Autophagy is Involved in Cardiac Remodeling in Response to Environmental Temperature Change.

Objectives: To study the reversibility of cold-induced cardiac hypertrophy and the role of autophagy in this process. Background: Chronic exposure to cold is known to cause cardiac hypertrophy independent of blood pressure elevation. The reversibility of this process and the molecular mechanisms involved are unknown. Methods: Studies were performed in two-month-old mice exposed to cold (4°C) for 24 h or 10 days. After exposure, the animals were returned to room temperature (21°C) for 24 h or 1 week. Results: We found that chronic cold exposure significantly increased the heart weight/tibia length (HW/TL) ratio, the mean area of cardiomyocytes, and the expression of hypertrophy markers, but significantly decreased the expression of genes involved in fatty acid oxidation. Echocardiographic measurements confirmed hypertrophy development after chronic cold exposure. One week of deacclimation for cold-exposed mice fully reverted the morphological, functional, and gene expression indicators of cardiac hypertrophy. Experiments involving injection of leupeptin at 1 h before sacrifice (to block autophagic flux) indicated that cardiac autophagy was repressed under cold exposure and re-activated during the first 24 h after mice were returned to room temperature. Pharmacological blockage of autophagy for 1 week using chloroquine in mice subjected to deacclimation from cold significantly inhibited the reversion of cardiac hypertrophy. Conclusion: Our data indicate that mice exposed to cold develop a marked cardiac hypertrophy that is reversed after 1 week of deacclimation. We propose that autophagy is a major mechanism underlying the heart remodeling seen in response to cold exposure and its posterior reversion after deacclimation.

Over 100-Year Preservation and Temporal Fluctuations of Cell Wall Polysaccharides in Marine Sediments.

Polysaccharides constitute an important carbon pool in marine systems, but much is still unknown about the fate and degradation of these compounds. They are derived partly from production in situ, and in coastal areas, they are partly terrestrially derived, originating from freshwater runoff from land. The aim of this study was to test the applicability of high-throughput polysaccharide profiling for plant and algal cell-wall compounds in dated sediment cores from a coastal marine environment, to examine the preservation of cell-wall polysaccharides and explore their potential as proxies for temporal environmental changes. Preserved compounds and remains of organisms are routinely used as paleoenvironmental proxies as the amount and composition of different compounds that can provide insight into past environmental conditions, and novel means for reporting environmental changes are highly sought.

Hypothalamic control of energy expenditure and thermogenesis.

Energy expenditure and energy intake need to be balanced to maintain proper energy homeostasis. Energy homeostasis is tightly regulated by the central nervous system, and the hypothalamus is the primary center for the regulation of energy balance. The hypothalamus exerts its effect through both humoral and neuronal mechanisms, and each hypothalamic area has a distinct role in the regulation of energy expenditure. Recent studies have advanced the understanding of the molecular regulation of energy expenditure and thermogenesis in the hypothalamus with targeted manipulation techniques of the mouse genome and neuronal function. In this review, we elucidate recent progress in understanding the mechanism of how the hypothalamus affects basal metabolism, modulates physical activity, and adapts to environmental temperature and food intake changes.

Hypoxia Induces Oxidative Injury and Apoptosis via Mediating the Nrf-2/Hippo Pathway in Blood Cells of Largemouth Bass (Micropterus salmoides)

Investigating how aquatic animals respond to hypoxia brought about by changes in environmental temperature may be of great significance to avoid oxidative injury and maintain the quality of farmed fish in the background of global warming. Here, we investigated the effects of hypoxia on oxidative injury and environment-sensing pathway in blood cells of Micropterus salmoides. The total blood cell count (TBCC) and Giemsa staining showed that hypoxia could lead to damage of blood cells. Flow cytometry analysis confirmed that the apoptosis rate, Ca2+ level, NO production and ROS of blood cells were significantly increased under hypoxia stress. Environment-sensing pathways, such as Nrf2 pathway showed that hypoxia resulted in significant up-regulation of hiF-1 alpha subunit (Hif-1α), nuclear factor erythroid 2-related factor 2 (Nrf2) and kelch-1ike ECH- associated protein l (Keap1) expression. Meanwhile, the expression of Hippo pathway-related genes such as MOB kinase activator 1 (MOB1), large tumor suppressor homolog 1/2 (Lats1/2), yes-associated protein/transcriptional co-activator with PDZ-binding motif (YAP/TAZ), protein phosphatase 2A (PP2A) were significantly increased in blood cells after hypoxia exposure. In addition, hypoxia stress also increased the expression of catalase (CAT) and glutathione peroxidase (GPx), but decreased the expression of superoxide dismutase (SOD). Consequently, our results suggested that hypoxia could induce oxidative injury and apoptosis via mediating environment-sensing pathway such as Nrf2/Hippo pathway in blood cells of M. salmoides.

Thermal Camouflaging MXene Robotic Skin with Bio‐Inspired Stimulus Sensation and Wireless Communication

AbstractCephalopod skin, which is capable of dynamic optical camouflage, environmental perceptions, and herd communication, has long been a source of bio‐inspiration for developing soft robots with incredible optoelectronic functions. Yet, challenges still exist in designing a stretchable and compliant robotic skin with high‐level functional integration for soft robots with infinite degrees of freedom. Herein, an emerging 2D material, Ti3C2Tx MXene, and an interfacial engineering strategy are adopted to fabricate the soft robotic skin with cephalopod skin‐inspired multifunctionality. By harnessing interfacial instability, the MXene robotic skin with reconfigurable microtextures demonstrates tunable infrared emission (0.30–0.80), enabling dynamic thermal camouflage for soft robots. Benefiting from the intrinsic Seebeck effect, crack propagation behaviors as well as high electrical conductivity, the MXene robotic skins are tightly integrated with thermal/strain sensation capabilities and can serve as a deformable antenna for wireless communication. Without additional electronics installed, the soft robots wearing the conformal MXene skins perform adaptive thermal camouflage based on the thermoelectric feedback in response to environmental temperature changes. With built‐in strain sensing and wireless communication capabilities, the soft robot can record its locomotion routes and wirelessly transmit the key information to the following soft robot to keep both in disguise under thermographic cameras.

Molecular physiology regulating cold tolerance and acclimation of Caenorhabditis elegans.

Many organisms can survive and proliferate in changing environmental temperatures. Here, we introduce a molecular physiological mechanism for cold tolerance and acclimation of the nematode Caenorhabditis elegans on the basis of previous reports and a new result. Three types of thermosensory neurons located in the head, ASJ, ASG, and ADL, regulate cold tolerance and acclimation. In ASJ, components of the light-signaling pathway are involved in thermosensation. In ASG, mechanoreceptor DEG-1 acts as thermoreceptor. In ADL, transient receptor potential channels are thermoreceptors; however, the presence of an additional unidentified thermoreceptor is also speculated. ADL thermoresponsivity is modulated by oxygen sensory signaling from URX oxygen sensory neurons via hub interneurons. ASJ releases insulin and steroid hormones that are received by the intestine, which results in lipid composition changing with cold tolerance. Additionally, the intestinal transcriptional alteration affects sperm functions, which in turn affects the thermosensitivity of ASJ; thus, the neuron–intestine–sperm–neuron tissue circuit is essential for cold tolerance.

Role of Melatonin in Temperature-Induced Activation of the Neuroendocrine Reproductive Axis in Garter Snakes

An animal’s ability to respond optimally to changing environmental conditions is paramount to successfully reproducing and thus maximizing fitness. Studies on photoperiod-induced changes in neural thyroid hormone metabolism have conclusively linked environmental cues to the neuroendocrine reproductive axis of birds and mammals. Whether this conserved mechanism also transduces changes in environmental temperature, however, has not been fully addressed. We investigated whether the hormone melatonin mediates the effects of low-temperature dormancy on thyroid hormone metabolism within the hypothalamus of red-sided garter snakes (Thamnophis sirtalis parietalis). To address this question, we used immunohistochemistry to assess changes in thyroid-stimulating hormone (TSH) in the infundibulum of the pituitary and deiodinase 3 (Dio3) and gonadotropin-releasing hormone (GnRH) in the hypothalamus. We also asked if changes in TSH, Dio3, and/or GnRH immunoreactivity are associated with changes in male courtship behavior. In contrast to our predictions, 6 weeks of dormancy at 4°C significantly decreased the number of TSH-labeled cells in the infundibulum. It is possible that the observed decrease in TSH is related to the release of snakes from temperature refractoriness, but this idea needs further testing. Treatment of snakes with the melatonin precursor 5-hydroxytryptophan during dormancy at 4°C both reversed the temperature-induced change in TSH immunoreactivity and disrupted the temporal pattern of male courtship behavior. These results suggest that TSH cells within the infundibulum are both modulated by temperature and sensitive to changes in melatonin. As predicted, male snakes hibernated at an elevated temperature of 12°C for 6 weeks and treated with vehicle showed no change in TSH-, Dio3-, or GnRH-immunoreactive cell number. Treatment of snakes with the melatonin receptor antagonist luzindole was not sufficient in rescuing the effects of dormancy at 12°C on TSH immunoreactivity or courtship behavior. However, luzindole-treated snakes showed a significant increase in GnRH-immunoreactive cell number, suggesting that melatonin exerts an inhibitory effect on GnRH in garter snakes. In summary, our results provide critical insights into the mechanisms that mediate the effects of temperature on reproductive physiology and behavior.

Correlation Between Changes in Environmental Temperature and Performance of High-Discharge Lithium-Polymer Batteries

Electric vehicles (EVs) have been developed to solve environmental problems and the depletion of energy resources, and batteries have been used as their main energy source. The battery used in this study was a lithium polymer battery with the same chemical structure as lithium-ion. The use of a solid electrolyte has advantages, such as lack of leakage of electrolyte, high density per size, and convenient shape design. In this study, we analyzed the heat generation and performance characteristics of EV batteries through experiments on changes in environmental temperature. There is a difference in the heat generation depending on the discharge rate, and the performance characteristics of the battery improves as the environmental temperature rises. In the experiment, the battery discharge performance and heat generation were meaningful at 40°C environmental temperature, the worst battery discharge performance and heat generation at −30°C environmental temperature were 25.1 W (1 C), 81.0 W (2 C), and 151.5 W (3 C). In this study, the heat generation and performance characteristics of the battery were analyzed according to the change in the environmental temperature and discharge rate of the battery. We proposed a relationship between heat generation and environmental temperature in terms of discharge rate. The relationship is significant in designing thermal management system for battery powered devices.

Cell-autonomous control of intracellular temperature by unsaturation of phospholipid acyl chains.

Intracellular temperature affects a wide range of cellular functions in living organisms. However, it remains unclear whether temperature in individual animal cells is controlled autonomously as a response to fluctuations in environmental temperature. Using two distinct intracellular thermometers, we find that the intracellular temperature of steady-state Drosophila S2 cells is maintained in a manner dependent on Δ9-fatty acid desaturase DESAT1, which introduces a double bond at the Δ9 position of the acyl moiety of acyl-CoA. The DESAT1-mediated increase of intracellular temperature is caused by the enhancement of F1Fo-ATPase-dependent mitochondrial respiration, which is coupled with thermogenesis. We also reveal that F1Fo-ATPase-dependent mitochondrial respiration is potentiated by cold exposure through the remodeling of mitochondrial cristae structures via DESAT1-dependent unsaturation of mitochondrial phospholipid acyl chains. Based on these findings, we propose a cell-autonomous mechanism for intracellular temperature control during environmental temperature changes.

A Robust Algorithm for State-of-Charge Estimation under Model Uncertainty and Voltage Sensor Bias

Accurate estimation of the state of charge (SOC) of zinc–nickel single-flow batteries (ZNBs) is an important problem in battery management systems (BMSs). A nonideal electromagnetic environment will usually cause the measured signal to contain nonnegligible noise and bias. At the same time, due to the influence of battery ageing, environmental temperature changes, and a complex reaction mechanism, it is difficult to establish a very accurate system model that can be applied to various complex working conditions. The unscented Kalman filter (UKF) is a widely used SOC estimation algorithm, but the UKF will reduce the estimation accuracy and divergence under the influence of inaccurate model and sensor errors. To improve the performance of the UKF, a robust desensitized unscented Kalman filter (RDUKF) is proposed to realize an accurate SOC estimation of batteries in the context of different disturbances. Then, the proposed method is applied to cases of error interference, such as Gaussian noise, voltage sensor drift, an unknown initial state, and inaccurate model parameters. The simulation and experimental results show that compared with the standard UKF algorithm, the proposed estimation algorithm can effectively suppress the influence of various errors and disturbances and achieve higher accuracy and robustness.

Latent Growth Curve Modeling for COVID-19 Cases in Presence of Time-Variant Covariate.

For the past two years, the entire world has been fighting against the COVID-19 pandemic. The rapid increase in COVID-19 cases can be attributed to several factors. Recent studies have revealed that changes in environmental temperature are associated with the growth of cases. In this study, we modeled the monthly growth rate of COVID-19 cases per million infected in 126 countries using various growth curves under structural equation modeling. Moreover, the environmental temperature has been introduced as a time-varying covariate to enhance the performance of the models. The parameters of growth curve models have been estimated, and accordingly, the results are discussed for the affected countries from August 2020 to July 2021.

The FtsH-Inactive Protein FtsHi5 Is Required for Chloroplast Development and Protein Accumulation in Chloroplasts at Low Ambient Temperature in Arabidopsis.

Chloroplasts are indispensable for higher plants. The growth and development of plants are very sensitive to environmental temperature changes, and chloroplast development is also regulated by adverse environmental temperatures. However, the molecular mechanism of how plants coordinate chloroplast development and environmental temperature changes remains largely unknown. Here, a temperature-conditioned chloroplast development defective mutant thermo-sensitive mutant in leaf color 2 (tsl2) of Arabidopsis was obtained through a forward genetic screening. The tsl2 mutant showed a weak yellowish phenotype at normal growth temperature (22°C), and the phenotype was more pronounced at low growth temperature (16°C) and largely rescued at high growth temperature (29°C). Bulk Segregant Analysis (BSA) revealed that TSL2 encodes FtsH-Inactive Protein 5 (FtsHi5). Genetic complementation analysis confirmed that complemented expression of FtsHi5 rescued the chlorophyll content and thylakoid development defects observed in tsl2 mutants at 16°C. Quantitative mass spectrometry analysis with Tandem Mass Tag (TMT) isobaric labeling revealed broad changes in the chloroplast proteome of tsl2 mutant plants at low temperature, which is agreed with the impaired chloroplast biogenesis and function in tsl2 plants. Together, our data demonstrates that FtsHi5/TSL2 plays an important role in chloroplast development and protein accumulation in chloroplasts, especially at low environmental temperatures in Arabidopsis.

Population genomics provides insights into the evolution and adaptation of tree shrews (Tupaia belangeri) in China.

Physiological adaptation of tree shrews (Tupaia belangeri) to changing environmental temperature has been reported in detail. However, the T. belangeri origin (mainland or island), population history, and adaptation to historical climate change remain largely unknown or controversial. Here, for the first time, we sequenced the simplified genome of 134 T. belangeri individuals from 12 populations in China and further resequenced one individual from each population. Using population genomic approaches, we first observed considerable genetic variation in T. belangeri. Moreover, T. belangeri populations formed obvious genetic structure and reflected different demographic histories; they generally exhibited high genetic diversity, although the isolated populations had relatively low genetic diversity. The results presented in this study indicate that T. b. modesta and T. b. tonquinia were separated recently and with a similar population dynamics. Second, physical barriers rather than distance were the driving factors of divergence, and environmental heterogeneity may play an important role in genetic differentiation in T. belangeri. Moreover, our analyses highlight the role of historical global climates in the T. belangeri population dynamics and indicate that the decrease of the T. belangeri population size may be due to the low temperature. Finally, we identified the olfaction-associated adaptive genes between different altitude populations and found that olfactory-related genes of high-altitude populations were selectively eliminated. Our study provides demographic history knowledge of T. belangeri; their adaption history offers new insights into their evolution and adaptation, and provides valuable baseline information for conservation measures.

Regulation of Calvin-Benson cycle enzymes under high temperature stress.

The Calvin-Benson cycle (CBC) consists of three critical processes, including fixation of CO2 by Rubisco, reduction of 3-phosphoglycerate (3PGA) to triose phosphate (triose-P) with NADPH and ATP generated by the light reactions, and regeneration of ribulose 1,5-bisphosphate (RuBP) from triose-P. The activities of photosynthesis-related proteins, mainly from the CBC, were found more significantly affected and regulated in plants challenged with high temperature stress, including Rubisco, Rubisco activase (RCA) and the enzymes involved in RuBP regeneration, such as sedoheptulose-1,7-bisphosphatase (SBPase). Over the past years, the regulatory mechanism of CBC, especially for redox-regulation, has attracted major interest, because balancing flux at the various enzymatic reactions and maintaining metabolite levels in a range are of critical importance for the optimal operation of CBC under high temperature stress, providing insights into the genetic manipulation of photosynthesis. Here, we summarize recent progress regarding the identification of various layers of regulation point to the key enzymes of CBC for acclimation to environmental temperature changes along with open questions are also discussed.

A real-time temperature responsive material based on partial inverse opal photonic crystals and cholesteric liquid crystals

In this paper, we fabricated a temperature-sensitive material with a partial inverse opal structure, which can reversibly and quickly identify small temperature changes at room temperature and show significant color differences. First, we synthesized silica microsphere and prepared silica photonic crystals (PCs), then we composited PCs with polymer. After covering the mask, we prepared partial inverse opal silica PCs template by using selective etching. The temperature-sensitive liquid crystals (LCs) composed of cholesterol oleyl carbonate (COC), cholesterol nonanoate (CPE) and cholesterol benzoate (CBZ) were filled in partial inverse opal silica PCs template. In the end, we sealed the film by methyl methacrylate (MMA) and polyethylene glycol diacrylate (PEGDA). In this way, we prepared the fast temperature-sensitive dual-opal PCs/LCs composite film, and studied the mechanism of temperature regulation on material display mode. As the temperature gone from 26 °C to 30 °C, the material changed colors rapidly from red to blue. The result showed that this PCs/LCs composite film had well temperature response performance, which can monitor slight environmental temperature change. By selective etching the material, we designed PCs film patterns. The different patterning states displayed and changed fast at different temperatures, showing that the material had a good application prospect in the fields of temperature sensor, anti-counterfeiting, and display device.

RNA thermometers and other regulatory elements: Diversity and importance in bacterial pathogenesis.

Survival of microorganisms depends to a large extent on environmental conditions and the occupied host. By adopting specific strategies, microorganisms can thrive in the surrounding environment and, at the same time, preserve their viability. Evading the host defenses requires several mechanisms compatible with the host survival which include the production of RNA thermometers to regulate the expression of genes responsible for heat or cold shock as well as of those involved in virulence. Microorganisms have developed a variety of molecules in response to the environmental changes in temperature and even more specifically to the host they invade. Among all, RNA-based regulatory mechanisms are the most common ones, highlighting the importance of such molecules in gene expression control and novel drug development by suitable structure-based alterations. Mechanical Mechanism Analysis of Warping and Bulging Of Track Slab of High-Speed Railway in Service

Due to warpage and ballooning of high-speed rail track slab caused by debonding and environmental temperature changes during the deterioration of CA mortar layer being extremely concerned in practical engineering, it is based on numerical simulation of typical working conditions and analysis of test model that the deformation dynamics mechanism of track slab of high-speed rail in service is studied in this paper. Firstly, three kinds of damage conditions of CA mortar layer are designed to simulate the partial stress state of track slab under normal, warping and bulging conditions, and the results of model test are compared with those of finite element analysis so that the accuracy and credibility of the numerical simulation method and results are verified. Then, through finite element numerical simulation, the dynamical mechanism of actual full-scale high-speed rail track slab under vibration load is studied. The results show that the warping deformation around the track slab and the bulging deformation in the middle part under the action of positive and negative temperature gradient load caused by environmental temperature change will have a great impact on the structural performance of itself and CA mortar layer; Bulging deformation of track slab is more destructive to its structure than warping deformation. It is of great practical significance to further study the critical position of track plate warpage and bulging deformation, and to optimize and strengthen the structure of this part; The research results are of great significance to further study the deterioration.

Regulation of collagen deposition in the trout heart during thermal acclimation.

The passive mechanical properties of the vertebrate heart are controlled in part by the composition of the extracellular matrix (ECM). Changes in the ECM, caused by increased blood pressure, injury or disease can affect the capacity of the heart to fill with blood during diastole. In mammalian species, cardiac fibrosis caused by an increase in collagen in the ECM, leads to a loss of heart function and these changes in composition are considered to be permanent. Recent work has demonstrated that the cardiac ventricle of some fish species have the capacity to both increase and decrease collagen content in response to thermal acclimation. It is thought that these changes in collagen content help maintain ventricle function over seasonal changes in environmental temperatures. This current work reviews the cellular mechanisms responsible for regulating collagen deposition in the mammalian heart and proposes a cellular pathway by which a change in temperature can affect the collagen content of the fish ventricle through mechanotransduction. This work specifically focuses on the role of transforming growth factor β1, MAPK signaling pathways, and biomechanical stretch in regulating collagen content in the fish ventricle. It is hoped that this work increases the appreciation of the use of comparative models to gain insight into phenomenon with biomedical relevance.

Season-sex interaction induces changes in the ecophysiological traits of a lizard in a high altitude cold desert, Puna region.

Functional traits are those characteristics of organisms that influence the ability of a species to develop in a habitat and persist in the face of environmental changes. The traits are often affected by a multiplicity of species-dependent and external factors. Our objective was to investigate thermal biology of Liolaemus ruibali in a high altitude cold desert at the arid Puna region, Argentina. We address the following question: do sex and seasonal variations in environmental temperature induce changes in the ecophysiological traits? We measured and compared the operative temperatures between fall and spring; and between sexes and seasons, we compared the ecophysiological traits of lizards, microenvironmental temperatures and thermoregulatory behavior. Air and operative temperatures were different between seasons. We found an effect of season-sex interaction on field body temperatures, preferred temperatures, panting threshold and thermal quality. The voluntary and critical temperatures presented seasonal variation in relation to changes in environmental temperatures, suggesting thermal acclimatization. We note behavioral changes between seasons, with the substrate being the main resource for gaining heat in spring. We conclude that Liolaemus ruibali is an efficient thermoregulator; it is a eurythermic lizard and presents phenotypic plasticity in different ecophysiological and behavioral traits induced by sex and seasonality. In addition, we predict that this population could buffer the effects of projected global warming scenarios.

Development of a Non-contact Skin Temperature Measurement System for Newborns Based on Intelligent Analysis of Thermometric Data

Measurement of body temperature is a vital parameter when monitoring newborns. The peculiarity of thermometry of newborns involves certain anatomical features of children, which make them vulnerable to changes in environmental temperature. These features include: the reduced amount of subcutaneous fat, thin epidermis, blood vessels located near the surface of the skin. This paper demonstrates the choice of the method of converting the original biometric data into the thermal map, as well as the analysis of the obtained experimental thermal imaging data of using the pyrometric method of scanning the incubator bed for newborns.
