Adsorption of fluoride and DB-53 dye onto PLA/rGO nanoparticles: Mathematical modeling and statistical studies

World Health Organization considered hazardous dye and fluoride removal to be an emergency issue in the water treatment process. In this investigation, fluoride and DB-53 dye adsorption was carried out using perovskite LaAlO3 (PLA) anchored with green synthesized rGO via the solvothermal process. BET characterization of PLA/rGO shows enhanced specific surface area (250 m2g−1) than PLA (5.91 m2g−1). PLA/rGO showed remarkable adsorption capacity of fluoride (qmax = 173.1 mgg−1) and DB-53 dye (qmax = 37.78 mgg−1) individually. The Fractal-like-pseudo-second order and Sips isotherm model well fitted with experimental statistical computation data R2 (1.000). The field water studies of fluoride removal and competing ions effect proved the novelty of the material. The potentiality of the PLA/rGO is proved by fluoride removal upto six effective cycles. Further, PLA/rGO exhibited high antibacterial properties compared to PLA because of their opposite electric charged nature of bacterial cell walls and surface charge of the nanocomposite. Hence, the present material study is multifunctional and interesting in the adsorption of fluoride and DB-53 dye with antibacterial properties.

Antimicrobial Properties of Polyaniline and Polypyrrole Decorated with Zinc-Doped Copper Oxide Microparticles.

Polyaniline (PANI) and polypyrrole (PPY) were synthesized by carbon dots (CDs) under UV irradiation and then sonicated together with zinc acetate and copper acetate to form the PANI-Zn@CuO and PPY-Zn@Cu composites. The former consisted of agglomerated spherical particles with diameters of 1–5 µm, whereas the latter displayed irregular stick shapes with similar diameters. The bacterial potency of the composites against Escherichia coli and Staphylococcus aureus was enhanced remarkably with Zn doping in the CuO matrix, designated as Zn0.11Cu0.89O, at 0.144 mg/mL. The cell death was mainly attributed to the release of reactive oxygen species (ROS) that would severely damage DNA, proteins, and lipids. Bacteria could adhere to neutral surfaces of the composites by van der Waals attractive forces. The binding event disrupted the native surface charge of bacterial cells to induce cell lysis and result in eventual cell death.

Promoting bacteria-anode interfacial electron transfer by palladium nano-complex in double chamber microbial fuel cell

ABSTRACT The slow electron transfer between microbial outer membrane and electrode surface is considered one of the limitations of Microbial Fuel Cell (MFC) performance. The aim of the present work is to assess the role of palladium α-lipoic acid nanocomplex compound (PLAC) in promoting bacteria-anode interfacial electron transfer, by studying the dielectric properties of Shewanella oneidensis WW-1 cell membrane and its contribution to biofilm formation on the anode. The results showed that adding PLAC increased bacterial cell membrane permeability and outer cell surface charge. Exopolysaccharides (EPS) and surface-bound proteins increased 2.27 and 1.14 fold, respectively upon adding 0.25% v/v PLAC. Dynamic Light Scattering (DLS) showed uniform distribution of Shewanella-PLAC biocomposite size while Zeta potential and Fourier Transform Infrared (FTIR) Spectroscopy results suggest that PLAC diffused inside the cells. Transmission Electron Microscope (TEM) images reveal Exopolysaccharide (EPS) mat around the cells when PLAC was added to the cells, also confirmed by the FTIR spectrum. Scanning Electron Microscope and Atomic Force Microscope (AFM) confirmed the thickness of biofilm in the presence of PLAC. The average voltage reached 492 mV (external resistance 1 KΩ) over 35 days using 0.25% v/v PLAC as compared to a few hours in MFCs lacking PLAC. The results suggest that the addition of PLAC assisted in interfacial direct electron transfer through enhancing biofilm formation, moreover, its hydrophilic/lipophilic nature facilitated the electron shuttling process from within the bacterial cell to the electrode surface suggesting the involvement of mediated electron transfer as well.

The role of zeta potential in the adhesion of E. coli to suspended intertidal sediments

The extent of pathogen transport to and within aquatic systems depends heavily on whether the bacterial cells are freely suspended or in association with suspended particles. The surface charge of both bacterial cells and suspended particles affects cell-particle adhesion and subsequent transport and exposure pathways through settling and resuspension cycles. This study investigated the adhesion of Faecal Indicator Organisms (FIOs) to natural suspended intertidal sediments over the salinity gradient encountered at the transition zone from freshwater to marine environments. Phenotypic characteristics of three E. coli strains, and the zeta potential (surface charge) of the E. coli strains and 3 physically different types of intertidal sediments was measured over a salinity gradient from 0 to 5 Practical Salinity Units (PSU). A batch adhesion microcosm experiment was constructed with each combination of E. coli strain, intertidal sediment and 0, 2, 3.5 and 5 PSU. The zeta potential profile of one E. coli strain had a low negative charge and did not change in response to an increase in salinity, and the remaining E. coli strains and the sediments exhibited a more negative charge that decreased with an increase in salinity. Strain type was the most important factor in explaining cell-particle adhesion, however adhesion was also dependant on sediment type and salinity (2, 3.5 PSU > 0, 5 PSU). Contrary to traditional colloidal (Derjaguin, Landau, Vervey, and Overbeek (DLVO)) theory, zeta potential of strain or sediment did not correlate with cell-particle adhesion. E. coli strain characteristics were the defining factor in cell-particle adhesion, implying that diverse strain-specific transport and exposure pathways may exist. Further research applying these findings on a catchment scale is necessary to elucidate these pathways in order to improve accuracy of FIO fate and transport models.

Streptomycin favors biofilm formation by altering cell surface properties.

Studies have shown that external stress induces biofilm formation, but the underlying details are not clearly understood. This study investigates the changes in cell surface properties leading to increase in biofilm formation by Staphylococcus aureus and Pseudomonas aeruginosa in the presence of streptomycin. Bacterial attachment in the presence and absence of streptomycin was quantified by fluorescence spectroscopy. In addition, cell surface charge and contact angle were measured and the free energy barrier for attachment was modeled using extended Derjaguin-Landau-Verwey-Overbeek (xDLVO) theory. Peptides from bacterial cell surface were shaved by protease treatment and identified with ultra-performance liquid chromatography (UPLC)-QTOF and a homology search program SPIDER. Biofilm formation increased significantly in the presence of streptomycin (10mg/L) in the culture. Bacterial cell surface charge reduced, and hydrophobicity increased leading to a net decrease in the free energy barrier for attachment. Extracellular matrix-binding protein was positively regulated in S. aureus under stress, indicating stronger interaction between bacterial cells and solid surface. In addition, several other proteins including biofilm regulatory proteins, multidrug efflux pumps, transporters, signaling proteins, and virulence factors were differentially expressed on bacterial cell surface, which is indicative of a strong stress response by bacteria to streptomycin treatment.

The Staphylococcus aureus Methicillin Resistance Factor FmtA Is a d-Amino Esterase That Acts on Teichoic Acids.

ABSTRACTThe methicillin resistance factor encoded by fmtA is a core member of the Staphylococcus aureus cell wall stimulon, but its function has remained elusive for the past two decades. First identified as a factor that affects methicillin resistance in S. aureus strains, FmtA was later shown to interact with teichoic acids and to localize to the cell division septum. We have made a breakthrough in understanding FmtA function. We show that FmtA hydrolyzes the ester bond between d-Ala and the backbone of teichoic acids, which are polyglycerol-phosphate or polyribitol-phosphate polymers found in the S. aureus cell envelope. FmtA contains two conserved motifs found in serine active-site penicillin-binding proteins (PBPs) and β-lactamases. The conserved SXXK motif was found to be important for the d-amino esterase activity of FmtA. Moreover, we show that deletion of fmtA (ΔfmtA) led to higher levels of d-Ala in teichoic acids, and this effect was reversed by complementation of ΔfmtA with fmtA. The positive charge on d-Ala partially masks the negative charge of the polyol-phosphate backbone of teichoic acids; hence, a change in the d-Ala content will result in modulation of their charge. Cell division, biofilm formation, autolysis, and colonization are among the many processes in S. aureus affected by the d-Ala content and overall charge of the cell surface teichoic acids. The esterase activity of FmtA and the regulation of fmtA suggest that FmtA functions as a modulator of teichoic acid charge, thus FmtA may be involved in S. aureus cell division, biofilm formation, autolysis, and colonization.

Surfactin restores and enhances swarming motility under heavy metal stress

The present work reports the importance of lipopeptide biosurfactant on swarming motility of multi-metal resistant (MMR) bacterium under heavy metal stress. The MMR bacteria strain CM100B, identified as Bacillus cereus, was isolated from the coal mine sample. The strain was able to grow and reduce several metals namely Cd2+, Co2+, Cu2+, Ni2+, Mn2+ and Pb2+ ions which are common environmental pollutants. Presence of toxic heavy metal ions in the swarming medium significantly altered the motility of CM100B. Presence of Cd2+ and Pb2+ ions inhibited development of peritrichous flagella, thus inhibiting swarming motility. However, the addition of anionic biosurfactant surfactin restored (in case of Cd2+ and Pb2+ ions) or enhanced (in case of Co2+, Cu2+, Ni2+ and Mn2+) the swarming ability of CM100B. Zeta potential studies for determining bacterial cell surface charge indicated that surfactin provided a suitable swarming environment to bacteria even under metal stress by chelating to cationic metal ions. Non-ionic surfactant Triton X-100 was unable to restore swarming under Cd2+ and Pb2+ ion stress. Thus, suggesting that surfactin can aid in motility not only by reducing the surface tension of swarming medium but also by binding to metal ions in the presence of metal ions stress.

Hydrophobic and Electrostatic Interaction Chromatography for Estimating Changes in Cell Surface Charge ofEscherichia coliCells Treated with Pulsed Electric Fields

Pulsed electric field (PEF) treatments, a nonthermal process, have been reported to injure and inactivate bacteria in liquid foods. However, the effect of this treatment on bacterial cell surface charge and hydrophobicity has not been investigated. Apple juice (pH 3.8) purchased from a wholesale distributor was inoculated with cocktail of Escherichia coli O157:H7 at 7.4 log CFU/mL, processed with a PEF at a field strength of 18.4 kV/cm and 32.2 kV/cm at 25°C, 35°C, and 45°C with a treatment time of 160 μs and a flow rate of 120 mL/min. Bacterial cell surface charge and hydrophobicity of untreated and PEF-treated E. coli O157:H7 were determined immediately and after storage at 5°C and 23°C using hydrophobic and electrostatic interaction chromatography. Similarly, the populations surviving the PEF treatments including injured cells were determined by plating 0.1 mL of the sample on sorbitol MacConkey agar and tryptic soy agar (TSA) plates. The surviving populations of E. coli cells after PEF treatment varied depending on field strength and treatment temperature used. Percent injury in the surviving populations was high immediately after PEF treatment and varied among treatment temperatures. Cell surface charge of E. coli bacteria before PEF treatment averaged 32.10±8.12. PEF treatments at 25°C, 35°C, and 45°C reduced the above surface charge to 26.34±1.24, 14.24±3.30, and 6.72±2.82, respectively. Similarly, the surface hydrophobicity of untreated E. coli cells at 0.194±0.034 was increased to an average of 0.268±0.022, 0.320±0.124, and 0.586±0.123 after PEF treatments at 25°C, 35°C, and 45°C, respectively. The results of this study indicate that PEF treatment affects the outer cell envelope of E. coli bacteria as evidenced by the changes in surface hydrophobicity and cell surface charge leading to injury and subsequent inactivation of the cells.

Growth-phase dependence of susceptibility to antimicrobial peptides in Staphylococcus aureus

Bacterial cell surface charge is responsible for susceptibility to cationic antimicrobial peptides. Previously, Staphylococcus aureus dlt and mprF were identified as factors conferring a positive charge upon cell surfaces. In this study, we investigated the regulation of cell surface charge during growth. Using a group of S. aureus MW2 mutants, which are gene-inactivated in 15 types of two-component systems (TCSs), we tested dltC and mprF expression and found that two TCSs, aps and agr, were associated with dltC and mprF expression in a growth phase-dependent manner. The first of these, aps, which had already been identified as a sensor of antimicrobial peptides and a positive regulator of dlt and mprF expression, was expressed strongly in the exponential phase, while its expression was significantly suppressed by agr in the stationary phase, resulting in higher expression of dltC and mprF in the exponential phase and lower expression in the stationary phase. Since both types of expression affected the cell surface charge, the susceptibility to antimicrobial peptides and cationic antibiotics was changed during growth. Furthermore, we found that the ability to sense antimicrobial peptides only functioned in the exponential phase. These results suggest that cell surface charge is tightly regulated during growth in S. aureus.

Relationship between the physicochemical properties of nonchitinolytic Listeria and Salmonella and their attachment to shrimp carapace.

Listeria and Salmonella are important foodborne pathogens normally associated with the shrimp production chain. This study investigated the potential of Salmonella Typhimurium, Salmonella Senftenberg, and Listeria monocytogenes (Scott A and V7) to attach to and colonize shrimp carapace. Attachment and colonization of Listeria and Salmonella were demonstrated. Shrimp abdominal carapaces showed higher levels of bacterial attachment (P < 0.05) than did head carapaces. Listeria consistently exhibited greater attachment (P < 0.05) than did Salmonella on all surfaces. Chitinase activity of all strains was tested and found not to occur at the three temperatures (10, 25, and 37 degrees C) tested. The surface physicochemical properties of bacterial cells and shrimp carapace were studied to determine their role in attachment and colonization. Salmonella had significantly (P < 0.05) more positive (-3.9 and -6.0 mV) cell surface charge than Listeria (-18 and -22.8 mV) had. Both bacterial species were found to be hydrophilic (<35%) when measured by the bacterial adherence to hydrocarbon method and by contact angle (theta) measurements (Listeria, 21.3 and 24.8 degrees, and Salmonella, 14.5 and 18.9 degrees). The percentage of cells retained by Phenyl-Sepharose was lower for Salmonella (12.8 to 14.8%) than it was for Listeria (26.5 to 31.4%). The shrimp carapace was found to be hydrophobic (theta = 74.5 degrees), and a significant (P < 0.05) difference in surface roughness between carapace types was noted. There was a linear correlation between bacterial cell surface charge (r2 = 0.95) and hydrophobicity (r2 = 0.85) and initial attachment (P < 0.05) of Listeria and Salmonella to carapaces. However, the same properties could not be related to subsequent colonization.

Variations in the Degree of d -Alanylation of Teichoic Acids in Lactococcus lactis Alter Resistance to Cationic Antimicrobials but Have No Effect on Bacterial Surface Hydrophobicity and Charge

An increase of the degree of d-alanylation of teichoic acids in Lactococcus lactis resulted in a significant increase of bacterial resistance toward the cationic antimicrobials nisin and lysozyme, whereas the absence of D-alanylation led to a decreased resistance toward the same compounds. In contrast, the same variations of the D-alanylation degree did not modify bacterial cell surface charge and hydrophobicity. Bacterial adhesion to polystyrene and glass surfaces was not modified either.

Relevance of Nontoxigenic Strains as Surrogates for Escherichia coli O157:H7 in Groundwater Contamination Potential: Role of Temperature and Cell Acclimation Time

Nontoxigenic bacteria are commonly used as indicators for predicting the contamination potential of pathogens in natural or engineered aqueous environments. In this study, column transport experiments were used to examine the relevance of two nontoxigenic strains of Escherichia coli O157:H7 as potential surrogates for the well-known pathogen. Experiments conducted at 11 degrees C indicate that only one of the nontoxigenic strains may be an appropriate surrogate for predicting the migration potential of the pathogen at low solution ionic strengths. Results of various bacterial characterization methods indicate that differences in cell attachment could qualitatively, but convincingly, be related to differences in cell surface charge. Additional experiments conducted at 22 degrees C reveal the influence of temperature on bacterial cell surface charge and cell attachmentto sand. The role of cell acclimation time to an artificial groundwater solution is also examined, showing little change in the degree of cell attachment over a period of several weeks.

Change of Collision Efficiency with Distance in Bacterial Transport Experiments

Understanding the mechanisms of bacterial transport in aquifers is important in developing bioremediation strategies. Collision efficiency (alpha) is one important parameter used in modeling bacterial transport. This study was undertaken to measure change in alpha with distance by performing a bacterial transport experiment in Oyster, Virginia. Following injection of a bacterium, Comamonas sp., into a well, water samples were collected at various distances along the flowpath and injected into columns packed with homogenized South Oyster focus area sediment. Zeta potentials of the bacteria in the samples were measured. Values of alpha were determined at various locations in the field in two ways: based on field breakthrough concentrations at the sampling points and based on column breakthrough concentrations. The alpha values estimated from field breakthrough decreased with distance, whereas those estimated from column breakthrough increased with distance. Bacterial cell surface charge became progressively more negative with distance in the field. We hypothesize that the apparent contradiction between field and column alpha values was caused by differences in the flow of the two systems. Flow in the columns was forced to occur through fine-grained zones of iron and aluminum hydroxide coatings that selectively removed the most negatively charged bacteria. In contrast, in the field, the injected cells did not come into contact with the positively charged coatings because the bulk solution bypassed them due to heterogeneous hydraulic properties. These results suggest that laboratory-based models may underestimate bacterial transport distance in the field. A more realistic approach may be necessary to capture the degree of heterogeneity.

An evaluation of the outer membrane charge and softness of Thiobacillus ferrooxidans by the Ohshima's electrophoretic model of a "soft" particle.

The surface charge of bacterial cells plays an important role in their interfacial physiology and adhesion to substrata mediated by the electrostatic double-layer interaction. The surface charge or potential of biological cells is generally calculated from the experimentally measurable electrophoretic velocity of these cells migrating in an external electric field, applying the well-known Smoluchowski equation which is valid for "hard" particles with a sharp interface. However, bacterial cells possessing a structured outer membrane of a finite thickness (dependent on the ionic strength and pH of the surrounding liquid medium) are expected to obey Ohshima's electrophoretic mobility equation derived recently for "soft" particles. The electrophoretic mobility of Thiobacillus ferrooxidans was measured here by the fully automated technique of electrophoretic light scattering, based on the proportionality between the mobility and the Doppler shift in the frequency of light scattered by electrophoresing cells. Agreement was obtained between the experimentally determined electrophoretic mobility expressed as a function of low ionic strength (60-6000 mumol/L) at different pH values and the best-fit theoretical predictions of the "soft" particle electrophoresis theory, which is better than in the case of applying the Smoluchowski formula. The best-fit surface-charge and softness parameters predict a rather rigid and low-charge outer membrane of the bacterium examined, as compared to the parameters obtained for other bacteria in media of high ionic strength.

Porin polypeptide contributes to surface charge of gonococci.

Each strain of Neisseria gonorrhoeae elaborates a single porin polypeptide, with the porins expressed by different strains comprising two general classes, Por1A and Por1B. In the outer membrane, each porin molecule folds into 16 membrane-spanning beta-strands joined by top- and bottom-loop domains. Por1A and Por1B have similar membrane-spanning regions, but the eight surface-exposed top loops (I to VIII) differ in length and sequence. To determine whether porins, and especially their top loop domains, contribute to bacterial cell surface charge, strain MS11 gonococci that were identical except for expressing a recombinant Por1A, Por1B, or mosaic Por1A-1B polypeptide were compared by whole-cell electrophoresis. These porin variants displayed different electrophoretic mobilities that correlated with the net numbers of charged amino acids within surface-exposed loops of their respective porin polypeptides. The susceptibilities of porin variants to polyanionic sulfated polymers correlated roughly with gonococcal surface charge; those porin variants with diminished surface negativity showed increased sensitivity to the polyanionic sulfated compounds. These observations indicate that porin polypeptides in situ contribute to the surface charge of gonococci, and they suggest that the bacterium's interactions with large sulfated compounds are thereby affected.

Kinetics of Pseudomonas aeruginosa adhesion to 304 and 316-L stainless steel: role of cell surface hydrophobicity

Fifteen different isolates of Pseudomonas aeruginosa were used to study the kinetics of adhesion to 304 and 316-L stainless steel. Stainless steel plates were incubated with approximately 1.5 X 10(7) CFU/ml in 0.01 M phosphate-buffered saline (pH 7.4). After the plates were rinsed with the buffer, the number of adhering bacteria was determined by a bioluminescence assay. Measurable adhesion, even to the electropolished surfaces, occurred within 30 s. Bacterial cell surface hydrophobicity, as determined by the bacterial adherence to hydrocarbons test and the contact angle measurement test, was the major parameter influencing the adhesion rate constant for the first 30 min of adhesion. A parabolic relationship between the CAM values and the logarithm of the adhesion rate constants (In k) was established. No correlation between either the salt aggregation or the improved salt aggregation values and the bacterial adhesion rate constants could be found. Since there was no significant correlation between the bacterial electrophoretic mobilities and the In k values, the bacterial cell surface charge seemed of minor importance in the process of adhesion of P. aeruginosa to 304 and 316-L stainless steel.
