Extracting Venom from the Parasitoid Wasp Trichogramma dendrolimi using an Artificial Host.

Parasitoid wasps are a diverse group of hymenopteran insects that serve as invaluable resources for pest biocontrol. To ensure successful parasitism, parasitoid wasps inject venom into their hosts to suppress their hosts' immunity, modulate hosts' development, metabolism, and even behavior. With over 600,000 estimated species, the diversity of parasitoid wasps surpasses that of other venomous animals, such as snakes, cone snails, and spiders. Parasitoid wasp venom is an underexplored source of bioactive molecules with potential applications in pest control and medicine. However, collecting parasitoid venom is challenging due to the inability to use direct or electrical stimulation and the difficulty in dissection because of their small size. Trichogramma is a genus of tiny (~0.5 mm) egg parasitoid wasps that are widely used for the biological control of lepidopteran pests in both agriculture and forests. Here, we report a method for extracting venom from T. dendrolimi using artificial hosts. These artificial hosts are created with polyethylene film and amino acid solutions and then inoculated with Trichogramma wasps for parasitism. The venom was subsequently collected and concentrated. This method enables the extraction of large amounts of Trichogramma venom while avoiding contamination from other tissues caused by dissection, a common issue in venom reservoir dissection protocols. This innovative approach facilitates the study of Trichogramma venom, paving the way for new research and potential applications.

Variation among populations of Trichogramma euproctidis (Hymenoptera: Trichogrammatidae) revealed by life table parameters: perspectives for biological control.

The successful mass-rearing of potential biological control agents is a prerequisite for sustainable pest control. In this study, the performance of 3 Trichogramma euproctidis (Girault) (Hymenoptera: Trichogrammatidae) populations collected from different locations in Khuzestan (Southwest Iran) were evaluated to optimize the egg parasitoid mass-rearing for augmentative biological control of lepidopteran pests. We aimed to investigate the effects of both population origin and host quality on biological traits of ovipositing females (number of parasitized eggs) and of their progeny (development time, survival rate, sex ratio, longevity, and fecundity). The effect of host quality was assessed by allowing the parasitoid to oviposit into 1, 2, 3, or 4-day-old Ephestia kuehniella Zeller (Lepidoptera: Pyralidae) eggs. The 3 T. euproctidis populations developed successfully regardless the age of the host eggs. However, we found significant variation among populations and a strong influence of host quality on the traits investigated. Progeny performance in all populations decreased with increasing host age. The best-performing population (collected in Mollasani) showed the highest parasitization rate, highest survival rate, and progeny sex ratio with the greatest percentage of females. A life table corroborated these findings with superior estimates of the net reproductive rate (R0), intrinsic rate of increase (r), and reduced generation time (T) for the Mollasani population on 1-day-old host eggs. We conclude that ample variation exists among T. euproctidis populations and that rearing the Mollasani population on young rather than old eggs of E. kuehniella would be recommended to implement the biological control programs to target lepidopteran pests in Southwestern Iran.

Map-based cloning and functional analysis revealed ABCC2 is responsible for Cry1Ac toxin resistance in Bombyx mori.

Bt toxins are parasporal crystals produced by Bacillus thuringiensis (Bt).They have specific killing activity against various insects and have been widely used to control agricultural pests. However, their widespread use has developed the resistance of many target insects. To maintain the sustainable use of Bt products, the resistance mechanism of insects to Bt toxins must be fully clarified. In this study, Bt-resistant and Bt-susceptible silkworm strains were used to construct genetic populations, and the genetic pattern of silkworm resistance to Cry1Ac toxin was determined. Sequence-tagged sitemolecular marker technology was used to finely map the resistance gene and to draw a molecular genetic linkage map, and the two closest markers were T1590 and T1581, indicating the resistance gene located in the 155 kb genetic region. After analyzing the sequence of the predicted gene in the genetic region, an ATP binding cassette transporter (ABCC2) was identified as the candidate gene. Molecular modeling and protein-protein docking result showed that a tyrosine insertion in the mutant ABCC2 might be responsible for the interaction between Cry1Ac and ABCC2. Moreover, CRISPR/Cas9-mediated genome editing technology was used to knockout ABCC2 gene. The homozygous mutant ABCC2 silkworm was resistant to Cry1Ac toxin, which indicated ABCC2 is the key gene that controls silkworm resistance to Cry1Ac toxin. The results have laid the foundation for elucidating the molecular resistance mechanism of silkworms to Cry1Ac toxin and could provide a theoretical basis for the biological control of lepidopteran pests.

Prepupae and pupae transcriptomic characterization of Trichogramma chilonis

The egg parasitoid, Trichogramma chilonis, has significant control effects on agriculture and forestry pests and is widely employed in southern China for the biological control of lepidopteran pests. In this study, transcriptomic analysis was used to gain a clear understanding of the molecular changes in prepupae and pupae of T. chilonis. A total of 16.88 Gb of clean data were obtained and finally assembled into 43,136 unigenes, 18,880 of which were annotated. After FPKM standardization, 117 and 838 specific expression genes were found in prepupae and pupae, respectively. There were 3129 differentially expressed genes between prepupae and pupae. Compared to pupae, 806 genes were up-regulated and 2323 were down-regulated in prepupae. Background on the T. chilonis transcriptome, the enriched GO function and KEGG pathway analysis of DEGs were considered. As indicated by GO classification, up-regulated genes were mainly involved in chitin metabolism, cell adhesion and endocytic, while most down-regulated genes were involved in synthesis of cell components, ion transport and biological regulation. KEGG enrichment analysis showed that 458 DEGs were enriched in 94 metabolic pathways. DEGs involved in nucleotide replication and transcription, substance metabolism, insect hormone biosynthesis, cell growth and death, reproductive metabolism, circadian rhythms and signal transduction pathways were up-regulated or down-regulated to different degrees, indicating that these genes played important roles during the process of metamorphosis in T. chilonis. This study provides a rich data source for the future study of T. chilonis molecular and biological mechanisms. A large number of genes related to metamorphosis were found based on comparison analysis between prepupae and pupae transcriptomes. This study lays a good foundation for in-depth study of gene transcription and regulation mechanisms during T. chilonis metamorphosis.

Effects of cold storage on the quality of Trichogramma dendrolimi Matsumura (Hymenoptera: Trichogrammatidae) reared on artificial medium.

Trichogramma dendrolimi Matsumura is an important egg parasitoid that can be used in the biological control of lepidopteran pests. The effects of storage temperature, developmental stage and storage duration on development and survival were investigated to determine the optimum storage conditions for rearing T. dendrolimi in vitro. The three investigated factors significantly impacted all the biological parameters except for parental G1 and offspring G2 sex ratios. The responses of in vitro reared T. dendrolimi to low temperatures differed depending on developmental stage and storage duration. Comparison of life table parameters under different storage treatments with the control group indicated that storing prepupae at 13°C for 2, 3 or 4weeks positively impacted the growth capacity of the parasitoid. In vitro reared T. dendrolimi can be reared following cold storage. The quality of the parasitoid was strongly affected by cold storage, likely due to the negative effects of storage on the artificial medium. Storage of the prepupal stage at 13°C for 4weeks is optimum for in vitro reared T. dendrolimi. Trichogramma reared in vitro were able to parasitize natural eggs after storage and, thus, have potential for use as biological control agents in the field. © 2018 Society of Chemical Industry.

Pupal parasitoids for the biological control of Diatraea saccharalis (Fabricius, 1794) (Lepidoptera: Crambidae)

Diatraea saccharalis (Fabricius, 1794) (Lepidoptera: Crambidae), popularmente conhecida como broca da cana-de-açúcar, é a maior praga da cultura de cana de açúcar e é responsável por consideráveis danos econômicos. O manejo dessa praga é complexo devido ao alojamento no interior do colmo; quando imatura, cria galerias que enfraquecem as plantas, reduz a biomassa e a qualidade do suco da cana, e perdas na produtividade agrícola. O uso excessivo de inseticidas químicos propicia resistência dessas pragas, danos ao meio ambiente, à fauna e à flora e alto custo. Dessa forma, um componente imprescindível do manejo de pragas é o controle biológico. Todas as espécies animais e vegetais apresentam inimigos naturais (parasitoides, predadores, entomopatógenos) que atacam diversos estágios de desenvolvimento. Parasitoides representam uma importante ferramenta no controle biológico de pragas lepidópteras. O conhecimento da atividade do parasitoide pupal é importante para o desenvolvimento de estratégias de controle biológico para essa praga, a qual ocasiona perdas significativas. Portanto, este artigo usa dados publicados para destacar a importância e a eficácia dos parasitoides contra a pupa da broca da cana-de-açúcar.

Bombyx mori epidermal growth factor receptor is required for nucleopolyhedrovirus replication.

Baculovirus-host interactions are important models for studying the biological control of lepidopteran pests. Research on baculovirus-host interactions has focussed on baculovirus manipulation of cellular signalling pathways, including the extracellular signal-regulated kinase (ERK) and phosphatidylinositol-3-kinases/protein kinase B (PI3K/Akt) signalling pathways. However, the mechanism underlying ERK and PI3K/Akt activation and function in response to baculovirus infection remains poorly understood. Here, we demonstrated that baculovirus activated the Bombyx mori ERK and PI3K/Akt signalling pathways via the B.mori epidermal growth factor receptor (BmEGFR). To further characterize the function of the BmEGFR/ERK signalling pathway in baculovirus replication, we calculated genome-wide changes in kinase-chromatin interactions for ERK after baculovirus infection using chromatin immunoprecipitation followed by high-throughput sequencing. A Gene Ontology analysis showed that virus infection had effects on the biological regulation, cellular process and metabolic process pathways. Moreover, ERK was shown to regulate the transcription of late viral genes. Taken together, our results suggest that baculoviruses manipulate components of the host cell machinery for replication via modulation of the BmEGFR signalling pathway.

Natural parasitism by Trichogramma spp. in agroecosystems of the Mid-North, Brazil

ABSTRACT: The micro-hymenopterans of genus Trichogramma are eggs parasitoids used in the biological control of lepidopteran pests. The objectives of this study was to record the interaction of species of Trichogramma and their hosts on crops in the Mid-North, in the states of Maranhão and Piauí, Brazil. Lepidopteran eggs were sampled on crops and non-crops. Trichogramma atopovirilia , T. manicobai , T. galloi and T. pretiosum occur naturally in eggs of eight lepidopteran species. Natural parasitism shown by Trichogramma species reveals the importance of these parasitoids as agents for biological control in the Mid-North region.

Parasitism and Suitability of Fertilized and Nonfertilized Eggs of the Rice Striped Stem Borer, Chilo suppressalis (Lepidoptera: Crambidae), for Trichogramma Parasitoids.

Trichogramma spp. are egg parasitoids largely used worldwide for biological control of lepidopteran pests. Host quality (related to host size and age, host species, etc.) can influence parasitism preference and fitness of the parasitoid offspring. However, few studies have documented the performance of Trichogramma parasitoids when they parasitize nonfertilized eggs of their lepidopteran hosts. In the present study, we investigated the performance of three indigenous Trichogramma species ( Trichogramma japonicum Ashmead, Trichogramma dendrolimi Matsumura, and Trichogramma chilonis Ishii) on fertilized and nonfertilized eggs of their native host, Chilo suppressalis (Walker). The results showed that the three Trichogramma species tested could recognize fertilized and nonfertilized eggs of C. suppressalis . The different fertilization status eggs of C. suppressalis were all accepted by T. japonicum , T. chilonis , and T. dendrolimi with a clear tendency that they preferred to parasitize fertilized eggs whether under no-choice or choice conditions. All Trichogramma species successfully completed development in parasitized eggs of C. suppressalis regardless if the host eggs were fertilized or not. There was similar adult emergence rate, development time, and female progeny between fertilized and nonfertilized eggs for all Trichogramma species with an exception of development for T. chilonis . Trichogramma chilonis had shorter developmental time in fertilized eggs than in nonfertilized ones. Whether in fertilized or nonfertilized eggs, T. dendrolimi had longer development time than other two Trichogramma species. However, T. dendrolimi had the largest percentage of female progeny on fertilized eggs, followed by T. japonicum , and T. chilonis had the least percentage of female progeny. The present study indicates that Trichogramma parasitoids (i) can distinguish fertilized vs. nonfertilized host eggs, and (ii) could use nonfertilized host eggs for successful offspring development (despite showing lower preference for such eggs) [corrected].

Maternal thermal effect on diapause inTrichogrammaspecies (Hymenoptera: Trichogrammatidae)

AbstractBoth direct thermal and maternal photoperiodic effects on diapause induction have been thoroughly investigated in many insect species, while maternal thermal effects have been infrequently studied. We studied the effect of temperature during development of maternal generation on the proportion of diapausing progeny in four species of the genusTrichogrammaWestw., minute egg parasitoids, widely used for biological control of lepidopteran pests. The maternal generations were reared at day lengths of 12 and 18 h and temperatures of 17, 20, 25 and 30°C, and their progeny developed under day length of 12 h and temperatures of 13 and 14°C. InT. evanescensandT. piceum, the proportion of diapausing progeny decreased with increasing temperature under all tested photoperiods and thermal regimes of progeny development; the high temperature of 30°C totally averted diapause of progeny. InT. buesiandT. principium,low temperatures of 17 and 20°C resulted in relatively high proportion of diapausing progeny only when the maternal generation developed under short‐day conditions. The threshold of the maternal thermal response varied from 17–18 to 22–23°C. Under field conditions,Trichogrammafemales are exposed to such high temperatures only during summer, when diapause in their progeny is in any case prevented by the maternal photoperiodic response and by the thermal response of the larvae. We conclude that the maternal thermal effect on diapause induction, although to a different extent, is inherent toTrichogrammaspecies but, at least as suggested by laboratory experiments, it does not play any role in the regulation of seasonal development under natural conditions. However, during mass rearing ofTrichogrammawasps, it should be taken into account that high temperature, even when combined with short photoperiod, can avert diapause in the next generation.

Natural Biological Control of Lepidopteran Pests by Ants

The predatory ants (Hymenoptera: Formicidade) are social insects and important natural enemies of pests in agroecosystems. Despite the importance of these predators, little is known about their role, especially in tropical regions. Among the major Lepidopteran pests of vegetables are Ascia monuste (Pieridae), Diaphania nitidalis (Crambidae), Neoleucinodes elegantalis (Crambidae) and Tuta absoluta (Gelechiidae). Thus, this work aimed to study the natural biological control of A. monuste, D. nitidalis, N. elegantalis and T. absoluta by ants. For this, we evaluated the natural biological control of A. monuste on kale and D. nitidalis on cucumber both species in the dry season. Whilst the natural biological control of N. elegantalis and T. absoluta on tomato plants were evaluated in the rainy and dry seasons. Ants preyed on Lepidoptera in the pupa stadium. They also preyed on eggs of D. nitidalis. The activity of predatory ants occurred mainly during the night. The ants were the main causes of pupae mortality of A. monuste, D. nitidalis and T. absoluta. Beyond the ants, the physiological disturbances and birds were also important factors of pupae mortality of N. elegantalis. The ants Labidus coecus and Solenopsis sp. were observed preying on pupae whereas the Paratrechina sp. was observed preying eggs of D. nitidalis. The pupae mortality of D. nitidalis and T. absoluta by ants were higher than the pupae mortality of N. elegantalis and A. monuste. The rate of pupae predation of N. elegantalis by ants was similar in rainy and dry seasons and the same occurred with T. absoluta.

Common Virulence Factors and Tissue Targets of Entomopathogenic Bacteria for Biological Control of Lepidopteran Pests.

This review focuses on common insecticidal virulence factors from entomopathogenic bacteria with special emphasis on two insect pathogenic bacteria Photorhabdus (Proteobacteria: Enterobacteriaceae) and Bacillus (Firmicutes: Bacillaceae). Insect pathogenic bacteria of diverse taxonomic groups and phylogenetic origin have been shown to have striking similarities in the virulence factors they produce. It has been suggested that the detection of phage elements surrounding toxin genes, horizontal and lateral gene transfer events, and plasmid shuffling occurrences may be some of the reasons that virulence factor genes have so many analogs throughout the bacterial kingdom. Comparison of virulence factors of Photorhabdus, and Bacillus, two bacteria with dissimilar life styles opens the possibility of re-examining newly discovered toxins for novel tissue targets. For example, nematodes residing in the hemolymph may release bacteria with virulence factors targeting neurons or neuromuscular junctions. The first section of this review focuses on toxins and their context in agriculture. The second describes the mode of action of toxins from common entomopathogens and the third draws comparisons between Gram positive and Gram negative bacteria. The fourth section reviews the implications of the nervous system in biocontrol.

Investigation of sex ratio and adult longevity of Habrobracon hebetor Say in relation to some conventional and biorational insecticides

Habrobracon hebetor Say (Hymenoptera: Braconidae) is one of the most important natural enemies that has been used for biological control of lepidopteran pests, so the sub-lethal effects of imidacloprid, abamactin and Bacillus thuringiensis Berliner (Bt) on progeny sex ratio (male/total) and adult longevity of H. hebetor were examined under laboratory conditions. It was found that the progeny sex ratio changed to male bias significantly by Bt treatment. Also, the adult longevity of the wasp was affected adversely by insecticides treatment and it was longest in Bt treatment. In conclusion, the current results indicated that the sub-lethal concentration of these insecticides affected the progeny sex ratio and adult longevity and the best strategies for reduction of hazard to the H. hebetor in combination with Bt is to avoid their application before Bt spraying.

Biological Characteristics ofTrichospilus diatraeae(Hymenoptera: Eulophidae) are Influenced by the Number of Females Exposed Per Pupa ofTenebrio molitor(Coleoptera: Tenebrionidae)

Different numbers of parasitoid females confined with a host can variously affect the number, sex ratio and other characteristics of the parasitoid's progeny. The objective of this study was to elucidate the effects of various ratios of Trichospilus diatraeae Cherian & Margabandhu (Hymenoptera: Eulophidae) females to pupae of one its hosts, Tenebrio molitor L. (Coleoptera: Tenebrionidae), primarily on the number of this parasitoid's progeny and their sex ratio. Both the parasitoid and the host used in this study were taken from cultures reared in the laboratory by standard methods. In order to minimize the effects of variations in host weight, 24 h-old T. molitor pupae weighing between 0.110 and 0.140 g were held as a single individuals in glass tubes (2.5 cm Ø × 14 cm L) with 48 h-old T. diatraeae females for 72 h to allow parasitization. After this period, female wasps were removed and the host pupa were transferred, one per glass tube, to a climate-controlled room at 25 ± 2 °C, 70 ± 10% RH and 12:12 h L:D. The experimental design was completely randomized with six treatments (parasitoid-host ratios) and 12 replicates per treatment. The ratios of T. diatraeae females per host used were: 1:1, 7:1, 14:1, 21:1, 28:1 and 32:1. The percentage parasitism of T. diatraeae on T. molitor pupae was 33.33, 82.00 and 83.33% at ratios of 1:1, 7:1 and 14:1, respectively, and 100% at all other ratios. The emergence of T. diatraeae adults from parasitized pupae was 75% at a parasitoid-host ratio of 1:1 and 100% at ratios of 21:1, 28:1 and 32:1. The duration of the parasitoid's life cycle ranged from 21.00 ± 2.22 to 24.00 ± 2.00 days at parasitoid-host ratios of 32:1 and 1:1, respectively. The number of T. diatraeae progeny per T. molitor pupa was highest at a ratio of 21:1 (246.50 ± 50.18). The proportion of T. diatraeae females in the offspring decreased as the parasitoid-host ratio increased, varying between 0.82 ± 0.06 and 0.97 ± 0.01. A parasitoid-host ratio of 21:1 T. diatraeae females per T. molitor pupa is considered the most adequate and appropriate for mass-rearing of this parasitoid. Tenebrio molitor appears to be a suitable alternate host for efficient mass-rearing of T. diatraeae for biological control of lepidopteran pests. At the parasitoid-host ratio of 21:1, each T. molitor pupa supported the production of 246.5 parsitoids of which 88% were females, i.e., 216.9 females and 29.6 males; each T. diatraeae female produced 9.55 ± 0.48 female progeny, and the developmental time from egg to adult was 20.4 days.

Effect of host instar and temperature on fitness-related traits in the solitary endoparasitoid, Meteorus pulchricornis

Meteorus pulchricornis is a parasitic wasp, which plays an important role in the biological control of lepidopteran pests. Through assessment of the effect of host instar and temperature on fitness-related traits of M. pulchricornis, this study ascertained the establishment potential of M. pulchricornis in the fields. Results showed that M. pulchricornis accepted Spodoptera exigua for oviposition and the progenies developed successfully in hosts of all ages at three constant temperatures, 20°C, 25°C and 30°C. Parasitoid survivorship from oviposition to adult eclosion was highest at 25°C, followed by 20°C and 30°C. Cocoon weight and hind tibia length differed significantly when the parasitoid was exposed to 20°C, 25°C and 30°C. The fitness-related traits of M. pulchricornis did not increase linearly with the host age at different temperatures. The host quality was found to be optimal at intermediate host instars, which the female parasitoid preferred to parasitize. The larval development rate increased with temperature in a linear fashion up to 30°C. The threshold for larval and pupal development of M. pulchricornis was 4.2°C and 1.6°C, respectively. These results have implications for predicting the population dynamics of M. pulchricornis in situ and optimizing mass rearing of the parasitic wasp under laboratory conditions.

Parasitoid and host egg ages have independent impact on various biological traits in a Trichogramma species

Trichogramma (Hymenoptera: Trichogram- matidae) are species used worldwide for the biological control of Lepidopteran pests, notably through inundative releases on millions of hectares. The optimal use of Trichogramma parasitoids in crop protection requires an accurate knowledge of their biology. More specifically, the importance of age factor in parasitoids during the time they forage in crops for host eggs (after initial release) and how the aging of host eggs could impact parasitoid biological traits may be important for overall efficiency in terms of crop protection. In this context, the importance of parasit- oid female and host egg ages on parasitism rate and the development of offspring was studied in laboratory con- ditions on Trichogramma cacoeciae Marchal (Hymenop- tera: Trichogrammatidae) and the eggs of the pest Lobesia botrana Denis and Schiffermuller (Lepidoptera: Tortrici- dae). Host eggs tested were 1-2- and 3-4-day-old, while the ages of T. cacoeciae adult females varied from 1-day- old to 4-day-old post-emergence. When L. botrana eggs were 3-4-day-old, they were less parasitized by T. cacoe- ciae than 1-2-day-old eggs, and this was not linked to the age of T. cacoeciae females. The age of parasitoid females has an effect on parasitism, as 1-day-old females produced fewer parasitized eggs than 2, 3, and 4-day-old females. For the total number of L. botrana eggs killed by T. cacoeciae, the two factors did not show significant effects. When L. botrana eggs were 1-2-day-old, parasitoid emergence increased according to the age of parasitoid females with the highest success observed for 3-day-old females. The lowest emergence rates were obtained with T. cacoeciae females 1-day-old. The development time was also longer with the young 1-day-old parasitoid females. This study demonstrated that both the aging of parasitoids and host eggs play a role in the subsequent development of parasitoid offspring. The importance of these results in the context of biological control programs involving Tricho- gramma parasitoids is discussed.

Natural occurrence of Wolbachia-infected and uninfected Trichogramma species in tomato fields in Portugal

Minute egg parasitoids of the genus Trichogramma (Hymenoptera; Trichogrammatidae) are promising candidates for biological control of lepidopteran pests in tomato in Portugal. This certainly applies to native Trichogramma strains that have thelytokous reproduction, i.e., produce only daughters. In Trichogramma wasps, thelytoky is mostly induced by the intracellular bacterium Wolbachia. In this study, we carried out a field survey of native Trichogramma species in four locations in Ribatejo, the main processing tomato region of Portugal, and determined the prevalence of Wolbachia in those species. Five Trichogramma species were found to emerge from lepidopteran eggs collected in the field, namely Trichogramma bourarache, Trichogramma cordubensis, Trichogramma evanescens, Trichogramma pintoi, and Trichogramma turkestanica. T. evanescens and T. pintoi were by far the dominating species representing, respectively, 64.9 and 26.4% of the trichogrammatids collected. Total natural parasitism rates of the collected lepidopteran eggs by Trichogramma wasps ranged from 28.2 to 64.6%. Three Trichogramma species were found to be infected with Wolbachia, namely T. cordubensis, T. evanescens, and T. turkestanica. All the wasp broods belonging to T. cordubensis were infected, whereas low infection rates were found in T. evanescens (0.9% of the broods) and T. turkestanica (4.5% of the broods). The latter represents the first record of a Wolbachia infection in T. turkestanica. Sequencing of the Wolbachia surface protein, wsp, revealed this Wolbachia infection to be related to other Wolbachia infections in Trichogramma wasps. As Wolbachia-infected thelytokous strains exist for T. evanescens, the most abundant Trichogramma species naturally occurring in the tomato fields of the Ribatejo region, this species offers interesting and powerful options for biological control of lepidopteran pests in processing tomato in this region.

Dissemination of the biocontrol agent Vairimorpha necatrix by the spined soldier bug, Podisus maculiventris

AbstractThe ability of the spined soldier bug, Podisus maculiventris (Say) (Heteroptera: Pentatomidae), to disseminate infective forms of two lepidopteran pathogens, Vairimorpha necatrix (Kramer) (Microspora: Microsporidia) and Lacanobia oleracea granulovirus (LoGV) was investigated. Individual female P. maculiventris that had fed on Lacanobia oleracea L. (Lepidoptera: Noctuidae) larvae, infected with V. necatrix, excreted approximately 6 × 108V. necatrix spores during the subsequent 7 days. Excreted spores were fed to L. oleracea larvae, causing 100% mortality, indicating that the spores remained viable after passing through the gut of the predator. Podisus maculiventris that had fed on V. necatrix or LoGV‐infected larvae were allowed to defecate on the foliage of tomato plants, prior to the infestation of the plants with L. oleracea or Spodoptera littoralis (Boisduval) (Lepidoptera: Noctuidae) larvae. This proved to be an effective way of infecting the pest larvae with the pathogens, particularly when five predatory bugs were used per plant. After 20 days, the number of S. littoralis and L. oleracea surviving on the plants was reduced by 75% and 61%, respectively. Female P. maculiventris maintained on V. necatrix‐infected prey showed reduced egg production and longevity, whilst those fed on LoGV‐infected prey showed only reduced egg production. The potential for P. maculiventris to disseminate insect pathogens is discussed in the context of improved biological control of lepidopteran pests.
