Maternal transfer of F-53B inhibited neurobehavior in zebrafish offspring larvae and potential mechanisms: Dopaminergic dysfunction, eye development defects and disrupted calcium homeostasis

Maternal exposure to environment toxicants is an important risk factor for neurobehavioral health in their offspring. In our study, we investigated the impact of maternal exposure to chlorinated polyfluoroalkyl ether sulfonic acids (Cl-PFESAs, commercial name: F-53B) on behavioral changes and the potential mechanism in the offspring larvae of zebrafish. Adult zebrafish exposed to Cl-PFESAs (0, 0.2, 2, 20 and 200 μg/L) for 21 days were subsequently mated their embryos were cultured for 5 days. Higher concentrations of Cl-PFESAs in zebrafish embryos were observed, along with, reduced swimming speed and distance travelled in the offspring larvae. Molecular docking analysis revealed that Cl-PFESAs can form hydrogen bonds with brain-derived neurotropic factor (BDNF), protein kinase C, alpha, (PKCα), Ca2+-ATPase and Na, K - ATPase. Molecular and biochemical studies evidenced Cl-PFESAs induce dopaminergic dysfunction, eye developmental defects and disrupted Ca2+ homeostasis. Together, our results showed that maternal exposure to Cl-PFESAs lead to behavioral alteration in offspring mediated by disruption in Ca2+ homeostasis, dopaminergic dysfunction and eye developmental defects.

Bioaccumulation, tissue distribution, and maternal transfer of novel PFOS alternatives (6:2 Cl-PFESA and OBS) in wild freshwater fish from Poyang Lake, China

As emerging alternatives to perfluorooctane sulfonate (PFOS), 6:2 chlorinated polyfluoroalkyl ether sulfonic acid (6:2 Cl-PFESA) and sodium p-perfluorous nonenox-benzenesulfonate (OBS) were frequently detected in the four freshwater fish species collected from Poyang Lake. Median concentrations of 6:2 Cl-PFESA and OBS in fish tissues were 0.046–6.0 and 0.46–5.1 ng/g wet weight, respectively. The highest concentrations of 6:2 Cl-PFESA was found in fish livers, whereas OBS was found in the pancreas, brain, gonads, and skin. The tissue distribution pattern of 6:2 Cl-PFESA is similar to that of PFOS. The tissue/liver ratios of OBS were higher than those of PFOS, suggesting that OBS has a greater tendency to transfer from the liver to other tissues. The logarithmic bioaccumulation factors (log BAFs) of 6:2 Cl-PFESA in three carnivorous fish species were greater than 3.7, whereas those of OBS were less than 3.7, indicating that 6:2 Cl-PFESA had a strong bioaccumulation potential. Notably, sex- and tissue-specific bioaccumulation of OBS has also been observed in catfish. Most tissues (except the gonads) exhibited higher OBS concentrations in males than in females. However, no differences were found for 6:2 Cl-PFESA and PFOS. Maternal transfer efficiency of OBS was higher than that of 6:2 Cl-PFESA and PFOS in catfish (p < 0.05), indicating that OBS presents a higher risk of exposure to males and offspring through maternal offloading.

Bioaccumulation and risk mitigation of legacy and novel perfluoroalkyl substances in seafood: Insights from trophic transfer and cooking method

Per- and polyfluoroalkyl substances (PFAS) have widespread application in industrial and civil areas due to their unique physical and chemical properties. With the increasingly stringent regulations of legacy PFAS, various novel alternatives have been developed and applied to meet the market demand. Legacy and novel PFAS pose potential threats to the ecological safety of coastal areas, however, little is known about their accumulation and transfer mechanism, especially after cooking treatment. This study investigated the biomagnification and trophic transfer characteristics of PFAS in seafood from the South China Sea, and assessed their health risks after cooking. Fifteen target PFAS were all detected in the samples, of which perfluorobutanoic acid (PFBA) was dominant with concentrations ranging from 0.76 to 4.12 ng/g ww. Trophic magnification factors (TMFs) > 1 were observed for perfluorooctane sulfonate (PFOS) and 6:2 chlorinated polyfluoroalkyl ether sulfonic acid (F-53B), indicating that these compounds experienced trophic magnification in the food web. The effects of different cooking styles on PFAS occurrence were further explored and the results suggested that ΣPFAS concentrations increased in most organisms after baking, while ΣPFAS amounts decreased basically after boiling and frying. Generally, there is a low health risk of exposure to PFAS when cooked seafood is consumed. This work provided quantitative evidence that cooking methods altered PFAS in seafood. Further, suggestions to mitigate the health risks of consuming PFAS-contaminated seafood were provided.

Integration of metabolomics and proteomics reveals the underlying hepatotoxic mechanism of perfluorooctane sulfonate (PFOS) and 6:2 chlorinated polyfluoroalkyl ether sulfonic acid (6:2 Cl-PFESA) in primary human hepatocytes

Perfluorooctane sulfonate (PFOS) and its alternative 6:2 chlorinated polyfluorinated ether sulfonate (6:2 Cl-PFESA) are ubiquitous in various environmental and human samples. They have been reported to have hepatotoxicity effects, but the potential mechanisms remain unclear. Herein, we integrated metabolomics and proteomics analysis to investigate the altered profiles in metabolite and protein levels in primary human hepatocytes (PHH) exposed to 6:2 Cl-PFESA and PFOS at human exposure relevant concentrations. Our results showed that 6:2 Cl-PFESA exhibited higher perturbation effects on cell viability, metabolome and proteome than PFOS. Integration of metabolomics and proteomics revealed that the alteration of glycerophospholipid metabolism was the critical pathway of 6:2 Cl-PFESA and PFOS-induced lipid metabolism disorder in primary human hepatocytes. Interestingly, 6:2 Cl-PFESA-induced cellular metabolic process disorder was associated with the cellular membrane-bounded signaling pathway, while PFOS was associated with the intracellular transport process. Moreover, the disruption effects of 6:2 Cl-PFESA were also involved in inositol phosphate metabolism and phosphatidylinositol signaling system. Overall, this study provided comprehensive insights into the hepatic lipid toxicity mechanisms of 6:2 Cl-PFESA and PFOS in human primary hepatocytes.

Disclosing the bioaccumulation and biomagnification behaviors of emerging per/polyfluoroalkyl substances in aquatic food web based on field investigation and model simulation

Emerging poly/perfluoroalkyl substances (PFASs) have been widely detected in the environment, but their bioaccumulation and biomagnification behaviors are not well understood. We collected surface water, sediment, and various aquatic organisms from Lake Taihu, China. Several emerging PFASs, such as fluorotelomer sulfonates (FTSs), hexafluoropropylene oxides (HFPOs), and chlorinated polyfluoroalkyl ether sulfonic acids (Cl-PFESAs) were frequently detected in water and sediment samples. The concentrations of HFPO trimer acid (HFPO-TA), 4,8-dioxa-3 H-per-fluorononanoate, and FTSs were remarkably higher than those reported previously, indicating that their application is increasing in Taihu Basin. These emerging PFASs displayed higher sediment/water partitioning coefficients (log Koc) than the corresponding perfluoroalkyl acids (PFAAs) with the same perfluorinated carbon chain length. HFPOs and Cl-PFESAs were more labile to deposit in fish livers than perfluorooctanoic and perfluorooctane sulfonic acids, respectively. Both field investigations and model simulations indicated that HFPO-TA and Cl-PFESAs, as well as the hydrogen-substituted analogs of 6:2 Cl-PFESA (6:2 H-PFESA), were biomagnified along the aquatic food chain. The bioaccumulation model simulation revealed that the accumulation of these emerging PFASs in fish was mainly through dietary intake, whereas gill respiration and fecal excretion facilitated their elimination. Metabolic transformation might also contribute to their elimination relative to the legacy ones.

Associations between emerging chlorinated polyfluoroalkyl ether sulfonic acids exposure and birth size

As an alternative to legacy perfluorooctane sulfonate (PFOS), the wide usage of chlorinated polyfluoroalkyl ether sulfonic acid (Cl-PFESA) in China has generated considerable concerns. The study aimed to investigate the associations between cord serum emerging Cl-PFESA (6:2 Cl-PFESA and 8:2 Cl-PFESA) levels and suboptimal birth size, including low birth weight (LBW), macrosomia, small or large for gestational age (SGA or LGA) among 1048 newborns in Wuhan, China. 6:2 Cl-PFESA and 8:2 Cl-PFESA were 0.76 ng/mL and 0.03 ng/mL, respectively, higher than previously reported levels in newborns from other cities in China. We did not find significant associations between the Cl-PFESAs and suboptimal birth size when considering each Cl-PFESA as a single exposure. However, when integrating emerging Cl-PFESAs and legacy PFASs (PFOS and perfluorooctanoate acid (PFOA)) as a mixture using Bayesian kernel machine regression (BKMR) models, we found that the mixture of legacy PFASs and emerging Cl-PFESAs had inverse associations with SGA and LGA. Two legacy PFASs and two Cl-PFESAs had similar contributions to the association with SGA, whereas the majority of the association with LGA was driven by PFOS. The results demonstrated these legacy PFASs were still the predominant PFASs associated with suboptimal birth size while emerging Cl-PFESAs had a less but nonnegligible contribution to the association with SGA. Further studies in different regions with varied Cl-PFESA levels are needed to replicate the associations.

Preconception exposure to perfluoroalkyl and polyfluoroalkyl substances and couple fecundity: A couple-based exploration

Numerous studies have examined the adverse health effects of perfluoroalkyl and polyfluoroalkyl substances (PFAS) but it remains unclear whether preconception exposure to PFAS affects couple fecundity. This prospective preconception cohort study with 936 Chinese couples aimed to comprehensively assess the effects of PFAS on couple fecundity [measured by the time to pregnancy (TTP)] and infertility (i.e., TTP > 12 menstrual cycles) with a focus on the effects of partner-specific exposure and joint-effects of couple-based exposure. Twenty-five PFAS were quantified in plasma from each partner, including seven branched isomers, two chlorinated polyfluoroalkyl ether sulfonic acids, four emerging PFAS replacements [i.e., 6:2 fluorotelomer phosphate diester (6:2 diPAP) and three short-chain alternatives: perfluoro-n-butanoicacid, perfluorobutane sulfonate and perfluoroheptanoic acid (PFHpA)]. Using a two-phase regression approach composed of elastic net regression and principal component analysis, we found that exposure to 6:2 diPAP and PFHpA rather than legacy PFAS in women and the couple-based exposure patterns characterized by high level of female 6:2 diPAP were significantly associated with reduced couple fecundity, which was independent of the adjustment of co-exposed PFAS homogenous from both partners. For example, a ln unit increase in female 6:2 diPAP was associated with 15 % [fecundity odds ratio (FOR) = 0.85, 95 %CI: 0.76, 0.96)] lower odds of couple fecundability (i.e., longer TTP) and 45 % increased risk of infertility [OR = 1.45 (95 %CI: 1.16, 1.81)], respectively. While most PFAS in men were not associated with couple fecundity, certain PFAS (e.g., perfluorohexane sulfonic acid) in men were negatively associated with infertility risk. However, the combined effects of PFAS mixture in couples were nonsignificant. Our findings suggest that PFAS in men and women may exert different impacts on couple fecundity. Preconception exposure to 6:2 diPAP and PFHpA in women may have the potential to impair couple fecundity.

Emerging and Legacy Per- and Polyfluoroalkyl Substances in an Elderly Population in Jinan, China: The Exposure Level, Short-Term Variation, and Intake Assessment.

Human exposure to per- and polyfluoroalkyl substances (PFASs) has gained worldwide attention due to their widespread presence in the environment and adverse health effects, but the exposure assessment in the elderly is still lacking. This study aimed to assess exposures to 3 emerging PFASs (chlorinated polyfluoroalkyl ether sulfonic acids, Cl-PFESAs) and 15 legacy PFASs. The temporal variability of internal exposures and intake amounts of these PFASs were evaluated among a population of 76 healthy elderly adults (age: 60-69) in Jinan, China over 5 consecutive months. Fifteen PFASs were detected in whole blood with the mean total concentration (ΣPFAS) at 20.1 ng/mL (range: 5.0-135.9 ng/mL) dominated by perfluorooctanoic acid (PFOA) (9.0 ng/mL), perfluorooctanesulfonic acid (PFOS) (5.3 ng/mL), and 6:2 Cl-PFESA (1.6 ng/mL). Across the 5 month assessment period, significant variation was only observed for short-chain (C4-C7) perfluoroalkyl carboxylic acids, and their variations ranged from 53 to 334%. The median intake of PFOA and PFOS was estimated to be 1.46 and 0.92 ng/kg bw/day, respectively. Regression analysis showed that dietary ingestion, especially fish, was likely an important exposure pathway for PFASs among the elderly adults. Various pathways (e.g., dietary, water, air, and dust) should thus be considered to fully understand human exposure to PFASs.

Human exposure to F-53B in China and the evaluation of its potential toxicity: An overview.

Chlorinated polyfluoroalkyl ether sulfonic acid (Cl-PFESAs, trade name F-53B), an alternative to perfluorooctane sulfonate (PFOS), has been widely used as a mist suppressant in the Chinese electroplating industry since the 1970s. Due to greater restrictions on PFOS globally in recent years, the production and use of F-53B correspondingly increased, consequently causing more emissions into the environment. In China, an increasing number of studies report frequent detection and broad exposure to F-53B in the natural environment, various wildlife and the human body. In human blood, the detection rate of F-53B is almost 80%, accounting for 8.69 to 28% of ∑per- and polyfluoroalkyl substances (PFASs). F-53B is the most biopersistent PFAS in humans to date, with a half-life of 15.3years. In addition, F-53B displays protein binding affinity and high human placental permeability. Recently, some epidemiological studies have reported the health risks associated with F-53B in humans, including abnormal serum lipid metabolism, vascular dysfunction, endocrine disorders and even adverse birth outcomes. Various in vivo and in vitro studies have demonstrated the toxicity of F-53B, such as hepatotoxicity, interference effects on the endocrine system, as well as reproductive and developmental toxicity. Our aims are to review studies on human F-53B exposure levels, trends and associated health effects; evaluate the potential toxicity; and predict directions for future research.

Particle size distribution, wet deposition and scavenging effect of per- and polyfluoroalkyl substances (PFASs) in the atmosphere from a subtropical city of China

Per- and polyfluoroalkyl substances (PFASs) as emerging organic pollutants have received great attention, but the scavenging efficiency of particulate PFASs by wet deposition was rarely studied. For the first time, we reported the scavenging efficiency of PFASs on different particle sizes. In this study, both rainwater and particle samples were collected for a whole year from Xiamen, a subtropical city of China. Particulate PFASs ranged from 4.11 to 67.41 pg m−3, with an average value of 26.56 pg m−3, and perfluorooctane sulfonic acid (PFOS), perfluorooctanoic acid (PFOA) were the main compounds. Perfluorocarboxylic acids (PFCAs) were predominantly observed on fine particles (<1 μm), while PFOS and 6:2 chlorinated polyfluoroalkyl ether sulfonic acid (6:2 Cl-PFESA) had large proportions on coarse particles (1–2.5 μm and 2.5–10 μm). In the rainwater, PFASs ranged from 0.20 to 180.65 ng L−1, with an average value of 10.71 ng L−1, and perfluorobutanoic acid (PFBA), PFOA were the main compounds. The wet deposition flux of ∑PFASs was 5200 mg km−2 yr−1, exhibiting high fluxes during the wet season (March to September). The scavenging efficiency of particulate PFOS and PFOA ranged from 68%–98% during the rainfall, and wash-out of the raindrops was found to be one of the main scavenging mechanisms. In addition, the precipitation duration and intensity influenced the scavenging efficiency. The scavenging capacity of PFCAs was large on fine particles, while for per-and polyfluoroalkyl sulfonic acids, the scavenging capacity was high on coarse particles. Our results showed that wet deposition effectively removed medium to long carbon chain (≥C6) PFASs in the atmosphere.

Chlorinated polyfluoroalkyl ether sulfonic acids in fish, dust, drinking water and human serum: From external exposure to internal doses.

Exposure to per- and polyfluoroalkyl substances (PFASs) has raised significant public health concerns due to their persistence and toxicity in the human body. Here, we aimed to investigate the characteristics of exposure to chlorinated polyfluoroalkyl ether sulfonic acids (Cl-PFESAs) and legacy PFASs and the relative contribution from different external pathways to human exposure. Three Cl-PFESAs and fourteen legacy PFASs were detected in human serum, fish, indoor dust and drinking water collected from Shijiazhuang, China. Results showed that 6:2 Cl-PFESA was the third most predominant compound, with an average concentration of 2.70ng/mL in serum, which was lower than those of perfluorooctane sulfonate (PFOS) and perfluorooctane acid (PFOA) (14.79 and 4.91ng/mL). The estimated daily intake of 6:2 Cl-PFESA via dust ingestion (mean: 0.008ng/kg bw/day) was found to be highest among all detected PFASs, while the highest value via fish and drinking water was found for PFOS and PFOA (0.438-9.799 and 0.034-0.155ng/kg bw/day), respectively. The similar composition of PFASs between serum and fish suggests that fish consumption is a major contributor to human exposure to PFASs. However, the increasing ratios of EDIindoor dust/EDIfish for PFOS, PFOA and 6:2 Cl-PFESA indicated that the contribution of dust ingestion for PFASs, especially 6:2 Cl-PFESA, could not be ignored. The modeled serum concentrations of 6:2 Cl-PFESA higher than its observed levels hint at its gradually increasing exposure levels in the general population. Combined with the lower modeled levels of PFOS than the observed levels, the substituent with 6:2 Cl-PFESA for PFOS is obvious in China. Therefore, further studies on contributions from more detailed external sources and risk assessments of Cl-PFESAs are recommended, especially for some vulnerable subpopulations, considering their widespread exposure and similar environmental behaviors compared with those of their predecessors.

Occurrence and Ecological Risk Assessment of Perfluoroalkyl Substances (PFASs) in Water and Sediment from an Urban River in South China.

The chemical substances in urban rivers influence municipal water systems and reflect the recent use of these chemicals by humans or industries around the urban center. In this study, seven perfluoroalkyl substances (PFASs)-perfluorohexanoic acid (PFHxA), perfluoroheptanoic acid (PFHpA), perfluorooctanoic acid (PFOA), perfluorononanoic acid (PFNA), 2-perfluorohexyl ethanol (6:2 FTOH), 2-perfluorooctyl ethanol (8:2 FTOH), and 6:2 chlorinated polyfluoroalkyl ether sulfonic acids (F-53B)-could be detected and quantified in river water and sediment samples collected from one tributary of the Liuxi River, which is part of Pearl River near Guangzhou in Guangdong province, South China. The fluxes of target PFASs into Liuxi River and their related ecological risks were further estimated. The total concentrations of PFASs (ΣPFASs) ranged from 506 to 3.16 × 103ng/L in water samples and 9.13 to 850ng/L in sediment samples. The two dominant PFAS compounds were 6:2 FTOH and PFHpA, which accounted for more than 90.0% of ΣPFASs in river water and sediment. Correlation analysis showed that there was significant positive correlation (p < 0.01) between two selected PFASs (e.g., between 6:2 FTOH and PFHpA). Correlation analysis of PFASs in river water and sediment indicated most PFASs in sediment were partitioned from river water. The ecological risk assessment indicated that the detected PFASs have a low risk (HQ < 0.1) in river water and sediment to Daphnia magna in the Liuxi River.

Exposure to novel and legacy per- and polyfluoroalkyl substances (PFASs) and associations with type 2 diabetes: A case-control study in East China.

Associations between per- and polyfluoroalkyl substances (PFASs) and the incidence of type 2 diabetes are controversial in epidemiological studies. In addition, limited data are available for assessing the health effects of novel PFAS alternatives. Our study evaluated the effects of PFAS exposure on type 2 diabetes by estimating the associations of PFASs in human serum with the risk of type 2 diabetes and levels of glycemic biomarkers and lipid fractions. The case-control study consisted of 304 participants from Shandong Province, East China, half of which were diagnosed with type 2 diabetes. Logistic regression showed that most PFASs were inversely associated with the risk of type 2 diabetes after adjusting for age, sex, and body mass index. However, concentrations of perfluorooctanoic acid (PFOA) in the control group were positively associated with fasting plasma glucose levels (β=0.04, 95% confidence interval (CI): 0.0003, 0.08), which may promote the development of type 2 diabetes. Furthermore, each log-unit increase in the concentrations of perfluorononanoic acid (PFNA), perfluoroundecanoic acid (PFUnDA), and 6:2 chlorinated polyfluoroalkyl ether sulfonic acid (Cl-PFESA) were associated with a total cholesterol increase (i.e., 17.49% (95% CI: 0.93%, 34.90%), 17.49% (95% CI: 4.71%, 31.83%), and 17.49% (95% CI: 4.71%, 31.83%), respectively). Positive associations were also observed between PFNA, PFUnDA, perfluorooctane sulfonate (PFOS), and 6:2 Cl-PFESA and low-density lipoprotein cholesterol. However, no associations between PFASs and hemoglobin A1c, triglycerides, or high-density lipoprotein cholesterol reached statistical significance, nor associations between PFAS mixtures and outcomes of interest. In conclusion, the significant correlations between serum PFASs and glycemic biomarkers and lipid fractions indicated that PFAS exposure may be a potential diabetogenic factor. To the best of our knowledge, this is the first study to assess the associations between novel Cl-PFESAs and type 2 diabetes, although the inverse associations observed require clarification in future studies.

The occurrence of PFAS in human placenta and their binding abilities to human serum albumin and organic anion transporter 4.

Both legacy and emerging per- and polyfluoroalkyl substances (PFAS) have been found to be threats to human health. In particular, fetuses are sensitive to xenobiotics and the placenta functions as a significant barrier for environmental pollutants. The placental transfer of PFAS is closely related to their interactions with proteins. In this study, 54 human placental samples were collected to investigate the occurrence of legacy and emerging PFAS in human placenta, including perfluorinated carboxylates (PFCAs), perfluorinated sulfonates (PFSAs), chlorinated polyfluoroalkyl ether sulfonic acids (Cl-PFESAs), and fluorotelomer sulfonates (FTSAs). Among the legacy PFAS, perfluorooctanesulfonate (PFOS), perfluorooctanoic acid (PFOA), and perfluorononanoic acid (PFNA) were detected in all samples, with PFOS and PFOA being the two predominant (mean: 0.457 and 0.242ng/g wet weight, respectively). Among the emerging PFAS, 6:2 Cl-PFESA was detected in all samples with the mean value of 0.104ng/g wet weight, while the detect frequency (DF) of 8:2 Cl-PFESAs was only 24%. The concentration and DF of the four FTSA congeners were low in the placentas. Molecular docking calculation results showed that the binding affinities of PFAS to the human serum albumin (HSA) were increased with chain length in each category except for the PFCAs, of which the perfluoroundecanoic acid (PFUnDA) was the turning point of binding affinity to HSA. For PFSAs, their binding affinities to organic anion transporter 4 (OAT4) were increased with the chain length except for the sodium perfluoro-1-heptanesulfonate (PFHpS) and sodium perfluoro-1-nonanesulfonate (PFNS). The calculation results demonstrated that the placental transfer of PFAS is closely related to chain length. The findings in the study can help better understand the occurrence of the PFAS in the human placenta and the placental transfer mechanisms of PFAS in human beings.

Per- and polyfluoroalkyl substances and their alternatives in black-tailed gull (Larus crassirostris) eggs from South Korea islands during 2012–2018

The temporal and spatial trends of sixteen per- and polyfluoroalkyl substances (PFAS) and their three alternatives, chlorinated polyfluoroalkyl ether sulfonic acid (F-53B), hexafluoropropylene oxide dimer acid (GenX), and dodecafluoro-3H-4,8-dioxanonanoate (ADONA) in whole eggs of black-tailed gulls from two South Korean islands, Baengnyeongdo (BLD) and Hongdo (HD), were investigated during 2012–2018. A total of 16 analyzed compounds were detected at concentrations of 21.3–47.8 ng/g ww in BLD and 11.2–40.0 ng/g ww in HD. Meanwhile, the mean levels of perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) were detected at 6.92 ± 4.72 ng/g ww and 0.674 ± 0.993 ng/g ww, respectively. In particular, F-53B, a major alternative to PFOS was detected in each year of the study period with a level of up to 6.66 ng/g ww in all egg samples. Significant increasing temporal trends were observed for PFOS, perfluorononanoic acid (PFNA) and one alternative (F-53B) during the investigated period suggesting continuous use or accumulation of these in the Korean environment. Moreover, distinctive spatial distribution patterns such as a significantly higher F-53B level in HD and an increased PFAS (< C11) in BLD were also observed.

Biomonitoring of chlorinated polyfluoroalkyl ether sulfonic acid in the general population in central and eastern China: Occurrence and associations with age/sex

Although chlorinated polyfluoroalkyl ether sulfonic acid (Cl-PFESA) has been reported to be widespread in different environmental matrices of China, its exposure data in the general Chinese population are very limited. In the present study, the serum-to-whole-blood ratio was first assessed for 6:2 Cl-PFESA (mean/median: 2.07/1.82) based on its paired concentrations (n = 36), which allows a comparison in different blood matrices. The exposure levels of Cl-PFESAs in the general population were investigated by collecting blood samples (n = 1516) from residents of seven cities in central and eastern China. 6:2 Cl-PFESA was observed as the third-highest contributing polyfluoroalkyl substance (PFAS) (8.69%), with the median concentration at 2.18 ng/mL, indicating its importance for assessing the human exposure risks of PFASs. The regional difference between 6:2 Cl-PFESA and perfluorooctane sulfonate (PFOS) can be explained by their use pattern in China. Overall, similar to PFOS, 6:2 Cl-PFESA displays significantly increasing levels with increasing age for both males and females, with significantly higher levels in males. However, a significant sex dependence was found for 6:2 Cl-PFESA in one specific age group (41–60), while there was no significance in the other groups although males display higher levels than females. Our study provides robust data regarding human exposure to 6:2 Cl-PFESA in the general population in central and eastern China.

Legacy and alternative per- and polyfluoroalkyl substances in a subtropical marine food web from the Beibu Gulf, South China: Fate, trophic transfer and health risk assessment

The usage of alternative per- and polyfluoroalkyl substances (PFASs) has been increasing due to the restriction and elimination of legacy PFASs. However, there is limited knowledge on bioaccumulation and trophic magnification of alternative PFASs, especially in subtropical ecosystems. In the present study, we performed a comprehensive survey to investigate the occurrence, bioaccumulation and trophic magnification of legacy and alternative PFASs in subtropical marine food webs in the Beibu Gulf, South China. Results showed that perfluorobutanoic acid (PFBA) and perfluorooctanoic acid (PFOA) were the predominant PFASs in water phase, while perfluorooctane sufonate (PFOS) contributed most to the sum of target PFASs in sediments and marine organisms. Of the investigated PFASs, PFOS and 6:2 chlorinated polyfluoroalkyl ether sulfonic acids (F-53B) exhibited the highest bioaccumulation factor with values > 5000, qualifying as very bioaccumulative chemicals. There was a significant positive correlation between log BSAF and the carbon chain length of perfluoroalkyl carboxylic acids (PFCAs). Trophic magnification (TMF) was observed for PFOS and F-53B, while the remaining PFASs were biodiluted through the present food web. The hazard ratios for PFOS and PFOA in all organisms were far less than unity, suggesting overall low PFAS risks for humans through consumption of marine organisms.

Occurrence and risk of chlorinated polyfluoroalkyl ether sulfonic acids (Cl-PFESAs) in seafood from markets in Beijing, China

In recent years, chlorinated polyfluoroalkyl ether sulfonic acid (Cl-PFESA, trade name: F-53B), one of the alternatives to perfluorooctane sulfonate (PFOS), has been widely detected in environmental matrices and organisms in China. However, sufficient information regarding its presence in seafood is not available. Therefore, we investigated the levels of Cl-PFESAs and legacy per- and polyfluoroalkyl substances (PFASs) in 276 seafood samples collected from markets in Beijing and assessed the human exposure risks of Cl-PFESAs through seafood intake. C8 Cl-PFESA was the dominant congener with significantly higher concentrations (<LOD–1.27 ng/g) than those of C10 Cl-PFESA (<LOD–0.109 ng/g) and C12 Cl-PFESA (<LOD–0.141 ng/g). The mean concentrations of ∑Cl-PFESAs (0.013–0.412 ng/g) contributed to 0.6–11.4% for ∑PFASs in different seafood species. Generally, the concentrations of C8 Cl-PFESA were lower than those of PFOS. However, their concentration trends in four categories of seafood were consistent, with the highest mean concentrations being in shrimps (0.279 and 0.749 ng/g), followed by crabs (0.275 and 0.714 ng/g), fish (0.141 and 0.317 ng/g), and mollusks (0.032 and 0.135 ng/g). A significantly positive correlation was observed between the concentrations of C8 Cl-PFESA and PFOS in all seafood samples (p < 0.001), indicating that they have similar sources and undergo similar environmental processes. The average daily intake of C8 Cl-PFESA (0.067 ng/kg-bw/day) for residents in Beijing via the consumption of seafood was lower than that of PFOS (0.167 ng/kg-bw/day) and far below the most stringent health-based reference dose of PFOS (1.86 ng/kg-bw/day), which implies a low risk for Beijing residents exposed to C8 Cl-PFESA by ingestion of seafood. However, considering the omnipresence of Cl-PFESAs and their stronger bioaccumulation than PFOS, further study on the potential environmental and human exposure risks of Cl-PFESAs is required.
