Magnetic Chitosan for the Removal of Sulfamethoxazole from Tertiary Wastewaters.

Magnetic chitosan nanoparticles, synthesized by in situ precipitation, have been used as adsorbents to remove sulfamethoxazole (SMX), a sulfonamide antibiotic dangerous due to its capacity to enter ecosystems. The adsorption of SMX has been carried out in the presence of tertiary wastewaters from a depuration plant to obtain more realistic results. The effect of pH on the adsorption capacity significantly changed when carrying out the experiments in the presence of wastewater. This change has been explained while taking into account the charge properties of both the antibiotic and the magnetic chitosan. The composition of wastewaters has been characterized and discussed as regards its effect on the adsorption capacity of the magnetic chitosan. The models of Elovich and Freundlich have been selected to describe the adsorption kinetics and the adsorption isotherms, respectively. The analysis of these models has suggested that the adsorption mechanism is based on strong chemical interactions between the SMX and the magnetic chitosan, leading to the formation of an SMX multilayer.

Enrichment of phosphopeptides by arginine-functionalized magnetic chitosan nanoparticles.

One of the most crucial and prevalent post-translational modifications is the phosphorylation of proteins. The study and examination of protein phosphorylation hold immense importance in comprehending disease mechanisms and discovering novel biomarkers. However, the inherent low abundance, low ionization efficiency, and coexistence with non phosphopeptides seriously affect the direct analysis of phosphopeptides by mass spectrometry. In order to tackle these problems, it is necessary to carry out selective enrichment of phosphopeptides prior to conducting mass spectrometry analysis. Herein, magnetic chitosan nanoparticles were developed by incorporating arginine, and were then utilized for phosphopeptide enrichment. A tryptic digest of β-casein was chosen as the standard substance. After enrichment, combined with matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF MS), the detection limit of the method was 0.4 fmol. The synthesized magnetic material demonstrated great potential in the detection of phosphopeptides in complex samples, as proven by its successful application in detecting phosphopeptides in skim milk and human saliva samples.

Recyclable malate dehydrogenase nanocatalyst based on chitosan for oxaloacetic acid green synthesis

Biocatalysis is a new, efficient and green method for oxaloacetic acid (OAA) synthesis. However, malate dehydrogenase (MDH) is unstable, which is the bottleneck for biocatalysis. Immobilization is a good strategy to solve this problem. In this study, chitosan magnetic nanoparticles were used as carriers, and the MDH-nanocatalyst was prepared by a one-pot method combined with electrostatic adsorption and covalent binding. The main factors affecting the performance of the MDH-nanocatalyst were studied in detail, and the optimum preparation process and conditions were obtained. The results show that the hydration particle size of MDH-nanocatalyst is 164 nm in size, with a PDI value of 0.458. It exhibited broad stability against various pH, thermal, organic solvent, metal ion and storage conditions. Finally, the MDH-nanocatalyst was used to synthesize OAA with a yield of 97.6 %. It also showed good circularity, with a remaining catalytic activity of 67.8 % after the 10th cycle. Thus, our findings demonstrate that MDH-nanocatalysts have several attractive features, such as easy preparation, excellent stability and reusability. Therefore, this work provides a foundation for the potential application of these nanocatalysts in the green synthesis of OAA.

Determination of bioactive flavonoids using β-cyclodextrin combined with chitosan-modified magnetic nanoparticles

To accurately determine flavonoids (rutin, quercetin or kaempferol), it is necessary to extract them from complex matrices. The ultrasound-assisted magnetic dispersion microsolid phase extraction technique has been predominantly used for separation and enrichment of the target analytes. The combination of magnetic chitosan nanoparticles and a deep eutectic supramolecular solvent (DESP) is likely to enhance the efficiency of flavonoid extraction from food. In this study, adsorbents were prepared by modifying chitosan with magnetic nanoparticles, and the eluent was a DESP derived from β-cyclodextrin and an organic acid. The successful preparation of these materials was confirmed by FTIR, XRD, FE-SEM and 1H NMR. The extraction recovery rates exceeded 93%, with limits of detection and quantitation ranging from 0.9 to 2.4μg/L and 2.7 to 7.2μg/L, respectively, and the flavonoid clearance rates for ABTS and DPPH radicals reached 100%. Therefore, the integration of magnetic chitosan nanoparticles with the DESP provides a new and efficient method for the extraction of flavonoids while also presenting a potential application of the DESP in separations.

Optimization of lead (II) removal from water and wastewater using a novel magnetic nanocomposite of aminopropyl triethoxysilane coated with carboxymethyl cellulose cross-linked with chitosan nanoparticles

The removal of heavy metals from industrial wastewater is nowadays one of the most interesting challenges in water remediation using adsorbent nanomaterials. The present study presents a novel adsorbent (Fe3O4@CS@CMC-SiNH2) nanocomposite (NC), by functionalization of the carboxymethyl cellulose (CMC) cross-linked to magnetic chitosan nanoparticles (Fe3O4@CS NPs) with aminopropyl triethoxysilane layer (APTES), and evaluate its performance toward lead (II) removal from wastewater. The synthesized NCs were characterized using several techniques. The optimization of the adsorption process was performed using the Response Surface Methodology with the Box-Behnken Design model. The optimal conditions for lead (II) ion removal were found to be; pH of 5.1, a dose of 0.10 g/L, and a temperature of 36 ℃ using Fe3O4@CS@CMC-SiNH2 NCs which proved to have superior adsorption performance with a maximum adsorption capacity of 555.56 mgPb(II)/gNC, and fast achievement of the adsorption equilibrium (30 min). Favorable monolayer chemisorption was estimated and proved to be a coordination bond between the NH amide and lead (II) ions using FTIR, as well as up to seven adsorption–desorption cycles were effective for lead (II) ion removal from aqueous solutions. In addition, the Fe3O4@CS@CMC-SiNH2 NCs showed 98.10%, and 97.21 % removal of lead (II) ions in the absence, and the presence of other contaminants of heavy metals such as cadmium (II) and mercury (II). Moreover, the Fe3O4@CS@CMC-SiNH2 NC exhibited high efficiency in the removal of lead (II) ions from wastewater, can be easily separated from water by magnetic separation, showed satisfactory reusability for five cycles of the adsorption–desorption process of lead (II) ion removal from tap water and wastewater. Consequently, Fe3O4@CS@CMC-SiNH2 NC is a prospective adsorbent nanomaterial for lead (II) ions removal from wastewater.

Activation of Insulin Gene Expression via Transfection of a CRISPR/dCas9a System Using Magnetic Peptide-Imprinted Nanoparticles.

A CRISPRa transcription activation system was used to upregulate insulin expression in HEK293T cells. To increase the delivery of the targeted CRISPR/dCas9a, magnetic chitosan nanoparticles, imprinted with a peptide from the Cas9 protein, were developed, characterized, and then bound to dCas9a that was complexed with a guide RNA (gRNA). The adsorption of dCas9 proteins conjugated with activators (SunTag, VPR, and p300) to the nanoparticles was monitored using both ELISA kits and Cas9 staining. Finally, the nanoparticles were used to deliver dCas9a that was complexed with a synthetic gRNA into HEK293T cells to activate their insulin gene expression. Delivery and gene expression were examined using quantitative real-time polymerase chain reaction (qRT-PCR) and staining of insulin. Finally, the long-term release of insulin and the cellular pathway related to stimulation by glucose were also investigated.

Neuroprotective effect of piracetam-loaded magnetic chitosan nanoparticles against thiacloprid-induced neurotoxicity in albino rats

Thiacloprid (TH) is a neurotoxic agricultural insecticide and potential food contaminant. The purpose of this study was to investigate the relationship between TH exposure and memory dysfunction in rats, as well as the potential protective effect of piracetam and piracetam-loaded magnetic chitosan nanoparticles (PMC NPs). Rats were divided into five equal groups (six rats/group). The control group received saline. Group II was treated with PMC NPs at a dose level of 200 mg/kg body weight (Bwt); Group III was treated with 1/10 LD50 of TH (65 mg/kg Bwt); Group IV was treated with TH (65 mg/kg Bwt) and piracetam (200 mg/kg Bwt); Group V was co-treated with TH (65 mg/kg Bwt) and PMC NPs (200 mg/kg Bwt). All animal groups were dosed daily for 6 weeks by oral gavage. Footprint analysis, hanging wire test, open field test, and Y-maze test were employed to assess behavioral deficits. Animals were euthanized, and brain tissues were analyzed for oxidative stress biomarkers, proinflammatory cytokines, and gene expression levels of glial fibrillary acidic protein (GFAP), amyloid-beta precursor protein (APP), B-cell lymphoma 2 (Bcl-2), and caspase-3. Brain and sciatic nerve tissues were used for the evaluation of histopathological changes and immunohistochemical expression of tau protein and nuclear factor kappa B (NF-κB), respectively. The results revealed that TH-treated rats suffered from oxidative damage and inflammatory effect on the central and peripheral nerves. The administration of PMC NPs considerably protected against TH-induced neuronal damage, increased antioxidant enzyme activity, decreased inflammatory markers, and improved behavioral performance than the group treated with piracetam. The neuroprotective effect of PMC NPs was mediated through the inhibition of GFAP, APP, caspase-3, Tau, and NF-κB gene expression with induction of Bcl-2 expression. In conclusion, TH could induce oxidative stress, inflammatory and neurobehavior impairment in rats. However, PMC NPs administration markedly mitigated TH-induced brain toxicity, possibly via oxidative and inflammatory modulation rather than using piracetam alone.

Removal of tetracycline from water using ethylenediamine-modified magnetic chitosan

In this work, the magnetic chitosan nanoparticles were prepared by in-situ co-precipitation method and then modified by glutaraldehyde and ethylenediamine. The effect dose of crosslinking agents for adsorbent preparation on tetracycline (TC) adsorption was investigated. The experiment results showed that the modified chitosan (EMMCS-G) had the best adsorption performance, which was more than twice that of raw magnetic chitosan. EMMCS-G was characterized by SEM, TEM, VSM, FTIR, XRD and XPS. The effects of experimental parameters such as time, temperature, pH, adsorbent dosage and Cu2+ on TC adsorption by EMMCS-G were discussed. The result indicated that the optimal pH was 6.5. Under the conditions of an initial TC concentration of 50 ​mg/L and an EMMCS-G dosage of 50 ​mg, the removal rate reached an impressive 94%. The isotherm and kinetic studies of adsorption process illustrated that adsorption of TC onto EMMCS corresponded to Freundlich isotherm and pseudo-second-order kinetic models. The maximum adsorption capacity was 84 ​mg/g. Thermodynamic analysis revealed that adsorption process was both endothermic and spontaneous. The regeneration experiment suggested that the adsorbent possessed excellent reusability for further use. Moreover, the adsorption mechanisms of TC onto EMMCS-G mainly involved pore filling, π-π interactions, hydrogen bonding and electrostatic attraction.

Nanoparticle-mediated CRISPR/dCas9a activation of multiple transcription factors to engineer insulin-producing cells.

Insulin may help to control blood glucose levels in diabetes; however, the long-term release of insulin is important for therapy. In this work, four guide RNAs (gRNA) for factors that promote specification and maturation of insulin-producing cells were synthesized: pancreatic and duodenal homeobox 1 (PDX1), protoendocrine factor (neurogenin 3, NGN3), NK6 homeobox 1 (NKX6.1), and musculoaponeurotic fibrosarcoma oncogene family A (MAFA). These gRNAs were used to form ribonucleoproteins (RNPs) with tracRNA and dCas9-VPR, and were then immobilized on magnetic peptide-imprinted chitosan nanoparticles, which enhanced transfection. The production and release of insulin from transfected cells were then measured using ELISA and staining with anti-insulin antibodies. The expression of the genes was evaluated using qRT-PCR; this was also used to investigate the cascade of additional transcriptional regulators. The magnitude and duration of insulin production were evaluated for single and repeated transfections (using different transfection schedules) to identify the most promising protocol.

Development of magnetic dispersive micro-solid phase extraction of four phenolic compounds from food samples based on magnetic chitosan nanoparticles and a deep eutectic supramolecular solvent

A magnetic dispersive micro-solid phase extraction technique (CS@Fe3O4-MD-μSPE-DESP) based on magnetic chitosan nanoparticles and a deep eutectic supramolecular solvent was developed and applied to determinations of four phenolic compounds in food samples. To prevent environmental pollution and the introduction of toxic substances, deep eutectic supramolecular solvents (DESPs), which exhibited greater desorption capacities than conventional organic solvents and deep eutectic solvents, were used as novel green eluents for the first time. Some important parameters were screened by the Plackett–Burman method and then further optimized with response surface methodology (RSM). Under the optimal conditions, the proposed method showed excellent methodological indices with linearity over the range 0.1–200.0 µg·mL-1, R2 > 0.9988, extraction recoveries above 94.8 %, and precision (RSD%) below 2.9 %. The established method finishes the process of adsorption and desorption in approximately 3 min and enhances the efficiency for determination of phenolic compounds.

Rapid determination of multiclass antibiotics and their metabolites in milk using ionic liquid-modified magnetic chitosan nanoparticles followed by UPLC-MS/MS

This research introduced a rapid and efficient method for simultaneous pre-concentration and determination of twenty-two antibiotics and their metabolites in milk samples using functional imidazole-based ionic liquid-modified magnetic chitosan nanoparticles followed by ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Different techniques characterized the synthesized nanoparticles. Experimental variables were studied and optimized by the one-factor-at-a-time approach and Box-Behnken design. All antibiotics showed good linearity, with correlation coefficients of more than 0.995. The matrix effects were greatly minimized using the proposed method. The range of average recoveries was from 85.9 to 107.5%, and precision values were presented as RSDs, which were below 9.2%. The limits of detection were 0.04–0.19 μg kg−1. The measurement uncertainty was estimated by the bottom-up approach. The conformity method was applied in the analysis of real milk samples. Antibiotic residues were found in 124 samples out of 241 milk samples. Nevertheless, it was found that 24 of 133 raw cow milk samples and 15 of 108 pasteurized milk samples were above maximum residue limits. Finally, the potential human health risk estimated using the hazard quotient approach, based on the estimated daily intake and acceptable daily intake, showed a negligible risk for human health from milk consumption.

Magnetic chitosan nanoparticles loaded with Amphotericin B: Synthesis, properties and potentiation of antifungal activity against common human pathogenic fungal strains

Amphotericin B has long been regarded as the gold standard for treating invasive fungal infections despite its toxic potential. The main objective of this research was to develop a novel IONPs@CS-AmB formulation in a cost-effective manner in order to enhance AmB delivery performance, with lowering the drug's dose and adverse effects. The chitosan-coated iron oxide nanoparticles (IONPs@CS) were synthesized afterward, AmB-loaded IONPs@CS (IONPs@CS-AmB) prepared and characterized by AFM, FT-IR, SEM, EDX, and XRD. Biological activity of the synthesized NPs determined and the cytotoxicity of IONPs@CS-AmB evaluated using the MTT and in vitro hemolysis tests. The IONPs@CS-AmB was synthesized using the coprecipitation method with core-shell structure in size range of 27.70 to ∼70 nm. The FT-IR, XRD and EDX pattern confirmed the successful synthesis of IONPs @CS-AmB. The IONPs@CS-AmB exhibited significant antifungal activity and inhibited the metabolic activity of Candida albicans biofilms. The hemolysis and MTT assays showed that IONPs@CS-AmB is biocompatible with high cell viability when compared to plain AmB and fungizone. The IONPs@CS-AmB is more effective, less toxic and may be a suitable alternative to conventional drug delivery. IONPs@CS-AmB may be a viable candidate for use as a microbial-resistant coating on the surfaces of biomedical devices.

Synthesis of Chitosan Magnetic Nanoparticles using Glutaraldehyde and Tripolyphosphate as Crosslinker

Chitosan is an abundant natural polymer, which able to adsorb Pb2+. However, chitosan has the drawback of being unstable in acid and alkaline conditions, so it needs cross-linking. In this article, we will compare the usage of two crosslinkers, glutaraldehyde (GD) and tripolyphosphate (TPP), to solve these issues. The synthesis of chitosan magnetic nanoparticles was carried out by co-precipitation with the addition of glutaraldehyde under acidic conditions and the addition of TPP under alkaline conditions. The results showed that the FTIR spectrum gave characteristic bands at 3131.96 cm-1 (OH and NH), 1635.86 cm-1 (C=O), 1401.97 cm-1 (CN), ~1110 cm-1 (COC), and ~617 cm-1 (Fe-O). SEM analysis showed that the surface morphology of CMNP-GA particle aggregation was clear more than that of CMNP-TPP. The size of the synthesized nanoparticles was determined by PSA analysis, which was 131.95 µm and 137.10 µm, respectively. The results of the XRD analysis showed that, in addition to the Fe3O4 and chitosan phases, the produced CMNP also contained γFe2O3 phase, which was considered to be caused by Fe3O4 oxidation

Synergic effect between TRAIL gene and curcumin in magnetic chitosan nanoparticles on cancer cells apoptosis enhanced by laser photoactivation

The Apoptosis is an intensely studied mechanism for cancer therapy. Various genes, drugs, and molecules have been combined to potentiate the apoptotic effect against tumor cells. Magnetic nanocomplexes with biopolymers have been proposed as nanocarriers for delivery of active molecules and improve apoptosis induction. To these purpose were formulated magnetic nanoparticles with curcumin coated chitosan (MNPs-CHI-TPP-CUR) and magnetic nanoparticles with pCEM-TRAIL plasmid coated chitosan (MNPs-CHI-TPP-TRAIL) for use alone or in combination to induce the apoptosis of B16F10 tumor cells because curcumin is capable of overexpressing receptors TRAIL required for apoptosis induction. Also, curcumin (CUR) has been used as a photosensitizer in photodynamic therapy (PDT). In this work, it was produced and characterized nanoparticles with curcumin and nanoparticles with TRAIL gene. Both types of nanoparticles were ~ 200 nm size. The results showed that the combination of (MNPs-CHI-TPP-CUR) and (MNPs-CHI-TPP-TRAIL) induced a significant increase in cellular death within 48 h compared with the effect of each nanocomplex individually, demonstrating a synergic effect. This is the first description the synergic interaction between TRAIL gene and curcumin drug to improve the apoptosis in cancer cells. Also, it was determined that cellular death was potentiated when the treated cells were exposed to laser photoactivation with wavelengths of 405 nm and 532 nm for 15 min.Graphical abstract

Grafting of Thiazole Derivative on Chitosan Magnetite Nanoparticles for Cadmium Removal-Application for Groundwater Treatment.

The synthesis and developments of magnetic chitosan nanoparticles for high efficiency removal of the cadmium ions from aquatic medium are one of the most challenging techniques. Highly adsorptive composite (MCH-ATA) was produced by the reaction of chitosan with formaldehyde and amino thiazole derivative. The sorbent was characterized by FTIR, elemental analyses (EA), SEM-EDX, TEM analysis, TGA and titration (volumetric). The modified material includes high nitrogen and sulfur contents (i.e., 4.64 and 1.35 mmol g−1, respectively), compared to the pristine material (3.5 and 0 mmol g−1, respectively). The sorption was investigated for the removal of Cd(II) ions from synthetic (prepared) solution before being tested towards naturally contaminated groundwater in an industrial area. The functionalized sorbent shows a high loading capacity (1.78 mmol Cd g−1; 200 mg Cd g−1) compared to the pristine material (0.61 mmol Cd g−1; 68.57 mg Cd g−1), while removal of about 98% of Cd with capacity (6.4 mg Cd g−1) from polymetallic contaminated groundwater. The sorbent displays fast sorption kinetics compared to the non-modified composite (MCH); 30 min is sufficient for complete sorption for MCH-ATA, while 60–90 min for the MCH. PFORE fits sorption kinetics for both sorbents, whereas the Langmuir equation fits for MCH and Langmuir and Sips for MCH-ATA for sorption isotherms. The TEM analysis confirms the nano scale size, which limits the diffusion to intraparticle sorption properties. The 0.2 M HCl solution is a successful desorbing agent for the metal ions. The sorbent was applied for the removal of cadmium ions from the contaminated underground water and appears to be a promising process for metal decontamination and water treatment.

The effect of surfactant on the hydrolysis of coconut husk using cellulase and xylanase enzyme immobilized on chitosan magnetic nanoparticles

This study aimed to investigate the effect of surfactant Tween 80 in the enzymatic hydrolysis of coconut husks by using immobilized cellulase from Aspergillus niger, Trichoderma reesei, and immobilized xylanase. Surfactant is common to enhance the yield of sugar in enzymatic process. However, its application in immobilized enzymes has not been well investigated. Coconut husk is an abundant resource of cellulose that can be converted to reducing sugar which can be utilized to produce various products such as biofuel, lactic acid, 5-hydroxymethyl furfural, and leluvinic acid. Chitosan magnetic nanoparticles as a support in the immobilization process were prepared using the coprecipitation method to obtain particles separated by magnetic field. Cellulase and xylanase were immobilized on magnetic particles through covalent bond directly and with Glutaral dialdehyde (GDA) as cross-linker. The immobilized cellulase and xylanase along with Tween 80 surfactants were applied for the hydrolysis of NaOH pre-treated coconut husk at 55°C for 48 hours. The reducing sugar content were identified with DNS methods. In this study, the highest reducing sugar content obtained from cellulase from A niger, cellulase from T reesei and xylanase were 0.412 mg/ml, 0.532 mg/mL and 1.166 mg/mL, respectively. The concentration of Tween 80 that yielding highest sugar concentration for immobilized cellulase from A niger, cellulase from T reesei and xylanase were 2%, 1% and 2% (w/v), respectively.

Using chitosan-based heterogeneous catalyst for degradation of Acid Blue 25 in the effective electro-Fenton process with rotating cathodes

Herein, the capability of glutaraldehyde cross-linked magnetic chitosan nanoparticles (Fe3O4/CS/GA NPs) was investigated as a heterogeneous electro-Fenton catalyst for the degradation of Acid Blue 25. A cylindrical reactor with rotating cathodes was used for performing the electro-Fenton process. Fe3O4/CS/GA NPs were characterized by Fourier-Transform Infrared Spectroscopy (FT-IR), X-ray Powder Diffraction (XRD), Field Emission Scanning Electron Microscope (FESEM), Energy Dispersive X-Ray (EDX), and Vibrating Sample Magnetometer (VSM). According to the results, the dye removal efficiency reached 94.83% after 90 min under the optimal conditions of Fe3O4/CS/GA NPs with the iron content of 15 mmol Fe/g chitosan and concentration of 0.3 g/L, natural pH of dye (6.8), initial dye concentration of 150 mg/L, the current intensity of 0.7 A, and rotational speed of 100 rpm. Also, Chemical Oxygen Demand (COD) and Total Organic Carbon (TOC) removal reached 74.19% and 61.53% after 120 min, respectively. Average oxidation state (AOS) and carbon oxidation state (COS) values were increased after treatment, indicating that wastewater's biodegradability was improved. Moreover, the heterogeneous electro-Fenton process using optimal Fe3O4/CS/GA NPs resulted in about 16% improvement in the removal efficiency compared to the homogeneous process. The catalyst showed good reusability up to 5 cycles. The amounts of leached iron into the solution were much less than allowable amounts of the Iranian wastewater discharge standard, thus no need for secondary treatment. Furthermore, scavengers studies under the optimum conditions showed the dye degradation occurred mainly by surface-bonded hydroxyl radicals. In short, this study proved, Fe3O4/CS/GA NPs are a promising heterogeneous electro-Fenton catalyst due to their low cost, excellent stability and reusability, negligible iron leaching, environmental compatibility, and superior catalytic activity in a wide range of pH.
