Wide‐Spectrum Nano‐Antibiotics Based on TPA‐Py@AuNCs⊂BSA for Multimodal Synergistic Therapy of Drug‐Resistant Bacteria and Wound Infections

ABSTRACTTo meet the high requirements of biomedical applications in antimicrobial agents, it is crucial to explore efficient nano‐antimicrobial agents with no resistance and good biocompatibility for treating infected wounds. In this study, composite nano‐antibiotic TPA‐Py@AuNCs⊂BSA nanoparticles (TAB NPs) are prepared using hollow mesoporous Au nanocages (AuNCs) loaded with a photosensitizer (namely TPA‐Py) with D‐π‐A structure showing aggregation‐induced emission properties. When TPA‐Py is encapsulated in the cavity of AuNCs, its fluorescence is suppressed. In the presence of photothermal induction, TPA‐Py can be released from the AuNCs, allowing for the restoration of fluorescence illumination and the specific imaging of Gram‐positive bacteria. TAB NPs demonstrate outstanding antimicrobial activity against a variety of bacteria, and this multimodal antimicrobial property does not lead to the development of bacterial resistance. In vitro experiments show that TAB NPs could eliminate bacteria and ablate bacterial biofilm. In vivo experiments show that the synergistic antimicrobial effect of TAB NPs has a significant positive impact on the treatment of infected wounds, including rapid antibacterial action, promotion of M2 macrophage polarization, and enhancement of chronic wound healing. This study provides an effective strategy for developing wide‐spectrum nano‐antibiotics for the ablation of bacterial biofilms and the treatment of infected wounds.

Polyphenol-mediated redox-active hydrogel with H2S gaseous-bioelectric coupling for periodontal bone healing in diabetes.

Excessive oxidative response, unbalanced immunomodulation, and impaired mesenchymal stem cell function in periodontitis in diabetes makes it a great challenge to achieve integrated periodontal tissue regeneration. Here, a polyphenol-mediated redox-active algin/gelatin hydrogel encapsulating a conductive poly(3,4-ethylenedioxythiopene)-assembled polydopamine-mediated silk microfiber network and a hydrogen sulfide sustained-release system utilizing bovine serum albumin nanoparticles is developed. This hydrogel is found to reverse the hyperglycemic inflammatory microenvironment and enhance functional tissue regeneration in diabeticperiodontitis. Polydopamine confers the hydrogel with anti-oxidative and anti-inflammatory activity. Theslow, sustained release of hydrogen sulfide from the bovine serum albumin nanoparticles recruits mesenchymal stem cells and promotes subsequent angiogenesis and osteogenesis. Moreover, poly(3,4-ethylenedioxythiopene)-assembled polydopamine-mediated silk microfiber confers the hydrogel with good conductivity, which enables it to transmit endogenous bioelectricity, promote cell arrangement, and increase the inflow of calcium ion. In addition, the synergistic effects of hydrogen sulfide gaseous-bioelectric coupling promotes bone formation by amplifying autophagy in periodontal ligament stem cells and modulating macrophage polarization via lipid metabolism regulation. This study provides innovative insights into the synergistic effects of conductivity, reactive oxygen species scavenging, and hydrogen sulfide on the periodontium in a hyperglycemic inflammatory microenvironment, offering a strategy for the design of gaseous-bioelectric biomaterials to promote functional tissue regeneration in immune-related diseases.

Berberine-Loaded Bovine Serum Albumin Nanoparticles: Fabrication, Characterization, and Drug Release Evaluation

Abstract Introduction/Objective Introduction: Numerous studies have highlighted the diverse biochemical and pharmacological properties of berberine (BER), including its anti-inflammatory, antioxidant, anticancer, and hypolipidemic effects. Nanoparticles play a crucial role in enhancing drug delivery by providing a larger surface area, facilitating quicker dissolution in the bloodstream. Methods/Case Report Methods: Blank bovine serum albumin nanoparticles (BSA NPs) were prepared using a desolvation process, followed by the fabrication of berberine-loaded BSA nanoparticles (BER-NP). Physicochemical characterization, including particle size and zeta potential, was conducted using a Malvern Zetasizer. The mean particle size and distribution were determined using photon correlation spectroscopy (PCS), and transmission electron microscope (TEM) images were obtained for visualization. Fourier transform infrared (FTIR) spectra and differential scanning calorimetry (DSC) thermograms were recorded to assess drug encapsulation efficiency (EE). Berberine release from BER-NPs was evaluated using dialysis membranes. Results (if a Case Study enter NA) Results: FTIR spectra and DSC thermograms confirmed successful drug loading into the nanoparticles. Encapsulation efficiency was determined by UV absorption spectroscopy, revealing high efficiency in the encapsulation process. Dialysis membranes demonstrated controlled release kinetics of berberine from BER-NPs, indicating their potential for targeted drug delivery. Conclusion Conclusion: Berberine-loaded bovine serum albumin nanoparticles exhibit favorable characteristics for drug delivery applications, including enhanced permeation across biological barriers. These nanoparticles hold promise for targeted therapy and combination drug regimens, offering potential benefits for various medical interventions.

Removal of the nalidixic acid antibiotic from aqueous solutions using bovine serum albumin nanoparticles

The presence of antibiotic pollutants in water and wastewater can cause significant risks to the environment in different aspects. Therefore, antibiotics need to be removed from water. This study investigates the adsorption of nalidixic acid (NA), a common antibiotic, using bovine serum albumin nanoparticles (BSA NPs). These NPs were synthesized via desolvation technique and characterized using SEM, DLS, FT-IR, and UV-Vis spectroscopy. The effects of adsorbent dosage (0.02–0.9 mg), initial NA concentration (30–80 mg L− 1) and contact time (0.5–24 h) on adsorption efficiency were considered. Adsorption isotherms and kinetics were determined experimentally. The Freundlich isotherm best described the adsorption equilibrium, while the pseudo-second-order kinetic model accurately represented the adsorption process. Thermodynamic parameters confirmed the spontaneous and exothermic nature of NA adsorption onto BSA NPs. Under optimal conditions, BSA NPs achieved a removal efficiency of 75% for NA with a maximum adsorption capacity of 240 mg g− 1. These results demonstrate the potential of BSA NPs as an effective adsorbent for removing NA from aqueous solutions.

Preparation and Characterization of Berberine-Loaded Bovine Serum Albumin Nanoparticles: Encapsulation Efficiency and Release Rate Determination

Nanoparticles serve as efficient carriers for drugs, offering increased surface area and enhanced dissolution in the bloodstream, thereby improving drug delivery. Methods/Case Report Methods: Blank Bovine Serum Albumin Nanoparticles (BSA NPs) were synthesized using a desolvation process, followed by the fabrication of Berberine-loaded Bovine Serum Albumin Nanoparticles (BER-NP). Encapsulation efficiency (EE) was determined by measuring the concentration of free BER in the supernatant after centrifugation, with EE calculated based on the difference between total added berberine and the measured supernatant concentration. Dialysis membranes were utilized to assess BER release from BER-NPs in vitro, with initial BER content in nanoparticles determining the release profile. Conclusion Conclusion: The prepared nanoparticles have demonstrated the potential to enhance drug permeation across epithelial and endothelial barriers. This approach holds promise for targeted drug delivery to specific sites, facilitating combined therapy with distinct modalities or drugs.

Investigations on the complexation and binding mechanism of bovine serum albumin with Ag-doped TiO2 nanoparticles.

It is essential to study the interactions between nanoparticles and proteins to better understand the biological interactions of nanoparticles. In this study, we studied the protein adsorption mode on the surface of Ag-doped TiO2 nanoparticles (NPs) using a model protein, bovine serum albumin (BSA). The mechanism of binding BSA to the Ag-doped TiO2 NPs was studied by applying fluorescence quenching, absorbance measurements, circular dichroism (CD) and Fourier transform infrared (FT-IR) spectroscopy techniques. The strong binding between BSA and Ag-doped TiO2 NPs was confirmed by a high value of binding constant (K = 2.65 × 105 L mol-1). We also studied the thermal stability of BSA in the presence of the Ag-doped TiO2 NPs. Thermodynamic parameters indicated that the adsorption of BSA on the Ag-doped TiO2 NPs was a spontaneous, natural and exothermic process. The effect of Ag-doped TiO2 NPs on the transportation function of BSA was also studied using a fluorescence spectroscopic technique. Fluorescence spectroscopic data suggested the existence of a strong interaction between BSA and the surface of the Ag-doped TiO2 NPs, which indicated that the binding affinities of some selected amino acids in BSA changed. This, in turn, clearly confirms that the Ag-doped TiO2 NPs affect the transportation capability of BSA in blood.

Genome-Wide Profiling of H3K27ac Identifies TDO2 as a Pivotal Therapeutic Target in Metabolic Associated Steatohepatitis Liver Disease.

H3K27ac has been widely recognized as a representative epigenetic marker of active enhancer, while its regulatory mechanisms in pathogenesis of metabolic dysfunction-associated steatotic liver disease (MASLD) remain elusive. Here, a genome-wide comparative study on H3K27ac activities and transcriptome profiling in high fat diet (HFD)-induced MASLD model is performed. A significantly enhanced H3K27ac density with abundant alterations of regulatory transcriptome is observed in MASLD rats. Based on integrative analysis of ChIP-Seq and RNA-Seq, TDO2 is identified as a critical contributor for abnormal lipid accumulation, transcriptionally activated by YY1-promoted H3K27ac. Furthermore, TDO2 depletion effectively protects against hepatic steatosis. In terms of mechanisms, TDO2 activates NF-κB pathway to promote macrophages M1 polarization, representing a crucial event in MASLD progression. A bovine serum albumin nanoparticle is fabricated to provide sustained release of Allopurinol (NPs-Allo) for TDO2 inhibition, possessing excellent biocompatibility and desired targeting capacity. Venous injection of NPs-Allo robustly alleviates HFD-induced metabolic disorders. This study reveals the pivotal role of TDO2 and its underlying mechanisms in pathogenesis of MASLD epigenetically and genetically. Targeting H3K27ac-TDO2-NF-κB axis may provide new insights into the pathogenesis of abnormal lipid accumulation and pave the way for developing novel strategies for MASLD prevention and treatment.

Two strategies of enhancing the therapeutic potential of a coumarin probe: Integration of a protonatable moiety and BSA-based nanoparticle formation

Photodynamic therapy (PDT) represents a promising non-invasive approach for cancer treatment, relying on photosensitizers (PSs) to generate reactive oxygen species (ROS) upon light activation. This study aims to enhance the therapeutic potential of a coumarin-based fluorescent probe by employing two strategic modifications: the integration of a protonatable lysosome-targeting moiety and the formation of bovine serum albumin (BSA)-based nanoparticles. The COM molecule was synthesized with a morpholine moiety to facilitate lysosome targeting and protonation in acidic environments, thereby enhancing ROS production and promoting lysosomal dysfunction for increased cancer cell apoptosis. Additionally, COM was encapsulated in BSA nanoparticles (COM-BSA NPs) to leverage hydrophobic interactions and hydrogen bonding, which enhance ROS generation by promoting the intersystem crossing (ISC) process. Comprehensive photophysical studies confirmed the enhanced ROS generation under acidic conditions and within BSA-based NPs. Cellular experiments demonstrated effective lysosome targeting and anticancer efficacy of both COM and COM-BSA NPs. These findings highlight the dual benefits of maintaining the bioimaging capabilities of coumarin while significantly improving its therapeutic efficacy in PDT applications.

Enhanced protection and bioavailability of Lycium barbarum leaf extract through encapsulation in whey protein isolate and bovine serum albumin nanoparticles

To improve the stability and bioavailabilityhe of polyphenolics in Lycium barbarum leaf, this study encapsulated L. barbarum leaf extracts (LLE) within whey protein isolate (WPI) and bovine serum albumin (BSA) nanoparticles (NPs) via self-assembly to enhance polyphenol distribution. The physicochemical properties of nanoparticles were characterized using transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), differential scanning calorimetry (DSC) and thermogravimetric (TG), respectively. The nanoparticles also showed good physical stability at various temperatures, different pH and NaCl concentrations. Compared with BSA-LLE NPs, WPI-LLE NPs exhibited strong physical stability with encapsulation efficiency of 70.6 %. The polyphenol nanoparticles demonstrated enhanced stability in the presence of stomach acid during in vitro simulated digestion. Additionally, the nanoparticles enhanced polyphenol stability during simulated gastrointestinal digestion. Following intestinal digestion, compared with LLE, the bioaccessibility of total phenolic increased by 53.67 % (WPI-LLE NPs), with specific enhancement in compounds like kaempferol, rutin, and chlorogenic acid.

Chitosan Oligosaccharide Modified Bovine Serum Albumin Nanoparticles for Improving Oral Bioavailability of Naringenin.

With the rapid development of nanotechnology, the research and development of nano-drugs have become one of the development directions of drug innovation. The encapsulation of the nanoparticles can change the biological distribution of the drug in vivo and improve the bioavailability of the drug in vivo. Naringenin is poorly soluble in water and has a low bioavailability, thus limiting its clinical application. The main purpose of this study was to develop a nano-sized preparation that could improve the oral bioavailability of naringenin. Chitosan oligosaccharide modified naringenin-loaded bovine serum albumin nanoparticles (BSA-COS@Nar NPs) were prepared by emulsification solvent evaporation and electrostatic interaction. The nanoparticles were characterized by HPLC, laser particle size analyzer, transmission electron microscope and X-ray diffraction analysis. The release in vitro was investigated, and the behavior of nanoparticles in rats was also studied. The caco-2 cell model was established in vitro to investigate the cytotoxicity and cellular uptake of nanoparticles. BSA-COS@Nar NPs were successfully prepared, and the first-order release model was confirmed in vitro release. In vivo pharmacokinetic results indicated that the area under the drug concentration- time curve (AUC) of BSA-COS@Nar NPs was 2.37 times more than free naringenin. Cytotoxicity and cellular uptake results showed that BSA-COS@Nar NPs had no significant cytotoxic effect on Caco- 2 cells and promoted cellular uptake of the drug. BSA-COS@Nar NPs could improve the in vivo bioavailability of naringenin.

Optimizing Desolvation Conditions for Glutathione-Cross-Linked Bovine Serum Albumin Nanoparticles: Implication for Intravenous Drug Delivery.

Protein-based nanocarriers, particularly albumin nanoparticles (NPs), offer numerous advantages when compared to other nanomaterials. These carriers are characterized by biocompatibility, biodegradability, reduced immunogenicity, and decreased cytotoxicity. Moreover, proteins possess an inherent ability to target tumor cells directly or indirectly. This study aims to investigate the impact of various organic solvents on the characteristics of synthesized bovine serum albumin NPs (BSA NPs). BSA NPs were produced using methanol, acetone, ethanol, dimethylsulfoxide (DMSO), and acetonitrile through the desolvation technique to achieve particles of acceptable size. Dynamic light scattering (DLS), blood compatibility assays, polyacrylamide gel electrophoresis (PAGE), and size exclusion chromatography (SEC) were employed to elucidate the properties of the generated NPs. The cytotoxicity of BSA NPs prepared under different conditions was assessed using Michigan Cancer Foundation - Mammary Adenocarcinoma - Breast Cancer 231 cells (MDA-MB-231 cells). The particle size of the synthesized NPs varied based on the organic solvent utilized, with the smallest size of 114.2 nm observed with methanol. Blood compatibility results indicated no abnormal interactions between BSA NPs and blood components. PAGE analysis revealed a strong band near 72 kDa for untreated BSA and BSA treated with all organic solvents. In SEC, the retention time of native albumin was 6.65 min, while the average retention times of the prepared BSA NPs ranged from 5.14 to 5.21 min, showing similarity to the native protein. Except for NPs produced with methanol and acetonitrile, cytotoxicity testing on MDA-MB-231 cells demonstrated no significant harmful effects at various concentrations (0-500 μg/mL). The choice of desolvating agent significantly influences the size of BSA NPs. Various factors, such as solvent characteristics like hydrogen bonds, polarity, dielectric constant, and functional groups, can affect the particle size and structure of BSA NPs. The compatibility of cross-linked BSA NPs with blood components suggests their potential for intravenous drug delivery applications.

Application of galactosylated albumin for targeted delivery of triptolide to suppress hepatocellular carcinoma progression through inhibiting de novo lipogenesis

Hepatocellular carcinoma (HCC) remains the fourth leading cause of cancer-associated death globally with a lack of efficient therapy. The pathogenesis of HCC is a complex and multistep process, highly reliant on de novo lipogenesis, from which tumor cells can incorporate fatty acids to satisfy the necessary energy demands of rapid proliferation and provide survival advantages. Triptolide (TP) is a bioactive ingredient exhibiting potent abilities of anti-proliferation and lipid metabolism regulation, but its clinical application is constrained because of its toxicity and non-specific distribution. The present study has developed galactosylated bovine serum albumin nanoparticles loaded with TP (Gal-BSA-TP NPs) to alleviate systemic toxicity and increase tumor-targeting and antitumor efficacy. Furthermore, Gal-BSA-TP NPs could inhibit de novo lipogenesis via the p53-SREBP1C-FASN pathway to deprive the fuel supply of HCC, offering a specific strategy for HCC treatment. In general, this study provided a biocompatible delivery platform for targeted therapy for HCC from the perspective of de novo lipogenesis.

Green synthesis of zinc ferrite nanoparticles from Nyctanthes arbor-tristis: unveiling larvicidal potential, protein binding affinity and photocatalytic activities.

Environmental pollution, being a major concern worldwide, needs a unique and ecofriendly solution. To answer this, researchers are aiming in utilizing plant extracts for the synthesis of nanoparticles. These NPs synthesized using plant extracts provide a potential, environmentally benign technique for biological and photocatalytic applications. Especially, plant leaf extracts have been safe, inexpensive, and eco-friendly materials for the production of nanoparticles in a greener way. In this work, zinc ferrite nanoparticles (ZnFe2O4 NPs) were prepared using Nyctanthes arbor-tristis leaf extract by hydrothermal method, and its biological and photocatalytic properties were assessed. The synthesized ZnFe2O4 NPs were characterized using powder X-ray diffraction (XRD), scanning electron microscopy (SEM), and Fourier-transform infrared spectroscopy (FT-IR). X-ray diffraction confirmed the arrangement of the fcc crystal structure of the nanoparticles and that some organic substances were encapsulated within the zinc ferrite. According to the SEM analysis, the resulting nanoparticles got agglomerated and spherical in shape. The ZnFe2O4 nanoparticles are in their pure form, and all of their elemental compositions were shown by the energy-dispersive X-ray analysis (EDAX) spectrum. The FTIR results revealed that the produced nanoparticles contained distinctive functional groups. Fluorescence spectroscopy was used to examine the binding affinities between bovine serum albumin (BSA) and ZnFe2O4 nanoparticles in terms of protein binding, stability, and conformation. The interaction between BSA and ZnFe2O4 NPs was examined using steady-state and time-resolved fluorescence measurements, and it was evident that static quenching occurred. The ability of ZnFe2O4 nanoparticles to kill Culex quinquefasciatus (C. quinquefasciatus) larvae was evaluated. The synthesized NPs demonstrated a noteworthy toxic effect against the fourth instar larvae of C. quinquefasciatus with LC50 values of 43.529µg/mL and LC90 values of 276.867µg/mL. This study revealed the toxicity of green synthesized ZnFe2O4 NPs on mosquito larvae, proving that these NPs are good and effective larvicides. Furthermore, the ZnFe2O4 NPs were utilized for dye degradation of methylene blue under visible light treatment and achieved 99.5% degradation.

Bovine Serum Albumin Nanoparticles as a Proposed Drug Formulation for the Delivery of 10H-2,7-diazaphenothiazine

The synthesis of new compounds and nanoparticles is one of many attempts to circumvent the drug resistance. Albumin nanoparticles are biocompatible drug carriers with an ability to incorporate drugs without modifications. 10H-2,7-diazphenothiazine (2,7-DAPT) is a newly phenothiazine derivative with an anticancer, immunomodulatory and anti-inflammatory activity with a low cytotoxicity toward normal splenocytes at the same time. Up to now, no administration route for 2,7-DAPT has been proposed, so the novelty of the study is synthesis of nanoparticles containing an active ingredient not yet used in the clinic. The aim of the study was to encapsulate 2,7-DAPT into bovine serum albumin (BSA) nanoparticles by desolvation method. This study was supplemented with spectroscopic studies of 2,7-DAPT, size and morphology measurements as well as release analysis at pH 7.4 and 5.6. 2,7-DAPT is a compound with high stability in solution and an ability to absorb at UV-Vis range. Based on the results of scanning electron microscopy, nanoparticles size oscillates around the value of 204 nm. The release of 2,7- DAPT from the nanoparticles was characterized by different mechanisms of release, which were dependent on the pH of the release buffer. The above results indicate the potential usefulness of the obtained nanoparticles. Due to the lack of studies of nanoparticles containing this substance, more detailed future analyses are required.

Silencing endomucin in bone marrow sinusoids improves hematopoietic stem and progenitor cell homing during transplantation.

Efficient homing of infused hematopoietic stem and progenitor cells (HSPCs) into the bone marrow (BM) is the prerequisite for successful hematopoietic stem cell transplantation. However, only a small part of infused HSPCs find their way to the BM niche. A better understanding of the mechanisms that facilitate HSPC homing will help to develop strategies to improve the initial HSPC engraftment and subsequent hematopoietic regeneration. Here, we show that irradiation upregulates the endomucin expression of endothelial cells in vivo and in vitro. Furthermore, depletion of endomucin in irradiated endothelial cells with short-interfering RNA (siRNA) increases the HSPC-endothelial cell adhesion in vitro. To abrogate the endomucin of BM sinusoidal endothelial cells (BM-SECs) in vivo, we develop a siRNA-loaded bovine serum albumin nanoparticle for targeted delivery. Nanoparticle-mediated siRNA delivery successfully silences endomucin expression in BM-SECs and improves HSPC homing during transplantation. These results reveal that endomucin plays a critical role in HSPC homing during transplantation and that gene-based manipulation of BM-SEC endomucin in vivo can be exploited to improve the efficacy of HSPC transplantation.

A do-it-yourself benchtop device for highly scalable flow synthesis of protein-based nanoparticles

Synthesis of nanoparticles is typically carried out in batch procedures, which offer limited control of parameters, and a narrow range of possible batch volumes. In contrast, flow synthesis systems, usually having a microfluidic chip as a crucial part, are devoid of these drawbacks. However, large scale devices – millifluidic systems – may offer several advantages over microfluidic systems, such as easier and cheaper production, enhanced throughput, and reduced channel clogging. Here we report a millifluidic system for the generation of protein nanoparticles, using the flow format of the original swift thermal formation technology (STF), which can process batch volume ranging from 100 µl to any practically significant amount. Capabilities of the system are demonstrated with model synthesis of Epirubicin-encapsulated BSA nanoparticles. A better degree of scalability of the synthesis over batch procedure is shown: with a 10-fold working volume increase, hydrodynamic diameter and loading capacity changed by only 10 % and 1 % respectively, compared to 60 % and 30 % for the batch synthesis. Additionally, we provide all engineering drawings, electrical circuits, programming code and nuances of assembly and operation, so that our findings can be easily reproduced. The ease of construction of the device and the superior characteristics of the resulting nanoparticles compared to the batch method indicate application potential in both the biomedical research and industrial spheres.

Experimental design of size variation in albumin nanoparticles synthesized by electron beam

Protein-based nanoparticles have garnered significant interest for their potential in theranostic applications, including cancer treatment and nuclear medicine. Plasma proteins are particularly appealing due to their ability to bypass the rapid clearance typically associated with synthetic particles. Human serum albumin (HSA), for example, is already utilized in diagnostic procedures such as lymphoscintigraphy and sentinel lymph node detection and in cancer treatments where it delivers therapeutic agents in nanoparticulate form. Among the various techniques for synthesizing protein nanoparticles, ionizing radiation stands out for its ability to maintain good size control and preserve the protein's three-dimensional structure. Factors such as the reaction precursor, pH, protein concentration, presence of a stabilizer, and irradiation dose can all influence the size and shape of the resulting nanoparticles. This study sought to identify a correlation between the size of albumin nanoparticles and reaction parameters, including protein concentrations, the ionic content of the buffer solution, and the e-beam irradiation dose. We conducted different synthesis methods with varying BSA concentrations, using two different buffers and varying radiation doses. The resulting nanoparticles were evaluated using Dynamic Light Scattering. The size data were statistically analyzed using SAS Studio, SAS JMP, and WEKA software to identify correlations among the synthesis variables. Our observations primarily revealed that lower protein concentrations consistently resulted in smaller particles. Additionally, using Tris-HCl buffer as a medium led to a more proportional growth of particles with increases in concentration and irradiation dose. These findings suggest that Tris buffer is more suitable for BSA nanoparticle synthesis, as increased albumin concentration and radiation dose resulted in more consistent size control.

Sustained release hypoxia-activated prodrug-loaded BSA nanoparticles enhance transarterial chemoembolization against hepatocellular carcinoma

Transarterial chemoembolization (TACE) is the standard of care for patients with advanced hepatocellular carcinoma (HCC), but facing the problem of low therapeutic effect. Conventional TACE formulations contain Lipiodol (LP) and chemotherapeutic agents characterized by burst release due to the unstable emulsion. Herein, we developed a novel TACE system by inducing bovine serum albumin (BSA) loaded hypoxia-activated prodrug (tirapazamine, TPZ) nanoparticle (BSATPZ) for sustained drug release. In the rabbit VX2 liver cancer model, TACE treatment induced a long-term hypoxic tumor microenvironment as demonstrated by increased expression of HIF-1α in the tumor. BSATPZ nanoparticles combined with LP greatly enhanced the anti-tumor effects of the TACE treatment. Compared to conventional TACE treatment, BSATPZ nanoparticle-based TACE therapy more significantly delayed tumor progression and inhibited the metastases in the lungs. The effects could be partially mediated by the rebuilt immune responses, as BSATPZ nanoparticle can served as an immunogenic cell death (ICD) inducer. Collectively, our results suggest that BSATPZ nanoparticle-based TACE therapy could be a promising strategy to improve clinical outcomes for patients with HCC and provide a preclinical rationale for evaluating TPZ therapy in clinical studies.

Targeting SUMOylation with an injectable nanocomposite hydrogel to optimize radiofrequency ablation therapy for hepatocellular carcinoma

BackgroundIncomplete radiofrequency ablation (iRFA) in hepatocellular carcinoma (HCC) often leads to local recurrence and distant metastasis of the residual tumor. This is closely linked to the development of a tumor immunosuppressive environment (TIME). In this study, underlying mechanisms and potential therapeutic targets involved in the formation of TIME in residual tumors following iRFA were explored. Then, TAK-981-loaded nanocomposite hydrogel was constructed, and its therapeutic effects on residual tumors were investigated.ResultsThis study reveals that the upregulation of small ubiquitin-like modifier 2 (Sumo2) and activated SUMOylation is intricately tied to immunosuppression in residual tumors post-iRFA. Both knockdown of Sumo2 and inhibiting SUMOylation with TAK-981 activate IFN-1 signaling in HCC cells, thereby promoting dendritic cell maturation. Herein, we propose an injectable PDLLA-PEG-PDLLA (PLEL) nanocomposite hydrogel which incorporates self-assembled TAK-981 and BSA nanoparticles for complementary localized treatment of residual tumor after iRFA. The sustained release of TAK-981 from this hydrogel curbs the expansion of residual tumors and notably stimulates the dendritic cell and cytotoxic lymphocyte-mediated antitumor immune response in residual tumors while maintaining biosafety. Furthermore, the treatment with TAK-981 nanocomposite hydrogel resulted in a widespread elevation in PD-L1 levels. Combining TAK-981 nanocomposite hydrogel with PD-L1 blockade therapy synergistically eradicates residual tumors and suppresses distant tumors.ConclusionsThese findings underscore the potential of the TAK-981-based strategy as an effective therapy to enhance RFA therapy for HCC.Graphic

Naringenin-Loaded Bovine Serum Albumin Nanoparticles Inhibit Reactive Oxygen Species (ROS) to Prevent Ulcer and Gastric Cancer

Gastric cancer is a malignance of digestive system and effective treatment measures are key to treatment of gastric cancer. In this experiment, we assessed the effect of bovine serum albumin nanoparticle (BASNP)-coated naringenin (NGN) on reactive oxygen species (ROS) in gastric cancer. After preparation of NGN-BASNP and animal model of gastric cancer, rats were administered with NGN-BASNP, ROS agonists and ROS inhibitors, when the model group was set. After one week of intervention, gastric ulcers in rats were measured and Hematoxylin-eosin (HE) staining was performed. Transwell chamber was used to detect cell invasion ability, while 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) was detected proliferation ability of gastric cancer cells. Real Time Quantitative Polymerase Chain Reaction (RT-qPCR) and Western blot determined TNF-α mRNA expression and ROS. With about 100 nm diameter of NGN-BASNP nanoparticles, the nanoparticles presented in good shape. Compared with lesions in the model group, NGN-BASNP treatment greatly improved the condition, relieving the ulcer and decreasing the ulcer area (P < 0.05). After adding ROS agonist, inhibitory effect of NGN-BASNP intervention was amplified (P < 0.01) when the ROS level in the NGN-BASNP+ROS agonist group was decreased and TNF-α expression decreased. Moreover, NGN-BASNP effectively suppressed gastric cancer cell proliferation and migration but induced apoptosis. NGN-BASNP inhibited the expression of TNF-α mRNA and ROS level in gastric cancer cells, thereby alleviating gastric ulcer and delaying gastric cancer cell growth.
