In vitro stress selection of nematophagous fungi for biocontrol of parasitic nematodes in ruminants

Laboratory experiments were designed to select nematophagous fungi that were able to survive in vitro conditions simulating passage through the gastro-intestinal tract of cattle. All of the tests were conducted at 39 degrees C. In a primary stress selection step in diluted rumen fluid, 21 isolates were obtained. Each of the primary stress selected isolates was tested in synthetic saliva, rumen fluid simulating the activity in the rumen, rumen fluid followed by pepsin-hydrochloric acid treatment simulating the additional effect of ruminal and abomasal activity, pepsin-hydrochloric acid solution simulating conditions in the abomasum and finally in a trypsin solution as an example of enzyme activity in the gut. The effect of the rumen fluid alone, or rumen fluid followed by pepsin-hydrochloric acid treatment, were responsible for the reduction in surviving fungal isolates. Only six of thirteen isolates belonging to the genus Arthrobotrys survived while seven of eight isolates of the genus Duddingtonia survived. Fourteen isolates were tested for their predatory capacity in a dung pat bioassay. Fungi of the genera Arthrobotrys and Duddingtonia reduced the development of Ostertagia ostertagi third stage larvae by approximately 75% and 96% respectively compared to the number of larvae that developed from fungus-free control pats.

Effects of feed intake on particle distribution, passage of digesta, and extent of digestion in the gastrointestinal tract of cattle

Four nonlactating Holstein dairy cows (means +/- SD BW = 692 +/- 49 kg) were used in a 4 x 4 Latin square design experiment to determine changes in distribution of particles within the ruminoreticulum (RR) and total digestive tract in cattle fed a forage-based diet at four intake levels. Relationships between chewing activities and rates of particle breakdown, passage, and digestion were also determined. Percentage of large particles increased linearly in the dorsal rumen (P = .001), ventral rumen (P = .004), reticulum (P = .007), duodenum (P = .007), and feces (P = .006) as DMI increased. Particles of sizes less than or equal to 2,000 microns, eligible to pass from the RR, constituted 60 to 92% of particle DM in the RR. Increased feed intake resulted in a longer (P = .003) eating but a shorter (P = .008) ruminating time per kilogram of DM intake, whereas total chewing time per kilogram of DMI was not affected (P = .12). Passage rate constants of NDF from the RR increased (P = .03) with intake. There was no change (P = .87) in the rate constant for particle breakdown in the dorsal rumen with increased intake. Neither fractional passage rate of NDF nor the rate constant for particle breakdown was related to time spent eating (P + .12; P = .34) or time spent ruminating (P = .11; P = .55). It was concluded that rate of passage of small particles from the RR was a determinant of passage from the RR and that changes in rates of passage and breakdown of particles could only be partially explained by changes in chewing activities.

The flow of undigested corn residues through the gastrointestinal tract of cattle.

Two experiments were conducted to measure effects of source and level of roughage on the flow of corn residues through the gastrointestinal tract of cattle. In Exp. 1, steers (195 kg) were fed diets of ground corn with 0, 30 or 60% of ground Coastal bermudagrass hay (Cynodon dactylon) [L.] Pers.) at intakes of 1, 1.5 or 2% of BW in a 9 x 9 Latin square. Experiment 2 consisted of two 4 x 4 Latin squares with either rice hulls (square 1) or ground Coastal bermudagrass hay (square 2) providing 0, 7.5, 15 or 30% of the total diet fed at 1.5% of BW. After a 28-d adjustment period, a portion of the corn in one meal was replaced with cracked corn stained with brilliant green. The concentrations of stained corn residues appearing in the feces subsequent to dosing were fitted to a one-compartment, age-dependent model and compartmental mean residence time (CMRT) and time delay (tau) were estimated. In Exp. 1, increasing the level of intake of the ration from 1% to 1.5 or 2.0% of BW increased (P less than .05) CMRT by 52% and reduced (P less than .05) tau by 41%. In Exp. 2, source of roughage had no effect (P = .95) on CMRT or tau. Combined results of the two experiments indicated that increasing proportion (P) of either roughage was associated with an exponential decline in CMRT of stained corn residues (CMRT = 1211 * e-.0315P) from rations consumed at 1.5 and 2.0% of BW. No consistent effect of roughage type or proportion was noted on time delay in the two experiments collectively. These results indicate that increasing the proportion of roughage in the diet exponentially reduces residence time of corn residues in the ruminoreticulum (CMRT) without affecting residence time in the postgastric segments (tau).

Equilibration and passage of water in the gastrointestinal tract of cattle in relation to estimating body water by compartmental kinetic models.

The volume of water in the rumen of four steers was increased (P less than .01) 47% when the level of ground cobs in the diet fed to the steers was changed from 10% to 50%. The difference in gut water content due to diet was not accurately (P greater than .10) estimated by deuterium oxide dilution using either one-, two- or three-compartmental models. Gut water was overestimated and empty body water underestimated when calculated by two-compartment models. The proportions of total body water within each compartment of a three-compartment model were quite variable among steers. When the two-compartment model was solved on the basis of measurements taken from either compartment, different compartment volumes were obtained. This indicated that the two-compartment model did not accurately describe the water equilibration process. Water in the contents of the proximal duodenum and terminal ileum equilibrated with blood in 30 min (range, 10 to 75 min), fecal water equilibrated in 4.3 h (range, 1.7 to 6.7 h) and water in rumen contents equilibrated in 8.3 h (range, 3.8 to 12.5 h). Diet did not affect (P greater than .10) equilibration time or the mean retention time of water in the gastrointestinal tract. Mean retention time was much longer than equilibration time; thus, the equilibration of water in the gastrointestinal tract contents was primarily dependent upon movement of water across the gut mucosa and not upon the flow of water through the gut. One-third of the water in the contents of the gastrointestinal tract was located outside the rumen. Compartmental modeling based only upon D2O disappearance from blood did not enable either gut water or rumen water to be accurately estimated.

The effects of dietary sucrose and the concentration of plasma urea and rumen ammonia on the degradation of urea in the gastrointestinal tract of cattle.

1. The rates of entry of urea into plasma, of urea degradation in the gastrointestinal tract, and the partition of that degradation between the rumen and post-ruminal tract were determined by use of [14C]urea and NaH14CO3 in Hereford steers receiving hay diets with or without sucrose. The concentrations of plasma urea and rumen ammonia were varied by infusions of urea into the rumen or abomasum. 2. For all diets, plasma urea concentration was related to urea entry rate, to degradation of urea in the whole gastrointestinal tract, and to its degradation in the post-ruminal tract, but the relationship with its degradation in the rumen was poor. 3. Degradation of urea in the rumen was related in a multiple regression in a curvilinear manner in three groups of diets (pasture-hay alone, pasture-hay--lucerne (Medicago sativa) mixtures, diets with sucrose), and negatively to rumen ammonia concentration for pasture-hay diets, and diets with sucrose. 4. Ruminal clearance of urea (rate of urea degradation per plasma urea concentration) was negatively related to the rumen ammonia concentration for steers given diets with sucrose, of pasture-hay with or without urea infusions. Provision of sucrose in the diet significantly increased clearance. 5. Enhanced urea degradation in the rumen associated with dietary sucrose supplements accounted for 0.4 of additional microbial N synthesis in the rumen. 6. The partition of transfer of urea to the rumen via saliva and through the rumen wall is discussed.

Effect of antimetabolites and actinomycin D on the replication of HADEN, a bovine parvovirus. Brief report.

A hemadsorbing enteric virus (HADEN), isolated from the gastrointestinal tract of cattle and previously classified as a bovine enterovirus, was found to induce Feulgen-positive intranuclear inclusions and to be inhibited in its replication by FUDR, BUDR and actinomycin D, indicating DNA as genetic material. Because of this fact and the other known properties of HADEN virus it should be considered a bovine parvovirus.

The copper status of some helminth parasites, with particular reference to host-helminth relationships in the gastro-intestinal tract of cattle

The copper content was determined in samples of 12 nematode, four cestode, and two trematode species collected from a variety of hosts infected either naturally or experimentally. Copper concentrations varied considerably from species to species, the lowest value recorded being 2 p.p.m. copper (dry weight basis) in the cestode Moniezia benedeni and the highest 604 p.p.m. in the nematode Haemonchus placei, both from cattle. Both the total content and the concentration of copper varied among mature individuals of the same population of the liver fluke, Fasciola hepatica, collected from cattle. Copper levels varied greatly in samples of H. placei and Oesophagostomum radiatum from cattle drawn from different host individuals, whereas values for Bunostomum phlebotomum and M. benedeni also from cattle varied little from host to host. Copper concentrations in samples of H. placei and O. radiatum, recovered from the abomasum and large intestine respectively of experimentally infected calves supplemented daily with copper sulphate, were considerably higher than those in samples from unsupplemented calves. With both species, the increases in worm copper levels were associated with increases in concentrations of soluble copper in the environment. It is suggested that soluble copper in environmental fluids can be absorbed through the cuticle of these helminths. Copper supplementation of the host's diet, on the other hand, had no effect on the copper content of B. phlebotomum Furthermore, the concentration of copper in solution in the contents of the duodenum and jejunum was not elevated significantly by the supplementatlon. The hypothesis that the intestinal helminths of bovines compete with the host for copper was examined, and it is concluded that in dairy calves in southeastern Queensland parasitic gastro-enteritis has a far greater significance as a disease entity in itself than as a factor contributing to the expression of a clinical state of copper deficiency.
