Advances in Immunomodulatory Mesoporous Silica Nanoparticles for Inflammatory and Cancer Therapies.

In recent decades, immunotherapy has been considered a promising treatment approach. The modulatable enhancement or attenuation of the body's immune response can effectively suppress tumors. However, challenges persist in clinical applications due to the lack of precision in antigen presentation to immune cells, immune escape mechanisms, and immunotherapy-mediated side effects. As a potential delivery system for drugs and immunomodulators, mesoporous silica has attracted extensive attention recently. Mesoporous silica nanoparticles (MSNs) possess high porosity, a large specific surface area, excellent biocompatibility, and facile surface modifiability, making them suitable as multifunctional carriers in immunotherapy. This article summarizes the latest advancements in the application of MSNs as carriers in cancer immunotherapy, aiming to stimulate further exploration of the immunomodulatory mechanisms and the development of immunotherapeutics based on MSNs.

The Facile Synthesis and Application of Mesoporous Silica Nanoparticles with a Vinyl Functional Group for Plastic Recycling.

Due to growing concerns about environmental pollution from plastic waste, plastic recycling research is gaining momentum. Traditional methods, such as incorporating inorganic particles, increasing cross-linking density with peroxides, and blending with silicone monomers, often improve mechanical properties but reduce flexibility for specific performance requirements. This study focuses on synthesizing silica nanoparticles with vinyl functional groups and evaluating their mechanical performance when used in recycled plastics. Silica precursors, namely sodium silicate and vinyltrimethoxysilane (VTMS), combined with a surfactant, were employed to create pores, increasing silica's surface area. The early-stage introduction of vinyl functional groups prevented the typical post-synthesis reduction in surface area. Porous silica was produced in varying quantities of VTMS, and the synthesized porous silica nanomaterials were incorporated into recycled polyethylene to induce cross-linking. Despite a decrease in surface area with increasing VTMS content, a significant surface area of 883 m2/g was achieved. In conclusion, porous silica with the right amount of vinyl content exhibited improved mechanical performance, including increased tensile strength, compared to conventional porous silica. This study shows that synthesized porous silica with integrated vinyl functional groups effectively enhances the performance of recycled plastics.

Effects of Skeleton Structure of Mesoporous Silica Nanoadjuvants on Cancer Immunotherapy.

Mesoporous silica nanoparticles (MSNs) have been widely praised as nanoadjuvants in vaccine/tumor immunotherapy thanks to their excellent biocompatibility, easy-to-modify surface, adjustable particle size, and remarkable immuno-enhancing activity. However, the application of MSNs is still greatly limited by some severe challenges including the unclear and complicated relationships of structure and immune effect. Herein, three commonly used MSNs with different skeletons including MSN with tetrasulfide bonds (TMSN), MSN containing ethoxy framework (EMSN), and pure -Si-O-Si- framework of MSN (MSN) are comprehensively compared to study the impact of chemical construction on immune effect. The results fully demonstrate that the three MSNs have great promise in improving cellular immunity for tumor immunotherapy. Moreover, the TMSN performs better than the other two MSNs in antigen loading, cellular uptake, reactive oxygen species (ROS) generation, lymph node targeting, immune activation, and therapeutic efficiency. The findings provide a new paradigm for revealing the structure-function relationship of mesoporous silica nanoadjuvants, paving the way for their future clinical application.

Mesoporous silica/organosilica nanoparticles for cancer immunotherapy.

Cancer is one of the fatal diseases in the history of humankind. In this regard, cancer immunotherapeutic strategies have revolutionized the traditional mode of cancer treatment. Silica based nano-platforms have been extensively applied in nanomedicine including cancer immunotherapy. Mesoporous silica nanoparticles (MSN) and mesoporous organosilica nanoparticles (MON) are attractive candidates due to the ease in controlling the structural parameters as needed for the targeted immunotherapeutic applications. Especially, the MON provide an additional advantage of controlling the composition and modulating the biological functions to actively synergize with other immunotherapeutic strategies. In this review, the applications of MSN, MON, and metal-doped MSN/MON in the field of cancer immunotherapy and tumor microenvironment regulation are comprehensively summarized by highlighting the structural and compositional attributes of the silica-based nanoplatforms.

Mesoporous Silica Nanoparticles as a Potential Nanoplatform: Therapeutic Applications and Considerations.

With advances in nanotechnology, nanoparticles have come to be regarded as carriers of therapeutic agents and have been widely studied to overcome various diseases in the biomedical field. Among these particles, mesoporous silica nanoparticles (MSNs) have been investigated as potential nanocarriers to deliver drug molecules to various target sites in the body. This review introduces the physicochemical properties of MSNs and synthesis procedures of MSN-based nanoplatforms. Moreover, we focus on updating biomedical applications of MSNs as a carrier of therapeutic or diagnostic cargo and review clinical trials using silica-nanoparticle-based systems. Herein, on the one hand, we pay attention to the pharmaceutical advantages of MSNs, including nanometer particle size, high surface area, and porous structures, thus enabling efficient delivery of high drug-loading content. On the other hand, we look through biosafety and toxicity issues associated with MSN-based platforms. Based on many reports so far, MSNs have been widely applied to construct tissue engineering platforms as well as treat various diseases, including cancer, by surface functionalization or incorporation of stimuli-responsive components. However, even with the advantageous aspects that MSNs possess, there are still considerations, such as optimizing physicochemical properties or dosage regimens, regarding use of MSNs in clinics. Progress in synthesis procedures and scale-up production as well as a thorough investigation into the biosafety of MSNs would enable design of innovative and safe MSN-based platforms in biomedical fields.

Nanoparticles are unique in diagnosis and therapy in that they do not affect the surroundings or show toxicity. Modern medicine has undergone further development with nanoscale materials providing advanced experimentation, clinical use, and applications. Nanoparticle use in imaging, drug delivery, and treatment is growing rapidly owing to their spectacular accuracy, bioavailability, and cellular permeability. Mesoporous silica nanoparticles (MSNs) play a significant role in nano therapy with several advantages such as easy synthesis, loading, controllability, bioavailability over various surfaces, functionalization, and biocompatibility. MSNs can be used as theranostics in immune-modulatory nano systems to diagnose and treat inflammatory diseases. The application of MSNs in the preparation of drug-delivery systems has been steadily increasing in recent decades. A systemic review on development of mesoporous nanoparticles as a vehicle for transdermal drug delivery.

Recent advances in drug delivery technologies utilizing a variety of carriers have resulted in a paradigm shift in the current approach to diagnosis and therapy. Mesoporous silica nanoparticles (MSNs) were developed in response to the need for materials with high thermal, chemical, and mechanical properties. The synthesis, ease of surface functionalization, tunable pore size, large surface area, and biocompatibility of MSNs make them useful in a variety of biomedical applications such as drug delivery, theranostics, and stem cell research. In addition, MSNs have a high capability of delivering actives ranging from small molecules such as drugs and amino acids to larger peptides, vaccines, and antibodies in general. Moreover, MSN-based transdermal delivery has sparked a lot of interest because of the increase in drug stability, permeation, and ease of functionalization. The functionalization of MSNs plays an important role in the efficient delivery of therapeutic agents in a highly controlled manner. This review introduced dermal and transdermal drug delivery systems, explained the anatomy of the skin, and summarized different barriers that affect the transdermal delivery of many therapeutic agents. In addition, the fundamentals of MSNs together with their physicochemical properties, synthesis approaches, raw materials used in their fabrication, and factors affecting their physicochemical properties will be covered. Moreover, the applications of MSNs in dermal and transdermal delivery, the biocompatibility of MSNs in terms of toxicity and safety, and biodistribution will be explained with the help of a detailed literature review. The review is covering the current and future perspectives of MSNs in the pharmaceutical field with therapeutic applications.

Key Parameters for the Rational Design, Synthesis, and Functionalization of Biocompatible Mesoporous Silica Nanoparticles.

Over the last few years, research on silica nanoparticles has rapidly increased. Particularly on mesoporous silica nanoparticles (MSNs), as nanocarriers for the treatment of various diseases because of their physicochemical properties and biocompatibility. The use of MSNs combined with therapeutic agents can provide better encapsulation and effective delivery. MSNs as nanocarriers might also be a promising tool to lower the therapeutic dosage levels and thereby to reduce undesired side effects. Researchers have explored several routes to conjugate both imaging and therapeutic agents onto MSNs, thus expanding their potential as theranostic platforms, in order to allow for the early diagnosis and treatment of diseases. This review introduces a general overview of recent advances in the field of silica nanoparticles. In particular, the review tackles the fundamental aspects of silicate materials, including a historical presentation to new silicates and then focusing on the key parameters that govern the tailored synthesis of functional MSNs. Finally, the biomedical applications of MSNs are briefly revised, along with their biocompatibility, biodistribution and degradation. This review aims to provide the reader with the tools for a rational design of biocompatible MSNs for their application in the biomedical field. Particular attention is paid to the role that the synthesis conditions have on the physicochemical properties of the resulting MSNs, which, in turn, will determine their pharmacological behavior. Several recent examples are highlighted to stress the potential that MSNs hold as drug delivery systems, for biomedical imaging, as vaccine adjuvants and as theragnostic agents.

Multi-functional pH-sensitive active and intelligent packaging based on highly cross-linked zein for the monitoring of pork freshness

Intelligent packaging not only protects food from environmental hazards but intuitively monitors the changes of food quality and safety. A novel intelligent packaging film with pH sensitivity and antibacterial and antioxidant effects was developed based on the highly cross-linked zein. The composite film with 0.05 g/g crosslinking agent had the best mechanical properties. The tensile strength (TS) and elongation-at-break (EBA) were 2.42 and 1.53 times of that of zein film, respectively. Moreover, the intelligent packaging showed longstanding antimicrobial and antioxidant effects because of the addition of the tea tree essential oil (TTEO)-loaded mesoporous silica nanoparticles (MSNs). The blueberry anthocyanin as colorimetric indicator was added in the packaging film to monitor the safety of meat products using a mobile phone. The color of the composite film as the packaging of the pork products changed from colorless to brown after 9-day storage to show the spoilage of the pork. To the best of our knowledge, this is the first-time report of 1) the application of TTEO-loaded MSNs for food packing, 2) the addition of TTEO as the antimicrobial agent for zein film, and 3) using the applications of mobile phone to measure the RBG value of the zein-based film. This study offers an example of the highly promising biodegradable intelligent packaging with multi-functions for the enhancement of food safety.

Recent advances in mesoporous silica nanoparticle-based targeted drug-delivery systems for cancer therapy.

Targeted drug-delivery systems are a growing research topic in tumor treatment. In recent years, mesoporous silica nanoparticles (MSNs) have been extensively studied and applied in noninvasive and biocompatible drug-delivery systems for tumor therapy due to their outstanding advantages, which includehigh surface area, large pore volume, tunable pore size, easy surface modification and stable framework. The advances in the application of MSNs for anticancer drug targeting are covered and highlighted in this review, and the challenges and prospects of MSN-based targeted drug-delivery systems are discussed. This review provides new insights for researchers interested in targeted drug-delivery systems against cancer.

Application of Mesoporous Silica Nanoparticles in Cancer Therapy and Delivery of Repurposed Anthelmintics for Cancer Therapy

This review focuses on the biomedical application of mesoporous silica nanoparticles (MSNs), mainly focusing on the therapeutic application of MSNs for cancer treatment and specifically on overcoming the challenges of currently available anthelmintics (e.g., low water solubility) as repurposed drugs for cancer treatment. MSNs, due to their promising features, such as tunable pore size and volume, ability to control the drug release, and ability to convert the crystalline state of drugs to an amorphous state, are appropriate carriers for drug delivery with the improved solubility of hydrophobic drugs. The biomedical applications of MSNs can be further improved by the development of MSN-based multimodal anticancer therapeutics (e.g., photosensitizer-, photothermal-, and chemotherapeutics-modified MSNs) and chemical modifications, such as poly ethyleneglycol (PEG)ylation. In this review, various applications of MSNs (photodynamic and sonodynamic therapies, chemotherapy, radiation therapy, gene therapy, immunotherapy) and, in particular, as the carrier of anthelmintics for cancer therapy have been discussed. Additionally, the issues related to the safety of these nanoparticles have been deeply discussed. According to the findings of this literature review, the applications of MSN nanosystems for cancer therapy are a promising approach to improving the efficacy of the diagnostic and chemotherapeutic agents. Moreover, the MSN systems seem to be an efficient strategy to further help to decrease treatment costs by reducing the drug dose.

Insight on the Dependence of the Drug Delivery Applications of Mesoporous Silica Nanoparticles on Their Physical Properties

SummaryMesoporous silica nanoparticles (MSNs) are fascinating due to their interesting properties and applications.PurposeThe optimization of MSNs for drug delivery applications was achieved by preparing different formulations of MSNs using different concentrations of ammonium hydroxide (NH4OH) (0.7, 1.4, 2.8, 4.2, and 5.6 mg/ml for MSN1, MSN2, MSN3, MSN4, and MSN5, respectively).MethodsIn the synthesis of MSNs, NH4OH was used as a catalyst while tetraethyl orthosilicate were used as a source of silica. Transmission electron microscopy (TEM) image revealed a linear increase in the size of the formed MSNs with increase in catalyst concentration. TEM images showed that all investigated nanoparticles were dispersed and spherical (changed to oval on addition of higher concentration of NH4OH).ResultsThe hydrodynamic sizes of prepared MSNs were (64.18 ± 6.8, 90.46 ± 7.1, 118.98 ± 7.01, 152.7 ± 1.7, and 173.9 ± 9.36 nm for MSN1, MSN2, MSN3, MSN4, and MSN5, respectively) assessed using the dynamic light scattering (DLS) technique. The negative values of zeta potential indicated high surface stability of the formed MSNs. N2-isotherm revealed that the pore volume of MSNs decreased with increase in the size of MSNs. In vitro drug release showed that all MSNs exhibited high encapsulation efficiency of doxorubicin. The encapsulation efficiency were 92.2%, 82.8%, 72.2%, 72.1% and 71.9%for MSN1, MSN2, MSN3, MSN4, and MSN5, respectively. ConclusionMSN1 and MSN2, with sizes of 64.18 ± 6.8 and 90.46 ± 7.1 nm, pore volume of 0.89 and 0.356 cc/g, encapsulation efficiency of 92.2% and 82.8%, and adequate drug release profiles, were probably the best choices for a drug carrier in drug delivery applications.

Effect of Surface Functionalization and Pore Structure Type on the Release Performance of Mesoporous Silica Nanoparticles

Based on the excellent physicochemical properties of mesoporous silica nanoparticles (MSN), the application of MSN in drug delivery has become a popular strategy. However, a wide variety of MSN structures and surface modification affect the drug loading and release, thus affecting the effectiveness of drug delivery system. Therefore, the effects of MSN type, pore structure and modified functional groups on drug release properties were investigated in this paper. A series of MSN was synthesized by sol-gel method, and the mesopore type and pore size were adjusted. Their surfaces were modified by amino groups (NH 2 ) and polydopamine (PDA) to change the surface charge. Anticancer Doxorubicin (DOX) was used as a model drug to study effects of different types of MSN on drug loading and release. MSN with hollow structure had a large inner cavity for DOX loading and tiny pore shell to prevent DOX loss, leading to higher loading capacity. MSN with surface-modified amine was positively charged and processed good pH responsiveness and accelerated DOX release. Thus hexagonal pore MSN-NH 2 and MSN-PDA were the anticancer DOX delivery carriers with better combined release performance in terms of pH-responsive performance and sustained release effect. Finally, several kinetic models were measured to fit the in vitro DOX release data and the results showed that release behaviors of different MSNs were more consistent with Korsmeyer-Peppas release kinetic model. All of them were Fick diffusion-controlled except for the second release stage of hollow MSN at pH 7.4, which was non-Fick diffusion release. • A series of MSNs with diverse structures and modification were studied. • Specific surface area, pore volume/type and surface charge affected drug loading. • Diverse kinetic models were employed to investigate mechanism of drug release. • MSNs with modification were drug delivery systems with better release performance.

How Advancing are Mesoporous Silica Nanoparticles? A Comprehensive Review of the Literature.

The application of mesoporous silica nanoparticles (MSNs) is ubiquitous in various sciences. MSNs possess unique features, including the diversity in manufacturing by different synthesis methods and from different sources, structure controllability, pore design capabilities, pore size tunability, nanoparticle size distribution adjustment, and the ability to create diverse functional groups on their surface. These characteristics have led to various types of MSNs as a unique system for drug delivery. In this review, first, the synthesis of MSNs by different methods via using different sources were studied. Then, the parameters affecting their physicochemical properties and functionalization have been discussed. Finally, the last decade’s novel strategies, including surface functionalization, drug delivery, and cancer treatment, based on the MSNs in drug delivery and cancer therapy have been addressed.

Recent Developments in Mesoporous Silica Nanoparticles for Tumor Theranostic Applications.

Due to the advantages of adjustable pore size, easy surface modification, high biocompatibility, and so on, mesoporous silica nanoparticles (MSNs) have attracted significant attention. Moreover, they are widely used in the fields of biology and medical research, mostly focusing on drug and gene delivery and bioimaging. This review introduces several commonly used synthetic methods of MSNs and the latest progress of MSNs in tumor therapy and diagnosis, mainly including the study about modified MSNs as drug carriers and the application of MSNs in bioimaging. The deficiencies of MSNs' application and prospects for its future clinical transformation are also discussed.

Mesoporous Silica Nanoparticles in Chemical Detection: From Small Species to Large Bio-Molecules.

A recompilation of applications of mesoporous silica nanoparticles in sensing from the last five years is presented. Its high potential, especially as hybrid materials combined with organic or bio-molecules, is shown. Adding to the multiplying effect of loading high amounts of the transducer into the pores, the selectivity attained by the interaction of the analyte with the layer decorating the material is described. Examples of the different methodologies are presented.

Our contributions to applications of mesoporous silica nanoparticles

Our contributions to mesoporous silica materials in the field of biomedicine are reported in this article. This perspective article represents our work in the basics of the material, preparing different ranges of mesoporous silica nanoparticles with different diameters and with varied pore sizes. We demonstrated the high loading capacity of these materials. Additionally, the possibility of functionalizing both internal and external surface with different organic or inorganic moieties allowed the development of stimuli-responsive features which allowed a proper control on the administered dose. In addition, we have demonstrated that these carriers are not toxic, and we have also ensured that the load reaches its destination without affecting healthy tissues. Statement of significanceThis paper presents my personal opinion and background on a hot topic as mesoporous silica nanoparticles for drug delivery. To this aim it provides a comprehensive and historical overview on the innovative contributions of my research group to this rapidly expanding field of research.
