Detection of bluetongue virus antibodies in sheep from Paraná, Brazil

Bluetongue (BT) is an infectious and non-contagious disease caused by bluetongue virus (BTV) belonging to the genus Orbivirus. It is transmitted by a hematophagous vector, Culicoides sp., to ruminants, particularly to sheep, which are most susceptible to this disease. The main serological tests are agar gel immunodiffusion (AGID), which is recommended by the World Organization for Animal Health (OIE), and the competitive enzyme-linked immunosorbent assay (cELISA), which has the advantage of no cross-reaction with other orbiviruses. The aim was to compare the results of these two tests by conducting them on sera collected from sheep in the state of Paraná, Brazil. From March to October 2017, serum samples were collected from 270 sheep from 10 farms in six mesoregions of Paraná. The samples were subjected to AGID and cELISA to detect antibodies against BTV. Based on the test results, we classified the sheep as low, moderate, and high occurrence. The results demonstrated that 64.81% (175/270) of the sheep were seropositive through the cELISA test, showing a high occurrence, and 41.11% (111/270) were seropositive through the AGID test, indicating a moderate occurrence. The concordance between the tests was moderate (0.51) as determined by the Kappa coefficient. Among the studied farms, 90% (9/10) presented at least one seropositive sheep, and the number of animals tested positive by the cELISA test was higher than those by the AGID test. Favorable climate, which favors the presence and multiplication of the culicoid vector and the occurrence of infection, was the biggest predominant factor responsible for the obtained results. The low occurrence in farms with milder climate suggest that the presence of antibodies also occurs due to the low pathogenicity of circulating serotypes in the different mesoregions studied. It is concluded that BTV infection is present in the sheep herds in Paraná, and the occurrence was moderate detected by AGID test and high detected by cELISA test.

Presence of Antibodies against Bluetongue Virus (BTV) in Sheep 5 to 7.5 Years after Vaccination with Inactivated BTV-8 Vaccines.

Thirty-six female sheep, previously vaccinated against Bluetongue virus serotype 8 (BTV-8) using inactivated vaccines, were included in this field study. In Germany, vaccination was compulsory in 2008 and 2009, voluntary in 2010 and early 2011, and later, was prohibited in 2011. Due to their age, eighteen sheep had been vaccinated for two or more consecutive years, while a further eighteen animals had only been vaccinated once or not at all. The sheep were blood sampled five (n = 31) to 7.5 years (n = 5) after their last vaccination. All serum samples (n = 36) were tested for BTV group-specific antibodies by an ELISA (IDScreen® Bluetongue Competition assay, ID Vet). In five of the animals, the BTV-8 serotype-specific antibody titers were measured by serum neutralization (SN). The majority of sheep that were vaccinated annually for two or more years showed a positive ELISA (14/18 sheep) and a SN (two of two sheep) result 5 years after their last vaccination. Most of the sheep vaccinated fewer than twice showed a negative ELISA result 5 to 7.5 years after their last vaccination (13/18 animals). The three animals in this group tested by SN showed one negative and two positive results. This short communication is the first to describe the presence of BTV antibodies in sheep 5 to 7.5 years after vaccination with inactivated BTV-8 vaccines.

Epidemiological investigation of bluetongue virus antibodies in sheep in Iran

AbstractAim: Bluetongue is a non-contagious, infectious viral disease of domestic and wild ruminants; which is believed to have originated in Africa. The epidemiology of Bluetongue virus infection is poorly defined in many parts of the world, including a wide range of Asia and the Middle East. This paper reports the results of a Bluetongue serological survey in sheep from some provinces of Iran during 2008-2009. Material and Methods: A total of 996 sheep sera were collected from 8 provinces in Iran and tested for Bluetongue virus specific using c-ELISA. Results: The results showed that the Bluetongue virus seroprevalence of sheep over the entire study areas was 34.93%, with the highest and lowest prevalence seen in West-Azerbaijan (64.86%) and Qom (12.1%) areas respectively. Conclusion: The results demonstrated a high prevalence of Bluetongue antibodies in Iranian sheep, giving serological evidence of extensive exposure to Bluetongue virus infection in some provinces of the country. Prevalence of bluetongue virus antibodies in sheep from Distrito Federal, Brazil

The aims of the present study were to determine the prevalence of bluetongue virus antibodies in sheep from Distrito Federal. Sera from 606 sheep of 18 herds were submitted to the agar-gel immunodiffusion for bluetongue virus antibodies. The prevalences of bluetongue infection found in Distrito Federal were 100% (CI 95%: 84.67 to 100.00) for flocks and 52.37% (389/606) (CI 95%: 35.76 to 68.98) for animals. Thus, data from the present study showed that infection by bluetongue virus is highly widespread in sheep flocks in the Distrito Federal, which intensifies the need for assessments on the impact of this disease in Brazil.

Seroprevalence of bluetongue virus antibodies in sheep in Kurdistan province in west of Iran

This study was conducted to determine the seroprevalence of bluetongue virus antibodies in sheep in Kurdistan province in west of Iran during 2011-2012. According to results, the seropositive rates of sheep in Kurdistan were 45.9%. The results showed that in area there is widespread presence of the bluetongue virus antibodies in the sheep population as 123 sheep’s out of 268 were affected.

High seroprevalence of bluetongue virus antibodies in Sheep, Goats, Cattle and Camel in different districts of Saudi Arabia

Aim: To estimate the prevalence and distribution of serum antibodies to BTV in different domesticated animals in different localities of Saudi Arabia. Materials and Methods: A total of 4845 field sera collected from different animal species within 10 districts in the Kingdom of Saudi Arabia were screened for the presence of group-specific BTV antibodies by competitive ELISA (c ELISA). Results: The overall BTV antibody prevalence was 54.1%, 53.3%, 44.8% and 25.7% in sheep, goat, cattle and camel respectively (at 95% confidence level). The Jizan and Eastern Province districts were the regions with the highest prevalence resulting 65.8% of sheep, 68.2% of goats, 49.3% of cattle, 44% of camel in Jizan and 65.8% of sheep, 62.5% of goats, 53.4% of cattle, 28.5% of camel in Eastern Province positive to c-ELISA. The second highest rate was in Najran district where the seropositivity for Bluetongue was found to be 60% of sheep, 57.9% of goats, 47.2% of cattle and 29.3% of camel. Our results recorded positive animals in all examined districts which indicate serological evidence of exposure to infection was widely distributed all over the country. Conclusions: These results demonstrate the high occurrence of the BTV that emphasize the necessity to a well-defined control strategy for preventing and controlling the BTV in Saudi Arabia.

Dot immunobinding assay for the detection of bluetongue virus antibodies in sheep experimentally inoculated with bluetongue virus type 1

Dot immunobinding assay (DIA) was evaluated for the detection of bluetongue virus (BTV) antibodies in sheep experimentally inoculated with BTV 1. Serum samples collected on 14, 21, 28, 43 and 60 days post infection (dpi) were positive for precipitating antibodies by the agar gel precipitation test (AGPT) while antibodies could be detected as early as 7 dpi by DIA and ELISA. Virus neutralizing antibodies were detected first at 14 dpi. The sensitivity of the four tests was compared on the same serum samples collected at different intervals. The results indicated that DIA was more sensitive than AGPT and the serum neutralization test and as sensitive as ELISA. Thus due to sensitivity simplicity and economy, DIA could replace AGPT for diagnosis and serological survey for BTV infection in animals.

Dot immunobinding assay in comparison with enzyme-linked immunosorbent assay for the detection of bluetongue virus antibodies in sheep

A total of 384 sheep serum samples collected from two organised sheep farms was tested by dot immunobinding assay (DIA) and indirect enzyme-linked immunosorbent assay (I-ELISA) for the presence of bluetongue virus (BTV) antibodies. The results of both these assays were compared to find a sensitive, specific, rapid, easily performed and economical test for the diagnosis of bluetongue disease. DIA detected BTV antibodies in 210 samples (54.94%) and I-ELISA detected 157 positive samples (40.88%). Competitive ELISA (C-ELISA) was performed to check the discrepancies in I-ELISA and DIA. On the basis of these tests the overall agreement, relative specificity and sensitivity between ELISA and DIA were 75%, 87.6% and 100%, respectively. DIA was found to be a rapid, sensitive, easily performed and economical test as compared to ELISA.
