Initial oil concentration affects hydrocarbon biodegradation rates and bacterial community composition in seawater

During oil spills in the field or for laboratory incubation studies, different oil concentrations are often encountered or applied, yet how initial oil concentration affects biodegradation rates of hydrocarbons and the development of oil degraders remains unclear. We incubated seawater for 50 d with different oil concentrations (0, 50, 100, 200, 400 and 800 ppm). n-Alkanes and polycyclic aromatic hydrocarbons (PAHs), and the bacterial community were analyzed periodically. Results show that the biodegradation rates of alkanes, derived from first order kinetics, decreased with increasing oil concentration, but percent residual was ~50% regardless of the initial concentration. In contrast, the biodegradation rates of PAHs increased with concentration, and the percent residual increased with oil concentration. Increasing oil concentration resulted in increased abundances of Rhodobacterales, Altererythrobacter, and Neptuniibacter. However, Alcanivorax abundance was barely detected in 400 and 800 ppm. Overall, oil concentration critically affected the degradation of hydrocarbons and the bacterial community.

Biodegradation of polyaromatic hydrocarbons and the influence of environmental factors during the co-composting of sewage sludge and green forest waste

The main aim of this research was to study the biodegradation of polycyclic aromatic hydrocarbons (PAHs) and the influence of environmental factors during composting in order to realize the utilization of PAHs contaminated sewage sludge. A production scale compost experiment was conducted with three mixtures of sewage sludge (SS): green forest waste (GFW) at quality ratios of 3:1 (T1), 3:2 (T2) and 3:3 (T3). The residual concentration of PAHs in the three treatments met the permissible limit prescribed by the Agricultural Sludge Pollutant Control Standard (GB 4284-2018). The biodegradation rates of PAHs were 0.0280, 0.0281 and 0.0218 mg/d, and removal efficiencies were 70.7%, 75.2% and 62.4% in T1, T2, T3, respectively, which followed the first-order rate kinetics. The residual concentration of PAHs in T2 was the lowest (1.81 mg/kg), while the germination index (GI) of Tall Fescues (~0.9) was the highest during 50 days of co-composting. The bacterial diversity negatively correlated with the residual PAHs content. The main genus, Bacillus, Pseudomonas and Methylotenera, with cellulose-degrading and PAHs co-metabolizing functions contributed significantly to sludge maturation and PAHs degradation. The dominant microorganisms, Pseudomonas, Sphingobacterium and Chitinophagaceae_uncultured, could support the high removal rate of PAHs in T2. Temperature, pH, total organic carbon (TOC), total nitrogen (TN) and carbon/nitrogen (C/N) ratio had significant positive effect on the compost maturity and quality, and these parameters correlated with the PAHs biodegradation efficiency during composting, especially in T2.

Solubilization, solution equilibria, and biodegradation of PAH’s under thermophilic conditions

Biodegradation rates of PAHs are typically low at mesophilic conditions and it is believed that the kinetics of degradation is controlled by PAH solubility and mass transfer rates. Solubility tests were performed on phenanthrene, fluorene and fluoranthene at 20 °C, 40 °C and 60 °C and, as expected, a significant increase in the equilibrium solubility concentration and of the rate of dissolution of these polycyclic aromatic hydrocarbons (PAHs) was observed with increasing temperature. A first-order model was used to describe the PAH dissolution kinetics and the thermodynamic property changes associated with the dissolution process (enthalpy, entropy and Gibb’s free energy of solution) were evaluated. Further, other relevant thermodynamic properties for these PAHs, including the activity coefficients at infinite dilution, Henry’s law constants and octanol–water partition coefficients, were calculated in the temperature range 20–60 °C. In parallel with the dissolution studies, three thermophilic Geobacilli were isolated from compost that grew on phenanthrene at 60 °C and degraded the PAH more rapidly than other reported mesophiles. Our results show that while solubilization rates of PAHs are significantly enhanced at elevated temperatures, the biodegradation of PAHs under thermophilic conditions is likely mass transfer limited due to enhanced degradation rates.

A molecular modeling analysis of polycyclic aromatic hydrocarbon biodegradation by naphthalene dioxygenase

A theoretical analysis was performed to examine the role of naphthalene dioxygenase (NDO) enzymes in determining differences in biodegradability and biodegradation rates of two- to four-ring polycyclic aromatic hydrocarbons (PAHs) via oxygenation and desaturation reactions. Investigation of the thermodynamics of PAH biodegradation reactions catalyzed by NDO revealed that enthalpies of reaction can explain reaction patterns or regioselectivity of the enzyme in limited cases. Molecular modeling analysis of the size and shape constraints of PAH-enzyme interactions suggests that PAHs bigger than approximately four rings and compounds with alpha substituents or other structural features contributing to increased width at the end of the substrate near the active site are expected to have binding difficulties. This explains some regioselectivity observations, in that thermodynamically favorable sites on some PAH molecules cannot be positioned correctly to be oxidized at the active site. The enzyme fit analysis also suggests that slower biodegradation rates are expected for compounds with larger widths because of the unique positioning that is required for reaction to occur. An inverse relationship between a molecular descriptor of compound width and previously obtained biodegradation rates suggests that this descriptor may be valuable for predicting relative biodegradation rates of PAHs with dioxygenases other than NDO.

Biodegradation of polycyclic aromatic hydrocarbons in the natural waters of the Yellow River: Effects of high sediment content on biodegradation

The contamination of polycyclic aromatic hydrocarbons (PAHs) has become one of the major problems in the Yellow River of China. As the Yellow River is the most turbid large river in the world, it remains unknown to which extent the high suspended sediment content in the river may affect the fate and effect of PAHs. Here we report the effect of sediment on biodegradation of chrysene, benzo( a)pyrene and benzo( g, h, i)perylene with phenanthrene as a co-metabolism substrate in natural waters from the Yellow River. Biodegradation kinetics of the PAHs in the river water with various levels of sediment contents were studied in the laboratory by fitting with a biodegradation kinetics model for organic compounds not supporting growth. The results indicated that the biodegradation rates of PAHs increased with the sediment content in the water. When the sediment contents were 0, 4 and 10 g/l, the biodegradation rate constants of chrysene with the initial concentration of 3.80 μg/l were 0.053, 0.084 and 0.111 d −1, respectively. Further studies suggested the enhanced biodegradation rate in the presence of sediment was caused by the following mechanisms: (1) the population of PAH-degrading bacteria in the water system was found to increase with the sediment content; the bacteria population on sediment phase was far greater than that on water phase during the cultivation process; (2) the sorption of PAHs on the sediment phase was well described by the dual adsorption–partition model. Although the sorption capacity of PAH per unit weight of sediment decreased with the increase of the sediment content, the amount of sorbed PAH increased with the sediment content; and, (3) the desorption of PAHs from the solid phase led to a higher concentration near the water–sediment interface. Since the bacteria were also attached to the interface, this resulted in an increased contact chance between the bacteria and PAHs.

Biodegradation of polycyclic aromatic hydrocarbons in oil-contaminated beach sediments treated with nutrient amendments.

Microbial biodegradation of polycyclic aromatic hydrocarbons (PAHs) during the process of bioremediation can be constrained by lack of nutrients, low bioavailability of the contaminants, or scarcity of PAH-biodegrading microorganisms. This study focused on addressing the limitation of nutrient availability for PAH biodegradation in oil-contaminated beach sediments. In our previous study, three nutrient sources including inorganic soluble nutrients, the slow-release fertilizer Osmocote (Os; Scotts, Marysville, OH) and Inipol EAP-22 (Ip; ATOFINA Chemicals, Philadelphia, PA), as well as their combinations, were applied to beach sediments contaminated with an Arabian light crude oil. Osmocote was the most effective nutrient source for aliphatic biodegradation. This study presents data on PAH biodegradation in the oil-spiked beach sediments amended with the three nutrients. Biodegradation of total target PAHs (two- to six-ring) in all treatments followed a first-order biodegradation model. The biodegradation rates of total target PAHs in the sediments treated with Os were significantly higher than those without. On Day 45, approximately 9.3% of total target PAHs remained in the sediments amended with Os alone, significantly lower than the 54.2 to 58.0% remaining in sediment treatments without Os. Amendment with Inipol or soluble nutrients alone, or in combination, did not stimulate biodegradation rates of PAHs with a ring number higher than 2. The slow-release fertilizer (Os) is therefore recommended as an effective nutrient amendment for intrinsic biodegradation of PAHs in oil-contaminated beach sediments.

Effect of non-aqueous phase liquid on biodegradation of PAHs in spilled oil on tidal flat

Biodegradation rates of polycyclic aromatic hydrocarbons (PAHs) in spilled oil stranded on tidal flats were studied using model reactors to clarify the effects of NAPL on the biodegradation of PAHs in stranded oil on tidal flat with special emphasis on the relationship between dissolution rates of PAHs into water and viscosity of NAPL. Biodegradation of PAHs in NAPL was limited by the dissolution rates of PAHs into water. Biodegradation rate of chrysene was smaller than that for acenaphthene and phenanthrene due to the smaller dissolution rates. Dissolution rates of PAHs in fuel oil C were smaller those in crude oil due to high viscosity of fuel oil C. Therefore, biodegradation rates of PAHs in fuel oil C were smaller than those in crude oil. Biodegradation rates of PAHs in NAPL with slow decrease rate like fuel oil C were slower than those in NAPL with rapid decrease like crude oil. The smaller decrease rate of fuel oil C than crude oil was due to higher viscosity of fuel oil C. Therefore, not only the dissolution rate of PAHs but also the decrease rates of NAPL were important factors for the biodegradation of PAHs.

Dissolution rates of PAHs in fuel oil C were smaller than those in crude oil due to high viscosity of fuel oil C. Hence, biodegradation rates of PAHs in fuel oil C were smaller than those in crude oil. Biodegradation rates of PAHs in NAPL with slow rate of decrease like fuel oil C was slower than those in NAPL with rapid rate of decrease like crude oil. The smaller rate of decrease of fuel oil C than crude oil was due to the higher viscosity of fuel oil C. Therefore, not only the dissolution rate of PAHs but also the rates of decrease of NAPL were important factors for the biodegradation of PAHs.
