Screening for probiotic properties and potential immunogenic effects of lactobacilli strains isolated from various food products.

Deceleration of disease progression and re-establishment of microbial balance in the gut is often achieved by application of lactobacilli strains. Their beneficial effects are associated with probiotic properties, which may be accompanied by immunomodulatory action at mucosal surfaces. To single out such strains, we screened almost three hundred lactobacilli isolates from eight genera and various food sources for acid and bile salt tolerance, adhesion to mucin as well as hemolytic activity and antibiotic susceptibility. Moreover, the immunomodulatory effects of cell-free supernatant (CFS) fractions of five lactobacilli strains were assessed using an in vitro cell line model. By our rationalized selection approach, we identified thirty-five strains with probiotic potential and biosafety features. Additionally, we showed that CFS from the Lactiplantibacillus L_4 strain downregulates proinflammatory cytokines IL-8 and IL-1β. In contrast, IL-8 expression was found to increase after treatment with CFSs of Lactiplantibacillus L_2 and L_5 and IL-1β was upregulated by CFSs of Lactiplantibacillus L_1 and Lactiplantibacillus L_3. Overall, our result delineate a rational approach of selecting lactobacilli strains for probiotic development to support the gut microbiota equilibrium and reinforce the host immune system.

Exploration of Antibiofilm Activities of Pediococci Strains Against Diarrheal Multi-Drug Resistant E.coli Strains

ABSTRACT The present study reports the potential of cell-free supernatant (CFS) from different probiotics for its antimicrobial and antibiofilm activity against Multi-Drug Resistant Diarrheal E. coli (MDR-DEC) strains. The CFS from Pediococcus acidilactici 100 exhibited maximum antimicrobial activity with Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of 23.33 mg/mL and 33.33 mg/mL, respectively. The CFS was able to inhibit the process of biofilm formation as well as eradication of preformed biofilms by 72–73%. A complete disintegration of aggregation of cells was observed in treated biofilms through advanced microscopic techniques. Atomic force microscopy (AFM) revealed a decrease in roughness (30%) and height of treated biofilms (2–17%). Organic acids, fatty acids, phenolic compounds and other derivatives were determined in CFS fractions using Liquid Chromatography Mass Spectrometry. The findings demonstrated the potential of CFS components from Pediococci strains against biofilms of MDR-DEC strains.

Enhancing the nutritional value and functional properties of mango pulp via lactic acid bacteria fermentation

The probiotic properties of four lactic acid bacteria (LAB) strains, Lacticaseibacillus paracasei OR1 and PP1 and Lacticaseibacillus rhamnosus MA3 and PP3, were evaluated. OR1 showed the highest levels of self-aggregation (61.27%) and coaggregation (17.70%) with Staphylococcus aureus. Compared to the crude cell-free supernatant (CFS) and ethyl acetate extract fraction, the water extract fraction of the CFS showed greater antimicrobial activity against Escherichia coli and Bacillus cereus. All LAB strains exhibited the highest levels of total carotenoids, phenolics, flavonoids and antioxidant activity in the CFS, suggesting that they can release water-soluble bioactive compounds into the growth medium. The phytochemical profile of Mahachanok mango, which is rich in bioactive compounds, was improved through LAB fermentation. LC–MS/MS analysis revealed 60 compounds, including phenolic acids (1.11-fold), organic acids (1.21-fold), and amino acid derivatives (1.28-fold), that were more abundant in the fermented Mahachanok mango pulp (FMP) samples than in the fresh samples. Interestingly, the antioxidant proline level increased 377.28-fold, and novel dipeptides and tripeptides were present in the FMP fermented with L. paracasei OR1. Mango pulp is a potential source of nutrients for LAB growth and metabolism; thus, LAB fermentation could be used to produce a functional food product rich in antioxidants and bioactive peptides.

Elemental sulfur enhances the anti-fungal effect of Lacticaseibacillus rhamnosus Lcr35

Lacticaseibacillus rhamnosus Lcr35 is a well-known bacterial strain whose efficiency in preventing recurrent vulvovaginal candidiasis has been largely demonstrated in clinical trials. The presence of sodium thiosulfate (STS) has been shown to enhance its ability to inhibit the growth of Candida albicans strains. In this study, we confirmed that Lcr35 has a fungicidal effect not only on the planktonic form of C. albicans but also on other life forms such as hypha and biofilm. Transcriptomic analysis showed that the presence of C. albicans induced a metabolic adaptation of Lcr35 potentially associated with a competitive advantage over yeast cells. However, STS alone had no impact on the global gene expression of Lcr35, which is not in favor of the involvement of an enzymatic transformation of STS. Comparative HPLC and gas chromatography-mass spectrometry analysis of the organic phase from cell-free supernatant (CFS) fractions obtained from Lcr35 cultures performed in the presence and absence of STS identified elemental sulfur (S0) in the samples initially containing STS. In addition, the anti-Candida activity of CFS from STS-containing cultures was shown to be pH-dependent and occurred at acidic pH lower than 5. We next investigated the antifungal activity of lactic acid and acetic acid, the two main organic acids produced by lactobacilli. The two molecules affected the viability of C. albicans but only at pH 3.5 and in a dose-dependent manner, an antifungal effect that was enhanced in samples containing STS in which the thiosulfate was decomposed into S0. In conclusion, the use of STS as an excipient in the manufacturing process of Lcr35 exerted a dual action since the production of organic acids by Lcr35 facilitates the decomposition of thiosulfate into S0, thereby enhancing the bacteria's own anti-fungal effect.

Mucin Binding Protein of Lactobacillus casei Inhibits HT-29 Colorectal Cancer Cell Proliferation.

Many Lactobacillus casei strains are reported to exhibit anti-proliferative effects on colorectal cancer cells; however, the mechanism remains largely unknown. While there has been considerable interest in bacterial small metabolites such as short chain fatty acids, prior reports suggested that larger-sized molecules mediate the anti-proliferative effect of L. casei. Here, other possible ways of communication between gut bacteria and its host are investigated. LevH1 is a protein displayed on the surface of L. casei, and its mucin binding domain is highly conserved. Based on previous reports that the cell-free supernatant fractions decreased colorectal cell proliferation, we cloned the mucin binding domain of the LevH1 protein, expressed and purified this mucin binding protein (MucBP). It has a molecular weight of 10 kDa, is encoded by a 250 bp gene, and is composed primarily of a β-strand, β-turns, and random coils. The amino acid sequence is conserved while the 36th amino acid residue is arginine in L. casei CAUH35 and serine in L. casei IAM1045, LOCK919, 12A, and Zhang. MucBP36R exhibited dose-dependent anti-proliferative effects against HT-29 cells while a mutation of 36S abolished this activity. Predicted structures suggest that this mutation slightly altered the protein structure, thus possibly affecting subsequent communication with HT-29 cells. Our study identified a novel mode of communication between gut bacteria and their host.

Lactate-driven macrophage polarization in the inflammatory microenvironment alleviates intestinal inflammation.

Lactate has long been considered an intermediate by-product of glucose metabolism. However, in recent years, accumulating evidence reveals that lactate has unique biological activities. In previous studies, lactate signaling was shown to inhibit inflammation. Furthermore, in vitro experiments have shown that lactate can promote the transformation of pro-inflammatory macrophages into anti-inflammatory macrophages. However, no in vivo studies have shown whether lactate can alleviate inflammation. RAW 264.7 macrophages were stimulated by LPS to induce an M1 phenotype, and cultured with low and high concentrations of lactate. The cells were then observed for phenotypic transformations and expression of inflammatory mediators and surface markers. The expression of inflammatory factors was also analyzed in the cell-free supernatant fraction. Further, a mouse model of DSS-induced colitis was established and treated with lactate. Colonic tissue injury was monitored by histopathological examinations. The in vitro experiments showed that lactate promoted the transformation of activated macrophages to M2 phenotype and decreased the expression of TLR4-mediated NF-κB signaling proteins and inflammatory factors. In the DSS-induced colitis mouse model, lactate promoted the phenotypic transformation of macrophages in colonic tissue, reduced inflammation and organ damage, inhibited the activation of TLR4/NF-κB signaling pathway, decreased the serum levels of pro-inflammatory factors, increased the expression of anti-inflammatory factors, promoted the repair of the intestinal mucosal barrier and reduced the severity of colitis. Lactate inhibits the TLR/NF-κB signaling pathway and the production of pro-inflammatory factors by promoting polarization of macrophages. In addition, lactate promotesthe repair of the intestinal mucosal barrier and protects intestinal tissue in inflammation. Furthermore, lactate is relatively safe. Therefore, lactate is a promising and effective drug for treating inflammation through immunometabolism regulation.

Antibacterial effect of cell-free supernatant fraction from Lactobacillus paracasei CH88 against Gardnerella vaginalis

Bacterial vaginosis (BV) is the most common vaginal infection in reproductive women, which is characterized by depleted level of lactic acid bacteria and overgrowth of anaerobes such as Gardnerella vaginalis spp. Lactic acid bacteria have been known to be beneficial for amelioration of BV, since they produce antimicrobial substances against G. vaginalis spp. The objectives of this study were to characterize different fractions of cell-free supernatant of Lactobacillus paracasei CH88 (LCFS) and investigate antibacterial activity of the LCFS fractions against G. vaginalis in-vitro and in-vivo. Antibacterial activity of the LCFS was stable during thermal treatment up to 120 °C for 30 min and maintained at pH ranging from 3.0 to 13.0 except pH 5.0. Fraction below 3 kDa of the LCFS partially lost its antibacterial activity after treatment with proteolytic enzymes. Precipitated protein fraction below 3 kDa of the LCFS (< 3 kDa LCFSP) inhibited the growth and biofilm formation of G. vaginalis. Treatment of L. paracasei CH88 or the < 3 kDa LCFSP attenuated G. vaginalis-induced BV in mice by inhibiting the growth of G. vaginalis, reducing exfoliation of vaginal epithelial cells, and regulating immune response. These results suggest that L. paracasei CH88 may have potential in ameliorating G. vaginalis-induced BV.

Comparison of efficacy and modes of action of two high-potential biocontrol &lt;i&gt;Bacillus&lt;/i&gt; strains and commercial biocontrol products against &lt;i&gt;Botrytis cinerea&lt;/i&gt; in table grapes

In table grapes (cv. Thomson Seedless), the antifungal activity against Botrytis cinerea was further analysed and compared between two high potential bacterial biological control agent (BCA) strains, Bacillus velezensis BUZ-14 and B. ginsengihumi S38. Two commercial biocontrol products (BPs), served as standards of comparison, Amylo-X® and Serenade Max®, also based on Bacillus BCA strains. The main mode of action quantified for all the strains was antibiosis due to hydrosoluble and volatile metabolites and their combinations. The BUZ-14 strain was the most active BCA strain, demonstrating significant disease reduction exceeding 60 % when used in the culture form grown in 863 medium, including living cells (LCs) and cell-free supernatant (CFS). Both BPs exhibited significantly reduced efficacy of their CFS fraction (&lt; 10 %) compared with that of the two BCA strains, confirming their high antibiosis potential. The novel methodology allowed us to demonstrate the significant effect of the BCA culture medium on volatilome (VOC) antagonist efficacy. The S38 strain achieved the highest disease reduction (90 %) owing to the greatest production of VOCs in the richest MOLP (Medium Optimum Lipopetide Production) culture medium, whereas grape juice was the least favourable medium for VOC efficacy for both bacterial strains (BUZ14 and S38). The overall poor activity of living Bacillus cells in all the BCA and BPs tested is discussed based on the low capacity of the BCA strains to grow in the berry. Then, the presence of living cells is also discussed with the possibility that these cells are not required in field applications of such BCA strains in this genus. Moreover, different environmental suboptimal conditions, including temperature (22 and 27 °C) and relative humidity (RH) (100 and 85–95 %), were tested, and BUZ-14 exhibited the highest Botrytis reduction at both temperatures and RH values. However, no significant differences were observed between temperatures or RH values for the same BCA. Further studies in vineyard conditions and applications, such as biofumigation or active packaging, will be performed to confirm the new findings reported in this investigation.

Construction of a Lactobacillus plantarum Strain Expressing the Capsid Protein of Porcine Circovirus Type 2d (PCV2d) as an Oral Vaccine.

Porcine circovirus type 2 (PCV2) is a pathogenic virus that causes high rates of porcine death, resulting in severe economic losses to the swine industry. In recent years, the prevalence of PCV2d genotype infection in pigs has increased, but most commercially available vaccines were developed against the PCV2a strain and do not ensure complete protection from PCV2d. Here, we first constructed an expression vector for the antigenic ORF2-encoded capsid protein of PCV2d (pLp3050-His6-tag-capsid). We then utilized Lactobacillus plantarum to express the protein at mucosal sites in orally vaccinated mice. After transducing L. plantarum with pLp3050-His6-tag-capsid, the expressed protein could be found in cell wall and cell-free supernatant fractions by Western blotting. Using flow cytometry, we found that L. plantarum cells with surface-displayed capsid protein increased with time after SppIP induction. Finally, mice that were orally immunized 18 times with capsid-expressing L. plantarum showed increased levels of capsid-specific sIgA and virus neutralizing activity at mucosal sites, suggesting mucosal immunity had been stimulated by the vaccine. Overall, our findings demonstrate the feasibility and utility of a PCV2d-based vaccine, which may be of great value in porcine agriculture.

Magnaporthe oryzae chrysovirus 1 strain D confers growth inhibition to the host fungus and exhibits multiform viral structural proteins

A Japanese isolate of Magnaporthe oryzae is infected by Magnaporthe oryzae chrysovirus 1-D (MoCV1-D), which is classified in cluster II of the family Chrysoviridae. The genome of MoCV1-D consists of five dsRNAs. dsRNAs 1–4 show high identity with those of related MoCV1 viruses, whereas dsRNA5 shows relatively low identity and is sometimes deleted during virus propagation. MoCV1-D causes growth inhibition of its host fungus, and the protein encoded by its dsRNA4 impairs cell growth when expressed in yeast cells. It also causes abnormal pigmentation and colony albinization, and we showed that these phenotypes are associated with reduced accumulation of the melanin biosynthesis intermediate scylatone. MoCV1-D exhibits multiform viral structural proteins during prolonged culture. The original host isolate is co-infected with MoCV1-D, a victorivirus, and a partitivirus, and these mycoviruses are detected in cell-free supernatant fractions after prolonged liquid culturing. Hyphal fusion experiments demonstrated that MoCV1-D is transmissible via anastomosis.

Lactobacillus plantarum USM8613 Aids in Wound Healing and Suppresses Staphylococcus aureus Infection at Wound Sites.

This study aimed to elucidate the targets and mechanisms of anti-staphylococcal effects from bioactive metabolites produced by lactic acid bacteria. We aimed to better understand the safety and efficacy of these bioactive metabolites in in vivo systems, typically at topical sites. The cell-free supernatant and protein-rich fraction from Lactobacillus plantarum USM8613 inhibited staphyloxanthin biosynthesis, reduced (p < 0.05) the cell number of Staphylococcus aureus by 106CFU/mL and reduced biofilm thickness by 55% in S. aureus-infected porcine skins. Genome-wide analysis and gene expression analysis illustrated the production of several plantaricins, especially the plantaricins EF and JK that enhanced the anti-staphylococcal effects of L. plantarum USM8613. In vivo data using rats showed that the protein-rich fraction from L. plantarum USM8613 exerted wound healing properties via direct inhibition of S. aureus and promoted innate immunity, in which the expression of β-defensin was significantly (p < 0.05) upregulated by 3.8-fold. The protein fraction from L. plantarum USM8613 also significantly enhanced (p < 0.05) the production of cytokines and chemokines through various stages of wound recovery. Using ∆atl S. aureus, the protein-rich fraction from L. plantarum USM8613 exerted inhibitory activity via targeting the atl gene in S. aureus. Taken altogether, our present study illustrates the potential of L. plantarum USM8613 in aiding wound healing, suppressing of S. aureus infection at wound sites and promoting host innate immunity.

Daucus carrota L. - seed germination and natural infection by Fusarium spp. affected by Pseudomonas spp.

The inhibitory effect of indigenous Pseudomonas strains on Fusarium spp. isolated from seeds of a new variety of carrot - ?Vizija? and their effects on seed germination were observed. Pseudomonas spp. strains were applied as bacterial suspensions (culture, 106 CFU mL-1) and cell-free supernatant (CFSa -106 and CFSb -108 CFU mL-1) fractions by sowing seeds during 7 (I) and 14 (II) days of incubation. The germination of control ?Vizija? seeds was 19% after the first and 40% after the second incubation period. Pseudomonas chlororaphis Q16 strain exhibited statistically significant increases in seed germination with all applied fractions and incubation periods, showing values of 43-62% and 55.5-91%, i.e. increments of 24-43% and 15.5-51% compared to the control, after 7 and 14 days of incubation, respectively. P. chlororaphis K35 showed lower but significantly different seed germination values (38-67%) for all variants, except for 7 days old culture. Pseudomonas sp. Ek1 had weaker seed germination potential, showing statistically significant increment only for CFSa,b (I) and CFSb (II) fractions. All tested Pseudomonas strains inhibited the growth of three Fusarium species isolated from ?Vizija? seeds: F. solani, F. oxysporum and F. subglutinans. Natural infection was observed in 20% (I) and 54% (II) of ?Vizija? seeds. P. chlororaphis K35 was the most efficient antifungal strain, reducing seed infection 97.5-100%, followed by Q16 with 95-100%, showing no statistically significant mutual difference. Pseudomonas sp. Ek1 showed a weaker antifungal activity and reduced seed infection by 85-96.75%. The application of P. chlororaphis Q16 and K35 as strains effective in improvement of carrot seed germination and growth inhibition of the seed pathogens F. solani, F. oxysporum and F. subglutinans, can be further tested in carrot production for more beneficial effects.

The Effect of Bacillus amyloliquefaciens subsp. plantarum IMV B-7524 Strain Exometabolites on the Induction of Defense Reactions in Winter Wheat Plants

To study the effect of antibiotic exometabolites of Bacillus amyloliquefaciens subsp. plantarum IMV B-7524 strain on the induction of defense responses in young plants of winter wheat. Microbiological, biochemical, phytopathological, statistical. It was found that, when pre-watering winter wheat plants with salicylic acid, the cell-free culture supernatant and lipopeptide fraction of IMV B-7524 strain exometabolites during the further infection with phytopathogen Bipolaris sorokiniana, the damaged surface area of leaves decreased more than 10 times compared to the control. It was shown that in the roots and shoots of winter wheat plants L-phenylalanine ammonia-lyase (PAL) activity increased about 3 times in 2 hours after treatment. Reduction of the damaged surface area of leaves by the action of fungal phytopathogen and change of PAL activity in plants indicate the induction of defense responses under the influence of antibiotic exometabolites of B. amyloliquefaciens subsp. plantarum IMV B-7524 strain.

Immune Activation in Patients With Irritable Bowel Syndrome

We set out to test the hypothesis that irritable bowel syndrome (IBS) is characterized by an augmented cellular immune response with enhanced production of proinflammatory cytokines. We further aimed to explore whether symptoms and psychiatric comorbidity in IBS are linked to the release of proinflammatory cytokines. We characterized basal and Escherichia coli lipopolysaccharide (LPS)-induced cytokine production in peripheral blood mononuclear cells (PBMCs) from 55 IBS patients (18 mixed-, 17 constipation-, 20 diarrhea-predominant) and 36 healthy controls (HCs). PBMCs were isolated by density gradient centrifugation and cultured for 24 hours with or without (1 ng/mL) LPS. Cytokine production (tumor necrosis factor [TNF]-alpha, interleukin [IL]-1beta, and IL-6) was measured by enzyme-linked immunosorbent assay. Abdominal symptoms and psychiatric comorbidities were assessed by using the validated Bowel Disease Questionnaire and the Hospital Anxiety and Depression Scale. IBS patients showed significantly (P < .017) higher baseline TNF-alpha, IL-1beta, IL-6, and LPS-induced IL-6 levels compared with HCs. Analyzing IBS subgroups, all cytokine levels were significantly (P < .05) higher in diarrhea-predominant IBS (D-IBS) patients, whereas constipation-predominant IBS patients showed increased LPS-induced IL-1beta levels compared with HCs. Baseline TNF-alpha and LPS-induced TNF-alpha and IL-6 levels were significantly higher in patients reporting more than 3 bowel movements per day, urgency, watery stools, and pain associated with diarrhea compared with patients without these symptoms (all P < .05). LPS-induced TNF-alpha production was associated significantly (r = 0.59, P < .001) with anxiety in patients with IBS. Patients with D-IBS display enhanced proinflammatory cytokine release, and this may be associated with symptoms and anxiety.

Conventional and real-time polymerase chain reaction assessment of the fate of transgenic DNA in sheep fed Roundup Ready®rapeseed meal

Conventional and real-time PCR were used to detect transgenic DNA in digesta, faeces and blood collected from six ruminally and duodenally cannulated sheep fed forage-based (F) or concentrate-based (C) diets containing 15% Roundup Ready (RR) rapeseed meal (n 3). The sheep were adapted for 14 d to F or C diets containing non-GM rapeseed, then fed the RR diets for 11 d. On day 12, they were switched back to non-GM diets for a further 11 d. Ruminal and duodenal fluids (RF, DF) and faecal samples were collected at 3 or 4 h intervals over the 4 d immediately following the last feeding of GM diets. DNA was isolated from whole RF and DF, from the cell-free supernatant fraction, and from culture fermentation liquid. Blood was collected on days 1, 5 and 9 of feeding the RR rapeseed meal. The 1363 bp 5-enolpyruvylshikimate-3-phosphate synthase transgene (epsps) was quantifiable in whole RF and DF for up to 13 h, and a 108 bp epsps fragment for up to 29 h. Transgenic DNA was not detectable in faeces or blood, or in microbial DNA. Diet type (F v. C) did not affect (P>0.05) the quantity of transgenic DNA in digesta. More (P<0.05) transgenic DNA was detected in RF than in DF, but there was an interaction (P<0.05) between sample type and collection time. In supernatant fractions from RF and DF, three different fragments of transgenic DNA ranging in size from 62 to 420 bp were not amplifiable.

UV Irradiation and Desiccation Modulate the Three-dimensional Extracellular Matrix of Nostoc commune (Cyanobacteria)

Cyanobacterium Nostoc commune can tolerate the simultaneous stresses of desiccation, UV irradiation, and oxidation. Acidic WspA, of approximately 33.6 kDa, is secreted to the three-dimensional extracellular matrix and accounts for greater than 70% of the total soluble protein. The wspA gene of N. commune strain DRH1 was cloned and found in a single genomic copy, in a monocistronic operon. Transcription of wspA and sodF (superoxide dismutase), and synthesis and secretion of WspA, were induced upon desiccation or UV-A/B irradiation of cells. Recombinant WspA binds the UV-A/B absorbing pigment scytonemin through non-covalent interactions. WspA peptide polymorphism, and heterogeneity of multiple wspA sequences within cells of a single colony, account for distinct WspA isoforms. WspA has no similarity to entries in the sequence databases and wspA, a possible xenolog, is restricted to a subset of strains in the "form species" N. commune characterized through group I intron phylogeny. We hypothesize that WspA plays a central role in the global stress response of N. commune through modulation of the structure and function of the three-dimensional extracellular matrix, particularly the transport, distribution, and/or macromolecular architecture of mycosporine and scytonemin UV-A/B absorbing pigment complexes.

Fluorescence assays for F-pili and their application

Conjugative pili are extracellular filaments elaborated by Gram-negative bacteria expressing certain type IV secretion systems. They are required at the earliest stages of conjugal DNA transfer to establish specific and secure cell-cell contacts. Conjugative pili also serve as adsorption organelles for both RNA and DNA bacteriophages. Beyond these facts, the structure, formation and function of these filaments are poorly understood. This paper describes a rapid, quantitative assay for F-pili encoded by the F plasmid type IV secretion system. The assay is based on the specific lateral adsorption of icosahedral RNA bacteriophage R17 by F-pili. Bacteriophage particles conjugated with a fluorescent dye, Alexa 488, and bound to F-pili defined filaments visible by immunofluorescence microscopy. F-pili attached to F+ cells and free F-pili were both visible by this method. For quantification, cell-bound bacteriophage were separated from free bacteriophage particles by sedimentation and released by suspending cell pellets in 0.1 % SDS. Fluorescence in cell-free supernatant fractions was measured by fluorometry. The authors present a characterization of this assay and its application to F-pilus formation by cells carrying mutations in the gene for the F-pilus subunit F-pilin. Each mutation introduced a cysteine, which F-pilin normally lacks, at a different position in its primary structure. Cysteine residues in the N-terminal domain I abolished filament formation as measured by fluorescent R17 binding. This was confirmed by measurements of DNA donor activity and filamentous DNA bacteriophage infection. With one exception (G53C), cysteines elsewhere in the F-pilin primary structure did not abolish filament formation, although some mutations differentially affected F-pilus functions.

Effect of feed additives on in vitro and in vivo rumen characteristics and frothy bloat dynamics in steers grazing wheat pasture

Research was conducted to determine the potential efficacy of feed additives (FA) to mitigate frothy bloat in yearling cattle grazing wheat pasture. Two experiments were conducted to: (1) quantify in vitro effect of FA on total rumen and methane gas production and foam potential and (2) quantify the influence of FA on rumen protein characteristics, bloat potential and weight gain of steers grazing wheat pasture. In Exp 1, duplicated analyses of in vitro gas production were measured as sequential plunger displacement (cc) at 0, 1, 2, 3, 4, 5, and 6 h incubation periods. In vitro rumen foam production and strength was measured at 0, 2, and 6 h incubation of minced wheat forage. In Exp 2, eight ruminally cannulated steers (386 ± 35.8 kg/steer) were randomly allocated to one of four FA treatments that included control, monensin, poloxalene, and condensed tannins (CT). Treatments were administered daily through rumen cannulla as pre-mixes with a mixed ration (300 mg/steer/day; as-fed basis). Steers grazed on wheat during a 2-week adaptation period prior to data collection from 05 March to 12 April, 2004. Rumen contents were collected 2 h post-FA infusion (1030 to 1130 h) on day −5, 0, 5, 15, and 22. Cattle were weighed at 28-days intervals. Bloat was visually scored weekly. In Exp 1, in vitro ruminal gas and methane gas production per gram of forage were similar between control and poloxalene, but were lower for monensin ( P<0.01) and CT ( P<0.05) treatments. The lowest ( P<0.01) in vitro foam strength occurred with the addition of poloxalene and the highest occurred in control and monensin treatments. CT was intermediate in rumen foam strength. Mean bloat score was lower in poloxalene and CT than that in monensin and control treatments. Among six-rumen protein components assayed, steers receiving CT after 10, 15, and 22 day had greater protein concentrations in whole rumen content, particulate matter, cheese-cloth filtrate, and protozoa and plant particle fractions than steers fed other treatments. Bacterial and cell-free supernatant protein fractions were comparable among treatments. Ruminal dry matter (DM) content, cell-free supernatant, protozoal and bacterial fractions were similar between the bloated and non-bloated steers. In bloated animals, ruminal pH was lower, and whole rumen content and cheese-cloth filtrate protein fractions tended to be greater ( P=0.08) than in non-bloated animals. Animal average daily gain (ADG) was not affected by FA during experimental period. The results of this trial suggest that wheat pasture bloat was associated with dietary protein and low ruminal pH. Feeding FA can be used to decrease either ruminal gas or foam production or both, and it may protect against the incidence of frothy bloat.

Spreading factors of Mycoplasma alligatoris, a flesh-eating mycoplasma.

Mycoplasma alligatoris causes lethal invasive disease of alligators and caimans. A homolog of the nagH gene, encoding a hyaluronidase secreted by Clostridium perfringens, and a C. perfringens hyaluronidase nagI or nagK pseudogene were discovered in the M. alligatoris genome. The nagH gene was detected by PCR in the closest relative of M. alligatoris, Mycoplasma crocodyli, but not in 40 other species representing the Mycoplasma hominis, Mycoplasma pneumoniae, and Spiroplasma phylogenetic clusters. The hyaluronidase activity in the cellular fraction of M. alligatoris and M. crocodyli SP4 broth cultures was equivalent to 10(-16) U of Streptomyces hyalurolyticus hyaluronidase CFU(-1). Negligible activity was present in the cell-free supernatant fraction. No chondroitinase activity was detected. There is also a novel homolog of the nanI gene, which encodes a sialidase secreted by C. perfringens, in the M. alligatoris genome. The signature YRIP and SXDXGXTW motifs and catalytic residues of the clostridial sialidase are conserved in the mycoplasmal gene, but the leader sequence necessary for its secretion by C. perfringens is absent. The gene was not detected by PCR in any other mycoplasma. Potent cell-associated sialidase activity was present in M. alligatoris colonies on agar but not in the cell-free supernatants of broth cultures or in M. crocodyli. The presence of hyaluronidase and sialidase in M. alligatoris is consistent with the rapid invasiveness and necrotizing effects of this organism, and the lack of sialidase in M. crocodyli is consistent with its comparatively attenuated virulence. This genetic and biochemical evidence suggests that the spreading factors hyaluronidase and sialidase, a combination unprecedented in mycoplasmas, are the basis of the virulence of M. alligatoris.

The cellulosome system of Acetivibrio cellulolyticus includes a novel type of adaptor protein and a cell surface anchoring protein.

A scaffoldin gene cluster was identified in the mesophilic cellulolytic anaerobe Acetivibrio cellulolyticus. The previously described scaffoldin gene, cipV, encodes an N-terminal family 9 glycoside hydrolase, a family 3b cellulose-binding domain, seven cohesin domains, and a C-terminal dockerin. The gene immediately downstream of cipV was sequenced and designated scaB. The protein encoded by this gene has 942 amino acid residues and a calculated molecular weight of 100,358 and includes an N-terminal signal peptide, four type II cohesions, and a C-terminal dockerin. ScaB cohesins 1 and 2 are very closely linked. Similar, but not identical, 39-residue Thr-rich linker segments separate cohesin 2 from cohesin 3 and cohesin 3 from cohesin 4, and an 84-residue Thr-rich linker connects the fourth cohesin to a C-terminal dockerin. The scaC gene downstream of scaB codes for a 1,237-residue polypeptide that includes a signal peptide, three cohesins, and a C-terminal S-layer homology (SLH) module. A long, ca. 550-residue linker separates the third cohesin and the SLH module of ScaC and is characterized by an 18-residue Pro-Thr-Ala-Ser-rich segment that is repeated 27 times. The calculated molecular weight of the mature ScaC polypeptide (excluding the signal peptide) is 124,162. The presence of the cohesins and the conserved SLH module implies that ScaC acts as an anchoring protein. The ScaC cohesins are on a separate branch of the phylogenetic tree that is close to, but distinct from, the type I cohesins. Affinity blotting with representative recombinant probes revealed the following specific intermodular interactions: (i) an expressed CipV cohesin binds selectively to an enzyme-borne dockerin, (ii) a representative ScaB cohesin binds to the CipV band of the cell-free supernatant fraction, and (iii) a ScaC cohesin binds to the ScaB dockerin. The experimental evidence thus indicates that CipV acts as a primary (enzyme-recognizing) scaffoldin, and the protein was also designated ScaA. In addition, ScaB is thought to assume the role of an adaptor protein, which connects the primary scaffoldin (ScaA) to the cohesin-containing anchoring scaffoldin (ScaC). The cellulosome system of A. cellulolyticus thus appears to exhibit a special type of organization that reflects the function of the ScaB adaptor protein. The intercalation of three multiple cohesin-containing scaffoldins results in marked amplification of the number of enzyme subunits per cellulosome unit. At least 96 enzymes can apparently be incorporated into an individual A. cellulolyticus cellulosome. The role of such amplified enzyme incorporation and the resultant proximity of the enzymes within the cellulosome complex presumably contribute to the enhanced synergistic action and overall efficient digestion of recalcitrant forms of cellulose. Comparison of the emerging organization of the A. cellulolyticus cellulosome with the organizations in other cellulolytic bacteria revealed the diversity of the supramolecular architecture.
