Frozen cultured sheets of epidermal keratinocytes in reepithelialization and repair of the cornea after photorefractive keratectomy

Purpose To determine whether frozen cultured sheets of human allogeneic epidermal keratinocytes (CEAK) improved wound repair after experimental corneal ablation by photorefractive keratectomy (PRK). Setting Hospital “Luis Sanchez Bulnes” de la Asociación para Evitar la Ceguera en Mexico, I.A.P, and Department of Cell Biology, CINVESTAV-IPN, Mexico City, Mexico. Methods Transepithelial PRK was performed in the right eye of male albino rabbits to obtain a 112 μm deep and 6.0 mm diameter ablation zone. In 17 eyes, the ablations were covered with frozen CEAK; in 11 eyes, the ablations were covered with a disposable contact lens without the cultured sheets; and in the control group (13 eyes), the ablations were not covered. Subepithelial fibrosis and reepithelialization of the ablated zone were evaluated in serial paraffin-embedded tissue sections from all wounds. Results Treatment with CEAK reduced fibroblast proliferation and the inflammatory response beneath the ablated zone and produced better organization of the newly formed epithelium by eliminating significant hyperplasia or discontinuities in the periodic acid Shiff-stained basement membrane. It also led to accelerated reepithelialization. Conclusions The use of frozen CEAK as a biologically active wound dressing improved tissue repair at 1 month in corneas ablated by transepithelial PRK in the male albino rabbit model. Treatment with CEAK could improve the outcome of PRK in humans.

New JCS Editor

We are delighted to announce the appointment of Kathleen Green as an Editor of Journal of Cell Science. Currently Joseph L. Mayberry Professor of Pathology and Toxicology at Northwestern University Medical School, Kathy is a leading figure in the field of cell adhesion. Her research career started at Washington University in St Louis, where she obtained a PhD in Cell and Developmental Biology in 1982. Kathy then moved to the Department of Cell Biology at Northwestern to pursue post-doctoral research on intermediate filaments. She subsequently joined the Departments of Pathology and Dermatology, where for several years a major focus of her research has been the structure and functions of desmosomes. Indeed, her work has been instrumental in illuminating the biology of plakins, a family of modular proteins that link cytoskeletal networks with adhesive junctions such as desmosomes and hemidesmosomes.Kathy has been a regular contributor to and reviewer for JCS for many years and joined the Editorial Board of the journal in 1992. On behalf of all the Editors and staff at JCS, we welcome Kathy to the editorial team and look forward to working with her.

A pod of botanists, an affinity of pharmacologists, an aggregate of structural biologists? No, the collective noun is 'department' by Caveman.

When you see a number of geese together you know that the collective noun is a ‘gaggle’; or when you look into the ocean and see fish swimming together you know that the collective noun is a ‘shoal’. But is the collective noun for a group of botanists a ‘pod’, or molecular biologists a ‘clone’ (two molecular biologists would, of course, be a ‘sub-clone’), or biochemists a ‘reaction’, or structural biologists an ‘aggregate’ (the very best, however, would be referred to as a ‘lattice’), or pharmacologists an ‘affinity’, or zoologists a ‘cage’? While these are interesting possibilities, and I could go on (a ‘network’ of neurobiologists, an ‘infection’ of virologists, a ‘plaque’ of bacteriologists?), the traditional collective noun for all groupings of scientists is ‘department’.Attempts have been made to specify ‘department’ for use as a collective noun for unique groupings of scientists by adding a simple descriptor - for example, Department of Biochemistry, Department of Genetics, or Department of Developmental Biology. Alternatively, the collective noun has been sub-specialized - for example, Department of Molecular Pharmacology. Or, an amalgam of collective nouns has been used - for example, Department of Cell Biology and Anatomy.The reason that these corruptions of ‘department’ do not work as collective nouns for specific scientists is that the groupings do not reflect the types of scientist in them. For example, Departments of Biochemistry are full of molecular biologists, developmental biologists, cell biologists - you name the speciality, and there is probably a Department of Biochemistry somewhere that has one - oh, and biochemists. When did you hear of a brood of hens containing a stray wildebeest, or a flock of starlings that contained a formation (a ‘squadron’?), of flying pigs?When ‘department’ is further sub-specialized (Molecular Pharmacology), there is really no real change in the motley grouping of scientists that make up the ‘department’. They did not suddenly add a ‘sub-clone’ of molecular biologists. Rather it is an attempt to make the old (Pharmacology) sound more modern (Molecular).Similarly, an amalgam of collective names (Cell Biology and Anatomy) does not necessarily reflect the inclusion of cell biologists into the old collective, but rather the attempt to make the old, original department (Anatomy) more attractive to students (Cell Biology?). In addition, other amalgams (Molecular, Cell and Developmental Biology) reflect the absorption of previous departments into a single collective (why not just call it ‘The Department’ when no other sub-departments remain?).It is the very fact that each ‘department’ comprises a group of scientists with broad and diverse interests that defies their collection under a more specialized noun. It is also what makes the ‘department’ so important as a collective. In the end, it is the scientists in the ‘department’ who define the style, teaching, training and research directions of the ‘department’ - not the (sub)name per se. As a cell biologist working on the actin cytoskeleton, would you benefit most by being surrounded by a group of other cell biologists working on the actin cytoskeleton or by a broad group of scientists that included molecular biologists, developmental biologists and biochemists? The answer depends a great deal on how parochial you are in terms of scientific interests and whether you need a security blanket around you of scientists who think like you, and identify with the same problems and research interests as you, whether you enjoy being stimulated by a smörgåsbord of science that, if you are lucky, will impact on the way that you think about your own work and move it in diverse directions, and on whether you like being challenged to convince scientists in very different research areas that your science is important and interesting. Personally, I like the presence of a diverse group of scientists around me that is summed up as a ‘department’ of scientists.Despite my reluctance to use a collective noun other than ‘department’ for scientists, the following sound not only useful but also rather intriguing: a ‘clone of pods’ (molecular botanists), a ‘cage of infections’ (animal virologists), a ‘reaction of plaques’ (biochemical bacteriologists) and an ‘affinity network’ (pharmaco-neurobiologists). By the way, I have from time-to-time belonged to a correlate, a reaction, an aggregate and a hereditary of scientists.

Abstracts presented at ISAC European International Conference for Flow Cytometry and Image Analysis, Epona, Hungary, May 13-16, 1999.

To facilitate research cooperation among the nations of Central and Eastern Europe, ISAC supported the Epona Cytometry Conference in eastern Hungary near Debrecen and the Hortobagy National Park. This was the first ISAC sponsored conference held in Hungary. The 150 attendees included scientists from Hungary, Austria, the Czech Republic, Croatia, Slovenia, Romania, Poland, Ukraine, and Russia. There was also representation from Western Europe, North America, and Japan. Presentations and posters were of typically high ISAC quality. In addition, the Epona meeting attracted an impressive selection of commercial exhibits that provided contacts and improved the awareness of advanced analytical cytology for the attendees. The Epona conference was organized with Hungarian passion and skill by Professor Sandor Damjanovich, Dr. Janos Szollosi, and the members of the Biophysics and Cell Biology Department of the Medical School of Debrecen. The Department has been active in cytometry for over 20 years, and in hosting the conference, it also celebrated the 20th anniversary of its installment of the first cell sorter in Eastern Europe. The successful meeting demonstrates that Hungarians are strong contenders for the organization of future ISAC meetings. During my 1999 Fulbright Scholarship in Professor Damjanovich's Department, I traveled extensively and can verify that there are other unique sites in Hungary—Sopron, Pecs, Lake Balaton, and Budapest—for ISAC meetings. The site of the conference was on the Hungarian Puszta or plain near the Hortobagy National Park, which offered tours to observe “Hungarian Cowboys,” long-horned cattle, twisted-horn sheep, hairy pigs, and water buffalo, which were brought into the Carpathian Basin by the Magyars. Located in beautiful grasslands on the edge of the Park, the Epona Village was an excellent site for the meeting and is distinctly Hungarian. The timing of the conference was dramatic. Hungary was admitted to NATO in April and the Kosovo conflict with Yugoslavia began a few weeks later. During the conference, NATO air raids into Kosovo were being conducted from bases in southern Hungary. There was much concerned discussion among the attendees of the Kosovo situation. All regretted the loss of life, and all shared the hope that creating cooperative projects in science, agriculture, business, and education will lower international tension. In supporting the Epona conference, ISAC contributed importantly to cooperative science.

Membrane trafficking and the cytoskeleton: an integrated view. ASCB/EMBO/Dudley Wright Summer Research Conference, Santa Maria Imbaro, Italy, June 26-30, 1999.

The beautiful grounds, facilities and location of the Consorzio Mario Negri Sud in Santa Maria Imbaro, on the Adriatic Coast of Italy, were the site at the end of June for a stimulating, interdisciplinary meeting entitled ‘Membrane trafficking and the cytoskeleton: an integrated view’. This meeting represented the fifth and final offering of biannual summer research conferences held this decade, arranged and supported jointly by the European Molecular Biology Organization, the American Society for Cell Biology (ASCB) and the H.Dudley Wright Foundation of Switzerland. Daniela Corda and Alberto Luini of the Department of Cell Biology and Oncology of the Consorzio hosted the meeting, supported by the excellent administrative assistance of Raffaella Bertazzi and Dorothy Doyle. The meeting topic was conceived and the sessions were organized by George Bloom (University of Texas Southwest Medical Center, Dallas, TX), Viki Allen (University of Manchester, Manchester, UK), Jennifer Lippincott‐Schwartz [National Institutes of Health (NIH) Division of Child Health and Human Development] and the late Thomas Kreis (University of Geneva, Geneva, Switzerland) who lost his life in the SwissAir crash last year. The meeting was attended by ∼175 principal investigators, postdoctoral and graduate students representing 14 European countries plus the USA, Canada, Japan, Israel and New Zealand. Additional funding for the meeting was provided by the National Cancer Institute of the NIH, the National Science Foundation and the Keith Porter Foundation. The purpose of these topic‐oriented summer research conferences, as envisioned by the late Mr Dudley Wright, is to foster scientific exchange between various groups of scientists isolated from each other by geographical constraints, or differences in fields or approaches to a problem. The meeting in Italy certainly succeeded in fulfilling Mr Wright's intent and was notable for its inclusion of many scientists at the postdoctoral and graduate student levels. Multiple docking sites on substrate proteins form a modular system that mediates recognition by ERK MAP kinase

MAP kinases phosphorylate specific groups of substrate proteins. Here we show that the amino acid sequence FXFP is an evolutionarily conserved docking site that mediates ERK MAP kinase binding to substrates in multiple protein families. FXFP and the D box, a different docking site, form a modular recognition system, as they can function independently or in combination. FXFP is specific for ERK, whereas the D box mediates binding to ERK and JNK MAP kinase, suggesting that the partially overlapping substrate specificities of ERK and JNK result from recognition of shared and unique docking sites. These findings enabled us to predict new ERK substrates and design peptide inhibitors of ERK that functioned in vitro and in vivo.

A coactivator of pre-mRNA splicing.

The nuclear matrix antigen recognized by the monoclonal antibody (mAb) B1C8 is a novel serine (S) and arginine (R)-rich protein associated with splicing complexes and is named here SRm160 (SR-related matrix protein of 160 kD). SRm160 contains multiple SR repeats, but unlike proteins of the SR family of splicing factors, lacks an RNA recognition motif. SRm160 and a related protein SRm300 (the 300-kD nuclear matrix antigen recognized by mAb B4A11) form a complex that is required for the splicing of specific pre-mRNAs. The SRm160/300 complex associates with splicing complexes and promotes splicing through interactions with SR family proteins. Binding of SRm160/300 to pre-mRNA is normally also dependent on U1 snRNP and is stabilized by U2 snRNP. Thus, SRm160/300 forms multiple interactions with components bound directly to important sites within pre-mRNA. The results suggest that a complex of the nuclear matrix proteins SRm160 and SRm300 functions as a coactivator of pre-mRNA splicing.
