Identification of Primary Metabolite Profiles Reveals Quality Characteristics of Citrus maxima ‘Shatian Yu’ from Different Origins

Citrus maxima ‘Shatian Yu’ displays varying nutritional profiles influenced by its production area. This study evaluated pomelo fruits from four primary and one developing ‘Shatian Yu’ production area. Notably, ‘Shatian Yu’ from the Guilin area exhibited higher sugar and alcohol content, suggesting enhanced taste. Principal component analysis and OPLS-DA revealed significant metabolite differences among production areas. In Guilin, variations were observed in a few substances, including sugars, alcohols, and phenolic acids. When compared with Rong City, Guilin showed a decrease in four phenolic acids and an increase in three organic acids, eighteen amino acids, eighteen lipids, and one vitamin. This comprehensive analysis provides valuable insights for the development of ‘Shatian Yu’ cultivation, highlighting the impact of production areas on fruit quality.

Genomic analysis provides insights into the origin and divergence of fruit flavor and flesh color of pummelo.

Pummelo (Citrus maxima) is one of the most important citrus crops and have genetically contributed sweet orange, lemon and most citrus cultivars. It has been cultivated for c. 4000 years in China and is also distributed in many Southeast Asian countries. Nevertheless, the origin and dispersal of pummelo remain elusive. We conducted whole-genome sequencing for 290 pummelo accessions from China and Southeast Asia (SEA). Our findings indicated that pummelo was originated in Yunnan province. The divergence of the China-SEA accessions occurred c. 2000 years ago and the divergence was likely facilitated through the Maritime Silk Road. We detected the divergence of genomic regions associated with fruit flavor and color, indicating different selection by human activities in different regions. A gene encoding lycopene cyclase 2 (LCYB2) exhibited a high degree of divergence in expression and sequence between red-flesh and white-flesh pummelos. A SNP in the coding region of LCYB2 resulted in a reduction in lycopene β-cyclizing enzyme activity, leading to the accumulation of lycopene and the development of the red-flesh trait. This study reveals the origin and evolutionary history of pummelo and provides insights into the genomic basis for the divergence of fruit flavor and color.

Evaluation of nitrogen use efficiency and N-supplying capacity on pomelo (Citrus grandis L. Osbeck) grown on alluvial soil in the Cuu Long Delta

The study was conducted to determine the nitrogen use efficiency (NUE) and the ability to provide nitrogen from alluvial soil for growing pomelo (Citrus grandis L. Osbeck) in the Mekong Delta. The experiment was arranged in a randomized complete block design including two treatments (NPK treatment: 673 N - 385 P2O5 -192 K2O g/plant and PK treatment: 0 N - 385 P2O5 - 192 K2O g/plant) and three replicates (3 plants for each replicate). The results showed that fruit yield was highest (39.1 kg/plant, N recovery efficiency of 53.8%) in the full NPK fertilization treatment and lowest in the PK fertilization treatment (18.5 kg/plant). Calculation results of the ability to supply nitrogen to pomelo trees in Chau Thanh, Hau Giang showed that alluvial soil had the ability to provide 49.6% of N and the remaining 50.4% from additional fertilizer sources for maximizing fruit yield.

Preparation, identification and α-glucosidase inhibitory activity of a high-methoxyl HG-type pectin from Liangping pomelo (Citrus maxima cv. Liangpin Yu) peel.

The peel from Liangping pomelo (Citrus maxima cv. Liangpin Yu) is generally discarded as waste during post-harvest handling and process, resulting in environmental pollution and waste. Pectin is the major component in pomelo peels and yields significant economic advantages. Thus, developing pomelo peel pectin (PPs) might be a feasible strategy to reduce environmental pollution caused by pomelo peel. The optimized PPs yield was 156.5 ± 2.5 g kg-1 under the inoculum size of 100 mg g-1, liquid-solid ratio of 31 mL g-1, fermentation time of 64 h, and fermentation temperature of 39 °C. PPs-6Aa, a pectin fraction from PPs purified with DEAE-52 cellulose, Sephadex G-100 and Sephadex G-75 column chromatography, showed higher α-glucosidase inhibitory activity, with an IC50 of 0.12 ± 0.03 mg mL-1. It was a high-methoxyl HG-type pectin of 42.8 kDa, and its repeat unit was →4)-α-GalpA-6-OMe-(1→4)-α-GalpA-6-OMe-(1→. Additionally, its α-glucosidase inhibitory activity might be related to hydrogen bonds formed with Lys-156, Glu-277, His-280, Asp-307, Arg-315, Asn-350, Asp-352 and Glu-411, and to hydrophobic interactions formed with Ser-157, Tyr-158, Asp-233, Gln-239, Ser-240, Phe-303, Thr-306, Leu-313, Phe-314, Gln-353 and Arg-442. These findings provide structural and bioactivity information on pectin from Liangping pomelo peel, which could be beneficial for the development of functional foods and pharmaceuticals. © 2024 Society of Chemical Industry.

Adaptation of rhizobacterial and endophytic communities in Citrus Grandis Exocarpium to long-term organic and chemical fertilization.

Organic fertilizers (OF) are crucial for enhancing soil quality and fostering plant growth, offering a more eco-friendly and enduring solution compared to chemical fertilizers (CF). However, few studies have systematically analyzed the effects of OF/CF on root microbiome of medicinal plants, especially in combination with active ingredients. In this study, we investigated the composition and function of bacteria and fungi in the rhizosphere or within the root of traditional Chinese medicinal plants, Citri Grandis Exocarpium (Huajuhong), which were treated with OF or CF over 1, 3, and 5 years (starting from 2018). Additionally, we conducted metabolome analysis to evaluate the effects of different fertilizers on the medicinal properties of Huajuhong. The results indicated that extended fertilization could enhance the microbial population and function in plant roots. Notably, OF demonstrated a stronger influence on bacteria, whereas CF enhanced the cohesion of fungal networks and the number of fungal functional enzymes, and even potentially reduced the proliferation of harmful rhizosphere pathogens. By adopting distancebased redundancy analysis, we identified the key physicochemical characteristics that significantly influence the distribution of endophytes, particularly in the case of OF. In contrast, CF was found to exert a more pronounced impact on the composition of the rhizosphere microbiome. Although the application of OF resulted in a broader spectrum of compounds in Huajuhong peel, CF proved to be more efficacious in elevating the concentrations of flavonoids and polysaccharides in the fruit. Consequently, the effects of long-term application of OF or CF on medicinal plants is different in many ways. This research provides a guide for OF/CF selection from the perspective of soil microecology and aids us to critically assess and understand the effects of both fertilizers on the soil environment, and promotes sustainable development of organic agriculture.

Reduced Fertilization and Magnesium Supplementation: Modulating Fruit Quality in Honey Pomelo (Citrus maxima (Burm.) Merr.)

The excessive use of chemical fertilizers in the Guanxi honey pomelo production area has led to severe soil acidification and magnesium (Mg) deficiency, adversely affecting pomelo fruit quality. To address this issue, an integrated nutrient optimization model crucial for ensuring the sustainable and environmentally friendly development of the Guanxi honey pomelo industry has been explored. In a three-year experiment, two fertilizer treatments were implemented: a farmer fertilizer practice (FP) and an NPK reduction plus foliar Mg fertilizer (OPT + fMg). We investigated the impact of this integrated optimized fertilization measure on pomelo fruit quality from three aspects: flavor (sugars and organic acids), nutrition (vitamin C and mineral elements), and antioxidant properties (phenolics, flavonoids, and phytic acid). The results revealed that the OPT + fMg treatment improved fruit flavor by reducing acidity (titratable acid, citric acid, and quinine), while having a minimal impact on sugar components (sucrose, fructose, and glucose). Additionally, the OPT + fMg treatment increased the total phenolics, total flavonoids, and phytic acid in the fruit peel, enhancing its potential antioxidant quality. However, the OPT + fMg treatment reduced the mineral nutrient quality (excluding calcium) in the fruit. As for the fruit developmental period, the OPT + fMg treatment significantly increased the total flavonoid concentration in the peel from the mid-expansion fruit stage, followed by notable increases in phytic acid in the peel during the mid-to-late expansion fruit stage. The total phenolic concentration in the peel significantly rose only during the late fruit development stage. The most pronounced effect was observed on phytic acid in both peel and pulp. The influence of the OPT + fMg treatment on the mineral nutrients (excluding calcium) primarily occurred during the mid-to-late expansion fruit stage. Overall, the OPT + fMg treatment significantly improved the comprehensive nutritional quality of pomelo fruit, providing valuable insights for scientifically reducing fertilizer application while enhancing fruit quality.

Coumarins rather than alkylamides evoke the numbing orosensation of pomelo peel

Liangpingyou, a well-known Chinese pomelo (Citrus grandis L.) variety, elicits a unique and uncharacterized numbing aftertaste. To understand the molecular bases and characteristics of the pomelo-induced numbing sensation, we first determined that hydroxyl sanshools, the major Sichuan pepper chemosensates, were not responsible via silylation-GC–MS analysis. Pomelo peel juice was then subjected to solid-phase extraction to form 4 fractions, and key sensory-active substances were screened via taste dilution analysis. Three simple coumarins, meranzin hydrate, isomeranzin, and marmin, were identified to induce numbing, which has not been previously reported. Sensory studies via extensively modified half-tongue tests and verification steps revealed recognition thresholds within 0.49–1.78, 0.32–1.56, and 0.43–1.46 μmol/L for numbness, pungency, and astringency, respectively. The temporal dominance trends showed the following taste notes: Meranzin hydrate–numbing dominated, isomeranzin–numbing and pungent, and marmin–astringent and numbing. Description of a new species of Schwiebea (Sarcoptiformes: Acaridae) from Thailand

The genus Schwiebea Oudemans (Sarcoptiformes: Acaridae) includes notable agricultural pests that are widely distributed across diverse ecological environments. Despite their economic significance, mites of this genus in Thailand have been insufficiently studied. This research describes a new species, Schwiebea citrae sp. nov., collected from the roots of pomelo, Citrus maxima (Burm.) Merr., contributing to the broader knowledge of acarid diversity.

Impact of Organic Fertilization Strategies on Soil Bacterial Community and Honey Pomelo (Citrus maxima) Properties

Soil health is a critical factor in sustainable agriculture, particularly in fruit production, where fertilization strategies play a vital role in maintaining the soil quality and enhancing fruit production and quality. This study investigates the effects of different fertilization strategies on soil bacterial communities and honey pomelo (Citrus maxima) properties in Ji’an City, Jiangxi Province, China. Three fertilization treatments were compared: conventional fertilization (CF: botanical organic plus chemical compound fertilizers), organic material fermented fertilization (OF: organic material including duck manure fermented fertilizer plus chemical compound fertilizer), and a special honey pomelo fertilizer (SF: organic material fermented fertilizer only during the whole honey pomelo growing season). Soil samples were collected at two depths (0–20 cm and 20–40 cm) from nine plots (three treatments × three replicates) and analyzed for their soil properties, bacterial community diversity and composition, and fruit characteristics. The results indicate that the OF and SF significantly improved the soil pH, soil organic matter (SOM), and nutrient availability compared to the CF. Additionally, the OF and SF treatments led to a 13.6% and 16.6% increase in fruit weight, respectively, and higher bacterial diversity, although no significant differences were observed in fruit quality parameters such as vitamin C, soluble sugar, and titratable acid. Acidobacteriota, Proteobacteria, Actinobacteria, and Chloroflexi were the dominant bacterial phyla. The soil bacterial composition structures were significantly different among the different fertilization strategies, and were well explained by soil properties such as the pH, SOM, total phosphorus, and available nutrients. Our study suggests that applying fermented organic fertilizers which use duck manure as part of the raw materials, either alone or in combination with chemical compound fertilizers, increases honey pomelo fruit production and improves soil health, contributing to the sustainable development of orchards.

Enhancement of microbial safety and shelf life of salmon fillets by Lactiplantibacillus plantarum 299 V fermented in pomelo (Citrus maxima) peel extract

This study demonstrated the selective support of pomelo (Citrus maxima) peel extract (PPE) on Lactiplantibacillus plantarum 299 V growth, of which the resultant enhanced microbial safety and shelf life of salmon fillets. Incorporating PPE fermented with L. plantarum 299 V for 48 h (FPPE-48h) into alginate coating inhibited both the spiked Listeria monocytogenes and the spoilage - causing bacteria (psychotrophic bacteria, Enterobacteriaceae, Pseudomonas spp.) on salmon fillets during the 4-day chilled storage, resulting in 1–1.5 log CFU/g lower bacterial counts (P < 0.05). FPPE-48h effectively suppressed biogenic amine-producing bacteria, and remarkably retarded the accumulation of putrescine, cadaverine, histamine, and tyramine (P < 0.05). Last, the key antimicrobial substances generated during the late stage of PPE fermentation were profiled using non-target ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) analysis. This study exemplifies a successful approach of fermenting probiotic utilizing food by-products/food waste to enhance the safety and shelf life of ready-to-eat foods.

Production of neohesperidin from hesperetin by an engineered strain of Escherichia coli

Neohesperidin is a flavonoid glycoside widely used in the food and pharmaceutical industries. The current production of neohesperidin mainly relies on extraction from plants. Microbial fermentation demonstrates a promising prospect as an environmentally friendly, efficient, and economical method. In this study, we designed and constructed the biosynthetic pathway of neohesperidin in an Escherichia coli strain by introducing the glycosyltransferase UGT73B2 from Arabidopsis thaliana, rhamnose synthase VvRHM-NRS from Vitis vinifera, and rhamnose transferase Cm1,2RhaT from Citrus maxima. After optimization of the module and the uridine diphosphate (UDP)-glucose synthetic pathway, the engineered strain produced 4.64 g/L neohesperidin in a 5 L bioreactor, and the molar conversion rate of hesperetin was 45.8%. This has been the highest titer reported to date for the biosynthesis of neohesperidin in microorganisms. This study lays a foundation for the construction and application of strains with high yields of neohesperidin and provides a potential choice for the microbial production of other flavonoid glycosides.

Modelling and nondestructive quality prediction of Guanxi Honey Red Pomelo (&lt;i&gt;Citrus grandis&lt;/i&gt; (L.) Osbeck) based on shape characteristics and bitterness

Currently, it is difficult to distinguish the inner quality of pachycarpous fruits using nondestructive instruments, and trace nutrients and flavor compounds are rarely considered in fruit sorting. In this study, the external features and internal quality indices of an extensively farmed citrus fruit, the Guanxi honey red pomelo, were measured. The correlation between the inner and external quality was analyzed using Pearson correlation, canonical correlation, and grey correlation analyses. This research innovatively introduced bitter substances and revealed that the height-diameter ratio could serve as a predictor for ultra-size fruits, which partially reflects inner quality indices. Several grey formulas were developed to express these correlations and were fitted to a matrix, which demonstrated accuracies at all level 1 (C &lt; 0.35, P &gt; 0.95). Based on the matrix, fruit quality analysis of white and red pomelo showed high prediction accuracy in TSS and bitter substance indices (R2 = 0.6 – 0.8) and indicated that mature and symmetrical pomelo (height-diameter ratio of 0.9 – 1.1) exhibit a similar quality pattern. This study achieved rapid quality assessment using easy measurable shape characteristics and explored the application of characteristic bitter substances in the quality evaluation model, providing theoretical references and potential application guidance for pachycarpous fruit sorting, as well as introducing a new idea to incorporating characteristic quality into modeling index.

CgLS mediates limonene synthesis of main essential oil component in secretory cavity cells of Citrus grandis ‘Tomentosa’ fruits

d-Limonene is the predominant component of essential oil from Exocarpium Citri Grandis, known for its antibacterial, antioxidant, insecticidal, and anti-inflammatory properties. The synthesis, transport, and accumulation of d-limonene in Citrus grandis ‘Tomentosa’ fruits are regulated by limonene synthase (LS) and its associated regulatory genes. This study addresses the gap in understanding the spatiotemporal cytological changes of LS and its regulatory genes. Through cytochemical techniques, we investigated the distribution of essential oil in the plastids, endoplasmic reticulum, and vacuoles of secretory cavity cells. We identified the LS-encoding gene CgLS via transcriptomics and demonstrated in vitro that CgLS catalyzes the formation of d-limonene from geranyl diphosphate (GPP). Transient overexpression of CgLS increased monoterpene limonene accumulation, while TRV virus-induced gene silencing reduced it. CgLS expression levels and d-limonene content showed spatiotemporal consistency with fruit development, with in situ hybridization revealing predominant expression in secretory cavity cells. Immunocytochemical localization indicated that CgLS is primarily located in the endoplasmic reticulum, plastids, and vacuoles. Our findings suggest that CgLS is translated in the endoplasmic reticulum and transported to plastids and vacuoles where d-limonene synthesis occurs. This study provides comprehensive insights into the characteristics of CgLS and its role in d-limonene synthesis at the tissue, cellular, and molecular levels in C. grandis ‘Tomentosa’.

Towards sustainable fruit production: Identifying challenges and optimization strategies

CONTEXTThe transition to sustainable fruit production is vital for global sustainability and agricultural development. However, implementing these insights into practice faces significant challenges. OBJECTIVESPomelo (Citrus maxima), a prominent citrus species in tropical and subtropical regions, has been cultivated to enhance farmer profitability but increasing yields often comes at the cost of environmental degradation. This study uses pomelo as a case study to assess key factors influencing pomelo production and establish optimal practices that meet consumption-based food while co-benefits for environmental and economic sustainability. METHODSA comprehensive study was conducted by combining 1155 farm surveys (from 2010, 2018, and 2022) with 31 field experiments. Life cycle assessment (LCA), classification and regression tree (CART) analysis, and scenario analysis were employed to develop pathways for sustainable pomelo production in Pinghe County, Fujian Province, China. RESULTS AND CONCLUSIONSResults show no significant differences in pomelo yield across the three years; however, pomelo production in 2022 demonstrated a 60 %–67 % lower in environmental footprints and a 64 %–76 % higher benefit-cost ratio (BCR) compared to 2010 and 2018. Tree age was identified as a critical factor influencing pomelo yield, with optimal yields observed in trees older than 12.5 years. The rate of phosphorus fertilizer application was a key determinant of the phosphorus footprint (PF), which was positively correlated with phosphorus fertilizer rates. Similarly, nitrogen fertilizer application significantly impacted both carbon and nitrogen footprints (CF and NF) as well as the BCR. CF and NF increased with higher nitrogen fertilizer rates, while BCR exhibited a negative correlation with nitrogen fertilizer application. Implementing Science and Technology Backyards (STB) followed by farmer training of slightly more than pomelo nutrient requirements practice (SNRP) or coordinate management by farmers and researchers of matching pomelo nutrients requirement practice (MNRP) had 50 %–91 % lower environmental footprints per ton of pomelo produced and 15 %–44 % higher BCR compared to current farming practices (FP) via reducing 46 %–77 % N, 49 %–92 % P2O5, and 42 %–73 % K2O per hectare. Moreover, adopting 100 % MNRP in pomelo production, along with sustainable fertilizer practices and the implementation of innovative fertilizers, could reduce greenhouse gas (GHG) emissions (0.76 Tg), reactive nitrogen (Nr) emissions (8.76 Gg), phosphorus (P) losses (0.84 Gg), and augment net ecosystem economic benefits (NEEB) (0.94 billion CNY) at the county scale. SIGNIFICANCEOur study provides evidence-based strategies for achieving sustainable pomelo production through multi-stakeholder collaboration of STB, government, enterprises, and smallholders.

Excessive boron fertilization-induced toxicity is related to boron transport in field-grown pomelo trees.

Boron (B) is an essential micronutrient for plant growth and development; however, the process of B toxicity in citrus production is still poorly understood. We proposed a hypothesis that B toxicity in citrus trees is related to the characteristics of B transport from soil to leaf or fruit. For this, a field experiment was conducted for two treatments, control (B free or without B) and B fertilizer treatment (100 g Na2B4O7·10H2O plant-1), to investigate the effects on plant growth, nutrient uptake, fruit yield and quality, and B transport in 10-year-old pomelo trees [Citrus grandis (L.) Osbeck cv. Guanximiyou]. Our results showed that excess B fertilization directly led to B toxicity in pomelo trees by dramatically increasing soil total B and water-soluble B contents. B toxicity induced interveinal chlorosis in leaves and decreased leaf biomass and function, resulting in a decreased 45.3% fruit yield by reducing 30.6% fruit load and 21.4% single fruit weight. Also, B toxicity induced changes in mineral elements between leaf positions and fruit parts, in which the concentrations of B, potassium, and magnesium were increased while those of nitrogen and iron were decreased. Under B toxicity conditions, fruit quality parameters of total soluble solids (TSS), TSS/titratable acidity (TA), total soluble sugar, sucrose, pH, vitamin C, and total phenol contents decreased, which were regulated by the lower carbohydrate production in new leaves and the lower transport capacity in old leaves. Moreover, B toxicity significantly increased the transfer factor and bio-concentration factor of B in pomelo plants, with higher levels in leaf organs than in fruit organs. Taken together, excess B fertilization-induced B toxicity in pomelo trees, with induced growth inhibition and nutrient disorder, results in reduced fruit yield and quality, which are related to B transport from soil to organs. The findings of this study highlight the understanding of B toxicity in citrus plants and strengthen B management in pomelo production for high yield and high quality.

An arabinan from Citrus grandis fruits alleviates ischemia/reperfusion-induced myocardial cell apoptosis via the Nrf2/Keap1 and IRE1/GRP78 signaling pathways

Citrus grandis fruit is a famous traditional Chinese medicine with various bioactivities, including cardioprotective effects. Polysaccharides are one of the key active ingredients responsible for its cardioprotective effects. This study aimed to investigate the structure and cardioprotective effect of a homogeneous polysaccharide from C. grandis fruit (CGP80-1) and explore its mechanism against myocardial ischemia-reperfusion (MI/R) injury. Structure analysis showed that CGP80-1 (11,917 Da) is an arabinan with compact coil chain conformation, containing →5)-α-L-Araf-(1→, →3,5)-α-L-Araf-(1→, and →2,3,5)-α-L-Araf-(1→ as the backbone, as well as →5)-α-L-Araf-(1→ and t-α-L-Araf as side-chains substituted at the C2 and C3 positions. Pharmacological experiments showed that pre-treatment with CGP80-1 could effectively alleviate MI/R injury by improving endogenous antioxidant enzymes and cardiac enzymes, reducing reactive oxygen species levels, and regulating apoptosis-related proteins such as caspase-3, Bax, and Bcl-2. The protective effects were correlated with the Nrf2/Keap1 and IRE1/GRP78 signaling pathways. Further analysis of structure-activity relationships revealed that the myocardial protection effects of CGP80-1 might be attributed to its appropriate molecular weight, high arabinose content, and unique compact coil chain conformation. Overall, our results provide insight into the chemical structure of CGP80-1 and its mechanism of action, suggesting that CGP80-1 could be a candidate drug for myocardial protection.

Exploring the chemotaxonomic significance of selected Citrus essential oils through GC-MS/FID and multivariate correlation analyses

The volatile chemical composition and qualitative as well as quantitative variation of the leaf and peel essential oils of five species belonging to Citrus genus were studied. The investigated Citrus species were Citrus aurantifolia (Christm.) Swingle, Citrus limon L. (Osbeck), Citrus maxima (Burm.f.) Merr. R, Citrus medica L. and Citrus medica *Citrus limon. Essential oils were isolated by hydrodistillation and analysed using GC-MS/FID. In total 49 compounds comprising of 83.1-99.0% of the volatile compositions were identified in five leaf essential oils whereas 45 compounds comprising of 97-99.4% of the volatile compositions were identified in peel essential oils. Oxygenated monoterpenes were the predominant composition present in the leaf oils of all Citrus species whereas sesquiterpene hydrocarbons were the main composition found in C. maxima leaf oil (39.9%). Monoterpene hydrocarbons were predominating in the isolated peel oils. Principal component analysis of Citrus species was employed to provide a comprehensive evaluation of essential oil components. Both PCA and cluster analysis of both leaf and peel essential oils were classified into two clusters and then subsequent subclusters, characterized according to the major essential oil components. Monoterpene hydrocarbons, oxygenated hydrocarbons and diterpenes were identified as the significant category of compounds characterised for Citrus essential oils.

Irradiation as a quarantine treatment on the quality and sensory evaluation of ‘Thong Dee’ Thai pomelo (Citrus maxima (Burm.) Merr.) during storage

Irradiation as a quarantine treatment on the quality and sensory evaluation of ‘Thong Dee’ Thai pomelo (<i>Citrus maxima</i> (Burm.) Merr.) during storage

Structural analysis and biological activity of cell wall polysaccharides and enzyme-extracted polysaccharides from pomelo (Citrus maxima (Burm.) Merr.)

Pomelo peel is a valuable source of pectin, but research on its cell wall polysaccharides is limited. This study compared the cell wall polysaccharides of pomelo peel, enzyme-extracted polysaccharides of pomelo peel, and enzyme-extracted polysaccharides of whole pomelo fruit. Cell wall polysaccharides, including water-soluble pectin (WSP), chelator-soluble pectin (CSP), sodium carbonate-soluble pectin (NSP), 1 mol/L KOH soluble hemicellulose (KSH-1), and 4 mol/L KOH soluble hemicellulose (KSH-2), were obtained by sequence-extraction method. Total polysaccharides from whole pomelo fruit (TP) and peel-polysaccharides from pomelo pericarps (PP) were obtained using enzyme-extraction method. The structural, thermal, rheological, antioxidant properties, and wound healing effect in vitro were described for each polysaccharide. WSP had a uniform molecular weight distribution and high uronic acid (UA) content, suitable for commercial pectin. NSP had the highest Rhamnose (Rha)/UA ratio and a rich side chain with highest viscosity and water retention. PP displayed the highest DPPH radical scavenging activity and reducing capacity at 0.1 to 2.0 mg/mL concentration range, with an IC50 of 1.05 mg/mL for DPPH free radicals. NSP also demonstrated the highest hydroxyl radical scavenging activity and promoted Human dermal keratinocyte proliferation and migration at 10 μg/mL, suggesting potential applications in daily chemical and pharmaceutical industries.
