Development and characterization of nanostructured lipid carriers for cannabidiol delivery

This study evaluated the physicochemical characteristics of nanostructured lipid carriers (NLCs) as a potential vehicle for cannabidiol (CBD), a lipophilic molecule with great potential to promote health benefits. NLCs were produced using hemp seed oil and fully-hydrogenated soybean oil at different proportions. The emulsifiers evaluated were soybean lecithin (SL), Tween 80 (T80) and a mixture of SL:T80 (50:50). CBD was tested in the form of CBD-rich extract or isolate CBD, to verify if it affects the NLCs characteristics. Based on particle size and polydispersity, SL was considered the most suitable emulsifier to produce the NLCs. All lipid proportions evaluated had no remarkable effect on the physicochemical characteristics of NLCs, resulting in CBD–loaded NLCs with particle size below 250 nm, high CBD entrapment efficiency and CBD retention rate of 100% for 30 days, demonstrating that NLCs are a suitable vehicle for both CBD-rich extract or isolate CBD.

Optimization and evaluation of microwave-assisted curcumin-loaded nanostructured lipid carriers: a green approach

BackgroundThe goal of current research work is to develop and optimize curcumin-encapsulated nanostructured lipid carriers and to enhance therapeutic effect of curcumin after oral administration.MethodCurcumin-loaded nanostructured lipid carriers were developed by a single-step one-pot microwave-assisted technique. The preparation of curcumin-loaded nanostructured lipid carriers was optimized by employing two factors and three levels central composite design (Design Expert® software) taking concentration of lipid blend and surfactant as independent variables and particle size, polydispersity index, and zeta potential as dependent variables, to investigate the effect of formulation ingredients on the physicochemical characteristics of nanostructured lipid carriers. The optimized batch was investigated by Fourier transform infrared spectroscopy, differential scanning calorimetry, X-ray diffraction, high-resolution transmission electron microscopy, in vitro drug release, stability studies, cytotoxicity, and in vivo anthelmintic studies.ResultsThe average particle size, polydispersity index, and zeta potential of the optimized batch were found to be 144 nm, 0.301, and − 33.2 mV, respectively, with an entrapment efficiency of 92.48%. The results of high-resolution transmission electron microscopy confirmed spherical shape of particles. In vivo antiparasitic studies included determining the duration of paralysis and eventual death of earthworms in the presence of test samples. The results of in vivo studies showed good anthelmintic potential for curcumin-loaded nanostructured lipid carriers as compared to albendazole in different concentrations. Cytotoxicity studies also confirmed the formulation to be nontoxic to Vero cells. In vitro drug release study showed 90.76 ± 0.01% release of curcumin in 24 h by following the Korsmeyer-Peppas model of release kinetics.ConclusionThe aforementioned results imply that microwave-developed nanostructured lipid carriers could be promising drug carriers and will aid in their fabrication for oral administration as a possible alternative for the treatment of other parasitic infections.Graphical

Because of the Non-specificity of anticancer drugs, they kill normal healthy cells along with cancer cells which lead to severe side effects. To minimize such limitations associated with conventional chemotherapy, nanostructured lipids carriers can be developed. These are nanocarriers consisting of a mixture of solid and liquid lipids along with surfactants. Lipids used in the formulation of NLCs are biocompatible and biodegradable. NLCs ensure high drug payload, less drug expulsion and more stability on storage. NLCs enhance the aqueous solubility of lipophilic anticancer drugs. Their surface modification can help to overcome drug resistance in cancer therapy. Controlled and targeted drug delivery of anticancer drugs can be possible by formulating them as NLCs. NLCs can play an important role in targeting anticancer drugs by different mechanisms. NANOSTRUCTURED LIPID CARRIERS FOR TRANSDERMAL DRUG DELIVERY

Transdermal drug delivery offers many advantages over oral delivery, such as avoiding first-pass metabolism, enhancing the bioavailability of poorly soluble drugs, and providing better patient compliance. However, only small lipophilic molecules can be delivered across the stratum corneum, the outermost layer of the skin. Unfortunately, the delivery of larger molecules remains a challenge. Nanostructured lipid carriers (NLCs) are second-generation lipid nanocarriers composed of biocompatible solid lipids, liquid lipids, surfactants, and co-surfactants. NLCs can be loaded with various classes of drugs, provide a controlled drug release profile, enhance drug stability, and be scaled up without needing organic solvents. This review article discusses the features, composition, formulation processes, and characterization of NLCs and their potential use in transdermal drug delivery.

Developed and characterization of nanostructured lipid carriers containing food-grade interesterified lipid phase for food application

The main objectives of our work were to produce new nanostructured lipid carriers (NLCs) from interesterified and simple lipid phases and to study the influence of lipid composition on the physical characteristics and stability of NLCs. We used conventional oils and fats already used in lipid-based foods, in addition to soy lecithin as a natural emulsifier. The NLCs were formulated using as lipid phase, simple and interesterified blends composed of soybean oil as liquid lipid and fully hydrogenated oils from palm, soybean, microalgae, and crambe as solid lipids. NLCs were produced using high-pressure homogenization. NLCs were influenced by chemical interesterification and by the composition of the solid lipid used, mainly in relation to the fatty acid chain size. NLC formulations developed with a simple lipid phase were 256–323 nm (d32) in size, with zeta potential values ranging from −36.93 to −42.87 mV after 60 days of storage. NLCs developed with the interesterified lipid phase were 250–288 nm (d32) in size, with zeta potential values ranging from −40.17 to −44.20 mV after 60 days of storage. NLCs produced with saturated fatty acids with larger chain sizes showed larger particle sizes but showed less variation in this parameter over storage. Interesterification reduced the melting temperature of NLCs, indicating decreased crystallinity and a less organized structure. Moreover, interesterification favored crystals in the β’ form, which is a positive characteristic for incorporating bioactive compounds. Thus, the systems developed in this study are innovative, mainly in terms of the composition of the NLCs, and have good potential for food applications.

Formulation, Optimization, and Ex-vivo Evaluation of Novel Lipid Carriers for Enhanced Transdermal Delivery of Hydroquinone

Background: HQ is used for hyper-pigmentation treatment using conventional creams and gels. These formulations show various disadvantages like poor skin permeation, allergic reactions, and repeated use and decreasing patient compliance. Objectives: The present work involved formulation, statistical optimization, and characterization of Nanostructured Lipid Carriers (NLCs) for efficient topical delivery of hydroquinone (HQ) for hyperpigmentation treatment. Methods: The NLCs were optimized exploring Box–Behnken Design (BBD) using three independent variables and two dependent variables. Formulation having the minimum size and maximum drug entrapment was considered as optimized formulation. The optimized formulation was evaluated for drug release followed by its freeze-drying. The freeze-dried formulation was subjected to Differential Scanning Calorimetry (DSC) analysis, X-Ray Diffraction (XRD) analysis, and Fourier Transform-Infrared spectroscopy (FT-IR) analysis. Furthermore, NLCs based gel was prepared by using Carbopol 934 as a gelling agent. NLCs based gel was evaluated for skin permeation, skin retention, and skin distribution (through confocal microscopic analysis) using pig ear skin. Results: Optimized NLCs showed smaller particle size [(271.9 ± 9) nm], high drug entrapment [(66.4 ± 1.2) %], tolerable polydispersity index (PDI) (0.221 ± 0.012), and zeta potential [(-25.9 ± 1.2) mV]. The FT-IR analysis revealed excellent compatibility between HQ and other excipients. The Carbopol 934 gel containing NLCs showed high transdermal flux [(163 ± 16.2) μg/cm2/h], permeability coefficient (0.0326 ± 0.0016), and skin permeation enhancement ratio (3.7 ± 0.4) compared to marketed cream of HQ. The results of confocal microscopic (CLSM) analysis revealed the accumulation of optimized NLCs in the lower epidermal layers of skin. Conclusion: NLCs based gel was considered effective in the topical delivery of HQ to treat hyperpigmentation due to high skin permeation, skin retention, and prolonged release of HQ.

NANOSTRUCTURED LIPID CARRIERS: A PROMISING APPROACH FOR TOPICAL DRUG DELIVERY SYSTEM

Nanostructured lipid carriers (NLCs) are innovative pharmaceutical formulations made up of physiological and biocompatible lipids, as well as surfactants and co-surfactant. The initial generation of lipid nanoparticles was Solid Lipid Nanoparticles (SLN), which had a longer-lasting activity and was better, suited to drug penetration. The NLC is a second-generation lipid nanoparticle designed to alleviate the limitations of SLN, such as limited drug loading capacity and solid lipid polymorphism. Many benefits of topical medication delivery include avoiding first-pass metabolism, focusing active components for a local effect, and patient compliance. When compared to typical topical dose forms, nanoparticles have a greater effect in transporting medications through the skin. The structure, composition, many formulation methods, and characterization of NLCs are all important aspects in formulating a stable drug delivery system, as discussed in this review paper. A variety of approaches are used to make solid lipid nanoparticles and nanostructured lipid carriers, which are discussed in this paper. Lipid nanoparticles have a variety of features that make them suitable for topical usage in cosmetics and medicinal formulations. Because of extensive positive benefits such as skin hydration, skin occlusion, and skin targeting, NLCs have a significant potential in the pharmaceutical market. Skin hydration is important for API topical distribution because it hydrates the skin, which causes the pores to open. Trans epidermal water loss decreases due to the occlusion nature of lipid nanoparticles, softening the skin. It is more suited since it uses biodegradable grade lipid, which does not cause toxicity like polymeric Nano formulations. Actually, because of their biodegradable composition, NLCs are a “Nano safe” carrier that has a lot of potential for overcoming the obstacles of topical distribution.

Design and characterization of Nanostructured lipid carriers (NLC) and Nanostructured lipid carrier-based hydrogels containing Passiflora edulis seeds oil

This study aims to design and characterize Nanostructured lipid carriers (NLC) and Nanostructured lipid carrier-based hydrogels with Passiflora edulis seeds oil, a by-product from Madeira Island food industry. NLC were prepared by the ultrasonication technique, using passion fruit seeds oil as a liquid lipid and glyceryl distearate as a solid lipid. These NLC were then gelled with Poly (acrylic acid). Long-term stability studies were conducted with NLC and NLC-based hydrogels stored for 12 months. The following tests were performed: morphology, encapsulation efficiency, particle size analysis, polydispersity index analysis, zeta potential, pH measurement, color analysis, viscosity studies, texture analysis, in vitro occlusion test, ex vivo skin penetration study, tyrosinase inhibition activity, in vitro skin permeation experiments and in vitro cytotoxicity studies. The developed NLC had spherical shape and narrow particle sizes distribution with mean sizes in the range of 150 nm and PDI below 0.3, Zeta potential values around −30 mV and high Encapsulation efficiency. The tyrosinase inhibitory activity and skin retention of the nanoparticles was superior to that of the non-encapsulated oil. The developed formulations did not show cytotoxicity towards HaCat cells and presented suitable viscosity and texture properties for skin application, proving to be good candidates as depigmenting agent.

Formulation, optimization and characterization of nanostructured lipid carrier of nitrofurantoin

Nitrofurantoin is effective against many urinary tract pathogens. It acts as bacteriostatic and/or bactericidal by inhibiting DNA-RNA protein& cell wall synthesis. Nanostructured Lipid Carriers (NLCs) of NFT was prepared by Hot Homogenization Process. Glyceryl Monostearate and Miglyol 812 were heated at 80ºC temperature on hot plate. In the melted lipid, drug was added with continuous stirring at high speed homogenization. Formulation NLC12B has % Entrapment efficiency 89.1 ± 0.5, PDI 0.11 ± 0.01 and mean particle size 237 ± 7nm represents narrow particle size distribution. Spherical feature of NLCs with better uniformity without aggregation of Nitrofurantoin loaded NLC was confirmed by TEM. Moreover, efficient miscibility of drug in lipids was confirmed by the absence of intense and characteristic peak of NFT in XRPD. After 6 month storage at 2-8°C there was no significant changes in the PDI as well as mean particle size.

Nanostructured Lipid Carriers: A Groundbreaking Approach for Transdermal Drug Delivery.

Nanostructured lipid carriers (NLCs) are novel pharmaceutical formulations which are composed of physiological and biocompatible lipids, surfactants and co-surfactants. Over time, as a second generation lipid nanocarrier NLC has emerged as an alternative to first generation nanoparticles. This review article highlights the structure, composition, various formulation methodologies, and characterization of NLCs which are prerequisites in formulating a stable drug delivery system. NLCs hold an eminent potential in pharmaceuticals and cosmetics market because of extensive beneficial effects like skin hydration, occlusion, enhanced bioavailability, and skin targeting. This article aims to evoke an interest in the current state of art NLC by discussing their promising assistance in topical drug delivery system. The key attributes of NLC that make them a promising drug delivery system are ease of preparation, biocompatibility, the feasibility of scale up, non-toxicity, improved drug loading, and stability.

Lipid Nanocarriers for Enhanced Delivery of Temozolomide to the Brain.

Brain disorders, a diverse range of conditions comprising of neurological and psychiatric conditions, are the leading cause of disability, severely affect the quality of life, and in many cases lead to mortality. The prime challenge in treatment of brain disorders is to deliver therapeutics by overcoming the blood-brain barrier (BBB), a unique anatomical and physiological barrier which restricts the passage of a number of molecules, proteins, and cells from the bloodstream. Lipid nanoparticles have emerged as promising drug delivery systems primarily because of biodegradability, low toxicity potential, and the ability to cross physiological barriers especially the BBB even without surface modifications.In this chapter we discuss the preparation and characterization of nanostructured lipid carriers of temozolomide, a chemotherapeutic drug. Evaluation of pharmacokinetics and biodistribution of the nanocarrier system in rats revealed improved delivery of the chemotherapeutic agent to the brain with the potential of lesser side effects.

Fabrication and characterization of nanostructured lipid carriers (NLC) using a plant-based emulsifier: Quillaja saponin.

Nanostructured lipid carriers (NLCs) are a type of colloidal delivery system that was developed in the pharmaceutical industry to combine the advantages and eliminate the shortcomings of oil-in-water (O/W) nanoemulsions and solid lipid nanoparticles (SLNs). The hydrophobic core of the particles within NLCs consists of a solidified fat phase with a partially disorganized structure, which inhibits morphological changes and bioactive expulsion. In the present study, we formulated NLCs using a hot-homogenization approach using fully hydrogenated soybean oil (HSO) as the lipid phase and quillaja saponins as a natural surfactant. The NLCs formed had a low viscosity and milky white appearance similar to that of O/W nanoemulsions. The fabrication conditions were optimized, including the number of passes through the microfluidizer, stirring conditions, cooling rate, and emulsifier level. Unlike bulk HSO, the emulsified form had to be supercooled substantially to promote crystallization of the lipid droplets, which was attributed to differences in nucleation behavior. The crystallization temperature decreased with increasing saponin concentration, which was probably because smaller droplets were formed at higher emulsifier levels. For instance, at 3, 6, 9, and 12 wt% saponin, the degree of supercooling was 10, 15, 18, and 18 °C, while the mean particle diameter was 0.82, 0.53, 0.41, and 0.44 μm, respectively. The melting and crystallization behavior of the NLCs was characterized using an optical microscope and differential scanning calorimetry (DSC), while the morphology of the NLCs was characterized using transmission electron microscopy (TEM). This analysis showed that the NLCs contained spherical particles with a crystallization temperature around 31 °C. This information may be useful for formulating NLC from natural ingredients for application in the food and beverage industry.

Comprehensive characterization of nanostructured lipid carriers using laboratory and synchrotron X-ray scattering and diffraction

The development of lipid nanoparticles requires knowledge on the crystalline structure, polymorphic transitions and lipid-drug interactions. This study aimed at introducing advanced techniques to characterize nanostructured lipid carriers (NLC) comprising palmitic acid, oleic acid, stabilizer and Domperidone. Crystallinity of single components and mixtures was investigated by laboratory Small Angle X-ray Scattering (SAXS). NLC were studied with laboratory Small and Wide Angle X-ray Scattering (SWAXS). Photon Correlation Spectroscopy and Freeze Fracture Transmission Electron Microscopy were used to monitor particle size, zeta potential and shape. Stability of NLC was investigated using synchrotron X-ray Diffraction (XRD) and SAXS and laboratory SAXS. Palmitic acid showed a lamellar structure (polymorph C), which was still present after particle preparation. Spherical 300 nm-sized particles with zeta potential values above −30 mV were obtained and Domperidone was incorporated in its amorphous form. During storage, no differences in synchrotron XRD spectra were seen. However, laboratory SAXS measurements showed a second lamellar structure, identified as polymorph B. Synchrotron SAXS temperature scans confirmed that polymorph B did not affect the morphology of the encapsulated drug or the shape of NLC. These results highlight the unique capabilities of laboratory and synchrotron X-ray Scattering and Diffraction for improved structural characterization of lipid nanoparticles.

Size and Vitamin E Release of Nanostructured Lipid Carriers with Different Liquid Lipids, Surfactants and Preparation Methods

Solid lipid nanoparticles have excellent properties as delivery systems for dermal cosmetic and pharmaceutical formulations because they have good compatibility with the skin, protect the encapsulated substance from the medium, and control its release at the site of action. This work describes the preparation and characterization of nanostructured lipid carriers (NLC) containing vitamin E (NLC‐VE). The emulsion‐solidification technique was used to prepare the particles. Three non‐ionic surfactants (Tween 80, Cremophor RH40 and Pluronic F‐127) were used in two concentrations (5 and 10% (w/w)) and three liquid lipids (avocado oil, coconut oil and medium chain triglycerides (MCT)) were mixed with the solid lipid (stearic acid) to form the oil phase. Some processing variables (emulsification procedure, and time and energy during sonication) were modified to find the most suitable NLC in terms of particle size and release profile. Higher surfactant concentration produced more homogeneous nanoparticles with smaller diameter. The liquid lipid and its proportion in the oil phase also modified the characteristics of NLC‐VE. Time and sonication energy did not strongly affect the particle size (24–28 nm), with the exception of the highest time and amplitude, which produced particles with 14 nm. NLC‐VE prepared with stearic acid as solid lipid, 10% (w/w) Tween 80 as surfactant, and MCT as liquid lipid in a 1:3 mass proportion of solid lipid‐to‐liquid lipid, had a unimodal size distribution and the best vitamin E release (50% in 24 h). Results indicate that this system can be used to improve the delivery of vitamin E.

Preparation and characterization of nanostructured lipid carriers as drug delivery system: Influence of liquid lipid types on loading and cytotoxicity

In this study, we aimed to investigate the influence of liquid lipid types on different features of NLC. Four variations of liquid lipids such as coconut oil, fish oil, black seed oil and linseed oil were used, while for all variations, cetyl palmitate was used as the solid lipid. Different NLC were characterized and compared in terms of particle size, zeta potential, polydispersity index (PDI), drug entrapment percentage and drug loading capacity. The results indicated that NLC containing black seed oil has the smallest size. Other features like PDI, zeta potential and entrapment efficiency were the same for all the liquid lipids. By close margins, the NLC containing black seed oil had the highest percent of drug release and antioxidant activity compared to the rest. Diffusion was the major mechanism of the drug release according to the drug release kinetic fitted by Higuchi’s model. Differential scanning calorimetry (DSC) and fourier transform infrared spectroscopy (FT-IR) confirmed no strong interaction between NLC constituents. The particles showed spherical shape morphology under atomic force microscopy (AFM). According to the cell viability assay on MCF-7 cell line, the curcumin loaded NLC composed of linseed oil showed better cytotoxic activity compared to the free curcumin.

Production and characterization of nanostructured lipid carriers and solid lipid nanoparticles containing lycopene for food fortification.

In this study, lycopene, was loaded on nanostructured lipid carrier and solid lipid nanoparticles using combination of high shear homogenization and ultrasonication method. Effect of applied lipids types, nanocarrier's type and lycopene loading on physicochemical properties of developed nanocarriers were studied. Particle sizes of developed nanocarriers were between 74.93 and 183.40nm. Encapsulation efficiency of nanostructured lipid carrier was significantly higher than solid lipid nanoparticles. Morphological study of developed nanocarriers using scanning electron microscopy showed spherical nanoparticles with smooth surface. Lycopene was entrapped in nanocarriers without any chemical interaction with coating material according to Fourier transform infrared spectroscopy spectrum and differential scanning calorimetry thermogram. Glycerol monostearate containing nanoparticles showed phase separation after 30days in 6 and 25°C, whereas this event was not observed in nanosuspensions that produced by glycerol distearate. Lycopene release in gastrointestinal condition was studied by the dialysis bag method. To evaluate nanocarrier's potential for food fortification, developed lycopene-loaded nanocarriers were added to orange drink. Results of sensory analysis indicated that nanoencapsulation could obviate the poor solubility and tomato after taste of lycopene. Fortified sample with lycopene nanocarriers didn't show significant difference with blank orange drink sample except in orange odor.

Development and characterization of nanostructured lipid carriers based chitosan thermosensitive hydrogel for delivery of dexamethasone

This study aims to explore a novel composite thermosensitive in situ gelling formulation which the nanostructured lipid carriers (NLC) was incorporated into hydroxypropyltrimethyl ammonium chloride chitosan (HACC)-based hydrogels, and the resulting formulation investigated for its potential to act as a potential sustained ocular delivery system. NLC formulation loaded with dexamethasone (DXM) were prepared using the melt-emulsification method. The particle size, zeta potential, encapsulation efficiency, and morphological properties of the NLC were characterized. The HACC was synthesized and structure was analyzed by FT-IR and 1H NMR. A thermosensitive hydrogel was designed and prepared by simply mixing HACC and β-glycerophosphate (β-GP). The obtained formulation showed a rapid solution-to-gel transition at 35°C. The NLC were then incorporated in HACC/β-GP hydrogel to form a NLC-loaded hydrogel carrier. In vitro release studies, 88.65% of total DXM was released from the NLC-HACC/GP gel within 3days, indicating DXM-based NLC-gel could release drug sustainably. Taken together, DXM-based NLC-HACC/GP gel is a promising drug delivery system.

Objective: The aim of this study was to investigate physical characteristics of nanostructured lipid carriers (NLCs) mixture of alpha tocopheryl acetate, cetyl palmitate, Tween 80 and propylene glycol using high shear homogenization technique on NLC preparation to predict the optimum ratio of alpha tocopheryl acetate-cetyl palmitate to produce good characteristics of NLC loaded coenzyme, higher % EE, good penetration, controlled release, and stable.Methods: Lipid characterizations were conducted by diffraction scanning calorimetry, X-ray diffraction, and Fourier transforms infrared spectrophotometry. Coenzyme Q10 concentration was measured by spectrophotometer at 275 nm. NLC characteristics based on their morphology was determined using transmission electron microscope, particle size, and its polydispersity index which were measured with Delsa Nano™ particle size analyzer. Percentage of coenzyme Q10 entrapped in NLC was determined by dialysis bag method. Coenzyme Q10 release profile was measured using with Franz cell for 12 hrs. The penetration depth of NLC coenzyme Q10 in abdominal skin of Wistar rat was determined with fluorescence microscopy using rhodamine B as marker. NLC physical stability based on minimum of particle size variation, pH and viscosity during 90 days storage.Results: The result showed that formula with ratio of cetyl palmitate-alpha tocopheryl acetate 70:30 (% w/w) produce good characteristics of NLCloaded coenzyme, higher % EE, good penetration, controlled release, and stable in 90 days storage.Conclusion: The coenzyme Q10 NLC system with cetyl palmitate and alpha tocopherol acetate as lipid matrixare characterized by small particle size, low crystallinity, spherical morphology of particle and high coenzyme Q10 entrapment efficiency.
