β-Hydroxybutyrate (BHB) and glucose concentrations in the blood of dairy calves as influenced by age, vaccination stress, weaning, and starter intake including evaluation of BHB and glucose markers of starter intake

The objective of this research was to determine how blood β-hydroxybutyrate (BHB) and glucose are affected by age, time of day, stress, weaning, forced intake restriction, and voluntary starter intake in calves between 1 and 9 wk of age, and to evaluate if either is an acceptable proxy for starter intake. Holstein calves were fed a 27% crude protein, 17% fat milk replacer at 660 g of dry matter daily along with free-choice starter and water. Calves were weaned on d 42. Jugular blood was sampled at 0800, 1200, and 1600 h, and within 5 min of sampling BHB, and glucose concentrations were estimated using test strips (Nova Max Plus meter, Nova Biomedical Corporation, Waltham, MA). Age effects and time of day were estimated by sampling blood weekly (d 6, 13, 20, 27, 34, 41, and 48). To determine vaccination stress, a Pasteurella vaccine was administered after blood sampling at 0800 h on d 36. Effect of voluntary starter intake was tested by selecting calves for low and high intakes (d 35 to 39) and sampling on d 40, 41, 43, and 44. Starter intake restriction was tested by restricting intake in half of the calves and sampling on d 60 and 61. Data were analyzed with repeated measurements in a mixed model procedure with either within-calf effect (day or week) or within-calf effects (hour, and day or week) included in the model. Time of day did not affect blood BHB and glucose in the first 6 wk. Blood BHB was greater in wk 7 versus wk 1 to 6. Blood glucose was greater in the first 5 wk compared with wk 6 and 7. Blood BHB increased and glucose decreased with increasing starter intake. Blood BHB declined due to vaccination, but glucose was unaffected. Starter intake restriction reduced BHB for 3 d and glucose for 2 d after restriction. Both were affected by time of day. Around weaning (d 40 to 44), BHB and glucose increased with increasing starter intake. In this research, neither blood BHB nor glucose was a good proxy for starter intake. Blood BHB was positively and glucose negatively related to starter intake; however, relationships were weak, variable, and affected by time of day, stress, and intake restriction. Over 30% of calves tested ≤0.2 mmol/L BHB when consuming >1,250 g/d of starter, and test strip increments were 0.1 mmol/L, which represented >25% of the mean blood BHB concentration. In this study, neither blood BHB nor glucose was an acceptable proxy for estimating starter intake.

Elevation of blood β-hydroxybutyrate concentration affects glucose metabolism in dairy cows before and after parturition

Recent studies in mid- and late-lactation dairy cows showed that β-hydroxybutyrate (BHB) infusion had a considerable effect on glucose metabolism and immune response during intramammary lipopolysaccharide challenge. The objective of the present study was to infuse BHB during the dry period and after parturition to investigate the effects of elevated plasma BHB concentrations on metabolism and endocrine changes in transition dairy cows. The hypothesis tested was that regulation of glucose metabolism would change at different physiological stages and an additional elevation of BHB concentration would alter glucose concentration. Multiparous Holstein cows in wk -2 (antepartum, a.p.; n = 6) and wk +2 (postpartum, p.p.; n = 8) relative to calving were infused (4 h from 0800 to 1200 h) with a BHB solution to increase plasma BHB concentration to 1.5 to 2.0 mmol/L (HyperB). The same period the next day without any infusion was considered the control period (CON). Blood samples were taken 1 h before the start of infusion as reference samples and every 30 min during the following 6 h (4 h of infusion and 2 h after infusion) in the HyperB and CON periods, and analyzed for glucose, BHB, insulin, and glucagon concentrations. During the steady state period (the latter 2 h of the 4-h infusion), plasma BHB concentration reached 1.87 ± 0.05 mmol/L (a.p.) and 1.93 ± 0.05 mmol/L (p.p.) in HyperB compared with 0.55 ± 0.06 mmol/L (a.p.) and 0.64 ± 0.04 mmol/L (p.p.) in CON, respectively. The 4-h average BHB infusion rate was 12.4 ± 1.0 and 13.3 ± 0.9 μmol/kg of BW per minute in wk -2 and +2, respectively. Infusion of BHB caused a decrease of plasma glucose concentrations relative to preinfusion levels both before and after parturition, although basal glucose concentrations were different before and after calving. Infusion of BHB increased plasma insulin concentrations a.p. but not p.p., despite a higher basal insulin concentration before than after parturition. These findings show that effects of hyperketonemia on plasma glucose concentrations are similar before and after calving but that endocrine adaptation to hyperketonemia differs before and after parturition. We assume that BHB is a metabolic key regulator in early lactating dairy cows and may affect glucose concentration by further pathways such as gluconeogenesis and altered lipolysis.

Blood BHBA declined (P < 0.05) due to vaccination but glucose was unaffected. Starter intake restriction reduced BHBA for 3 d (P < 0.05) and glucose for 2 d (P < 0.05) after intake restriction. Around weaning (d 40 to 44), BHBA and glucose increased (P < 0.05) with increasing starter intake. Blood BHBA was positively and glucose negatively related with starter intake; however, relationships were weak, variable, and impacted by time of day, stress, and intake restriction. Over 30% of calves tested ≤ 0.2 mmol/L BHBA when consuming > 1250 g/d of starter, and test strip increments were 0.1 mmol/L which represented > 25% of the mean blood BHBA concentration. Niacin feeding to fresh dairy cows: immediate effects on health and milk production

Early lactation dairy cows are frequently in negative energy balance and susceptible to ketosis, fatty liver and metritis. Because of its anti-lipolytic properties, the B-vitamin niacin could reduce negative energy balance by reducing non-esterified fatty acids for ketogenesis, thereby reducing hyperketonemia. We determined effects of feeding ruminally protected niacin (RPNi) on lipolysis during the fresh period using blood non-esterified fatty acids concentrations as a ketosis indicator, blood β-hydroxybutyrate concentrations as an indicator of lipid mobilisation, as well as dry matter (DM) intake, milk and milk component yields, in 906 multi-parity Holstein cows from ~14 days before calving through the immediate fresh period. Prior to calving, cows were co-mingled in one pen and fed the same total mixed ration without RPNi. Between 24 and 36 h postpartum, cows were assigned to fresh pens and fed the same fresh cow total mixed ration, except for RPNi at 0, 3.5, 7 or 14 g niacin/cow.day. During the close-up and fresh periods, cows were sampled for tail vein blood. Milk yield and composition was measured twice at a 140-days interval in the fresh pens postpartum. The 3.5 g/day RPNi feeding tended to decrease ketosis prevalence (% of cows with β-hydroxybutyrate ≥ 1.44 mg/dL) from 36% to 20% (P = 0.06) and also tended (P = 0.07) to increase DM intake from 19.3 to 21.5 kg DM/day versus Control. The RPNi effect tended to increase with duration of RPNi feeding, with no effects at 7 ± 3.9 days in milk, but milk (P = 0.10), milk fat (P = 0.11) and milk energy (P = 0.07) yields tending to be higher at 21 ± 3.9 days in milk. Conversely, 14 g/day RPNi had no effect on ketosis prevalence or DM intake. However, milk (P = 0.10), milk fat (P = 0.11) and milk energy (P = 0.07) yields tended to decrease versus Control. Overall, low level RPNi feeding was judged to improve health and production in fresh cows, but higher feeding levels had clear negative impacts.

Relación entre concentración sanguínea de β-hidroxibutirato e indicadores productivos, reproductivos y de salud en hatos Jersey y Holstein

Se correlacionó la concentración sanguínea de β-hidroxibutirato (βHBA) con indicadores productivos, reproductivos y de salud, en 2 hatos de vacas de alta producción, de las razas Jersey y Holstein, ubicados en Oreamuno (Jersey) y La Unión (Holstein) de Cartago, Costa Rica; ambos con vacas primíparas y multíparas, en sistemas de pastoreo. Se tomaron muestras de sangre de vasos coccígeos de 117 y 114 vacas Jersey a los 8±3 y 30±3 días de lactancia (DL), y de 72 y 50 Holstein a 5±3 y 30±3 DL, respectivamente. En cada muestra se analizó in situ la concentración de βHBA, y la información productiva, reproductiva y de salud del hato se registró en el programa VAMPP Bovino 3.0. Mediante análisis de asociación, se establecieron relaciones entre concentración de βHBA e indicadores de interés. Un análisis de razón de oportunidades (“odds ratio”) señaló a las Jersey que sufrieron hipocalcemia clínica con 3,63 veces la probabilidad de tener concentraciones de βHBA superiores a 1,4 mmol.l-1, en contraste con las sanas. Sin embargo, la información obtenida sugiere que concentraciones promedio de βHBA de 0,66 (IC 95%: 0,59-0,74) a los 8±3 DL en vacas Jersey y de 0,59 (IC 95%: 0,52-0,65) a los 5±3 DL en las vacas Holstein, así como concentraciones promedio de βHBA de 0,91 (IC 95%: 0,78-1,05) en las Jersey y de 0,69 (IC 95%: 0,58-0,80) en las Holstein a los 30±3 DL no afectaron la producción láctea ni la reproducción en hatos cuya alimentación se basó en uso intensivo de forrajes tropicales más 2,5 a 3 kg por vaca por día de un alimento balanceado (1,9 Mcal ENL.kg-1, 48% almidón).

Measurement of β-hydroxybutyrate in capillary blood obtained from an ear to detect hyperketonemia in dairy cows by using an electronic handheld device

The primary objective of the present study was to test whether capillary blood obtained by puncturing the skin of an ear with a minimal invasive lancet technique is able to detect hyperketonemia (HYK) in dairy cows. Furthermore, test characteristics of a new available handheld device, the FreeStyle Precision Neo (FSP-Neo, Abbott GmbH & Co. KG, Wiesbaden, Germany) for determination of β-hydroxybutyrate (BHB) concentrations in bovine blood were evaluated by comparing the measurements with a laboratory reference. The BHB concentration was determined with the FSP-Neo device in 720 capillary blood samples from 3 different sampling sites (left, right ear, and repeated measurement) and in 240 samples from a coccygeal vessel. The concentration of BHB in serum harvested from the coccygeal blood samples was analyzed at the laboratory and was used as reference. The Spearman correlation coefficient (ρs) between the BHB concentrations in capillary blood measured with the handheld device and the reference test was between 0.76 and 0.81. Using capillary blood, the mean ± standard deviation BHB difference compared with the reference test was 0.20±0.47 mmol/L for all 3 sampling locations at the ears. The receiver operating characteristic analyses for the FSP-Neo device resulted in an optimized threshold for the detection of subclinical ketosis (SCK) in capillary blood of 1.3 mmol/L (left and right ear) and 1.2 mmol/L (repeated measurements). Applying these adjusted threshold sensitivities (Se) for all 3 capillary sampling sites at the ear were 100%, and specificities (Sp) ranged between 93 and 94%. Hence, we conclude that all sampling locations were suitable to identify cows suffering from SCK. The reference test compared with BHB measurements in coccygeal blood resulted in a ρs of 0.92 with a mean ± standard deviation of 0.02±0.21 mmol/L. The receiver operating characteristic analyses for the FSP-Neo device resulted in an optimized threshold for the detection of SCK in coccygeal blood of 1.1 mmol/L, with a corresponding Se and Sp of 100 and 95%, respectively. Because capillary blood is easily achievable from an ear, particularly if animals are fixed in headlocks for routine checkups, this technique is considered as an additional minimally invasive method for the identification of dairy cows suffering from HYK.

Randomized clinical trial of a calcium supplement for improvement of health in dairy cows in early lactation

Prophylactic Ca supplementation immediately after calving is a common strategy to prevent clinical and subclinical hypocalcemia in parturient dairy cows. The objective of this study was to evaluate the effect of prophylactic administration of an injected Ca supplement on blood Ca concentration at 24 and 48h after treatment, incidence risk of clinical disease and culling, milk production in early lactation, and probability of pregnancy at first insemination. Cows without signs of visible milk fever (n=984) from 7 farms were blocked by parity and randomly assigned to receive either Ca gluconate (35% wt/vol) in combination with Ca glucoheptonate (10% wt/vol; TheraCalcium, Vétoquinol Canada Inc., Lavaltrie, Quebec) or a placebo (medication vehicle solution with no Ca) at first contact with each cow after calving and again 12 to 24h later. Each dose was 120mL injected subcutaneously over 2 sites. Total serum Ca concentration (tCa) was measured from coccygeal blood samples before (time 0) and 24 and 48h after first treatment in a subsample of cows (n=129). Blood β-hydroxybutyrate concentrations were measured from all cows twice between 3 and 16d in milk at weekly visits and cows were evaluated for vaginal discharge once between 28 and 42d in milk. Disease events, production data from the first 3 Dairy Herd Improvement milk tests, reproduction, and culling data were collected from each herd. For cows that had received 1 injection of Ca before the blood sample at 24h (n=95), tCa was significantly higher in the treated cows: mean ± standard error, 2.03±0.03 versus 1.90±0.03mmol/L, accounting for tCa at time of enrollment and a treatment by tCa at enrollment interaction. At 48h, no significant difference was found in tCa between treatment and control (mean ± SE, 2.12±0.02 and 2.10±0.03mmol/L, respectively). Cows treated with the Ca product were significantly less likely to have received intravenous, subcutaneous, or oral supplemental Ca for exhibiting clinical signs of hypocalcemia than control cows (5.0 vs. 8.4%). No effect was found of treatment on retained placenta, metritis, hyperketonemia, prevalence of purulent vaginal discharge, culling from the herd, early lactation production, probability of pregnancy to first artificial insemination, or time to pregnancy. With this subcutaneous prophylactic Ca treatment regimen, blood Ca levels were temporarily increased at 24h after treatment, but no effect was observed of supplemental Ca on the risk of disease or culling, milk production, or reproductive performance.

The effect of subclinical ketosis on activity at estrus and reproductive performance in dairy cattle

Our aims were to investigate the influence of subclinical ketosis (SCK) on physical activity at estrus using a neck accelerometer device and on future reproductive performance. Two hundred three Holstein-Friesian cows were studied on 3dairy farms in Northwest England between September 2013 and March 2014. Seventeen percent (35 of 203) of the enrolled cows were affected with SCK between 7 and 21d in milk, defined as a blood β-hydroxybutyrate concentration of 1.2 to 2.9mmol/L. Time to event analyses and multivariable regression analyses were used to assess the effect of SCK on reproductive performance and activity at estrus. The SCK cows exhibited a lower peak activity (measured as the number of standard deviations above mean activity) and shorter duration in activity clusters associated with first estrus and first insemination postpartum, compared with non-SCK cows. Peak activity and cluster duration associated with the insemination that led to a pregnancy were not different between SCK and non-SCK cows. Calving to first estrus, calving to first insemination, and calving to pregnancy intervals were prolonged in SCK cows. First insemination was 4.3 times (95% confidence interval=1.6 to 15.0) less likely to be successful in SCK cows compared with non-SCK cows. Adjusted mean number of inseminations per pregnancy was 2.8 for SCK cows and 2.0 for non-SCK cows. The current study confirms the long-lasting effects of SCK on reproductive efficiency. Furthermore, it is indicated that physical activity around estrus is reduced by SCK in early lactation, but this negative effect appears to diminish as cows progress through lactation.

Effect of different alfalfa hay levels on growth performance, rumen fermentation, and structural growth of Holstein dairy calves.

Introducing forage in dairy calf diets during the preweaning period stimulates development of rumen capacity and function and gut wall integrity. It was hypothesized that calves fed alfalfa hay (AH) at greater levels (up to 25% of DM) would result in an increase in starter diet intake and growth performance. The objective of this study was to determine the effects of increasing levels of supplemental AH on growth performance, rumen fermentation, blood metabolites, and feeding behavior in intact male dairy calves. For this purpose, 45 Holstein male calves (3 d of age and 41 ± 2.5 kg of BW) were randomly assigned to 1 of the following 3 starter diets differing in their forage level: 1) diet without AH (control), 2) diet with 12.5% AH on a DM basis, and 3) diet with 25% AH on a DM basis. Supplementation of AH in the starter diets linearly increased total DMI and ADG during the postweaning ( < 0.01) and overall ( = 0.02) periods. Alfalfa hay supplementation linearly increased ( < 0.01) ruminal pH on d 35 and 70 of the study. The concentrations of total VFA ( = 0.85), acetate ( = 0.74), propionate ( = 0.93), and butyrate ( = 0.91) were not affected by treatments. Furthermore, forage supplementation tended (linearly, = 0.08) to increase the acetate to propionate ratio compared with the control calves. Blood β-hydroxybutyrate (BHBA) concentrations in the control calves were less ( = 0.03) than in the AH-fed calves on d 70 of the study. Among all the measured variables of skeletal growth, supplementation of AH in the starter diets linearly increased heart girth ( = 0.05) and abdominal girth ( < 0.01) on d 70 of the study. These results indicate that the feed intake, feeding behavior, rumen fermentation parameters, and blood BHBA concentration may be affected by rations differing in AH level so that providing calves with 25% AH can improve calf performance.

Technical note: Validation of a handheld meter for measuring β-hydroxybutyrate concentrations in plasma and serum from dairy cows

The use of handheld meters for cow-side tests for β-hydroxybutyrate (BHB) concentrations in whole blood has become common. The aim of this study was to compare serum BHB (sBHB) and plasma BHB (pBHB) concentrations analyzed using either a “gold standard” enzymatic laboratory method (LM; Randox Laboratories Ltd., Antrim, UK; cat. no. RB1007) or a handheld meter (PX; Precision Xtra, Abbott Diabetes Care Inc., Alameda, CA). Results from 374 (187 serum and 187 plasma) samples taken from Holstein cows from 11 d before (52 samples) to 5 d after parturition (137 samples) were used for the analysis. Statistical analysis was performed using the MIXED, REG, and LOGISTIC procedures of SAS (v9.4; SAS Institute Inc., Cary, NC). A linear mixed model with repeated measures was fitted for LM and PX. Regression and correlation analyses were completed to estimate the relationship and agreement between the 2 methods. Lin’s concordance correlation coefficient (CCC) and Bland-Altman plots were used to evaluate agreement between LM and PX. Cross-validation by randomly splitting the data in model-building and validation sets was performed to estimate and validate the equation that predicted the LM results using PX. Receiver operating characteristic (ROC) curves were used to estimate the sensitivity (Se) and specificity (Sp) of PX at different threshold levels. The CCC was 0.74 for pBHB and 0.68 for sBHB. The 95% confidence interval of agreement of the Bland-Altman plot encompassed 96% of the difference between LM and PX for pBHB and 95% for sBHB. The mean difference for pBHB was −0.50±0.25mmol, and that for sBHB was −0.63±0.41mmol. The highest Se and Sp for PX were achieved when the threshold for ketosis was set to 1.8mmol/L for pBHB and 2.1mmol/L for sBHB. The area under the ROC curve was 0.97 for pBHB and 0.96 for sBHB. The negative bias shown by the Bland-Altman plots suggested that PX yielded higher pBHB and sBHB concentrations than the LM. However, the excellent test characteristics and area under the ROC curve close to 1 indicated that PX at the adjusted thresholds was able to accurately classify between samples <1.2mmol/L and those ≥1.2mmol/L based on the gold standard test.

Comparative Clinical and Haematological Investigations in Lactating Cows with Subclinical and Clinical Ketosis

Abstract Ketosis of lactating cows is among the most common metabolic diseases in modern dairy farms. The economic importance of the disease is caused by the reduced milk yield and body weight loss, poor feed conversion, lower conception rates, culling and increased mortality of affected animals. In the present study, a total of 47 high-yielding dairy cows up to 45 days in milk (DIM) are included. All animals were submitted to physical examination wich included checking the rectal body temperature, heart rate, respiratory and rumen contraction rates, and inspection of visible mucous coats. The body condition was scored, and blood β-hydroxybutyrate (BHBA) concentrations were assayed. The cows were divided into 3 groups: first group (control) (n=24) with blood β-hydroxybutyrate level &lt;1.2 mmol/l, second group (n=15) with blood β-hydroxybutyrate between 1.2-2.6 mmol/l (subclinical ketosis) and third group (n=8) with blood β-hydroxybutyrate &gt;2.6 mmol/l (clinical ketosis). Whole blood samples were obtained and analyzed for Red Blood Cell (RBC, 1012/l), Hemoglobin (HGB, g/l), Hematocrit (HCT, %), Mean Corpuscular Volume (MCV, fl), Mean Corpuscular Hemoglobin (MCH, pg), Mean Corpuscular Hemoglobin Concentration (MCHC, g/l), White Blood Cell (WBC, 109/l), Lymphocytes (LYM, 109/l), Monocytes (MON, 109/l), Granulocytes (GRA, 109/l), Red Blood Distribution Width (RDW, %), Red Blood Cell Distribution Width Absolute (RDWa, fl), Platelets (PLT, 109/l) and Mean Platelet Volume (MPV, fl). In this study, deviations in the clinical parameters in the control group and in those with subclinical ketosis were not identified. The cows from the third group (clinical ketosis) exhibited hypotonia, anorexia and body weight loss vs. control group. Hematological analysis showed leukocytosis and lymphocytosis in cows with subclinical ketosis vs. control group. In cows with clinical ketosis WBC counts decreased (leukopenia), while hemoglobin content and hematocrit values are higher vs. control group. Blood BHBA values are higher in both groups of ketotic cows vs. the control group. The other analyzed parameters (RBC, MCH, MCHC, MCV, RDW, RDWa, MON, GRA, PLT and MPV) were close to control values.

Pregnancy toxaemia in ewes: Development of an experimental model and potential interactions with gastrointestinal nematode infections

Objective of the present study was to develop an experimental model that could be used for the study of pregnancy toxaemia in ewes. The study included 28 ewes, which initially had received effective anthelmintic treatment and, then, received a mixture of trichostrongylid infective larvae. Until the 60th day of pregnancy, per ewe daily ration was 1.30kg of a concentrate feed (net energy: 0.844 FUL) plus 2.50kg of alfalfa hay. From the 60th to 100th day of pregnancy, per ewe daily ration was 0.60kg of the same concentrate plus 2.00kg of hay. From the 100th day of pregnancy, per ewe daily ration was 0.50kg of a reduced energy concentrate feed (net energy: 0.748 FUL) plus 0.50kg of alfalfa hay for ewes with one foetus; during that period, respective figures for ewes with two foetuses were 0.60kg and 0.50kg and for ewes with three foetuses were 0.80kg and 0.50kg. In total, 16 ewes developed increased β-hydroxybutyrate blood concentrations, characteristic of pregnancy toxaemia. There was a significant reverse correlation (P=0.016) between blood β-hydroxybutyrate concentrations and lamb birth bodyweight. Ewes with pregnancy toxaemia had greater faecal epg counts than ewes with no pregnancy toxaemia (P<0.025); there was also a significant reverse correlation between faecal epg counts and lamb birth bodyweight (P=0.03) and a significant correlation between blood β-hydroxybutyrate concentrations and faecal epg counts (P<0.001). Ewes with pregnancy toxaemia also had smaller blood concentrations of glucose than ewes with no pregnancy toxaemia (P=0.033). In conclusion, administration of a concentrate feed with reduced energy content during the last stage of pregnancy induced pregnancy toxaemia in ewes, at the same time covering satiation requirements of the animals and thus maintaining welfare standards. Parasitism might have further contributed to improving the efficacy of the model.

A randomised trial of a medium-chain TAG diet as treatment for dogs with idiopathic epilepsy.

Despite appropriate antiepileptic drug treatment, approximately one-third of humans and dogs with epilepsy continue experiencing seizures, emphasising the importance for new treatment strategies to improve the quality of life of people or dogs with epilepsy. A 6-month prospective, randomised, double-blinded, placebo-controlled cross-over dietary trial was designed to compare a ketogenic medium-chain TAG diet (MCTD) with a standardised placebo diet in chronically antiepileptic drug-treated dogs with idiopathic epilepsy. Dogs were fed either MCTD or placebo diet for 3 months followed by a subsequent respective switch of diet for a further 3 months. Seizure frequency, clinical and laboratory data were collected and evaluated for twenty-one dogs completing the study. Seizure frequency was significantly lower when dogs were fed the MCTD (2·31/month, 0-9·89/month) in comparison with the placebo diet (2·67/month, 0·33-22·92/month, P=0·020); three dogs achieved seizure freedom, seven additional dogs had ≥50 % reduction in seizure frequency, five had an overall <50 % reduction in seizures (38·87 %, 35·68-43·27 %) and six showed no response. Seizure day frequency were also significantly lower when dogs were fed the MCTD (1·63/month, 0-7·58/month) in comparison with the placebo diet (1·69/month, 0·33-13·82/month, P=0·022). Consumption of the MCTD also resulted in significant elevation of blood β-hydroxybutyrate concentrations in comparison with placebo diet (0·071 (sd 0·035) v. 0·053 (sd 0·028) mmol/l, P=0·028). There were no significant changes in serum concentrations of glucose (P=0·903), phenobarbital (P=0·422), potassium bromide (P=0·404) and weight (P=0·300) between diet groups. In conclusion, the data show antiepileptic properties associated with ketogenic diets and provide evidence for the efficacy of the MCTD used in this study as a therapeutic option for epilepsy treatment.

Rumen fermentation, blood metabolites, and growth performance of calves during transition from liquid to solid feed: Effects of dietary level and particle size of alfalfa hay

This study evaluated the effects of particle size (PS) and dietary level of alfalfa hay (AH) on rumen fermentation parameters, blood metabolites, eating behavior, and growth performance in dairy calves during transition from liquid to solid feed. Sixty newborn dairy calves (41±2.5,kg of body weight) were used in a 2×2 factorial arrangement with the factors dietary AH level (medium, 12.5%, or high, 25%, on DM basis) and PS (fine=1mm or medium=3mm, as geometric means) of AH. Hence, the dietary treatments were (1) medium level of AH with fine PS (M-FPS), (2) medium level of AH with medium PS (M-MPS), (3) high level of AH with fine PS (H-FPS), and (4) high level of AH with medium PS (H-MPS). Particle size of AH did not affect total DMI (TDMI) during the preweaning period, although TDMI was greater for calves fed MPS than in those fed FPS during the postweaning and overall periods. Calves fed MPS spent more time eating solid feed and ruminating and less time on nonnutritive oral behaviors compared with FPS calves. The dietary level of AH did not affect behavioral parameters. Average daily gain of calves was not affected by dietary treatment before weaning. During the postweaning and overall periods, average daily gain was greater in calves fed MPS than in those fed FPS at the 25% AH level, but this effect was absent with 12.5% AH. Furthermore, the rumen pH values on d 35 and 70 of the study were greater for MPS than for FPS, regardless of the dietary level of AH. Effects of AH level, PS, and their interaction did not affect blood glucose concentrations in developing calves. These results indicate that feed intake, feeding behavior, rumen fermentation parameters, and blood β-hydroxybutyrate concentration may be affected by rations differing in forage PS; thus, providing calves with MPS can improve calf performance and reduce their nonnutritive oral behaviors.

Effects of precalving body condition score and prepartum feeding level on production, reproduction, and health parameters in pasture-based transition dairy cows

Precalving feeding level alters postcalving energy balance, dry matter intake, the liver and adipose tissue transcriptome, hepatic lipidosis, and the risk of metabolic diseases in both high-production cows consuming total mixed rations and moderate-production cows grazing pasture. We hypothesized that the reported benefits of a controlled restriction before calving are dependent on precalving body condition score (BCS): low BCS animals would not benefit from reduced feeding levels precalving, but high BCS cows would have metabolic and immunomodulatory profiles indicative of an improved health status. One hundred sixty-one days before calving, 150 cows were allocated randomly to 1 of 6 treatment groups (n=25) in a 2×3 factorial arrangement: 2 precalving BCS categories (4.0 and 5.0; based on a 10-point scale: BCS4 and BCS5, respectively) and 3 levels of energy intake during the 3 wk preceding calving (75, 100, and 125% of estimated requirements). Cows in the BCS4 and BCS5 groups were managed through late lactation to ensure that target calving BCS was achieved at dry off. Cows were then fed to maintain this BCS target until 3 wk before expected calving date, at which point they were managed within their allotted precalving energy intake treatments by offering different allowances of fresh pasture/cow per day. Milk production, body weight, and BCS were measured weekly; blood was sampled weekly before and after calving and on d 0, 1, 2, 3, and 4 relative to calving. Aspirated plasma was assayed for nonesterified fatty acids, β-hydroxybutyrate, total protein, albumin, cholesterol, haptoglobin, IL-1β, IL-6, total antioxidant capacity, and reactive oxygen species. Liver was sampled wk 1, 2, and 4 postcalving for triacylglycerol analysis. Results confirm that precalving BCS and precalving feeding level have both independent and interdependent effects on production and health characteristics of transition dairy cows. Irrespective of precalving BCS, a controlled restriction precalving reduced the net release of nonesterified fatty acids from adipose tissue postpartum and increased plasma calcium concentrations, reducing the risk of milk fever. Fatter cows produced more milk but lost more BCS postcalving and had greater blood β-hydroxybutyrate concentrations and increased hepatic lipidosis. In comparison, after calving, indicators of reduced immune competence were accentuated in BCS4 cows subjected to a feed restriction before calving, probably increasing the risk of infectious diseases. It would appear from these results that optimally conditioned cows will benefit from a short-term (2–3 wk) controlled feed restriction (75–90% of requirements), whereas cows in less than optimal condition should be fed to requirements before calving.

Ketonemia in dairy goats: Effect of dry period length and effect on lying behavior

In dairy animals, a successful transition from one lactation to the next includes minimizing negative energy balance. Cows experiencing excessive negative energy balance typically develop metabolic complications following parturition (e.g., ketosis); does are also susceptible before kidding (e.g., pregnancy toxemia). It is not known to what extent the provision and the length of the dry period affect these conditions in does. Furthermore, whereas clinical symptoms of these conditions include lethargy, behavioral changes resulting from ketosis and pregnancy toxemia have not been quantified in small ruminants. The aims of this study were to (1) describe the relationship between the dry period and negative energy balance, and (2) determine if lying behavior changes are indicative of the metabolic status of dairy goats. A total of 420 does on 10 commercial dairy goat farms in southern Ontario, Canada, were enrolled in the study (mean ± SD: 42±18 does/farm). Each doe was affixed with a data logger to measure lying behavior from 12 d before to 12 d after kidding. Blood samples were collected at least once before and at least once following kidding to determine blood β-hydroxybutyrate (BHBA) concentration as an indicator of negative energy balance. Does were categorized as healthy (HLTH; both pre- and postkidding samples BHBA <0.9 mmol/L), PREGTOX (prekidding BHBA ≥1.7 mmol/L), or KET (postkidding BHBA ≥1.7 mmol/L). Behaviors were analyzed according to 5 periods: P−2 (d −12 to d −2 relative to kidding), P−1 (d −1 relative to kidding), P0 (d 0, kidding day), P1 (d 1 relative to kidding), and P2 (d 2 to 12 relative to kidding). Dry period length and milk production after kidding were recorded when available. Farms ranged from 0 to 15% and 0 to 50% in prekidding and postkidding ketonemia, respectively. The HLTH does had shorter dry periods compared with PREGTOX and KET does (43 vs. 55 d, SE of the differences of means=4 d). One farm kept some does milking, while drying off others; on this farm more PREGTOX and KET does (11/28) were found in the dry group versus the continuously milked group (1/16). Overall, does that had ketonemia before kidding (PREGTOX) spent more time lying down compared with healthy does (16.1 vs. 12.7 h/d, SE of the differences of means=0.9). Both PREGTOX and HLTH does had increased lying bouts in P−2 compared with the day before (P−1) kidding [(mean (95% CI): 16.8 (15.8–17.8) vs. 20.5 (19.4–21.8) bout/d]. Compared with HLTH does, animals that were ketonemic following kidding (KET) had higher mean lying times throughout the study (14.5 vs. 13.5 h/d, SE=0.4). Previous work has shown a relationship between shorter and skipped dry periods and improved energy balance in dairy cows; the current study is the first to indicate a similar relationship in dairy goats. Furthermore, goats that developed ketonemia, particularly before kidding, displayed decreased activity, including longer lying times and fewer lying bouts in the days around kidding.

Factors associated with survival in the herd for dairy cows following surgery to correct left displaced abomasum

Left displaced abomasum (LDA) is a common problem in dairy cows. There have been numerous studies focused on predicting prognosis for right displaced abomasal corrective surgery, but a paucity of studies exist focused on more common LDA surgeries. Our objective was to determine if survival to 60d or 1yr after surgery could be predicted from the physical exam findings, periparturient disease status, and a biochemical profile from a blood sample obtained at the time of LDA diagnosis. Blood β-hydroxybutyrate (BHBA) concentrations were measured immediately using a hand-held meter. Data obtained from CanWest DHI (Guelph, ON, Canada) for all of the study subjects (n=179 cases, by 24 veterinarians from 4 clinics), including cull date, cull reason, and test-day milk production. Cows were classified based on whether or not they were culled within 60d or 1yr of surgery. Based on logistic regression, cows that had dystocia [odds ratio (OR)=13, 95% confidence interval (CI)=7–26] or were not ketotic (blood BHBA <1.2mmol/L; OR=3, 95% CI=1.03–9) at the time of corrective surgery were more likely to be culled within 60 d. Higher serum concentrations of BHBA (OR=0.95, 95% CI=0.92–0.98), nonesterified fatty acids (OR=0.81, 95% CI=0.75–0.88), and Mg (OR=0.49, 95% CI=0.35–0.68) all had a protective effect against culling within 1yr of LDA surgery. Based on survival analysis, longevity in the herd for 365d following corrective surgery was associated with higher BHBA and Mg at the time of LDA diagnosis before surgery, as well as milk production following surgery.

Effects of a single glucocorticoid injection on propylene glycol-treated cows with clinical ketosis

This study investigated the metabolic effects of glucocorticoids when administered to propylene glycol-treated cows with clinical ketosis. Clinical ketosis was defined by depressed feed intake and milk production, and a maximal score for acetoacetate in urine. All cows received 250 mL oral propylene glycol twice daily for 3 days and were randomly assigned to a single intramuscular injection with sterile isotonic saline solution (n = 14) or dexamethasone-21-isonicotinate (n = 17). Metabolic blood variables were monitored for 6 days and adipose tissue variables for 3 days. β-Hydroxybutyrate (BHBA) concentrations in blood decreased in all cows during treatment, but were lower in glucocorticoid-treated cows. Cows treated with glucocorticoids had higher plasma glucose and insulin concentrations, whereas concentrations of non-esterified fatty acids, 3-methylhistidine and growth hormone were unaffected. mRNA expression of hormone-sensitive lipase, BHBA receptor and peroxisome proliferator-activated receptor type γ in adipose tissue was not affected. This shows that lipolytic effects do not appear to be important in ketotic cows when glucocorticoids are combined with PG. Plasma 3-methyl histidine concentrations were similar in both groups, suggesting that glucocorticoids did not increase muscle breakdown and that the greater rise in plasma glucose in glucocorticoid-treated cows may not be due to increased supply of glucogenic amino acids from muscle.

Hyperketonemia in early lactation dairy cattle: A deterministic estimate of component and total cost per case

The purpose of this study was to develop a deterministic economic model to estimate the costs associated with (1) the component cost per case of hyperketonemia (HYK) and (2) the total cost per case of HYK when accounting for costs related to HYK-attributed diseases. Data from current literature was used to model the incidence and risks of HYK (defined as a blood β-hydroxybutyrate concentration ≥1.2mmol/L), displaced abomasa (DA), metritis, disease associations, milk production, culling, and reproductive outcomes. The component cost of HYK was estimated based on 1,000 calvings per year; the incidence of HYK in primiparous and multiparous animals; the percent of animals receiving clinical treatment; the direct costs of diagnostics, therapeutics, labor, and death loss; and the indirect costs of future milk production losses, future culling losses, and reproduction losses. Costs attributable to DA and metritis were estimated based on the incidence of each disease in the first 30 DIM; the number of cases of each disease attributable to HYK; the direct costs of diagnostics, therapeutics, discarded milk during treatment and the withdrawal period, veterinary service (DA only), and death loss; and the indirect costs of future milk production losses, future culling losses, and reproduction losses. The component cost per case of HYK was estimated at $134 and $111 for primiparous and multiparous animals, respectively; the average component cost per case of HYK was estimated to be $117. Thirty-four percent of the component cost of HYK was due to future reproductive losses, 26% to death loss, 26% to future milk production losses, 8% to future culling losses, 3% to therapeutics, 2% to labor, and 1% to diagnostics. The total cost per case of HYK was estimated at $375 and $256 for primiparous and multiparous animals, respectively; the average total cost per case of HYK was $289. Forty-one percent of the total cost of HYK was due to the component cost of HYK, 33% to costs attributable to metritis, and 26% to costs attributable to DA. The high total cost of HYK at reported incidences of 40 to 60% highlights the importance of appropriate transition cow nutrition and management to decrease the effect of HYK.

Trematode infections in pregnant ewes can predispose to mastitis during the subsequent lactation period

Objective was to investigate if trematode infections predispose ewes to mastitis and/or metritis. We used 80 trematode-infected ewes: primigravidae in group P-A and multigravidae in M-A remained untreated, primigravidae in P-B and multigravidae in M-B were drenched with netobimin and multigravidae in M-C were given rafoxanide. We collected faecal samples for parasitological examination, blood samples for β-hydroxybutyrate concentration measurement and uterine content, teat duct material and milk samples for bacteriological examination. We found significant differences in blood β-hydroxybutyrate concentrations between M-A, M-B and M-C during pregnancy (P⩽0.002). We did not observe significant differences between groups regarding development of metritis (P>0.83). We found that for M-A, M-B and M-C ewes, respectively, median time to first case of mastitis was 5.75, 21 and 6.75days after lambing (P=0.003) and incidence risk of mastitis was 0.308, 0.069 and 0.222 (P=0.047). We postulate that trematode infections predispose ewes to mastitis; perhaps, increased β-hydroxybutyrate blood concentrations adversely affect mammary cellular defences. This is the first report associating parasitic infections with mastitis in sheep.
