Effects of staged rehabilitation training on inflammatory factor levels and red blood cell distribution width followingcardiac valve replacement

BackgroundThe current study was conducted aimed atexploring the effects of staged rehabilitation training on the levels of inflammatory factors and red blood cell distribution in patients who underwent cardiac valve replacement.MethodsA total of 140 patients who underwent cardiac valve replacement at The First Hospital of Hebei Medical University between April 2021 and November 2022 were included in this study. During the postoperative rehabilitation phase, the patients were randomly assigned to either the control group or the experimental group. The experimental group received staged rehabilitation training (n = 70), while the control group received conventional care and rehabilitation suggestions without specialized staged rehabilitation training (n = 70). Informed consent was obtained from all patients prior to theirinclusion in the study. Clinical data of the patients were collected andanalyzed. RDW was measured using an automated blood cell analyzer on postoperative day 1, 14, and 28. Levels ofTNF-α, IL-6 and CRP were measured using ELISA. Quality of life was evaluated usingthe WHOQOL-BREF questionnaire. The effects of postoperative rehabilitation were assessed using the 6MWD test. The occurrence of adverse events in the postoperative periodwas alsoanalyzed.ResultsThere were no significant differences in the general characteristics of the two groups of patients (P > 0.05). On the first day after surgery, no significant differences were seen in RDW between the two groups (P > 0.05). However, on the 14th and 28th day after surgery, the experimental group exhibited a significant reduction in RDW compared to the control group (P < 0.05). On the first day after surgery, the levels of serum TNF-α, IL-6 and CRP were comparable between the two groups (P > 0.05). However, on the 14th and the 28th after surgery, the experimental group showed evidently lower levels of TNF-α, IL-6 and CRP compared to the control group (P < 0.05). The experimental group demonstrated higher scores in the domains of physical health, psychological state, social relationships, and environment in the WHOQOL-BREF questionnaire compared to the control group (P < 0.05). Furthermore, the experimental group exhibited increased average,minimum,maximum walking distances in the6-minute walking test compared to the control group (P < 0.05). There were no significant differences in the incidence of postoperative adverse events between the two groups of patients (P > 0.05).ConclusionStaged rehabilitation training exerteda positive effect on the levels of inflammatory factors and red blood cell distribution in patients following cardiac valve replacement. This type of rehabilitation training facilitated the patient’s recovery process by reducing the inflammatory response and improving the condition of red blood cells. Additionally, it enhanced the quality of life and rehabilitation outcomes for patients.

Differences and significance of peripheral blood interleukin-6 expression between patients with granulomatous lobular mastitis and those with benign breast tumors.

It is unclear whether the mechanism of the interleukin (IL)-6 signaling pathway is similar between granulomatous lobular mastitis (GLM) and benign breast tumors. This study aimed to explore the differences and significance of peripheral blood IL-6 and related cytokines, routine blood test results, and C-reactive protein (CRP) levels between patients with GLM and benign breast tumors. Seventy-three inpatients with GLM who underwent surgery and 60 patients with benign breast tumors diagnosed based on pathological findings between November 2022 and May 2023 were included. The white blood cell (WBC) and neutrophil (NEU) counts were determined using an automatic blood cell analyzer, the CRP level was determined by an immunoturbidimetric assay, and serum IL-6 and related cytokine levels were determined by an enzyme-linked immunosorbent assay. The WBC, NEU, and CRP values in patients with GLM were significantly higher than those in patients with benign breast tumors (P < 0.01). Serum IL-6 levels were significantly higher in patients with GLM than in those with benign breast tumors (P < 0.01). There were no significant differences in the serum concentrations of IL-1β, IL-7, and interferon (IFN)-γ between patients with GLM and those with benign breast tumors (P > 0.05), but the tumor necrosis factor (TNF)-α level was higher in patients with GLM than in those with benign breast tumors (P < 0.01). In patients with GLM, the Pearson correlation analysis showed that the IL-6 level was positively correlated with NEU, NEU%, CRP, IL-17, and TNF-α values (P < 0.01). Additionally, the IL-6 level was weakly positively correlated with WBC and IFN-γ values. Conversely, in patients with benign breast tumors, the IL-6 level was not significantly correlated with the aforementioned indicators in routine blood tests but was positively correlated with IL-17, IFN-γ, and TNF-α values (P < 0.01). IL-6, NEU, NEU%, and CRP values were significantly elevated in patients with GLM compared to those with benign breast tumors, indicating that IL-6 plays an important role in the development and onset of GLM. The correlation between these cytokines and the development and progression of benign breast tumors needs to be further explored, as cytokines such as IL-6 may provide effective markers for the treatment of GLM.

Leukocyte index of intoxication according to V. K. Ostrovskii as a criterion for assessing bacterial infection

The OBJECTIVE was choose an informative, convenient and accessible in clinical practice index for assessing the level of endogenous intoxication associated with a bacterial infection.METHODS AND MATERIALS. In the groups of patients with surgical inflammatory bacterial diseases (104 people), with intracellular non-bacterial infection (254 people), and in healthy people (94 people), the degree of LII correlation according to Kalf-Kalif (as a "standard" of indices) was compared with more simple and easy-to-calculate indices: LII according to Ostrovsky, simplified LII according to Ostrovsky ¬ without taking into account plasma cells (sLII according to Ostrovsky), the blood leukocyte shift index according to Yabluchansky, the Harkavy index and the Krebs index.RESULTS. Within all groups of observations, the closest correlation with LII according to Kalf-Kalif was established for LII according to Ostrovsky, slightly less for LII according to Ostrovsky. The latter was chosen for further study due to the possibility of calculation based on the data of the 5diff automatic blood cell analyzer. Compared with the group of healthy people (1.34 (0.93; 1.59)) sLII according to Ostrovsky did not increase in the group of patients with intracellular non-bacterial infections (1.21 (0.82; 1.49)), moderately increased in subgroup of urological and gynecological patients with chronic local inflammatory processes (2.35 (1.49; 2.99)) and was many times higher in subgroups with acute inflammatory and destructive diseases of the abdominal organs (7.64 (2.05; 11.77 )), sepsis (8.58 (2.96; 13.29)) and septic shock (7.34 (4.07; 9.48)).CONCLUSION. The calculation of LII according to Ostrovsky or its simplified version without taking into account plasma cells (neutrophils + immature granulocytes)/(lymphocytes + monocytes + eosinophils + basophils) is appropriate and easily applicable in wide clinical practice for assessing a bacterial infection and the development vector of an infectious process.

Hematological and molecular characteristics of a novel α-globin variant Hb Liangqing (HBA2:c.224A>G)

ABSTRACTObjectivesTo report the hematological and molecular characteristics of a novel α-globin variant found among Chinese families.MethodsThis study was done on two unrelated families (F1 and F2). Hematological results were obtained by an automated blood cell analyzer. Hemoglobin (Hb) fraction analysis was carried out using capillary electrophoresis (CE) and high-performance liquid chromatography (HPLC). Gap-PCR and reverse dot blot (RDB) methods were used to detect the common α-thalassemia mutations in the Chinese population. The Hb variants were defined by Sanger sequencing.ResultsHb fraction analysis of F2 cord blood using HPLC showed an abnormal peak (3.5%) of the S-window, but CE presented a 12.2% abnormal peak at zone 5(S). Similar results of CE were observed from the F1 twin's cord blood. Compared with newborns, Hb analysis of F2 father using HPLC demonstrated an abnormal peak (16.9%) of S-window and an unknown peak (0.5%) at a retention time of 4.60 min, respectively. In contrast, CE revealed a high Hb F peak at zone 7 and an unknown peak at zone 1. There was no abnormality detected with Gap-PCR and RDB in these patients. However, Sanger sequencing confirmed the presence of a new heterozygous mutation (GAC>GGC) at codon 74 of the HBA2 gene (HBA2:c.224A>G), resulting in a novel Hb variant. We named it Hb Liangqing for the birthplace of the proband.ConclusionThis is the first report of Hb Liangqing detected by HPLC and CE. The normal hematological phenotype suggests that it may be a benign Hb variant.

Analysis of the Influencing Factors of Tumor Volume, Body Immunity, and Poor Prognosis after 125I Particle Therapy for Differentiated Thyroid Cancer.

To analyze the influencing factors of tumor volume, body immunity, and poor prognosis after 125I particle therapy for differentiated thyroid cancer. A total of 104 patients with differentiated TC who were treated with 125I particles during January 2020 to January 2021 was picked. These subjects were graded as low-dose group (80Gy-110Gy) and high-dose group (110Gy-140Gy) according to the minimum dose received by 90% of the target volume (D90) after surgery. The tumor volume before and after treatment was compared, and fasting venous blood was collected before and after treatment. The content of thyroglobulin (Tg) was detected by electrochemiluminescence immunoassay. The levels of absolute lymphocyte count (ALC), lymphocytes, neutrophils, and monocytes were detected on automatic blood cell analyzer. The lymphocyte to monocyte ratio (LMR), neutrophil to lymphocyte ratio (NLR) and platelet to lymphocyte ration (PLR) were calculated. The changes in the condition of patients were closely observed, and the occurrence of adverse reactions in the two groups were compared. The risk factors influencing the efficacy of 125I particle therapy for differentiated TC were analyzed through multivariate logistic regression analysis. The total effective rate of patients in the low- and high-dose groups was 78.85% and 82.69%, respectively (P > 0.05). Compared with the pretreatment period, the tumor volume and Tg level in both groups were much lower (P < 0.05), and the differences in tumor volume and Tg level had no statistically significant difference between the two groups before and after treatment (P > 0.05). At 1 week of the treatment, the total incidence of adverse reactions such as nausea, radiation gastritis, radiation parotitis, and neck discomfort was obviously higher in the high-dose group than in the low-dose group (P < 0.05). At 1 month of treatment, the incidence of adverse reactions such as nausea was markedly higher in the high-dose group than in the low-dose group (P < 0.05). After treatment, serum NLR and PLR contents were memorably elevated and LMR level was sharply decreased in both groups, and serum NLR and PLR contents were higher and LMR content was lower in the high-dose group than in the low-dose group (P < 0.05). Multivariate logistic regression analysis showed that the pathological type of follicular adenocarcinoma, tumor size ≥ 2 cm, clinical stage of III~IV, distant metastasis, and high TSH level before 125I particle treatment were all risk factors related to the efficacy of 125I particle treatment of TC (P < 0.05). The efficacy of low-dose and high-dose 125I particles in the treatment of differentiated thyroid cancer is comparable, among which low-dose 125I particles have fewer adverse effects and have less impact on the immunity of the body, which is well tolerated by patients and can be widely used in clinical practice. In addition, the pathological type of follicular adenocarcinoma, tumor size ≥ 2 cm, clinical stage III~IV, distant metastasis, and high TSH level before 125I particle treatment are all risk factors that affect the poor effect of 125I particles on thyroid cancer treatment, and early monitoring of the above index changes can help evaluate the prognosis.

An Optimized Method of Collecting Murine Peripheral Blood and Dilution Correction for Accurate Blood Cell Enumeration.

Accurate measurement of whole blood counts from mice is an essential quantitative tool across the fields of vascular cell biology. In particular, the measurement of platelet counts can be challenging as the process relies upon good phlebotomy technique, the inclusion of a sufficient amount of the appropriate anticoagulant, and very often dilution of the sample to meet the sample volume requirements of an automated analyzer. To minimize sample dilution, blood collection tubes pre-coated with the anticoagulant can be used; however, these are expensive and prone to blood clotting issues. Here, we describe a simple dilution correction method that accurately calculates blood-to-anticoagulant dilutions to generate appropriate volumes for automated blood cell analysis while minimizing blood clotting. We also discuss some simple steps that can be incorporated into blood collection methods to avoid artefacts during blood collection. Blood count data analysis involving volume correction and clot exclusion can significantly reduce variable blood cell count values among healthy untreated littermates. It also detects subtle changes in blood cell counts, mainly of platelets and RBCs in experimental settings, which can be masked in the absence of careful and precise volume correction. Blood count analysis with volume correction precisely determines mouse whole blood cell counts for investigators. The decreased variability in cell count values reduces the number of experimental animals required for meaningful analysis. © 2023 The Authors. Current Protocols published by Wiley Periodicals LLC. Ferroptosis inhibition attenuates inflammatory response in mice with acute hypertriglyceridemic pancreatitis.

Ferroptosis is involved in developing inflammatory diseases; yet, its role in acute hypertriglyceridemic pancreatitis (HTGP) remains unclear. To explore whether ferroptosis is involved in the process of HTGP and elucidate its potential mechanisms. An HTGP mouse model was induced using intraperitoneal injection of P-407 and caerulein (CAE). Then, pancreatic tissues from the model animals were subjected to proteome sequencing analysis. The pathological changes and scores of the pancreas, lung, and kidney were determined using hematoxylin-eosin staining. The levels of serum amylase (AMY), triglyceride, and total cholesterol were measured with an automatic blood cell analyzer. Additionally, the serum levels of tumor necrosis factor (TNF)-α, interleukin (IL)-6, and IL-1β were determined by enzyme linked immunosorbent assay. Malonaldehyde (MDA), glutathione (GSH), and Fe2+ were detected in the pancreas. Finally, immunohistochemistry was performed to assess the expression of ferroptosis-related proteins. Proteome sequencing revealed that ferroptosis was involved in the process of HTGP and that NADPH oxidase (NOX) 2 may participate in ferroptosis regulation. Moreover, the levels of serum AMY, TNF-α, IL-6, and IL-1β were significantly increased, MDA and Fe2+ were upregulated, GSH and ferroptosis-related proteins were reduced, and the injury of the pancreas, lung, and kidney were aggravated in the P407 + CAE group compared to CAE and wild type groups (all P < 0.05). Notably, the inhibition of ferroptosis and NOX2 attenuated the pathological damage and the release of TNF-α, IL-6, and IL-1β in the serum of the mice. Ferroptosis was found to have an important role in HTGP and may be considered a potential target for clinical treatment.

Clinical significance of immature reticulocyte fraction (IRF) and reticulocyte maturity indices in differential diagnosis of anemia

Background: Anemia is most common public health problems in developing countries. It is important to know the cause of anemia to treat the patients. The Immature reticulocyte fraction (IRF) value is an early marker for evaluating the regeneration of erythropoiesis. In addition to conventional reticulocyte measurement, the fluorescence method allows the classification of reticulocytes into three maturation stages – LFR, MFR and HFR. Aims: To determine clinical significance of immature reticulocyte fraction (IRF) and reticulocyte maturity indices (LFR, MFR, HFR) in differential diagnosis of anemic patients. Material and Method: IRF and other reticulocyte parameters were measured by automated blood cell analyzer, Horiba Pentra XLR using fluorescence marker that labels RNA and DNA (such as thiazole orange or polymethines), in 124 patients with anemia. The anemia group consist of 50 iron deficiency anemia (IDA) patients, 29 cases of thalassemia trait patients and 45 patients with anemia due to chromic kidney diseases (CKD). In this retrospective and prospective type study, the values of IRF and other parameters were analysed and compared in each group for the duration of one year. Result: Total of 124 cases were evaluated with median age of age of 32.8 years showed that mean value of IRF was low in IDA (0.096±0.058) and in CKD (0.092±0.111) compare to thalassemia (0.116±0.094). The mean values of MFR and HFR were also found to be significantly lower in IDA group and in CKD when compared to the thalassemic group. Values of LFR were higher in IDA and in CKD compare to thalassemia. Conclusions: IRF assesses reticulocyte maturation by the intensity of the staining that reflects the mRNA content. The evaluation of IRF and reticulocyte maturity indices by automated cell counter analyzer seemed to be accurate and clinically useful for the early diagnosis of anemia and the differentiation of IDA and CKD from thalassemia.

Effect of Anti-Oxidative of Ethyl Pyruvate and Taurine on the Red Blood Cell Storage at 4 ℃

To investigate the anti-oxidative effect of ethyl pyruvate (EP) and taurine (TAU) on the quality of red blood cells stored at 4±2 ℃, hemolysis, energy metabolism and lipid peroxidation of the red blood cells in the preservation solution were studied at different intervals. At 4±2 ℃, the deleukocyte red blood cells were stored in the citrate-phosphate-dextrosesaline-adenine-1 (CPDA-1) preservation (control group), preservation solution with EP (EP-AS), and TAU (TAU-AS) for long-term preservation. The enzyme-linked immunoassay and automatic blood cell analyzer were used to detect hemolysis and erythrocyte parameters. Adenine nucleoside triphosphate (ATP), glycerol 2,3-diphosphate (2,3-DPG) and malondialdehyde (MDA) kits were used to test the ATP, 2,3-DPG and MDA concentration. During the preservation, the rate of red blood cell hemolysis in EP-AS and TAU-AS groups were significantly lower than that in CPDA-1 group (P<0.01). The MCV of EP-AS group was increased with the preservation time (r=0.71), while the MCV of the TAU-AS group was significantly lower than that in the other two groups (P<0.05). The concentration of ATP and MDA in EP-AS and TAU-AS groups were significantly higher than that in CPDA-1 group at the 14th day (P<0.01). The concentrations of 2,3-DPG in the EP-AS and TAU-AS groups were significantly higher than that in the CPDA-1 group from the 7th day (P<0.01). EP and TAU can significantly reduce the red blood cell hemolysis rate, inhibit the lipid peroxidation level of red blood cells, and improve the energy metabolism of red blood cells during storage. The mechanism of EP and TAU may be related to their antioxidation and membrane protection effect, so as to improve the red blood cell quality and extend the preservation time.

Frequency of Anemia in Chronic Obstructive Pulmonary Disease (COPD)

Background: Multiple western studies have focused on anemia in COPD and observed that anemia is more prevalent in COPD patients than Polycythemia. Anemia significantly increases the morbidity and deteriorates the functional status of COPD patients. Objectives: The objective of current study was to determine the frequency of anemia in Chronic Obstructive Pulmonary Disease (COPD) patients present in a tertiary care hospital. Study Design: Retrospective Material and Methods: Two hundred COPD patients fulfilling the inclusion criteria were selected for the study. Hemoglobin (Hb) level test was done on all patients. Hb level was calculated by Automated Blood Cell Analyzer and anemia was labeled at Hb&lt;13g/dL Results: The mean of patients’ age was 56.65+ 7.95 years. 181 (90.5%) patients were male and 19 (9.5%) patients were female. The mean Hemoglobin level (Hb) of the patients was 13.46 + 1.45 g/dL. 48 patients (24%) were anemic and 152 patients (76%) were non anemic. Conclusion: The conclusion of the study is there is frequent occurrence of anemia in patients with chronic obstructive pulmonary disease (COPD), as in current study the frequency of anemia was found in 24.0% patients. The Relationship Between the Neutrophil to Lymphocyte Ratio, The Platelet to Lymphocyte Ratio, and Cardiac Syndrome X.

ObjectiveThis study aims to investigate the relationship between the neutrophil to lymphocyte ratio (NLR), the platelet to lymphocyte ratio (PLR), and cardiac syndrome X (CSX).MethodsA total of 102 patients with CSX who were hospitalized in the Cardiology Department of our hospital from December 2018 to December 2020 were enrolled in the CSX group, and 102 subjects who underwent physical examinations during the same period were included in the control group. An automatic blood cell analyzer was adopted to detect the neutrophil count (NC), lymphocyte count (LC), and number of platelets (PLT) in the whole blood of the subjects in both groups, and the NLR and PLR were calculated. Electrocardiography was conducted on the subjects in both groups to detect whether any abnormality existed in the ST segment. The receiver operating curve (ROC) was used to evaluate the diagnostic value of each indicator of CSX, and multivariate logistic regression analysis was adopted for the analysis of the influencing factors.ResultsNo significant differences existed in age, gender, smoking history, or family history of diabetes mellitus, hypertension, and tumors between the two groups (p > 0.05). When compared with the control group, the NC, PLT, NLR, PLR, and rate of abnormality of the ST segment on the electrocardiogram were significantly higher, and the LC was significantly lower in the CSX group (p < 0.05). Multivariate logistic regression analysis showed that the ST-segment abnormality (3.95 [2.10~7.41]; NLR > 2.21, 3.46 [1.87~6.39]; and PLR > 119.77, 3.66 [1.99~6.73]) was a correlated risk factor for the occurrence of CSX (p < 0.05).ConclusionBoth the NLR and PLR in patients with CSX were significantly elevated, and both have a certain predictive value for the occurrence of CSX and are expected to be effective biomarkers for CSX.

Predictive Value of the Peripheral Blood Parameters for Preeclampsia.

Excessive systemic inflammation plays a vital role in pathophysiology of preeclampsia (PE). The aim is to clarify the predictive value of the peripheral blood parameters including white blood cell (WBC), neutrophils, lymphocyte, monocyte, platelet count, mean platelet volume (MPV), plateletcrit, platelet distribution width (PDW), platelet-large cell ratio (PLCR), and the ratio value for PE. This retrospective study enrolled 170 PE patients, 123 healthy control pregnant women, and 122 non-pregnant women. When pregnant women were admitted to the hospital for delivery, peripheral complete blood cell count was detected by an automatic blood cell analyzer. Clinical signs and demographic characteristics were recorded. The receiver operating characteristic (ROC) curve was used to determine the cutoff value and analyze the predictive significances for PE. Furthermore, the risk factors of PE were tested by univariate and stratified analyses. This study showed that WBC, neutrophil count, neutrophil percentage, NLR, NMR, and PLR# were significantly increased in PE patients as compared with pregnant control patients (p < 0.001), whereas lymphocyte percentage, monocyte percentage, and PNR were decreased. In addition, there was no significant difference in the rest of the peripheral blood parameters between women with and without PE. The ROC curve result revealed that WBC and neutrophil count had a higher AUC value than the rest of peripheral blood variables. WBC and neutrophil count are positively correlated MAP. Moreover, the WBC and neutrophil count were indicated as independent risk factors for the development of PE. This study clarifies that peripheral blood parameters of WBC and neutrophil count have good applied value with high sensitivity and specificity in predicting the development of PE and are also independent risk factors for the development of PE.

Taurine Alleviates LPS-Induced Acute Lung Injury by Suppressing TLR-4/NF-κB Pathway.

Taurine has the function of immune regulation, relieving acute and chronic inflammation caused by various agents, and maintaining cell homeostasis. This investigation focused on the protective functions of taurine targeting acute lung injury (ALI) induced by LPS. Sixty male SD rats aged 6-7weeks were segregated at random: blank control group (C group), taurine control group (T group), ALI model group (LPS group), and taurine prevention groups (LPST1, LPST, LPST3 Groups). C group and LPS group were given normal drinking water, while T group and LPST group were given 2% taurine in drinking water. LPST1 group was given 1% taurine in drinking water while. LPST3 group was given 3% taurine in drinking water. On the 14th and 28th day, LPS group and LPST1, LPST, and LPST3 groups were subjected to injection of LPS (25 mg/kg) into the trachea of rats. Serum, peripheral blood, lung tissue, and bronchoalveolar lavage fluid (BALF) were collected at 6h post-LPS injection. The wet/dry ratio (W/D) of lung was measured by hot drying method. The population of white blood cells and the abundance of inflammatory-related cells within peripheral blood were counted by an automatic blood cell analyzer. The population of white blood cells within BALF was counted by a white blood cell counting plate combined with Swiss Giemsa staining, while the proportion of related white blood cells was calculated. BCA reagent was used to determine the protein concentration in BALF. The levels of pro-inflammatory factors (IL-1 β, IL-6, IL-18, TNF - α), anti-inflammation factors (IL-10, IL-4), and taurine within serum and lung tissue were detected by ELISA. Lung structural tissue alterations were observed through HE staining techniques. Myeloperoxidase (MPO) activities within lung tissue were detected through colorimetry. Protein expression levels of TLR4, MyD88, NF-κ Bp65, NF-κ Bp-p65, MCP-1, together with CD68 within lung tissue, were analyzed by Western blot (WB) and immunohistochemistry (IHC). The taurine pretreatment group contained significantly reduced W/D, MPO activity, and the number of inflammatory cells in BALF induced by LPS. In addition, compared with ALI model group, the taurine pretreatment group contained significantly reduced levels of pro-inflammatory factors in lung tissue, increased levels of anti-inflammatory factors, and decreased expression levels of key proteins in TLR-4/NF-κ B pathway. Taurine can protect rats from ALI by inhibiting the activation of neutrophils, macrophages, and TLR-4/NF-κ B signaling pathway.

Study of platelet indices and their role in evaluation of thrombocytopenia.

Thrombocytopenia may result from mechanisms such as marrow hypoplasia, increased destruction of platelets, and splenic sequestration. The gold standard method for discriminating the causes of thrombocytopenia is bone marrow examination, but it is invasive and expensive. Therefore, an alternative method should be introduced as a first-line diagnostic procedure. Of late, the automated blood cell analyzer has made it possible to assess the cause of thrombocytopenia through various machine-derived parameters, known as platelet indices, which include the mean platelet volume (MPV), platelet distribution width (PDW), and plateletcrit (PCT), which are provided as a part of routine complete blood count. The objectives of the present study are to study the variation and effectiveness of platelet indices in establishing the etiology of thrombocytopenia. An observational, prospective, and comparative study was conducted on 134 patients with thrombocytopenia, and 67 cases were taken as the normal group. The study group was classified into two groups: hypo-productive and hyper-destructive. Platelet indices were recorded and compared in the two groups along with the normal group. The mean platelet count (10^3 μL) in the normal, hypo-productive, and hyper-destructive groups was 232.03 ± 74.84, 73.00 ± 36.52, and 68.28 ± 38.24, respectively. The MPV and mean PCT in the normal, hypo-productive, and hyper-destructive groups were 9.46 ± 1.68fL, 8.99 ± 1.49fL, and 11.35 ± 1.35fL and 0.22 ± 0.06%, 0.07 ± 0.04%, and 0.08 ± 0.05%, respectively. The mean PDW in the normal, hypo-productive, and hyper-destructive groups was 15.66 ± 1.76fL, 17.63 ± 1.01fL, and 18.32 ± 1.10fL, respectively. In the present study, platelet indices such as MPV, PCT, and PDW are higher in the hyper-destructive group and may discriminate hyper-destructive from hypo-productive causes of thrombocytopenia.

Correlation between GEM Premier 3000 and Vitros5.1+5600, SYSMEX XN-9000 in Detecting Electrolytes and Red Blood Cell Volume

Objective: To explore the consistency and relevance of the results of the bedside rapid blood gas analyzer GEM premier 3000, the Vitros5.1+5600 biochemical immunoassay analyzer and the SYSMEX XN-9000 automatic blood cell analyzer in the central laboratory detecting serum potassium (K+), serumsodium (Na+), blood glucose (Glu), hemoglobin (Hb) and red blood cell volume (Hct). And to provide a reference for the accurate interpretation of the bedside blood gas analysis report. Method: Usually, ICU patients will be taken arterial blood gas, biochemical and blood samples through the arterial indwelling needle; at the same time patients’ potassium, serum sodium, blood glucose, hemoglobin and red blood cell volume will be detected. This study implemented paired t-test and correlation regression analysis on each group of data, and used the analysis quality requirements (allowable total error) of CLIA’88 proficiency testing program as the criteria for clinical acceptance. Results: The paired t-test showed that the serum potassium, serum sodium and blood glucose detected by GEM premier 3000 and Vitros5.1+5600 were significantly different; and the hemoglobin and red blood cell volume detected by GEM premier 3000 and SYSMEX XN-9000 were significantly different (P +, Na+, Glu, Hb and Hct detected by the two sets of instruments is less than the allowable error of CLIA’88, and the SE of blood Na+ and Hb is less than half of the allowable error of CLIA’88. Conclusion: The test results of GEM premier 3000, the central laboratory Vitros5.1+5600 and SYSMEX XN-9000 have good correlation, but the consistency is not good. The test results of GEM premier 3000 cannot replace the central laboratory.

Performance evaluation of the new platelet measurement channel on the BC-6800 Plus automated hematology analyzer.

To evaluate the performance of new optical platelet measurement channel on the BC-6800 Plus automated blood cell analyzer. The basic PLT count performance of the BC-6800 Plus was evaluated according to the requirements of the Clinical Laboratory and Standards Institute (CLSI) Document H26-A2. In addition, low-PLT-value specimens, red blood cell debris specimens, small red blood cell specimens, and giant PLT specimens were detected with the blood cell analyzer and a flow cytometer. Whole-blood specimens in ethylenediaminetetraacetic acid(EDTA) or sodium citrate anticoagulant tubes from 20 patients with EDTA-dependent PLT aggregation were determined in CDR mode of the analyzer. Blank counting and the carryover contamination rate of PLTs using the BC-6800 Plus both met the technical requirements. For abnormal PLT specimens, PLT-O 8× and PLT-I both exhibited high comparability with flow cytometry. The comparability of PLT-O 8× with flow cytometry was better than that of PLT-I. In EDTA-anticoagulated blood specimens from 20 patients with EDTA-dependent PLT aggregation, the results of PLT-O were significantly higher than those for PLT-I using samples from the same tubes (P<.001). However, the PLT counts were similar between these two methods for sodium citrate-anticoagulated blood specimens (P=.263). The performance of PLT-O 8× in the BC-6800 Plus met the technical requirements. PLT-O 8× exhibited better reproducibility than did PLT-I for low-PLT-value samples. Reexamination of abnormal PLT specimens using PLT-O 8× yielded more accurate results. PLT-O performed significantly better than PLT-I in the detection of EDTA-dependent PLT-aggregation specimens.

Effect of Solanum gilo leaf extract on some haematological indices of albino Wistar rats

Solanum gilo (eggplant) is an edible vegetable whose leaf is reported to possess numerous medicinal properties due to its rich constituents. Against this backdrop, there is possibility that its consumption may have effect on haematological indices. Therefore this research sought to assess the effect of Solanum gilo leaf diet on some haemotological indices of albino wistar rats. About twenty four (24) albino wistar rats weighing between 120-150mg were randomly assigned into four groups of six animals each. Each rat was allowed to drinking water ad labitum. Control group received normal rodent chow while group 2 was fed 5% Solanum gilo diet. Group 3 was fed 10% Solanum gilo diet and group 4 received 20% Solanum gilo diet. The different diets were prepared by mixing different proportions of ground S. gilo leaves with rodent chow. Automated blood cell analyzer (Model PCE 210, Japan) was used for analysis of haematological parameters. Preliminary study on its proximate composition, showed the appreciable presence of crude fat, protein, fibre and carbohydrate. Consumption of S. gilo caused significant improvement in some blood parameters; notably packed cell volume (PCV), white blood cell (WBC) and platelet counts. Hence, eggplant is a nutritious vegetable with healing, the therapeutic and immunity properties. If exploited in human diet has the potentials of improving quality of life.

Detection of nucleated red blood cells using the Mindray BC-6800Plus hematology analyzer: a clinical performance evaluation.

With the development of instrument technology, the functions and detection methods of automatic blood cell analyzers have become more complex. To ensure optimal clinical applicability, it is crucial to select an automatic blood cell analyzer with excellent clinical detection performance. This study evaluated the latest Mindray BC-6800Plus automatic blood cell analyzer and assessed its performance in the detection of nucleated red blood cells (NRBCs). A total of 490 clinical blood samples were used to assess the performance of the instrument, including parameters such as precision, linearity, conformity rate of manual microscopic examination, carryover, and limit of quantitation. The instrument showed a small carryover (≤0.02) and excellent linearity (R2≥0.9986). The reproducibility of the sample tests was satisfactory, and the coefficient of variation (CV) of the test results [0.98-1.72% and 0.62-6.97% for white blood cells (WBCs) and NRBCs, respectively] were significantly lower than that declared by the manufacturer (2.5% and 20% for WBCs and NRBCs, respectively). Thus, the BC-6800Plus satisfies the requirements of clinical testing. Two separate Mindray BC-6800Plus machines were tested and found to be in good agreement with each other and with manual microscopy methods. Furthermore, WBC and NRBC counts were highly consistent with results obtained using the XN-9100 blood analyzer. The Mindray BC-6800Plus is an excellent analyzer that can provide timely and accurate reports for clinical laboratory detection of NRBC.

Evaluation of humoral pattern detection performance of Mindray BC-6000PLUS blood analyzer based on WS/T 662-2020.

Manual microscopic examination is the gold standard of humoral cell count test. However, it has some limitations and cannot fully meet clinical needs. Compared with the manual method, the automatic blood cell analyzer has the advantages of a high degree of automation, minimal error, high speed, high precision, and easy standardization. This study intends to verify the detection performance of the body fluid model of the Mindray BC-6000PLUS automatic hematology analyzer. This study was performed in accordance with the International Committee for Standardization in Haematology (ICSH) Hematology Analyzer Evaluation Guide (version 2014) and the requirements of WS/T662-2020 "Clinical humoral examination technique". The humoral white blood cell-body fluid (WBC-BF), humoral red blood cell-body fluid (RBC-BF), monocyte (MN), polymorphonuclear (PMN) were measured to verify the performance indicators of the instrument, including background counting, intra-batch precision, accuracy, carrying contamination rate, and linear range. Referring to the WS/T514-2017 (Establishment and verification of detection capability for clinical laboratory measurement procedures), the limit of blank (LoB) and limit of detection (LoD) values of WBC-BF and RBC-BF in the humoral mode of the instrument were established. The blank count of WBC-BF and RBC-BF, and contamination of Mindray BC-6000PLUS analyzer were zero; the coefficient of variation (CV) of intra-batch precision at different levels of each item was less than 10%. There was a high correlation between instrument test results and manual microscopic examination results (r>0.95). The linear range of the instrument was wide, and the linear verification parameters was good (R2>0.999). The LoB value and LoD value of WBC-BF established by the instrument were 0×109/L and 0.004×109/L, respectively. The LoB value and LoD value of the RBC-BF established by the instrument were 0×1012/L and 0.004×1012/L, respectively. The lower detection limits of WBC-BF and RBC-BF were set as 0.004×109/L and 0.004×1012/L, respectively. All performance indicators of the Mindray BC-6000PLUS automatic blood analyzer met the requirements of the manufacturer's criteria. This instrument can fulfill the requirement of body fluid sample routine test in clinical practice.
