17O Solid-State NMR Spectroscopy of Lipid Membranes.

Despite the limitations posed by poor sensitivity, studies have reported the unique advantages of 17O based NMR spectroscopy to study systems existing in liquid, solid, or semisolid states. 17O NMR studies have exploited the remarkable sensitivity of quadrupole coupling and chemical shift anisotropy tensors to the local environment in the characterization of a variety of intra- and intermolecular interactions and motion. Recent studies have considerably expanded the use of 17O NMR to study dynamic intermolecular interactions associated with some of the challenging biological systems under magic angle spinning (MAS) and aligned conditions. The very fast relaxing nature of 17O has been well utilized in cellular and in vivo MRS (magnetic resonance spectroscopy) and MRI (magnetic resonance imaging) applications. The main focus of this Review is to highlight the new developments in the biological solids with a detailed discussion for a few selected examples including membrane proteins and nanodiscs. In addition to the unique benefits and limitations, the remaining challenges to overcome, and the impacts of higher magnetic fields and sensitivity enhancement techniques are discussed.

Drop That Activation Energy: Tetragonal to Cubic Transformations in Na3PS4−xSex for Solid State Sodium Ion Battery Materials

AbstractSodium‐containing chalcogenide materials are emerging as a class of solid electrolytes for application in inexpensive all‐solid‐state sodium‐ion batteries due to their high ionic conductivity, abundance, and degree of synthetic and structural variability. Members of the solid solution Na3PS4−xSex, which are promising solid electrolytes for sodium‐ion batteries, are prepared by reaction at high temperature. With increasing substitution of S by Se, the structure transforms from tetragonal (space group P21c for x = 0, 1) to cubic (space group I3m for x = 2, 3, 4). Within the solid solution, the S and Se atoms are completely disordered in the environments around the P atoms, in accordance with a binomial distribution, as inferred by the 31P nuclear magnetic resonance (NMR) spectra. In 23Na NMR experiments conducted at different magnetic fields and temperatures, quadrupolar lineshapes are observed that are influenced by sodium ion dynamics; the activation energies decrease from 0.21 to 0.15 eV on progressing from the S‐ to the Se‐rich members. A dynamic model is proposed to account for the changes in the 23Na quadrupolar lineshapes by switching the orientations of the electric field gradient and chemical shift anisotropy tensors when Na+ ions hop to the four nearest Na sites.

Atomic-Scale Resolution Insights into Structural and Dynamic Differences between Ofloxacin and Levofloxacin.

This study employs advanced solid-state NMR techniques to investigate the atomic-level structure and dynamics of two enantiomers: ofloxacin and levofloxacin. The investigation focuses on critical attributes, such as the principal components of the chemical shift anisotropy (CSA) tensor, the spatial proximity of 1H and 13C nuclei, and site-specific 13C spin-lattice relaxation time, to reveal the local electronic environment surrounding specific nuclei. Levofloxacin, the levo-isomer of ofloxacin, exhibits higher antibiotic efficacy than its counterpart, and the dissimilarities in the CSA parameters indicate significant differences in the local electronic configuration and nuclear spin dynamics between the two enantiomers. Additionally, the study employs the 1H-13C frequency-switched Lee-Goldburg heteronuclear correlation (FSLGHETCOR) experiment to identify the presence of heteronuclear correlations between specific nuclei (C15 and H7 nuclei and C13 and H12 nuclei) in ofloxacin but not in levofloxacin. These observations offer insights into the link between bioavailability and nuclear spin dynamics, underscoring the significance of NMR crystallography approaches in advanced drug design.

Determination of the relative orientation between 15N-1H dipolar coupling and 1H chemical shift anisotropy tensors under fast MAS solid-state NMR

In this work, we have proposed a proton-detected three-dimensional (3D) 15N-1H dipolar coupling (DIP)/1H chemical shift anisotropy (CSA)/1H chemical shift (CS) correlation experiment to measure the relative orientation between the 15N-1H dipolar coupling and the 1H CSA tensors under fast magic angle spinning (MAS) solid-state NMR. In the 3D correlation experiment, the 15N-1H dipolar coupling and 1H CSA tensors are recoupled using our recently developed windowless C-symmetry-based C331-ROCSA (recoupling of chemical shift anisotropy) DIPSHIFT and C331-ROCSA pulse-based methods, respectively. The 2D 15N-1H DIP/1H CSA powder lineshapes extracted using the proposed 3D correlation method are shown to be sensitive to the sign and asymmetry of the 1H CSA tensor, a feature that allows the determination of the relative orientation between the two correlating tensors with improved accuracy. The experimental method developed in this study is demonstrated on a powdered U-15N L-Histidine.HCl·H2O sample.

Quantifying quadrupole effects in the NMR spectra of spin-1/2 nuclei in rotating solids.

Quantifying the NMR spectra of spin I = 1/2 nuclei coupled to quadrupolar spins (nuclei with spin quantum number, S > 1/2) has remained an arduous task in solid-state magic angle spinning (MAS) NMR experiments. In particular, extraction of chemical shift anisotropy (CSA) tensors from line-shapes of spin I = 1/2 nuclei coupled to quadrupolar spin (S = 1) in MAS experiments has remained challenging owing to the simultaneous presence of both heteronuclear dipolar interactions and quadrupolar interactions. Unlike experiments that involve only spin-1/2 nuclei, both faster spinning frequencies and stronger decoupling field strengths on the quadrupolar spins are essential to average/minimize the contributions from heteronuclear dipolar interactions. To this end, a quantitative theory based on the concept of "effective fields" is proposed to deduce optimal conditions in experiments that involve simultaneous recoupling and decoupling of heteronuclear dipolar interactions. Through analytic expressions, the spectral frequencies and intensities observed in experiments are quantified and rigorously verified. Since extraction of molecular constraints in NMR experiments involves iterative fitting of experimental data, we believe that the analytic expressions derived would speed up and be beneficial in quantifying such experiments.

NMR Crystallographic Approach to Study the Variation of the Dynamics of Quinine and Its Quasienantiomer Quinidine

The structure and dynamics of quinine and its quasienantiomer quinidine were studied at the atomic resolution by measuring the chemical shift anisotropy (CSA) tensor and site-specific spin–lattice relaxation time. For quinine, there are three crystallographically independent molecules “a”, “b”, and “c” in an asymmetric unit since its 13C CP-MAS SSNMR spectrum features three distinct resonance peaks for certain carbon nuclei. The 13C assignments are fulfilled by DFT calculations. The experimental 13C isotropic chemical shifts well match the calculated values. These variations of isotropic chemical shift for three independent molecules are also observed by two-dimensional 13C–1H heteronuclear correlation spectroscopy (HETCOR) of quinine. The spin–lattice relaxation time, and the principal components of CSA parameters are also varied substantially for certain carbon nuclei of “a”, “b”, and “c” molecules. For quinidine, its 13C CP-MAS SSNMR spectrum is remarkably different from that of quinine despite, their almost identical solution NMR spectra. Furthermore, the remarkable change in the structure and dynamics of quasienantiomers are also observed including the steric effect of the substituent vinyl group, the variation of helical motifs, and the variation of the strength of the intermolecular hydrogen bonds. The variation of the structure and dynamics of quasienantiomers are thoroughly studied by solid-state NMR measurements. These types of studies will enrich the field of NMR crystallography.

Molecular Structures of the Silicon Pyridine-2-(thi)olates Me3Si(pyX), Me2Si(pyX)2 and Ph2Si(pyX)2 (py = 2-Pyridyl, X = O, S), and Their Intra- and Intermolecular Ligand Exchange in Solution

A series of pyridine-2-olates (pyO) and pyridine-2-thiolates (pyS) of silicon was studied in solid state and in solution. The crystal structures of Me3Si(pyO) (1a), Me3Si(pyS) (1b), Me2Si(pyO)2 (2a), Me2Si(pyS)2 (2b), Ph2Si(pyO)2 (3a) and Ph2Si(pyS)2 (3b) were determined by X-ray diffraction. For that purpose, crystals of the (at room temperature) liquid compounds 1a and 1b were grown in a capillary on the diffractometer. Compounds 1a, 1b, 2a, 2b and 3a feature tetracoordinate silicon atoms in the solid state, whereas 3b gave rise to a series of four crystal structures in which the Si atoms of this compound are hexacoordinate. Two isomers (3b1 with all-cis arrangement of the C2N2S2 donor atoms in P1¯, and 3b2 with trans S-Si-S axis in P21/n) formed individual crystal batches, which allowed for their individual 29Si NMR spectroscopic study in the solid state (the determination of their chemical shift anisotropy tensors). Furthermore, the structures of a less stable modification of 3b2 (in C2/c) as well as a toluene solvate 3b2 (toluene) (in P1¯) were determined. In CDCl3, the equimolar solutions of the corresponding pairs of pyO and pyS compounds (2a/2b and 3a/3b) showed substituent scrambling with the formation of the products Me2Si(pyO)(pyS) (2c) and Ph2Si(pyO)(pyS) (3c), respectively, as minor components in the respective substituent exchange equilibrium.

Exploring Molecular Dynamics of Adsorbed CO2 Species in Amine-Modified Porous Silica by Solid-State NMR Relaxation.

Previous studies on CO2 adsorbents have mainly addressed the identification and quantification of adsorbed CO2 species in amine-modified porous materials. Investigation of molecular motion of CO2 species in confinement has not been explored in depth yet. This work entails a comprehensive study of molecular dynamics of the different CO2 species chemi- and physisorbed at amine-modified silica materials through the determination of the rotating frame spin–lattice relaxation times (T1ρ) by solid-state NMR. Rotational correlation times (τC) were also estimated using spin relaxation models based on the Bloch, Wangsness, and Redfield and the Bloembergen–Purcell–Pound theories. As expected, the τC values for the two physisorbed CO2 species are considerably shorter (32 and 20 μs) than for the three identified chemisorbed CO2 species (162, 62, and 123 μs). The differences in molecular dynamics between the different chemisorbed species correlate well with the structures previously proposed. In the case of the physisorbed CO2 species, the τC values of the CO2 species displaying faster molecular dynamics falls in the range of viscous liquids, whereas the species presenting slower dynamics exhibit T1ρ and τC values compatible with a CO2 layer of weakly interacting molecules with the silica surface. The values for chemical shift anisotropy (CSA) and 1H–13C heteronuclear dipolar couplings have also been estimated from T1ρ measurements, for each adsorbed CO2 species. The CSA tensor parameters obtained from fitting the relaxation data agree with the experimentally measured CSA values, thus showing that the theories are well suited to study CO2 dynamics in silica surfaces.

Valinate and SiMe2 – An interesting couple in pentacoordinate Si-complexes: Templated generation of the dipeptide val-val and formation of an organosilicon-ammonia-adduct

Reaction of dichlorodimethylsilane and L-valin in molar ratio 1:2 (with addition of a supporting base, Et3N) afforded a complex mixture of products, from which the pentacoordinate silicon complex (val-val)SiMe2 (1) could be isolated and identified by single-crystal X-ray diffraction analysis. This complex features the di-anion of dipeptide valinylvalin as a tridentate (N,N‘,O)-ligand. Using a stochiometric ratio of starting materials adjusted to the formation of 1 (the use of dichlorodimethylsilane and L-valin in molar ratio 1:1, the additional silane serving as a water trap to foster peptide condensation) allowed for the preparation of this compound in moderate yield. Using 1,1,3,3,5,5-hexamethylcyclotrisilazane, which may serve both as a source of SiMe2 moieties and deprotonator for valin, afforded another pentacoordinate Si-complex, i.e., (val)SiMe2(NH3) (2). In addition to the chelating di-anion of valin its Si atom carries an ammonia molecule as an axial ligand within a trigonal-bipyramidal coordination sphere, as confirmed by single-crystal XRD. For both solids 1 and 229Si CP/MAS NMR spectra were recorded and the chemical shift anisotropy tensors were determined therefrom.

A Description of the Local Structure and Dynamics of Ketoconazole Molecule by Solid‐State NMR Measurements and DFT Calculations: Proposition for NMR Crystallography

AbstractAn azole class antifungal agent, ketoconazole, is widely used in the treatment of mucosal fungal infections that arise due to AIDS immunosuppression, organ transplantation, and cancer chemotherapy. The structure and dynamics of various molecular moieties of ketoconazole are thoroughly studied by measuring chemical shift anisotropy tensor and site‐specific spin‐lattice relaxation time. The local correlation time at crystallographically different carbon nuclei sites is also calculated. The chemical shift anisotropy (CSA) parameters at the crystallographically distinct sites of ketoconazole are determined by the two‐dimensional phase‐adjusted spinning sideband (2D PASS) cross‐polarization magic angle spinning (CP‐MAS) solid‐state NMR experiment. The site‐specific spin‐lattice relaxation time is measured by the method outlined by Torchia (T1CP). The values of the principal components of CSA parameters extracted by the 2DPASS CP‐MAS ssNMR experiment are supported by density functional theory (DFT) calculations. The CSA parameters are high for those carbon nuclei, whose spin‐lattice relaxation rate is slow, and it is low for those carbon nuclei whose spin‐lattice relaxation rate is fast. It suggests that the spin‐lattice relaxation mechanism is mainly governed by chemical‐shift anisotropy interaction for these carbon nuclei. A huge variation of the spin‐lattice relaxation time and the local correlation time are observed for numerous carbon nuclei that reside on the side‐chain of the molecule. The spin‐lattice relaxation time varies from 500 s to 8 s, and the order of the local correlation time varies from s to s. These types of studies in which the chemical shift anisotropy (CSA) parameters and spin‐lattice relaxation time provide information about the electronic configuration and the spin dynamics at the various crystallographic locations of the drug molecule will enrich the field “NMR crystallography.” It will also help to develop the strategies for the administration of antifungal drugs by providing information about the structure and dynamics of various parts of the drug molecule.

Effective Local Geometry Descriptor for 29Si NMR Q4 Anisotropy

The nuclear shielding anisotropy, ζ, is a useful nuclear magnetic resonance (NMR) shielding tensor parameter in describing the extent of electron cloud distortion about an atom. Despite the advantages afforded by NMR in structural characterization, the relationship between ζ and local structure of an atom in high-symmetry environments, such as Si–Q4 sites, is poorly understood. Here, we use a data-driven approach combining random forest feature ranking and the Sure Independence Screening and Sparsifying Operator (SISSO) approach to derive a simple and accurate geometric descriptor for ζ with a root-mean-squared prediction error of 6.77 ppm and an R2 of 0.761. We then apply this descriptor to describe the local geometric distortion of zeolites Sigma-2 and silica-ZSM-5 whose chemical shift anisotropy tensor has been reported. We envision that this geometric descriptor will allow for structural description and refinement in previously difficult-to-describe materials.

Study of the structure and dynamics at various parts of the antibacterial drug molecule cefpodoxime proxetil

The structure and dynamics of cefpodoxime proxetil are elucidated by measuring chemical shift anisotropy (CSA) tensor, spin-lattice relaxation time, and local correlation time at twenty-one crystallographically different 13C nuclei sites. The principal components of CSA tensor of cefpodoxime proxetil are extracted by the two-dimensional phase adjusted sinning sideband (2DPASS) cross-polarization magic angle spinning (CP-MAS) solid-state NMR experiment, and the spin-lattice relaxation time is measured by the method outlined by Torchia(T1CP). The local correlation time is calculated by bearing in mind that the spin-lattice relaxation mechanism of 13C nuclei is mainly governed by the CSA interaction and the heteronuclear dipole-dipole interaction. The aminothiazole ring, β-lactam ring, and dihydrothiazine ring provide stability to the drug molecule and increase the affinity of the drug to penicillin-binding proteins (PBPs) receptors. The principal components of CSA parameters, spin-lattice relaxation time, and local correlation time vary substantially for carbon nuclei residing on these three rings. These signify that not only the electronic environment, but the molecular conformation, and the local dynamics are also altered within the ring. The substitution of the acyl side chain, oxime group, and the aminothiazole ring at the C7 position of the β-lactam ring enhances the antibacterial activity and the binding affinity of the drug. A huge variation of the spin-lattice relaxation time and local correlation time is observed in those regions. The change in the electron charge distribution and nuclear spin dynamics at different parts of the drug molecule is clear by CSA and spin-lattice relaxation measurements, which will enrich the field “NMR crystallography”.

Structure and dynamics of sodium alginate as elucidated by chemical shift anisotropy and site-specific spin-lattice relaxation time measurements.

The biocompatible, biodegradable, linear copolymer sodium alginate is fabricated from [Formula: see text] linked [Formula: see text]-D-mannuronic acid (M block) and [Formula: see text]-L-guluronic acid (G-block). It has wide applications in drug delivery, cell encapsulation, and commercial application in the textile, cosmetics, paper, food, biomedical, and pharmaceutical industries. The structure and dynamics of sodium alginate were here investigated by measuring chemical shift anisotropy (CSA) parameters, spin-lattice relaxation time, and molecular correlation time. The principal components of the CSA tensor were determined by two-dimensional phase-adjusted spinning sideband (2DPASS) cross-polarization magic angle spinning (CP-MAS) SSNMR. The alternating M and G blocks of both equatorial and axial links are associated with greater overall flexibility. The molecular correlation time of the carboxyl carbon of both G and M blocks is faster than for the anomeric carbon and pyranose carbon. This is further experimental evidence of the coexistence of two different dynamics within the polysaccharide chains of sodium alginate, which was previously established by 1H-13C dipolar profile analysis. The relaxation time of the para-crystalline region of sodium alginate is comparable with that of chitosan, but it is much shorter than that of cellulose and chitin. The order of the molecular correlation time of sodium alginate and chitosan is also the same. Hence, it can be concluded that sodium alginate exhibits greater flexibility than cellulose and chitin. These types of investigation into the local electronic configuration and nuclear spin dynamics at various carbon nuclei sites of the biopolymer at atomic-scale resolution will help in the design of biomimetic materials.

Investigation on the Effect of Charge Carriers in Chemical Shift Anisotropy of Si Single Crystal

The Chemical Shift Anisotropy (CSA) of single crystalline silicon (Si) was investigated using 29Si static NMR for commercially procured p-type and n-type silicon. The observed CSA has been distributed between -80 ppm to -90 ppm for n-type and p-type Si. The NMR parameters such as Isotropic Chemical Shift (5iso), Chemical Shift Anisotropy tensor (ΔS) and the asymmetry parameter (77) of the CSA were extracted by using DMFIT with the CSA-static model. The ΔS and rj are found to be -1.73 ppm and 0.3 ppm respectively for the n-type and the asymmetry parameter of the CSA was found one order different from p-type material. The changes observed in CSA pattern and spin-lattice relaxation (T 1) time are ascribed to strength (mobility) of the shallow donor level and shallow acceptor level effects in the Fermi contact term.

Investigation of the Structure and Dynamics of Antiviral Drug Adefovir Dipivoxil by Site-Specific Spin-Lattice Relaxation Time Measurements and Chemical Shift Anisotropy Tensor Measurements.

Adefovir is regarded as a potential antiviral agent. However, it cannot be considered as a valuable drug candidate due to its high polarity that limits its permeability across the human intestinal mucosa. When the ribose phosphate group of adefovir is replaced by the isopolar phosphonomethyl ether functionality, it neutralizes the negative charge of the drug. This makes the drug lipid-soluble and potent to diffuse across the cell membrane. The prodrug adefovir dipivoxil is regarded as a potent antiviral drug against hepatitis B virus (HBV), human immunodeficiency virus (HIV), Rauscher murine leukemia virus (R-MuLV), murine cytomegalovirus (MCMV), herpes simplex virus (HSV), simian immunodeficiency virus (SIV), and feline immunodeficiency virus (FIV). The correlation between the structure and the dynamics of adefovir dipivoxil is determined by measuring the principal components of chemical shift anisotropy (CSA) tensor, site-specific spin–lattice relaxation time, and molecular correlation time at crystallographically different carbon nuclei sites. The CSA parameters, spin–lattice relaxation time, and molecular correlation time of phosphorous nucleus of the organophosphate group of adefovir dipivoxil molecule are also determined. The spin–lattice relaxation time of carbon nuclei varies from 1 to 107 s. The range of molecular correlation time also varies from 10–4 to 10–8 s. These remarkable diversities of motional dynamics of the molecules imply that there exist various motional degrees of freedom within this valuable drug and these motional degrees of freedom are independent of each other, which may be the reason for the biological activities exhibited by the drug. The correlation between structure and dynamics of such an important antiviral drug adefovir dipivoxil can be visualized by these types of extensive spectroscopic measurements, which will enlighten the path of inventing advanced medicine in the pharmaceutical industry, and it will also illuminate the understanding of the structure–activity relationships of antiviral drug.

Relaxation-induced dipolar exchange with recoupling (RIDER) distortions in CODEX experiments.

Chemical shift anisotropy (CSA) and dipolar CODEX (Cenralband Only Detection of EXchange) experiments enable abundant quantitative information on the reorientation of the CSA and dipolar tensors to be obtained on millisecond-second timescales. At the same time, proper performance of the experiments and data analysis can often be a challenge since CODEX is prone to some interfering effects that may lead to incorrect interpretation of the experimental results. One of the most important such effects is RIDER (relaxation-induced dipolar exchange with recoupling). It appears due to the dipolar interaction of the observed nuclei with some other nuclei, which causes an apparent decay in the mixing time dependence of the signal intensity reflecting not molecular motion, but spin flips of the adjacent nuclei. This may hamper obtaining correct values of the parameters of molecular mobility. In this contribution we consider in detail the reasons why the RIDER distortions remain even under decoupling conditions and propose measures to eliminate them. That is, we suggest (1)using an additional filter between the cross-polarization (CP) section and the CODEX recoupling blocks that suppresses the interfering anti-phase coherence responsible for the -H RIDER and (2)recording only the cosine component of the CODEX signal since it is less prone to the RIDER distortions in comparison to the sine component. The experiments were conducted on rigid model substances as well as microcrystalline H N-enriched proteins (GB1 and SH3) with a partial back-exchange of labile protons. Standard CSA and dipolar CODEX experiments reveal a fast-decaying component in the mixing time dependence of N nuclei in proteins, which can be misinterpreted as a slow overall protein rocking motion. However, the RIDER-free experimental setup provides flat mixing time dependences, meaning that the studied proteins do not undergo global motions on the millisecond timescale.

Endohedral Plumbaspherenes of the Group 9 Metals: Synthesis, Structure and Properties of the [M@Pb12]3− (M=Co, Rh, Ir) Ions

The icosahedral [M@Pb12 ]3- (M=Co(1), Rh(2), Ir(3)) cluster ions were prepared from K4 Pb9 and Co(dppe)Cl2 (dppe=1,2-bis(diphenylphosphino)ethane)/[Rh(PPh3 )3 Cl]/[Ir(cod)Cl]2 (cod=1,5-cyclooctadiene), respectively, in the presence of 18-crown-6/ 2,2,2-cryptand in ethylenediamine/toluene solvent mixtures. The [K(2,2,2-cryptand)]+ salt of 1 and the [K(18-crown-6)]+ salt of 3 were characterized via X-ray crystallography; the ions 1 and 3 are isostructural and isoelectronic to the [Rh@Pb12 ]3- (2) ion as well as to the group 10 clusters [M'@Pb12 ]2- (M'=Ni, Pd, Pt). The ions are all 26-electron clusters with near perfect icosahedral Ih point symmetry. Clusters 1-3 show record downfield 207 Pb NMR chemical shifts due to σ-aromaticity of the cluster framework. Calculated and observed 207 Pb NMR chemical shifts and 207 Pb-x M J-couplings (x M=59 Co, 103 Rh, 193 Ir) are in excellent agreement and DFT analysis shows that the variations of 207 Pb NMR chemical shifts for the [M@Pb12 ]2, 3- ions (M=Co, Rh, Ir, Ni, Pd, Pt) are mainly governed by the perpendicularly oriented σ11 component of the chemical shift anisotropy tensor. The laser desorption ionization time-of-flight (LDI-TOF) mass spectra contain the molecular ions as well as several new gas phase clusters derived from the parents. The DFT-minimized structures of these ions are described.

Elucidating natural product structures using a robust measurement of carbon residual chemical shift anisotropy combined with DFT.

Determination of configurations and conformations is an important step in the structural characterization of small molecules. Apart from utilizing isotropic J-couplings and nuclear overhauser effect (NOEs) measured in isotropic solution, anisotropic Nuclear Magnetic resonance (NMR) data such as residual dipolar couplings and residual chemical shift anisotropies (RCSAs) were also used to elucidate complex small molecule structures. Measuring RCSA has always been historically difficult due to the isotropic shift effect accompanied by molecular alignment and therefore only occasionally applied in a few examples. Here, we present a robust measurement of carbon RCSAs using a smaller gel-stretching device to determine the structures of a few small molecules. A systematic study on how different density functional theory computed anisotropies of the chemical shift anisotropy tensors impact RCSA data interpretation has also been discussed. We also discuss the effect of utilizing various carbons as reference nuclei for RCSA data extraction as well as the orientation behavior of estrone in orthogonal alignment media.

An atomic resolution description of folic acid using solid state NMR measurements.

The chemical shift anisotropy tensor and site-specific spin-lattice relaxation time of folic acid were determined by a 13C 2DPASS CP-MAS NMR experiment and Torchia CP experiment respectively. The molecular correlation time at various carbon nuclei sites of folic acid was evaluated by assuming that the 13C spin-lattice relaxation mechanism is mainly governed by chemical shift anisotropy interaction and hetero-nuclear dipole–dipole coupling. CSA parameters are larger for the carbon nuclei residing at the heteroaromatic ring and aromatic ring, and those attached to double-bonded electronegative oxygen atoms. It is comparatively low for C9, C19, C21, and C22. The molecular correlation time is of the order of 10−4/10−5 s for C9, C19, C21 and C22 carbon nuclei, whereas it is of the order of 10−3 s for the rest of the carbon nuclei sites. Spin lattice relaxation time varies from 416 s to 816 s. For C23 and C14, the value is 816 s, and it is 416 s for C7 nuclei. The correlation between structure and dynamics on an atomic scale of such an important drug as folic acid can be visualized by these types of extensive spectroscopic measurements, which will help to develop an advanced drug for DNA replication.

Determination of chemical shift anisotropy tensor and molecular correlation time of proton pump inhibitor omeprazole by solid state NMR measurements

The correlation between the structure and dynamics of omeprazole is portrayed by extracting CSA parameters through the 13C 2DPASS CP-MAS SSNMR experiment, site specific spin–lattice relaxation time by Torchia CP experiment, and calculation of the molecular correlation time.
