Total Synthesis, Revised Structure, and Cytotoxic Activities of Iubol

To unambiguously establish the stereostructure of the marine cytotoxic bromotriterpenoid iubol, a member of the thyrsiferol family, asymmetric chemical synthesis has been carried out. The synthesis features a one-pot process for the tetrahydropyranyl D ring construction through a stoichiometric Sharpless asymmetric epoxidation of allylic alcohols followed by titanium chelation-assisted 6-exo oxacyclization. In this paper, we report the asymmetric total synthesis of iubol, revision of the proposed structure to the C22-epimer, and preliminary cytotoxic activities of synthetic compounds against some tumor cells.

Sharpless Asymmetric Epoxidation: Applications in the Synthesis of Bioactive Natural Products

This review discusses an important synthetic tool proposed by K.B. Sharpless in 1980, known as the Sharpless asymmetric epoxidation of allylic alcohols, and examines its use in the total synthesis of representative exponents of biologically active natural products. Focus is given to the synthesis of simple to highly complex secondary metabolites, including lactones, amino acids, diterpenes, and macrolides. The Sharpless approach involves the use of a catalyst, titanium tetra isopropoxide [Ti(OiPr)4], dialkyl tartrate as a chiral ligand, and tert-butyl hydroperoxide (TBHP) as an oxidizing agent. The method allows converting allylic alcohols to epoxides, which are chiral building blocks and versatile intermediates in the synthesis of natural products. The biological and synthetic importance of epoxides lies in the susceptibility of the three-membered heterocyclic ring to stereoand regioselective opening by nucleophilic or acidic reagents, providing oxygen adducts.

Step-by-Step Assembly of 2D Confined Chiral Space Endowing Achiral Clusters with Asymmetric Catalytic Activity for Epoxidation of Allylic Alcohols.

Endowing achiral polyoxometalates (POMs) with asymmetric catalytic properties is always an intriguing but challenging topic because of their high catalytic activities yet highly symmetrical molecular structures. In this work, a novel strategy was proposed to fabricate a series of two-dimensional chiral POM catalysts. Following the steps of exfoliation, covalent modification, and reassembly, the achiral POMs were orderly confined into the chiral interstitial domains of chiral ionic liquid (CIL)-modified layered double hydroxide materials with a decreased molecular symmetry. The chirality of POM molecules was induced by the l- or d-pyrrolidine-type CILs, and their asymmetric catalytic activity was enhanced by the confinement effect. Compared with the reported chiral POM-based catalysts [e.g., 8 turnover frequency (TOF) and 79% enantiomeric excess (ee) for chiral POM-based metal-organic frameworks], the constructed chiral POM catalysts showed a significantly higher TOF and enantioselectivity (up to 240 TOF and 93% ee) for the asymmetric epoxidation of allylic alcohols. The facilitated mass transfer in the IL channels and the increased binding efficiency between the chiral catalytic sites and reactants render this strategy highly promising for constructing efficient chiral catalysts from the catalytically active while achiral building blocks.

An immobilized vanadium-binaphthylbishydroxamic acid complex as a reusable catalyst for the asymmetric epoxidation of allylic alcohols

An immobilized polymer-supported vanadium-binaphthylbishydroxamic acid (PS-VBHA) has been developed as an efficient, reusable catalyst for the asymmetric epoxidation of allylic alcohols. This PS-VBHA catalyst shows comparable catalytic performance to that of the parent V-BBHA catalyst and can be reused five times without significant loss of enantioselectivity.

Substrate Directed Asymmetric Reactions.

Historically, reagent controlled reactions (mechanism controlled reactions) have played a significant role in the asymmetric synthesis of complex structures. In contrast, today's asymmetric synthesis is greatly dependent on substrate directed approaches. In this approach, a polar functional group, namely, a "directing group", in the vicinity of the reactive site inside the substrate has been documented to preassociate with the chiral catalyst, which exerts stereodirecting influence by directing the reacting partner toward one of the enantiotopic faces of the reaction center. Those reactions usually proceed through exceptionally ordered transition states and result in extraordinary levels of stereoselection. Within the last four decades, the substrate directed approach has become an indispensible tool for the preparation of complex chiral frameworks starting directly from relatively simple achiral substrate molecules via asymmetric induction or various resolution techniques or both. Likewise, the substrate directed approach has been applied to functionalize enantiopure substrates bearing pre-exisiting stereocenters into complex structures as a single diastereomer. A classical example is Sharpless asymmetric epoxidation of allylic alcohols in which the free hydroxy function acts as an active anchor to a dimeric Ti-catalyst that controls the stereochemical outcome of the epoxidation process by transferring the oxidant enantioselectively. The principal aim of the present review is to give a general overview of substrate directed asymmetric transformations, a topic that has not yet been documented in the form of a concise review of recently developed approaches. Due to the large number of related applications, only recent advances that have been documented within the last two decades have been reviewed. Furthermore, in the current review, we have mainly highlighted asymmetric reactions that are controlled by abundant and frequently used directing groups such as hydroxy, amide, and sulfonamide groups. In addition, selected examples of a few important substrate-directed chemo-, regio-, and diastereoselective reactions have also been included in this review.

Total synthesis of myxol and deoxymyxol stereoisomers and their application to determining the absolute configurations of the natural products

The total synthesis of myxol stereoisomers 1a,b and deoxymyxol (plectaniaxanthin) stereoisomers 2a,b was accomplished by Wittig reaction of (S)- and (R)-C10-phosphonium salts 8a,b bearing a silyl-protected 1,2-dihydroxy-ψ end group with C30-apocarotenals 6 and 7. The phosphonium salts 8a,b were derived from aldehydes 11a,b possessing a cyclopentylidene ketal moiety, prepared via Sharpless asymmetric epoxidation of allylic alcohol 12 followed by regioselective cleavage of the oxirane ring. We established an analytical HPLC method using a chiral column to separate stereoisomers 1a,b and 2a,b and thus determined the absolute configurations of the natural products. The HPLC analyses established that both myxol and deoxymyxol isolated from bacteria have the 2′S-configuration.

Synthesis of the C8'-epimeric thymine pyranosyl amino acid core of amipurimycin.

The C8’-epimeric pyranosyl amino acid core 2 of amipurimycin was synthesized from D-glucose derived alcohol 3 in 13 steps and 14% overall yield. Thus, the Sharpless asymmetric epoxidation of allyl alcohol 7 followed by trimethyl borate mediated regio-selective oxirane ring opening with azide, afforded azido diol 10. The acid-catalyzed 1,2-acetonide ring opening in 10 concomitantly led to the formation of the pyranose ring skeleton to give 2,7-dioxabicyclo[3.2.1]octane 12. Functional group manipulation in 12 gave 21 that on stereoselective β-glycosylation afforded the pyranosyl thymine nucleoside 2 – a core of amipurimycin.

Validity of Inorganic Nanosheets as an Efficient Planar Substituent To Enhance the Enantioselectivity of Transition‐Metal‐Catalyzed Asymmetric Synthesis

Effectively enhancing the enantioselectivity is a persistent challenge in heterogeneous asymmetric catalysis. Here, the validity of a layered double hydroxides (LDH) nanosheet as an efficient planar substituent to enhance the enantioselectivity has been investigated theoretically; first in vanadium-catalyzed asymmetric epoxidation of allylic alcohols, and then in zinc-catalyzed direct asymmetric aldol addition. The computational predication is further confirmed experimentally in zinc-catalyzed direct asymmetric aldol addition by controlling the location of catalytic sites.

Nanosheet‐Enhanced Enantioselectivity in the Vanadium‐Catalyzed Asymmetric Epoxidation of Allylic Alcohols

The use of suitable chiral ligands is an efficient means of producing highly enantioselective transition-metal catalysts. Herein, we report a facile, economic, and effective strategy for the design of chiral ligands that demonstrate enhanced enantioselectivity and catalytic efficacy. Our simple strategy employs naturally occurring or synthetic inorganic nanosheets as huge and rigid planar substituents for, but not limited to, naturally available α-amino-acid ligands; these ligands were successfully used in the vanadium-catalyzed asymmetric epoxidation of allylic alcohols. The crucial role of the inorganic nanosheets as planar substituents in improving the enantioselectivity of the reaction was clearly revealed by relating the observed enantiomeric excess with the distribution of the catalytic centers and the accessibility of the substrate molecules to the catalytic sites. DFT calculations indicated that the LDH layer improved the enantioselectivity by influencing the formation and stability of the catalytic transition states, both in terms of steric resistance and H-bonding interactions.

An efficient enantioselective synthesis of florfenicol via a vanadium-catalyzed asymmetric epoxidation

An efficient and highly enantioselective synthesis of florfenicol 1 was achieved with 37% overall yield starting from commercially available 4-methylthiobenzaldehyde. A key feature of the synthesis is the vanadium-catalyzed asymmetric epoxidation of allylic alcohol 5 with aq tert-butyl hydroperoxide to form (2 S,3 S)-epoxide 6.

Abstract A highly stereoselective synthesis of [(S)-coriolic acid] ( 1 ) and first total synthesis of (S)-15,16-didehydrocoriolic acid ( 2 ) by a Pd o -CuI catalysed coupling of (S )-halovinylalcohol with acetylenic moiety is described. The required optically pure chlorovinylalcohols or bromovinylalcohols have been prepared from epoxy chlorides, obtained by Sharpless asymmetric epoxidation of allylic alcohols.

Oxidation Catalysis of Nb(salan) Complexes: Asymmetric Epoxidation of Allylic Alcohols Using Aqueous Hydrogen Peroxide as an Oxidant

Several optically active Nb(salan) complexes were synthesized, and their oxidation catalysis was examined. A dimeric mu-oxo Nb(salan) complex that was prepared from Nb(OiPr)(5) and a salan ligand was found to catalyze the asymmetric epoxidation of allylic alcohols using a urea-hydrogen peroxide adduct as an oxidant with good enantioselectivity. However, subsequent studies of the time course of this epoxidation and of the relationship between the ee of the ligand and the ee of the product indicated that the mu-oxo dimer dissociates into a monomeric species prior to epoxidation. Moreover, monomeric Nb(salan) complexes prepared in situ from Nb(OiPr)(5) and salan ligands followed by water treatment were found to catalyze the epoxidation of allylic alcohols better using aqueous hydrogen peroxide in CHCl(3)/brine or toluene/brine solution with high enantioselectivity ranging from 83 to 95% ee, except for the reaction of cinnamyl alcohol that showed a moderate ee of 74%. This is the first example of the highly enantioselective epoxidation of allylic alcohols using aqueous hydrogen peroxide as an oxidant.

An enantioselective synthesis of the naturally occurring lipid, guggultetrol, is described with an overall yield of 24% starting from commercially available l-pentadecanol in ten linear steps. The key chiral-inducing steps include a Sharpless asymmetric epoxidation of allylic alcohol and a dihydroxylation of an α,β-unsaturated ester.

Modification of chiral dimethyl tartrate through transesterification: Immobilization on POSS and enantioselectivity reversal in sharpless asymmetric epoxidation

Modification of dimethyl tartrate has been investigated through transesterification with aminoalcohols to provide reactive functionalities for the covalent bonding of chiral tartrate to polyhedral oligomeric silsesquioxanes. The transesterification of dimethyl tartrate has been widely studied using different catalytic systems and reaction conditions. Through the proper selection of both the catalytic system and the reaction conditions, it is possible to achieve monosubstituted or bis-substituted tartrate derivatives as sole products. All the intermediate chiral tartrate-derived ligands were successfully used in the homogeneous enantioselective epoxidation of allylic alcohols providing moderate enantiomeric excess over the products. Attached amine groups have been used to support the modified tartrate ligands on to a haloaryl-functionalized silsesquioxane moiety. This final chiral tartrate ligand displays reverse enantioselectivity in the asymmetric epoxidation of allylic alcohols with regard to the starting dimethyl tartrate ligand, both molecules having the same chiral sign. However, the POSS-containing ligand can be easily recovered in almost quantitative yield and reused in asymmetric epoxidation reactions. In addition, recovered silsesquioxane-pendant ligand, though displaying decreasing catalytic activity in recycling epoxidation tests, showed very stable enantioselective behavior.

HPLC with polysaccharide chiral stationary phase in polar‐organic phase mode: Application to the asymmetric epoxidation of allylic alcohols

A simple and rapid HPLC method using a polysaccharide-based chiral stationary phase (Chiralpak AD-H) in polar-organic phase mode has been developed for direct resolution of glycidyl nitrobenzoate (GNB) and 2-methyl glycidyl nitrobenzoate (MGNB) enantiomers. ACN and methanol were used as mobile phase and the effects of the addition of ethanol and 2-propanol as organic modifier in the mobile phase, flow rate and the column temperature were tested. The optimized conditions were: methanol/ethanol (80:20) at a flow rate of 0.9 mL/min and 40 degrees C. Analysis time was < or = 13 min and the chiral resolution was > or = 2. The method was validated and resulted to be selective, precise and accurate. The method was found to be linear in 2-300 microg/mL range (R(2) >0.999) with an LOD nearly 0.5 microg/mL for four enantiomers. GNB and MGNB enantiomers were obtained by asymmetric epoxidation of allyl alcohol and 2-methyl allyl alcohol, respectively, using chiral titanium-tartrate complexes as catalyst and dichloromethane as solvent after in situ derivatization of the intermediate glycidols derivatives. The quite simple and rapid validated method was applied successfully for direct determination of the enantiomeric excess (> or = 90%) and yield obtained in real samples of asymmetric epoxidation of allylic alcohols without further purification, workup or solvent removal. The method provides a useful and value-added tool for controlling the enantiomeric purity of the synthesized epoxides.
