In silico toxicity assessment and trace level quantification of two genotoxic impurities in silodosin using capillary gas chromatography

A capillary gas chromatographic method using flame ionization detection was developed and validated for the trace quantification of 2-bromoethanol (2-BE) and 2-bromoethylmethanesulfonate (2-BEM) in silodosin, used in the treatment of benign prostatic hyperplasia. Chromatographic separation was performed in spilt mode using nitrogen as carrier gas on a column containing crosslinked polyethylene glycol (30 m × 0.32 mm, 0.25 µm) stationary phase modified with nitroterephthalic acid. A simple matrix precipitation strategy was implemented to eliminate the sample overload and the matrix interference problems. The developed method was linear and accurate in the concentration range of 24–3000 ppm for 2-BE and 24–300 ppm for 2-BEM with r2 ˃ 0.999 and percent recoveries greater than 90% for both the analytes. The developed method was precise for both the analytes with RSD(%) of not more than 4.5%. In silico genotoxicity and carcinogenicity potential of 2-BEM were assessed using ICH M7 principles. The developed method can be applied in the quality control laboratories of pharmaceutical industries for trace level quantification of 2-BE and 2-BEM in silodosin.

Substantiation of technological parameters of the oil extract obtained from Urtica dioica roots based on the yield determination of phytosterols using capillary gas chromatography method

Due to the content of phytosterols, extractive preparations of Urtica dioica roots are able to show antiandrogenic effect in the case of external therapy of men and women with androgenic alopecia. Oil extracts (OE) are characterized by several advantages when applied to the skin of the scalp compared to water-alcohol extracts. For the development of OE technology from Urtica dioica roots, it is important to choose the optimal extraction parameters, which are based on the quantitative determination of phytosterols in the extractant and the studied samples of extracts.
 The aim of the work is to choose the optimal parameters for obtaining OE from Urtica dioica roots based on quantitative determination of phytosterols content in experimental samples of OE by gas capillary chromatography.
 Materials and methods. Objects of the research – Urtica dioica root, refined corn oil, refined sunflower oil, samples of oil extracts. Determination of phytosterol content in experimental samples was carried out by gas capillary chromatography (chromatograph “Crystal 2000”, manufacturer – research and production company “Analytics”).
 Results. 5 different compounds of steroid structure (stigmasterol, β-sitosterol, etc.) were identified in sunflower oil by gas liquid chromatography, and 10 (campesterol, 2-α stigmasterol, β-sitosterol, Δ5-avenosterol, etc.) were identified in sunflower oil. The quantitative content of β-sitosterol in the sum of sterols of corn oil was significantly higher compared to the content of this substance in sunflower oil and amounted to 59.33 %. Optimal technological parameters were established considering the peculiarities of extraction with oil extractant and quantitative determination of the amount of phytosterols and β-sitosterol in experimental samples of OE. The total content of plant sterols in OE, including considering their amount in the extractant, was in the range of 7880 mg/kg; the amount of β-sitosterol was 4638 mg/kg.
 Conclusion. The choice of optimal parameters for obtaining OE from UDR based on determination of phytosterol yield by gas capillary chromatography was experimentally substantiated, namely: extractant – corn oil, raw material-extract ratio – 1: 5, extraction time – 6 h, extraction method – maceration

THE BOUILLON TABLES FROM TURKEY: DETERMINATON OF CIS-TRANS FATTY ACID PROFILES BY CAPILLARY GAS CHROMATOGRAPHY

Nowadays, commmercial bouillon tablets are mostly used as a flavor enhancer substant or an instant product in Turkish cuisine. As chemical structure, Trans FAs are unsaturated fatty acids having at least one double bond in its trans geometric configuration. Trans FA are occured by the partial hydrogenation of vegetable oils in the manufacturing of margarine and vegetable shortening. In this study, industrially produced commmercial bouillon tablet (n=14) samples from Turkey were analyzed by capillary (DB 23 column) gas chromatography method and an under controlled temperature oven programm with particular emphasis on cis–trans fatty acid profiles. The bouillon samples were collected as two goups (first group containing animal additives [AAB n=12] and second group including vegatable additives [VAB n=2]). There is no detailed information on fatty acid (FA) compounds, including trans fatty acids (TFAs), of consumed several commercial boullions in Turkey. There is no “trans fatty acid-free” declaration on various bouillon labels in Turkey. Large variations were observed among the cis–trans FA profiles of the boullion samples from Turkey,despite the fact that the samples are produced in the same production conditions.The palmitic (PAM) levels (predominant cis saturated FAs for all samples) of AAB samples (32.63–44.44%) were more high rather than the VAB samples (47.62–50.64%). Other major saturated FAs for AAB and VAB samples were determined stearic ( (5.28–9.90% AAB and 6.33–6.73% VAB), lauric (0.19– 5.84% AAB and 1.13–1.30% VAB) and myristic acids (0.86–2.61% AAB and 1.39–1.45% VAB).The changes of oleic (OLA) acid (from other predominant cis monounsaturated FA) of VAB samples (30.75–33.45%) were high than those of AABs (12.64–29.35%). The ranges of LO, an essential /nutritional fatty acid and the predominant FA of PUFAs, and PUFAs of AAB – except two samples– commercial bouillon samples (0.45–9.82% and 0.45–9.97% ) were low rather than VAA samples (7.97–8.14% and 8.14–8.35 %). Conjugated Linoleic (CLA) FAs, having nutritional FAs having anti-carcinogenic, antioxidative and anti-atherosclerotic effects, were determined in small amounts for only six samples (0.03% to 0.06% ). Elaidic acid (C18:1 trans acid) content, the mainly trans FA in all boullion, was within the range of 20.00–27.32% in the AAB samples, and it was significantly higher than the range in VAB samples (0.08–0.14%). Total trans fatty acids (TFAs) were another important major (second) FAs for all bouillon samples. The changes of Trans/Cis ratio ranged between 0.26 and 0.50 for AAB samples but VB samples has a little values from 0.002 to 0.003. Turkish vegetable boullion. samples have low total trans FA contents (0.19–0.26%) than the animal boullion samples (20.19–28.78%).There are a large variation and significant (P<0.05) differences were statistically determined among major cis FAs (PAM, SA, OLA, LO and LN) and their involved parameters (SFAs, MUFAs and PUFAs). The consumed several commercial boullions in Turkey were classified and characterized chemometric method (Principal Component Analysis, PCA) based on some fatty acid profiles and their parameters. Applying PCA to the all bouillon samples data determined the percentage of total variance explained by the first two PCs were 49.3% and 21.8% (totally 71.1%), respectively.

Fabrication of a monolithic adsorbent with multi-sites and its application in the extraction of active components from traditional Chinese medicine formula

A vinylic functionalized covalent organic framework (V-COF) was introduced to the preparation of a porous organic polymer (POP)/V-COF monolithic cartridge, via one-step polymerization reaction in a 50-mm-long and 4.6-mm-inner diameter stainless steel column. The fabricated POP/V-COF monolith was characterized with scanning electron microscope, Fourier transform infrared spectrometer and specific surface area measuring instrument, which indicate that the monolithic material in the cartridge occupies porous structure, multiple functional groups and high specific surface area of 188.2 m2 g−1. The POP/V-COF monolith was used as a solid-phase extraction (SPE) cartridge, which was online combined with an octadecyl-silane analytical column of high-performance liquid chromatography (HPLC) to extract, enrich and detect dihydrotanshinone Ⅰ, cryptotanshinone, tanshinone I and tanshinone ⅡA from traditional Chinese formula of POP/V-COF adsorbent. Furthermore, the POP/V-COF cartridge was used to extract and enrich the bornyl acetate from the Salvia Miltiorrhiza Drink by offline SPE procedure, and the extractive solution was subsequently analysed using capillary-gas chromatography method. These results show that the POP/V-COF SPE cartridge can remove the matrices of the Salvia Miltiorrhiza Drink, and simultaneously can extract the target active components with specific selectivity. Quantitative methodology validation indicates the applicable of this method for determination of the four active components in Salvia Miltiorrhiza Drink with the fabricated online SPE-HPLC. It is worth noting that the present cartridge can be reused for at least 100 times, with the RSD values based on the peak areas of the four components are less than 6.3% (n = 100).

The use of bacterial cultures to increase the content of conjugated linoleic acid in milk and dairy products

The bioactive properties of conjugated linoleic acid (CLA) are associated with human health promotion processes, including anticarcinogenic, antiatherogenic, antiinflammatory and antidiabetic activity, and the ability to reduce body fat. The functional properties of the bioactive component of the fat phase of cow’s milk, called conjugated linoleic acid (CLA), are described in this article. The content of conjugated linoleic acid in raw milk was studied using the capillary gas chromatography (GC) method. The content of CLA in raw milk was evaluated, taking into account the influence of the diet of feeding cows. Many samples of raw milk were studied in summer and winter for this purpose. It was determined that the concentration of conjugated linoleic acid in the fat phase of milk in winter is lower than in summer, when the diet of cows is dominated by green feed. The content of conjugated linoleic acid in the summer period ranged from 122.3 to 372.2 mg/dm 3 , and in winter it ranged from 54.7 to 136.47 mg/dm 3 . Experimental data of the possibility of increasing the content of conjugated linoleic acid in the fat phase of milk by inserting bacterial cultures is also presented. Re sults of the parameters of production conjugated linoleic acid under the influence of Lactobacillus acidophilus and Lactobacillus delbrueckii subsp . bulgaricus were obtained. The use of these bacterial cultures allowed to increase the content of conjugated linoleic acid by 23–36% in the studied samples.

Determination of oxidant dyes in hair dyes by gas chromatography

A capillary gas chromatographic method was adapted for the determination of nine dye components in oxidative hair dyes. An appropriate amount of sample was dissolved in 10 ml of alcohol containing 0.75% of diphenyl as internal standard. The solution was directly injected into an RTX-35 column, with temperatures ranging from 130-160°C. The recoveries achieved by this method, the effect of temperature and a survey of commercial hair dyes are discussed. The recoveries of nine oxidative hair dye components, i. e., three isomers of aminophenol, three isomers of phenylenediamine, catechol, resorcinol and pyrogallol, were in the range of 91.4-118.4% and 85.5-103.2% in liquid and cream samples, respectively. The temperature of storage affected the degradation of the oxidative hair dyes. Those stored at 5°C were degraded more slowly than those stored at 40°C. Fifty commercial hair dye smples were purchased between September 1993 and April 1994 from drugstores and supermarkets in Taipei. Sixty-six percent of these samples contained oxidative hair dye components. Sixteen percent of the smples were badly labeled. On the analyses of dyes, twenty-four percent of these samples were detected beyond the registration level (100±10%) . Two samples without registration contained oxidative hair dyes Nevertheless, all of the samples were within the hygiene requirements (aminophenol below 2%, phenylenediamine below 4%) .

Біохімічні особливості плодів лимонника китайського (Schizandra chinensis (Turcz.) Baill.)

Introduction. Today the searching of new innocuous plant substances of the introduced non-traditional plants is increasingly important due to its possibility of versatile use in the pharmaceutical industry. These include Chinese Magnolia vine (Schizandra chinensis (Turcz.) Baill.) – the valuable source of bioactive compounds, which are widely used in folk and scientific medicine.The aim of the study – to learn biochemical characteristics of fruits of Chinese Magnolia vine, introduced in Wood-steppe of Ukraine. The lipid fraction, the content of volatile compounds and the mineral composition of the Magnolia-vine (S. chinensis) fruits (the cultivar Sadovy – 1), selected in M. Hryshko National Botanic Garden of NAS ofUkraine, were carried out.Research Methods. As a raw material we used the Magnolia-vine fruits (the cultivar Sadovy – volatile compounds was implemented with chromatographic method, the content of fatty acids – with the method of capillary gas chromatography, the content of macro- and microelements – with X-ray fluorestcent method.Results and Discussions. The chemical composition of the S. chinensis fruits is characterized by a combination of the important bioactive components, including fatty and essential oils with the rich mineral complex of the macro- and microelements. The lipid complex of Magnolia-vine seeds is revealed a high content of unsaturated fatty acids with the highest percentage of linoleic acid (69.97 %). The 65 components are founded in volatile fractions of the fruits, and 75 components – in the seeds, the main of which are nerolidol, β-elemen, D. hermakren. The mineral complex is represented with 13 macro- and microelements, the main of which are potassium, calcium, sulfur, iron, manganese and zinc.Conclusions. The results confirm the high value of the fruits of Magnolia-vine, introduced in conditions of the Wood-steppe ofUkraine. The chemical composition of the fruit is distinguished by a combination of important bioactive components, including fatty and essential oils, rich mineral complex macro- and microelements, which has a wide range of pharmacological action. This gives the base to widespread cultivation of Magnolia-vine as a promising source of biologically active compounds that can be used to create the food additives and phyto-compositions of medical-prophylactic action.

A novel ultrashort capillary gas chromatography method using on-column injection and detection

A novel method for ultrashort capillary column gas chromatography (GC) analysis is introduced, which employs on-column injection and detection and rapid temperature programming. Using 10–20 cm long capillary columns, results showed that the method provides efficient and very rapid separations for relatively simple mixtures. Moreover, the on-column aspect of the method used here is demonstrated to avoid the extra column analyte degradation that can occur in traditional approaches to such separations. As a result, the developed method allows for the first time the GC analysis of some very large and (or) highly thermally labile analytes, such as polypeptides and drug molecules that are normally prone to decomposition. As an application, this method is further used to monitor pharmaceutical degradant formation as a function of temperature and was found to provide similar results to those obtained from conventional high-performance liquid chromatography analysis. Overall, the findings indicate that this ultrashort GC column approach could be useful in these areas and potentially others, where relatively simple GC analysis and universal flame ionization detection is desirable.

A Quantitative Assay for Fatty Acid Composition of Castor Seed in Different Developmental Stages

The aim of this study was to detect and investigate the fatty acid composition of castor seed under different development stage using soxhlet extraction and capillary gas chromatography methods to find the dynamic change of fatty acid composition and their relationships. The results showed that oil content and twelve fatty acid compositions were identified in the seed development, while the content of oil and compositions were significantly different and monitored during seed development. Oil content displayed a linear increase from the beginning of seed formation to maturation. Several compositions, mytistic acid, behenic acid and lignoceric acid, were just found at initial seed filling phase and significantly declined with seed maturity, suggesting that fatty acid composition of castor seed in the initial phases of seed formation differed substantially form that of the mature seeds. The correlation analysis results revealed significant positive correlations of seed development time with mostly fatty acids, further demonstrating that the fatty acids were closely correlated with seed development. These obtained results will benefit for breeding research in high oil content of castor.

Determination of ethylene glycol in workplace air by capillary column gas chromatography

To establish the method of capillary column gas chromatography for determination of ethylene glycol in workplace air. Ethylene glycol in workplace air was collected with silicone tube, desorbed with methanol, separated with FFAP (nitroterephthalic acid-modified polyethylene glycol)capillary column, and measured with flame ionization detector. The detection limit of ethylene glycol was 0.41 mg/L, the lower limit of quantification was 1.4 mg/L, the range of measurement was 1.4~163.9 mg/L, and the minimum detectable concentration was 0.3 mg/m3 (1.5 L of air was collected as the sample). This method had a good repeatability, the relative standard deviation was 1.4%~5.2%, the average desorption efficiency was 94.4%~101.7%, and the sampling efficiency was 99.2%~100%. The penetrating capacity of 200 mg silicone was higher than 6.9 mg, and the samples could be preserved for 14 days at room temperature. The method has a low detection limit, high accuracy, and good precision, which is feasible for determination of ethylene glycol in workplace air.

Trace Level Determination of Mesityl Oxide and Diacetone Alcohol in Atazanavir Sulfate Drug Substance by a Gas Chromatography Method.

A capillary gas chromatography method with a short run time, using a flame ionization detector, has been developed for the quantitative determination of trace level analysis of mesityl oxide and diacetone alcohol in the atazanavir sulfate drug substance. The chromatographic method was achieved on a fused silica capillary column coated with 5% diphenyl and 95% dimethyl polysiloxane stationary phase (Rtx-5, 30 m x 0.53 mm x 5.0 µm). The run time was 20 min employing programmed temperature with a split mode (1:5) and was validated for specificity, sensitivity, precision, linearity, and accuracy. The detection and quantitation limits obtained for mesityl oxide and diacetone alcohol were 5 µg/g and 10 µg/g, respectively, for both of the analytes. The method was found to be linear in the range between 10 µg/g and 150 µg/g with a correlation coefficient greater than 0.999, and the average recoveries obtained in atazanavir sulfate were between 102.0% and 103.7%, respectively, for mesityl oxide and diacetone alcohol. The developed method was found to be robust and rugged. The detailed experimental results are discussed in this research paper.

Enantioseparation of Methyl 2-Hydroxypropionate with Two Peracylated β-Cyclodextrin Derivatives as CGC Chiral Stationary Phases

Two peracylated β-cyclodextrin derivatives, 2,3,6-tri-O-valeryl-β-cyclodextrin and 2,3,6-tri-O-octanoyl-β-cyclodextrin, were synthesized and used as capillary gas chromatography (CGC) chiral stationary phases. The two peracylated β-cyclodextrin derivatives showed enantioseparation abilities to some chiral compounds tested, such as enantiomers of methyl 2-chloropropionate, 5-hydroxy-4, 4-dimethyl-dihydrofuran-2-one, and methyl 2-hydroxypropionate. As CGC chiral stationary phases, the two peracylated β-cyclodextrin derivatives showed excellent enantioseparation abilities to methyl 2-hydroxypropionate (i.e., methyl lactate). On 2,3,6-tri-O-valeryl-β-cyclodextrin, the enantiomer separation factor and resolution of methyl lactate were 1.21 and 6.08, respectively, and on 2,3,6-tri-O-octanoyl-β-cyclodextrin, 1.22 and 5.49, respectively. The enantioseparation results of methyl lactate on the two peracylated β-cyclodextrin derivatives were better than on most reported β-cyclodextrin derivatives, including a commercial chiral column of permethylated β-cyclodextrin tested in this study. Results suggest that CGC methods using 2,3,6-tri-O-valeryl-β-cyclodextrin or 2,3,6-tri-O-octanoyl-β-cyclodextrin as chiral stationary phase could be used for enantioseparation and evaluating the enantiomeric optical purity of methyl lactate.

Development and Validation of a Gas Chromatography Method for the Trace Level Determination of Allylamine in Sevelamer Hydrochloride and Sevelamer Carbonate Drug Substances

A capillary gas chromatography method using a flame ionization detector has been developed for the trace analysis of allylamine (AA) in sevelamer hydrochloride (SVH) and sevelamer carbonate (SVC) drug substances. The method utilized a mega bore capillary column DB-CAM (30 m × 0.53 mm × 1.0 μm) with a bonded and cross-linked, base-deactivated polyethylene glycol stationary phase and was validated for specificity, sensitivity, precision, linearity, and accuracy. The detection and quantitation limits obtained for allylamine were 2 μg/g and 6 μg/g, respectively. The method was found to be linear in the range between 6 μg/g and 148 μg/g with a correlation coefficient of 0.9990. The average recoveries obtained in SVH and SVC were 93.9% and 99.1%, respectively. The developed method was found to be robust for the determination of AA in sevelamer drug substances and also the specificity was demonstrated with a gas chromatograph coupled with a mass spectrometer.

Analysis and Sensory Evaluation of Jostaberry (Ribes x nidigrolaria Bauer) Volatiles

Volatiles of jostaberries (Ribes x nidigrolaria Bauer)--a hybrid of black currant (Ribes nigrum L.) and gooseberry (Ribes uva-crispa L.)--were isolated via vacuum headspace extraction and analyzed by capillary gas chromatographic methods for the first time. (E)-Hex-2-enal, (E)-hex-2-en-1-ol, (Z)-hex-3-enal, (Z)-hex-3-en-1-ol, methyl butanoate, ethyl butanoate, 2-methylbut-3-en-2-ol, and 1,8-cineol turned out to be the most dominant volatiles. The variability of the volatile profile was shown by the analysis of jostaberries harvested from different locations in Southern Germany and in different years. In addition to ripe jostaberries, underripe berries were also investigated and changes in the volatile profile were followed during the ripening process. By using sensory analysis, key aroma compounds were elucidated. An aroma model prepared by mixing most odor active compounds ((Z)-hex-3-enal, 1,8-cineol, ethyl butanoate, (E)-hex-2-enal, (E)-hex-3-enal, hexanal, pent-1-en-3-one, methyl butanoate, ethyl hexanoate, and oct-1-en-3-one) in their naturally occurring concentrations showed an overall aroma very similar to that of fresh jostaberries.

Determination of Residual Organic Solvents in Antitumor di-phenyl-di-(2,4-dichlorobenzohydroxamato)tin by Capillary Gas Chromatography

Abstract. To determine the residual organic solvents(methanol, dichloromethane, normal hexane ) in antitumor crude drug di-phenyl-di-(2,4-dichlorobenzohydroxamato)tin(DPDCT), capillary gas chromatography (GC) method was established. The HP-5 (30m×320μm×0.25μm) capillary column and flame ionization detector (FID) detector were adopted. The carrier gas was nitrogen with flow rate 5.08 ml·min-1. The column temperature was 50.0°C and column pressure was 20.00psi. The distributary ratio was 20:1. 1, 2-dichloroethane was used as the internal standard substance while toluene as the solvent to determine the residual organic solvents. The calibration curves for methanol, dichloromethane and normal hexane are linear in the rang of 180.6～481.5μg·ml-1(r2 = 0.9974), 35.78～95.40μg·ml-1(r2 = 0.9950), 17.82～47.52μg·ml-1(r2 = 0.9995), respectively. The average recoveries of methanol, dichloromethane and normal hexane are 100.4 %( RSD=1.8%), 100.3 %( RSD=1.7%), 99.6 %( RSD=1.5%), respectively. The method is a simple and accurate method with high sensitivity and repeatability. It is suitable for the residual solvent determination of antitumor DPDCT.

Spatial patterns of total petroleum hydrocarbons in the terrestrial environment at McMurdo Station, Antarctica

AbstractFossil fuels are used throughout the United States Antarctic Program. Accidental releases of petroleum hydrocarbons are the leading source of environmental contamination. Since 1999 McMurdo Station has been the site of the most extensive environmental monitoring programme in Antarctica. Nearly 2500 surface soil samples were collected from 1999–2007 to determine the spatial “footprint” of petroleum hydrocarbons. Total petroleum hydrocarbons (TPH) concentrations were measured using a high-resolution capillary gas chromatographic method with flame ionization detection. Three distinct TPH patterns were detected: low molecular weight gasoline/JP5/AN8, residual weathered petroleum and an unresolved complex mixture of high molecular weight material. Overall TPH concentrations were low with 38% of the samples having TPH concentrations below 30 ppm and 58% below 100 ppm. Total petroleum hydrocarbon concentrations above 30 ppm are largely confined to the central portions of the station, along roads and in other areas where elevated TPH would be expected. Peripheral areas typically have TPH concentrations below 15 ppm. Areas of elevated TPH concentrations are patchy and of limited spatial extent, seldom extending over distances of 100 m. This environmental monitoring programme is ongoing and can serve as an example to other Antarctic programmes concerned with monitoring environmental impacts.

Determination of glyceryl trinitrate and its two main metabolites in human plasma using a new sensitive gas chromatography method

The aim of the present study was to determine the concentrations of nitroglycerin (glyceryl trinitrate, GTN, CAS 55-63-0) and its two main stable metabolites; 1,2-dinitroglycerin (1,2-glyceryl dinitrate, GDN, CAS 621-65-8) and 1,3-dinitroglycerin (1,3-GDN, CAS 623-87-0) in human plasma using a capillary gas chromatography method with an electron capture detection. Using the GC conditions, linear calibrations were obtained for 1,3-GDN from 0.14 to 3 ng/mL, for 1,2-GDN from 0.06 to 6 ng/mL, and for GTN from 0.01 to 0.3 ng/mL in plasma samples by the following calibration curve equations: [y = 0.1924x - 0.0088 (r = 0.999)], [y = 0.2273x + 0.0164 (r = 0.995)], [y = 17.434x - 0.0751] for 1,3-GDN, 1,2-GDN, and GTN respectively. The calculated limits of quantification values for GTN, 1,2-GDN, and 1,3-GDN were 0.03 ng/mL, 0.2 ng/mL, and 0.15 ng/mL respectively. This method was verified with a bioequivalence study of an Iranian brand of oral sustained release nitroglycerin with an innovator formulation.

Compositional features of carbohydrate compound from rhizoma ligustici wallichii and ethanol extract of danshen and its bioactivity

Abstract Water extraction was applied to prepare carbohydrate compound of rhizoma ligustici wallichii. Four main fractions, fraction-I, fraction-II, fraction-III, and fraction-IV, were obtained by membranes of 1.0 × 10 −4 mm pore size and normal molecular-weight cut-off of 50 kDa. The resulting four preparations were further analysed by capillary gas chromatography method. Thin layer chromatography (TLC) analysis showed that carbohydrate compound of rhizoma ligustici wallichii was composed of five types of monosaccharides, namely glucose, rhamnose, mannose, galactose and arabinose. Gas chromatography (GC) analysis showed that fraction I of rhizoma ligustici wallichii was composed of four types of monosaccharides, namely glucose, mannose, galactose and arabinose at a molar ratio of 521:1:4.6:3.3. Furthermore, the protective effect of the Rhizoma ligustici wallichii polysaccharides and ethanol extract of danshen against ischemia–reperfusion (IR) induced renal injury were evaluated. The findings imply that carbohydrate compound of the Rhizoma ligustici wallichii and ethanol extract of danshen play a causal role in IR-induced renal injury probably by the radical scavenging and antioxidant activities. Moreover, ethanol extract of danshen displayed stronger renoprotective effect than that of carbohydrate compound of the Rhizoma ligustici wallichii.

Effect of sowing time on grain yield, oil content, and fatty acids in rapeseed (Brassica napus subsp. oleifera)

Sowing time is an important factor affecting crop yield and quality, especially in arid and semi-arid regions. The aim of this research was to investigate the effects of sowing time on the growth, yield, and quality of rapeseed genotypes and to determine genotype × sowing time interactions for grain yield and its related traits. The study was carried out using 8 winter rapeseed genotypes (H604049, H604038, H604041, Viking, Elan, Titan, Lorenz, and Trabant) and 4 sowing times (10 October, 20 October, 30 October and 10 November) during the 2005-2006 and 2006-2007 growing seasons. An NIRS system was used to determine oil, protein, glucosinolate and sinapic acid esters. Fatty acid analyses were done by the capillary gas chromatography method. Significant differences were found between sowing times for most of the traits measured. The lowest average seed yield (1027.40 kg ha^{-1}) was obtained from the latest sowing time, whereas the highest average seed yield (2437.50 kg ha^{-1}) was obtained from the earliest sowing time. The genotype H604038 produced the highest seed yield (1988.4 kg ha^{-1}), and it was followed by Trabant (1980.8 kg ha^{-1}) and Titan (1963.8 kg ha^{-1}). The highest oil content, at 42.0%, was obtained from genotype Lorenz at the first sowing time and from genotype Trabant at the second sowing time. No interaction was found between genotype and sowing time for oil content, but significant interactions were found for seed yield and glucosinolates. Seed yield significantly decreased as sowing time was delayed. The effects of sowing time on fatty acid composition were also significant. As a result, it was found that sowing time is an important factor for seed yield and quality in rapeseed.
