How does silicon alleviate Cd-induced phytotoxicity in barley, Hordeum vulgare L.?

Toxic heavy metal accumulation in edible plants has become a problem worth worrying about for human health. Cadmium is one of the most toxic metals presenting high bioavailability in the environment. The main route of transfer of Cd to humans is the consumption of contaminated food which suggests that reducing of Cd absorption by plants could reduce this risk. In this context, it was suggested that silicon supply would be able to limit the transfer of Cd to the plants. Thus, this work evaluated the effects of 0.5 mM Si on Cd absorption and accumulation in barley (Hordeum vulgare L.). Plants were grown hydroponically for 21 days in the presence of 0 and 15 μM Cd2+ combined or not with 0.5 mM Si. Analyses were related to growth and photosynthesis parameters, Cd accumulation in organs and Cd subcellular distribution in the shoots. Results showed that, under Cd alone, plants showed severe toxicity symptoms as chlorosis and necrosis and produced significantly less biomass as compared to control. 0.5 mM Si in the medium culture significantly reduced Cd-induced toxicity by mitigating symptoms and restoring growth, photosynthesis, and nutrition. Si also induced a significant reduction of Cd concentration in plants and changed its sub-cellular compartmentalization by enhancing fixation to cell walls and reducing the Cd concentration in the cytoplasmic and organelles fractions. These data suggest that the application of Si could significantly increase Cd tolerance and reduce the risk of the Cd accumulation in edible plants.

Elucidating the role of atmospheric deposition on uptake and subcellular distribution of As, Cd, and Pb through root in A. paniculata

The present study examined the impact of atmospheric deposition on the accumulation, translocation, and subcellular distribution of As, Pb, and Cd via root in A. paniculata. Two experimental conditions were set in the study: i. Plants grown in metal-contaminated and without metals-contaminated soils were exposed to atmospheric deposition, and ii. Plants grown in metal-contaminated and without metals-contaminated soils were not exposed to atmospheric deposition. The PM10 in the air and dry deposition on the surface of leaves were collected and characterized. The uptake kinetics and subcellular distribution of As, Cd, and Pb, growth parameters, stress enzymes, and nutrient content in plants were also investigated. The PM10 concentration at the site varied between 142 and 469 μg/m3 and demonstrated the presence of Al, B, Ba, Ca, Cu, Fe, K, Mg, Mn, Zn, Pb, As, and Cd. The dry deposition on leaves displayed the presence of Pb, Cd, Al, B, Ba, Ca, Cu, Fe, K, Mg, Mn, and Zn. Results displayed no translocation of atmospheric Pb and Cd from leaves to stem and root in plants but increased uptake velocity and binding affinity of As, Cd, and Pb in the stem and root tissues from soil. An increase in As, Cd, and Pb content in the cell wall and organelle portion of all plant tissues was observed after atmospheric dust exposure. Alterations in the stomata opening and aperture size in leaves and reduction in plant growth and secondary metabolite content were observed after atmospheric dust exposure.

SpCTP3 from the hyperaccumulator Sedum plumbizincicola positively regulates cadmium tolerance by interacting with SpMDH1

Cadmium (Cd) is a heavy metal pollutant mainly originating from the discharge of industrial sewage, irrigation with contaminated water, and the use of fertilizers. The phytoremediation of Cd polluted soil depends on the identification of the associated genes in hyperaccumulators. Here, a novel Cd tolerance gene (SpCTP3) was identified in hyperaccumulator Sedum plumbizincicola. The results of Cd2+ binding and thermodynamic analyses, revealed the CXXC motif in SpCTP3 functions is a Cd2+ binding site. A mutated CXXC motif decreased binding to Cd by 59.93%. The subcellular localization analysis suggested that SpCTP3 is primarily a cytoplasmic protein. Additionally, the SpCTP3-overexpressing (OE) plants were more tolerant to Cd and accumulated more Cd than wild-type Sedum alfredii (NHE-WT). The Cd concentrations in the cytoplasm of root and leaf cells were significantly higher (53.75% and 71.87%, respectively) in SpCTP3-OE plants than in NHE-WT. Furthermore, malic acid levels increased and decreased in SpCTP3-OE and SpCTP3-RNAi plants, respectively. Moreover, SpCTP3 interacted with malate dehydrogenase 1 (MDH1). Thus, SpCTP3 helps regulate the subcellular distribution of Cd and increases Cd accumulation when it is overexpressed in plants, ultimately Cd tolerance through its interaction with SpMDH1. This study provides new insights relevant to improving the Cd uptake by Sedum plumbizincicola.

Effect of Phosphorus Application on Subcellular Distribution and Chemical Morphology of Cadmium in Eggplant Seedlings under Cadmium Stress

Soil cadmium (Cd) contamination poses a serious threat to ecosystems, and the application of phosphorus fertilizers can reduce Cd toxicity. However, the specific effects of different phosphorus fertilizers on the subcellular distribution and chemical morphology of Cd in eggplant grown in calcareous Cd-contaminated soil remain unclear. This study examined the impact of various types and levels of phosphate fertilizers on the subcellular distribution and chemical morphology of cadmium in eggplant seedlings using a two-factor analysis. The investigation was conducted via a pot experiment utilizing a two-factor analysis. The application of 0.35 g kg−1 dicalcium phosphate significantly decreased the Cd content in the subcellular distribution and induced notable alterations in the chemical morphology of Cd in eggplant roots. Specifically, the ethanol-extracted Cd state decreased by 65.45%, and the sodium chloride-extracted Cd state decreased by 64.65%. Conversely, Cd extracted by deionized water, acetic acid, hydrochloric acid, and the residue state increased by 6.20%, 4.01%, 20.87%, and 17.85%, respectively. The application of 0.35 g kg−1 dicalcium phosphate resulted in the most significant reduction in Cd content in eggplant and modification of subcellular Cd distribution and chemical morphology in roots.

NtGCN2 confers cadmium tolerance in Nicotiana tabacum L. by regulating cadmium uptake, efflux, and subcellular distribution

General control non-derepressible-2 (GCN2) is widely expressed in eukaryotes and responds to biotic and abiotic stressors. However, the precise function and mechanism of action of GCN2 in response to cadmium (Cd) stress in Nicotiana tabacum L. (tobacco) remains unclear. We investigated the role of NtGCN2 in Cd tolerance and explored the mechanism by which NtGCN2 responds to Cd stress in tobacco by exposing NtGCN2 transgenic tobacco lines to different concentrations of CdCl2. NtGCN2 was activated under 50 μmol·L−1 CdCl2 stress and enhanced the Cd tolerance and photosynthetic capacities of tobacco by increasing chlorophyll content and antioxidant capacity by upregulating NtSOD, NtPOD, and NtCAT expression and corresponding enzyme activities and decreasing malondialdehyde and O2·- contents. NtGCN2 enhanced the osmoregulatory capacity of tobacco by elevating proline (Pro) and soluble sugar contents and maintaining low levels of relative conductivity. Finally, NtGCN2 enhanced Cd tolerance in tobacco by reducing Cd uptake and translocation, promoting Cd efflux, and regulating Cd subcellular distribution. In conclusion, NtGCN2 improves the tolerance of tobacco to Cd through a series of mechanisms, namely, increasing antioxidant, photosynthetic, and osmoregulation capacities and regulating Cd uptake, translocation, efflux, and subcellular distribution. This study provides a scientific basis for further exploration of the role of NtGCN2 in plant responses to Cd stress and enhancement of the Cd stress signaling network in tobacco.

Unveiling the variability in cadmium accumulation and tolerance characteristics: a comparative study of Basma and Yunyan 87 tobacco varieties

ABSTRACT Tobacco (Nicotiana tabacum L.) shows promise for remediating Cd-contaminated soil due to its significant Cd accumulation capabilities. Although various tobacco varieties exhibit distinct Cd bioaccumulation capacities, a comprehensive understanding of the underlying mechanisms is lacking. This study, conducted using hydroponics, explores differences in Cd accumulation and tolerance mechanisms between two tobacco varieties, Basma and Yunyan 87. The results showed that Cd stress reduced the dry weight, tolerance index, and root morphology for both varieties. Basma exhibited a relatively smaller decline in these indices compared to Yunyan 87. Moreover, Basma demonstrated a higher Cd bioconcentration factor (BCF), concentration, and accumulated content, signifying its superior tolerance and bioaccumulation capacity to Cd compared to Yunyan 87. The Carbonyl Cyanide3-ChloroPhenylhydrazone (CCCP) addition resulted in reduced Cd accumulation and BCFs in both tobacco species. This effect was more pronounced in Basma, suggesting that Basma relies more on an active transport process than Yunyan 87. This could potentially explain its enhanced bioaccumulation ability. Subcellular Cd distribution analysis revealed Basma's preference for distributing Cd in soluble fractions, while Yunyan 87 favoured the cell wall fractions. Transmission electron microscope showed that Basma's organelles were less damaged than Yunyan 87's under Cd stress, possibly contributing to the superior tolerance of Basma. Therefore, these results provided a theoretical foundation for development of Cd-contaminated soil tobacco remediation technology.

Phosphorus deficiency-induced cell wall pectin demethylesterification enhances cadmium accumulation in roots of Salix caprea

Regions affected by heavy metal contamination frequently encounter phosphorus (P) deficiency. Numerous studies highlight crucial role of P in facilitating cadmium (Cd) accumulation in woody plants. However, the regulatory mechanism by which P affects Cd accumulation in roots remains ambiguous. This study aims to investigate the effects of phosphorus (P) deficiency on Cd accumulation, Cd subcellular distribution, and cell wall components in the roots of Salix caprea under Cd stress. The results revealed that under P deficiency conditions, there was a 35.4% elevation in Cd content in roots, coupled with a 60.1% reduction in Cd content in shoots, compared to the P sufficiency conditions. Under deficient P conditions, the predominant response of roots to Cd exposure was the increased sequestration of Cd in root cell walls. The sequestration of Cd in root cell walls increased from 37.1% under sufficient P conditions to 66.7% under P deficiency, with pectin identified as the primary Cd binding site under both P conditions. Among cell wall components, P deficiency led to a significant 31.7% increase in Cd content within pectin compared to P sufficiency conditions, but did not change the pectin content. Notably, P deficiency significantly increased pectin methylesterase (PME) activity by regulating the expression of PME and PMEI genes, leading to a 10.4% reduction in the degree of pectin methylesterification. This may elucidate the absence of significant changes in pectin content under P deficiency conditions and the concurrent increase in Cd accumulation in pectin. Fourier transform infrared spectroscopy (FTIR) results indicated an increase in carboxyl groups in the root cell walls under P deficiency compared to sufficient P treatment. The results provide deep insights into the mechanisms of higher Cd accumulation in root mediated by P deficiency.

Inhibitory effects of Pseudomonas sp. W112 on cadmium accumulation in wheat grains: Reduced the bioavailability in soil and enhanced the interception by plant organs

Microorganisms play an important role in heavy metal bioremediation and soil fertility. The effects of soil inoculation with Pseudomonas sp. W112 on Cd accumulation in wheat were investigated by analyzing the transport, subcellular distribution and speciation of Cd in the soil and plants. Pseudomonas sp. W112 application significantly decreased Cd content in the roots, internode and grains by 10.2%, 29.5% and 33.0%, respectively, and decreased Cd transfer from the basal nodes to internodes by 63.5%. Treatment with strain W112 decreased the inorganic and water-soluble Cd content in the roots and increased the proportion of residual Cd in both the roots and basal nodes. At the subcellular level, the Cd content in the root cell wall and basal node cytosol increased by 19.6% and 61.8%, respectively, indicating that strain W112 improved the ability of the root cell wall and basal node cytosol to fix Cd. In the rhizosphere soil, strain W112 effectively colonized and significantly decreased the exchangeable Cd, carbonate-bound Cd and iron-manganese oxide-bound Cd content by 43.5%, 27.3% and 17.6%, respectively, while it increased the proportion of residual Cd by up to 65.2%. Moreover, a 3.1% and 23.5% increase in the pH and inorganic nitrogen content in the rhizosphere soil, respectively, was recorded. Similarly, soil bacterial community sequencing revealed that inoculating with strain W112 increased the abundance of Pseudomonas, Thauera and Azoarcus, which are associated with inorganic nitrogen metabolism, and decreased that of Acidobacteria, which is indicative of soil alkalinization. Hence, root application of Pseudomonas sp. W112 improved soil nitrogen availability and inhibited Cd accumulation in the wheat grains in a two-stage process: by reducing the Cd availability in the rhizosphere soil and by improving Cd interception and fixation in the wheat roots and basal nodes. Pseudomonas sp. W112 may be a suitable bioremediation agent for restoring Cd-contaminated wheat fields.

Cadmium induced changes in rhizosphere microecology to enhance Cd intake by Ligusticum sinense cv. Chuanxiong

As the most famous and widely used traditional Chinese medicine (TCM), Ligusticum sinense cv. Chuanxiong (L. Chuaniong) has been affected by cadmium (Cd) exceeding with high ability of Cd accumulation. There is relatively little research on Cd absorption and storage process in L. Chuanxiong, which is an important reason for the poor remediation efficiency. Hence, this study takes L. Chuanxiong as the point of penetration to explore how L. Chuanxiong affects rhizobacteria through root exudates to alter soil Cd intake, as well as to explore the migration and storage of Cd in its body with 0.10 (T0), 5.00 (T5), 10.00 (T10) mg/kg Cd contaminations. The results showed that the relative abundance of amino acids and phospholipids secreted from L. Chuanxiong root noticeably increased with increasing Cd levels, which directly activated soil Cd or extremely significantly (P < 0.01) recruited bacteria such as Bacillus, Arthrobacter to indirectly increase Cd availability. Under the interaction of root exudates and rhizobacteria, Cd bioavailability increased by 80.00% in rhizosphere soil and Cd accumulation in L. Chuanxiong increased 5.44–6.65 mg/kg. Cd subcellular distribution analysis demonstrated that Cd was mainly stored in the root (10-fold more than in the leaf), whose Cd content was cytoderm>cytoplasm>organelle in tissues. The sequential extraction results found that non-soluble phosphate and protein-chelated Cd dominated (85.00–90.00%) in the cell, while Cd cheated with alcohol soluble protein, amino acid salts, water-soluble organic acid in cell was minimal (5.50%). The phenomenon indicated that L. Chuanxiong fixed Cd in root (the medical part) with low translocation ability. This study can provide theoretical support for the high-quality production of L. Chuanxiong and other root medical plant in heavy metal influenced sites.

Two-stage Inhibition Effects of Burkholderia sp. Y4 Application on Cadmium Uptake and Transport in Wheat

In order to evaluate the feasibility of using Burkholderia sp. Y4 as a cadmium （Cd）-reducing bacterial agent in contaminated wheat fields， the changes in the rhizosphere soil microbial community and Cd available state， as well as the content and transport characteristics of Cd in the wheat root， basal node， internode， and grain under the treatment of strain Y4 were tested using microbial high-throughput sequencing， step-by-step extraction， subcellular distribution， and occurrence analyses. The results showed that root application of strain Y4 significantly reduced the root and grain Cd content of wheat by 7.7% and 30.3%， respectively， compared with that in the control treatment. The Cd content and Cd transfer factor results in wheat vegetative organs showed that strain Y4 reduced the Cd transfer factor from basal node to internode by 79.3%， and Cd content in the wheat internode stem also decreased by 50.9%. The study of Cd occurrence morphology showed that strain Y4 treatment increased the proportion of residual Cd in roots and basal ganglia， decreased the contents of inorganic and water-soluble Cd in roots， and increased the content of residual Cd in basal ganglia. Further examination of the subcellular distribution of Cd showed that the Cd content in root cell walls and basal ganglia cell fluid increased by 21.3% and 98.2%， respectively， indicating that the Cd fixation ability of root cell walls and basal ganglia cell fluid was improved by the strain Y4 treatment. In the rhizosphere soil， it was found that the microbial community structure was changed by strain Y4 application. Under the Y4 treatment， the relative abundance of Burkholderia increased from 9.6% to 11.5%， whereas that of Acidobacteriota decreased. Additionally， the relative abundance of Gemmatimonadales， Pseudomonadales， and Chitinophagales were also increased by strain Y4 treatment. At the same time， the application of strain Y4 increased the pH value of rhizosphere soil by 8.3%. The contents of exchangeable Cd， carbonate-bound Cd， and iron-manganese oxide-bound Cd in the soil decreased by 44.4%， 21.7%， and 15.9%， respectively， whereas the proportion of residual Cd reached 53.6%. Root application of strain Y4 increased the contents of nitrate nitrogen and ammonium nitrogen in the soil by 22.0% and 21.4%， respectively， and the contents of alkaline nitrogen also increased to a certain extent. In conclusion， the root application of strain Y4 not only improved soil nitrogen availability but also inhibited Cd transport and accumulation from contaminated soil to wheat grains in a "two-stage" manner by reducing Cd availability in rhizosphere soil and improving Cd interception and fixation capacity of wheat roots and basal nodes. Therefore， Burkholderia Y4 has application potential as a Cd-reducing and growth-promoting agent in wheat.

Physiological and biochemical characteristics of high and low Cd accumulating Brassica napus genotypes.

Phytoremediation is a widely used and cost-effective technique for in situ remediation of heavy metals. Brassica napus L. genotype with high Cd accumulation and strong Cd tolerance is an ideal candidate for phytoremediation. In this study, a hydroponic experiment was conducted to select a Brassica napus genotype with either high or low Cd accumulation from a panel of 55 genotypes. The physiological mechanisms governing Cd accumulation and Cd tolerance were then explored. BN400 and BN147 were identified as the high and low Cd accumulating genotypes, respectively. Additionally, BN400 exhibited greater tolerance to Cd stress compared to BN147. Root morphology analysis revealed that BN400 exhibited longer root length, smaller root surface area and root volume, and less root tips but bigger root diameter than BN147. Subcellular Cd distribution showed that the Cd concentrations in the cell wall and vacuole in shoot were significantly higher in BN400 than in BN147, whereas the opposite trend was observed in the roots.. Pectate/protein-integrated Cd was found to be the predominant form of Cd in both shoots and roots, with significantly higher levels in BN400 compared to BN147 in the shoot, but the opposite trend was observed in the roots. These results suggest that the long fine roots play a role in Cd accumulation. The high Cd accumulating genotype was able to retain Cd in leaf cell walls and vacuoles, and Cd was mainly present in the form of pectate/protein-integrated Cd, which contributes to its strong Cd tolerance. These findings have important implications for the screening and breeding of Brassica napus genotypes with high Cd accumulation for phytoremediation purposes.

Effects of magnetic field on cd subcellular distribution and chemical speciation in Noccaea caerulescens

Implementing an external magnetic field of suitable strength has been reported to increase Cd uptake by Noccaea caerulescence. However, only a few mechanisms promoting this efficiency have been reported. A series of culture experiments was conducted to explore how Cd subcellular distribution and speciation vary within the tissue of N. caerulescens when subjected to external magnetic fields of different intensities. Without a magnetic field, over 80% of the Cd was deposited in the cell wall and cytoplasm, indicating that cell wall retention and cytoplasm isolation are significant mechanisms for the detoxification of Cd. An external magnetic field (120 mT) increased the Cd concentrations deposited in the cytoplasm and water-soluble inorganic Cd in the roots, increasing the cell wall-bound Cd and undissolved Cd phosphate in the shoots. Meanwhile, the magnetic field increased carbonic anhydrase activity in plant shoots, except at 400 mT. These results indicated that an external field can elevate the Cd decontamination capacity of N. caerulescens by changing the subcellular compartmentalization and speciation of Cd in different tissues.

Exogenous Xyloglucan Oligosaccharides Alleviate Cadmium Toxicity in Boehmeria nivea by Increasing the Cadmium Fixation Capacity of Cell Walls

Xyloglucan is an important component of hemicellulose, and xyloglucan oligosaccharides (Xh), which are metabolized by xyloglucan, play an important role in plant growth and development. However, the regulatory effects of the external application of Xh under cadmium (Cd) stress have not been determined. In this study, we evaluated the mechanism by which Xh contributes to resistance to Cd stress in ramie, a candidate plant species for toxic ion removal. The external application of Xh effectively attenuated the effects of Cd on ramie growth and photosynthetic pigments. Cd stress can also inhibit the activity of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX), resulting in a significant increase in the extent of membrane lipid peroxidation. After the external application of Xh, antioxidant enzyme activity was up-regulated, and damage to membranes in plants was reduced. In addition, the external application of Xh increased Cd retention in roots, thereby significantly decreasing Cd content in shoots. The external application of Xh also regulated the subcellular distribution of Cd and increased the Cd content of the cell wall. In particular, a root cell wall analysis revealed that Cd+Xh treatment significantly increased the hemicellulose content in the cell wall and the amount of Cd retained. In summary, the external application of Xh alleviates Cd toxicity in ramie by increasing the hemicellulose content and the Cd fixation ability of the cell wall and by reducing membrane lipid peroxidation via antioxidant enzymes.

Characteristics and Mechanisms of Soil Co-Contamination Affecting the Transfer of Cadmium and Arsenic in Peanut (Arachis hypogaea L.)

Soil co-contamination with cadmium (Cd) and arsenic (As) occurs frequently and has caused increasing concern. This study aimed to explore the transfer characteristics and the chemical forms, subcellular distribution of Cd and As, as well as the synthesis of phytochelatins (PCs) and other chelates in peanut (Arachis hypogaea L.) plants grown in a Cd and As co-contaminated soil, shedding light on the mechanisms involved. Compared with the single Cd contamination, Cd–As co-contamination led to a higher accumulation of Cd in peanut plants. Conversely, compared to the single As contamination, the As content increased in peanut shoots but decreased in roots and grains under Cd–As co-contamination. Furthermore, the Cd–As interaction resulted in notable changes in peanut plants’ physiological and biochemical responses. In the roots and shoots, there was an 81.8% and 60.0% increase in water-soluble Cd. In the roots, metallothioneins (MTs) content increased by 50%, while PCs increased by 6.4% in the shoots. These changes promoted the translocation of Cd from roots to grains. The Cd–As interaction also influenced the synthesis of MTs in the roots, showing a 41.2% increase, and facilitated the transfer of As to the shoots. In peanut shoots, Cd increased the cell wall fraction of As by 34.5%, decreased the proportion of water-soluble As by 31.8%, and increased PCs content by 6.9%. These changes inhibited the migration of As from shoots to grains. Overall, Cd–As co-contamination increased Cd in peanut grains by increasing water-soluble forms and MTs in roots, while Cd–As co-contamination decreased As in peanut grains by increasing cell wall fractions and PCs in shoots. These findings provide a theoretical basis for understanding Cd–As interactions in soil–peanut systems.

Mechanisms underlying the alleviated cadmium toxicity in marine diatoms adapted to ocean acidification

Anthropogenic activities have significantly increased the influx of carbon dioxide and metals into the marine environment. Combining ocean acidification (OA) and metal pollution may lead to unforeseen biological and ecological consequences. Several studies have shown that OA reduces cadmium (Cd) toxicity in marine diatoms. Although these studies have shed light on the physiological and transcriptomic responses of diatoms exposed to Cd, many aspects of the mechanisms underlying the reduced metal accumulation in diatoms remain unknown. This study aims to address this unresolved question by comparing Cd subcellular distribution, antioxidant enzyme activity, relative expression of metal transporters, surface potential, surface composition, and transmembrane potential in the diatom Phaeodactylum tricornutum grown under ambient and 1200 µatm pCO2 conditions. Our findings reveal that diatoms grown in acidified seawater exhibit higher surface potential and higher plasma membrane depolarization. These changes and the competing effects of increased H+ concentration result in a blunted response of P. tricornutum to the Cd challenge. Consequently, this study offers a new explanation for mitigating Cd toxicity by marine diatoms adapted to OA.

Selenate reduced wheat grain cadmium accumulation by inhibiting cadmium absorption and increasing root cadmium retention

Selenium (Se) fertilizer has been recently used to reduce cadmium (Cd) accumulation in plant. A pot culture was performed to analyze Cd uptake, translocation, and distribution in wheat plants during the reproductive growth period in a Cd–contaminated soil after selenate was applied to the soil, and a hydroponic culture was carried out to investigate the effects of selenate application on Cd2+ influx, subcellular Cd distribution, and Cd accumulation in wheat seedlings. Results showed that selenate application had no significant effect on DTPA–Cd and Cd fraction in soil. The application of selenate greatly inhibited the whole–plant Cd absorption by 14%–23%. In addition, selenate prompted the retention of Cd in root by increasing the Cd distribution in the vacuole, which reduced the root–to–shoot Cd translocation by 18%–53%. The application of selenate increased the Cd concentration in nodes, inhibited Cd remobilization from nutritive organs to grain, and ultimately reduced Cd accumulation in wheat grain. Further, heading to grain filling was the key growth stage for exogenous selenate to regulate grain Cd accumulation. In summary, soil selenate application is an effective method to reduce grain Cd concentration in wheat, which provided scientific basis for remediation of Cd–contaminated soil.

Potential use of Rorippa sylvestris (L.) Besser. as a cadmium hyperaccumulator for phytoremediation

Rorippa sylvestris (L.) Besser. (yellow fieldcress) is a hyperaccumulator of cadmium (Cd) discovered recently. There are several studies in the topic of Cd tolerance capacity and detoxification mechanism of plants, but the detoxification mechanism of R. sylvestris is not clear. Root tiller seedlings of R. sylvestris were planted in the brown bottles filled with culture solution containing different levels of Cd (0, 10, 25, 50, 100, 200 μmol·L−1, respectively) for 8 days in order to make sure the physiological responses, Cd subcellular distribution and chemical forms under Cd stress. The results showed that the growth increased in lower Cd concentration and declined in higher Cd concentration. Physiological characteristics such as photosynthesis and MDA were not significantly affected compared with the control at Cd concentratetion ≤ 50 μmol·L−1. The content of soluble sugar and free proline increased with the increasing of Cd concentration for the purpose of reducing the Cd toxicity to plant. R. sylvestris had a strong capability of Cd accumulation in different Cd treatments. The concentration of Cd in plant tissues (root, stem, leaf) was in the order of root>stem>leaf. The Cd subcellular distribution in plant tissues of R. sylvestris was in the order of soluble fractions>cell wall>protoplast>mitochondria. Cd was mainly extracted by deionized water and 80% ethanol (60.91–69.76%), followed by sodium chloride (16.14–25.12%), which indicated that R. sylvestris achieved to enhance Cd tolerance capacity and detoxification mechanism through root retention and vacuolar compartmentalization.

Boron decreases cadmium accumulation in water spinach by enhancing cadmium retention in the root cell walls.

Cadmium (Cd) contamination and boron (B) deficiency are two major challenges associated with the farmland soils in Southern China. Therefore, this study was conducted to examine the impacts of B supply on Cd accumulation in water spinach (Ipomoea aquatica) using a cultivar (T308) with high Cd accumulation. The study further investigated the physiological mechanism behind the changes in Cd accumulation due to B supply. The findings revealed that B supply substantially reduced the Cd concentration in the leaves of water spinach by 41.20% and 37.16% under the Cd stress of 10μM and 25μM, respectively. Subcellular distribution of Cd showed that the Cd content as well as its proportion in root cell wall (RCW) increased significantly after B supply. Fourier transform infrared spectroscopy showed significant enrichment of negatively charged groups (such as -OH, -COOH, and -NH2) in the RCW after B supply. Overall, B supply also enhanced covalently bound pectin (CSP) content as well as the Cd content linked with CSP under Cd stress. These observations revealed that B regulated the Cd chelation in RCW, thereby reducing the amassment of Cd in water spinach.

Transcriptome analysis reveals the molecular mechanism of different forms of selenium in reducing cadmium uptake and accumulation in wheat seedlings

Selenium (Se) can counteract cadmium (Cd) toxicity in wheat, but the molecular mechanism of different Se forms reducing Cd uptake and accumulation in wheat seedlings remain unclear. Here, a hydroponic experiment was conducted to investigate the effects of three Se forms (selenite (Se(IV)), selenate (Se(VI)) and seleno−L−methionine (SeMet)) on Cd2+ influx, Cd subcellular distribution, and Cd accumulation in wheat seedlings, and the underlying molecular mechanisms were investigated through transcriptome analysis. Consequently, Se(IV) and Se(VI) addition significantly reduced root Cd concentration by 74.3% and 80.8%, respectively, and all Se treatments significantly decreased shoot Cd concentration by approximately 34.2%–74.9%, with Se(IV) addition having the most pronounced reducing effect. Transcriptome analysis showed the reduction of Cd accumulation after Se(IV) addition was mainly due to the downregulation of Cd uptake genes. The inhibition of Cd accumulation after Se(VI) addition was not only associated with the downregulation of Cd uptake genes, but also related to the sequestration of Cd in vacuole. For SeMet addition, the reduction of Cd accumulation was mainly related to the sequestration of Cd in vacuole as GSH−Cd. The above findings provide novel insights to understand the effects of different forms of Se on Cd uptake and accumulation and tolerance in wheat.

The Role of Lignin in the Compartmentalization of Cadmium in Maize Roots Is Enhanced by Mycorrhiza.

In nature, arbuscular mycorrhizal fungi (AMF) play a crucial role in the root systems of plants. They can help enhance the resistance of host plants by improving the compartmentalization of toxic metal contaminants in the cell walls (CWs). However, the functions and responses of various CW subfractions to mycorrhizal colonization under Cd exposure remain unknown. Here we conducted a study to investigate how Cd is stored in the cell walls of maize roots colonized by Funneliformis mosseae. Our findings indicate that inoculating the roots with AMF significantly lowers the amount of Cd in the maize shoots (63.6 ± 6.54 mg kg-1 vs. 45.3 ± 2.19 mg kg-1, p < 0.05) by retaining more Cd in the mycorrhized roots (224.0 ± 17.13 mg kg-1 vs. 289.5 ± 8.75 mg kg-1, p < 0.01). This reduces the adverse effects of excessive Cd on the maize plant. Additional research on the subcellular distribution of Cd showed that AMF colonization significantly improves the compartmentalization of 88.2% of Cd in the cell walls of maize roots, compared to the 80.8% of Cd associated with cell walls in the non-mycorrhizal controls. We observed that the presence of AMF did not increase the amount of Cd in pectin, a primary binding site for cell walls; however, it significantly enhanced the content of lignin and the proportion of Cd in the total root cell walls. This finding is consistent with the increased activity of lignin-related enzymes, such as PAL, 4CL, and laccase, which were also positively impacted by mycorrhizal colonization. Fourier transform infrared spectroscopy (FTIR) results revealed that AMF increased the number and types of functional groups, including -OH/-NH and carboxylate, which chelate Cd in the lignin. Our research shows that AMF can improve the ability of maize plants to tolerate Cd by reducing the amount of Cd transferred from the roots to the shoots. This is achieved by increasing the amount of lignin in the cell walls, which binds with Cd and prevents it from moving through the plant. This is accomplished by activating enzymes related to lignin synthesis and increasing the exposure of Cd-binding functional groups of lignin. However, more direct evidence on the immobilization of Cd in the mycorrhiza-altered cell wall subfractions is needed.
