Holocene climate regulates multiple sulfur isotope compositions of pyrite in the East China sea via sedimentation rate

Marine authigenic pyrite sulfur isotope (δ34Spyr) is one of the most important proxies for reconstructing remarkable changes of Earth's surface in deep time. Recent observations of unexpectedly large δ34Spyr variations in Quaternary sediments complicate the application of this isotopic proxy, and a mechanistic understanding of such variabilities is needed. In this study, we measured multiple sulfur isotopic compositions of pyrite in well dated sediments spanning the last 7500 years from the East China Sea (ECS) inner shelf to study the mechanism how Holocene climate controls pyrite sulfur isotopic compositions. We found that variabilities of pyrite multiple sulfur isotopes, δ34Spyr and Δ33Spyr (ranging from −38.3‰ to 2.3‰ and from −0.01‰ to 0.21‰, respectively) are significantly correlated with large fluctuations of sedimentation rates (0.03–2.09 cm/yr). In tandem with box model calculations and compiled data, the large δ34Spyr variations up to 40‰ are primarily ascribed to fluctuated anaerobic oxidation of methane coupled sulfate reduction (AOM-SR). Further analyses suggest that heightened distillation of interstitial sulfate, likely propelled by high methane flux coupled with increased sedimentation rates, can produce such negatively correlated δ34Spyr-Δ33Spyr pattern and large variations in δ34Spyr. Possible changes in isotope fractionation factors caused by varied cell-specific sulfate reduction rates result in a maximum δ34Spyr variation of <20‰ and therefore play a minor role in the large variation in δ34Spyr (>40‰). The major and minor isotopic compositions of pyrite align with sulfide produced through pure sulfate reduction, indicating little evidence of additional oxidative sulfur cycling. Sedimentation rates in the ECS are controlled by the East Asian winter monsoon that influences the sediment transport capacity from longshore currents on a multicentennial timescale, and also by the coupled East Asian summer monsoon that affects the sediment availability from the Yangtze River on a millennial timescale. Holocene climate changes alter sediment supplies at the ECS and therefore play a vital role in controlling multiple sulfur isotope compositions of sedimentary pyrite in this region. This study has the potential to provide new insights into δ34S excursions in seawater sulfate that occurred during periods of notable acceleration in sedimentation rates throughout the geological history.

Large sulfur isotope fractionation during abiotic consumption of hydrogen sulfide after cessation of bacterial growth in batch culture experiments of sulfate reducing bacteria

Microbial sulfate reduction is a key process in the oceanic sulfur cycle and imparts a large sulfur isotope fractionation. The mechanism of sulfur isotope fractionation during microbial sulfate reduction has been studied from the geochemical and biochemical aspects since the 1950s. Recently, however, the large sulfur isotope fractionation, exceeding 47‰, has been observed in some pure culture experiments of sulfate reducing bacteria and the tentative understanding of microbial sulfur isotope fractionation has not well explained the mechanism. Here we quantified growth phase dependent sulfur isotope fractionation of a type sulfate reducer Desulfovibrio desulfuricans (DSM642) and revealed that the magnitude of isotope fractionation increased from -13.4 ± 3.6‰ in early exponential phase to -65.9 ± 21.0‰ in later exponential phase having clear negative correlation with cell specific sulfate reduction rate. Our results show the importance of cells growth phase and states that control cell specific sulfate reduction rate and sulfur isotope fractionation. Although microbial sulfate reduction likely continued to stationary phase of cells, microbial sulfur isotope fractionation could not be quantified because of the decreasing total dissolved sulfide (ΣH2S: H2S, HS−, and S2−) concentration after stationary phase. A non-microbial ΣH2S-consuming reaction is interpreted to have occurred during the stationary phase and this had large sulfur isotope fractionations, -10.5 ± 1.1‰ and -45.6 ± 12.4‰. Besides, the non-microbial ΣH2S-consuming reaction decreased Δ33S’ value of the ΣH2S. While the end-product of the non-microbial ΣH2S-consuming reaction in the stationary phase remains unidentified, we observed precipitates of some sulfide minerals and organic sulfur in the stationary phase media. Regardless of the end-product, the reaction increases the sulfur isotopic composition of dissolved ΣH2S, which can account for the high sulfur isotopic compositions of ΣH2S compared to pyrite and organic sulfur observed in some modern marine sediments.

Black blooms-induced adaptive responses of sulfate reduction bacteria in a shallow freshwater lake

Decomposing cyanobacterial bloom-induced black blooms been seen as an issue in the management of freshwater ecosystems, but its effect on sulfate-reducing bacteria (SRB) in shallow freshwater lakes is not clear. The objective of this study is to present an in-depth investigation of black bloom effects on the activities and composition of SRB, as well as the interactions between SRB and other bacteria. Water and surface sediments samples were collected from a shallow freshwater lake during black and non-black blooms. Sulfate reduction rates (SRRs) in the water column were determined from the linear regression of sulfate depletion with time. Quantitative real-time polymerase chain reactions (qPCRs), targeting the dsrA gene and Illumina sequencing of 16S rDNA, were used to estimate the SRB population and SRB community structures, respectively. Our data indicate that although a higher abundance of SRB was responsible for the higher SRR in the bottom water (34.09 ± 2.37 nmol mL−1 day−1) than in the surface water (14.57 ± 2.91 nmol mL−1 day−1) during black blooms, cell-specific sulfate reduction rates (csSRRs) in the distinct water layers were not significantly different (P = 0.95), with the value of approximately 0.017 fmol cell−1 day−1. Additionally, Desulfomicrobium and Desulfovibrio were the two main genera of SRB in the water column during black bloom season, while Desulfobulbus, Desulfobacca and Desulfatiglans genera were identified in the sediments of both the black and non-black blooms in genera pools. Each SRB genus preferentially associated with bacteria for specific functions in the bacterial co-occurrence network, regardless of whether black booms occurred or not. These results extend our knowledge on the importance of SRB during black blooms and the adaptation of SRB to environmental changes in freshwater lakes.

Sedimentary pyrite sulfur isotope compositions preserve signatures of the surface microbial mat environment in sediments underlying low-oxygen cyanobacterial mats.

The sedimentary pyrite sulfur isotope (δ34 S) record is an archive of ancient microbial sulfur cycling and environmental conditions. Interpretations of pyrite δ34 S signatures in sediments deposited in microbial mat ecosystems are based on studies of modern microbial mat porewater sulfide δ34 S geochemistry. Pyrite δ34 S values often capture δ34 S signatures of porewater sulfide at the location of pyrite formation. However, microbial mats are dynamic environments in which biogeochemical cycling shifts vertically on diurnal cycles. Therefore, there is a need to study how the location of pyrite formation impacts pyrite δ34 S patterns in these dynamic systems. Here, we present diurnal porewater sulfide δ34 S trends and δ34 S values of pyrite and iron monosulfides from Middle Island Sinkhole, Lake Huron. The sediment-water interface of this sinkhole hosts a low-oxygen cyanobacterial mat ecosystem, which serves as a useful location to explore preservation of sedimentary pyrite δ34 S signatures in early Earth environments. Porewater sulfide δ34 S values vary by up to ~25‰ throughout the day due to light-driven changes in surface microbial community activity that propagate downwards, affecting porewater geochemistry as deep as 7.5cm in the sediment. Progressive consumption of the sulfate reservoir drives δ34 S variability, instead of variations in average cell-specific sulfate reduction rates and/or sulfide oxidation at different depths in the sediment. The δ34 S values of pyrite are similar to porewater sulfide δ34 S values near the mat surface. We suggest that oxidative sulfur cycling and other microbial activity promote pyrite formation in and immediately adjacent to the microbial mat and that iron geochemistry limits further pyrite formation with depth in the sediment. These results imply that primary δ34 S signatures of pyrite deposited in organic-rich, iron-poor microbial mat environments capture information about microbial sulfur cycling and environmental conditions at the mat surface and are only minimally affected by deeper sedimentary processes during early diagenesis.

Isotopic Fractionation Associated With Sulfate Import and Activation by Desulfovibrio vulgaris str. Hildenborough.

The use of stable isotopes to trace biogeochemical sulfur cycling relies on an understanding of how isotopic fractionation is imposed by metabolic networks. We investigated the effects of the first two enzymatic steps in the dissimilatory sulfate reduction (DSR) network – sulfate permease and sulfate adenylyl transferase (Sat) – on the sulfur and oxygen isotopic composition of residual sulfate. Mutant strains of Desulfovibrio vulgaris str. Hildenborough (DvH) with perturbed expression of these enzymes were grown in batch culture, with a subset grown in continuous culture, to examine the impact of these enzymatic steps on growth rate, cell specific sulfate reduction rate and isotopic fractionations in comparison to the wild type strain. Deletion of several permease genes resulted in only small (∼1‰) changes in sulfur isotope fractionation, a difference that approaches the uncertainties of the measurement. Mutants that perturb Sat expression show higher fractionations than the wild type strain. This increase probably relates to an increased material flux between sulfate and APS, allowing an increase in the expressed fractionation of rate-limiting APS reductase. This work illustrates that flux through the initial steps of the DSR pathway can affect the fractionation imposed by the overall pathway, even though these steps are themselves likely to impose only small fractionations.

The effect of temperature on sulfur and oxygen isotope fractionation by sulfate reducing bacteria (Desulfococcus multivorans).

Temperature influences microbiological growth and catabolic rates. Between 15 and 35°C the growth rate and cell specific sulfate reduction rate of the sulfate reducing bacterium Desulfococcus multivorans increased with temperature. Sulfur isotope fractionation during sulfate reduction decreased with increasing temperature from 27.2 ‰ at 15°C to 18.8 ‰ at 35°C which is consistent with a decreasing reversibility of the metabolic pathway as the catabolic rate increases. Oxygen isotope fractionation, in contrast, decreased between 15 and 25°C and then increased again between 25 and 35°C, suggesting increasing reversibility in the first steps of the sulfate reducing pathway at higher temperatures. This points to a decoupling in the reversibility of sulfate reduction between the steps from the uptake of sulfate into the cell to the formation of sulfite, relative to the whole pathway from sulfate to sulfide. This observation is consistent with observations of increasing sulfur isotope fractionation when sulfate reducing bacteria are living near their upper temperature limit. The oxygen isotope decoupling may be a first signal of changing physiology as the bacteria cope with higher temperatures.

Oxygen isotope effects during microbial sulfate reduction: applications to sediment cell abundances

The majority of anaerobic biogeochemical cycling occurs within marine sediments. To understand these processes, quantifying the distribution of active cells and gross metabolic activity is essential. We present an isotope model rooted in thermodynamics to draw quantitative links between cell-specific sulfate reduction rates and active sedimentary cell abundances. This model is calibrated using data from a series of continuous culture experiments with two strains of sulfate reducing bacteria (freshwater bacterium Desulfovibrio vulgaris strain Hildenborough, and marine bacterium Desulfovibrio alaskensis strain G-20) grown on lactate across a range of metabolic rates and ambient sulfate concentrations. We use a combination of experimental sulfate oxygen isotope data and nonlinear regression fitting tools to solve for unknown kinetic, step-specific oxygen isotope effects. This approach enables identification of key isotopic reactions within the metabolic pathway, and defines a new, calibrated framework for understanding oxygen isotope variability in sulfate. This approach is then combined with porewater sulfate/sulfide concentration data and diagenetic modeling to reproduce measured 18O/16O in porewater sulfate. From here, we infer cell-specific sulfate reduction rates and predict abundance of active cells of sulfate reducing bacteria, the result of which is consistent with direct biological measurements.

Diversity decoupled from sulfur isotope fractionation in a sulfate-reducing microbial community.

The extent of fractionation of sulfur isotopes by sulfate-reducing microbes is dictated by genomic and environmental factors. A greater understanding of species-specific fractionations may better inform interpretation of sulfur isotopes preserved in the rock record. To examine whether gene diversity influences net isotopic fractionation in situ, we assessed environmental chemistry, sulfate reduction rates, diversity of putative sulfur-metabolizing organisms by 16S rRNA and dissimilatory sulfite reductase (dsrB) gene amplicon sequencing, and net fractionation of sulfur isotopes along a sediment transect of a hypersaline Arctic spring. In situ sulfate reduction rates yielded minimum cell-specific sulfate reduction rates<0.3×10-15 moles cell-1 day-1 . Neither 16S rRNA nor dsrB diversity indices correlated with relatively constant (38‰-45‰) net isotope fractionation (ε34 Ssulfide-sulfate ). Measured ε34 S values could be reproduced in a mechanistic fractionation model if 1%-2% of the microbial community (10%-60% of Deltaproteobacteria) were engaged in sulfate respiration, indicating heterogeneous respiratory activity within sulfate-reducing populations. This model indicated enzymatic kinetic diversity of Apr was more likely to correlate with sulfur fractionation than DsrB. We propose that, above a threshold Shannon diversity value of 0.8 for dsrB, the influence of the specific composition of the microbial community responsible for generating an isotope signal is overprinted by the control exerted by environmental variables on microbial physiology.

Proteomic and Isotopic Response of Desulfovibrio vulgaris to DsrC Perturbation.

Dissimilatory sulfate reduction is a microbial energy metabolism that can produce sulfur isotopic fractionations over a large range in magnitude. Calibrating sulfur isotopic fractionation in laboratory experiments allows for better interpretations of sulfur isotopes in modern sediments and ancient sedimentary rocks. The proteins involved in sulfate reduction are expressed in response to environmental conditions, and are collectively responsible for the net isotopic fractionation between sulfate and sulfide. We examined the role of DsrC, a key component of the sulfate reduction pathway, by comparing wildtype Desulfovibrio vulgaris DSM 644T to strain IPFG07, a mutant deficient in DsrC production. Both strains were cultivated in parallel chemostat reactors at identical turnover times and cell specific sulfate reduction rates. Under these conditions, sulfur isotopic fractionations between sulfate and sulfide of 17.3 ± 0.5‰ or 12.6 ± 0.5‰ were recorded for the wildtype or mutant, respectively. The enzymatic machinery that produced these different fractionations was revealed by quantitative proteomics. Results are consistent with a cellular-level response that throttled the supply of electrons and sulfur supply through the sulfate reduction pathway more in the mutant relative to the wildtype, independent of rate. We conclude that the smaller fractionation observed in the mutant strain is a consequence of sulfate reduction that proceeded at a rate that consumed a greater proportion of the strains overall capacity for sulfate reduction. These observations have consequences for models of sulfate reducer metabolism and how it yields different isotopic fractionations, notably, the role of DsrC in central energy metabolism.

Influence of Phosphorus and Cell Geometry on the Fractionation of Sulfur Isotopes by Several Species of Desulfovibrio during Microbial Sulfate Reduction.

We investigated the influence of organic substrates and phosphate concentration on the rates of dissimilatory microbial sulfate reduction and the 34S/32S isotopic fractionation produced by several Desulfovibrio species. Our experiments corroborate the previously reported species-specific correlation between sulfur isotope fractionation and cell-specific sulfate reduction rates. We also identify cell size as a key factor that contributes to the species-effect of this correlation. Phosphate limitation results in larger cells and contributes to a small decrease in sulfur isotope fractionation concomitant with an apparent increase in cell-specific sulfate reduction rates. Sulfur isotope fractionation in phosphate-limited cultures asymptotically approaches a lower limit of approximately 5‰ as cell-specific sulfate reduction rates increase to >100 fmol cell−1 day−1. These experimental results test models that link the reversibilities of enzymatic steps in dissimilatory sulfate reduction to sulfur isotope fractionation and show that these models can provide consistent predictions across large variations in physiological states experienced by sulfate reducing bacteria.

Metabolic rates and sulfur cycling in the geologic record

In PNAS, Leavitt et al. (1) describe the shared dependence of cell-specific sulfate reduction rate and sulfur isotope fractionations on the availability of a single electron donor for the model sulfate reducer, Desulfovibrio vulgaris Hildenborough. These authors then use their findings in a unique way to calibrate past sedimentary sulfate reduction rates (SedSRR) using sulfur isotope data from sedimentary pyrite and sulfate. This approach reveals evidence for changes in SedSRR over the past 200 million years that played a critical role in determining sulfur isotope fractionations in marine sediments.

Effects of Iron and Nitrogen Limitation on Sulfur Isotope Fractionation during Microbial Sulfate Reduction

Sulfate-reducing microbes utilize sulfate as an electron acceptor and produce sulfide that is depleted in heavy isotopes of sulfur relative to sulfate. Thus, the distribution of sulfur isotopes in sediments can trace microbial sulfate reduction (MSR), and it also has the potential to reflect the physiology of sulfate-reducing microbes. This study investigates the relationship between the availability of iron and reduced nitrogen and the magnitude of S-isotope fractionation during MSR by a marine sulfate-reducing bacterium, DMSS-1, a Desulfovibrio species, isolated from salt marsh in Cape Cod, MA. Submicromolar levels of iron increase sulfur isotope fractionation by about 50% relative to iron-replete cultures of DMSS-1. Iron-limited cultures also exhibit decreased cytochrome c-to-total protein ratios and cell-specific sulfate reduction rates (csSRR), implying changes in the electron transport chain that couples carbon and sulfur metabolisms. When DMSS-1 fixes nitrogen in ammonium-deficient medium, it also produces larger fractionation, but it occurs at faster csSRRs than in the ammonium-replete control cultures. The energy and reducing power required for nitrogen fixation may be responsible for the reverse trend between S-isotope fractionation and csSRR in this case. Iron deficiency and nitrogen fixation by sulfate-reducing microbes may lead to the large observed S-isotope effects in some euxinic basins and various anoxic sediments.

Effect of electron donors on the fractionation of sulfur isotopes by a marine Desulfovibrio sp.

Sulfur isotope effects produced by microbial dissimilatory sulfate reduction are used to reconstruct the coupled cycling of carbon and sulfur through geologic time, to constrain the evolution of sulfur-based metabolisms, and to track the oxygenation of Earth’s surface. In this study, we investigate how the coupling of carbon and sulfur metabolisms in batch and continuous cultures of a recently isolated marine sulfate reducing bacterium DMSS-1, a Desulfovibrio sp ., influences the fractionation of sulfur isotopes. DMSS-1 grown in batch culture on seven different electron donors (ethanol, glycerol, fructose, glucose, lactate, malate and pyruvate) fractionates 34S/ 32S ratio from 6‰ to 44‰, demonstrating that the fractionations by an actively growing culture of a single incomplete oxidizing sulfate reducing microbe can span almost the entire range of previously reported values in defined cultures. The magnitude of isotope effect correlates well with cell specific sulfate reduction rates (from 0.7 to 26.1 fmol/cell/day). DMSS-1 grown on lactate in continuous culture produces a larger isotope effect (21–37‰) than the lactate-grown batch culture (6‰), indicating that the isotope effect also depends on the supply rate of the electron donor and microbial growth rate. The largest isotope effect in continuous culture is accompanied by measurable changes in cell length and cellular yield that suggest starvation. The use of multiple sulfur isotopes in the model of metabolic fluxes of sulfur shows that the loss of sulfate from the cell and the intracellular reoxidation of reduced sulfur species contribute to the increase in isotope effects in a correlated manner. Isotope fractionations produced during sulfate reduction in the pure culture of DMSS-1 expand the previously reported range of triple sulfur isotope effects ( 32S, 33S, and 34S) by marine sulfate reducing bacteria, implying that microbial sulfur disproportionation may have a smaller 33S isotopic fingerprint than previously thought.

A ‘rare biosphere’ microorganism contributes to sulfate reduction in a peatland

Methane emission from peatlands contributes substantially to global warming but is significantly reduced by sulfate reduction, which is fuelled by globally increasing aerial sulfur pollution. However, the biology behind sulfate reduction in terrestrial ecosystems is not well understood and the key players for this process as well as their abundance remained unidentified. Comparative 16S rRNA gene stable isotope probing in the presence and absence of sulfate indicated that a Desulfosporosinus species, which constitutes only 0.006% of the total microbial community 16S rRNA genes, is an important sulfate reducer in a long-term experimental peatland field site. Parallel stable isotope probing using dsrAB [encoding subunit A and B of the dissimilatory (bi)sulfite reductase] identified no additional sulfate reducers under the conditions tested. For the identified Desulfosporosinus species a high cell-specific sulfate reduction rate of up to 341 fmol SO42− cell−1 day−1 was estimated. Thus, the small Desulfosporosinus population has the potential to reduce sulfate in situ at a rate of 4.0–36.8 nmol (g soil w. wt.)−1 day−1, sufficient to account for a considerable part of sulfate reduction in the peat soil. Modeling of sulfate diffusion to such highly active cells identified no limitation in sulfate supply even at bulk concentrations as low as 10 μM. Collectively, these data show that the identified Desulfosporosinus species, despite being a member of the ‘rare biosphere’, contributes to an important biogeochemical process that diverts the carbon flow in peatlands from methane to CO2 and, thus, alters their contribution to global warming.

Temperature effect on the sulfur isotope fractionation during sulfate reduction by two strains of the hyperthermophilic Archaeoglobus fulgidus

Sulfur isotope fractionation during dissimilatory sulfate reduction by two strains of the thermophilic archaeon Archaeoglobus fulgidus (strains VC-16 and Z) was explored over the entire temperature range of growth. The optimal cell-specific sulfate reduction rate (14 fmol cell(-1) h(-1)) was found at 82-84 degrees C but growth was measured as low as 54 degrees C. The fractionation ranged between 0.52 per thousand and 27 per thousand, with largest fractionations were found at intermediate temperatures and the smallest fractionations at the lowest and highest temperatures. There was an inverse relationship between the cell-specific sulfate reduction rate and fractionation, and the cell-specific rate was a good indicator of the expected fractionations regardless of whether temperature or substrate concentrations controlled the rate. Comparison of the fractionation trend found in this study with similar measurements for seven other sulfate-reducers showed that sulfate-reducing organisms respond to temperature in three different ways and this correlated with their maximum fractionation value, but not with the cell-specific sulfate reduction rate. A sulfur isotope model was used to reproduce the observed variation of fractionation with temperature. This approach predicted the rate of internal sulfur transformations as having the major influence on the observed fractionations in the intermediate temperature range, whereas the exchange of sulfate across the cell membrane controls fractionation at low and high temperatures.

Oxygen isotope biogeochemistry of pore water sulfate in the deep biosphere: Dominance of isotope exchange reactions with ambient water during microbial sulfate reduction (ODP Site 1130)

Microbially mediated sulfate reduction affects the isotopic composition of dissolved and solid sulfur species in marine sediments. Experiments and field data show that the δ 18 O SO 4 2 - composition is also modified in the presence of sulfate-reducing microorganisms. This has been attributed either to a kinetic isotope effect during the reduction of sulfate to sulfite, cell-internal exchange reactions between enzymatically-activated sulfate (APS), and/or sulfite with cytoplasmic water. The isotopic fingerprint of these processes may be further modified by the cell-external reoxidation of sulfide to elemental sulfur, and the subsequent disproportionation to sulfide and sulfate or by the oxidation of sulfite to sulfate. Here we report δ 18 O SO 4 2 - values from interstitial water samples of ODP Leg 182 (Site 1130) and provide the mathematical framework to describe the oxygen isotope fractionation of sulfate during microbial sulfate reduction. We show that a purely kinetic model is unable to explain our δ 18 O SO 4 2 - data, and that the data are well explained by a model using oxygen isotope exchange reactions. We propose that the oxygen isotope exchange occurs between APS and cytoplasmic water, and/or between sulfite and adenosine monophosphate (AMP) during APS formation. Model calculations show that cell external reoxidation of reduced sulfur species would require up to 3000 mol/m 3 of an oxidant at ODP Site 1130, which is incompatible with the sediment geochemical data. In addition, we show that the volumetric fluxes required to explain the observed δ 18 O SO 4 2 - data are on average 14 times higher than the volumetric sulfate reduction rates (SRR) obtained from inverse modeling of the porewater data. The ratio between the gross sulfate flux into the microbes and the net sulfate flux through the microbes is depth invariant, and independent of sulfide concentrations. This suggests that both fluxes are controlled by cell density and that cell-specific sulfate reduction rates remain constant with depth.

Desulfobacter psychrotolerans sp. nov., a new psychrotolerant sulfate-reducing bacterium and descriptions of its physiological response to temperature changes

A psychrotrolerant acetate-oxidizing sulfate-reducing bacterium (strain akvb(T)) was isolated from sediment from the northern part of The North Sea with annual temperature fluctuations between 8 and 14 degrees C. Of the various substrates tested, strain akvb(T) grew exclusively by the oxidation of acetate coupled to the reduction of sulfate. The cells were motile, thick rods with round ends and grew in dense aggregates. Strain akvb(T) grew at temperatures ranging from -3.6 to 26.3 degrees C. Optimal growth was observed at 20 degrees C. The highest cell specific sulfate reduction rate of 6.2 fmol cell(-1) d(-1) determined by the (35)SO(2-)(40) method was measured at 26 degrees C. The temperature range of short-term sulfate reduction rates exceeded the temperature range of growth by 5 degrees C. The Arrhenius relationship for the temperature dependence of growth and sulfate reduction was linear, with two distinct slopes below the optimum temperatures of both processes. The critical temperature was 6.4 degrees C. The highest growth yield (4.3-4.5 g dry weight mol(-1) acetate) was determined at temperatures between 5 and 15 degrees C. The cellular fatty acid composition was determined with cultures grown at 4 and 20 degrees C, respectively. The relative proportion of cellular unsaturated fatty acids (e.g. 16:1omega7c) was higher in cells grown at 4 degrees C than in cells grown at 20 degrees C. The physiological responses to temperature changes showed that strain akvb(T) was well adapted to the temperature regime of the environment from which it was isolated. Phylogenetic analysis showed that strain akvb(T) is closest related to Desulfobacter hydrogenophilus, with a 16S rRNA gene sequence similarity of 98.6%. DNA-DNA-hybridization showed a similarity of 32% between D. hydrogenophilus and strain akvb(T). Based on phenotypic and DNA-based characteristics we propose that strain akvb(T) is a member of a new species, Desulfobacter psychrotolerans sp. nov.

A model for oxygen and sulfur isotope fractionation in sulfate during bacterial sulfate reduction processes

We present a model of bacterial sulfate reduction that includes equations describing the fractionation relationship between the sulfur and the oxygen isotope composition of residual sulfate (δ 34S SO4_residual, δ 18O SO4_residual) and the amount of residual sulfate. The model is based exclusively on oxygen isotope exchange between cell-internal sulfur compounds and ambient water as the dominating mechanism controlling oxygen isotope fractionation processes. We show that our model explains δ 34S SO4_residual vs. δ 18O SO4_residual patterns observed from natural environments and from laboratory experiments, whereas other models, favoring kinetic isotope fractionation processes as dominant process, fail to explain many (but not all) observed δ 34S SO4_residual vs. δ 18O SO4_residual patterns. Moreover, we show that a “typical” δ 34S SO4_residual vs. δ 18O SO4_residual slope does not exist. We postulate that measurements of δ 34S SO4_residual and δ 18O SO4_residual can be used as a tool to determine cell-specific sulfate reduction rates, oxygen isotope exchange rates, and equilibrium oxygen isotope exchange factors. Data from culture experiments are used to determine the range of sulfur isotope fractionation factors in which a simplified set of equations can be used. Numerical examples demonstrate the application of the equations. We postulate that, during denitrification, the oxygen isotope effects in residual nitrate are also the result of oxygen isotope exchange with ambient water. Consequently, the equations for the relationship between δ 34S SO4_residual, δ 18O SO4_residual, and the amount of residual sulfate could be modified and used to calculate the fractionation-relationship between δ 15N NO3_residual, δ 18O NO3_residual, and the amount of residual nitrate during denitrification.

Sulfur isotope fractionation during growth of sulfate-reducing bacteria on various carbon sources

Stable sulfur isotope fractionation during microbial sulfate reduction is a potential tool to estimate sulfate reduction rates at field sites. However, little is known about the influence of the utilized carbon source on the magnitude of sulfur isotope fractionation. To investigate this effect, both a pure culture (strain PRTOL1) and enrichment cultures from a petroleum hydrocarbon (PHC)–contaminated aquifer were used and grown in batch cultures on various carbon sources with an initial sulfate concentration of 1 mmol/L. As sole carbon sources the PHC components naphthalene, 1,3,5-trimethylbenzene, and heating oil (enrichment culture) and the organic acids acetate, pyruvate, benzoate, and 3-phenylpropionate (enrichment culture and PRTOL1) were used. Sulfate reduction rates of all cultures ranged from 6 ± 1 nmol cm −3 d −1 (enrichment culture grown on 1,3,5-trimethylbenzene) to 280 ± 6 nmol cm −3 d −1 (enrichment culture grown on pyruvate). Cell-specific sulfate reduction rates ranged from 1.1 × 10 −14 mol cell −1 d −1 (PRTOL1 grown on pyruvate) to 1.5 × 10 −13 mol cell −1 d −1 (PRTOL1 grown on acetate). Sulfur isotope enrichment factors (ε) for the enrichment culture ranged from 16.1‰ (3-phenylpropionate) to 34.5‰ (1,3,5-trimethylbenzene) and for PRTOL1 from 30.0‰ (benzoate) to 36.0‰ (pyruvate). Cultures of PRTOL1 always showed higher ε values than the enrichment culture when grown on the same carbon source due to culture-specific properties. Higher ε values were obtained when the enrichment culture was grown on PHC components than on organic acids. No relationship between ε values and cell-specific sulfate reduction rate existed when all data were combined. When comparing the magnitude of ε values determined in this laboratory study with ε values measured at contaminated and uncontaminated field sites, it becomes evident that a multitude of factors influences ε values at field sites and complicates their interpretation. The results of this study help us assess some of the general parameters that govern the magnitude of ε in sulfate-reducing environments.

Controls on stable sulfur isotope fractionation during bacterial sulfate reduction in Arctic sediments

Sulfur isotope fractionation experiments during bacterial sulfate reduction were performed with recently isolated strains of cold-adapted sulfate-reducing bacteria from Arctic marine sediments with year-round temperatures below 2°C. The bacteria represent quantitatively important members of a high-latitude anaerobic microbial community. In the experiments, cell-specific sulfate reduction rates decreased with decreasing temperature and were only slightly higher than the inferred cell-specific sulfate reduction rates in their natural habitat. The experimentally determined isotopic fractionations varied by less than 5.8‰ with respect to temperature and sulfate reduction rate, whereas the difference in sulfur isotopic fractionation between bacteria with different carbon oxidation pathways was as large as 17.4‰. Incubation of sediment slurries from two Arctic localities across an experimental temperature gradient from −4°C to 39°C yielded an isotopic fractionation of 30‰ below 7.6°C, a fractionation of 14‰ and 15.5‰ between 7.6°C and 25°C, and fractionations of 5‰ and 8‰ above 25°C, respectively. In absence of significant differences in sulfate reduction rates in the high and low temperature range, respectively, we infer that different genera of sulfate-reducing bacteria dominate the sulfate-reducing bacterial community at different temperatures. In the Arctic sediments where these bacteria are abundant the isotopic differences between dissolved sulfate, pyrite, and acid-volatile sulfide are at least twice as large as the experimentally determined isotopic fractionations. On the basis of bacterial abundance and cell-specific sulfate reduction rates, these greater isotopic differences cannot be accounted for by significantly lower in situ bacterial sulfate reduction rates. Therefore, the remaining isotopic difference between sulfate and sulfide must derive from additional isotope effects that exist in the oxidative part of the sedimentary sulfur cycle.
