Effect of induced electric field parameters on physiochemical characteristics and microorganism of orange juice

AbstractThe induced electric field (IEF) was produced via an oscillating magnetic field, for direct heating of orange juice within a winding coil. During the process, the applied magnetic field was at the range from 0.07 to 1.43 T with 50 kHz. Then, the numerical relationships between excitation voltage, magnetic field, IEF, induced current density, magnetomotive force and energy efficiency were investigated. Since a rectangular wave of excitation voltage was applied, the sawtooth waveform of induced voltage or IEF loaded on the juice was observed. As the magnetic field increased, outlet temperature and induced current density in the juice was improved, the maximum values were 99.5°C and 0.50 A/cm2, respectively, at a conductivity 4.53 mS/cm for the residence time of 4.5 min. However, the efficiency of the process exhibited a negative correlation with the escalation of excitation voltage. At initial colony count about 4.50 log colony‐forming units/mL, the IEF pasteurization reduced all the aerobic microorganisms, molds, and yeasts in the juice, which were evaluated at detectable limits below 1 log colony‐forming units/mL. Moreover, all treatments had no significant effects on pH, total soluble solids and titratable acidity of the juice (p > 0.05). In particular, there was also no significant change in color parameters after IEF pasteurization (p > 0.05). It indicated a significant decrease in vitamin C and total phenolic content of the control groups (p > 0.05), but no significant change in carotenoid content was observed in all treated juice (p > 0.05). heating technology using IEF has the potential to the pasteurization of fruit juice.Practical applicationsInduction electric field technology has progressed rapidly in the past 2 years, it can use oscillating alternating magnetic field to realize the direct heating of liquid food. The process has thermal and non‐thermal effects, at 65–70°C, within 15–30 s to achieve the commercial asepsis of acidic liquid food. After the pasteurization, it can better maintain the color and flavor of the juice. We have assembled a laboratory prototype for testing in the early stage, the processing capacity is at the range of 0–10 L/h. Currently, the system has been able to reach the pilot production scale with the processing capacity of 100–500 L/h. Through the use of highly permeable magnetic materials, the energy consumption per ton of juice processing is controlled at 120 k·Wh level for the pasteurization and is expected to achieve a larger processing capacity in the future.

Effects of different light intensity on leaf color changes in a Chinese cabbage yellow cotyledon mutant.

Leaf color is one of the most important phenotypic features in horticultural crops and directly related to the contents of photosynthetic pigments. Most leaf color mutants are determined by the altered chlorophyll or carotenoid, which can be affected by light quality and intensity. Our previous study obtained a Chinese cabbage yellow cotyledon mutant that exhibited obvious yellow phenotypes in the cotyledons and the new leaves. However, the underlying mechanisms in the formation of yellow cotyledons and leaves remain unclear. In this study, the Chinese cabbage yellow cotyledon mutant 19YC-2 exhibited obvious difference in leaf color and abnormal chloroplast ultrastructure compared to the normal green cotyledon line 19GC-2. Remarkably, low-intensity light treatment caused turn-green leaves and a significant decrease in carotenoid content in 19YC-2. RNA-seq analysis revealed that the pathways of photosynthesis antenna proteins and carotenoid biosynthesis were significantly enriched during the process of leaf color changes, and many differentially expressed genes related to the two pathways were identified to respond to different light intensities. Remarkably, BrPDS and BrLCYE genes related to carotenoid biosynthesis showed significantly higher expression in 19YC-2 than that in 19GC-2, which was positively related to the higher carotenoid content in 19YC-2. In addition, several differentially expressed transcription factors were also identified and highly correlated to the changes in carotenoid content, suggesting that they may participate in the regulatory pathway of carotenoid biosynthesis. These findings provide insights into the molecular mechanisms of leaf color changes in yellow cotyledon mutant 19YC-2 of Chinese cabbage.

Molecular and Photosynthetic Performance in the Yellow Leaf Mutant of Torreya grandis According to Transcriptome Sequencing, Chlorophyll a Fluorescence, and Modulated 820 nm Reflection.

To study the photosynthetic energy mechanism and electron transfer in yellow leaves, transcriptomics combined with physiological approaches was used to explore the mechanism of the yellow leaf mutant Torreya grandis ‘Merrillii’. The results showed that chlorophyll content, the maximal photochemical efficiency of PSII (Fv/Fm), and the parameters related to the OJ phase of fluorescence (φEo, φRo) were all decreased significantly in mutant-type T. grandis leaves. The efficiency needed for an electron to be transferred from the reduced carriers between the two photosystems to the end acceptors of the PSI (δRo) and the quantum yield of the energy dissipation (φDo) were higher in the leaves of mutant-type T. grandis compared to those in wild-type leaves. Analysis of the prompt fluorescence kinetics and modulated 820 nm reflection showed that the electron transfer of PSII was decreased, and PSI activity was increased in yellow T. grandis leaves. Transcriptome data showed that the unigenes involved in chlorophyll synthesis and the photosynthetic electron transport complex were downregulated in the leaves of mutant-type T. grandis compared to wild-type leaves, while there were no observable changes in carotenoid content and biosynthesis. These findings suggest that the downregulation of genes involved in chlorophyll synthesis leads to decreased chlorophyll content, resulting in both PSI activity and carotenoids having higher tolerance when acting as photo-protective mechanisms for coping with chlorophyll deficit and decrease in linear electron transport in PSII.

Analyses of key gene networks controlling carotenoid metabolism in Xiangfen 1 banana

BackgroundBanana fruits are rich in various high-value metabolites and play a key role in the human diet. Of these components, carotenoids have attracted considerable attention due to their physiological role and human health care functions. However, the accumulation patterns of carotenoids and genome-wide analysis of gene expression during banana fruit development have not been comprehensively evaluated.ResultsIn the present study, an integrative analysis of metabolites and transcriptome profiles in banana fruit with three different development stages was performed. A total of 11 carotenoid compounds were identified, and most of these compounds showed markedly higher abundances in mature green and/or mature fruit than in young fruit. Results were linked to the high expression of carotenoid synthesis and regulatory genes in the middle and late stages of fruit development. Co-expression network analysis revealed that 79 differentially expressed transcription factor genes may be responsible for the regulation of LCYB (lycopene β-cyclase), a key enzyme catalyzing the biosynthesis of α- and β-carotene.ConclusionsCollectively, the study provided new insights into the understanding of dynamic changes in carotenoid content and gene expression level during banana fruit development.

The genes crucial to carotenoid metabolism under elevated CO2 levels in carrot (Daucus carota L.)

The CO2 saturation point can reach as high as 1819 μmol· mol−1 in carrot (Daucus carota L.). In recent years, carrot has been cultivated in out-of-season greenhouses, but the molecular mechanism of CO2 enrichment has been ignored, and this is a missed opportunity to gain a comprehensive understanding of this important process. In this study, it was found that CO2 enrichment increased the aboveground and belowground biomasses and greatly increased the carotenoid contents. Twenty genes related to carotenoids were discovered in 482 differentially expressed genes (DEGs) through RNA sequencing (RNA-Seq.). These genes were involved in either carotenoid biosynthesis or the composition of the photosystem membrane proteins, most of which were upregulated. We suspected that these genes were directly related to quality improvement and increases in biomass under CO2 enrichment in carrot. As such, β-carotene hydroxylase activity in carotenoid metabolism and the expression levels of coded genes were determined and analysed, and the results were consistent with the observed change in carotenoid content. These results illustrate the molecular mechanism by which the increase in carotenoid content after CO2 enrichment leads to the improvement of quality and biological yield. Our findings have important theoretical and practical significance.

Plasma‐activated water to improve the stress tolerance of barley

AbstractIt is shown that plasma‐activated water (PAW) improves tolerance against combined low temperature and hypoxia stresses during germination. Due to PAW's effect on the antioxidant system, PAW pretreatment mitigates the reducing effect on plant growth of the hydroponically applied NaCl. PAW pretreatment reduces the salt‐stress‐induced decrease of the cysteine and γ‐glutamylcysteine content and improves the plant's recovery after NaCl stress. PAW pretreatment also modifies the NaCl‐induced changes in carotenoid content, especially, β‐carotenes and xanthophylls. These compounds, together with glutathione can reduce salt‐induced damage through the removal of excess reactive oxygen species, which could be the basis of the protective effect of PAW.

Raman spectroscopy has so far proved to be a suitable method for the analysis of carotenoids in a wide range of plant samples. This study aimed to determine whether the influence of mesotrione on the changes in carotenoid content of leaves in treated weed species Chenopodium album and Abutilon theophrasti can be detected using Raman spectroscopy. When the plants reached the targeted growth stages (C. album: 4 leaves, A. theophrasti: 2 leaves), Skaut herbicide (a.i. mesotrione 480 g l-1, SC) was applied in the amount of 120 g a.i. ha-1 in the tank mix with the adjuvant Alteox wet 40 (a.i. mineral oil (500 g L-1) + polyoxy alcohol (500 g L-1)), in the amount of 1 L ha-1. Leaf analysis, using Raman spectroscopy, was performed four times: on the 1st, 2nd, 3rd, and 7th day after the application of mesotrione. The intensity of the bands in the obtained Raman spectra, at positions 1007, 1155, and 1522 cm-1 in untreated and treated plants was analysed to determine the relative carotenoid content. The obtained results have shown that changes in the relative carotenoid content in the leaves of treated C. album plants can be detected as early as 3 days after the application of mesotrione, compared to the leaves of untreated plants of the same age. Meanwhile, a statistically significant difference for the same parameter in the leaves of treated A. theophrasti plants was evident at a later stage than in C. album, on the 7th day after the application of mesotrione. Consequently, this study has confirmed that Raman spectroscopy can be used as an adequate method for the rapid detection of the effect of herbicides on the content of carotenoids in the leaves of treated weed species.

The Impact of an Inert Gas Atmosphere on the Kinetics of Changes in the Physical and Chemical Properties of Carrot Lyophilisate

AbstractThe purpose of this study was to analyse the impact of blanching, the atmosphere of nitrogen and storage time on carotenoids content, colour and sorption properties of freeze-dried carrot. The material was stored for up to 16 weeks, in the dark, at room temperature. The colour was measured on the surface and in cross sections. As the storage time increased, there was a continual decrease in the carotenoids content in the carrot packed in atmospheric air. In the carrots packed in the atmosphere of nitrogen, no changes in carotenoid content were observed. The colour parameters correlated with the changes in the carotenoids content. The colour parameters changed on the surface and in cross sections in the same manner. During storage, a significant decrease in sorption properties of freeze-dried carrots was noted after 2 weeks of storage.

Evolution, Development and Function of Vertebrate Cone Oil Droplets.

To distinguish colors, the nervous system must compare the activity of distinct subtypes of photoreceptors that are maximally sensitive to different portions of the light spectrum. In vertebrates, a variety of adaptations have arisen to refine the spectral sensitivity of cone photoreceptors and improve color vision. In this review article, we focus on one such adaptation, the oil droplet, a unique optical organelle found within the inner segment of cone photoreceptors of a diverse array of vertebrate species, from fish to mammals. These droplets, which consist of neutral lipids and carotenoid pigments, are interposed in the path of light through the photoreceptor and modify the intensity and spectrum of light reaching the photosensitive outer segment. In the course of evolution, the optical function of oil droplets has been fine-tuned through changes in carotenoid content. Species active in dim light reduce or eliminate carotenoids to enhance sensitivity, whereas species active in bright light precisely modulate carotenoid double bond conjugation and concentration among cone subtypes to optimize color discrimination and color constancy. Cone oil droplets have sparked the curiosity of vision scientists for more than a century. Accordingly, we begin by briefly reviewing the history of research on oil droplets. We then discuss what is known about the developmental origins of oil droplets. Next, we describe recent advances in understanding the function of oil droplets based on biochemical and optical analyses. Finally, we survey the occurrence and properties of oil droplets across the diversity of vertebrate species and discuss what these patterns indicate about the evolutionary history and function of this intriguing organelle.

Development of a green procedure of citrus fruits waste processing to recover carotenoids

In this study, an original and green procedure of processing waste of the citrus fruits was developed using the concept of bio-refinery, innovative techniques “ultrasound”and “micro-wave”, and a green sol- vent “limonene”to recover carotenoids. Essential oil extraction was performed by Solvent Free Microwave Extraction (SFME) and compared to steam distillation (SD). The essential oil yields were comparable for both processes: 4.02 ±0.23% for SFME and 4.16 ±0.05% for SD. After that, carotenoid extraction from cit- rus peels was performed by ultrasound-assisted extraction (UAE) and conventional extraction (CE) using d -limonene obtained starting from essential oil, as a solvent, and then compared to n-hexane extract. Response surface methodology (RSM) using central composite designs (CCD) approach was launched to investigate the influence of process variables on the ultrasound-assisted extraction (UAE). The statisti- cal analysis revealed that the optimized conditions of ultrasound power, temperature and time were 208 W cm −2 , 20 °C and 5 min giving carotenoid content of 11.25 mg L −1 . Compared to the conventional ex- traction, (UAE) gave an increase of 40% in carotenoid content. The comparison to n-hexane extract gave no significant changes in carotenoid content. Combination of microwave, ultrasound and d -limonene ob- tained from a bio-refinery of a by-product of citrus fruits industry allow us to develop a very good envi- ronmental green approach giving high added values compounds, with a saving of time, and a complete valorisation of waste.

The effect of various sapropel concentrations on content and fluorescence of photosynthetic pigments of bean

Tarosiskio lake sapropel impact on beans (Vicia faba L.) photosynthesis parameters (content of chlorophylls and chlorophyll a fluorescence) was investigated in this study. Plants were sown in 1 L volume pots by 5 seeds with light loamy soil and grown in growth chambers. Considering sapropel composition, 2.2; 3.1 and 4 t/ha dry sapropel concentrations were chosen for investigation. Sapropel affected plants were compared with plants grown in unfertilised and with synthetic fertiliser fertilised soils. On 14th day of experiment, when the plants reached two leaves growths stage (BBCH 12), chlorophyll a fluorescence parameters of investigated plants were measured. Then plants were grown for 4 weeks more and at stem elongation stage (BBCH 31) chlorophyll a fluorescence parameters were measured repeatedly and also the content of chlorophylls and carotenoids were investigated. The efficiency II photosystem of bean increased by increasing sapropel concentration in soil, but insignificantly (p>0.05). Photosynthesis performance index was increasing (r=0.91; p 0.05). At BBCH 12 stage the highest non photochemical quenching (NPQ) value was measured in control beans, while at BBCH 31 stage, opposite trend was noticed (r=0.86; p 0.05), and the changes in carotenoid contents were similar to chlorophyll a+b.DOI: http://dx.doi.org/10.5755/j01.erem.72.4.16585

Profiles of gene family members related to carotenoid accumulation in citrus genus

Citrus fruit are an important reservoir of carotenoids. Numerous studies have been carried out to identify and profile the members of gene families involved in carotenoid biosynthetic pathway to explain the diversity of coloration in citrus fruit. It was found that gene expression analysis could not always explain the changes in carotenoid content and composition, indicating that other unknown genes and mechanisms should be operative. This review summarizes and updates the current knowledge on gene families involved in the citrus carotenoid biosynthetic pathway and their roles on the regulation of carotenoid biosynthesis, as well as provides insightful questions leading to future experimentation.

Effect of increasing manganese concentration in nutrient solution on the antioxidant activity, vitamin C, lycopene and polyphenol contents of tomato fruit

ABSTRACTThis study evaluated the effect of increasing manganese (Mn) nutrition on the content of antioxidative compounds such as vitamin C, lycopene and polyphenols, and the antioxidant activity of tomato (Lycopersicon esculentum Mill., cvs ‘Alboney F1’ and ‘Emotion F1’) fruit. Plants were grown in rockwool using a nutrient solution with the following content of Mn (mg dm–3): 0.0, 0.3, 0.6, 1.2, 2.4, 4.8, 9.6 and 19.2. The level of vitamin C and lycopene decreased with the increasing Mn nutrition. Since the colour of fruits was correlated with the change in carotenoid content, the decrease in lycopene content promoted the reduction of redness and increase of yellowness of fruits. However, total polyphenol content and antioxidant activity significantly increased when plant were exposed to toxic levels of Mn. Observed changes could be the result of the oxidative stress induced by high concentrations of Mn. Polyphenolic compounds play a crucial role in the plant’s response to Mn stress and affect predominantly the total antioxidant properties of fruits, which could be used as a source of phenolics. Moreover, total phenolic content measurement, as an easy and inexpensive method, could be used as an indicator of Mn-induced stress in fruits of tomato.

The effects of peeling and cooking on the mineral content and antioxidant properties in carrots enriched with potassium iodate and/or selenite (SeIV) and selenite (SeVI)

Research covered six variants: control, unfertilized carrots and carrots fertilized with: KIO3, Na2SeO4, Na2SeO3, KIO3 and simultaneously with Na2SeO4, and fertilized with KIO3 and simultaneously Na2SeO3. Carrots enriched with iodate or selenite, or both iodate and selenite, were characterized by higher amount of these minerals. Changes to the content of micro- and macroelements, during the cooking time of the carrots, both in peeled and unpeeled carrots, did not head in the same direction (increase, decrease and no change). However, cooking an unpeeled carrot generally resulted in the increased content of polyphenol and carotenoids. On the other hand, cooking peeled carrots led to a decrease in the content of polyphenol and a general lack of change in carotenoid content in relation to the unpeeled cooked carrot. During cooking, the antioxidant activity of the carrot being assessed changed together with the direction of changes in polyphenol content but not in line with the direction of changes in carotenoids.

Spatial and Temporal Variation of Carotenoids in Four Species ofTrentepohlia(Trentepohliales, Chlorophyta)

TrentepohliaMartius, a dominant green subaerial alga, grows abundantly in Shillong and imparts the yellow, red, and orange colours to most of the tree barks, walls, rocks, and electric poles because of high accumulation of carotenoids in their filaments. This study emphasised the seasonal changes in carotenoid content amongst four different species ofTrentepohlia, that is,T. diffracta,T. arborum,T. umbrina, andT. abietina, collected from four different substrata, that is, wall, rock, electric pole, and three types of tree bark (smooth, fissured, and rough). Quantitative estimation of different carotenoids, namely,β-cryptoxanthin, lutein, andβ-carotene, from HPLC chromatogram peak showed a significant seasonal variation in all of the four species andβ-carotene was threefold more in winter compared to summer. Amongst the selected species,T. diffractacollected from wall contained the highest amount ofβ-carotene both in summer and in winter, followed byT. arborumfrom rock andT. abietinafrom bark, and least amount was inT. umbrinacollected from electric pole. Comparing the carotenoid content inTrentepohlia abietinagrowing in different types of barks, sample from smooth bark had the highest amount of carotenoids both in summer and in winter, followed by the sample from rough bark, and lowest amount was noted in fissured bark.

Variations in the carotenoid and anthocyanin contents of Korean cultural varieties and home-processed sweet potatoes

The sweet potato is an important industrial crop and a source of food that contains useful dietary fiber and vitamins. Recently, orange- and purple-fleshed varieties have come under the spotlight due to their healthful components, carotenoids and anthocyanins, respectively. In this study, an HPLC-DAD method was applied to determine the carotenoid composition and content in nine Korean cultural varieties of sweet potato. Changes in carotenoid contents and composition were also observed during home-processing of an orange-fleshed cultivar with high carotenoid content (530±60μg/g of dry weight, DW as all-trans-β-carotene). A loss of the carotenoids was observed for all of the home-processing methods examined; the baked or boiled or steamed sweet potatoes had higher amounts of all-trans-β-carotene (246±34, 253±29 and 240±21μg/g DW, respectively) than pressure-cooked, sautéed and fried ones (194±21, 201±28 and 111±19μg/g DW, respectively). Interestingly, cis-isomer of the all-trans-β-carotene, 13Z-β-carotene was found in elevated amounts in all of the processed samples, particularly in baked, pressure-cooked and steamed sweet potatoes compared to control. Variations in anthocyanin content in the nine cultural varieties and home-processed sweet potatoes were also determined by an HPLC-DAD method.

Quality Evaluation of Enzyme Liquefied Papaya Juice Concentrate (PJC) Stored at Various Temperatures

This paper aims to study the effect of storage on enzyme liquefied papaya juice concentrate. Papaya pulp was liquefied using pectinase enzyme for juice extraction. The enzyme liquefied juice was subjected to concentration process by adopting Single Effect Tubular Evaporator (SETE) at 50°C with 24 inches vacuum. The Papaya Juice Concentrate (PJC), 68°brix was packed in high density polyethylene (HDPE) jars (200 g capacity) by leaving one inch head space and packed PJC jars were stored at various temperatures such as room temperature (RT, 29±1°C), low temperature (LT, 4°C) and frozen storage temperature (FT, -18°C). The PJC samples were analyzed for physicochemical characteristics and microbial load at 15 days intervals using methods reported in the literature. During storage, the PJC samples stored at RT showed significant increase in L, a and b values and browning as compared to LT and FT, while slight change in carotenoid content was observed. At the end of one month storage, the microbial counts (colony-forming unit/g) like total plate counts, yeast and molds counts and Staphylococcus counts in the PJC samples were low and Escherichia coli was absent. The study showed that enzymatic liquefaction can be used for better juice extraction from papaya and PJC prepared from this juice was physicochemically and microbiologically stable at various temperatures for one month.

Variations in carotenoid and anthocyanin compositions in sweet potato according to cultural varieties and home-processing methods

A high-performance liquid chromatography method was applied to determine the content of carotenoid composition in nine cultural varieties of sweet potato (Ipomoea batatas Lam.) and home-processed one by most common cooking methods. The changes in carotenoid contents and composition were also observed during home-processing of an orange-fleshed cultivar showing the highest carotenoid content (529.7µg/g DW as β-carotene). The loss of the carotenoids occurred through the all home-processing, and the baked or boiled or steamed sweet potato showed higher amount of β-carotene (245.8, 253.0 and 239.9µg/g DW) than pressure-cooked, sauteed and fried ones (194.1, 200.6 and 110.9µg/g DW). Interestingly, a cis-isomer of the all-trans-β-carotene, 13-cis-β-carotene was found in raised amounts in all processed samples, especially in boiled, steamed and pressure-cooked ones. The higher content of another isomer, 9-cis-β-carotene was detected in pressure-cooking and frying processed sweet potato than in the other samples. For a purple-fleshed cultivar, the variations in anthocyanin content during home-processing were also evaluated.
