rat’s gonad histology, oxidative stress and improve both sperm parameters (count and motility) and serum testosterone levels. In conclusion, our results suggested that Opuntia ficus indica cladodes extract improved MTX-induced testicular injury and possess potent fertility boosting effects in rats. The cactus cladode (Opuntia ficus indica) modulates uricemia, improves endothelial dysfunction and corrects oxidative damage in rats fed a cafeteria diet

PurposeThe effects of Opuntia ficus indica (OFI) cladodes on uricemia level, endothelial dysfunction and oxidative damage were studied in young rats fed a cafeteria diet (CD).Design/methodology/approachA total of 16 young male Wistar rats (weighing 110 ± 20 g and four weeks old) were divided into two homogenous groups. The first group received a CD containing 50% of hyperlipidic diet and 50% of junk food mix (processed mix: hyper-fat, hyper-salted and sweetened) (CD group), and the second group (CD + OFI nopalitos) received the same diet supplemented with 50 g of fresh OFI nopalitos (young cladodes) for 30 days.FindingsOFI nopalitos regulate the hyperuricemia, improve the endothelial dysfunction by raising the bioavailability of nitric oxide(NO) and reduce prooxidant markers by reducing lipid peroxidation and protein oxidation (p &lt; 0.05) and boosting antioxidant capacity and enhancing the antioxidant enzymes activities (p &lt; 0.05) in blood and aorta tissues of rats early fed with a high-fat diet /junk food.Social implicationsBy-products of OFI have specific functional properties that may be beneficial in metabolic disorders and offer a better alternative with an economic and sustainable development perspective.Originality/valueBy-products of OFI highlight potential functional properties mainly based on its potent antioxidant capacity. By-products of OFI can be used as a promising nutraceutical resource to prevent various metabolic disorders in relation with cardiovascular diseases or hyperuricemia in subjects consuming junk food and or living in the Western society to reach the objectives of health policy and maintain a sustainable health system development.

Prospective antimycotoxigenic action of wild Opuntia ficus-indica by-products

Numerous natural compounds perform pharmaceutical, antimycotic and antitoxic purposes in the body system. The aim was to evaluate new phytoconstituents that provide antimycotoxigenic properties against mycotoxins. Bioactive materials chosen were fruit peels and cladodes of wild Opuntia ficus-indica containing a measurable quantity of bioactive phytochemicals. The highest concentration of bioactive metabolites was recorded for protocatechuic and t-cinnamic acids. A reduction effect of bioactives was estimated against aflatoxins, ochratoxin A and zearalenone in a simulated body system. Antifungal activity was determined in liquid media to evaluate antimycotic properties. Lyophilised extracts caused an aflatoxin reduction in media by 14.65% to 23.77% for fruit peels and cladodes, respectively. It caused a decrease of 59% zearalenone and 51% of ochratoxin A in a in a simulated body fluid. The cladode extract manifested better antimycotic and antimycotoxigenic characteristics due to its bioactive contents. These results support a modern antimycotoxin trend of food preservation that has a considerable impact on food safety.

The Effect of Cladode Drying Techniques on the Prebiotic Potential and Molecular Characteristics of the Mucilage Extracted from Opuntia ficus-indica and Opuntia joconostle

The dry, powdered cladodes of Opuntia ficus-indica are often-used in over-the-counter (OTC) pharmaceutical formulations. Gentle drying techniques, such as lyophilization and vacuum drying are compared with convection drying for the cladodes and also compared with another species of economic importance, Opuntia joconostle. The heteropolysaccharide purified from the mucilage extracted from the dried powders were investigated in their monosaccharide composition (HPAEC-PAD, TLC), mineral and protein content, molecular dimensions (SEC) and fermentability by probiotic bacteria (Bioscreen technique) for evaluation of the prebiotic potential of the mucilage. The heteropolysaccharide is composed of galactose, arabinose, xylose, galacturonic acid and rhamnose. O. ficus-indica includes an additional 13% of glucose coming from an α-glucan. The content of Ca (0.3%) and Mg (0.4%) is relatively low in both species; the content of protein adds up to 1.5% in O. ficus-indica but is significantly lower in O. joconostle with 0.8%. The average molecular mass Mw of the extracted mucilage ranges from 3.7 to 4.7 × 105 g∙mol−1 for both species; only the mucilage from long-time convection drying (C2) delivers a lower average Mw of 2.6 × 105 g∙mol−1, due to partial breakdown of the mucilage matrix. All tested probiotic strains utilized the mucilage to some extent; C2 being the most active, and thus confirms the prebiotic potential of cladode’s powder and its derived products. In general, the molecular dimensions and prebiotic potential are not extremely sensitive to the drying treatment, yet temperature and drying time can modify the cladode’s powder to a profile with better prebiotic characteristics.

Opuntia ficus-indica as a durable and eco-friendly corrosion inhibitor on AISI 410 stainless steel in $$0.5~\hbox {M}\,\hbox {H}_{{2}}\hbox {SO}_{{4}}$$

In the dental office, surgical instruments must be sterilized for each patient, following their contamination by saliva and blood. Repeated sterilization cycles cause corrosion. A worn out and corroded instrument may cause toxicity to the patient. The aim of our study is to improve the corrosion resistance of orthodontic pliers during sterilization using green inhibitors. The behaviour of AISI 410 stainless steel with respect to corrosion was studied in a $$0.5~\hbox {M H}_{{2}}\hbox {SO}_{{4}}$$ , using cladodes of Opuntia ficus-indica (OFI) as a green inhibitor. Temperature effect on OFI efficacy was studied for different ranges (25, 35 and $$45^{\circ } \hbox {C}$$ ). The study was carried out using the weight loss method, the stationary electrochemical techniques (polarization curves), the transients (electrochemical impedance spectroscopy), and the scanning electron microscopy associated with X-ray dispersive energy spectroscopy (SEM–EDX) and atomic force microscopy (AFM) observations. OFI behaves as a mixed inhibitor, with a physisorbed adsorption obeying to Langmuir isotherm and, with an efficacy ranging from 95.12 to 15% (v/v) obtained after 12 h immersion at a temperature of $$25^{\circ } \hbox {C}$$ . The SEM–EDX and AFM observations confirm the obtained results. In conclusion, OFI improves corrosion resistance of AISI 410 stainless steels in $$0.5~\hbox {M H}_{{2}}\hbox {SO}_{{4}}$$ at $$25^{\circ } \hbox {C}$$ .

Evaluation of Biological Activities of Opuntia Ficus Indica Cladodes Extract Against Cadmium-Induced Osteoporosis in Male Wistar Rats

The cactus Opuntia ficus indica (L.) Mill. (Cactaceae) is widely used in Tunisian medicine for the treatment of various illnesses. The purpose of the present study was to investigate the antioxidant activities of cactus cladode extract (CCE) and to assess the protective effects of Opuntia ficus indica against osteoporosis induced by cadmium chloride in male Wistar rats. Adult male rats were divided into 4 groups of 9 each: a control group, a group injected with cadmium (3.5mg/kg) for 10 weeks, a group orally given a O. ficus indica cladodes aqueous extract (CCE) (100 mg /kg/day) for 10 weeks then treated with cadmium, and a group receiving only (CCE) for 10 weeks. Bone toxicity was estimated by examining femoral length and weight, calcium, phosphorus, alkaline phosphatase (ALP) and total tartrate-resistant acid phosphatase (ACP) levels in serum. Also, bone levels of calcium, phosphorus and vitamin C and bone mineral density (BMD) of femur diaphysis were measured. Alterations of these bone biomarkers and decreased BMD confirmed cadmium-induced bone toxicity. However, when cadmium was administered in rats given CCE, all the biological parameters underwent much less alteration. Administration of CCE was found to be beneficial by attenuating cadmium-induced femur damage. The protective effect of the plant is mainly attributed to its phenolic compounds that orchestrate antioxidant properties, as highlighted by HPLC-based analysis.

Phytochemical study and evaluation of the antioxidant activity of extracts of Opuntia Ficus-Indica cladodes from the Oriental of Morocco and the effect of microwave activation on the drying time of the plant

In the present work, the phytochemical study and evaluation of the antioxidant activities of the crude extracts of various solvents with increasing polarity (ethanol> acetone> dichloromethane > ethyl acetate > hexane) on fine dried cladode powder was studied. Each microwave oven power (500, 350, 160 and 90 W), of an Opuntia ficus-indica (OFI) medicinal species of the Cactaceae family collected in the rural commune of Bni Rais in Taourirt province of Oriental of Morroco. The phytochemical screening showed the presence of the studied chemical compounds (Flavonoids, Tannins, Quinones and Steroids) rich in powers of 500W and 350 W, especially in extracts with ethanol and acetone. The antioxidant activity of OFI cladodes extracts was evaluated by the DPPH (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging method. The results obtained show that the acetone extracts have a very important activity, especially at the power of 500 W, IC50 = 0.29 mg/ml is found followed by the ethanol extract with a value of IC50 equal to 0.34 mg/ml. After the results we realize that the power 500 W showing antioxidant activity of each solvent extract (Et, Ac, Dc, AE and Hx) greater than that of the power 350, 160 and 90 W respectively (IC50: 500 W <350W <160 W <90 W). Also, the effect of microwaves on the duration of drying of the plant was assessed (40 min under microwave activation compared to 17.5h of conventional heating). The cleaning activities of all extracts, however, were significantly low compared to the reference standard of ascorbic acid with a sequence IC50 = 0.114 mg/ml, used at the same dose.

Physico-chemical changes in cladodes of Opuntia ficus-indica as a function of the growth stage and harvesting areas

The physicochemical properties of cladodes Opuntia ficus-indica (OFI) vary widely during maturation. However, few studies have been performed to evaluate changes in chemical characteristics on the pad depending on the maturity stages. Chemical proximate analysis like the content of total sugars, protein, dry matter, pH, conductivity and ash, depending on the variation of weight of the pad and the growing region was done to determine the changes of characteristics of nopal. Significant changes in physicochemical properties were observed among harvesting sites and during the growing stages. The principal component analysis was then performed to evaluate the relationship between harvesting sites, growth stages, and physicochemical properties. Results highlighted that Sidi El Aidi can be categorized as a poor ecotype among the studied ecotypes, besides, overall harvesting sites pH and proteins had a significant negative correlation with ash, total sugars, reducing sugars, conductivity proteins decrease with increasing age of nopals while total sugars, reducing sugars, and ash content showed the opposite trend. The high total sugar content, minerals of nopal pad (weight higher than 200 g) and proteins (weight lower than 200 g) could be an interesting source of these important components for human diets and also could be used in food, cosmetics, and pharmaceutical industry.

Evaluation of the Molluscicidal, Artemicidal and Cytotoxic Activities of the Lectin from Opuntia ficus-indica Cladodes (OfiL)

Background: Biomphalaria glabrata is an intermediate host for the larvae of Schistosoma mansoni, which is distributed widely in the tropics. B. glabrata control is important to minimize the spread of schistosomiasis and natural compounds have been sought for use against this disease. The Artemia salina bench-top bioassay has been used to investigate the ecotoxicity of many natural compounds, and its results also correlate well with the in vitro cytotoxicity of natural compounds to tumor cells.&#x0D; Aims: To evaluate deleterious effects of the Opuntia ficus-indica lectin (OfiL) on B. glabrata, A. salina and human cancer cell lines.&#x0D; Methods: OfiL was isolated following a previously established protocol. The effects of OfiL on B. glabrata were investigated by determining survival of adults as well as development and hatching of embryos. The concentration required to kill 50% (LC50) of A. salina nauplii was determined. The cytotoxicity was determined using the human cell lines Hep-2 (human larynx epidermoid carcinoma), NCI-H292 (human lung mucoepidermoid carcinoma) and K562 (chronic myelocytic leukemia).&#x0D; Results: The development of most embryos (92.5–97.5%) treated with 1, 10 and 100 µg/mL of OfiL was found to be delayed, and dead (2.2–3.3%) and malformed (0.3–5.2%) embryos were also observed. OfiL did not kill B. glabrata adults, but a high percentage (30–45%) of the embryos generated by snails incubated with the lectin exhibited malformations. OfiL exhibited toxicity against A. salina (LC50: 61.02 µg/mL) but did not display cytotoxicity against the tumor cell lines evaluated.&#x0D; Conclusion: In conclusion, this study showed that OfiL can be a tool for schistosomiasis control that acts by impairing the viability of B. glabrata eggs and the fecundity of adult snails.
