Which distribution to choose for deriving a species sensitivity distribution? Implications from analysis of acute and chronic ecotoxicity data

Species sensitivity distributions (SSDs) estimated by fitting a statistical distribution to ecotoxicity data are indispensable tools used to derive the hazardous concentration for 5 % of species (HC5) and thereby a predicted no-effect concentration in environmental risk assessment. Whereas various statistical distributions are available for SSD estimation, the fundamental question of which statistical distribution should be used has received limited systematic analysis. We aimed to address this knowledge gap by applying four frequently used statistical distributions (log-normal, log-logistic, Burr type III, and Weibull distributions) to acute and chronic SSD estimation using aquatic toxicity data for 191 and 31 chemicals, respectively. Based on the differences in the corrected Akaike’s information criterion (AICc) as well as visual inspection of the fitting of the lower tails of SSD curves, the log-normal SSD was generally better or equally good for the majority of chemicals examined. Together with the fact that the ratios of HC5 values of other alternative SSDs to those of log-normal SSDs generally fell within the range 0.1–10, our findings indicate that the log-normal distribution can be a reasonable first candidate for SSD derivation, which does not contest the existing widespread use of log-normal SSDs.

Probabilistic risk assessment of microplastics in Tai Lake, China

Microplastics are a global environmental concern, especially in freshwater ecosystems. Despite the studies in specific regions of Tai lake, a gap persists in understanding the comprehensive risk of MPs across the entire watershed. Therefore, this study offers an overview of MPs abundance and assesses ecotoxicological risk by employing acute and chronic species sensitivity distributions, which consider the effects triggered by MPs. The concentrations of MPs ranged from 0 to 18.6 particles/L within the lake, 1.56 to 1.42 × 102 particles/L in the rivers, and 0.16 to 0.7 particles/L in the estuaries. Certain areas, particularly the northwest and southeast regions, exhibit higher concentrations. Using existing toxicity data, this study calculated predicted no effect concentrations for acute and chronic exposure of MPs to freshwater species, resulting in values of 11.5 and 31.72 particles/L, respectively. The probabilistic risk assessment indicates that the average risk possibility of MPs in Tai lake was 16 %. Moreover, the risk characterization ratio indicated that 22 % of the locations in Tai lake showed an acute ecological risk, while 7.4 % exhibit a chronic ecological risk. The assessment concluded that MPs reported in the literature could pose a considerable risk to Tai lake biota. However, the risk associated with MPs followed descending order: river >lake > estuary waters. Our research supplies valuable insights for the assessment of ecological risks associated with MPs on a whole watershed scale.

Hazard screening of contaminants of emerging concern (CECs) in Sweden’s three largest lakes and their associated rivers

Persistent, mobile, and toxic (PMT) substances have recently garnered increased attention by environmental researchers, the water sector and environmental protection agencies. In this study, acute and chronic species sensitivity distributions (SSDs) were retrieved from literature data for previously quantified contaminants of emerging concern (CECs) in Swedish surface waters (n = 92) and risk quotients (RQ) were calculated. To better understand the characteristics of the detected CECs in non-urban lake sites (n = 71), these compounds were checked against established criteria for potentially toxic PMs (PM(T)s) and occurrence in the aquatic environment, respectively. For the CECs with missing SSDs (n = 15 [acute], n = 41 [chronic]), ecotoxicity data were extracted for eight taxonomic groups, and if data were sufficient (n ≥ 3), SSDs were derived. The retrieved and newly developed SSDs were then used in an environmental hazard assessment (EHA) in the investigated Swedish rivers and lakes. In the rivers, 8 CECs had RQ> 1 in at least one location, and 20 CECs posed a moderate risk (0.01 < RQ < 1). In total, 21 of the 71 detected substances had already been identified as PM(T)/vPvM substances. Our study shows the importance of studying field data at large spatial scale to reveal potential environmental hazards far from source areas.

Influence of climate change and pesticide use practices on the ecological risks of pesticides in a protected Mediterranean wetland: A Bayesian network approach

Pollution by agricultural pesticides is one of the most important pressures affecting Mediterranean coastal wetlands. Pesticide risks are expected to be influenced by climate change, which will result in an increase of temperatures and a decrease in annual precipitation. On the other hand, pesticide dosages are expected to change given the increase in pest resistance and the implementation of environmental policies like the European ´Farm-to-Fork` strategy, which aims for a 50 % reduction in pesticide usage by 2030. The influence of climate change and pesticide use practices on the ecological risks of pesticides needs to be evaluated making use of realistic environmental scenarios. This study investigates how different climate change and pesticide use practices affect the ecological risks of pesticides in the Albufera Natural Park (Valencia, Spain), a protected Mediterranean coastal wetland. We performed a probabilistic risk assessment for nine pesticides applied in rice production using three climatic scenarios (for the years 2008, 2050 and 2100), three pesticide dosage regimes (the recommended dose, and 50 % increase and 50 % decrease), and their combinations. The scenarios were used to simulate pesticide exposure concentrations in the water column of the rice paddies using the RICEWQ model. Pesticide effects were characterized using acute and chronic Species Sensitivity Distributions built with toxicity data for aquatic organisms. Risk quotients were calculated as probability distributions making use of Bayesian networks. Our results show that future climate projections will influence exposure concentrations for some of the studied pesticides, yielding higher dissipation and lower exposure in scenarios dominated by an increase of temperatures, and higher exposure peaks in scenarios where heavy precipitation events occur right after pesticide application. Our case study shows that pesticides such as azoxystrobin, difenoconazole and MCPA are posing unacceptable ecological risks for aquatic organisms, and that the implementation of the ´Farm-to-Fork` strategy is crucial to reduce them.

Large-scale monitoring and risk assessment of microplastics in the Amazon River

Microplastics (MPs) are one of the most widespread contaminants worldwide, yet their risks for freshwater ecosystems have seldom been investigated. In this study, we performed a large monitoring campaign to assess the presence and risks of MPs in Amazonian freshwater ecosystems. We investigated MP pollution in 40 samples collected along 1500 km in the Brazilian Amazon, including the Amazon River, three major tributaries, and several streams next to the most important urban areas. MPs in the 55–5000 µm size range were characterized (size, shape, color) by microscopy and identified (polymer composition) by infrared spectroscopy. Ecotoxicological risks were assessed using chronic Species Sensitivity Distributions for effects triggered by food dilution and tissue translocation using data alignment methods that correct for polydispersity of environmental MPs and bioaccessibility. This study shows that MPs are ubiquitous contaminants in Amazonian freshwater ecosystems, with measured concentrations (55–5000 µm) ranging between 5 and 152 MPs/m3 in the Amazon River and its main tributaries, and between 23 and 74,550 MPs/m3 in urban streams. The calculated Hazardous Concentration for the 5% of species (HC5) derived from the SSDs for the entire MP range (1–5000 µm) were 1.6 × 107 MPs/m3 (95% CI: 1.2 × 106 – 4.0 × 108) for food dilution, and 1.8 × 107 MPs/m3 (95% CI: 1.5 × 106 – 4.3 × 108) for translocation. Rescaled exposure concentrations (1–5000 µm) in the Amazon River and tributaries ranged between 6.0 × 103 and 1.8 × 105 MPs/m3, and were significantly lower than the calculated HC5 values. Rescaled concentrations in urban streams ranged between 1.7 × 105 and 5.7 × 108 MPs/m3, and exceeded both calculated HC5 values in 20% of the locations. This study shows that ecological impacts by MP contamination are not likely to happen in the Amazon River and its major tributaries. However, risks for freshwater organisms may be expected in near densely populated areas, such as the cities of Manaus or Belem, which have limited wastewater treatment facilities.

Chronic effects of clothianidin to non-target soil invertebrates: Ecological risk assessment using the species sensitivity distribution (SSD) approach

This study aimed to assess the chronic toxicity and risk of clothianidin in a seed dressing formulation to non-target soil invertebrates. The toxicity assays were performed with two oligochaetes (earthworms Eisenia andrei and enchytraeids Enchytraeus crypticus) and three collembolans (Folsomia candida, Proisotoma minuta and Sinella curviseta) species following ISO protocols. Risk assessment (via Hazard Quotient approach – HQ) was based on the hazardous concentrations for 95% of the species (HC5), derived from chronic Species Sensitivity Distributions (SSD) for clothianidin, and on its predicted environmental concentrations (PEC). Four SSD scenarios were generated with literature and/or this study data, following different data selection criteria (i.e., general, only data from tests using similar formulations, similar soils, or identical soil/formulation). In our experiments, a higher clothianidin toxicity (EC50-based) was found for collembolans (varying from 0.11 to 0.28 mg kg−1 between species) followed by the earthworms (4.35 mg kg−1), while the enchytraeids were the least sensitive (33.5 mg kg−1). HQ indicated a significant risk of clothianidin to soil invertebrates because the estimated PEC were at least 16.6 times higher than HC5 and are expected to affect the whole group of collembolans. Despite the criteria for data inclusion have influenced the HC5 values, no substantial changes were observed for the risk outcomes. To our knowledge, this is the first study assessing the chronic ecological risk of clothianidin to beneficial soil fauna based on a probabilistic SSD approach. Data from this study can help to derive more reliable protection thresholds for clothianidin in soils.

Can We Reasonably Predict Chronic Species Sensitivity Distributions from Acute Species Sensitivity Distributions?

Estimation of species sensitivity distributions (SSDs) is an essential way to estimate the hazardous concentration for 5% of the species (HC5) and thus to derive a "safe" concentration. Here, we examined whether we can reasonably predict SSDs based on chronic no-observed-effect concentration or level (chronic SSDs) from SSDs based on acute median effective/lethal concentration (acute SSDs) by analyzing log-normal SSDs of 150 chemicals. Chronic SSD means were, on average, 10 times lower than acute SSD means. The standard deviations (SDs) of acute and chronic SSDs closely overlapped. Our detailed analysis suggests that the acute SSD SD can be used as an initial estimate of the chronic SSD SD if the number of tested species is ≥10. There were no significant differences in the ratios of chronic to acute SSD means or SDs among three different modes of action. The HC5 of chronic SSDs was, on average, 10 times lower than the acute SSD HC5. We suggest that multiplication of the acute HC5 by a factor of 0.1 is a defensible way to obtain a first approximation of the chronic HC5, particularly when relative ecological risks of chemicals are being evaluated. Further study is needed to develop methods for a more accurate estimation of chronic SSDs.

Species Sensitivity Distributions for Nonylphenol to Estimate Soil Hazardous Concentration.

Nonylphenol is an endocrine-disrupting chemical that mimics estrogenic activity. Few studies have investigated the soil ecotoxicity of nonylphenol in the environment, based on probabilistic approaches. The present study generated soil toxicity data for nonylphenol through bioassays that determined the acute and chronic species sensitivity distributions and estimated the hazardous concentrations of nonylphenol in soil in order to protect soil ecosystems. We used eight soil-based organisms from six taxonomic groups for acute assays and five soil-based organisms from four taxonomic groups for chronic assays. The hazardous concentration values of nonylphenol in soil, based on acute and chronic species sensitivity distributions, were estimated using compiled data from the present study, as well as previous studies. This is the first study that generated sufficient data to develop species sensitivity distributions for nonylphenol in soil, and to determine hazardous concentrations of nonylphenol for soil environments.

Estimation of vanadium water quality benchmarks for the protection of aquatic life with relevance to the Athabasca Oil Sands region using species sensitivity distributions.

Elevated vanadium (V) concentrations in oil sands coke, which is produced and stored on site of some major Athabasca Oil Sands companies, could pose a risk to aquatic ecosystems in northern Alberta, Canada, depending on its future storage and utilization. In the present study, V toxicity was determined in reconstituted Athabasca River water to various freshwater organisms, including 2 midge species (Chironomus dilutus and Chironomus riparius; 4-d and 30-d to 40-d exposures) and 2 freshwater fish species (Oncorhynchus mykiss and Pimephales promelas; 4-d and 28-d exposures) to facilitate estimation of water quality benchmarks. The acute toxicity of V was 52.0 and 63.2 mg/L for C. dilutus and C. riparius, respectively, and 4.0 and 14.8 mg V/L for P. promelas and O. mykiss, respectively. Vanadium exposure significantly impaired adult emergence of C. dilutus and C. riparius at concentrations ≥16.7 (31.6% reduction) and 8.3 (18.0% reduction) mg/L, respectively. Chronic toxicity in fish presented as lethality, with chronic 28-d LC50s of 0.5 and 4.3 mg/L for P. promelas and O. mykiss, respectively. These data were combined with data from the peer-reviewed literature, and separate acute and chronic species sensitivity distributions (SSDs) were constructed. The acute and chronic hazardous concentrations endangering only 5% of species (HC5) were estimated as 0.64 and 0.05 mg V/L, respectively. These new data for V toxicity to aquatic organisms ensure that there are now adequate data available for regulatory agencies to develop appropriate water quality guidelines for use in the Athabasca Oil Sands region and elsewhere. Until then, the HC5 values presented in the present study could serve as interim benchmarks for the protection of aquatic life from exposure to hazardous levels of V in local aquatic environments. Deriving acute and chronic predicted no effect concentrations of pharmaceuticals and personal care products based on species sensitivity distributions

Pharmaceuticals and personal care products (PPCPs), as emerging contaminants, have been detected in various environmental matrices and caused adverse effects on human health and the ecosystem. But water quality criterias (WQCs) of PPCPs for protecting aquatic environment are lacking, which hinders the environmental management of these emerging contaminants. In the present study, in order to support their WQC derivation, acute and chronic hazardous concentrations for 5% of species (HC5s) of some frequently detected PPCPs in China were calculated based on acute and chronic species sensitivity distributions (SSDs), respectively, using both parametric (log-normal and log-logistic) and nonparametric bootstrap approaches. The groups of aquatic species used in SSDs included planktons, zooplanktons, invertebrates and vertebrates. Acute and chronic predicted no effect concentrations (PNECs) were derived from the HC5s. The acute PNECs of the selected PPCPs were in a range from 1.1 to 4993μg/L. While the chronic PNECs were one or two orders of magnitude lower than the acute PNECs, with a range from 0.02 to 298μg/L. Among these PPCPs, the compound with the highest acute effect on the aquatic environment was clarithromycin while erythromycin was the one with the highest chronic toxicity effect. Among the studied PPCPs, erythromycin caused a relatively higher aquatic ecological risk in China. This study helps derive WQCs of PPCPs in the aquatic environment, which is essential for environmental managment of these emerging contaminants.

A microcosm study to support aquatic risk assessment of nickel: Community‐level effects and comparison with bioavailability‐normalized species sensitivity distributions

The aquatic risk assessment for nickel (Ni) in the European Union is based on chronic species sensitivity distributions and the use of bioavailability models. To test whether a bioavailability-based safe threshold of Ni (the hazardous concentration for 5% of species [HC5]) is protective for aquatic communities, microcosms were exposed to 5 stable Ni treatments (6-96 μg/L) and a control for 4 mo to assess bioaccumulation and effects on phytoplankton, periphyton, zooplankton, and snails. Concentrations of Ni in the periphyton, macrophytes, and snails measured at the end of the exposure period increased in a dose-dependent manner but did not indicate biomagnification. Abundance of phytoplankton and snails decreased in 48 μg Ni/L and 96 μg Ni/L treatments, which may have indirectly affected the abundance of zooplankton and periphyton. Exposure up to 24 μg Ni/L had no adverse effects on algae and zooplankton, whereas the rate of population decline of the snails at 24 μg Ni/L was significantly higher than in the controls. Therefore, the study-specific overall no-observed-adverse-effect concentration (NOAEC) is 12 μg Ni/L. This NOAEC is approximately twice the HC5 derived from a chronic species sensitivity distribution considering the specific water chemistry of the microcosm by means of bioavailability models. Thus, the present study provides support to the protectiveness of the bioavailability-normalized HC5 for freshwater communities.

Application of U.S. EPA guidelines in a bioavailability‐based assessment of ambient water quality criteria for zinc in freshwater

The United States Environmental Protection Agency's (U.S. EPA) current ambient water quality criteria (AWQC) for zinc in freshwater are hardness-based and were last updated in 1995. The acute and chronic freshwater toxicity databases have since expanded substantially and the U.S. EPA's minimum phylogenetic diversity requirements for chronic zinc toxicity are now met (an acute:chronic ratio was previously required). Additionally, several acute and chronic biotic ligand models (BLMs) for zinc have since been developed and validated for freshwater organisms. Using the expanded toxicity database and existing BLMs, we developed a unified zinc BLM that could efficiently predict both acute and chronic toxicity over a wide range of zinc bioavailabilities. The unified BLM, developed by objectively averaging the biotic ligand binding constants for zinc (Zn(2+)) and competing cations (Ca(2+), Mg(2+), Na(+), H(+)) from existing BLMs, performed better in predicting toxicity to a diverse set of organisms than any individual existing BLM. Performance of the unified BLM was further improved by optimizing the biotic ligand binding constant for the ZnOH(+) species. The updated freshwater zinc toxicity database and unified BLM were then used to estimate the fifth percentiles of the acute and chronic species sensitivity distributions following the U.S. EPA guidelines for AWQC development.

Discrepancies in the acute versus chronic toxicity of compounds with a designated narcotic mechanism

In this study, it was illustrated that even for certain simple organic compounds with a designated mode of action (MOA) (i.e. narcotic toxicity) unexpected differences in acute and chronic toxicity can be observed. In a first part of the study, species sensitivity distributions (SSDs) based on either acute or chronic toxicity data of three narcotic test compounds (methanol, ethanol and 2-propanol) were constructed. The results of the acute SSDs were as expected for narcotic compounds: rather similar sensitivity and small differences in toxicity were observed among different species. On the contrary, the chronic SSDs of methanol and ethanol indicated larger interspecies variation in sensitivity. Furthermore, the chronic toxicity trend (ethanol>methanol>2-propanol) was unexpectedly different from the acute toxicity trend (2-propanol>ethanol>methanol) and acute versus chronic extrapolation could not be successfully described for methanol and ethanol using an ACR of 10 (as suggested for narcotic compounds). In contrast to the interspecies approach in the first part of this study, the second part of the study was focused on the assessment of acute and chronic toxicity of the three test compounds in Daphnia magna, which was identified as one of the most sensitive organisms to methanol and ethanol. Here, the differences in acute and chronic toxicity trend were in accordance to the results of the SSDs. The enhancement of membrane penetration due to the small molecular size of methanol and ethanol, in combination with the higher toxicity of their respective biotransformation products were suggested as potential causes of the increased chronic toxicity. Furthermore, it was stressed that larger awareness of these irregularities in acute to chronic extrapolations of narcotic compounds is required and should receive additional attention in further environmental risk assessment procedure.

Relevance of Risk Predictions Derived from a Chronic Species Sensitivity Distribution with Cadmium to Aquatic Populations and Ecosystems

Criteria to protect aquatic life are intended to protect diverse ecosystems, but in practice are usually developed from compilations of single-species toxicity tests using standard test organisms that were tested in laboratory environments. Species sensitivity distributions (SSDs) developed from these compilations are extrapolated to set aquatic ecosystem criteria. The protectiveness of the approach was critically reviewed with a chronic SSD for cadmium comprising 27 species within 21 genera. Within the data set, one genus had lower cadmium effects concentrations than the SSD fifth percentile-based criterion, so in theory this genus, the amphipod Hyalella, could be lost or at least allowed some level of harm by this criteria approach. However, population matrix modeling projected only slightly increased extinction risks for a temperate Hyalella population under scenarios similar to the SSD fifth percentile criterion. The criterion value was further compared to cadmium effects concentrations in ecosystem experiments and field studies. Generally, few adverse effects were inferred from ecosystem experiments at concentrations less than the SSD fifth percentile criterion. Exceptions were behavioral impairments in simplified food web studies. No adverse effects were apparent in field studies under conditions that seldom exceeded the criterion. At concentrations greater than the SSD fifth percentile, the magnitudes of adverse effects in the field studies were roughly proportional to the laboratory-based fraction of species with adverse effects in the SSD. Overall, the modeling and field validation comparisons of the chronic criterion values generally supported the relevance and protectiveness of the SSD fifth percentile approach with cadmium.

First Derivation of Predicted-No-Effect Values for Freshwater and Terrestrial Ecosystems Exposed to Radioactive Substances

The FASSET Radiation Effects Database (FRED) constitutes a unique structured resource of the biological effects of ionizing radiation on non-human species mainly from temperate ecosystems, encompassing 26,000 primary data entries. Quality-assessed data were extracted from FRED and dose-effect relationships were constructed to provide estimates of ED50 and EDR10. These estimates are Doses (or Dose Rates) related to the percent change in the average level of the endpoint for a particular effect (50% or 10% for acute or chronic exposure regimes, respectively). Acute and chronic Species Sensitivity Distributions (SSDs) were built on the basis of these data sets, and the Assessment Factor Method (AFM) was applied when data were too scarce. The Hazardous Dose corresponding to 5% of species acutely affected at the 50% effect level varied from 1 to 5.5 Gy according to the ecosystem. For chronic gamma external irradiation exposure, no-effect values varied from 10 microGy/h for freshwaters through application of the AFM to 67 microGy/h for terrestrial ecosystems, corresponding to the 5th percentile of the non-weighted SSD (vs 229 microGy/h when trophic weights are applied). These values are higher by ca. x50 to x100 than the upper bound of natural background, and lower than dose rates triggering effects at individual levels on contaminated sites.

Acute‐to‐chronic species sensitivity distribution extrapolation

Seeking to make greater use of available data for risk assessment of substances, we constructed, for the situation in which chronic data are limited or even nonexistent but acute data are relatively large, an acute to chronic transformation (ACT) methodology based on the concept of species sensitivity distributions (SSDs). This ACT methodology uses a comparison of acute and chronic SSDs, separately for vertebrate data (with 22 substances) and for invertebrate data (with 15 substances). Rather than comparing an acute toxicity value with a chronic value, as when calculating an acute to chronic ratio (ACR), samples of acute and chronic data corresponding to the same category of species were compared. Starting from a sample of acute data, the ACT methodology showed relationships that enable the creation of a sample of predicted chronic values. This sample can then be used to calculate a predicted chronic hazardous concentration potentially affecting 5% of species (HC5%), just as with a sample of real chronic toxicity values. This ACT approach was tested on 11 substances. For each substance, the real chronic HC5% and the predicted chronic HC5% were calculated and compared. The ratio between chronic HC5% and ACT HC5% was, on average, 1.6 and did not exceed 4.4 for the 11 substances studied.

Assessing acute and chronic copper risks to freshwater aquatic life using species sensitivity distributions for different taxonomic groups

Using copper as an example, we present a method for assessing chemical risks to an aquatic community using species sensitivity distributions (SSDs) for different taxonomic groups. This method fits probability models to chemical exposure and effects data to estimate the percentage of aquatic species potentially at risk and expands on existing probabilistic risk assessment methodologies. Due to a paucity of chronic toxicity data for many chemicals, this methodology typically uses an acute-chronic ratio (ACR) to estimate the chronic effects distribution from the acute effects distribution. We expanded on existing methods in two ways. First, copper SSDs were developed for different organism groups (e.g., insects, fish) that share similar sensitivities or ecological functions. Integration of exposure and effects distributions provides an estimate of which organism groups may be at risk. These results were then compared with a site-specific food web, allowing an estimation of whether key food web components are potentially at risk and whether the overall aquatic community may be at risk from the perspective of ecosystem function. Second, chronic SSDs were estimated using the relationship between copper ACRs and acute toxicity (i.e., the less acutely sensitive a species, the larger the ACR). This correction in the ACR removes concerns previously identified with use of the ACR and allows evaluation of a significantly expanded chronic data set with the same approach as that for assessing acute risks.
