Determination of fatty acid composition after saponification of common oil pharmaceutical excipients by supercritical fluid-evaporative light scattering method and its application in oil identification

Oils and fats are commonly used in the pharmaceutical industry as solvents, emulsifiers, wetting agents, and dispersants, and are an important category of pharmaceutical excipients. Fatty acids with unique compositions are important components of oil pharmaceutical excipients. The Chinese Pharmacopoeia provides clear descriptions of the fatty acid types and limits suitable for individual oil pharmaceutical excipient. An unqualified fatty acid composition or content may indicate adulteration or deterioration. The fatty acid composition, as a key indicator for the identification and adulteration evaluation of oil pharmaceutical excipients, can directly affect the quality and safety of oil pharmaceutical excipients and preparations. Gas chromatography is the most widely used technique for fatty acid analysis, but it generally requires derivatization, which affects quantitative accuracy. Supercritical fluid chromatography (SFC), an environmentally friendly technique with excellent separation capability, offers an efficient method for detecting fatty acids without derivatization. Unlike other chromatographic methods, SFC does not use nonvolatile solvents (e. g., water) as the mobile phase, rendering it compatible with an evaporative light-scattering detector (ELSD) for enhanced detection sensitivity. However, the fatty acids in oil pharmaceutical excipients exist in the free and bound forms, and the low content of free fatty acids in these oil pharmaceutical excipients not only poses challenges for their detection but also complicates the determination of characteristic fatty acid compositions and contents. Moreover, the compositions and ratios of fatty acids are influenced by environmental factors, leading to interconversion between their two forms. In this context, saponification provides a simpler and faster alternative to derivatization. Saponification degrades oils and fats by utilizing the reaction between esters and an alkaline solution, ultimately releasing the corresponding fatty acids. Because this method is more cost effective than derivatization, it is a suitable pretreatment method for the detection of fatty acids in oil pharmaceutical excipients using the SFC-ELSD approach. In this study, we employed SFC-ELSD to simultaneously determine six fatty acids, namely, myristic acid, palmitic acid, stearic acid, arachidic acid, docosanoic acid, and lignoceric acid, in oil pharmaceutical excipients. Saponification of the oil pharmaceutical excipients using sodium hydroxide methanol solution effectively avoided the bias in the determination of fatty acid species and contents caused by the interconversion of fatty acids and esters. The separation of the six fatty acids was achieved within 12 min, with good linearity within their respective mass concentration ranges. The limits of detection and quantification were 5-10 mg/L and 10-25 mg/L, respectively, and the spiked recoveries were 80.93%-111.66%. The method proved to be sensitive, reproducible, and stable, adequately meeting requirements for the analysis of fatty acids in oil pharmaceutical excipients. Finally, the analytical method was successfully applied to the determination of six fatty acids in five types of oil pharmaceutical excipients, namely, corn oil, soybean oil, coconut oil, olive oil, and peanut oil. It can be combined with principal component analysis to accurately differentiate different types of oil pharmaceutical excipients, providing technical support for the rapid identification and quality control of oil pharmaceutical excipients. Thus, the proposed method may potentially be applied to the analysis of complex systems adulterated with oil pharmaceutical excipients.

Lower Plasmatic Levels of Saturated Fatty Acids and a Characteristic Fatty Acid Composition in the Ovary Could Contribute to the High-Fertility Phenotype in Dummerstorf Superfertile Mice.

In recent decades, fertility traits in humans as well as in farm animals have decreased worldwide. As such, it is imperative to know more about the genetics and physiology of increased or high fertility. However, most of the current animal models with reproductive phenotypes describe lower fertility or even infertility (around 99%). The “Dummerstorf high-fertility lines” (FL1 and FL2) are two unique mouse lines selected for higher reproductive performances, more specifically for higher number of pups per litter. We recently described how those superfertile mice managed to increase their reproductive phenotype by doubling the ovulation rate and consequently the litter size compared to the unselected mice of the same founder population. FLs show an unusual estrous cycle length and atypical levels of hormones that link reproduction and metabolism, such as insulin in FL1 and leptin in FL2. Moreover, we described that their higher ovulation rate is mostly due to a higher quality of their oocytes rather than their sheer quantity, as they are characterized by a higher quantity of high-quality oocytes in antral follicles, but the quantity of follicles per ovary is not dissimilar compared to the control. In the present study, we aimed to analyze the lipid composition of the fertility lines from plasma to the gonads, as they can connect the higher reproductive performances with their metabolic atypicalities. As such, we analyzed the fat content of FLs and fatty acid composition in plasma, liver, fat, oocytes of different quality, and granulosa cells. We demonstrated that those mice show higher body weight and increased body fat content, but at the same time, they manage to decrease the lipid content in the ovarian fat compared to the abdominal fat, which could contribute to explaining their ovarian quality. In addition, we illustrate the differences in fatty acid composition in those tissues, especially a lower level of saturated fatty acids in plasma and a different lipid microenvironment of the ovary. Our ongoing and future research may be informative for farm animal biology as well as human reproductive medicine, mostly with cases that present characteristics of lower fertility that could be reversed following the way-of-managing of Dummerstorf high-fertility lines.

Effect of Flaxseed (Linum usitatissimum L.) Supplementation on Vascular Endothelial Cell Morphology and Function in Patients with Dyslipidaemia-A Preliminary Observation.

Context: Flaxseed has a characteristic fatty acids composition and unique phytonutrient profile that may have health-promoting properties. Objective: This study aimed to determine the effects of 10 weeks of supplementation with the flaxseed (28 g/day) on endothelial cells (EC) function, serum lipids and proinflammatory mediators in patients with mild and severe dyslipidaemia. Materials and methods: Eleven lean patients with severe dyslipidaemia treated with apheresis (group 1; 10 weeks treated in four phases: (i) ordinary diet, (ii) ordinary diet + flaxseed, (iii) ordinary diet (wash out), (iv) ordinary diet + placebo) and eleven obese patients with mild dyslipidaemia—not treated with apheresis (group 2; 10 weeks treated in two phases: (i) ordinary diet, (ii) low fat diet + flaxseed). Flaxseed was given blindly. Serum was collected at the end of each phase of the study. ECs were exposed in vitro to the medium supplemented with pooled serum taken from patients from both groups to detect their morphological changes using light and electron microscopy. ECs proliferation was also measured at the end of each study phase. Results: Serum vascular endothelial growth factor was decreased after flaxseed supplementation but only in group 1. ECs proliferation was increased after flaxseed supplementation only in obese patients. ECs exposed to medium supplemented with obese patients’ serum revealed the following cellular abnormalities: accumulation of lipid droplets, changes of rough endoplasmic reticulum and mitochondria, and flaxseed did not reverse observed changes. At the same time, flaxseed supplementation decreases total cholesterol in both tested groups, low-density lipoprotein cholesterol in group 1 and triglycerides in group 2. Conclusions: Our findings support the potential role of flaxseed in treating dyslipidaemia but indicate only a slight impact on endothelial cell function.

Occurrence of Cis-11,12-Methylene-Hexadecanoic Acid in the Red Alga Solieria pacifica (Yamada) Yoshida

Fatty acids in marine algae have attracted the attention of natural chemists because of their biological activity. The fatty acid compositions of the Solieriaceae families (Rhodophyceae, Gaigartinales) provide interesting information that unusual cyclic fatty acids have been occasionally found. A survey was conducted to profile the characteristic fatty acid composition of the red alga Solieria pacifica (Yamada) Yoshida using gas chromatography-mass spectrometry (GC-MS), infrared spectroscopy (IR), and proton nuclear magnetic resonance spectroscopy (1H-NMR). In S. pacifica, two cyclopentyl fatty acids, 11-cyclopentylundecanoic acid (7.0%), and 13-cyclopentyltridecanoic acid (4.9%), and a cyclopropane fatty acid, cis-11,12-methylene-hexadecanoic acid (7.9%) contributed significantly to the overall fatty acid profile. In particular, this cyclopropane fatty acid has been primarily found in bacteria, rumen microorganisms or foods of animal origin, and has not previously been found in any other algae. In addition, this alga contains a significant amount of the monoenoic acid cis-11-hexadecenoic acid (9.0%). Therefore, cis-11,12-methylene-hexadecanoic acid in S. pacifica was likely produced by methylene addition to cis-11-hexadecenoic acid.

Two Blends of Refined Rice Bran, Flaxseed, and Sesame Seed Oils Affect the Blood Lipid Profile of Chinese Adults with Borderline Hypercholesterolemia to a Similar Extent as Refined Olive Oil

Individual vegetable oils have a characteristic fatty acids (FA) composition and unique phytonutrient profiles, enabling formulation of oil blends that may have health-promoting effects. The primary objective of this study was to investigate effects of 2 oil blends made with refined rice bran, flaxseed, and sesame oils, with distinct monounsaturated to saturated FA, polyunsaturated to saturated FA, and omega-3 (n-3) to omega-6 FA ratios and different phytonutrient concentrations on blood lipid profile, compared with refined olive oil as a control. The secondary outcomes were other markers of cardiometabolic health. A parallel-design, randomized controlled trial compared consumption of 30g of allocated intervention oil per day for a period of 8 wk. The study recruited 143 borderline hypercholesterolemic (LDL cholesterol: 3.06-4.51mmol/L) Chinese volunteers between 50 and 70 y old and with a BMI (kg/m2) ≤27.5. All outcomes were measured every 2 wk, and the time × treatment interactions and the main effects of treatment and time were analyzed using an intention-to-treat approach. Compared with baseline (week 0), there were significant reductions during the post-intervention time points in serum total cholesterol (-3.47%; P<0.0001), LDL cholesterol (-4.16%; P<0.0001), triglycerides (-10.3%; P<0.0001), apoB (-3.93%; P<0.0001), total to HDL-cholesterol (-3.44%; P<0.0001) and apoB to apoA1 (-3.99%; P<0.0001) ratios, systolic and diastolic blood pressures (-3.32% and -3.16%, respectively; both P<0.0001), and serum glucose (-1.51%; P<0.05) and a small but significant increase in body weight (+0.7%; P<0.001) for all 3 intervention oils but no effects of intervention on HDL-cholesterol or apoA1 concentration. No significant effects of treatment or time × treatment interactions were found. Using blended vegetable oils that are extensively consumed in Asia, this study found that specific oil blends can improve blood lipid profile and other cardiometabolic parameters, to a similar extent as refined olive oil, in Chinese adults with borderline hypercholesterolemia. Mineral and fatty acid composition of milk from native Jutland and Danish Red 1970 cattle breeds

ABSTRACTMineral and fatty acid composition in milk from two minor native dairy breeds in Denmark; Danish Red 1970 (RDM-1970) and Jutland cows were analyzed. Results showed that for most compositional traits breed only played a minor role, whereas management and feeding played a significant role. Biodynamic management and very low milk yield resulted in a very characteristic fatty acid composition and a strongly elevated n-3/n-6 ratio (ratio 0.9 compared to 0.3). For milk minerals, RDM-1970 had significantly higher contents of P, Mg, Mn, whereas milk from Jutland cows had significantly higher K content.

Edible Oil Production From Microalgae: A Review

Microalgae are considered an interesting candidate for the sustainable production of edible oils. Because microalgae contain abundant polyunsaturated fatty acids (PUFA) and have high photosynthetic rates and high oil productivity, they are superior to terrestrial oleaginous crops for producing edible oils. Microalgae are cultivated in aquatic environments, and thus, oil extraction methods for microalgae must consider the influence of water. This paper provides an overview of the suitability of microalgal lipid compositions and proposes a selection of species for edible oil production. The numerous technologies currently available for pretreatment, wet extraction and the refining of edible oils from microalgae are collated and presented, with a primary focus on the key parameters influencing process performance. Furthermore, the paper presents a proposed solution for a microalgae integrated system for edible oil production, which is a possible future direction to develop an economical and environmentally friendly process for the resource utilization of microalgal biomass.Practical Application: Microalgae might be the most promising source of edible oils, with potential applications in food industrialization, due to its rich oil content and its characteristic fatty acid composition. The review provides a selection criterion of microalgal species to help researchers exploit microalgae to produce superior edible oils for human consumption. Various process parameters and wet extraction methods are discussed that may be conducive to improving process performance and achieving high oil recovery yields. By considering the maximum utilization of microalgal biomass, an integrated system in this review would assist in economical production during further studies.Microalgae are the most promising candidates for edible oil production. Cell disruption, extraction, and refining are important processes for microalgal oils and on this basis, wet extraction and resource utilization can further promote the economic feasibility of microalgal process.

Decreased 16:0/20:4-phosphatidylinositol level in the post-mortem prefrontal cortex of elderly patients with schizophrenia

The etiology of schizophrenia includes phospholipid abnormalities. Phospholipids are bioactive substances essential for brain function. To analyze differences in the quantity and types of phospholipids present in the brain tissue of patients with schizophrenia, we performed a global analysis of phospholipids in multiple brain samples using liquid chromatography electrospray ionization mass/mass spectrometry (LC-ESI/MS/MS) and imaging mass spectrometry (IMS). We found significantly decreased 16:0/20:4-phosphatidylinositol (PI) levels in the prefrontal cortex (PFC) in the brains from patients with schizophrenia in the LC-ESI/MS/MS, and that the 16:0/20:4-PI in grey matter was most prominently diminished according to the IMS experiments. Previous reports investigating PI pathology of schizophrenia did not identify differences in the sn-1 and sn-2 fatty acyl chains. This study is the first to clear the fatty acid composition of PI in brains from patients with schizophrenia. Alteration in the characteristic fatty acid composition of PI may also affect neuronal function, and could play a role in the etiology of schizophrenia. Although further studies are necessary to understand the role of reduced 16:0/20:4-PI levels within the prefrontal cortex in the etiology of schizophrenia, our results provide insight into the development of a novel therapy for the clinical treatment of schizophrenia.

Dietary DHA supplementation causes selective changes in phospholipids from different brain regions in both wild type mice and the Tg2576 mouse model of Alzheimer's disease

Alzheimer's disease (AD) is of major concern in ageing populations and we have used the Tg2576 mouse model to understand connections between brain lipids and amyloid pathology. Because dietary docosahexaenoic acid (DHA) has been identified as beneficial, we compared mice fed with a DHA-supplemented diet to those on a nutritionally-sufficient diet.Major phospholipids from cortex, hippocampus and cerebellum were separated and analysed. Each phosphoglyceride had a characteristic fatty acid composition which was similar in cortex and hippocampus but different in the cerebellum. The biggest changes on DHA-supplementation were within ethanolamine phospholipids which, together with phosphatidylserine, had the highest proportions of DHA. Reciprocal alterations in DHA and arachidonate were found. The main diet-induced alterations were found in ethanolamine phospholipids, (and included their ether derivatives), as were the changes observed due to genotype. Tg mice appeared more sensitive to diet with generally lower DHA percentages when on the standard diet and higher relative proportions of DHA when the diet was supplemented. All four major phosphoglycerides analysed showed age-dependent decreases in polyunsaturated fatty acid contents.These data provide, for the first time, a detailed evaluation of phospholipids in different brain areas previously shown to be relevant to behaviour in the Tg2576 mouse model for AD. The lipid changes observed with genotype are consistent with the subtle alterations found in AD patients, especially for the ethanolamine phospholipid molecular species. They also emphasise the contrasting changes in fatty acid content induced by DHA supplementation within individual phospholipid classes.

Novel, Rapid Identification, and Quantification of Adulterants in Extra Virgin Olive Oil Using Near-Infrared Spectroscopy and Chemometrics.

A new, rapid Fourier transform near infrared (FT-NIR) spectroscopic procedure is described to screen for the authenticity of extra virgin olive oils (EVOO) and to determine the kind and amount of an adulterant in EVOO. To screen EVOO, a partial least squares (PLS1) calibration model was developed to estimate a newly created FT-NIR index based mainly on the relative intensities of two unique carbonyl overtone absorptions in the FT-NIR spectra of EVOO and other mixtures attributed to volatile (5280cm(-1)) and non-volatile (5180cm(-1)) components. Spectra were also used to predict the fatty acid (FA) composition of EVOO or samples spiked with an adulterant using previously developed PLS1 calibration models. Some adulterated mixtures could be identified provided the FA profile was sufficiently different from those of EVOO. To identify the type and determine the quantity of an adulterant, gravimetric mixtures were prepared by spiking EVOO with different concentrations of each adulterant. Based on FT-NIR spectra, four PLS1 calibration models were developed for four specific groups of adulterants, each with a characteristic FA composition. Using these different PLS1 calibration models for prediction, plots of predicted vs. gravimetric concentrations of an adulterant in EVOO yielded linear regression functions with four unique sets of slopes, one for each group of adulterants. Four corresponding slope rules were defined that allowed for the determination of the nature and concentration of an adulterant in EVOO products by applying these four calibration models. The standard addition technique was used for confirmation.

Fatty acid composition as biomarkers of freshwater microalgae: analysis of 37 strains of microalgae in 22 genera and in seven classes

The fatty acid (FA) composition of algae is an important determinant of their food quality for consumers, and FAs can also be used as biomarkers for biochemical and energetic pathways in food webs. FA analyses of 7 freshwater algal classes and 37 strains showed clear sim- ilarity within classes and strong differences amongst classes. Class was a dominant factor (66.4%) explaining variation in FA signatures of microalgae. The 7 algal classes comprised 4 separate groups according to their FA profiles: (1) Chlorophyceae and Trebouxiophyceae, (2) Bacillario- phyceae, (3) Cryptophyceae, Chrysophyceae, and Raphidophyceae, and (4) Euglenophyceae. Each group had a characteristic FA composition, although the proportional abundance of individ- ual FAs also differed between species and with environmental conditions. FAs found to be partic- ularly representative for each group (i.e. diagnostic biomarkers) were as follows: 16:4ω3 and 16:3ω3 for Chlorophyceae and Trebouxiophyceae; 16:2ω7, 16:2ω4, 16:3ω4, 16:4ω1, and 18:4ω4 for Bacillariophyceae; 22:5ω6 and 18:4ω3 for Cryptophyceae and Chrysophyceae (Synurales), 16:3ω1 for Chrysophyceae (Ochromonadales), 16:2ω4, 16:3ω4, 16:3ω1, and 20:3ω3 for Raphidophyceae; and 15:4ω2, 20:4ω3, 20:2ω6, 20:3ω6, and 22:4ω6 for Euglenophyceae. FAs thus offer a powerful tool to track different consumer diets in a lacustrine food web. Based on the 20:5ω3 (eicosapen- taenoic acid) and 22:6ω3 (docosahexaenoic acid) content among the investigated freshwater algal classes, Chlorophyceae, Trebouxiophyceae, and Chrysophyceae are of intermediate food quality for zooplankton, and Cryptophyceae, Bacillariophyceae, Euglenophyceae, and Raphidophyceae should be excellent resources for zooplankton.

Changes in tissue and mitochondrial membrane composition during rapid growth, maturation and aging in rainbow trout, Oncorhynchus mykiss

Membrane compositions, particularly of mitochondria, could be critical factors in the mechanisms of growth and aging processes, especially during phases of high oxidative stress that result in molecular damage. In the present study, liver and mitochondrial membrane phospholipid (PL) compositions were analyzed in rainbow trout during its four first years of life, a period characterized by rapid growth and high oxidative stress. Specifically, farmed fish of three ages (1-, 2- and 4-years) were studied, and PL compositions of whole liver and liver mitochondria, and fatty acid compositions of individual PL classes were determined. Liver mitochondrial membranes showed a PL composition different to that of the whole tissue suggesting adaptation of cell and subcellular membranes to specific functions. Individual PL had characteristic fatty acid compositions that were similar in whole liver and mitochondrial membranes. Whole liver and mitochondria showed increased lipid peroxidation with age along with changes in membrane PL fatty acid compositions. Most PL classes showed similar changes in fatty acid composition among the age groups, with reduced proportions of docosahexaenoic acid (DHA) and, generally, concomitantly increased levels of monounsaturated fatty acids, which together resulted in reduced peroxidation index (PIn). However, total polyunsaturated fatty acid (PUFA) content did not change significantly with age due to increased eicosapentaenoic acid, docosapentaenoic acid and, in most PL, increased n-6 PUFA. These results suggest there may be oxidation of PL DHA with compensatory mechanisms to maintain membrane fluidity and function. However, modification of fatty acid composition of specific PLs, such as cardiolipin, could affect the electron transport chain efficiency and propagate the oxidative reaction throughout the cell. In addition, both the content and fatty acid composition of sphingomyelin, which has been suggested as a possible mediator of cell dysfunction and apoptosis, changed with age differently to the other PL classes. Moreover, these changes showed different trends between mitochondria and whole liver. These data suggest there is marked oxidative stress associated with rapid growth and maturation in rainbow trout. Changes observed in membrane lipids point to their possible participation in the processes involved in this species response to oxidative stress and damage accumulation rate.

The influence of storage conditions on flavour changes in human milk

The effects of freeze-storage on human milk have been investigated with regard to sensory changes and modifications in the molecular composition of constituent odour-active volatiles. Sensory evaluation showed that fishy-metallic odour attributes are generated during storage, as are rancid-sweaty odour notes, which were described by panellists to be highly unpleasant. To monitor the dynamics of off-odour development in human milk, sensory evaluation in both orthonasal and retronasal mode was performed at defined time intervals of storage, with parallel evaluation of human milk samples that had been heated prior to freeze-storage. Heating was found to induce slight flavour changes, with generation of a predominantly egg-white-like note, but with no further generation of off-odour formation upon subsequent storage. This suggests that the formation of the off-odour in non-heated milk resulted from metabolic processes in the milk, although may also be due to the specific composition of human milk. In comparison, similar off-odour development was not observed in bovine milk. Molecular identification of the off-odour substances, most specifically free fatty acids and oxidation derivatives of polyunsaturated fatty acids, indicated both lipolytic and peroxidative processes of components of the native human milk. Finally, these results indicate that human milk is obviously a highly sensitive food material, specifically with regard to its characteristic fatty acid composition.

Novel Mutations of the FASN Gene and Their Effect on Fatty Acid Composition in Japanese Black Beef

Eight novel and four known mutations were detected in the coding sequence of the bovine fatty acid synthase (FASN) gene of an F2 population from Japanese Black and Limousin cattle. Two mutations, g.16024A>G and g.16039T>C, detected in exon 34, which determine amino acid substitutions of threonine (T) to alanine (A) and tryptophan (W) to arginine (R), were clearly separated in the parental breeds. The haplotypes (TW and AR) segregated in F2 individuals and had a significant effect on the fatty acid composition of backfat, intermuscular fat, and intramuscular fat. The TW haplotype was associated with increasing C18:0 and C18:1 content and the ratio of monounsaturated to saturated fatty acids, and decreasing C14:0, C14:1, C16:0, and C16:1 content. The two mutations were screened in two commercial Japanese Black half-sibling populations and similarly determined the contribution to the fatty acid composition of intramuscular fat. The frequency of the TW haplotype was markedly higher in Japanese Black (0.67) than in Holstein (0.17), Angus (0.02), and Hereford (0.07). We conclude that these mutations may contribute to the characteristic fatty acid composition of Japanese Black beef.

Comparative study of the fatty acid composition of some groups of purple nonsulfur bacteria

The fatty acid composition (FAC) of 43 strains of purple nonsulfur bacteria belonging to six genera--Rubrivivax, Rhodopseudomonas, Rhodoplanes, Blastochloris, Rhodobium, and Rhodomicrobium--was studied by capillary gas chromatography. The cultures were grown on standard medium under standard conditions. Automatic identification of the fatty acid methyl esters and statistical processing of the results were performed by the computerized Microbial Identification System (M.I.S). Significant differences between the FACs of different genera, species, and, sometimes, strains were revealed. 16S rRNA genes of some of the new isolates, primarily those having a specific FAC, were sequenced. The taxonomic status of a number of the strains in question was determined using the FAC characteristics as one of the criteria. It was shown that the FAC characteristics may be used both for affiliating the isolates to known species and for revealing new taxa.

Lipid composition of the red alga Tichocarpus crinitus exposed to different levels of photon irradiance

We examined variations in the lipid composition of the marine red alga Tichocarpus crinitus exposed to different levels of photon irradiance: 70–80% and 8–10% of the incident photosynthetically active radiation (PAR). The content of storage and structural lipids was significantly affected by the light intensity. Exposure of T. crinitus to low light conditions induced an increase in the abundance of structural components of the cell membranes, especially sulfoquinovosyldiacylglycerol, phosphatidylglycerol (PG) and phosphatidylcholine, while growth of algae at high light intensity resulted in a 1.5-folder increase in the level of storage lipids, i.e. triacylglycerols. There were no significant differences in the fatty acid composition of the total lipid pool in algae grown under different light conditions. However, the content of the most unsaturated acid, 20:5 n − 3, was slightly higher in T. crinitus under 8–10% PAR compared to those at 70–80% PAR. Each lipid class was found to have a characteristic fatty acid composition. The relative proportions of fatty acids esterifying monogalactosyldiacylglycerol (MGDG) and PG were significantly affected by irradiance conditions. Exposure of algae to low light resulted in increase in the content of 20:5 n − 3 in MGDG and in decrease in the level of this acid in PG. The concentration of trans-16:1 acid in PG increased in algae grown under high light intensity. Light conditions influenced on total lipid content, which made up 4.2 ± 0.5 and 3.4 ± 0.3 mg g −1 fresh weight in algae exposed to 8–10% PAR and 70–80% PAR, respectively. We suggest that variations in the lipid composition of T. crinitus exposed to different levels of light intensity may be a response of alga to light conditions and it can be considered as one of the mechanisms of adaptation of T. crinitus to varying incident light intensity.

Effect of solar irradiance on lipids of the green alga Ulva fenestrata Postels et Ruprecht

We examined variations in lipid composition of the marine green alga Ulva fenestrata grown under different irradiance conditions (80% of incident photosynthetically active radiation [PAR] and 24% PAR). The ratio of storage lipids (triacylglycerol) to major structural chloroplast lipids (glycolipids and phosphatidylglycerol) was influenced considerably by the irradiance. Triacylglycerol predominated in U. fenestrata exposed to 80% PAR (58.7±1.7% of total lipids). In contrast, when the alga was grown at 24% PAR, the amount of monogalactosyldiacylglycerol, sulfoquinovosyldiacylglycerol and phosphatidylglycerol increased two to three and a half times compared to algae cultivated at 80% PAR. The content of digalactosyldiacylglycerol and betaine lipid in U. fenestrata showed little dependence on light intensity. Each lipid class had a characteristic fatty acid composition. However, the relative proportions of fatty acids present in triacylglycerol, monogalactosyldiacylglycerol and sulfoquinovosyldiacylglycerol did not depend on irradiance conditions. The variation in fatty acid composition of digalactosyldiacylglycerol and phosphatidylglycerol and changes in the amounts of lipids were responsible for the differences in total fatty acid composition of U. fenestrata among light intensities. The most sensitive to the change in quantity of incident light were the 16:4n-3 and 16:0 acids. The highest level of 16:4n-3 acid was detected in algae grown under 24% PAR, whereas 16:0 acid was highest at 80% PAR. However, the total lipid content in the algae was relatively constant at different light intensities (5.0±1.0 mg/g fresh weight at 80% PAR and 5.2±1.1 mg/g at 24% PAR). These results suggest that changes in lipid composition of U. fenestrata can be considered as a mechanism of adaptation and survival of thalli subjected to variations in solar irradiance.

Lipid and Fatty Acid Composition during Vegetative and Resting Stages of the Marine Diatom Chaetoceros salsugineus

The lipid and fatty acid compositions of the marine diatom Chaetoceros salsugineus were investigated during the life cycle, including the vegetative and resting stages. The relative proportion of the lipid classes and their fatty acid composition varied during the life cycle. The amount of polar lipids, structural components of the cell membranes, increased rapidly in the period of the resting cell formation. Individual lipid classes were found to have a characteristic fatty acid composition. Changes in the proportion of polar lipids contributed to a difference in the total fatty acids. In polar lipids, polyunsaturated fatty acids, predominantly 16:3(n-4) and 20:5(n-3), composed up to 59% of the total fatty acids, peaking during resting cell formation. The results suggest that in resting cells the structural components of photosynthetic and cellular membranes accumulate, thus ensuring the immediate growth and development of cells upon the return of favourable environmental conditions.

Total lipid and fatty acid composition of eight strains of marine diatoms

Fatty acid composition and total lipid content of 8 strains of marine diatoms (Nitzschia frustrula, Nitzschia closterium, Nitzschia incerta, Navicula pelliculosa, Phaeodactylum tricornutum, Synedra fragilaroides) were examined. The microalgae were grown under defined conditions and harvested at the late exponential phase. The major fatty acids in most strains were 14∶0 (1.0%–6.3%), 16∶0 (13.5–26.4%), 16∶1n−7 (21.1%–46.3%) and 20∶5n−3 (6.5%–19.5%). The polyunsaturated fatty acids 16∶2n−4, 16∶3n−4, 16∶4n−1 and 20∶4n−6 also comprised a significant proportion of the total fatty acids in some strains. The characteristic fatty acid composition of diatoms is readily distinguishable from those of other microalgal groups. Significant concentration of the polyunsaturated fatty acid 20∶5n−3 (eicosapentaenoic acid) was present in each strain, with the highest proportion in B222 (19.5%).

Seasonal variation in lipid and fatty acid composition of ice algae from the Barents Sea

The fatty acid compositions of neutral lipid, glycolipid and phospholipid fractions from ice algae sampled from the Barents Sea in spring and autumn were examined for seasonal differences. The ice-algal assemblages were dominated by diatoms. In spring, Nitzschia frigida was the most common species whereas resting stages of Thalassiosira bioculata and Actinocyclus cf curvatulus predominated in autumn. With the exception of one spring sample, neutral lipids predominated over glycolipids and phospholipids in all algal samples. The lipid fractions displayed characteristic fatty acid compositions. In the spring samples the major fatty acids of the neutral lipid fraction were 16:0, 16:1(n-7) and 20:5(n-3) whilst the glycolipid fraction was characterised by higher levels of 20:5(n-3) and C16 polyunsaturated fatty acids, particularly 16:4(n-1). Phospholipids contained higher levels of 22:6(n-3) than the other two lipid fractions although 20:5(n-3) was still the major polyunsaturated fatty acid. In the autumn samples, the neutral lipid fraction contained higher proportions of saturated fatty acids and 16:1(n-7) than the two polar lipid fractions and 22:6(n-3) was most abundant in phospholipids. As with the spring samples, 20:5(n-3) was the major polyunsaturated fatty acid in all lipid fractions of the autumn algae. Overall, the fatty acid compositions of the lipid fractions from spring and autumn algal samples were similar and are consistent with diatoms being the predominant group in the ice algae studied. The high level of neutral lipids observed in both spring and autumn samples suggests that the production of neutral lipids is characteristic of ice algae regardless of season. Nevertheless, some species-specific differences in lipid production may exist since the neutral lipid content of autumn samples containing mainly A. curvatulus was substantially higher than those in which T. bioculata predominated.
