Cytosine-Phosphate-Guanine Oligodeoxynucleotides Containing GACGTT Motifs Enhance the Immune Responses Elicited by Keyhole Limpet Hemocyanin Antigen in Dairy Cattle

Adjuvants are important components of vaccine formulations. Effective adjuvants line innate and adaptive immunity by signaling through pathogen recognition receptors. Synthetic cytosine-phosphate-guanine (CpG) oligodeoxynucleotides (ODNs) have been shown to have potentials as adjuvants for vaccines. However, the immunostimulatory effect of CpG is species-specific and depends on the sequence of CpG motifs. A CpG ODN (2135), containing 3 identical copies of GTCGTT motif, was previously reported to have the strongest effects on bovine peripheral blood mononuclear cells (PBMC). Based on the sequence of 2135, we replaced the GTCGTT motif with 11 other sequences containing CG and investigated their effects on bovine lymphocyte proliferation. Results showed that the CpG ODNs containing 3 copies of GACGTT motif had the highest lymphocyte stimulation index (7.91±1.18), which was significantly (P<0.05) higher than that of 2135 (4.25±0.56). The CpG ODNs containing 3 copies of GACGTT motif also significantly increased the mRNA expression of interferon (IFN)-α, interleukin (IL)-12, and IL-21 in bovine PBMC. When dairy cows were immunized with the keyhole limpet hemocyanin (KLH) antigen formulated with CpG ODNs containing 3 copies of GACGTT, production of KLH-specific antibodies in serum and in milk whey was significantly (P<0.05) enhanced. IFN-γ in whole blood stimulated by KLH was also significantly (P<0.05) increased in cows immunized with KLH plus CpG ODNs. Our results indicate that CpG ODNs containing 3 copies of the GACGTT motifs is a potential adjuvant for bovine vaccines.

Use of bovine lymphocytes to assess the immunomodulatory effect of natural extracts

A simple and rapid method based on the in vitro bovine lymphocyte proliferation was developed to assess the immunomodulatory activity of natural compounds extracted from plant and foods. The proliferation of lymphocytes was measured by the MTT [3-(4, 5- dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide] assay. This test was used to measure the immuno-enhancing activity of 2 plant extracts and 2 samples of fermented milk after 48 h of incubation. To evaluate the effect of the cow physiological phase on the assay response, the tests were repeated using lymphocytes taken from non-pregnant dry, pregnant dry and post partum cows. The assay was not affected by the cow physiological phase and the results obtained were comparable. Both plant extracts enhanced lymphocyte growth, in particular the extract 2 was slightly more potent than extract 1. The fermentation of milk with proteolytic starter cultures, such as FAIR E-63 and LA 2, can generate peptidic compounds from milk protein with potential immunostimulating activity. These samples tested in cell culture inhibited lymphocyte proliferation because they showed an acidic pH of 4.5. However, this bioassay was enough sensitive to detect biological activity of different compounds also at low concentrations.

Staphylococcus aureus Enterotoxin D Is Secreted in Milk and Stimulates Specific Antibody Responses in Cows in the Course of Experimental Intramammary Infection

An enterotoxin D (SED)-producing strain of Staphylococcus aureus was used to infect one mammary gland of each of 17 lactating dairy cows. All glands became infected and shed bacteria over a sampling period of 3 weeks. Serum and milk antibodies specific for SED were monitored by an enzyme-linked immunosorbent assay for 12 weeks. Elevated anti-SED antibodies were detected in all cows after infection, and immunoglobulin of the G2 subclass comprised most of the specific serum response. SED was detected in mastitic milk samples from two cows at levels of 5 to 10 ng/ml. An in vitro lymphocyte proliferation assay showed that SED at levels below 10 pg/ml induced proliferation of bovine lymphocytes and that sheep antiserum specific for SED neutralized this proliferative response. Sera obtained from the cows pre- and postinfection inhibited lymphocyte proliferation at SED concentrations of 10 and 50 ng/ml, respectively. The addition of SED to whole blood or to isolated neutrophils had no significant effect on neutrophil function in vitro. The results show that SED is secreted during mammary gland infection, is mitogenic for bovine lymphocytes, and stimulates the production of specific antibodies.

Characterization of a lympho-inhibitory peptide produced by Mycoplasma bovis

Mycoplasma bovis is able to inhibit the mitogen-induced proliferation of bovine lymphocytes. Herein is described the isolation of an immuno-inhibitory peptide from M. bovis. Using size exclusion chromatography, three lympho-suppressive fractions were isolated from M. bovis free supernatant. MALDI-TOF analysis revealed a common peak throughout the suppressive fractions. The purest of these fractions was subjected to N-terminal sequencing, revealing an 84% homologous match with the C-terminus of the M. bovis surface protein VspL (variable surface protein—L). A recombinant of the 26 amino acid peptide was also able to suppress Concanavalin A (ConA)-induced proliferation of bovine lymphocytes. This describes a unique immunosuppressive peptide produced by the bovine respiratory pathogen, M. bovis.

Modulation of bovine lymphocyte response by salivary gland extracts of the stable fly, Stomoxys calcitrans (Diptera: Muscidae).

The effect of salivary gland extract of the stable fly, Stomoxys calcitrans (L), on bovine lymphocyte proliferation was determined, and antibody reactivity to salivary gland proteins was characterized in cattle exposed to stable flies. Salivary glands were dissected from male and female flies (4-8 d after eclosion), and protein extracts were made by freeze-thaw cycles. Salivary gland extract (SGE, 1 and 5 microg) significantly inhibited mitogen-driven proliferation of bovine lymphocytes, compared with 1 and 5 microg of identically prepared midgut extract (ANOVA, P < 0.05). Phytohemagglutinin A (PHA) stimulated lymphocyte responses were suppressed by 61.7 and 79.5% (mean values) with 1 and 5 microg of SCE, whereas concanvalin A (Con A) stimulated responses were suppressed by 62.9 and 77.1% (1 and 5 microg). In contrast, midgut extract (1 and 5 microg) minimally suppressed PHA (12.7% +/- 12.6 and 18.7% +/- 15.5) and Con A-driven responses (13.8% +/- 20.5 and 24.6% +/- 14.9), respectively. Viability studies using propidium iodide and flow cytometry demonstrated that SGE was not cytotoxic. Two-color immunofluorescence studies identified T and B lymphocytes as the nonviable cells in the cultures. Western blot analysis of serum collected from five dairy cows during periods of low and high fly exposure identified an immunodominant 27 kDa protein among the salivary gland proteins. These results indicate that exposure of cattle to stable fly saliva during blood feeding results in an antibody response to salivary proteins and that the saliva has a potential to modulate T lymphocyte function.

Monoclonal antibody BLT-1, specific for the bovine homologue of CD5, reacts with the majority of mature T cells, a subpopulation of B cells and stimulates t cell proliferation

Monoclonal antibody (MAb) BLT-1, an IgG2a with kappa light chains, reacted strongly with 21% of bovine thymocytes, weakly with 15% of thymocytes, with a subpopulation of peripheral blood B cells that also expressed CD20 and with peripheral blood T cells. Practically all of the reactive thymocytes were of a large cell subpopulation. By immunoprecipitation, BLT-1 was shown to recognize a membrane molecule with a molecular mass of 67 kDa. In competitive assays for lymphocyte surface binding, BLT-1 and MAb CC-29 (which had been shown previously to react with bovine CD5) blocked one another, indicating that the epitopes recognized were identical or extensively overlapping. In contrast, another CD5-reactive MAb, CC-17, did not block BLT-1 reactivity with lymphocytes although the reactivity of CC-17 was blocked by BLT-1, suggesting partial overlap of the epitopes or steric hinderance by BLT-1 but not by CC-17. BLT-1 was able to induce proliferation of bovine lymphocytes in culture alone if monocytes were present or in the absence of monocytes synergized with PMA. The results indicate that BLT-1 recognizes an epitope of the bovine homologue of CD5 and that perturbation of the epitope by MAb binding results in signal transduction to bovine lymphocytes.

Effects of In Vivo Administration of 1,25-Dihydroxyvitamin D3 on In Vitro Proliferation of Bovine Lymphocytes

Continuous infusion of 1,25-dihydroxyvitamin D3 in the bovine elevated 1,25-dihydroxyvitamin D3 in plasma. Plasma 1,25-dihydroxyvitamin D3 remained elevated until infusions were discontinued. Increased 1,25-dihydroxyvitamin D3 was associated with hypercalcemia (11 to 12 mg/dl) that continued for several days postinfusion. Elevated 1,25-dihydroxyvitamin D3 also was associated with a significant 70% enhancement of lymphocyte proliferation in cells treated with concanavalin A. Cessation of infusion of 1,25-dihydroxyvitamin D3 resulted in the return of 1,25-dihydroxyvitamin D3 in plasma and lymphocyte proliferative response to preinfusion amounts. Proliferation in response to pokeweed mitogen was significantly inhibited on d 3 of infusions of 1,25-dihydroxyvitamin D3. Administration of parathyroid hormone for 4 d resulted in a sustained hypercalcemia (≈ 13 mg/dl) and a transient 1-d increase in 1,25-dihydroxyvitamin D3 in plasma. Parathyroid hormone treatment generally did not affect the response of bovine lymphocytes to mitogen. In vivo elevation of 1,25-dihydroxyvitamin D3 is either moderately immunostimulating or exerts no effect on mitogen-stimulated proliferation of lymphocytes. These results contrast with the in vitro results suggesting that 1,25-dihydroxyvitamin D3 is only immunosuppressive.

Sodium dodecyl sulfate- and salt-extracted antigens from various Brucella species induce proliferation of bovine lymphocytes.

One- and two-dimensional gel electrophoresis and cellular immunoblotting were used to compare the protein profiles and immunogenic capabilities of salt- and sodium dodecyl sulfate-extracted components isolated from different Brucella species. Cellular immunoblotting demonstrated that freshly isolated bovine peripheral blood mononuclear cells proliferated to similar molecular mass components from the various Brucella extractions. One- and two-dimensional gel electrophoresis showed similarities in protein profiles among the different Brucella species that correlated with lymphocyte reactivity. The results presented in this study provide preliminary evidence that common proteins that induce lymphocyte proliferation are present among different Brucella species, suggesting that a genuswide subunit vaccine may be feasible.

Immunomodulatory effects of fusarochromanones TDP-1 and TDP-2.

An in vitro peripheral lymphocyte blastogenesis system was used to investigate the biological activities of the fungal toxin fusarochromanone (TDP-1) and its monoacetyl derivative TDP-2. Briefly, cultures of human or bovine peripheral lymphocytes were exposed to TDP-1 or TDP-2 and a mitogen (PHA, Con A or PWM). After a standard incubation time, cell proliferation was quantified using the MTT bioassay. Human and bovine lymphocyte proliferation was inhibited by high concentrations of TDP-1; however, bovine lymphocyte proliferation was significantly increased at low concentrations of TDP-1. TDP-2 has similar but less pronounced effects on lymphocyte proliferation.

Effects of Ketones, Acetate, Butyrate, and Glucose on Bovine Lymphocyte Proliferation ,

Blood leukocytes from age-matched heifers were used to determine effects of ketones, acetate, butyrate, and glucose on in vitro lymphocyte proliferation. Lymphocytes stimulated with concanavalin A, phytohemagglutinin-P, or pokeweed mitogen were cultured in the presence or absence of β-hydroxybutyrate, acetoacetate, acetone, acetate, butyrate, and glucose. Only supraphysiological levels of β-hydroxybutyrate inhibited proliferation in cultures of mitogen-stimulated lymphocytes, whereas mixtures of β-hydroxybutyrate and acetoacetate at levels seen in severe ketosis stimulated concanavalin A and phytohemagglutinin-P-driven proliferation. Because acetoacetate was a lithium salt, lithium chloride served as a negative control. Results suggest the enhanced proliferation by cultures containing lithium acetoacetate was due to lithium, not acetoacetate. Butyrate (at concentrations greater than seen in bovine plasma) and acetate at normal levels inhibited proliferation. Concanavalin A-and pokeweed-mitogen-driven proliferation was greater in cultures containing lower glucose levels, but acetate added to cultures containing low glucose inhibited concanavalin A-stimulated proliferation. Proliferation by pokeweed mitogen-stimulated cultures containing acetate, β-hydroxybutyrate, and acetoacetate was suppressed at the lower concentrations of glucose tested. In conclusion, ketones, butyrate, and glucose at concentrations occurring in vivo had minimal effects on bovine lymphocyte proliferation in vitro. Levels of acetate associated with ketosis suppressed lymphocyte function and may alter immune responsiveness in vivo.

Preliminary investigation of the mechanism of inhibition of bovine lymphocyte proliferation by Pasteurella haemolytica A1 leukotoxin

Pasteurella haemolytica A1 leukotoxic culture supernate has been shown to inhibit bovine lymphocyte blastogenesis induced by concanavalin A (Con A), pokeweed mitogen (PWM) and purified protein derivative (PPD). The various mechanisms by which this inhibition could be overcome were investigated in an effort to determine at which stage of cell activation the leukotoxin exerted its inhibitory effect. For both Con A and PWM stimulated cultures, the addition of partially purified bovine interleukin 1 reduced the leukotoxin-induced inhibition. Recombinant interleukin 2 had a similar effect. Addition of the glycolipid, monosialoganglioside was also able partially to overcome the inhibition.

Effect of in vitro selenium repletion on bovine lymphocyte proliferation

Deficient or toxic levels of selenium (Se) are known to impair immune function. Peripheral blood mononuclear cells (PBMC) were isolated from Holstein heifers of adequate Se status. PBMC were plated at densities of 0.5, 1.0, and 2.0 × 10 5 cells/well in 96-well microtiter plates and stimulated with T-cell mitogens, concanavalin A (ConA) and phytohemagglutinin-P (PHAP), and pokeweed mitogen (PWM), a T-dependent B-cell mitogen. Sera incorporated into the incubation system were from two newborn calves born to Se-deficient dams and averaged 20 ng Se/ml. Se additions of 50-400 ng/ml resulted in significant enhancement of PWM-stimulated proliferation relative to controls (no added Se). Enhancement of proliferation was cell density dependent, occurring predominately at a density of 0.5 × 10 5 cells/well. In contrast, proliferative responses of PBMC to ConA and PHAP were generally not enhanced by repletion with the lower levels of Se. Supplementation with higher levels of Se (1600–3200 ng/ml) inhibited proliferative responses to all mitogens tested regardless of cell density in culture. In summary, repletion with physiological levels of Se selectively stimulated B-cell proliferation an effect which was dependent on cell density.

Inhibition of Cell Proliferation by Basic Proteins from Bull Seminal Plasma

The fraction of basic proteins has been shown to suppress the concanavalin A-induced proliferation of bovine lymphocytes effectively in a dose-dependent manner. The inhibitory effect was observed within a concentration range where the fraction of basic proteins was not cytotoxic. By separating the basic proteins, only two proteins, seminal ribonuclease and seminal antimicrobial protein were found to be active in inhibition of lymphocyte proliferation. Both proteins appear to function differently. At a concentration of 56 micrograms/ml, where both proteins inhibited DNA synthesis, seminal ribonuclease also inhibited RNA synthesis by greater than 50% whereas seminal antimicrobial protein had no effect on RNA synthesis. It was furthermore observed that the suppressive effect of the basic proteins was independent of the mitogenic stimulus and proliferation of both, T- as well as B-lymphocytes was inhibited by those proteins.

1,25-Dihydroxyvitamin D 3 enhancement of concanavalin-A induced bovine lymphocyte proliferation: Requirement of monocytes

The effect of 1,25-(OH)2D3 on Concanavalin-A (ConA) mediated bovine lymphocyte proliferation was dependent on the magnitude of the proliferative response. Lymphocytes from cows with high inherent proliferative responses (134,904 +/- 15,488 cpm/well) were unaffected by 1,25-(OH)2D3; whereas, lymphocytes from cows with low inherent proliferative responses (46,317 +/- 6000 cpm/well) were stimulated 75% (P less than 0.01) by 10(-10)M 1,25-(OH)2D3. At low initial proliferation rates, seen with low density cultures enriched for lymphocytes, the effect of 1,25-(OH)2D3 on proliferation changed from inhibition in the absence of added monocytes to stimulation at 18-23% monocyte addition. These results suggest that 1,25-(OH)2D3 possesses both stimulatory and inhibitory effects on ConA-induced proliferation that are monocyte dependent.

Suppression of antigen- and mitogen-induced proliferation of bovine lymphocytes by excretory-secretory products of Oesophagostomum radiatum

Excretory-secretory products (ESP) isolated from in vitro-grown stage-3 to -4 larvae of Oesophagostomum radiatum were found to inhibit both the in vitro antigen-specific proliferation of keyhole limpet hemocyanin- and ovalbumin-primed lymphocytes and the proliferation induced by the T-cell mitogen concanavalin A. As little as 50 ng of ESP protein per culture resulted in 50% reductions of subsequent proliferative responses. Antigen-induced responses were 100 to 1,000 times more sensitive to inhibition than were mitogen-induced responses. The inhibitory activity was found to affect the induction of proliferation as evidenced by the observation that complete inhibition was seen when ESP were added to cultures within the first 24 h. ESP were found to have no inhibitory activity when added 72 h after the initiation of the cultures. The inhibition was not a result of a direct action upon macrophages because pulsing of adherent cells with ESP had no more effect on a subsequent proliferative response than did a pulsing of the culture vessel itself. The inhibitory activity eluted from high-pressure liquid chromatography columns in the same fractions as protein standards with molecular weights of 25,000 to 35,000. Of special interest is the fact that this inhibitor of the expansion of immunoreactive clones of lymphocytes is found associated with the stages of parasites most intimately associated with host tissues, namely larval stages 3 and 4.
