Blood flow disturbances in the brain may lead to stroke and continuous decrease in blood flow compromises memory. The latter will lead to confusion and progression of dementia. In addition, N-methyl-D-aspartate (NMDA) receptor system in the brain has been implicated in neuronal plasticity, neurotoxicity, learning and memory. The present study aims to evaluate the role of NMDA-receptor system particularly glutamate-mediated long term potentiation (LTP) in the processes of learning and memory formation in chronic cerebral hypoperfusion rats. Cerebral hypoperfusion was developed via permanent ligation of the common carotid arteries to decrease blood flow to the brain. Common carotid arteries of the male Sprague-Dawley rats (250-300g) were permanently ligated (2VO) and sham operated-group were undergone the same surgery without ligation. LTP was recorded on day 28 after surgery in urethane-anesthesized rats. The field excitatory postsynaptic potential (fEPSP) was found to be improved in rats treated with NMDA receptor antagonist, MK801 (0.25mg/kg, intraperitoneal injection) compared to non-treated rats recorded in the hippocampus CA1. The findings confirm the important role of the NMDA-receptor system in reversing the cognitive function of chronic cerebral hypoperfusion rats.
