Simultaneous efflux of endogenous D‐ser and L‐glu from single acute hippocampus slices during oxygen glucose deprivation

D-serine and L-glutamate play crucial roles in excitotoxicity through N-methyl-D-aspartate receptor coactivation, but little is known about the temporal profile of efflux during cerebral ischemia. We utilized a newly designed brain slice microperfusion device coupled offline to capillary electrophoresis laser-induced fluorescence to monitor dynamic efflux of endogenous D-ser and L-glu in response to oxygen glucose deprivation (OGD) in single acute hippocampus slices. Efflux profiles with 2-min temporal resolution in response to 24-min OGD show that efflux of D-ser slightly precedes efflux of L-glu by one 2-min sampling interval. Thus both coagonists are available to activate NMDA receptors by the time when glu is released. The magnitude of D-ser efflux relative to baseline values is, however, less than that for L-glu. Peak efflux during OGD, expressed as pre-OGD baseline values, was as follows: D-ser 254% +/- 24%, L-glu 1,675% +/- 259%, L-asp 519% +/- 128%, and L-thr 313% +/- 33%. L-glutamine efflux was shown to decrease significantly in response to OGD. The microperfusion/CE-LIF approach shows several promising attributes for studying endogenous chemical efflux from single, acute brain slices.

Determination of NAD+and NADH in a Single Cell under Hydrogen Peroxide Stress by Capillary Electrophoresis

A capillary electrophoresis (CE) method based on an enzymatic cycling reaction is developed to determine both NAD(+) and NADH in a single cell in a single run. The detection limit can reach down to 0.2 amol of NAD(+) and 1 amol of NADH with a homemade capillary electrophoresis laser-induced fluorescence (CE-LIF) setup. This method shows good reproducibility and specificity. After an intact cell is injected into the capillary and lysed using a Tesla coil, intracellular NAD(+) and NADH were separated, incubated with the cycling buffer, and quantified by recording the amount of fluorescent product generated. Cellular NAD(+) and NADH levels of a rat myoblast cell line were determined using this method. Both NAD(+) and NADH levels decreased when the cells were exposed to oxidative stress induced by H(2)O(2). This may be due to the activation of the DNA repair enzyme, poly(ADP-ribose) polymerase, in response to the oxidative damage imposed on DNA, since pretreatment of the cells with an inhibitor of these enzymes prevented the reduction of cellular NAD(+) and NADH levels.

Separation and Characterization of an IgG2 Antibody Containing a Cyclic Imide in CDR1 of Light Chain by Hydrophobic Interaction Chromatography and Mass Spectrometry

Hydrophobic interaction chromatography (HIC) was used to separate populations of recombinant IgG2 antibody that were created as a result of prolonged incubation at 40 degrees C. Antibody was separated by HIC into three major and seven minor fractions. All but one fraction was composed of antibody with distinct chemical modifications that resulted from exposure to elevated temperature. The results of intact and reduced mass analysis as well as peptide map data derived from the three major HIC fractions indicated that the antibody was being chromatographically separated into populations containing a succinimidyl intermediate in complementarity determining region 1 (CDR1) on zero, one, and two light chain arms. Lower level species purified by HIC were analyzed by intact and reduced mass analysis and laser-induced fluorescence capillary electrophoresis (CE-LIF) and consisted of an antibody that was clipped in four different places in the heavy chain as well as misfolded and aggregated antibody. The potency of the recombinant antibody containing a succinimidyl intermediate on zero, one, and two light chain arms was analyzed by LANCE binding assay and a cell based in vitro bioassay, and the occurrence of this modification on one or both light chain arms was associated with a reduction in the binding affinity of the molecule to the target by approximately 10%. We show that HIC has the unique ability as a first step purification method to separate populations of antibody which are covalently modified under stability programs. The method conditions that have been developed for the HIC assay are ideal for purifying antibodies with labile modifications for the purpose of further characterization.

Capillary Electrophoresis with Laser-Induced Fluorescence Detection as a Tool for Enzyme Characterization and Inhibitor Screening

An effective, rapid and economical CE/LIF (capillary electrophoresis/laser-induced fluorescence) method was developed and applied to the characterization of signal peptidase (SPase) enzyme, which is a target for the screening of new drug candidates. In this method, CE separates the product from the substrate and LIF selectively detects the fluorescence-labeled product and substrate. By measuring the increase of the product as a function of time, one can monitor the progression of the enzyme reaction. The progression curves were also used for screening inhibitors for this enzyme. The effects of various reaction conditions were also studied and discussed. In addition, this CE/LIF method was applied to the determination of the enzyme activity, the quality control of the substrate and/or enzymes, and the cross-reactivity of inhibitors to the enzyme. It can be concluded that this method is suitable for high throughput screening (HTS) assays because it can deliver fast, sensitive, quantitative, and reliable results.

Use of cyclodextrins as modifiers of fluorescence in the detection of mycotoxins

Cyclodextrins, cyclic oligosaccharides composed of amylose subunits, are known to interact with mycotoxins. The interactions may be useful to analytical chemists by altering the properties of the mycotoxin of interest, namely the chromatographic properties, electrophoretic properties, fluorescence, or absorption of these fungal metabolites. Practical applications of these effects have been the incorporation of cyclodextrins into high-performance liquid chromatography and capillary electrophoresis methods for mycotoxin detection. Specific mycotoxins include those with a native fluorescence such as the aflatoxins, ochratoxin A (OTA) and zearalenone (ZEN) as well as those that can be rendered fluorescent through derivatization, such as T-2 toxin. The literature describing the applications of cyclodextrins in mycotoxin analysis is reviewed and an attempt to extend the use of cyclodextrins to the detection of labelled T-2 toxin is presented. Twenty cyclodextrins were evaluated for their ability to enhance the fluorescence emission of T-2 toxin derivatized with pyrene-1-carbonyl cyanide (T2-Pyr). This evaluation revealed that heptakis (2,6-di-O-methyl)-β-cyclodextrin (DIMEB), in particular, enhanced T2-Pyr fluorescence. DIMEB was used as a buffer modifier in a capillary electrophoresis-laser-induced fluorescence (CE-LIF) method for detecting T-2 in maize. Because of the effects that certain cyclodextrins have, especially under aqueous conditions, they may make useful additives for a variety of mycotoxin analytical methods.

Protective action of bacterial melanin against DNA damage in full UV spectrums by a sensitive plasmid-based noncellular system

The purpose of this study was to introduce a simple and sensitive plasmid-based noncellular system to evaluate the photoprotection of bacterial melanin on DNA damage against ultraviolet (UV) radiation. Plasmid DNA was used to assess the role of melanin in different ranges of UV using a series of in vitro assays. Fluorometric measurements suggested that melanin could efficiently scavenge reactive oxygen species (ROS) generated by UVA irradiation in solution, and the scavenging capability was proportional to the pigment concentration. The protective effect of melanin on plasmid DNA under UVB irradiation was confirmed by the transformation efficiency of the protected DNA, which was at least 10-fold higher than that of the non melanin protected DNA. After the UVC irradiation, the DNA damage of strand breaks was quantified by laser-induced fluorescence capillary electrophoresis. The percentage of supercoiled plasmid was reduced from 80% to less than 5% without melanin protection. In contrast, the percentage of supercoiled DNA only decreased to about 40% in the presence of melanin under the same radiation conditions. All these results demonstrated that bacterial melanin did protect DNA from being damaged throughout full UV irradiation. This system, avoiding the potential interference by cellular DNA repair machinery and intracellular substances, may provide a sensitive in vitro means to evaluate the functions of melanin and other photoprotective compounds from different sources.

Taurine determination by capillary electrophoresis with laser-induced fluorescence detection: from clinical field to quality food applications

In this work we describe a new method for taurine quantification in plasma by capillary electrophoresis laser-induced fluorescence detection. Taurine is derivatized with fluorescein isothiocyanate at 100 degrees C in 20 min. These conditions allow to reduce the pre-analytical times and to derivatize quantitatively the taurine contained in the reaction mixture, contrary to the room temperature derivatization commonly adopted. FITC-taurine adduct is analyzed in an uncoated fused-silica capillary, 75 mum ID and 40 cm effective length using a 20 mmol/L tribasic sodium phosphate buffer pH 11.8, at 22 kV. To avoid the typical problems due to instability of FITC-adduct, we use the homocysteic acid as internal standard. The loss of FITC-taurine signal during the sequence analysis is compensated by the same loss of FITC-internal standard adduct, thus giving a noteworthy improvement in the assay precision. The method shows a good reproducibility of the migration times (coefficient of variation, CV%, 1.93) and the peak areas (CV%, 3.65). Intra- and interassay CV were 4.63 and 6.44%, respectively, and analytical recovery was between 98.1 and 102.3%. Assay application was tested measuring taurine plasma levels in 50 healthy volunteers in which a mean value of 60.2 +/- 17.9 micromol/L was found. Moreover, the applicability of the method was also checked on energy drinks and milk.

Effects of sulpiride on mRNA levels of steroid 5α-reductase isozymes in prostate of adult rats

Prolactin (PRL) is implicated in prostate growth and in the development and regulation of benign prostatic hypertrophy (BPH) and prostate cancer (PCa). PRL may exert its effects on prostate in synergism with androgens. The most active androgen in the prostate is the 5alpha-dihydrotestosterone (DHT) obtained from testosterone by the 5alpha-reductase (5alpha-R) enzyme, which is expressed in the prostate as two isozymes, 5alpha-R1 and 5alpha-R2. In this study, sulpiride, a prolactin-secretion inductor, was administered to male rats. mRNA levels of 5alpha-R1 and 5alpha-R2 were measured in prostate of controls and sulpiride-treated rats, using one-step quantitative RT-PCR coupled with laser-induced fluorescence capillary electrophoresis (LIF-CE). Results demonstrated that sulpiride-induced hyperprolactinemia is associated with an increase in mRNA levels of both 5alpha-R1 and 5alpha-R2 in prostate of adult rats. Although a direct effect of sulpiride on prostate gland cannot be ruled out, hyperprolactinemia may be a factor to be considered in aging males, in whom prostatic diseases such as BPH and PCa are more frequent.

Laser-Induced Fluorescence−Capillary Electrophoresis and Fluorescence Microplate Reader Measurement: Two Methods to Quantify the Effect of Antibiotics

Putative antibiotic drugs have to be classified according to their bactericidal potential. Two new methods by means of fluorescence spectroscopy (FS) using a fluorescence microplate reader (FMR) and laser-induced fluorescence capillary electrophoresis (LIF-CE), respectively, were developed for the assessment of the bactericidal efficiency using the LIVE/DEAD BacLight viability kit composed of the two fluorescent nucleic acid stains, SYTO9 (stains all cells green) and propidium iodide (stains cells with damaged membrane red). By correlation of the ratio of green and red fluorescence with the percentage of live cells by employing FS and LIF-CE, respectively, dose response curves of drug-treated Pseudomonas spp. and Streptococcus spp. samples were calculated, which allowed for the assessment of ED50 values. Both methods led to similar results which were in agreement with the minimum inhibitory concentrations (MICs) obtained by conventional broth microdilution. The application of the BacLight viability kit on drug-treated bacteria cultures presents a rapid method of assessing the antibiotic potency which is of great importance for high throughput screening in the development of new antibiotics. Additionally, the new LIF-CE method, which based on the use of a second unlabeled bacteria injection as a stacking front, allowed drawing conclusions from the electrophoretic profile about the constitution of the bacterial population. Thus, the tendency of bacterial chain formation and alterations in the live/dead ratio of the bacterial composition can be directly observed in the presence of different antibiotics.

Detection of G Protein Coupled Receptor Mediated Adenylyl Cyclase Activity by Capillary Electrophoresis Using Fluorescently Labeled ATP

A capillary electrophoresis (CE) laser-induced fluorescence (LIF) assay was developed for the detection of G protein coupled receptor mediated adenylyl cyclase (AC) activity using BODIPY FL ATP (BATP) as substrate. In the assay, cell membranes coexpressing the stimulatory G protein fused to the beta2 adrenergic receptor (beta2AR) and AC were incubated with BATP, the resultant mixture injected, and BATP separated from product BODIPY FL cAMP (BcAMP) by CE. AC activity was quantified by measuring the rate of BcAMP formation. beta2AR agonists isoproterenol and terbutaline increased basal AC activity with EC50s of 2.4 +/- 0.2 and 60 +/- 9 nM, respectively. The antagonist propranolol competed with terbutaline for beta2AR binding sites and expectedly right-shifted the terbutaline dose-response curve to 8 +/- 3 microM. The high sensitivity of the assay was demonstrated by detection of small changes in AC activity, with the partial agonist alprenolol increasing (22 +/- 1%) and the inverse agonist ICI 118,551 decreasing (19 +/- 2%) basal activity. The simplicity and automation of the CE-LIF assay offers advantages over the more traditional assay using radiochemical ATP and column chromatography.

Effects of swim stress on mRNA and protein levels of steroid 5α-reductase isozymes in prefrontal cortex of adult male rats

The metabolite of progesterone, allopregnanolone, is among the most potent known ligands of the γ-aminobutyric acid receptor complex (GABA A-R) in the central nervous system. This neuroactive steroid is markedly increased in an animal model of acute stress. Allopregnanolone is synthesized from progesterone by steroidogenic enzymes 5α-reductase (5α-R) and 3α-hydroxysteroid dehydrogenase (3α-HSD), with the former being the rate-limiting enzyme in this reaction sequence. In this paper, a quantitative RT-PCR method coupled to laser-induced fluorescence capillary electrophoresis (LIF-CE) and Western blot were used to measure both mRNA and protein levels of 5α-R type 1 (5α-R1) and 5α-R type 2 (5α-R2) isozymes in prefrontal cortex of male rats after acute swim stress situations. Our results demonstrate that both 5α-R isozymes are significantly higher in prefrontal cortex of male rats after acute swim stress in comparison with control rats. These data may open up a new research line that could improve our understanding of the role of 5α-R isozymes in processes that accompany stress situations.

Analysis of amino acid neurotransmitters by capillary electrophoresis and laser-induced fluorescence using a new fluorescein-derived label

A capillary electrophoresis laser-induced fluorescence detection method (CE-LIF) was developed for the separation of eight neurotransmitters tagged on their amino function with 6-oxy-(N-succinimidyl acetate)-9-(2′-methoxycarbonyl) fluorescein (SAMF), a new fluorescent reagent synthesized in our lab. Derivatization was performed in boric acid buffer (pH = 7.75) at 37 °C over 15 min. The pH-independent fluorescence of SAMF (pH 4–9) permits background buffers over a wide range of pH. It was demonstrated that an acidic running buffer offers a better resolution compared to basic medium in terms of resolution and peak shapes. Employing Cu2+ as the additive, the molecules were baseline-separated using a running buffer consisting of 40 mM sodium acetate and 2 mM Cu2+ (pH 6.0). The detection limits ranged from 1 to 2 × 10−10 M. The method has been validated for the characterization of lymphocyte samples. The results obtained illustrate the advantages of combining SAMF derivatization with CE-LIF for determining neurotransmitters.

Capillary electrophoresis of the mycotoxin zearalenone using cyclodextrin-enhanced fluorescence

Certain of the cyclodextrins are capable of significantly enhancing the native fluorescence of the estrogenic mycotoxin zearalenone (ZEN). Twenty-two cyclodextrins (CDs) were screened for their ability to enhance the fluorescence of ZEN in a capillary electrophoresis-laser induced fluorescence (CE-LIF) format. Of the CDs that were examined heptakis (2,6-di-O-methyl)-β-CD gave the greatest enhancement. The heptakis (2,6-di-O-methyl)-β-CD was applied to the development of a CE-LIF method for detection of ZEN in maize. The resulting method was capable of detecting ZEN with a limit of quantitation of 5ng/g maize. Recoveries of ZEN from maize spiked over the range from 5ng/g to 500ng/g averaged 103.1±8.5% (n=20). The CE-LIF method will be useful for future studies of ZEN in maize.

Plasma Homocysteine and Cysteine Levels in Retinal Vein Occlusion

To determine plasma homocysteine and cysteine levels in patients with retinal vein occlusion (RVO) and in healthy subjects and to ascertain whether there are statistically significant differences between patients and control subjects. In this case-control study, the study group consisted of 75 consecutive patients with RVO: 33 had central retinal vein occlusion (CRVO), and 42 had branch retinal vein occlusion (BRVO). Seventy-two apparently healthy age- and sex-matched subjects served as control subjects. Homocysteine and cysteine levels were measured with a new laser-induced fluorescence capillary electrophoresis (CE-LIF) METHOD: Wilcoxon or Student's t-test was used, when appropriate, to determine differences between groups. There were no significant differences in median plasma homocysteine between patients with RVO and control subjects, nor were there any statistically significant differences when patients were categorized by type of vein occlusion (CRVO or BRVO). Similarly, there were no significant differences in mean plasma cysteine between patients with RVO and control subjects. However, when categorized by type of vein occlusion, mean plasma cysteine was significantly higher in CRVO patients than in control subjects (P = 0.034). This study failed to demonstrate an association between increased plasma homocysteine and RVO. Mean plasma cysteine was significantly higher in patients with CRVO, suggesting that hypercysteinemia may contribute to the pathogenesis of this retinal vascular disorder.

Effects of testosterone on brain mRNA levels of steroid 5α-reductase isozymes in early postnatal life of rat

The enzyme 5α-reductase (5α-R) (EC 1.3.99.5) exists as two isoforms, 5α-R type 1 (5α-R1) and 5α-R type 2 (5α-R2), and both are present in the brain. 5α-R1 has been proposed as a constitutive enzyme that essentially plays a catabolic and neuron protective role whereas 5α-R2 has been associated with sexually dimorphic functions of the male. In this work, we studied the effects of testosterone (T), the masculinizing hormone of the central nervous system (CNS), on mRNA levels of both 5α-R isoforms in the prefrontal cortex of male and female rats during the postnatal sexual differentiation of the CNS in the rat, using one-step quantitative RT-PCR coupled with laser-induced fluorescence capillary electrophoresis (LIF-CE). We found an increase in 5α-R2 mRNA levels in both male and female rats after T treatment, while 5α-R1 mRNA levels were decreased in the same experimental conditions. Our results clearly indicated that T regulates the expression of both 5α-R1 and 5α-R2 genes in an opposite manner and independently of the sex. This could point to a crucial role of T in the sexual dimorphism for both 5α-R isozymes in the neonatal brain. These results open up a new research line that could improve understanding of the role of 5α-R isozymes in the physiology of the CNS.

Capillary electrophoresis laser-induced fluorescence for screening combinatorial peptide libraries in assays of botulinum neurotoxin A

Botulinum neurotoxin serotype A (BoNT/A) is a proteolytic enzyme that induces muscle paralysis. It is a cause of food poisoning, a potential bioterrorist threat and, in low doses an emerging pharmaceutical product. No effective treatment is currently available for BoNT intoxication. Previously we developed a BoNT/A light chain enzyme assay using a peptide substrate based on the SNAP-25 protein target, with HPLC separation and UV detection of assay products, and applied the method to screen combinatorial peptide libraries for inhibitory activity to BoNT/A. We now report on development of a capillary electrophoresis laser-induced fluorescence (CE-LIF) method for measuring BoNT/A activity. The enzyme assay products were labeled with CBQCA dye followed by CE separation on a bare fused silica column in a HEPES-based buffer and LIF detection. All assay products were separated in CE within 8 min compared to incomplete separation of assay products within 1h by HPLC. The labeled products showed linear dependence of intensity versus concentration, and quantitative mole-fraction assignments. We used the CE-LIF method to screen combinatorial peptide libraries for potential modulating effects on BoNT/A peptidase activity. With some of the libraries, peptides co-migrated with assay products and interfered with quantitation. In such cases, interference was reduced by substituting sodium dodecyl sulfate (SDS) for Tween-20 in the running buffer. Separation in the capillaries then occurred by micellar electrokinetic chromatography (MEKC). The CE-LIF method is quick and lends itself to high-throughput or microfluidic formats.

Capillary electrophoresis–mass spectrometry as a characterization tool for therapeutic proteins

With the increasing use of capillary electrophoresis (CE) in the biotechnology industry, there is a demand for analytical tools and methodology that can be used to characterize CE profiles. This article describes the implementation and optimization of a robust online CE–mass spectrometry (CE–MS) system used for the characterization of several CE assays developed at Genentech Inc. These assays include CE as a complement to reverse-phase peptide mapping for the identification of small peptides eluting in the void volume, profiling N-linked glycopeptide heterogeneity, and determining O-linked site occupancy. In addition, CE–MS was used to confirm major 8-aminopyrene-1,3,6-trisulfonate (APTS)-labeled glycans released from recombinant antibodies that are routinely profiled by CE–laser-induced fluorescence (CE–LIF). For each study, CE–MS was able to successfully identify components seen in UV or LIF electropherograms, thereby expanding the capability of CE and CE–MS for profiling biomolecules.

Steroid 5α-reductase isozymes in the adult female rat brain: central role of dihydrotestosterone

The enzyme 5alpha-reductase (5alpha-R) (EC 1.3.99.5) exists as two isoforms, 5alpha-R type 1 (5alpha-R1) and 5alpha-R type 2 (5alpha-R2). 5alpha-R1 has been associated with catabolic functions whereas 5alpha-R2 has been associated with sexually dimorphic functions of the male. We recently demonstrated that both 5alpha-R isozymes are present in the central nervous system (CNS) of the adult male rat and are regulated in an opposing way by androgens. This finding raises the question as to whether both isozymes play a role in the sexual dimorphism of the CNS, besides other functions. To test this hypothesis, it is essential to study the regulation of both isozymes by androgens in the female. In this work, we studied the effects of testosterone (T) and dihydrotestosterone (DHT) on mRNA levels of both 5alpha-R isoforms in the prefrontal cortex of the adult female rat by one-step quantitative RT-PCR coupled with laser-induced fluorescence capillary electrophoresis. Our results demonstrate for the first time that 5alpha-R2 mRNA is slightly regulated by T and DHT in females. Surprisingly, 5alpha-R1 mRNA is not regulated by T in the intact female, whereas it is very positively regulated by DHT, a more potent androgen than T. These data indicate the great sexual dimorphism in the CNS with respect to both 5alpha-R isozymes, and suggest a crucial role of DHT in the sexual dimorphism of the CNS in the female. These results open up a new research line that may lead to a better understanding of the physiology of the CNS.

Detection of Adenylyl Cyclase Activity Using a Fluorescent ATP Substrate and Capillary Electrophoresis

A capillary electrophoresis laser-induced fluorescence (CE-LIF) assay was developed for detection of adenylyl cyclase (AC) activity using BODIPY FL ATP (BATP) as substrate. In the assay, BATP was incubated with AC and the resulting mixture of BATP and enzyme product (BODIPY cyclic AMP, BcAMP) separated in 5 min by CE-LIF. Substrate depletion and product accumulation were simultaneously monitored during the course of the reaction. The rate of product formation depended upon the presence of AC activators forskolin or Galpha(s)-GTPgammaS as evidenced by a more rapid BATP turnover to BcAMP compared to basal levels. The CE-LIF assay detected EC50 values for forskolin and Galpha(s)-GTPgammaS of 27 +/- 6 microM and 317 +/- 56 nM, respectively. These EC50 values compared well to those previously reported using [alpha-32P]ATP as substrate. When AC was concurrently activated with 2.5 microM forskolin and 25 nM Galpha(s)-GTPgammaS, the amount of BcAMP formed was 3.4 times higher than the additive amounts of each activator alone indicating a positively cooperative activation by these compounds in agreement with previous assays using radiolabeled substrate. Inhibition of AC activity was also demonstrated using the AC inhibitor 2'-(or-3')-O-(N-methylanthraniloyl) guanosine 5'-triphosphate with an IC50 of 9 +/- 6 nM. The use of a fluorescent substrate combined with CE separation has enabled development of a rapid and robust method for detection of AC activity that is an attractive alternative to the AC assay using radioactive nucleotide and column chromatography. In addition, the assay has potential for high-throughput screening of drugs that act at AC.

Single-cell analysis by intracellular immuno-reaction and capillary electrophoresis with laser-induced fluorescence detection

A novel method for single-cell analysis was developed by combining electroporation for intracellular immuno-reaction and capillary electrophoresis (CE) with laser-induced fluorescence (LIF) detection. Human interferon-gamma (IFN-γ) in natural killer (NK) cells was chosen as the test antigen. Two forms of IFN-γ in single cells could be well separated and detected with a limit of detection of zeptomole. In this assay, the anti-IFN-γ monoclonal antibody labeled with fluorescein isothiocyanate (Ab*) was introduced into NK cells by electrophoration for intracellular immuno-reaction. After completion of the intracellular immuno-reaction, the NK cells were chemically pre-perforated with digitonin to lyse easily. Then, one NK cell containing the complexes of IFN-γ isoantigens with Ab* was electrokinetically injected into the capillary. The cell adsorbed on the tip of capillary was lysed by ultrasonication. Finally, the complexes of the different forms of IFN-γ in the cell were separated and detected by CE–LIF detection.
