Association of Occupational and Leisure-Time Physical Activity With Allostatic Load

IntroductionLeisure-time physical activity (LTPA) decreases allostatic load (AL), a measure of burden of chronic stress. However, the role of occupational PA (OPA) is unknown. This study examined associations of OPA and LTPA with AL among workers in the United States (US). MethodsThis cross-sectional study included 6944 US workers aged 20–64 years from the National Health and Nutrition Examination Survey (2007–2018). PA was assessed using the Global Physical Activity Questionnaire. AL was calculated using biomarkers of cardiovascular health, metabolic function and immune response. Associations of OPA and LTPA were examined with AL using negative binomial regressions. Analyses were conducted between August 2022 and March 2023. ResultsVigorous LTPA inversely associated with AL among all workers (count ratio = 0.68, 95% CI: 0.62–0.76) and in each sex- and age- stratified group (i.e., females aged 20–44 years, females aged 45–64 years, males aged 20–44 years, and males aged 45–64 years), as well as in each race/ethnicity- stratified group (i.e., Whites, Blacks, Hispanics). Vigorous OPA positively associated with AL only among females aged 20–44 years [1.38 (1.10–1.73)]. Inverse associations of vigorous LTPA with AL were similar in young females with high [0.61 (0.50–0.75)] or low [0.66 (0.60–0.74)] vigorous OPA. ConclusionsIncreasing vigorous LTPA is associated with a lower AL for all workers, while increasing vigorous OPA is associated with a higher AL only in young females. Promoting vigorous LTPA reduces AL among young females with either low or high vigorous OPA.

Abstract P307: Differences in Cardiovascular Disease Burden, Screening, Education, and Care by Clinic Type in the 2022 Health Center Patient Survey (HRSA)

Background: Persons experiencing homelessness (PEH) have a 2-3-fold risk for cardiovascular disease (CVD) mortality. Evidence has repeatedly shown elevated chronic disease burden, reduced access to care, and lower utilization of medication to control CVD risk factors in this setting. Migrant communities also experience reduced access to care and the impacts from deferring care. There are targeted, federally-funded health clinics in place for both settings. However, these systems are frequently overwhelmed and limited by their scope of care to primary care. There is scarce evidence on differences between access, quality, and experiences of care delivered relative to other safety-net models. Method: The 2022 Health Center Patient Survey (HCPS) was collected on behalf of the Health Resources and Services Administration (HRSA). The HCPS is a nationally-representative, three-staged, sample-based survey collected via 1:1 interviews with clinic patients. The survey assessed sociodemographics, health conditions and behaviors, access to and utilization of care, and patients’ experiences with comprehensive services they received at HRSA-funded health centers, including: community health centers (CHC), healthcare for the homeless (HCH) clinics, migrant health centers (MHC), and public housing primary care (PHPC) clinics. A total of 103 unique awardees and 318 health center sites were recruited, and 4,414 patient interviews were completed. Investigators analyzed patient characteristics and multiple survey items related to AHA’s Essential 8 metrics for differences across clinic type. Results: MHC and HCH clinics had fewer elderly patients (~7%) than CHC and PHPC clinics (~17%). There were differences across sex, race, and ethnicity across all four clinic types. Cardiovascular disease history ranged from 4.9% (MHC) to 14.7% (HCH). Reported 7-day physical activity measures, average sleep below 7 hours per day, and Lifetime smoking (>100 cigarettes; OR=4.2, p<0.001) were all greatest among HCH patients. Fewer HCH and MHC patients reported ever having or recent lipid tests (both p<0.001). HCH patients were more likely to report hypertension (p=0.003), but less likely to report receiving nutrition advice (all p<0.05). MHC patients were less likely to be prescribed hypertension medication (p=0.044) and HCH patients were less likely to be taking medication even if it was prescribed (p<0.001). Adjustment for differences in age or CVD history were able to explain some observed differences, but increased the magnitude of other disparities. Conclusion: CVD burden differs across the various HRSA funding mechanisms for clinics, as do demographics and multiple metrics of health behaviors and biomarkers of cardiovascular health. Greater disease burden in HCH and MHC patients is likely compounded by increased risk factors and under performance in providing health education interventions.

Comparative assessment of erythrocyte sphingolipid levels as potential cardiovascular health markers in women from Libya and Serbia: a small-scale study

ABSTRACT Aim: Cardiovascular diseases (CVDs) represent the major cause of morbidity and mortality worldwide including Libya, where they account for 43% of all deaths. Sphingolipids are involved in the pathology of numerous diseases including cardiovascular diseases and are proposed as potential biomarkers of cardiovascular health that could be more effective compared to traditional clinical biomarkers. The aim of this study was to determine the sphingolipid content in the erythrocyte membrane of Libyan migrant and Serbian resident women. In addition, to examine if sphingolipid levels could be used as a novel indicator of cardiovascular risk, we evaluated possible correlations with some well-established biomarkers of cardiovascular health. Materials and Methods: A total of 13 Libyan and 15 Serbian healthy women participated in the study. The high-performance version thin-layer chromatography (HPTLC) using the image analysis tool JustTLC was applied for quantification of erythrocytes’ sphingolipids. Results: Lower mean values of erythrocytes’ sphingolipids and cholesterol concentrations were found in the group of Libyan emigrants compared to Serbian resident women. Besides, in this group of apparently healthy women (n = 28), the sphingolipid content of erythrocytes was inversely related to the Omega-3 index (r =-0.492, p = 0.008) and directly linked to vitamin D status (r = 0.433, p = 0.021) and membrane cholesterol levels (r = 0.474, p = 0.011). Conclusion: The erythrocytes’ sphingolipid levels should be measured/assessed as an additional biomarker of CV health, by applying a simple and routine method. Still, further investigation in a larger population-specific context is warranted.

Effects of long-term consumption of two plant-based dietary supplements on cardiovascular health and low-grade inflammation in middle-aged and elderly people: study protocol for a randomised, controlled trial

BackgroundAgeing is a process characterised by chronic low-grade inflammation and oxidative stress which could lead to increased prevalence of both physical and mental age-related chronic conditions. A healthy balanced diet, rich in fruit and vegetables as well as omega-3 polyunsaturated fatty acids (n3 PUFA), could reduce oxidative stress and improve markers of low-grade inflammation. Nonetheless, considering that a large part of the population struggles to meet current guidelines on fruit and vegetable and n3 PUFA recommendations, fruit and vegetable concentrate supplements and mixed omega fatty acid supplements could be an effective strategy to bridge the gap between actual and recommended intakes.MethodsIn this randomised, controlled, open-labelled, parallel-grouped clinical trial, 112 participants will be allocated to one of four arms (n = 28 on each arm): an encapsulated juice powder concentrate, a plant-based omega fatty acid supplement, both or a control group. We aim to investigate whether long-term separate or combined ingestion of the two can affect biomarkers of cardiovascular health, low-grade inflammation and indicators of ageing, including cognitive function, in middle-aged and elderly people. We will additionally explore the effect of the different supplementations on plasma levels of vitamins, carotenoids and fatty acids. Intervention will last 2 years and participants will be assessed at baseline and at follow-up visits at 6, 12, 18 and 24 months.DiscussionThis study will provide evidence whether long-term, plant-based dietary supplementation can support cardiovascular health, anti-inflammatory processes, immunity and nutritional status in ageing.Trial registration This trial was registered at ClinicalTrials.gov (NCT04763291) on February 21, 2021.

Testicular function is associated with cardiometabolic health markers: A cross-sectional study of 2289 young men.

Testicular function, including compensated Leydig cell function, has been indicated to be an early marker of morbidity. To study the association of testicular function and markers of metabolic and cardiovascular health in a population of young men. A cross-sectional study of 2289 men (median age 19 years, 5-95 percentile 18.4-22.2) from the general population examined between 2012 and 2019. Participants answered a questionnaire, had a blood sample drawn for assessment of reproductive hormone levels and health markers (lipids, glycosylated hemoglobin), delivered a semen sample, underwent physical examination including blood pressure measurements, and dual-energy X-ray absorptiometry scan for assessment of body composition. Associations were assessed in both crude and adjusted linear regression analyses. The men were within the normal reference intervals of their age for reproductive and health biomarkers. Compared to the lowest quartile, having luteinizing hormone levels in the highest quartile was associated with higher mean arterial pressure (1.6 [95% confidence interval: 0.8; 2.5] mmHg), cholesterol (0.1 [95% confidence interval: 0.02; 0.18] mmol/L), and total body fat percentage (1.1 [95% confidence interval: 0.4; 1.8] %-points). Higher serum testosterone levels were associated with more advantageous cardiometabolic health markers and higher total sperm count with a healthier body composition and lower glycosylated hemoglobin. In this study of young men, unselected regarding reproductive hormones and semen quality, higher luteinizing hormone was associated with cardiovascular risk factors. Higher testosterone and total sperm count were associated with more favorable cardiometabolic indices. Thus, serum reproductive hormones and semen quality may be early appearing biomarkers of cardiovascular health even among young healthy men, which could potentially be useful for preventive initiatives to reduce the excess mortality and morbidity risk among infertile men. However, our study was cross-sectional and cannot determine causation. Future longitudinal studies of reproductive health in young men are warranted.

Ambivalence is associated with decreased physical activity and cardiorespiratory fitness among adolescents with critical congenital heart disease

BackgroundAdolescents with congenital heart disease (CHD) are insufficiently physically active. Given that increasing physical activity may reduce their cardiovascular risk, it is important to identify correlates of this behavior. Perceived benefits of and barriers to physical activity are associated with physical activity engagement. Existing research has only considered these constructs separately. This population may be ambivalent toward physical activity (i.e., perceive both strong benefits and barriers). The association of ambivalence and physical activity related outcomes is unknown among this at-risk population. ObjectiveDetermine the association of ambivalence and sedentary behavior, moderate-to-vigorous physical activity (MVPA), and cardiorespiratory fitness (VO2Peak) among adolescents with CHD. MethodsThe present study is an analysis of data from an eligibility assessment for a randomized clinical trial of an intervention to promote MVPA among adolescents aged 15 to 18 years with moderate or complex CHD. Participants (N = 84) completed a survey assessing perceived benefits and barriers from which ambivalence toward physical activity was calculated, an exercise stress test to measure VO2Peak, and wore an accelerometer for one week to determine their engagement in sedentary behavior and MVPA. Linear regression analyses determined associations between ambivalence and physical activity related outcomes. ResultsGreater ambivalence toward physical activity was associated with increased sedentary behavior, decreased MVPA, and reduced VO2Peak, adjusting for demographic and clinical covariates. ConclusionsAmbivalence is associated with objectively measured physical activity (sedentary behavior, MVPA) and a biomarker of cardiovascular health (VO2Peak). Screening for ambivalence may help clinicians identify those most likely to benefit from physical activity-related education.

PHYSICAL ACTIVITY TRENDS IN A NATIONWIDE MHEALTH PROGRAM: A POPULATION-BASED COHORT STUDY

Abstract Physical inactivity is a global public health challenge, leading to an increase in chronic diseases. Effective large-scale physical activity interventions are needed. We examined the effectiveness of a nationwide mHealth intervention in Singapore, National Steps Challenge Season 3 (NSC3). NSC3 included gain-framed financial incentives for reaching pre-defined daily step targets, nudging via reminders and real-time feedback on physical activity levels. Our study includes 411,528 participants, with 53,371 (13.0%) participants aged 59 and above. Regression discontinuity design examined changes to daily step counts prior to and during NSC3. NSC3 was associated with an overall mean increase of 1437 steps per day (95% CI: 1408 to 1467). Participants in older age groups were associated with larger mean step increases. Females aged between 59 to 68 and 69 and above were associated with average increases of 1640 steps per day (95% CI: 1510 to 1770) and 2300 steps per day (95% CI: 2050 to 2550), respectively. Males in the same age groups were associated with average increases of 1360 steps per day (95% CI: 1200 to 1520) and 1640 steps per day (95% CI: 1370 to 1900), respectively. We provide real-world evidence that suggests NSC3 improved participants’ step counts, with a larger increase among older adults. As older adults have a higher risk of chronic diseases, lower physical inactivity has potential health benefits such as reducing the incidence of hypertension and improving cardiovascular health biomarkers. While results are promising, further investigations are necessary to ensure sustained engagement among older adults.

Effect of short-term exposures to traffic air pollution and temperature on heart rate among healthy female students during commuting

Findings showed that studies on the interaction effect of air pollution and temperature on biomarkers of cardiovascular health in a micro-environment (MiE) are still very limited. Therefore, this study aims to determine the interaction effect between air pollution, temperature, and biomarkers of cardiovascular outcomes, including Heart Rate/HR, Systolic Blood Pressure/SBP, and Diastolic Blood Pressure/DBP among healthy female students during commuting in a MiE, namely a public transport. A ParticleScan™ CR device (IQAir Group, Inc., Swiss) was used to measure Particulate Matter (PM). Temperature, Relative Humidity (RH), Nitrogen Dioxide (NO2), Nitric Oxide (NO), Carbon Monoxide (CO), and Total Volatile Organic Compounds (TVOC) were measured using a YES Plus LGA multi-sensor device (Critical Environment Technologies Canada, Inc., Canada). The heart rates of the samples were assessed using an electronic blood pressure monitor Mediatech (Shanzen ZhengKang Technology Co., Ltd., China), while the vital lung capacity was obtained with a portable spirometer MicroLab (Micro Direct, Inc., USA). The data obtained in this study were analyzed using multiple linear regression. Furthermore, normality tests were carried out for all dependent variables, namely HR, SBP, and DBP using the Kolmogorov-Smirnov test and the result shows normal distribution. The beta coefficients (β) were used to express the association between the data. For the single effects, PM ≥ 0.3, PM ≥ 0.5, PM ≥ 0.7, PM ≥ 1.0, PM ≥ 2.0, PM0.3-0.5, PM0.5-0.7, PM0.7-1.0, PM1.0-2.0, and PM2.0-5.0 NO2, and TVOC had a significant negative effect on HR; NO affected SBP and DBP negatively; and NO2 had a positive influence on DBP. For the interaction effects, PM ≥ 0.3, PM ≥ 0.5, PM ≥ 0.7, PM0.3-.05, PM0.5-0.7, and PM0.7-1.0 had a significant positive interaction effect with temperature on HR.

Increase in colonic PRopionate as a method of prEVENTing weight gain in adults aged 20–40 years (iPREVENT): protocol of a multi-centre, double-blind, randomised, parallel-group trial to investigate the efficacy of inulin-propionate ester versus inulin (control) in the prevention of weight gain over 12 months

Introduction: Overweight and obesity affects over 70% of the UK population and is a major risk factor for the development of co-morbidities, including type 2 diabetes and cardiovascular disease. There now exists a considerable evidence base for the management of obesity. However, this is not the case for the prevention of obesity. Preventing weight gain in periods of life where there is an elevated risk of fat mass expansion could be beneficial to preventing associated diseases in later life. This protocol investigates the impact of novel food ingredient inulin propionate ester (IPE) in the prevention of weight gain. This trial aims to investigate the primary hypothesis that IPE has a superior effect on preventing body weight gain, compared with inulin, in young (<40 years old) adults over 12 months, whilst also investigating several complementary mechanisms that may explain the prevention of weight gain and improved long-term energy balance from consuming IPE. Methods: In this multi-centre, double-blind, randomised, parallel-group study, eligible participants will be randomly assigned to consume 10g IPE or 10g inulin (control) daily for 12 months. Study visits will be conducted at baseline, two-month, six-month and 12-month time points. The primary outcome is weight gain from baseline to 12 months. Secondary outcomes will examine changes in metabolic and cardiovascular health biomarkers, body composition and appetite. A mechanistic sub-group will explore causal mechanisms around energy balance, body composition, appetite regulation and the gut microbiota. Based on the power calculation, the sample size required is 270 participants or 135 per study group. Ethics and dissemination: The trial protocol and participant-facing documents have been reviewed and approved, by the London Hampstead Ethics Committee (REC Reference 19/LO/0095, 29th January 2019). Upon completion, the trial results will be published in peer-reviewed journals and presented at scientific conferences. Trial registration number: ISRCTN16299902, 1st March 2018.

A Portable Sweat Sensor Based on Carbon Quantum Dots for Multiplex Detection of Cardiovascular Health Biomarkers.

The future of personalized diagnostics and treatment of cardiovascular diseases lies in the use of portable sensors. Portable sensors can acquire biomarker information in biological fluids such as sweat, an approach that mitigates the shortcomings of conventional hospital-centered healthcare. Low sensitivity, selectivity, and specificity remain bottlenecks for the widespread use of portable sensors. Herein, we demonstrate a portable sensor that simultaneously detects Na+, ascorbic acid, and human neuropeptide Y in sweat, all useful biomarkers to index cardiovascular health. The portable sensor comprises a six-electrode system containing three working electrodes, two reference electrodes, and one counter electrode. The working electrodes were prepared by depositing sensing components on carbon quantum dot (CQD) electrodes. The sensing mechanisms were based on selective ion recognition, enzyme catalytic reaction, and immune response, which guarantees specificity to corresponding targets. The CQDs offer massive reactive sites and effectively reduce the interfacial impedance during the sensing reaction, thereby enhancing the three biomarkers' detection sensitivity. As evidence of portable sensor capability, we demonstrate herein its effective simultaneous detection of the three biomarkers in a real sweat from healthy volunteers during routine activities including exercise, extra ascorbic acid ingestion, and extra Na+ ingestion. As such, the sensor shows promise for real-time noninvasive personalized medical diagnostics and metabolic wellness management.

Can low-carbohydrate diets be recommended for reducing cardiovascular risk?

This review provides a rationale for implementing carbohydrate restriction as a dietary therapy to improve biomarkers of cardiovascular health and suggests that this will require a paradigm shift away from what is currently promulgated as a 'heart-healthy' diet. Type 2 diabetes mellitus (T2DM), metabolic syndrome, and related co-morbidities are major risk factors for cardiovascular disease (CVD). Ideally, then, a diet intended to support cardiovascular health should be one that improves or reverses these underlying risk factors. Carbohydrate restriction is effective for this purpose as well as for favorably impacting atherogenic dyslipidemia. Recent consensus reports from select national organizations have endorsed low-carbohydrate diets for improving glycemia and cardiovascular risk. Reluctance among public health organizations and some clinicians to more widely promote this therapeutic nutritional approach is driven primarily by the increase in serum low-density lipoprotein cholesterol (LDL-C) observed in a proportion of individuals who adopt a low-carbohydrate diet. Here we explore the rationale for using carbohydrate restriction to improve cardiovascular health by way of favorably impacting T2DM and insulin resistance, and why this salutary effect outweighs the potential adverse effects of an increase in serum LDL-C. Carbohydrate restriction is a logical foundation for a dietary intervention intended to reduce CVD risk, particularly among individuals with T2DM or metabolic syndrome.

Local Arterial Stiffness Assessment: Comparison of Pulse Wave Velocity Assessed by Ultrafast Ultrasound Imaging versus the Bramwell-Hill Equation

Arterial stiffness is a major biomarker of cardiovascular health and a key predictor of cardiovascular events.1 In the clinical setting, pulse wave velocity (PWV) is used as a noninvasive surrogate for arterial stiffness. Local PWV represents the stiffness of a small segment of a given vessel. This is an important assessment for monitoring the progression of local and heterogenous disease such as atherosclerosis, connective tissue disorders, and the iatrogenic impacts of radiation. Currently, the most common tool for measuring local PWV is the theoretical Bramwell-Hill (B-H) equation.

Relaxin-2 as a Potential Biomarker in Cardiovascular Diseases.

The pleiotropic hormone relaxin-2 plays a pivotal role in the physiology and pathology of the cardiovascular system. Relaxin-2 exerts relevant regulatory functions in cardiovascular tissues through the specific receptor relaxin family peptide receptor 1 (RXFP1) in the regulation of cardiac metabolism; the induction of vasodilatation; the reversion of fibrosis and hypertrophy; the reduction of inflammation, oxidative stress, and apoptosis; and the stimulation of angiogenesis, with inotropic and chronotropic effects as well. Recent preclinical and clinical outcomes have encouraged the potential use of relaxin-2 (or its recombinant form, known as serelaxin) as a therapeutic strategy during cardiac injury and/or in patients suffering from different cardiovascular disarrangements, especially heart failure. Furthermore, relaxin-2 has been proposed as a promising biomarker of cardiovascular health and disease. In this review, we emphasize the relevance of the endogenous hormone relaxin-2 as a useful diagnostic biomarker in different backgrounds of cardiovascular pathology, such as heart failure, atrial fibrillation, myocardial infarction, ischemic heart disease, aortic valve disease, hypertension, and atherosclerosis, which could be relevant in daily clinical practice and could contribute to comprehending the specific role of relaxin-2 in cardiovascular diseases.

Flow-mediated dilation reference values for evaluation of endothelial function and cardiovascular health.

Endothelial function is essential for cardiovascular health, and flow-mediated dilation (FMD) is an established technique to measure it. This paper aims to assess FMD values in apparently healthy individuals and provides reference values to facilitate wider clinical use. In 1,579 apparently healthy individuals (aged 18-76), fasted FMD values (data from 44 studies, 6 institutions, 22 operators) were normally distributed and inversely univariately correlated with age, body mass index, glucose, cholesterol, blood pressure, and brachial artery (BA) diameter. Significant multivariate predictors of FMD were age (-0.4%/decade), BMI (0.04%/kg/m2), smoking (-0.7%), and BA diameter (-0.44%/mm) that together explained 19% of the variability independent of operator, institution or ultrasound machine. Individuals in the high FMD tertile (>6.8%) were younger, had smaller BA diameter, lower blood pressure and cholesterol. In individuals with low- and intermediate fatal cardiovascular risk (SCORE), 26% and 53% of individuals, respectively, had FMD values in the low tertile (<5.4%). After adding data from 385 patients with stable coronary artery disease (CAD), ROC analysis (c = 0.841, P < 0.001) showed that FMD of >6.5% excluded CAD (95% sensitivity; 60% specificity) and FMD <3.1% excluded 95% healthy individuals (95% specificity, 31% sensitivity). A meta-analysis and meta-regression of 82 clinical trials (11 countries, n = 3,509) using similar FMD methodology showed that despite considerable heterogeneity (I2 = 0.97) FMD in healthy individuals was on average 6.4% (95%CI: 6.2%, 6.7%) with no significant differences between countries but a significant age-dependent decline (-0.3%/decade, R2 = 0.13). We provide an age-adapted frame of FMD reference intervals in apparently healthy individuals for use as a biomarker of cardiovascular health. As the degree of vascular endothelial function integrates environmental and genetic factors with classical CV risk factors, FMD may more comprehensively classify individuals with and without standard modifiable cardiovascular risk factors and serve as a target for cardiovascular prevention.

The effects of a structured-exercise training program on erectile function, cardiovascular and respiratory parameters in men following prostate cancer treatment: Preliminary results from a randomised, controlled clinical trial in prostate cancer survivorship

ABSTRACT Introduction Prostate cancer (PC) treatments can cause significant cardiovascular and sexual dysfunctions. There is limited data on the effects of structured exercise interventions specifically on the improvements of erectile function (EF) in the context of cardiovascular health and PC survivorship. It is postulated that structured physical exercise programs can result in positive changes in biomarkers of cardiovascular health through cardiovascular remodelling and in turns, will improve penile haemodynamic and EF recovery in PC men with erectile dysfunction (ED). Objective To explore the efficacy of structured-exercise training on EF, cardiorespiratory fitness (V̇O2peak), exercise capacity (peak power output; PPO) and vascular health (pulse wave velocity; PWV) in men with ED in the context of PC survivorship. Methods Following ethics approval, men who underwent radical prostatectomy and reported mild-to-moderate ED (defined as International Index of Erectile Function (IIEF-5) score of 8 to 21) were recruited and randomised to complete a thrice-weekly 12-week of structured aerobic and resistance high-intensity interval training (HIIT) intervention, or usual care. Changes in EF (based on IIEF-5 scores), V̇O2peak, PPO and PWV were assessed at baseline and 12 weeks follow-up. Results Of the 33 patients referred to the study, seven were ineligible due to severe (n=5) or normal (n=2) erectile function, with 12 agreeing to participate and blindly randomise into structured HIIT and usual care groups. Attendance to the 144 scheduled exercise sessions was 100%, with no study-related adverse events. At baseline, 75% of participants had a low V̇O2peak compared to normative data. After 12 weeks, EF (mean improvement in IIEF-5 by 5 points), V̇O2peak (INT=+3.7 mL/kg/min, CON=+0.4 mL/kg/min), absolute PPO (INT=+35 W, CON=+3 W) and PWV (INT=-3.16 m/s, CON=-0.95 m/s) were greater in the intervention group compared with the usual care group. Conclusions A 12-week structured HIIT exercise program is feasible, safe and has been shown to improve EF, cardiorespiratory fitness, exercise capacity and vascular health in men with PC. Zinc as a Biomarker of Cardiovascular Health.

The importance of zinc (Zn) for cardiovascular health continuously gains recognition. As shown earlier, compromised Zn homeostasis and prolonged inflammation are common features in various cardiovascular diseases (CVDs). Similarly, Zn biochemistry alters several vascular processes, and Zn status is an important feature of cardiovascular health. Zn deficiency contributes to the development of CVDs; thus, Zn manipulations, including Zn supplementation, are beneficial for preventing and treating numerous cardiovascular (CV) disorders. Finally, additional long-term, well-designed studies, performed in various population groups, should be pursued to further clarify significant relationships between Zn and CVDs.

Effects of Two Plant-Based Dietary Supplements on Cardiovascular Health and Low-Grade Inflammation in the Elderly: Study Protocol for a Randomised Controlled Trial

Abstract Objectives To assess whether the long-term separate ingestions of an encapsulated juice powder concentrate and a plant-based omega fatty acid supplement, or a combined ingestion of the two, can affect biomarkers of cardiovascular health, low-grade inflammation and indicators of aging in the elderly. Methods This is a randomised, controlled, open-labelled, parallel-grouped clinical trial, consisting of 4 arms. One hundred and twelve, overweight and obese (BMI 25–35 kg/m2) subjects, aged 60 to 80 years will be recruited and randomly allocated to: i) control group; ii) Fruit, Vegetable and Berry (FVB) group, where participants have to ingest a fruit, vegetable and berry supplement (Juice Plus+® capsules); iii) Omega group, where participants have to ingest a plant-based fatty acid supplement (Juice Plus+® Omega Blend); and iv) Fruit, Vegetable, Berry and Omega (FVBO) group, where participants have to ingest the fruit, vegetable and berry supplement, together with the plant-based fatty acid supplement. Participants in all groups will continue their habitual diet and lifestyle. Those allocated to one of the three intervention groups, will be asked to ingest the supplements for a total duration of two years. Participants will be assessed at baseline and at follow-up visits at 6, 12, 18 and 24 months. Primary outcomes include markers of cardiovascular health and low-grade inflammation. Secondary outcomes include selected indicators of aging, cognitive function, immunity and quality of life. Finally, plasma levels of vitamins, carotenoids and fatty acids will also be assessed. Results N/A Conclusions This study will provide evidence whether long-term, plant-based dietary supplementation can support cardiovascular health, anti-inflammatory processes, immunity and nutritional status in aging. Effects of dim artificial light at night on locomotor activity, cardiovascular physiology, and circadian clock genes in a diurnal songbird

Artificial light is transforming the nighttime environment and quickly becoming one of the most pervasive pollutants on earth. Across taxa, light entrains endogenous circadian clocks that function to synchronize behavioral and physiological rhythms with natural photoperiod. Artificial light at night (ALAN) disrupts these photoperiodic cues and has consequences for humans and wildlife including sleep disruption, physiological stress and increased risk of cardiovascular disease. However, the mechanisms underlying organismal responses to dim ALAN, resembling light pollution, remain elusive. Light pollution exists in the environment at lower levels (<5 lux) than tested in many laboratory studies that link ALAN to circadian rhythm disruption. Few studies have linked dim ALAN to both the upstream regulators of circadian rhythms and downstream behavioral and physiological consequences. We exposed zebra finches (Taeniopygia gutatta) to dim ALAN (1.5 lux) and measured circadian expression of five pacemaker genes in central and peripheral tissues, plasma melatonin, locomotor activity, and biomarkers of cardiovascular health. ALAN caused an increase in nighttime activity and, for males, cardiac hypertrophy. Moreover, downstream effects were detectable after just short duration exposure (10 days) and at dim levels that mimic the intensity of environmental light pollution. However, ALAN did not affect circulating melatonin nor oscillations of circadian gene expression in the central clock (brain) or liver. These findings suggest that dim ALAN can alter behavior and physiology without strong shifts in the rhythmic expression of molecular circadian pacemakers. Approaches that focus on ecologically-relevant ALAN and link complex biological pathways are necessary to understand the mechanisms underlying vertebrate responses to light pollution.

Common Genetic Variations Involved in the Inter-Individual Variability of Circulating Cholesterol Concentrations in Response to Diets: A Narrative Review of Recent Evidence.

The number of nutrigenetic studies dedicated to the identification of single nucleotide polymorphisms (SNPs) modulating blood lipid profiles in response to dietary interventions has increased considerably over the last decade. However, the robustness of the evidence-based science supporting the area remains to be evaluated. The objective of this review was to present recent findings concerning the effects of interactions between SNPs in genes involved in cholesterol metabolism and transport, and dietary intakes or interventions on circulating cholesterol concentrations, which are causally involved in cardiovascular diseases and established biomarkers of cardiovascular health. We identified recent studies (2014–2020) that reported significant SNP–diet interactions in 14 cholesterol-related genes (NPC1L1, ABCA1, ABCG5, ABCG8, APOA1, APOA2, APOA5, APOB, APOE, CETP, CYP7A1, DHCR7, LPL, and LIPC), and which replicated associations observed in previous studies. Some studies have also shown that combinations of SNPs could explain a higher proportion of variability in response to dietary interventions. Although some findings still need replication, including in larger and more diverse study populations, there is good evidence that some SNPs are consistently associated with differing circulating cholesterol concentrations in response to dietary interventions. These results could help clinicians provide patients with more personalized dietary recommendations, in order to lower their risk for cardiovascular disease.
