Protein kinetics in human endotoxaemia and their temporal relation to metabolic, endocrine and proinflammatory cytokine responses.

Sepsis is associated with profound alterations in protein metabolism. The unpredictable time course of sepsis and the multiplicity of confounding factors prevent studies of temporal relations between the onset of endocrine and proinflammatory cytokine responses and the onset of protein catabolism. This study aimed to determine the time course of whole-body protein catabolism, and relate it to the endocrine, metabolic and cytokine responses in a human endotoxaemia model of early sepsis. Six healthy male volunteers were studied twice in random order, before and for 600 min after administration of either an intravenous bolus of Escherichia coli lipopolysaccharide (LPS) or sterile saline. Whole-body protein synthesis, breakdown and net protein breakdown were measured by amino acid tracer infusion, and related to changes in plasma levels of growth hormone, glucagon, cortisol, insulin-like growth factor (IGF) 1, tumour necrosis factor (TNF) α and interleukin (IL) 6. Protein synthesis, breakdown and net protein breakdown increased and peaked 120 min after LPS administration (P < 0·001), the alterations persisting for up to 480 min. These peaks coincided with peaks in plasma growth hormone, TNF-α and IL-6 concentrations (P = 0·049, P < 0·001 and P < 0·001 for LPS versus saline), whereas plasma cortisol concentration peaked later. No alterations in plasma insulin or glucagon concentrations, or in the IGF axis were observed during the period of abnormalities of protein metabolism. LPS administration induced an early protein catabolic response in young men and this coincided with changes in plasma growth hormone, TNF-α and IL-6 concentrations, rather than changes in cortisol, glucagon, insulin or the IGF axis. Surgical relevance Sepsis in surgical patients is common and remains associated with substantial mortality. Although sepsis is a heterogeneous condition and its pathophysiology therefore difficult to study, a universal and profound clinical problem is protein catabolism not responsive to nutritional support. Human experimental endotoxaemia is a promising model of clinical sepsis that can be used to elucidate underlying pathophysiology and explore novel therapeutic approaches. This study demonstrates that human experimental endotoxaemia replicates the changes in whole-body protein turnover seen in clinical sepsis. Frequent measurements allowed identification of tumour necrosis factor (TNF) α, interleukin (IL) 6 and growth hormone as putative mediators. Human experimental endotoxaemia is a valid model for further study of mechanisms and putative therapies of catabolism associated with sepsis. In particular, effects of TNF-α and IL-6 blockade should be evaluated.

Postprandial changes in plasma growth hormone, insulin, insulin-like growth factor (IGF)-I, and IGF-binding proteins in coho salmon fasted for varying periods

We examined postprandial changes in circulating growth hormone (GH), insulin, insulin-like growth factor (IGF)-I, and IGF-binding proteins (IGFBPs) in yearling coho salmon under different feeding regimes. Fish were initially fasted for 1 day, 1 wk, or 3 wk. Fasted fish were then fed, and blood was collected at 4-h intervals over 26 h. After the various periods of fasting, basal levels of insulin were relatively constant, whereas those of IGF-I, IGFBPs and GH changed in proportion to the duration of the fast. A single meal caused a rapid, large increase in the circulating insulin levels, but the degree of the increase was influenced by the fasting period. IGF-I showed a moderate increase 2 h after the meal but only in the regularly fed fish. Plasma levels of 41-kDa IGFBP were increased in all groups within 6 h after the single meal. The fasting period did not influence the response of 41-kDa IGFBP to the meal. IGFBP-1 and GH decreased after the meal to the same extent among groups regardless of the fasting period. The present study shows that insulin and IGF-I respond differently to long (weeks)- and short (hours)-term nutritional changes in salmon; insulin maintains its basal level but changes acutely in response to food intake, whereas IGF-I adjusts its basal levels to the long-term nutritional status and is less responsive to acute nutritional input. IGFBPs maintain their sensitivity to food intake, even after prolonged fasting, suggesting their critical role in the nutritional regulation of salmon growth.

Effect of diet and ration on the relationship between plasma GH and IGF-1 concentrations in Arctic charr, Salvelinus alpinus (L.)

The purpose of the study was to investigate whether dietary ration or diet composition influence the relationship between plasma growth hormone (GH) and insulin-like growth factor-1 (IGF-1) in Arctic charr (Salvelinus alpinus L.). The pattern of changes in plasma GH and IGF-1 concentrations was examined in fish fed at different ration levels (0%, 0.35% and 0.70% BW day−1) for 5 weeks, and in fish fed diets containing different lipid:crude protein (LCP) ratios. Ration level significantly affected plasma GH and IGF-1 concentrations; at 5 weeks the levels of both hormones in the food-deprived group were significantly lower than in fish fed the 0.70% BW day−1 ration. Also, plasma IGF-1 levels in fish of each ration treatment group were significantly correlated with individual final body weight; no such correlation was found for GH. To examine the effects of dietary LCP ratios, fish were fed for up to 18 weeks, with one of four formulated diets that had LCP ratios (dry matter basis) of 0.35 (Diet 1), 0.43 (Diet 2), 0.51 (Diet 3) or 0.59 (Diet 4), or a commercial diet (Diet 5) which had an LCP ratio of 0.38. Statistical differences in plasma GH and IGF-1 concentrations were found only after 18 weeks. Growth hormone was significantly lower in fish fed Diets 1 and 2 compared with Diets 3 and 5, and IGF-1 was significantly lower in fish fed Diet 1 compared with Diets 2 and 5. Significant correlations between plasma GH and IGF-1 concentrations were found only for fish fed Diets 1 and 5, suggesting that the influence of diet composition on the relationship between GH and IGF-1 varies with the dietary LCP ratio in this species. The decline in plasma IGF-1 concentrations during food deprivation is similar to that described in other species; however, the unexpected decrease in plasma GH during food deprivation in this study may represent a species-specific response.

Establishment of a time-resolved fluoroimmunoassay for measuring plasma insulin-like growth factor I (IGF-I) in fish: effect of fasting on plasma concentrations and tissue mRNA expression of IGF-I and growth hormone (GH) in channel catfish ( Ictalurus punctatus)

A time-resolved fluoroimmunoassay (TR-FIA) was established and validated that allows for the determination of plasma concentrations of insulin-like growth factor I (IGF-I) in three domestically cultured fishes: channel catfish ( Ictalurus punctatus), hybrid striped bass ( Morone chrysops × M. saxatilis), and rainbow trout ( Oncorhynchus mykiss). Sensitivity of the assay was 0.20 ng/ml. Intra- and inter-assay coefficients of variation (CV) were <7 and <12%, respectively. Serial dilutions of plasma from each species were parallel to the standard curve. Recovery of IGF-I from spiked plasma samples was >90% for all three species of fishes. The IGF-I TR-FIA was biologically validated via its use to determine the effect of fasting on circulating IGF-I levels in channel catfish. Fasting-induced changes in plasma growth hormone (GH), hepatic IGF-I mRNA expression, and pituitary GH mRNA expression were also determined. Fasted channel catfish lost 5.6 and 15.6% body mass after 2 and 4 weeks of fasting, respectively. Plasma IGF-I concentrations were depressed ( P < 0.05) relative to fed controls following 2 and 4 weeks of fasting. Plasma GH concentrations were not different ( P > 0.05) in fasted fish after 2 weeks, but significantly increased ( P < 0.05) by 4 weeks of fasting. Hepatic IGF-I mRNA expression after 2 and 4 weeks of fasting was reduced ( P < 0.05) relative to fed controls. Pituitary GH mRNA expression was similar ( P > 0.05) between 2-week-fasted catfish and fed controls, but was increased ( P < 0.05) in 4-week-fasted catfish. The IGF-I TR-FIA was sensitive, accurate, and precise for all three species of fishes, and provided a low-cost, and non-radioisotopic method for quantifying plasma IGF-I levels in fed and fasted channel catfish.

Changes in Plasma Growth Hormone (GH) and Secretion Patterns of GH and Luteinizing Hormone in Buffaloes (Bubalus bubalis) During Growth

To assess the changes of plasma growth hormone (GH) and secretion patterns of GH and luteinizing hormone (LH) during growth in buffaloes, six growing female Murrah buffalo calves (mean age 6 ± 0.9 months and body weight 66 ± 6 kg) were selected. Plasma samples were collected twice a week for 52 weeks for GH and LH assay. To examine for pulsatile secretion samples were collected at 15 minutes interval for 9 hr at weeks 6 and 42 for GH and LH measurements. Plasma progesterone was also estimated in twice‐a‐week samples to assess whether any of the buffalo had begun ovarian cyclicity. The body weight of all animals was recorded at weekly interval. Plasma GH concentration decreased (P < 0.01) only up to week 29 and showed an increasing trend (P < 0.01) thereafter. The ratio of plasma GH to body weight declined (P < 0.01) throughout the entire experimental period. Plasma GH showed a declining trend only up to when the animals attained 155 kg body weight and thereafter showed an increasing trend (P < 0.01). Plasma GH revealed distinct pulsatile patterns of release, with a mean of 6 and 5 pulses in the 6‐week and 42‐week samples, respectively. The plasma LH concentrations around the 42‐week time period were significantly higher (P < 0.01) than at the 6‐week time period, and they exhibited pulsatility. No animal reached puberty until the end of the experiment. In summary, plasma GH levels have a definite pattern of change during growth and patterns of secretion of plasma GH and LH also have a relation with body weight in this species of animal.

Influences of photoperiod and alternate days of feeding on plasma growth hormone and thyroid hormone levels in juvenile rainbow trout

The diurnal patterns of plasma growth hormone (GH), thyroid hormone and cortisol concentrations in rainbow trout Oncorhynchus mykiss held under three photoperiod (L : D) regimes (6 : 18, 12 : 12 and 18 : 6), and fed either daily (DF) or on alternate days (ADF) with 2·0% body mass per day of a commercial trout diet were determined. The ADF groups had reduced total mass gain and specific growth rates compared with DF fish, but photoperiod had no affect on growth for either of the feeding regime groups. In the ADF groups, the mean 24 h plasma thyroxine (T4) and triiodothyronine (T3) concentrations were significantly lower, both on days of feeding and days of fasting, than in DF fish held under all three photoperiod regimes, but for GH, only the 18L : 6D DF group was higher than the comparable ADF groups. There were no significant differences in mean 24 h plasma cortisol concentrations of DF and ADF groups. Diurnal patterns of plasma GH, cortisol, T4 and T3 were found in DF fish held under all three photoperiod regimes. Increases in plasma cortisol changes were associated with the onset of the light phase; elevations in plasma GH and T4 concentrations were more closely associated with clock time, regardless of photoperiod; increases in plasma T3 concentrations were strongly associated with time of feeding. In ADF groups, these diurnal changes in plasma GH, T4 and T3 concentrations were suppressed for both the fed and fast days, and plasma cortisol concentrations were suppressed on the fasting day. The observations are discussed in terms of the proposed anabolic, catabolic and growth regulating roles of these hormones in different growth and metabolic modifying situations in teleosts.

Influence of physical activity on plasma insulin-like growth factor-1 and insulin-like growth factor binding proteins in healthy older women

It was examined whether physical activity could alter the bioavailability of insulin-like growth factor-1 (IGF-1) which is dependent upon plasma IGF-1 and insulin-like growth factor binding protein (IGFBPs) levels. The potential role that growth hormone (GH) and insulin play in this process was also examined. Seven healthy 62–69-year-old women performed four bouts of physical activity on separate occasions at either a low (L; heart rate=100 bpm) or moderate intensity (M; heart rate=120 bpm) for either 25 (S) or 50 (L) min (LS, low intensity/short duration; LL, low intensity/long duration; MS, moderate intensity/short duration; ML, moderate intensity/long duration). GH levels were elevated immediately following the physical activity from 1.3 to 2.6-fold ( P<0.05) whereas IGF-1 levels were not affected by any activity condition. Plasma insulin levels decreased about 35% under all activity conditions ( P<0.05). Plasma levels of IGFBP-1 (BP-1) were decreased immediately following the ML (−47%; P<0.05) and the LL (−21%) activity bouts and remained lower than initial values 1 h after these activity bouts (−25 and 34%, respectively, P<0.05). The ML exercise bout resulted in significant ( P<0.05) increases in IGFBP-2 (BP-2) and IGFBP-3 (BP-3) immediately following activity (+31, and +30%, respectively) and these binding proteins remained elevated following the activity (+28, and +48%, respectively). No relationship was found between any changes in plasma GH or insulin, and changes in plasma IGFBPs. Thus, moderate intensity physical activity of long duration may modulate the bioavailability of IGF-1 in the elderly via alterations in BP-1, -2 and -3. However, changes in circulating levels of GH, insulin or IGF-1 do not appear to be regulating IGF-1 bioavailabilty in response to physical activity.

Effects of N-methyl-D,L-aspartate (NMA) on growth hormone and thyroid hormone levels in steroid-primed immature rainbow trout (Oncorhynchus mykiss)

In this study we investigate whether the stimulatory action of the glutamate agonist, N-methyl-D,L-aspartate (NMA) on growth hormone (GH) levels in rainbow trout (Oncorhynchus mykiss) is steroid hormone-dependent. NMA was administered to sexually immature juvenile rainbow trout that had been primed with testosterone (T), 17β-estradiol (E2) or 5α-dihydrotestosterone and changes in plasma growth hormone (GH) concentrations were measured. NMA had no significant effect on the plasma GH concentrations of non-steroid hormone-primed trout (controls), although in both T- and E2-primed fish plasma GH levels were elevated 6 hours following NMA administration. The response had disappeared by 24 hours in the T-primed group, but was still evident in the E2-primed fish. In comparison, in the 5αDHT-primed fish, NMA significantly depressed plasma GH levels 6 hours post-injection (P < 0.05), but this effect had disappeared 24 hours after NMA administration. NMA had no effect on plasma T4 levels in any treatment group, and although plasma T3 levels were significantly (P < 0.05) depressed in T-primed fish 6 hours post-injection, this response was not seen in any other group. These results suggest that NMA has no effect on plasma thyroid hormone levels but it does have a steroid hormone-dependent action on plasma GH levels in rainbow trout. J. Exp. Effect of Gonadal Steroid Hormones on Plasma Growth Hormone Concentrations in Sexually Immature Rainbow Trout,Oncorhynchus mykiss

In the present study we investigated the effects of gonadal steroid hormones on plasma growth hormone (GH), triiodothyronine (T3), and thyroxine (T4) levels in steroid-primed sexually immature rainbow trout,Oncorhynchus mykiss,in order to determine whether changes in plasma GH and thyroid hormone levels during sexual maturation are a result of elevated gonadal steroid levels or are an altered condition associated with reproduction (as proposed by Sumpteret al.,1991b). We found no significant correlation between plasma GH and condition factor in either of two cohorts of rainbow trout (1+ and 2+ year old fish) examined. To evaluate the effects of gonadal steroids on plasma GH and thyroid hormone levels, fed and fasted (4 weeks fasted) fish were given a slow-release implant of testosterone (T), 17β-estradiol (E2), 17α-methyltestosterone (17αMT), or 5α-dihydrotestosterone (5αDHT) dissolved in coconut oil, or coconut oil alone (control). Blood samples were taken after 2 weeks and analyzed for GH, T3, and T4. Neither 17αMT nor 5αDHT had any effect on plasma GH levels in fed or fasted fish. However, plasma GH levels were elevated after E2treatment in both fed and fasted (P≤0.01) fish, and after T treatment in fasted but not fed fish (P<0.01). The effects of gonadal steroid hormones on thyroid hormone levels were less clear. T3levels were depressed in fed E2-primed fish (P<0.05) and elevated in fasted T- and 17αMT-primed fish (P<0.05). T4levels were depressed in T- and 5αDHT-primed fed fish (P<0.01) and elevated in fasted E2-primed fish (P<0.01). These elevated GH levels associated with sexual maturation may be more clearly linked to elevated gonadal steroid levels rather than to changes in nutritional status. The effects of gonadal steroids on thyroid hormone levels are less clear.

Do changes in growth hormone levels correlate with IGF-I levels in patients undergoing IVF-ET?

It has been suggested that adjunctive growth hormone (GH) therapy improves ovarian response and in vitro fertilization (IVF) outcome in specific groups of patients. The correlation between insulin-like growth factor (IGF) and GH is well established. The aim of this study was to determine whether changes in plasma GH correlate with IGF blood levels in patients during IVF treatment. Thirty-six women undergoing IVF and embryo transfer (ET) were examined. Ovarian stimulation was carried out by gonadotropin-releasing hormone agonists (GnRHa) and gonadotropins. Blood was drawn at the early and late follicular phase, on the day of human chorionic gonadotropin (hCG) injection and at the mid-and the late luteal phases. The samples were assayed for IGF-I, IGF-II, IGF-binding protein-3 (IGF BP-3), GH and estradiol. According to the IGF-I and GH plasma levels, patients were divided into three major groups: Group I consisted of patients in whom peak levels of GH reached more than 4 ng/ml and IGF-I decreased significantly. In this group, estradiol levels were 1863 ± 149pg/ml. Group II consisted of patients in whom peak blood GH levels did not exceed 2.5 ng/ml and the IGF-I level remained unchanged. In this group estradiol levels were 630 ± 57pg/ml. Group III consisted of patients in whom blood GH levels were low and remained unchanged while estradiol levels were 1600 ± 420 pg/ml. In this group no significant increase in IGF levels were observed. There was no significant change in the levels of either IGF-II or IGF BP-3 in any of the groups. We can conclude that (1) there is a negative correlation between GH and IGF-I plasma levels in patients undergoing controlled ovarian hyperstimulation (COH)-IVF, when levels of estradiol and GH are elevated; (2) plasma levels of IGF-I under ovarian hyperstimulation are probably regulated by a multifactorial system; and (3) no correlation was found between the plasma levels of IGF-I and those of IGF-II and IGF BP-3 in all patient groups.

Increased galanin and neuropeptide-Y immunoreactivity within the hypothalamus of ovariectomised ewes following a prolonged period of reduced body weight is associated with changes in plasma growth hormone but not gonadotropin levels.

Manipulation of diet is known to affect the secretion of the gonadotropins and growth hormone (GH). The former are under the direct regulation of hypothalamic gonadotropin-releasing hormone (GnRH) and the latter is under the dual control of GH-releasing hormone (GHRH) and somatostatin (SRIH). At the level of the hypothalamus, both galanin (GAL) and neuropeptide Y (NPY) are thought to regulate the secretion of the above releasing and inhibiting factors. Both peptides are also potent orexigenic agents. We have studied ovariectomised ewes that were either well-fed (HIGHs) or underfed (LOWs) and used immunocytochemistry and image analysis to measure the levels of GAL and NPY in hypothalamic nuclei in which GnRH, GHRH and SRIH are found and which are also involved in the regulation of appetite and feeding. The sheep were given a normal diet or a restricted diet for 15 months. Four pairs of ewes were then blood-sampled to measure GH, luteinising hormone (LH), and follicle-stimulating hormone (FSH) and then killed for recovery of the brains. After perfusion, cryostat sections were cut through the entire hypothalamus, mounted, and stained fro NPY or GAL. All treatments and analyses were performed in pairs. The number of immunoreactive cells, density of terminals and total immunoreactivity (IR) were quantified by image analysis by sampling 6-16 subareas (depending on region) on sections through the pre-optic area (POA), paraventricular nucleus (PVN), arcuate nucleus (ARC) and median eminence (ME). Mean (+/- SEM) live weight of the LOWs was significantly (p < 0.0001) lower than that of the HIGHs (37.6 +/- 0.6 kg vs. 60.6 +/- 0.5 kg). There was no difference in the plasma levels of LH and FSH but the area under the GH curve (ng/ml/h) was significantly (p < 0.0001) greater in the LOWs (320 +/- 40.9 vs. 67.3 +/- 16.1). There was an increased number of cells staining for NPY but not GAL in the ARC/ME of the LOWs. Nevertheless, the oveall level of immunostaining for both peptides was increased in the LOWs. GAL IR was restricted to the mediobasal hypothalamus. In the LOWs, the density of NPY terminal fields in each area of the ARC was significantly (p < 0.05) increased. Food restriction also increased the density of NPY terminals in the POA and PVN (p < 0.025) but not in the ME. These data indicate that a dietary manipulation which affects GH secretion but not the gonadotropins may be mediated by NPY and GAL neuronal systems in specific brain regions within the hypothalamus.

Inverse control of growth hormone and prolactin secretion in clonidine-stimulated dairy cattle.

Clonidine is a specific alpha-2-adrenoreceptor agonist that stimulates growth hormone (GH) release in animals and humans. This drug was used to study the GH and prolactin (PRL) secretory response in dairy cows and heifers. An i.v. infusion of 10 micrograms/kg body weight induced GH release to a peak concentration after 30-60 min, while 2 micrograms/kg had no effect on GH secretory patterns. Plasma PRL decreased significantly (P < 0.01) starting 15-60 min after both doses of clonidine, this effect lasting up to 6 h. Clonidine significantly lowered plasma insulin (P < 0.01) and raised plasma glucose (P < 0.01). The changes in plasma GH, PRL, insulin and glucose differed significantly between doses, the 10 micrograms/kg dose being more effective (P < 0.01). The results of our investigation in dairy cattle provide evidence of (i) an increase in GH release after 10 micrograms/kg clonidine; (ii) a concomitant decrease in PRL secretion, hence GH and PRL secretion in cattle appear inversely controlled; (iii) a significant difference between the effects of the 2 and 10 micrograms/kg doses and (iv) no relationship between the changes in plasma GH and PRL after clonidine and plasma hormone levels before treatment.

Seasonal Changes in Plasma Growth Hormone and Prolactin Concentrations of the Frog Rana esculenta

Plasma growth hormone (G/D, prolactin (PRL), and vitellogenin (VTG) concentrations were determined during the annual reproductive cycle of the frog Rana esculenta. Plasma GH and PRL were measured using a RIA that employed purified bullfrog ( Rana catesbeiana) GH and PRL as standards and radioligand, and their respective antibodies. Using ELISA, plasma VTG titers were related to ovarian weight. GH, PRL, and VTG displayed different trends related to season and sex. In male frogs the GH and PRL trends have been found parallel, showing the highest concentrations (35 and 85 ng/ml, respectively) during the winter months. In the female frogs, the GH trend behaved differently from that in males; in fact, plasma GH changed during the annual reproductive cycle, showing two main peaks occurring during the reproductive period and the autumn ovarian recrudescence that are well correlated with the vitellogenesis as well as with the main changes in ovarian weight.

Hormonal treatments modulate pulsatile plasma growth hormone, gonadotrophin and osteocalcin levels in postmenopausal women

Oestrogen plays a role in modulating growth hormone (GH), luteinizing hormone (LH), folliclestimulating hormone (FSH) and osteocalcin secretion in women. Indeed, the postmenopausal period is characterized by changes in plasma GH, LH, FSH and osteocalcin levels. The aim of the present study was to investigate the changes in the secretory patterns of these hormones in postmenopausal women under different therapeutic regimens. A total of 20 subjects took part in the study. They were subdivided into four groups comprising 5 untreated postmenopausal women (time since menopause 1–5 years), 5 postmenopausal women receiving steroid hormone replacement therapy, 5 postmenopausal women receiving salmon calcitonin and 5 fertile women with regular menstrual cycles. Blood samples were collected every 15 min for 4 h and hormone levels were measured by radioimmunoassay. Plasma GH levels fell in the postmenopausal women, but were restored under oestrogen-progestogen treatment. The pulsatile patterns of plasma LH and FSH did not show significant differences in the treated and untreated women. Plasma osteocalcin levels showed episodic fluctuations and mean levels decreased under steroid hormone or calcitonin treatment. No significant correlation was observed between plasma GH and osteocalcin or gonadotrophin levels or body mass index.

Growth hormone (GH)-binding protein in normal and GH-deficient dwarf rats.

There are GH-binding proteins (GHBPs) present in the blood of many species, and these correspond to the extracellular GH-binding domain of the GH receptor. In the rat, GHBP arises by alternative splicing of the GH receptor mRNA, but little is known of the physiological role of circulating GHBP, or its relationship with episodic GH secretion. We have developed a sensitive radioimmunoassay based on recombinant GHBP, and have measured rat GHBP levels in small samples of plasma from normal and GH-deficient dwarf rats. In normal adult rats, GHBP levels were two- to threefold higher in females than in males (16.6 +/- 0.8 vs 6.4 +/- 0.4 micrograms/l, P < 0.001), but this sex difference was not seen in dwarf rats. A continuous infusion of human GH in dwarf males raised plasma GHBP to 23.5 +/- 3.5 micrograms/l compared with 6.7 +/- 0.5 micrograms/l in sham-infused animals, whereas suppression of GH by continuous infusion of a long-acting somatostatin analogue in female dwarf rats had no effect on GHBP. In anaesthetized rats, large changes in plasma GH caused by i.v. administration of rat GH, somatostatin or GH-releasing factor did not affect GHBP acutely. Both GH and GHBP were also measured in serial blood samples from conscious normal and dwarf rats. A sexually dimorphic GH secretory pattern was observed in both strains. Males showed peaks and troughs of GH every 3 h varying over a 100-fold range, whereas females exhibited more continuous GH secretion. Despite the large fluctuations in endogenous GH, GHBP levels remained relatively constant in individual normal or dwarf males, as well as in females of both strains, and there was no significant correlation between GH and GHBP either in individual rats or as a group. Our results suggest that GHBP is GH-dependent in the longer term, and that the higher GHBP levels in female rats require their continuous GH secretory pattern. However, plasma GHBP levels remain stable and are not affected by acute changes in endogenous or exogenous GH.

Plasma growth hormone levels in fed and fasted rainbow trout (Oncorhynchus mykiss) are decreased following handling stress

Plasma growth hormone concentrations of rainbow trout,Oncorhynchus mykiss, fasted for six weeks, were significantly (p < 0.01) higher than in comparable fed animals; in the fasted fish, the levels fell progressively following acute stress (by displacing the fish within their home aquarium), with significant differences from pre-stressed fish evident between one and thirty-two hours after application of the stressor. Plasma growth hormone concentrations also fell significantly in the fed group, but differences were evident only between two and eight hours after stressor application.Plasma cortisol concentrations in pre- and post-stressed fed and fasted fish were similar. There was a bimodal response to stressor application in both groups, with significantly higher values relative to the pre-stressed sample evident one and eight hours after disturbance, but not after two, four or thirty-two hours. The changes in plasma cortisol levels between the initial (09:00h) sample and the sample taken eight hours later resembles the diet pattern seen in trout given access to self-demand feeders.Plasma glucose concentrations in pre-stressed fed animals were higher than in pre-stressed fasted fish. This relationship was also evident between one and four hours and thirty-two hours after stressor application. The post-stress rise in plasma glucose concentration was evident between one and four hours in the fed group, and between four and eight hours in fasted fish.The diel changes in plasma growth hormone and glucose concentrations could not be attributed to normal circadian patterns, and there was no apparent correlation between changes in plasma growth hormone and cortisol concentrations. There was a significant inverse correlation between plasma glucose and growth hormone concentrations when the total data set were analyzed, but these correlations were not apparent when the treatment groups were analyzed separately.

Effects of thymosin alpha-1 on pituitary hormone release.

The thymosins are a family of hormone-like products of epithelial cells of the thymus which are important in maintenance and function of the immune system. Thymosin fraction 5, a partially purified extract of calf thymus, can influence pituitary hormone release. We have studied the effects of thymosin alpha 1 (T alpha 1), the first peptide isolated from thymosin fraction 5, on thyrotropin (TSH), adrenocorticotropin (ACTH), prolactin (Prl) and growth hormone (GH) release. To evaluate its effect in vivo we injected the peptide into the third ventricle of conscious male rats and measured the concentration of the pituitary hormones in plasma at different times after the injection. Following third-ventricular injection of T alpha 1, there was a significant decrease in plasma TSH and ACTH concentrations in comparison with values of control groups injected with diluent. The decrease in plasma TSH was of longer duration and was obtained with a lower dose of T alpha 1 than that of ACTH. Also, a significant decrease in plasma Prl was observed, with the same dose as for TSH. On the other hand, there were no significant changes in plasma GH. To examine if there is any direct effect of T alpha 1 at the pituitary level, we incubated hemipituitaries from male rats in vitro with different concentrations of the peptide. In this system T alpha 1 evoked a dose-dependent release of TSH and ACTH, while there was no effect on the release of Prl and GH.(ABSTRACT TRUNCATED AT 250 WORDS)

Mechanism of Growth Retardation following Chronic Administration of β-Adrenoceptor Antagonists to Developing Rats

A total of 112 3-week old Wistar rats were separated into eight groups: control groups I-IV (n = 62) and propranolol-treated groups V-VIII (n = 50). Propranolol hydrochloride (100 mg/kg) was present in the rats' drinking water until 26 weeks of age and growth rates of all groups were monitored daily until 53 days of age and thereafter every third day throughout the study. Chronic oral propranolol administration produced growth retardation (P less than 0.05) in both sexes that was reversible when treatment was discontinued. Organ weights were generally smaller in propranolol-treated rats; on the other hand, the ratio of most organ weights per 100 g of body weight was greater in propranolol-treated rats (especially females). The radio-immunological determination of plasma growth hormone showed increased concentrations of growth hormone in propranolol-treated rats (P less than 0.05), whereas hypothalamic somatostatin content was not significantly changed. The results showed that the retarded growth rate following chronic oral propranolol administration to growing rats was independent of changes in plasma growth hormone and hypothalamic somatostatin concentrations, and that retardation was entirely reversible when the beta-adrenoceptor antagonist was discontinued.

Measurement of developmental changes in plasma insulin-like growth factor-I levels of broiler chickens by radioreceptor assay and radioimmunoassay

The main purpose of this study was to examine the relationship between insulin-like growth factor-I (IGF-I) and growth hormone (GH) during embryonic and posthatching development of broiler chickens. Two heterologous assays were validated for measurement of IGF-I in chicken and turkey plasma. A radioreceptor assay (RRA), utilizing microsomal membranes prepared from human placenta, was modified and validated for measurement of IGF peptide (mainly IGF-I). A double-antibody radioimmunoassay (RIA) was validated for measurement of immunoreactive IGF-I levels in chicken and turkey plasma. In both assay systems, recombinant-derived human IGF-I was used for standards and trace hormone. Hypophysectomy in turkey poults reduced plasma levels of IGF (RRA) by 35% and IGF-I (RIA) by 59% as compared to intact control turkeys. In Experiment 1,14 chicken embryos were bled at 15, 17, 19, and 21 days of incubation and at 1 week of age to determine plasma levels of IGF-I and GH. Plasma IGF levels (RRA) remained constant during late incubation, but increased significantly ( P < 0.05) at 1 week of age. Plasma IGF-I levels (RIA) declined 2 days before hatching; however, plasma levels of IGF-I were sharply elevated ( P < 0.05) at 1 week of age. Plasma GH concentrations were low in embryos and were greatly elevated ( P < 0.05) at hatching (21 days of incubation) and at 1 week of age. In Experiment 2, 12 different broiler cockerels were weighed and then bled by cardiac puncture each week from hatching (1 day of age) to 7 weeks of age. The plasma profiles of IGF, IGF-I, GH, triiodothyronine (T 3), and thyroxine (T 4) were each compared to relative growth rate by analysis of covariance. Plasma IGF and IGF-I levels increased progressively from 0 to 3 weeks of age and were maintained in a plateau from 3 to 7 weeks of age. Plasma GH levels reached a peak at 4 weeks of age, but declined sharply thereafter, while IGF and IGF-I levels remained elevated. Plasma T 3 concentrations were progressively increased and reached peak concentrations at 3 weeks of age, while plasma T 4 levels increased only at 6 and 7 weeks of age. There was a high correlation ( P < 0.01) between relative growth rate and age-related changes in plasma levels of IGF ( r = 0.96), IGF-I ( r = 0.97), and T 3 ( r = 0.94); however, there was no correlation between relative growth rate and changes in plasma GH or T 4. These observations suggest that thyroid activity could be more important than GH in maintaining normal growth rate and plasma IGF-I levels in broiler chickens.
