Morphological and molecular identification of fungi isolated from selected brri rice varieties

A total of 19 fungal species were isolated from the seeds of selected rice varieties (BRRI dhan 90 to BRRI dhan 99) following Tissue planting method and Blotter method. The isolated fungi were Aspergillus niger, A. ochraceus, A. oryzae, A. tamarii, A. terreus, Chaetomium globosum, Cladosporium oxysporum, Colletotrichum gloeosporioides, Corynespora cassiicola, Curvularia lunata, Curvularia soli, Daldinia eschscholtzii, Fusarium solani, Penicillium oxalicum,Penicillium sclerotiorum, Pestalotiopsis guepinii, Pyricularia oryzae and Rhizopus stolonifer. Fourteen fungi were selected for molecular identification. Out of the 19 fungal isolates, 14 were confirmed up to species level through ITS sequence based molecular analysis. Among the isolated fungi Pnicillium sclerotiorum and Curvularia soli are the new record for Bangladesh. Association of Daldinia eschscholtzii with rice seeds is also recorded first time from world. Trait analysis of short duration boro rice (Oryza sativa L.) varieties in northern region of Bangladesh: Insights from heatmap, correlation and PCA

The role of genetic variation is utmost importance in the selection of a desirable genotype for breeding programs. In the northern region of Bangladesh, the performance of five short duration boro rice varieties (BRRI dhan28, BRRI dhan74, BRRI dhan81, BRRI dhan88, and Bangabandhu dhan100) on yield and yield-contributing traits were evaluated in four locations. Yield attributes tiller hill-1 (TN), panicle hill-1 (PN), filled grain panicle-1 (FG), panicle length (PL), 1000-grain weight (TGW), and harvest index (HI), showed a positive correlation with grain yield (GY). Conversely, spikelet sterility (SSP) and unfilled grains panicle-1 (UFG) showed negative associations with grain yield. Principal component analysis (PCA) revealed that the first three components accounted for 89.60% of variability. The highest positive eigenvalue was observed in DF, DM, BY, HI, PN, SY, TN, PL and GY in PC1, indicating their significant influence on the overall genotype variations. PC2 was primarily driven by plant height (PH) and FG, while PC3 driven by TGW and UFG. The results suggest that grain yield is highly influenced by FG, PH, DM, DF, PN, TN, PL, TGW, SY, BY, and HI. These components highlighted the importance in distinguishing rice genotypes with higher grain yield potential. By considering these traits, BRRI dhan74 have outperformed other varieties, followed by BRRI dhan88 and Bangabandhu dhan100. BRRI dhan74 is most suitable for all four location followed by BRRI dhan88. The traits such as PN, PL, FG, TGW, PH, & HI significantly influence rice yield and should be prioritized in breeding programs aimed at developing high-yielding rice varieties.

Enhancing Yield and Nitrogen Utilization of Boro Rice with Urea Super Granules

A field experiment was conducted at the Soil Science Field Laboratory of Bangladesh Agricultural University, Mymensingh during boro season to assess the comparative performance of prilled urea (PU) and urea super granule (USG) on the growth, yield, and nitrogen uptake of BRRI dhan29. The experiment was laid out in a randomized complete block design with three replications. The treatment combinations were T1: Control, T2: PU at 7 days after transplanting (DAT)+27 DAT+47 DAT, T3: PU at 10 DAT+30 DAT+50 DAT, T4: PU at 15 DAT+35 DAT+55 DAT, T5: USG at transplanting, T6: USG at 7 DAT, T7: USG at 10 DAT and T8: USG at 10 DAT+30 DAT where PU and USG were applied @ 216 kg N ha-1. PU was applied as surface broadcasting whereas USG was placed at 6-8 cm depth by hand between four hills at every alternate row. The results revealed that all the treatments showed better performances than control (T1). Results found that yield components, yield, and nitrogen uptake of BRRI dhan29 responded significantly to deep placement of USG than PU. Treatment T6 (USG at 7 DAT) produced the highest grain and straw yields of 6.56 t ha-1 and 6.95 t ha-1 respectively. Treatment T6 (USG at 7 DAT) also showed the highest grain yield increase (57.16%) and straw yield increase (52.37%) over control. The highest grain, straw, and total N uptake of 87.28, 32.71 and 120 kg ha-1 respectively were obtained in the treatment T6 (USG at 7 DAT). The overall results clearly indicate that deep placement of USG at 7 DAT in rice field increases rice yield and efficient uptake of the applied N. So, treatment T6 (USG at 7 DAT) could be recommended as best treatment for reducing N losses, conserving environment and improving sustainable production of rice.

Evaluation of Some Popular Rice Varieties of Bangladesh for Physical and Cooking Quality Properties

Rice is the primary staple meal in many parts of the world. Therefore, both consumers and producers must pay close attention to the physical and cooking characteristics of grains. The present study was conducted to evaluate the physical and cooking quality characters in twenty popular Bangladeshi boro rice genotypes along with one japonica rice genotype. A high degree of significant variation was observed for all the characters studied. Genotypes BRRI dhan55, BR16, BRRI dhan57, BINA dhan-6 and BINA dhan-14 has the maximum length of grain before cooking (L1). BRRI dhan36, BRRI dhan57, BRRI dhan55 and BINA dhan-6 were the best performer in terms of grain length after cooking (L2). BRRI dhan57 and BRRI hybrid dhan1 were best for width of grain before cooking (W1). BRRI dhan57, BRRI dhan29 and BRRI dhan36 were exhibited minimum width of grain after cooking (W2). Yukihikari, BRRI hybrid dhan3 and BRRI dhan36 were best for grain elongation ratio. Highest grain length/width ratio before cooking (L1/W1) was observed in genotype BRRI dhan57, BRRI hybrid dhan1 and BINA dhan-14. The highest grain length/width ratio after cooking (L2/W2) was recorded in genotype BRRI dhan57 and BRRI dhan36. Best genotype for percent water uptake (WU %) after cooking was BRRI dhan28 (180%). The best genotypes for solid in cooking water (SCW) were BRRI dhan36 (0.897%), BRRI dhan28 (0.956%), Yukihikari (1.043%) and BRRI dhan29 (1.048%), respectively. The prominent performer genotypes for CT were BRRI dhan58 (18.38 minutes) followed by BR 14 (19.12 minutes), BRRI dhan28 (20.00 minutes) and BRRI dhan57 (19.82 minutes). Best genotypes for amylase content (AC) BRRI dhan28 (24.93%), BRRI dhan55 (24.91%), BRRI dhan59 (24.90%). Most of the rice genotypes had found low cooking time (GT) except BRRI dhan57, BRRI dhan59, Yukihikari which shows intermediate gelatinization temperature (GT). The comparative information about physical and cooking qualities of rice might be used as reference to develop high grain quality rice cultivars in future. Res. Agric. Livest. Fish. BRRI Dhan107: High Protein Premium Quality Rice Variety for Irrigated Ecosystem in Bangladesh

A newly released high protein enriched, premium quality type high yielding, extra-long slender grain containing rice variety BRRI dhan107 which suitable for irrigated ecosystem of Bangladesh, is an improvement over existing high protein enriched and premium quality type rice varieties for Bangladesh. The variety has satisfactorily passed the Proposed Variety Trial (PVT) conducted in the farmers’ field in Boro 2022-23 season. As a result National Seed Board (NSB) approved this variety for commercial cultivation in the Boro season (dry season) in 2024. It has modern plant type with 103 cm plant height and matures by 148 days. The proposed variety showed 1.20 t/ha higher yield than check variety BRRI dhan50. Identifying characters of this variety are green leaf, intermediate plant height, semi-erect flag leaf, extra-long slender grain, intermediate leaf senescence. On an average it can produce 8.19 t/ha grain yield, although with appropriate management under favorable environment it can give up to 9.57 t/ha yield. Thousand grain weight of the variety is 26.1 gm and head rice yield is 60.1%. High protein (10.02%) enriched BRRI dhan107 has high amylose (29.1%) with extra-long slender (7.6 mm) type grain. The high protein enriched, premium quality type rice variety BRRI dhan107 is a superb variety for cultivating in the Boro season (dry) and farmers as well as the country can be nutritionally and economically benefited by the cultivation of BRRI dhan107.

Detection and quantification of aflatoxins in farmers’ stored rice seeds in some selected areas of Bangladesh

Detection and quantification of total aflatoxins in farmers’ stored rice seeds at different storage intervals in two cropping seasons (Boro 2017–2018: cv. BRRI Dhan28 and cv. BRRI Dhan29 and Aman 2017: cv. BR 11 and cv. BRRI dhan49) was focused in this study. Aflatoxin was quantified in two cropping seasons (Boro and Aman). Blotter incubation test was conducted for fungal prevalence assessment and a one-step lateral flow immune-chromatographic assay was used to quantify the aflatoxins. Seed moisture content ranged from 11.59 to 13.32% and aflatoxin concentrations ranged from 2.41 to 11.12 ppb. Variation in fungal contamination in stored seeds was correlated with seed moisture content. The moisture content in stored rice seeds increased with storage time and was positively correlated with fungal infestation and toxin production. The findings of this study indicate that moisture content of stored rice seeds and storage duration are determining factors for aflatoxin contamination.

Synergistic Benefits of Combined Application of Manures and Chemical Fertilizers: Enhancing Growth and Yield of Rice and Wheat

The incorporation of compost and manure with chemical fertilizer has become a key concern to sustain soil fertility. Because the nutrients are continuously depleting from the soil and consequently creating a risk to preserving soil health. Aims: This study assesses the response of different doses of cowdung (CD), municipal solid waste compost (MSWC), oil cake (OC), and poultry manure (PM), incorporated with fertilizers on the growth, yield and protein content of BRRI dhan49 rice and BARI Gom-24. Study Design: RCBD with 8 treatments and 3 replications. Place and Duration of Study: Field experiment and nutrient analysis were carried out at Bangladesh Agricultural University (BAU), Mymensingh. Wheat was grown during the rabi season from October 2018 to March 2019. Again, rice was cultivated during kharif II season from August 2019 to December 2019 on the same field. Then the laboratory analysis was performed till June 2020. Methodology: Eight treatments i.e., T0 Control (no fertilizer), T1 Recommended Fertilizer Dose, T2 (CD + NPK), T3 (MSWC + NPK), T4 (OC + NPK), T5 (PM + N), T6 (MSWC + N), T7: (CD + OC) were accommodated. Data on the growth parameters including grain and straw yields were documented after the harvest of the fully matured crop simultaneously determining the nitrogen (N) and protein content. Results: Adoption of various combinations of compost and manures with fertilizer accorded an increase in yield contributing characters as well as the N and protein content. Integrating MSWC @ 10 t ha-1+N projected the highest grain yield (7.11 t ha-1 in rice; 4.84 t ha-1 in wheat) and straw yield (8.8 t ha-1 in rice; 5.55 t ha-1 in wheat). It also promisingly secured the highest protein content compared to all other treatments. Conclusion: The appliance of MSWC @ 10 t ha-1 with the recommended dose of N may be endorsed for rice and wheat cultivation in the AEZ-9 region and retaining the physicochemical properties of Old Brahmaputra Floodplain soils.

Role of Trichoderma asperelloides and Trichoderma brevicompactum in improving drought tolerance in rice

Drought is considered as one of the major abiotic stresses affecting rice production under rainfed ecologies of Bangladesh hampering the country's self-sufficiency in rice production through reduced crop yield and quality. In the present study, we studied the effect of two species of plant growth promoting Trichoderma v iz; Trichoderma asperelloides and Trichoderma brevicompactum on rice plant development under drought stress and their role in inducing tolerance to drought at physiological and biochemical level. Results reveal that both species are capable of inducing plant tolerance to drought stress when tested on drought tolerant (BRRI dhan56) and drought susceptible (IR64) varieties. As the number of drought stress days increased, there was an increase in root length of both the varieties but increase in root length was more in BRRI dhan56 as compared to IR 64 when seeds of both varieties were primed with Trichoderma. There was significant change in proline, malondialdehyde (MDA), total Phenolic content (TFC), H2O2 content in Trichoderma treated versus non-treated plants for both varieties. These biochemical changes were verified with the expression of aquaporin (AQU), dehydrin (DHN) and DREB genes in both rice varieties treated with both Trichoderma species. Interaction studies of Trichoderma species with two rice genotypes suggested that AQU-2 expressed more in tolerant variety (BRRI dhan56) when primed with Trichoderma asperelloides whereas, both Trichoderma spp. up regulated the expression of DHN-4 and DREB genes in both rice genotypes under drought conditions. The present findings suggest that the both Trichoderma asperelloides and Trichoderma brevicompactum are most effective in improving drought tolerance in rice, and its prospective application will contribute to the advancement of rice genotypes to sustain crop productivity under drought stress.

Energy potential of biomass from rice husks in bangladesh: An experimental study for thermochemical and physical characterization

Rice husks, abundant leftovers from rice production, offer immense potential as a renewable energy source through thermochemical conversion processes. However, their efficient utilization hinges on understanding their unique thermal properties and reaction kinetics. This study addresses a critical gap by meticulously analyzing four prominent Bangladeshi rice husk varieties: BR22, BRRI Dhan46, BRRI Dhan47, and BRRI Dhan49. Through rigorous experimentation, we unveil a comprehensive dataset encompassing their physical, chemical, and thermochemical characteristics. Our investigation reveals favorable alignment of moisture content with various conversion technologies, alongside suitable bulk density for efficient handling. Proximate analysis sheds light on crucial components like ash, volatile matter, and fixed carbon, vital for optimizing combustion efficiency. Furthermore, elemental analysis not only highlights the presence of ash-forming elements but also assures low nitrogen and sulfur content, suggesting potential environmental benefits compared to conventional fuels. Delving into thermochemical characteristics, we measured higher heating values ranging from 13.31 MJ/kg to 14.42 MJ/kg, confirming the viability of these rice husk varieties for energy conversion. Thermogravimetric and kinetic analyses further illuminate their unique decomposition behavior, with BRRI Dhan47 exhibiting the highest decomposition rate, emphasizing the distinct reactivity profiles of different varieties. This extensive dataset empowers researchers and industry professionals with valuable insights for informed decision-making. By understanding the unique attributes of each rice husk variety and their behavior during conversion, we can optimize operational parameters for various thermochemical methods. Ultimately, this study bridges a critical research gap and paves the way for more efficient and sustainable utilization of rice husks as a renewable energy source, contributing to a greener future.

Assessment of phenotypic and genotypic diversity in rice for salinity tolerance at reproductive stage

Soil salinity has turned out to be an important global concern, which affects tenable rice production in many agricultural countries like Bangladesh. A cross was made between Binadhan-7 with FL-478 for developing salt tolerant advance lines to assess phenotypic and genotypic diversity in rice for salinity tolerance. Twenty advance rice lines (Binadhan-7/FL-478) along with check varieties were screened for salinity tolerance at reproductive stage using sea site saline water having electrical conductivity (EC) 10 dS m-1. Among the twenty rice lines, seven lines were tolerant, three were moderately tolerant, six were susceptible, four were highly susceptible on the basis of phenotypic evaluation. Seven simple sequence repeat (SSR) markers linked with salt tolerance quantitative trait loci were used for investigation of salt tolerant rice lines. The result revealed that an average number of 6.286 alleles per locus were detected, with polymorphism information content (PIC) values ranging from 0.672 (RM490) to 0.838 (RM562). The highest gene diversity value (0.812) was found in RM562 and the lowest (0.684) was in RM490. A dendrogram constructed from the genetic distance of the genotypes produced four distinct clusters of twenty rice genotypes. Considering both phenotypic and genotypic observation, seven genotypes viz., Binadhan-10, FL-478, SL-51, SL-56, SL-77, EFSD-59 and IZSD-45 were identified as salt-tolerant; on the other hand, EFSD-21, SL-28, SL-32, SL-10, BRRI dhan28, and Binadhan-7 were identified as salt-susceptible. Evaluation of two advanced generation boro rice mutants based on morpho-physiological criteria

Experiments were performed at farmers’ fields of five locations viz., Mymensingh, Sherpur, Jamalpur, Natore and Naogaon districts with two promising Boro rice mutants, RM-16(N)-8, RM-16(N)-10 and a check, BRRI dhan58 to evaluate some morpho-physiological features and its impact on grain yield. The experiment was setup in RCB design with three replications. Results revealed that high yielding genotypes, in general, showed superiority in morpho-physiological characters (leaf area, leaf area index, total dry mass production, absolute growth rate, relative growth rate, chlorophyll, total sugar and soluble protein content in leaves and harvest index) than the low yielding one. Results further indicated that genotype improvement efforts have achieved higher grain yield by higher growth rate at early growth stages and better assimilate partitioning to economic yield and short stature plants are unlikely to improve harvest index and resistant to lodging in rice. The mutant, RM-16(N)-10 had medium plant stature with greater biomass production capacity, superiority in growth and biochemical parameters and improve dry matter partitioning to economic yield which resulted higher number of grains panicle-1, thereby grain yield. In contrast, RM-16(N)-8 showed inferiority in morpho-physiological characters and performed the lowest yield attributes and grain yield. Yield Performance of Binadhan-24 Over Two Selected Locations in Bangladesh

Boro is the most important cereal crops in terms of production to the farmers in Bangladesh. An experiment was conducted to examine the suitability and productivity of two rice varieties cultivating in Boro season. The experiment was consisted of Binadhan-24 and BRRI dhan58 and it was conducted at the farmer’s field of Mymensingh and Kishoreganj districts from December, 2022 to May, 2023. The experiment was performed in Randomized Complete Block Design (RCBD) with three replications. Higher yield (7.03 tha-1) was recorded in Binadhan-24 compared to the variety BRRI dhan58 (6.53 tha-1). A higher day to maturity was found in BRRI dhan58 (152.50 days) compared to the varieties Binadhan-24. The highest effective tiller hill-1 (24.90) and number of filled grain panicle-1 (142) were recorded in Binadhan-24 where in BRRI dhan58, effective tiller hill-1 and number of filled grain panicle-1 were 22.19 and 136.80 respectively. Higher 1000 seed wt. (24.38 g) was found in Binadhan-24 compared to another variety. And the maximum yield potential (7.03 tha-1) was recorded in Binadhan-24. This experiment will be useful to select Boro genotypes with high yield potential and future breeding stock in area specific suitable locations and all over the world.

YIELD PERFORMANCE AND COST-EFFECTIVENESS OF DRY DIRECT SEEDED BORO RICE IN RESPONSE TO SOWING DATE AND IRRIGATION REGIME

Dry direct seeding is a new technology to address the water scarcity problem faced in rice culture in boro season. An experiment was run at Bangladesh Agricultural University in Mymensingh, to determine the impact of the sowing date and irrigation schedule on the yield potential and cost effectiveness of boro rice grown under a dry direct seeding system. The experiment was laid out in a split-plot design and consisted of two sowing dates viz. 10 February and 02 March; and five irrigation regimes viz., no irrigation, one irrigation at 25% field capacity (FC), two irrigations (at 25% FC and at 1 week after 25% FC), three irrigations (at 25% FC, at 1 and at 2 weeks after 25% FC) and four irrigations (at 25% FC, 1, 2 and 3 weeks after 25% FC) replicated thrice assigning sowing date in main plots and irrigation in sub-plots. The result showed that BRRI dhan58 sown on 10 February with four irrigations (at 25% FC, 1, 2 and 3 weeks after 25% FC) produced the highest grain yield and economic return which was statistically comparable to those for two (at 25% FC and at 1 week after 25% FC), and three irrigations (at 25% FC, at 1 and at 2 weeks after 25% FC). The present study concludes that rice variety BRRI dhan58 can be cultivated successfully to achieve maximum yield and economic return by sowing the seed during 10 February to 02 March and applying 2 - 3 irrigations at the vegetative phase in dry direct seeding system.

Development of Submergence Tolerant Rice Variety BRRI dhan79 for Flash Flood Ecosystem of Bangladesh

Tolerance to submergence stress is an important breeding objective for the areas where rice cultivars are subjected to complete inundation for a week or more. Submergence tolerance is an important trait for rice (Oryza sativa L.) in the flash flood-prone ecosystem. The trait is largely controlled by a major gene designated as Sub1 located on chromosome 9. Submergence of rice (Oryza sativa) by flash flooding is a major constraint to rice production in Bangladesh. Quantitative trait loci analyses have revealed that a large portion of this variation in submergence tolerance can be explained by Sub1. A newly released submergence tolerant rice variety namely BRRI dhan79 was developed by hybridization between BRRI dhan49*6/BRRI dhan52 through Marker Assisted Backcrossing and selection method where BRRI dhan52 (submergence tolerant mega variety in Bangladesh) used as Sub1 donor. It can tolerate upto three weeks complete submergence at vegetative stage whereas Sub1 donor mega variety BRRI dhan52 can tolerate on an average two weeks complete submergence condition. It can also tolerate 50-60 cm water stagnation for 15-20 days after de-submergence. The variety has satisfactorily been passed in the proposed variety trial conducted in flash flood prone farmers' field in Bangladesh. As a result, the National Seed Board (NSB) of Bangladesh approved this rice variety for commercial cultivation for the flash flood affected rainfed lowland ecosystem (Transplanted Aman season) in 2017. It meet up all the trait of modern plant likely medium plant heihght (112 cm), strong and stature culm structure, erect flag leaf with stay green leaf at maturity stage and high yield potential with 140 days growth duration (in case of three weeks submergence mature by 160 days). It can produce 5.5 t/ha grain yield under non-stress condition and 4.0-4.5 t/ha yield under three weeks submergence condition. The grain shape of the variety is medium bold. The rice production scenario in flash flood affected region of Bangladesh has been remarkably improved after releasing this rice variety. It has not only change the socio-economic status of farmers but also sustaining food security.

Growth Performance of Rice Genotypes at the Seedling Stage under Different Salinity Stresses

Background: Worldwide coastal rice growing areas are severely affected by salinity which has become a major constraint to rice production. Salinity is a serious soil problem in rice-growing countries, severely limiting global rice production. Methods: In this study, the seedlings of 21 rice genotypes were tested against seven different levels of salinity concentrations (0.3, 2, 4, 6, 8, 10 and 12 dSm-1) under a hydroponic system in a glasshouse condition at Field 10, Faculty of Agriculture, Universiti Putra Malaysia (UPM). Data on seedling growth and salinity injury of the tested rice genotypes were recorded. Result: The results showed that Pokkali, FL478 and Binadhan-10 were tolerant at 12 dSm-1; BRRI dhan73, BRRI dhan61, Binadhan-8, BRRI dhan67 and BRRI dhan47 were tolerant at 10 dSm-1; Putra-1 were moderately tolerant at 8 dSm-1 and MR263, MR284, MR211 and MRQ74 were tolerant at 4 dSm-1 and rest of the genotypes were salt susceptible. The lower amount of Na: K and Na: Ca were measured from rice genotypes, Pokkali and FL478 at maximum salinity levels. The promising rice genotypes that were tolerant at 12 dSm-1 would be taken into consideration for a hybridization program to develop a new salt-tolerant rice variety.

AGRO-MORPHOLOGICAL CHARACTERIZATION AND GENETIC DIVERSITY ASSESSMENT OF NINETEEN BRRI-RELEASED RICE VARIETIES

Knowledge on genetically determined several growth parameters including morpho- physiological and developmental parameters are essential for rice breeding programs. Genetic divergence helps in the development of superior recombinants, which is prerequisite in any crop improvement program. The aim of the present study was to characterize 19 rice varieties based on agro-morphological descriptors and genetic diversity. The experiment was conducted at Bangladesh Rice Research Institute farm, Gopalganj, using a randomized complete block design with three replications. Both the qualitative and quantitative agro-morphological traits were diversified. The observed variations in the qualitative data were primarily accounted for by the blade pubescence, anthocyanin color of the leaf sheath, lemma-palea color, grain tip color, and awning. The top five axes of PCA contributed 81.04% of the total variation. The varieties were divided into five groups based on the cluster. With six genotypes, Cluster IV was the largest group, whereas Cluster I was the lowest with just one genotype. Cluster IV had the greatest intra-cluster distance, whereas clusters IV and I had the greatest inter-cluster distances. Contrarily, cluster IV had the highest cluster means for most of the quantitative characteristics noticed. Once more, grain length was the trait that contributed most to divergence. While considering results from genetic distance, the varieties under cluster IV (BRRI dhan29, BRRI dhan89, BRRI dhan92 BRRI dhan97, BRRI hybrid dhan3, BRRI hybrid dhan5) might be selected for additional breeding after taking the results of the genetic distance into consideration.
