Interactions between Root Hair Development and Arbuscular Mycorrhizal Fungal Colonization in Trifoliate Orange Seedlings in Response to P Levels

Both arbuscular mycorrhizal (AM) fungi and root hairs are crucial in facilitating plant uptake of phosphorus (P), while it is unclear whether and how they respond to varying P supplies. In order to explore how AM fungal colonization and root hair development are affected by substrate P supply, trifoliate orange (Poncirus trifoliata) seedlings were inoculated with AM fungus Rhizophagus intraradices and grown under low, moderate, and high P conditions; then, root hair morphological features and AM fungal colonization were measured. Following 120 days of AM fungal inoculation, root hair density, root hair length, AM fungal colonization rate, arbuscule colonization rate, and AM fungal colonization frequency all increased significantly under P-deficient conditions but decreased under high P conditions. Moreover, the colonization of AM fungi had a major impact on root hair formation by altering the expression of related genes and the growth of epidermal cells. The effect of AM fungi was dependent on P supply levels, as evidenced by the fact that root hair density and length increased at high P levels but decreased at low P levels. As a result, root hairs may serve as a preferential site for AM fungal colonization, and their morphology could influence the early stage of AM symbiosis establishment.

Computer vision models enable mixed linear modeling to predict arbuscular mycorrhizal fungal colonization using fungal morphology

The presence of Arbuscular Mycorrhizal Fungi (AMF) in vascular land plant roots is one of the most ancient of symbioses supporting nitrogen and phosphorus exchange for photosynthetically derived carbon. Here we provide a multi-scale modeling approach to predict AMF colonization of a worldwide crop from a Recombinant Inbred Line (RIL) population derived from Sorghum bicolor and S. propinquum. The high-throughput phenotyping methods of fungal structures here rely on a Mask Region-based Convolutional Neural Network (Mask R-CNN) in computer vision for pixel-wise fungal structure segmentations and mixed linear models to explore the relations of AMF colonization, root niche, and fungal structure allocation. Models proposed capture over 95% of the variation in AMF colonization as a function of root niche and relative abundance of fungal structures in each plant. Arbuscule allocation is a significant predictor of AMF colonization among sibling plants. Arbuscules and extraradical hyphae implicated in nutrient exchange predict highest AMF colonization in the top root section. Our work demonstrates that deep learning can be used by the community for the high-throughput phenotyping of AMF in plant roots. Mixed linear modeling provides a framework for testing hypotheses about AMF colonization phenotypes as a function of root niche and fungal structure allocations.

Prairie restoration promotes the abundance and diversity of mutualistic arbuscular mycorrhizal fungi.

Predicting how biological communities assemble in restored ecosystems can assist in conservation efforts, but most research has focused on plants, with relatively little attention paid to soil microbial organisms that plants interact with. Arbuscular mycorrhizal (AM) fungi are an ecologically significant functional group of soil microbes that form mutualistic symbioses with plants and could therefore respond positively to plant community restoration. To evaluate the effects of plant community restoration on AM fungi, we compared AM fungal abundance, species richness, and community composition of five annually cultivated, conventionally managed agricultural fields with paired adjacent retired agricultural fields that had undergone prairie restoration 5-9 years prior to sampling. We hypothesized that restoration stimulates AM fungal abundance and species richness, particularly for disturbance-sensitive taxa, and that gains of new taxa would not displace AM fungal species present prior to restoration due to legacy effects. AM fungal abundance was quantified by measuring soil spore density and root colonization. AM fungal species richness and community composition were determined in soils and plant roots using DNA high-throughput sequencing. Soil spore density was 2.3 times higher in restored prairies compared to agricultural fields, but AM fungal root colonization did not differ between land use types. AM fungal species richness was 2.7 and 1.4 times higher in restored prairies versus agricultural fields for soil and roots, respectively. The abundance of Glomeraceae, a disturbance-tolerant family, decreased by 25% from agricultural to restored prairie soils but did not differ in plant roots. The abundance of Claroideoglomeraceae and Diversisporaceae, both disturbance-sensitive families, was 4.6 and 3.2 times higher in restored prairie versus agricultural soils, respectively. Species turnover was higher than expected relative to a null model, indicating that AM fungal species were gained by replacement. Our findings demonstrate that restoration can promote a relatively rapid increase in the abundance and diversity of soil microbial communities that had been degraded by decades of intensive land use, and community compositional change can be predicted by the disturbance tolerance of soil microbial taxonomic and functional groups.

Effects of Claroideoglomus etunicatum Fungus on the Growth Parameters of Maize (Zea mays L.) Plants under Boron Toxicity and Salt Stress

Soil salinity is an emerging phenomenon threatening arid and semiarid areas due to changing climatic events. Salinity, in combination with other elemental contaminants, can often harm crop performance and productivity. This experiment was conducted to evaluate the mitigating effect of Claroideoglomus etunicatum, an arbuscular mycorrhizal fungus (AMF), on combined boron (B) toxicity and salt stress symptoms in maize plants. After the stress and AMF treatments, plants were subjected to a wide range of analyses, such as AMF colonization rates, ion leakage, plant biomass, and concentration of B, phosphorus, sodium, potassium, iron, zinc, copper, and manganese in root and shoot tissues. The results showed that the combined stress did not affect the AMF colonization rate. AMF inoculation significantly increased plant biomass, the K+/Na+ ratio, and shoot B, sodium, and copper concentrations, but reduced root B concentrations and ion leakage. AMF inoculation slightly increased root dry weight and the sodium, potassium, zinc, copper and Mn contents in shoots under combined B and salinity stress, while AMF reduced the electrolyte leakage in leaves. It is inferred that AMF can ameliorate B toxicity in maize by improving biomass and reducing B concentration in plant tissues. Our research implies that C. etunicatum could be a valuable candidate for assisting in the remediation of boron-contaminated and saline soils.

Improved Waterlogging Tolerance in Roots of Cucumber Plants after Inoculation with Arbuscular Mycorrhizal Fungi

Mycorrhizal symbiosis enhances host plant resistance to various unfavorable environmental stresses, but whether and how it also enhances waterlogging tolerance in cucumber plants is not known. The objective of this study was to analyze the effect of Paraglomus occultum inoculation on biomass production, osmolyte levels, and the expression of 12 heat shock protein 70 (Hsp70) genes and 14 plasma membrane intrinsic protein (PIP) genes in the roots of cucumber plants under a short-term waterlogging stress (WS) (5 days) condition. Although the short-term WS treatment significantly inhibited the arbuscular mycorrhizal fungal colonization of roots, the inoculation with arbuscular mycorrhizal fungi (AMFs) significantly increased leaf, stem, and root biomass under WS. AMF inoculation also significantly increased root glucose, sucrose, betaine, and proline contents, along with decreased fructose levels, compared with the uninoculated control. More CsHsp70 and CsPIP genes were up-regulated in AMF-inoculated plants than in AMF-uninoculated plants in response to WS. AMF inoculation showed no significant effect on the expression of any of the examined CsHsp70 genes under no-waterlogging stress, but it did raise the expression of 11 of 12 CsHsp70 genes under WS. AMF colonization also down-regulated or had no effect on CsPIP expression under no-waterlogging stress, whereas it up-regulated the expression of 12 of the 14 CsPIP genes under WS. It is concluded that AMF inoculation enhances waterlogging tolerance in cucumber plants by increasing osmolyte levels and stress-responsive gene (CsPIP and CsHsp70) expression.

Mycorrhizal Symbiosis Can Change the Composition of Secondary Metabolites in Fruits ofSolanum nigrumL.

Solanum nigrum is a common weed in arable land, while being used in traditional medicine around the world due to its remarkable levels of valuable secondary metabolites. Agronomic and biological techniques can alter the production of a specific metabolite by influencing plant growth and metabolism. The effects of colonization with three arbuscular mycorrhizal fungi (AMF), including Funneliformis mosseae, Rhizoglomus intraradices, and Rhizoglomus fasciculatum, on the chemical composition of S. nigrum fruits were evaluated by gas chromatography-mass spectrometry (GC-MS) analysis. More than 100 different chemical constituents were evaluated by GC-MS. Our study revealed that the levels of phenols (quinic acid), benzenes (hydroquinone), sulfur-containing compounds, lactone and carboxylic acids were improved by R. intraradices. In contrast, hydroxymethylfurfural increased by 68% in R. fasciculatum inoculated with uninoculated S. nigrum plants, and this species was also the most efficient in inducing sugar compounds (D-galactose, lactose, and melezitose). Our results suggest that AMF colonization is an effective biological strategy that can alter the chemical composition and improve the medicinal properties of S. nigrum.

Arbuscular mycorrhiza and rhizosphere soil enzymatic activities as modulated by grazing intensity and plant species identity in a semi-arid grassland

Understanding the symbiosis of arbuscular mycorrhizal fungi (AMF) with plants in relation to soil nutrients and enzyme activities under different grazing intensities can be an important guide for the management and protection of semi-arid grasslands. The aim of the present study was to evaluate how the interaction of grazing intensity and plants shapes the composition of AMF communities and enzyme activities in a semi-arid grassland ecosystem in Iran. Sampling focused three dominant plant species (i.e., Salsola laricina, Artemisia siberia, and Stipa hohenackeriana) at sites with different grazing intensities. Soil chemical properties, enzyme activities, root colonization by AMF and AMF communities in the roots were evaluated. Potassium and nitrogen, as well as alkaline phosphatase and urease enzymatic activities were significantly increased at the heavily grazed site, whereas root colonization by AMF was reduced by the high grazing intensity. In addition, AM fungal root colonization is dependent on the host plant species and easier to measure as a sensitive indicator of sustainable grazing. Neither plant species nor grazing intensity affected AM fungal diversity in roots, which could be due to the overall low phylogenetic diversity of AMF in the grassland and the lack of significant differences in soil humidity, pH and organic carbon between the sites. However, plant species and soil properties were the two factors explaining variation in AMF community composition, while grazing had no significant effect. Therefore, AMF communities in root of the semi-arid grassland plants responded largely to plant type rather than to grazing intensity.

Tripartite interactions among free‐living, N‐fixing bacteria, arbuscular mycorrhizal fungi, and plants: Mutualistic benefits and community response to co‐inoculation

AbstractInteractions between arbuscular mycorrhizal (AM) fungi and free‐living nitrogen fixers (FLNF) occur in the rhizosphere where they can enhance plant nutrient acquisition, impact plant growth, and affect soil processes. Tripartite mutualism commonly occurs between nodule‐forming plants, symbiotic diazotrophs, and AM fungi, and can occur between non‐nodulating plants, FLNF, and AM fungi. However, information on the extent of, and controls on, tripartite mutualism in non‐nodulating plant systems is limited to a small number of crop plants and culturable microbial inoculum, mostly in greenhouse growing conditions. We conducted a systematic literature review to synthesize the current understanding of the responses of plants, AM fungi, and FLNF to co‐inoculation, as well as the conditions affecting tripartite mutualism and the magnitude and range of benefits conferred. Our review shows that plants generally benefit from co‐inoculation with AM fungi and FLNF taxa, but benefits are highly variable and context dependent, ranging from 94% reduction in plant shoot biomass to 255% increase in total plant biomass. Additionally, the presence of AM fungi can increase abundance of FLNF and the presence of FLNF can increase AM fungal root colonization, but these responses also vary widely. Major factors influencing variation in response to co‐inoculation by all organisms include plant phenology/age, soil type and nutrient availability, and partner pairing. There is potential for leveraging these tripartite mutualisms to improve plant productivity and soil microbial function, but successful application is more likely with a thorough understanding of the environmental and mechanistic controls on these relationships and testing of field‐scale implementation.

Can Arbuscular Mycorrhizal Fungi Enhance Crop Productivity and Quality in Hydroponics? A Meta-Analysis

Finding environmentally friendly solutions for crop growth and productivity has been gaining more attention recently. This meta-analysis aims to understand the combined application of arbuscular mycorrhizal fungi (AMF) and hydroponic systems compared to AMF in conventional (soil) systems. The analysis of up-to-date studies revealed that the root colonization, calculated as the proportion of colonized root segments relative to the total root length, by AMF in conventional (soil-based) culture exceeded hydroponic (or soilless) culture systems by 16.8%. The mean root colonization by AMF was determined to be 52.3% in hydroponic systems and 61.1% in conventional systems. Within hydroponic systems, the root colonization ranged from 2% to 20% after 10 days of inoculation, and notably, it exceeded 50% after 30 to 65 days, depending on the growing substrate and species. Under hydroponics, AMF application had a higher (compared to none-inoculated) positive effect on crop biomass and yield than fruit and leaf quality (antioxidants, phenols, and sugars) as well as leaf nutrients. However, AMF do not always have the potential to improve crop growth, quality and productivity in hydroponics. Among the studies analyzed in this review, approximately 34% (no effect: 29%; negative: 5%) reported no discernible positive effect on biomass or yield, 37% (no effect: 16%; negative: 21%) on fruit or leaf quality, and 60% (no effect: 47%; negative: 13%) on nutrient levels within plant tissues. To improve the performance of AMF in hydroponic systems, the meta-analysis recommended maintaining phosphorus levels in the nutrient solution within the range of 0.15 to 15.5 mg L−1 as elevated levels (40–75 mg L−1) were found to significantly reduce AMF colonization. Additionally, it was observed that certain hydroponic techniques, such as the presence of air bubbles generated by air pumps in floating hydroponic systems (Deep Flow technique) and continuous circulation of the nutrient solution (Ebb and Flow systems), may create dynamic conditions that could potentially hinder the introduction of AMF spores into hydroponic systems and potentially compromise the integrity of the spores and hyphae.

Distinct effects of canopy vs understory and organic vs inorganic N deposition on root resource acquisition strategies of subtropical Moso bamboo plants

Atmospheric nitrogen (N) deposition inevitably alters soil nutrient status, subsequently prompting plants to modify their root morphology (i.e., adopting a do-it-yourself strategy), mycorrhizal symbioses (i.e., outsourcing strategy), and root exudation (i.e., nutrient-mining strategy) linking with resource acquisition. However, how N deposition influences the integrated pattern of these resource-acquisition strategies remains unclear. Furthermore, most studies in forest ecosystems have focused on understory N and inorganic N deposition, neglecting canopy-associated processes (e.g., N interception and assimilation) and the impacts of organic N on root functional traits. In this study, we compared the effects of canopy vs understory, organic vs inorganic N deposition on eight root functional traits of Moso bamboo plants. Our results showed that N deposition significantly decreased arbuscular mycorrhizal fungi (AMF) colonization, altered root exudation rate and root foraging traits (branching intensity, specific root area, and length), but did not influence root tissue density and N concentration. Moreover, the impacts of N deposition on root functional traits varied significantly with deposition approach (canopy vs. understory), form (organic vs. inorganic), and their interaction, showing variations in both intensity and direction (positive/negative). Furthermore, specific root area and length were positively correlated with AMF colonization under canopy N deposition and root exudation rate in understory N deposition. Root trait variation under understory N deposition, but not under canopy N deposition, was classified into the collaboration gradient and the conservation gradient. These findings imply that coordination of nutrient-acquisition strategies dependent on N deposition approach. Overall, this study provides a holistic understanding of the impacts of N deposition on root resource-acquisition strategies. Our results indicate that the evaluation of N deposition on fine roots in forest ecosystems might be biased if N is added understory.

Transcriptome Analysis Reveals the Impact of Arbuscular Mycorrhizal Symbiosis on Toona ciliata var. pubescens Seedlings

Toona ciliata var. pubescens, known as “Chinese mahogany”, has high commercial value and is classified as a level II priority protected wild plant in China. However, due to overexploitation and its poor natural regeneration capacity, natural T. ciliata var. pubescens forests show varying degrees of decline in habitat adaptability. Arbuscular mycorrhizal fungi (AMF) symbiosis presents a potential strategy to enhance its regeneration. In this study, T. ciliata var. pubescens seedlings were inoculated with Septoglomus viscosum, followed by RNA-Seq analysis to compare gene expression differences between AMF-inoculated (AMI) and non-mycorrhizal (NM) treatments three months post-inoculation. A total of 16,163 differentially expressed genes (DEGs) were upregulated by AMF colonization, constituting 96.46% of the total DEGs. Specifically, 14,420 DEGs were exclusively expressed in the AMI treatment, while 35 DEGs were completely silenced. Most of the upregulated DEGs were located on the cell membrane, nucleus, and cytoskeleton and functioned in protein binding, S-adenosylmethionine-dependent methyltransferase activity, and lipid binding during cellular/macromolecule/protein localization, intracellular/protein transport, the cell cycle, and signal transduction. Additionally, lots of key genes related to oxidative stress responses, nutrient transport, and small GTPase-mediated signal transduction were found to be upregulated. These results suggest that AMF inoculation may enhance root cell growth by activating genes involved in nutrient uptake, stress responses, signal transduction, and substance transportation. This study elucidates the molecular mechanisms underlying the growth promotion of T. ciliata var. pubescens through AMF symbiosis, laying a foundation for the future application of AMF in its natural forest regeneration.

Community responses of arbuscular mycorrhiza fungi to hydrological gradients in a riparian Phragmites australis wetland.

The hydrological regime is considered to be the major factor that affects the distribution of arbuscular mycorrhiza (AM) fungi in wetlands. We aimed to investigate the responses of AM fungal community to different hydrological gradients. Illumina Miseq sequencing technology was used to study the AM fungal community structure in roots and rhizosphere soils of Phragmites australis in different moisture areas (dry area, alternating wet and dry area, and flooded area) in Mengjin Yellow River wetland. The rhizosphere soils and roots hosted different AM fungal communities. In roots, the AM fungal colonization and Chao1 richness in dry area were significantly higher than that in alternating wet and dry area and flooded area, but the community composition did not vary clearly under different water conditions. In rhizosphere soils, the Chao1 richness of AM fungi in flooded area was significantly higher than that in alternating wet and dry area and dry area, and the AM fungal community structure obviously differed across different areas. The redundancy analyses indicated that changes in the AM fungal community in soils were associated with altered soil properties, and the abundance of the dominant genus Glomus was mostly positively correlated with alkali-hydrolyzable nitrogen in soils. This study helps us to understand the responses of AM fungal community to hydrological gradients in wetlands.

OsEXO70L2 is required for large lateral root formation and arbuscular mycorrhiza establishment in rice

As a major subunit of the exocyst complex, members of the EXO70 family have mainly been shown to play roles in cell polarity and morphogenesis in Arabidopsis, but their roles in plant endosymbiosis, such as with arbuscular mycorrhizal fungi (AMF), have rarely been reported. Here, using knockout and overexpression lines, we show that OsEXO70L2, which encodes a divergent EXO70 protein in rice, controls the number of primary roots and is essential for large lateral root formation. Furthermore, the OsEXO70L2 mutant sr1 displayed rare internal AMF hyphae and no arbuscules. We also found that AMF sporulation can occur in roots despite low colonization and that AMF colonization and sporulation are modulated by photoperiod and co-culture with clover. Finally, genes related to auxin homeostasis were found to be affected in the OsEXO70L2 knockout or overexpression lines, suggesting that auxin is at least partly responsible for the phenotypes. This study provides new perspectives on the role of the exocyst complex during root development and AM in rice.

Molybdenum-mediated nitrogen accumulation and assimilation in legumes stepwise boosted by the coexistence of arbuscular mycorrhizal fungi and earthworms

Molybdenum (Mo) is a critical micronutrient for nitrogen (N) metabolism in legumes, yet the impact of Mo on legume N metabolism in the context of natural coexistence with soil microorganisms remains poorly understood. This study investigated the dose-dependent effect of Mo on soil N biogeochemical cycling, N accumulation, and assimilation in alfalfa under conditions simulating the coexistence of arbuscular mycorrhizal fungi (AMF) and earthworms. The findings indicated that Mo exerted a hormetic effect on alfalfa N accumulation, facilitating it at low concentrations (below 29.98 mg/kg) and inhibiting it at higher levels. This inhibition was attributed to Mo-induced constraints on C supply for nitrogen fixation. Concurrently, AMF colonization enhanced C assimilation in Mo-treated alfalfas by promoting nutrients uptake, particularly Mg, which is crucial for chlorophyll synthesis. This effect was further amplified by earthworms, which improved AMF colonization (p < 0.05). In the soil N cycle, these organisms exerted opposing effects: AMF enhanced soil nitrification and earthworms reduced soil nitrate (NO3−-N) reduction to jointly increase soil phyto-available N content (p < 0.05). Their combined action improved alfalfa N assimilation by restoring the protein synthesis pathway that is compromised by high Mo concentrations, specifically the activity of glutamine synthetase. These findings underscored the potential for soil microorganisms to mitigate N metabolic stress in legumes exposed to elevated Mo levels.

Effect of arbuscular mycorrhizal symbiosis on growth and biochemical characteristics of Chinese fir (Cunninghamia lanceolata) seedlings under low phosphorus environment.

The continuous establishment of Chinese fir (Cunninghamia lanceolata) plantations across multiple generations has led to the limited impact of soil phosphorus (P) on tree growth. This challenge poses a significant obstacle in maintaining the sustainable management of Chinese fir. To investigate the effects of Arbuscular mycorrhizal fungi (AMF) on the growth and physiological characteristics of Chinese fir under different P supply treatments. We conducted an indoor pot simulation experiment in the greenhouse of the Forestry College of Fujian Agriculture and Forestry University with one-and-half-year-old seedlings of Chinese fir from March 2019 to June 2019, with the two P level treatment groups included a normal P supply treatment (1.0 mmolL-1 KH2PO4, P1) and a no P supply treatment (0 mmolL-1 KH2PO4, P0). P0 and P1 were inoculated with Funneliformis mosseae (F.m) or Rhizophagus intraradices (R.i) or not inoculated with AMF treatment. The AMF colonization rate in the root system, seedling height (SH), root collar diameter (RCD) growth, chlorophyll (Chl) photosynthetic characteristics, enzyme activities, and endogenous hormone contents of Chinese fir were estimated. The results showed that the colonization rate of F.m in the roots of Chinese fir seedlings was the highest at P0, up to 85.14%, which was 1.66 times that of P1. Under P0 and P1 treatment, root inoculation with either F.m or R.i promoted SH growth, the SH of R.i treatment was 1.38 times and 1.05 times that of F.m treatment, respectively. In the P1 treatment, root inoculation with either F.m or R.i inhibited RCD growth. R.i inhibited RCD growth more aggressively than F.m. In the P0 treatment, root inoculation with F.m and R.i reduced the inhibitory effect of phosphorus deficiency on RCD. At this time, there was no significant difference in RCD between F.m, R.i and CK treatments (p < 0.05). AMF inoculation increased Fm, Fv, Fv/Fm, and Fv/Fo during the chlorophyll fluorescence response in the tested Chinese fir seedlings. Under the two phosphorus supply levels, the trend of Fv and Fm of Chinese fir seedlings in different treatment groups was F.m > R.i > CK. Under P0 treatment, The values of Fv were 235.86, 221.86 and 147.71, respectively. The values of Fm were 287.57, 275.71 and 201.57, respectively. It increased the antioxidant enzyme activity and reduced the leaf's malondialdehyde (MDA) content to a certain extent. It is concluded that AMF can enhance the photosynthetic capacity of the host, regulate the distribution of endogenous hormones in plants, and promote plant growth by increasing the activity of antioxidant enzymes. When the P supply is insufficient, AMF is more helpful to plants, and R.i is more effective than F.m in alleviating P starvation stress in Chinese fir.

Mycorrhizas Affect Physiological Performance, Antioxidant System, Photosynthesis, Endogenous Hormones, and Water Content in Cotton under Salt Stress.

Saline-alkali stress seriously endangers the normal growth of cotton (Gossypium hirsutum). Arbuscular mycorrhizal fungi (AMF) could enhance salt tolerance by establishing symbiotic relationships with plants. Based on it, a pot experiment was conducted to simulate a salt environment in which cotton was inoculated with Paraglomus occultum to explore its effects on the saline-alkali tolerance of cotton. Our results showed that salt stress noticeably decreased cotton seedling growth parameters (such as plant height, number of leaves, dry weight, root system architecture, etc.), while AMF exhibited a remarkable effect on promoting growth. It was noteworthy that AMF significantly mitigated the inhibitory effect of salt on cotton seedlings. However, AMF colonization in root and soil hyphal length were collectively descended via salt stress. With regard to osmotic regulating substances, Pro and MDA values in roots were significantly increased when seedlings were exposed to salt stress, while AMF only partially mitigated these reactions. Salt stress increased ROS levels in the roots of cotton seedlings and enhanced antioxidant enzyme activity (SOD, POD, and CAT), while AMF mitigated the increases in ROS levels but further strengthened antioxidant enzyme activity. AMF inoculation increased the photosynthesis parameters of cotton seedling leaves to varying degrees, while salt stress decreased them dramatically. When inoculated with AMF under a salt stress environment, only partial mitigation of these photosynthesis values was observed. Under saline-alkali stress, AMF improved the leaf fluorescence parameters (φPSII, Fv'/Fm', and qP) of cotton seedlings, leaf chlorophyll levels, and root endogenous hormones (IAA and BR); promoted the absorption of water; and maintained nitrogen balance, thus alleviating the damage from salt stress on the growth of cotton plants to some extent. In summary, mycorrhizal cotton seedlings may exhibit mechanisms involving root system architecture, the antioxidant system, photosynthesis, leaf fluorescence, endogenous hormones, water content, and nitrogen balance that increase their resistance to saline-alkali environments. This study provide a theoretical basis for further exploring the application of AMF to enhance the salt tolerance of cotton.

Interactions between foredune morphologies and vegetation: Exploring the occurrence of arbuscular mycorrhizal fungi

Vegetation is a key trait for understanding the foredune morphologies. However, the rhizosphere in coastal dunes imposes high levels of water and nutrient stress on plants. In these conditions, arbuscular mycorrhizal fungi (AMF) are prominent root symbionts that play a crucial role in improving to abiotic stress and promoting plant growth. On the south coast of the state of São Paulo, foredunes were studied for their floristic composition, relative frequency of AMF and grain size of rhizosphere. Foredune was classified regarding the morphology as terrace, ridge I, and ridge II. The terrace has a low height (<0.5 m) and is composed of native species canopies low (<0.2 m) and high (0.5–1.5 m) characterized by AMF colonization close to 60%. In contrast, ridge I showed wavy crests that were about 1 m high and ∼30 m wide, with a predominance of plant species with low canopy and 68% AMF colonization. The morphology ridge II reaches the greatest height (3 m), to the detriment of a smaller width (up to 15 m) and the plants present high canopy (>0.5 m) with close to 75% AMF. The increase of the relative frequency AMF from the terrace to ridges I and II may indicate that the increase of sand accumulation with continuous burial of roots is rather positive to plant growth. Yet, varied canopy heights of the plants species control the verticalization and horizontalization of the foredune. Low-canopy species, gradually slow down wind flow, causing spatialized deposition and widening. Conversely, species with highest canopies swiftly reduce wind flow due to the density of their aerial portion (leaves), resulting in deposition just below their structure fostering foredune vertical growth.

Arbuscular mycorrhizal fungi induce Zn uptake and antioxidant efficiency in broccoli (Brassica oleracea L.) exposed to Zn deficiency

Zinc (Zn) is an essential micronutrient for plant growth and development, and its deficiency in soil can be a significant problem for broccoli (Brassica oleracea L.) production. While the genetic efforts to breed Zn-efficient broccoli are complex, harnessing microbiomes is an emerging way to induce mineral efficiency in crops. In this study, broccoli, a non-host mycorrhizal species, was cultivated with or without zinc deficiency in the presence or absence of arbuscular mycorrhizal fungi (AMF), confirmed by the colonization efficiency in roots. Zn deficiency caused a significant decrease in morphological parameters and photosynthetic attributes being consistent with the decreased Zn levels in root and shoot relative to Zn-sufficient plants. However, the broccoli plants inoculated with AMF showed a substantial improvement in morphological and photosynthetic parameters in Zn-deficient conditions due to the significant increase in tissue Zn levels. In addition, AMF-inoculated plants under Zn deficiency showed a significant decrease in cell death (%), electrolyte leakage, and H2O2 levels compared to Zn-deficient plants, which further suggests the improvements in cellular status due to the AMF colonization. The real-time PCR experiments showed a significant induction in the expression of BoZIP1 and BoNRAMP1 genes in the roots of Zn-deficient broccoli inoculated with AMF, suggesting that AMF may be associated with the induction of these genes responsible for Zn-uptake and transport. We further observed the induction of POD (peroxidase), APX (ascorbate peroxidase) and SOD (superoxide dismutase), and S-metabolites (cysteine and glutathione) predominantly in the roots of Zn-starved broccoli inoculated with AMF which may confer tolerance to Zn-deficiency induced oxidative damage. This is the first report on the role of AMF in mitigating Zn-deficiency in broccoli which may promote the microbiome-aided improvement in plant health suffering from Zn-deficiency in broccoli and other vegetable crops.

Zea mays cultivation, biochar, and arbuscular mycorrhizal fungal inoculation influenced lead immobilization.

Plant cultivation can influence the immobilization of heavy metals in soil. However, the roles of soil amendments and microorganisms in crop-based phytoremediation require further exploration. In this study, we evaluated the impact of Zea mays L. cultivation, biochar application, and arbuscular mycorrhizal fungi (AMF) inoculation on soil lead (Pb) immobilization. Our results indicated that biochar addition resulted in a significant, 42.00%, reduction in AMF colonization. Plant cultivation, AMF inoculation, and biochar addition all contributed to enhanced Pb immobilization, as evidenced by decreased levels of diethylenetriaminepentaacetic acid- and CaCl2-extractable Pb in the soil. Furthermore, soil subjected to plant cultivation with AMF and biochar displayed reduced concentrations of bioavailable Pb. Biochar addition altered the distribution of Pb fractions in the soil, transforming the acid-soluble form into the relatively inert reducible and oxidizable forms. Additionally, biochar, AMF, and their combined use promoted maize growth parameters, including height, stem diameter, shoot and root biomass, and phosphorus uptake, while simultaneously reducing the shoot Pb concentration. These findings suggest a synergistic effect in Pb phytostabilization. In summary, despite the adverse impact of biochar on mycorrhizal growth, cultivating maize with the concurrent use of biochar and AMF emerges as a recommended and effective strategy for Pb phytoremediation.IMPORTANCEHeavy metal contamination in soil is a pressing environmental issue, and phytoremediation has emerged as a sustainable approach for mitigating this problem. This study sheds light on the potential of maize cultivation, biochar application, and arbuscular mycorrhizal fungi (AMF) inoculation to enhance the immobilization of Pb in contaminated soil. The findings demonstrate that the combined use of biochar and AMF during maize cultivation can significantly improve Pb immobilization and simultaneously enhance maize growth, offering a promising strategy for sustainable and effective Pb phytoremediation practices. This research contributes valuable insights into the field of phytoremediation and its potential to address heavy metal pollution in agricultural soils.
