Recent advances in stereoselective construction of fluorinated quaternary carbon centers from fluorinated compounds.

There are many fluorinated quaternary carbon structural units in pharmaceuticals and bioactive compounds. Organic chemists are interested in the stereoselective synthesis of fluorinated quaternary carbon structural units. Constructing a fluorinated quaternary carbon stereocenter can be achieved directly and efficiently by the asymmetric catalytic reaction of fluorinated compounds as substrates. This approach aims to construct fluorinated quaternary carbon stereocenters while diversifying the types of fluorinated compounds. This review introduces a series of reactions for synthesizing fluorinated quaternary carbon chiral centers through asymmetric organic catalysis and transition metal catalysis, including alkylation, arylation, Mannich, Michael addition, and allylation reactions. This work will contribute to the development of the synthesis of fluorinated quaternary carbon chiral center-containing compounds in the future.

Nickel‐Catalyzed Enantioselective Synthesis of Dienyl Sulfoxide

AbstractSulfoxides are widely used in the pharmaceutical industry and as ligands in asymmetric catalysis. However, the efficient asymmetric synthesis of this structural motif remains limited. In this study, we disclosed a Ni‐catalyzed enantioconvergent reaction that utilizes both racemic allenyl carbonates and β‐sulfinyl esters. Our method employs cheap and more sustainable Ni(II) as a precatalyst and successfully overcomes the challenging poisoning effect and instability of sulfenate generated in situ. This enables the synthesis of a series of dienyl sulfoxides with enantioselectivity of up to 98 % ee. The product exhibits tremendous potential in various applications, including diastereoselective Diels–Alder reactions, coordination with transition metals, and incorporation into medicinal compounds, among others. Using a combination of experimental and computational methods, we have uncovered an interesting associated outersphere mechanism that contrasts with conventional mechanisms commonly observed in asymmetric transition metal catalysis.

Interrogating the Crucial Interactions at Play in the Chiral Cation-Directed Enantioselective Borylation of Arenes.

Rendering a common ligand scaffold anionic and then pairing it with a chiral cation represents an alternative strategy for developing enantioselective versions of challenging transformations, as has been recently demonstrated in the enantioselective borylation of arenes using a quinine-derived chiral cation. A significant barrier to the further generalization of this approach is the lack of understanding of the specific interactions involved between the cation, ligand, and substrate, given the complexity of the system. We have embarked on a detailed computational study probing the mechanism, the key noncovalent interactions involved, and potential origin of selectivity for the desymmetrizing borylation of two distinct classes of substrate. We describe a deconstructive, stepwise approach to tackling this complex challenge, which involves building up a detailed understanding of the pairwise components of the nominally three component system before combining together into the full 263-atom reactive complex. This approach has revealed substantial differences in the noncovalent interactions occurring at the stereodetermining transition state for C-H oxidative addition to iridium for the two substrate classes. Each substrate engages in a unique mixture of diverse interactions, a testament to the rich and privileged structure of the cinchona alkaloid-derived chiral cations. Throughout the study, experimental support is provided, and this culminates in the discovery that prochiral phosphine oxide substrates, lacking hydrogen bond donor functionality, can also give very encouraging levels of enantioselectivity, potentially through direct interactions with the chiral cation. We envisage that the findings in this study will spur further developments in using chiral cations as controllers in asymmetric transition-metal catalysis.

Asymmetric Allylic Substitution-Isomerization for the Modular Synthesis of Axially Chiral N-Vinylquinazolinones.

Axially chiral N-substituted quinazolinones are important bioactive molecules, which are presented in many synthetic drugs. However, most strategies toward their atroposelective synthesis are mainly limited to the axially chiral arylquinazolinone frameworks. The development of modular synthetic methods to access diverse quinazolinone-based atropisomers remains scarce and challenging. Herein, we report the regio- and atroposelective synthesis of axially chiral N-vinylquinazolinones via the strategy of asymmetric allylic substitution-isomerization. The catalysis system utilized both asymmetric transition-metal catalysis and organocatalysis to efficiently afford trisubstituted or tetrasubstituted N-vinylquinazolinone atropisomers, respectively. With the meticulous design of β-substituted allylic substrates, configurationally stable Z- and E-tetrasubstituted axially chiral N-vinylquinazolinones were obtained in good yields and high enantioselectivities.

Facile Synthesis of Enantiomerically Pure P-Chiral 1-Alkoxy-2,3-dihydrophospholes via Nucleophilic P-N Bond Cleavage of a 1-Phospha-2-azanorbornene.

The reactive P-N bond in a racemic mixture of endo-1-phospha-2-azanorbornene (PAN) (RP /SP )-endo-1 is readily cleaved with enantiomerically pure lithium alkoxides followed by protonation to afford diastereomeric mixtures of P-chiral 1-alkoxy-2,3-dihydrophosphole derivatives. The isolation of these compounds is rather challenging due to the reversibility of the reaction (elimination of alcohols). However, methylation of the sulfonamide moiety of the intermediate lithium salts and sulfur protection of the phosphorus atom prevent the elimination reaction. The resulting air-stable P-chiral diastereomeric 1-alkoxy-2,3-dihydrophosphole sulfide mixtures can be readily isolated and fully characterized. The diastereomers can be separated by crystallization. The 1-alkoxy-2,3-dihydrophosphole sulfides are readily reduced with Raney nickel to afford phosphorus(III) P-stereogenic 1-alkoxy-2,3-dihydrophospholes with potential use in asymmetric homogeneous transition metal catalysis.

Metal Stereogenicity in Asymmetric Transition Metal Catalysis.

Chiral transition metal catalysts represent a powerful and economic tool for implementing stereocenters in organic synthesis, with the metal center providing a strong chemical activation upon its interaction with substrates or reagents, while the overall chirality of the metal complex achieves the desired stereoselectivity. Often, the overall chiral topology of the metal complex implements a stereogenic metal center, which is then involved in the origin of the asymmetric induction. This review provides a comprehensive survey of reported chiral transition metal catalysts in which the metal formally constitutes a stereocenter. A stereogenic metal center goes along with an overall chiral topology of the metal complex, regardless of whether the ligands are chiral or achiral. Implications for the catalyst design and mechanism of asymmetric induction are discussed for half-sandwich, tetracoordinated, pentacoordinated, and hexacoordinated chiral transition metal complexes containing a stereogenic metal center. The review distinguishes between chiral metal catalysts originating from the coordination to chiral ligands and those which are solely composed of optically inactive ligands (achiral or rapidly interconverting enantiomers) prior to complexation (dubbed "chiral-at-metal" catalysts).

Photoinduced Cobalt-Catalyzed Desymmetrization of Dialdehydes to Access Axial Chirality

Enantioselective metallaphotoredox catalysis, which combines photoredox catalysis and asymmetric transition-metal catalysis, has become an effective approach to achieve stereoconvergence under mild conditions. Although many impressive synthetic approaches have been developed to access central chirality, the construction of axial chirality by metallaphotoredox catalysis still remains underexplored. Herein, we report two visible light-induced cobalt-catalyzed asymmetric reductive couplings of biaryl dialdehydes to synthesize axially chiral aldehydes (60 examples, up to 98% yield, >19:1 dr, and >99% ee). This protocol shows good functional group tolerance, broad substrate scope, and excellent diastereo- and enantioselectivity.

Enantioselective transition-metal catalysis via an anion-binding approach.

Asymmetric transition-metal catalysis represents a powerful strategy for accessing enantiomerically enriched molecules1-3. The classical strategy for inducing enantioselectivity with transition-metal catalysts relies on direct complexation of chiral ligands to produce a sterically constrained reactive metal site that allows formation of the major product enantiomer while effectively inhibiting the pathway to the minor enantiomer through steric repulsion4. The chiral-ligand strategy has proven applicable to a wide variety of highly enantioselective transition-metal-catalysed reactions, but important scenarios exist that impose limits to its successful adaptation. Here, we report a new approach for inducing enantioselectivity in transition-metal-catalysed reactions that relies on neutral hydrogen-bond donors (HBDs)5,6 that bind anions of cationic transition-metal complexes to achieve enantiocontrol and rate enhancement through ion pairing together with other non-covalent interactions7-9. A cooperative anion-binding effect of a chiral bis-thiourea HBD is demonstrated to lead to high enantioselectivity (up to 99% enantiomeric excess) in intramolecular ruthenium-catalysed propargylic substitution reactions10. Experimental and computational mechanistic studies show the attractive interactions between electron-deficient arene components of the HBD and the metal complex that underlie enantioinduction and the acceleration effect.

Biocatalytic Enantioselective Synthesis of Atropisomers.

Atropisomeric compounds are found extensively as natural products, as ligands for asymmetric transition-metal catalysis, and increasingly as bioactive and pharmaceutically relevant targets. Their enantioselective synthesis is therefore an important ongoing research target. While a vast majority of known atropisomeric structures are (hetero)biaryls, which display hindered rotation around a C-C single bond, our group's long-standing interest in the control of molecular conformation has led to the identification and stereoselective preparation of a variety of other classes of "nonbiaryl" atropisomeric compounds displaying restricted rotation around C-C, C-N, C-O, and C-S single bonds.Biocatalytic transformations are finding increasing application in both academic and industrial contexts as a result of a significant broadening of the range of biocatalytic reactions and sources of enzymes available to the synthetic chemist. In this Account, we summarize the main biocatalytic strategies currently available for the asymmetric synthesis of biaryl, heterobiaryl, and nonbiaryl atropisomers. As is the case with more traditional synthetic approaches to these compounds, most biocatalytic methodologies for the construction of enantioenriched atropisomers follow one of two distinct strategies. The first of these is the direct asymmetric construction of atropisomeric bonds. Synthetically applicable biocatalytic methodologies for this type of transformation are limited, despite the extensive research into the biosynthesis of (hetero)biaryls by oxidative homocoupling or cross-coupling of electron-rich arenes. The second of these is the asymmetric transformation of a molecule in which the bond that will form the axis already exists, and this approach represents the majority of biocatalytic strategies available to the synthetic organic chemist. This strategy encompasses a variety of stereoselective techniques including kinetic resolution (KR), desymmetrization, dynamic kinetic resolution (DKR), and dynamic kinetic asymmetric transformation (DYKAT).Nondynamic kinetic resolution (KR) of conformationally stable biaryl derivatives has provided the earliest and most numerous examples of synthetically useful methodologies for the enantioselective preparation of atropisomeric compounds. Lipases (i.e., enzymes that mediate the formation or hydrolysis of esters) are particularly effective and have attracted broad attention. This success has led researchers to broaden the scope of lipase-mediated transformations to desymmetrization reactions, in addition to a limited number of DKR and DYKAT examples. By contrast, our group has used redox enzymes, including an engineered galactose oxidase (GOase) and commercially available ketoreductases (KREDs), to desymmetrize prochiral atropisomeric diaryl ether and biaryl derivatives. Building on this experience and our long-standing interest in dynamic conformational processes, we later harnessed intramolecular noncovalent interactions to facilitate bond rotation at ambient temperatures, which allowed the development of the efficient DKR of heterobiaryl aldehydes using KREDs. With this Account we provide an overview of the current and prospective biocatalytic strategies available to the synthetic organic chemist for the enantioselective preparation of atropisomeric molecules.

Electrolyte Induced Solvent Cage Effects for Enantioselective Electrosynthesis

Electrochemistry provides a tunable, regioselective, and green alternative to traditional synthetic organic methods, and access to reactive intermediates. Problematically, electrochemical redox events often go through planar radical intermediates, thus destroying enantioselectivity. As such, researchers have sought the “chiral electron”, a general methodology to impart enantioselectivity to electroorganic reactions. One strategy has been asymmetric transition metal catalysis while replacing the typical stochiometric redox reagent needed with electricity, thus providing a chiral pathway for elecoorganic reactions. However, the general physical parameters that govern enantioselectivity at electrochemical interfaces, remains poorly understood.Here, we focus on the effects of supporting electrolyte in synthetic organic electrochemistry, specifically its role in enantioselective reactions. Cyclic voltammetry provides a tool to investigate the mechanistic consequences of changes in electrolyte. Using the model reaction of enantioselective carboxylation with a cobalt catalyst, we observe changes in mechanism as the electrolyte size is varied. Specifically, electrolyte identity effects the lifetime of the chiral Co-alkyl intermediate. These fundamental electroanalytical studies provide a sound mechanistic basis for the origin of enantioselectivity in electroorganic reactions. In summary, these results of a general interest as a strategy to tune and improve enantioselectivity in electrochemical transformations.

Birds of a Feather—Asymmetric Organocatalysis Meets Asymmetric Transition Metal Catalysis

The results of recent studies on the mechanism of stereoinduction in asymmetric hydrogenation catalyzed by transition metal complexes suggest that hydrogen activation by metal atoms and the generation of enantioselectivity by organic ligands proceed independently. Hence, these reactions can be considered as variants of a cooperative organocatalytic reaction. This conclusion opens a broader view on rational catalyst design, suggesting that the structural ideas from different fields can be exploited reciprocally.

Macrocylases as synthetic tools for ligand synthesis: enzymatic synthesis of cyclic peptides containing metal-binding amino acids.

Improving the sustainability of synthesis is a major goal in green chemistry, which has been greatly aided by the development of asymmetric transition metal catalysis. Recent advances in asymmetric catalysis show that the ability to control the coordination sphere of substrates can lead to improvements in enantioselectivity and activity, in a manner resembling the operation of enzymes. Peptides can be used to mimic enzyme structures and their secondary interactions and they are easily accessible through solid-phase peptide synthesis. Despite this, cyclic peptides remain underexplored as chiral ligands for catalysis due to synthetic complications upon macrocyclization. Here, we show that the solid-phase synthesis of peptides containing metal-binding amino acids, bipyridylalanine (1), phenyl pyridylalanine (2) and N,N-dimethylhistidine (3) can be combined with peptide macrocylization using peptide cyclase 1 (PCY1) to yield cyclic peptides under mild conditions. High conversions of the linear peptides were observed (approx. 90%) and the Cu-bound cyclo(FSAS(1)SSKP) was shown to be a competent catalyst in the Friedel-Crafts/conjugate addition of indole. This study shows that PCY1 can tolerate peptides containing amino acids with classic inorganic and organometallic ligands as side chains, opening the door to the streamlined and efficient development of cyclic peptides as metal ligands.

Recent Advances in Asymmetric Iron Catalysis.

Asymmetric transition-metal catalysis represents a fascinating challenge in the field of organic chemistry research. Since seminal advances in the late 60s, which were finally recognized by the Nobel Prize to Noyori, Sharpless and Knowles in 2001, the scientific community explored several approaches to emulate nature in producing chiral organic molecules. In a scenario that has been for a long time dominated by the use of late-transition metals (TM) catalysts, the use of 3d-TMs and particularly iron has found, recently, a widespread application. Indeed, the low toxicity and the earth-abundancy of iron, along with its chemical versatility, allowed for the development of unprecedented and more sustainable catalytic transformations. While several competent reviews tried to provide a complete picture of the astounding advances achieved in this area, within this review we aimed to survey the latest achievements and new concepts brought in the field of enantioselective iron-catalyzed transformations.

Complementing Pyridine-2,6-bis(oxazoline) with Cyclometalated N-Heterocyclic Carbene for Asymmetric Ruthenium Catalysis.

A strategy for expanding the utility of chiral pyridine‐2,6‐bis(oxazoline) (pybox) ligands for asymmetric transition metal catalysis is introduced by adding a bidentate ligand to modulate the electronic properties and asymmetric induction. Specifically, a ruthenium(II) pybox fragment is combined with a cyclometalated N‐heterocyclic carbene (NHC) ligand to generate catalysts for enantioselective transition metal nitrenoid chemistry, including ring contraction to chiral 2H‐azirines (up to 97 % ee with 2000 TON) and enantioselective C(sp3)−H aminations (up to 97 % ee with 50 TON).

Nickel/Photo-Cocatalyzed Asymmetric Acyl-Carbamoylation of Alkenes.

An unprecedented asymmetric acyl-carbamoylation of pendant alkenes tethered on aryl carbamic chlorides with both aliphatic and aromatic aldehydes has been developed via the cooperative catalysis of a chiral nickel-PHOX complex and tetrabutylammonium decatungstate. This reaction represents the first example of merging hydrogen-atom-transfer photochemistry and asymmetric transition metal catalysis in difunctionalization of alkenes. Using this protocol, a variety of oxindoles bearing a challenging quaternary stereogenic center are furnished under mild conditions in highly enantioselective manner.

Recent Developments in Highly Stereoselective Michael Addition Reactions Catalyzed by Metal Complexes

Achieving high enantioselectivity and diastereoselectivity simultaneously­ is a rather challenging task for asymmetric catalytic synthesis­. Thanks to the rapid development of asymmetric transition-metal catalysis, significant progress has been made during recent years in achieving highly enantio- and diastereoselective conjugate addition reactions with a diverse combination of Michael donors and acceptors. This short review surveys the advances in transition-metal-catalyzed asymmetric diastereoselective Michael addition including diastereodivergent catalysis developed between 2015 and 2019. Enantioselective hydrophosphinylation of 1-alkenylphosphine oxides catalyzed by chiral strong Brønsted base.

A catalytic enantioselective addition of diarylphosphine oxides to 1-alkenyl(diaryl)phosphine oxides was achieved by using a chiral ureate as a chiral strong Brønsted base catalyst. The reaction followed by the reduction of phosphine oxide moieties provided chiral 1,2-diphosphinoalkanes, which are a family of useful chiral ligands for asymmetric transition metal catalysis.

Chiral-at-Iron Catalyst: Expanding the Chemical Space for Asymmetric Earth-Abundant Metal Catalysis.

A new class of chiral iron catalysts is introduced that contains exclusively achiral ligands with the overall chirality being the result of a stereogenic iron center. Specifically, iron(II) is cis-coordinated to two N-(2-pyridyl)-substituted N-heterocyclic carbene (PyNHC) ligands in a bidentate fashion in addition to two monodentate acetonitriles, and the dicationic complex is complemented by two hexafluorophosphate ions. Depending on the helical twist of the PyNHC ligands, the metal center adopts either a Λ or Δ absolute configuration. Importantly, the two PyNHC ligands are constitutionally and configurationally inert, while the two acetonitriles are labile and allow asymmetric transition metal catalysis. This is demonstrated with an enantioselective Cannizzaro reaction (96% yield, 88% ee) and an asymmetric Nazarov cyclization (89% yield, >20:1 dr, 83% ee).
