Chemotherapy of Experimental Schistosomiasis. I. Drug Activity and Mode of Action of a New Thioxanthone Derivative

In addition to the antimonials, only a few thioxanthone derivatives have been employed in the chemotherapy of schistosomiasis. Nevertheless, the results obtained with the drugs now available are relatively poor and there exists a great need for new schistosomicidal agents. Raison and Standen (1954, 1955) have recently reported marked schistosomicidal activity of symmetrical diaminodiphenoxyalkanes. Caldwell and Standen (1956) and Hill (1956) demonstrated that p-aminophenoxyalkane derivatives and some compounds of the homologous series of diphenoxyalkanes are also active against S. mansoni. Ring substituents of diphenoxyalkanes are less effective (Standen and Walls, 1956). Some rhodanines, hydantoins and N-(9-Xan thenyl) amides have been demonstrated by Luttermoser and Bond (1954) and Bond and Luttermoser (1954) to have therapeutic activity. The results obtained in mice experimentally infected with S. inansoni and treated with a new thioxanthone derivative are reported in this paper, along with some observations concerning the mode of drug action.

Effect of Feeding Antimony Compounds to Mice Experimentally Infected with Schistosoma Mansoni 1,2,3,4

In past work from this laboratory on the effects of various chemical compounds on experimental Schistosoma mansoni infections in mice many compounds were fed but only two were found to be of some promise when administered by this route. These were butyl antimonyl gallate and phenyl mercuric gluconate. Since the treatment of human schistosomiasis by a drug that could be administered orally would be of great advantage, further work has been carried on to explore the effects of other antimonials when fed to infected mice. Several other antimonials have been found which completely clear infected mice of schistosoma worms when these compounds, mixed with their food, are fed to the mice. Since it has been found necessary to discontinue work in this laboratory on experimental chemotherapy of schistosomiasis a few other results with oil soluble antimonials are also included in the present report as well as some work on the treatment of experimentally infected golden hamsters.

Effect of Drugs during Earliest Stages of Experimental Schistosomiasis Mansoni in Mice 1

In connection with work previously reported from this Laboratory on the chemotherapy of schistosomiasis in experimentally infected mice, the suggestion was made by Dr. H. E. Meleney of this University that it might be well to do some screening work testing the effect of drugs administered to mice during a short period of time immediately following the exposure of these mice to infection. The aim of such experiments would be to find drugs to prevent the development of the disease by stopping the development of the larval worms to adults. Since the war was still in progress at the time these experiments were begun the possibility of immediate application of any findings to actual field conditions was sufficient warrant for such an investigation. It takes about a week after the penetration of the cercariae into the body of their definitive host before they begin to collect in the liver in any considerable numbers.

Conditions for Drug Testing in Experimental Schistosomiasis Mansoni in Mice 1

Some studies of limited scope have been published on the experimental chemotherapy of schistosomiasis. Faust and Meleney (1) in 1924 reported the cure with tartar emetic of two dogs infected with Schistosoma japonicum. Fairley (2) experimentally infected goats with S. spindalis and showed that they could be cured by repeated injections of tartar emetic. Kawamura (3) in 1929 and Ozawa (4) in 1930 described good results using stibonal on a variety of animals experimentally infected with S. japonicum. It is impossible to judge the value of this work as no details are given in the English references available. Eichholtz (5) in 1930 and Szidat and Wigand (6) in 1931 reported attempts to cure ducks of natural infections of Bilharziella polonica by injections of fuadin and of emetine. They concluded that these drugs had no therapeutic effect and that, therefore, such ducks were of no value as test animals for human schistosomes.
