Application of nanotechnology and proteomic tools in crop development towards sustainable agriculture

AbstractThe increase in global population which translates to increased demand for food called for urgent attention from key players and policy makers in agricultural sector. Also, the effects of climate change and its consequent biotic and abiotic stresses in plants has greatly affect the sustainability of agriculture and production of food. These challenges require novel and sustainable approaches to improve the quality and yield of crops. The vast application of nanoparticles in different fields of study is attributed to their distinct chemical and physical characteristics. However, in agriculture, their application is limited because of their safety concern. Currently, research is tailored to study the response of plants to various nanoparticle treatments; however, these studies are inconclusive due to limited knowledge on the mechanisms of plant–nanoparticle interactions. Recently, studies on nanobiotechnology have taken a new dimension from preliminary bioassay experiments to more complex, research-oriented studies using various omics tools. Changes in protein expression caused by plant–nanoparticle interaction at any developmental stages, or tissue types may be investigated using suitable proteomics techniques. This review discussed the various applications of nanotechnology and proteomic tools in plant growth and development. Plant–nanoparticle interactions and the application of nanoparticles and proteomic tools in genetic engineering of plants to attain agricultural sustainability and food safety are also discussed. Informative and thorough understanding of plant–nanoparticle interaction will serve as a blueprint that will enable plant scientist and genetic engineers to develop plant biomarkers and explore their potential application for crop improvement.

In Situ Plasma Studies Using a Direct Current Microplasma in a Scanning Electron Microscope

AbstractMicroplasmas can be used for a wide range of technological applications and to improve the understanding of fundamental physics. Scanning electron microscopy, on the other hand, provides insights into the sample morphology and chemistry of materials from the mm‐ down to the nm‐scale. Combining both would provide direct insight into plasma‐sample interactions in real‐time and at high spatial resolution. Up till now, very few attempts in this direction have been made, and significant challenges remain. This work presents a stable direct current glow discharge microplasma setup built inside a scanning electron microscope. The experimental setup is capable of real‐time in situ imaging of the sample evolution during plasma operation and it demonstrates localized sputtering and sample oxidation. Further, the experimental parameters such as varying gas mixtures, electrode polarity, and field strength are explored and experimental V–I curves under various conditions are provided. These results demonstrate the capabilities of this setup in potential investigations of plasma physics, plasma‐surface interactions, and materials science and its practical applications. The presented setup shows the potential to have several technological applications, for example, to locally modify the sample surface (e.g., local oxidation and ion implantation for nanotechnology applications) on the µm‐scale.

Biogenic synthesized SeONPs by Bacillus paramycoides as antimicrobial, anticancer, antioxidant, apoptotic and hepatorenal treating agent.

Employing bacteria in nanoparticles (NPs) biosynthesis has appeared as a fast-emerging research area in green nanotechnology applications. The biosynthesis processes optimization can lead to obtaining NPs with desired sizes and suitable quantities. The goal of the current work was to synthesize selenium oxide nanoparticles (SeONPs) from Bacillus paramycoides (MCCC 1A04098), a locally isolated terrestrial bacteria, and in vitro investigate its antibacterial, anticancer, and anti-cytotoxic properties. Rats with hepatorenal injury triggered by carbon tetrachloride (CCl4) were used for the in vivo evaluation. Oxidative stress markers, lipid profile, liver and kidney function indices, as well as their histological features were measured and compared with silymarin as a reference drug. SeONPs revealed antimicrobial effect against Bacillus cereus, Staphylococcus aureus, Escherichia coli, and Candida albicans. It also showed anticancer effect on hepatic (HepG2) and breast (MCF-7) cancer cell lines in a dose dependently with apoptotic effect rather than necrosis. Treatment of CCl4 injured rats with SeONPs restored all the biochemical parameters to normal values with variable degrees and the results were confirmed by the histological pictures of liver and kidney. In conclusion, the obtained biogenic SeONPs from the local isolate of Bacillus paramycoides strain (MCCC 1A04098) showed a remarkable biomedical application including in vitro antimicrobial and anticancer activities. It also recorded in vivo antioxidant, anti-hypercholesterolemic and anti-cytoxic activities against hepatorenal injury in rats. Therefore, SeONPs synthesis by ecofriendly technique may be considered as a promising nutraceutical agent.

Motility of an autonomous protein-based artificial motor that operates via a burnt-bridge principle

Inspired by biology, great progress has been made in creating artificial molecular motors. However, the dream of harnessing proteins – the building blocks selected by nature – to design autonomous motors has so far remained elusive. Here we report the synthesis and characterization of the Lawnmower, an autonomous, protein-based artificial molecular motor comprised of a spherical hub decorated with proteases. Its “burnt-bridge” motion is directed by cleavage of a peptide lawn, promoting motion towards unvisited substrate. We find that Lawnmowers exhibit directional motion with average speeds of up to 80 nm/s, comparable to biological motors. By selectively patterning the peptide lawn on microfabricated tracks, we furthermore show that the Lawnmower is capable of track-guided motion. Our work opens an avenue towards nanotechnology applications of artificial protein motors.

The Effects of Silver Nanoparticles on the Antimicrobial and Biodegradation of Cornstarch Bioplastic

The development of bioplastics is currently increasing, because bioplastics are an effort to reduce landfill waste. One of the bioplastics that has good degradation ability is cornstarch. The addition of nanoparticles was carried out to improve the properties of bioplastic packaging. One example of the application of nanotechnology in food packaging is silver nanoparticles (AgNP), known as antimicrobial substances. This research was conducted to determine the effect of adding AgNP (0%, 1%, and 2%) on the antimicrobial and biodegradation of cornstarch bioplastics. Bioplastics are made by casting method. AgNP was used from the synthesis of silver nitrate (AgNO3) and trisodium citrate dihydrate (C6H5Na3O7.2H2O) as a reducing agent and stabilizer by chemical reduction method, which was then analyzed by FTIR. The results obtained showed that cornstarch bioplastic AgNP 1% has the ability to estimate the fastest degradation time among other concentrations with an addition of 103 days. Cornstarch bioplastic AgNP 2% had the best ability to inhibit bacterial growth, with antibacterial inhibition zone diameters of 11.03 mm (Staphylococcus aureus) and 10.61 mm (Escherichia coli). However, AgNP could not inhibit the mold growth of Aspergillus niger. The addition of AgNP to cornstarch bioplastics can increase the degradation capabilities and antibacterial activity of bioplastics.

"Turbo-Charged" DNA Motors with Optimized Sequence Enable Single-Molecule Nucleic Acid Sensing.

DNA motors that consume chemical energy to generate processive mechanical motion mimic natural motor proteins and have garnered interest due to their potential applications in dynamic nanotechnology, biosensing, and drug delivery. Such motors translocate by a catalytic cycle of binding, cleavage, and rebinding between DNA "legs" on the motor body and RNA "footholds" on a track. Herein, we address the well-documented trade-off between motor speed and processivity and investigate how these parameters are controlled by the affinity between DNA legs and their complementary footholds. Specifically, we explore the role of DNA leg length and GC content in tuning motor performance by dictating the rate of leg-foothold dissociation. Our investigations reveal that motors with 0 % GC content exhibit increased instantaneous velocities of up to 150 nm/sec, three-fold greater than previously reported DNA motors and comparable to the speeds of biological motor proteins. We also demonstrate that the faster speed and weaker forces generated by 0 % GC motors can be leveraged for enhanced capabilities in sensing. We observe single-molecule sensitivity when programming the motors to stall in response to the binding of nucleic acid targets. These findings offer insights for the design of high-performance DNA motors with promising real-world biosensing applications.

Recent status and trends of nanotechnology in cervical cancer: a systematic review and bibliometric analysis.

Cervical cancer is currently the second leading cause of death among women. However, there is a lack of effective treatment methods, and the existing treatments often result in significant adverse reactions and high chances of recurrence, which ultimately impact the prognosis of patients. As a result, the application of nanotechnology, specifically nanoparticle-based approaches, in the diagnosis and treatment of cervical cancer has gained significant attention.This study aims to examine the current research status and future development trends of nanotechnology in relation to cervical cancer using a bibliometric perspective. A bibliometric analysis was performed to gather relevant research papers from the Web of Science database. VOSviewer and CiteSpace were utilized to conduct quantitative analysis and identify hot topics in the field, focusing on countries, institutions, journals, authors, and keywords. A total of 997 eligible literature were retrieved. From January 1, 2014 to September 20, 2023, the overall number of publications showed an upward trend. The paper mainly comes from China (n=414). The main institution is the Chinese Academy of Sciences (n=62), and 60% of the top 10 institutions in the number of documents issued are from China. First authors Ma, Rong (n=12) and Alifu, Nuernisha (n=12). The journal with the highest publication volume is ACS Applied Materials&INTERFACES (n=35), and the journal with the highest citation frequency is BIOMATERIALS (n=508). "Nanoparticles (n=295)", "cervical cancer (n=248)", and "drug delivery (n=218)" are the top three most frequently occurring keywords. In recent years, photothermal therapy and indocyanine green have become research hotspots. The application of nanotechnology in the field of cervical cancer has garnered considerable attention. Nanoparticles-based methods for diagnosis, administration, and treatment have proven to be instrumental in enhancing the sensitivity of cervical cancer detection, improving the accuracy and efficiency of administration, and reducing drug toxicity. Enhancing treatment efficacy and improving patient prognosis have emerged as current research priorities and future directions.

REVIEW: REVIEW ARTICLE LIPID-BASED NANOTECHNOLOGY

Nanotechnology is defined as engineering the creation of materials, functions, and devices on the nanometer scale. Nanoscience is increasingly developing and becoming a part of various fields, such as electronics, materials, and biology. Lipid nanoparticles are a major application in nanotechnology. In formulas II and III, smaller results were obtained compared with the other formulas. Based on the results of this study, it can be concluded that the nanostructured lipid carrier system with solid lipid poloxamer and stearic acid with liquid soybean oil lipids obtained good characteristics, and each test met the range. From the explanation above, it can be concluded that lipid-based nanotechnology has great potential in various fields such as medicine, cosmetics, and agriculture. Its use can provide great benefits such as increasing the effectiveness of drug administration, increasing the absorption of nutrients in food, and developing cosmetic products. which one is safer and more effective. To compare several types of nanoparticle methods, such as Liposomes, Neosomes, Etosomes, SLN, and NLC for selection of the lipid base, namely NLC. Nanotechnology-based delivery systems to overcome drug resistance in cancer.

Cancer nanomedicine is defined as the application of nanotechnology and nanomaterials for the formulation of cancer therapeutics that can overcome the impediments and restrictions of traditional chemotherapeutics. Multidrug resistance (MDR) in cancer cells can be defined as a decrease or abrogation in the efficacy of anticancer drugs that have different molecular structures and mechanisms of action and is one of the primary causes of therapeutic failure. There have been successes in the development of cancer nanomedicine to overcome MDR; however, relatively few of these formulations have been approved by the United States Food and Drug Administration for the treatment of cancer. This is primarily due to the paucity of knowledge about nanotechnology and the fundamental biology of cancer cells. Here, we discuss the advances, types of nanomedicines, and the challenges regarding the translation of invitro to invivo results and their relevance to effective therapies.

Critical characteristics of spin − 1/2 four-leg nanotube: Monte Carlo Simulation

In this study, we aim to investigate theoretically the critical characteristics of spin −1/2 four-leg nanotube for the nanotechnology applications. The Ising model simulated by Monte Carlo simulation is adopted with the first exchange coupling J , the second exchange coupling between the second nearest neighbor atomic spins J ′ and the transverse exchange coupling J z , and the external magnetic field h as parameters. Herein, we examine the local properties of spin −1/2 four-leg nanotube by analyzing the magnetization, susceptibility and hysteresis loop behavior as function of J ′ , J Z and h . The obtained results show that the isotropic ‐ 1 / 2 four-leg nanotube Ising model presents critical behavior at ferromagnetic to paramagnetic phase transition where the Curie temperature T C increases according to vertical and second nearest neighbor exchange coupling. Furthermore, under external magnetic field, the magnetization keeps the second order transition and the magnetic susceptibility declines with the increase of applied external magnetic field value. In addition to that, we have found single ferromagnetic hysteresis loops, for which the shape increases by increasing the second nearest neighbor exchange coupling when T = J . Finally, the critical temperature shows perfect mirror symmetry relative to the vertical and increases with the second nearest neighbor exchange coupling.

Doping-induced changes in the structural and mechanical properties of germanene monolayers: A DFT-Based study

In the current study, the density functional theory is utilized to investigate the elastic, plastic and electronic properties of the 2×2 and 3×3 pristine and transition metal (TM) doped germanene. Different atoms, including V, Co, Fe, Mn, Cr, Ti, Ni and Sc are selected for this purpose. It is shown that doping of the transition metal atoms would result in the reduction of the Young's and bulk moduli of the germanene. In addition, the isotropic behavior of these nanosheets were shown by comparing the Young's moduli of both pristine and doped structures in armchair and zigzag directions. Furthermore, the plastic behavior of these structures were investigated by increasing the applied loading. It was seen that except for the 2×2 Co-doped monolayer, the yield strain of both 2×2 and 3×3 nanosheets were reduced under uniaxial and biaxial loading. In 2×2 doped nanosheets under uniaxial loading, the highest reduction occurs for Ti-doped germanene, and in 3×3 cases, the highest reduction happens for Co, Ni, Fe and Sc-doped germanene. Electron localization function revealed the ionic nature of the boding between TM atoms and germanium while the density of state indicated that doping of transition metals would change the semi-metallic behavior of germanene to metallic except for Ti-doped structure. The findings provide valuable information for potential applications in nanotechnology, guiding the development of new materials with specific characteristics based on the identified impacts of different transition metals.

Field-induced macroscopic flow of a dilute self-assembling magnetic colloid under rotating magnetic fields

Flow generation by colloidal motors activated by external stimuli is an important issue for active matter physics and several nanotechnological or biomedical applications. For instance, flow recirculation generated by rotating magnetic self-assemblies allows effective ‘pumping’ of a thrombolytic drug towards a blood clot along a blocked vessel. However, the physics of the flow generation in this case remains still poorly explored. This study is focused on the generation of a recirculation flow of a magnetic colloid (aqueous suspension of iron oxide nanoparticles with partially screened electrostatic repulsion) within a closed microfluidic channel via application of an external rotating magnetic field. The colloid undergoes reversible phase separation manifested through the appearance of micron-sized elongated aggregates. They synchronously rotate with the magnetic field and can generate macroscopic flows only in the presence of gradients of the aggregate concentration across the channel induced by superposition of a weak magnetic field gradient to the homogeneous rotating field. We achieve recirculation flows with a characteristic speed ${\sim} 5{-}8\;{\rm \mu}\textrm{m}\;{\textrm{s}^{ - \textrm{1}}}$ at low magnetic field amplitude and frequency ( ${H_0} \approx 3{-}10\;\textrm{kA}\;{\textrm{m}^{ - 1}}$ , ${f = 5{-}15\ \textrm{Hz}}$ ) at low nanoparticle volume fraction ${\varphi _p} = (1.6{-}3.2) \times {10^{ - 3}}$ . The concentration and velocity profiles have been assessed experimentally through particle tracking and particle image velocimetry, and have also been computed using the hydrodynamic diffusion approach coupled with the momentum balance equation with a magnetic torque term. The model correctly reproduces the shape of the experimental concentration and velocity fields and explains complex behaviours of the average recirculation speed as a function of governing parameters ( ${H_0}$ , f, ${\varphi _p}$ , channel size).

Advances and Emerging Trends in Horticultural Production and Management

The potential of advanced technologies like automation, robotics, artificial intelligence, and genetics in transforming horticulture is discussed. Challenges for sustainable intensification of horticultural systems are examined. The review highlights how cutting-edge sciences, digital integration, and ecological approaches will shape the future of horticulture with more productive, efficient, and climate-resilient production.

Biosynthesis of NPs CuS/Cu2S and self-assembly with C. beijerinckii for improving lignocellulosic butanol production in staged butyrate-butanol fermentation process

A staged butyrate-butanol fermentation process (SBBFP) has been developed to efficiently produce biobutanol from renewable lignocellulose. The SBBFP combines the fermentation of bioaugmented degradation consortium for butyrate (BDCB) and butanol production of Clostridium beijerinckii, but is still challenged by the lower specific butanol yield (SBY). In this study, a novel technology for synthesizing semiconductor nanoparticles (NPs) and their self-assembly with Clostridium beijerinckii was proposed to enhance the SBY. Results showed that supplement of 0.1 g/L CuSO4 or 0.2 g/L ZnSO4 were more favorable for the formation of butyrate and NPs (CuS/Cu2S or ZnS) in the acidogenic fermentation process of BDCB. Butyrate production achieved 19–21 g/L in the fed-batch fermentation mode from NaOH-pretreated rice straw. Only CuS/Cu2S can be assembled with NPs@C. beijerinckii hybrid system, and a butanol production of 14.6 g/L was obtained using acidogenic fermentation broth and saccharide liquid from rice straw as the co-substrates, with the SBY of 0.17 g/g initial pretreated rice straw. The enhanced butanol synthesis was likely attributed to the upgraded intracellular NADH/NAD+ and specific activity of enzyme CoA-transferase (ctfAB) in C. beijerinckii. The research work provides an efficient approach for biofuels and biochemical production from lignocellulose, along with the extended application of biological nanotechnology.

The C-terminus is essential for the stability of the mycobacterial channel protein MspA.

Outer membrane proteins perform essential functions in uptake and secretion processes in bacteria. MspA is an octameric channel protein in the outer membrane of Mycobacterium smegmatis and is structurally distinct from any other known outer membrane protein. MspA is the founding member of a family with more than 3000 homologs and is one of the most widely used proteins in nanotechnological applications due to its advantageous pore structure and extraordinary stability. While a conserved C-terminal signal sequence is essential for folding and protein assembly in the outer membrane of Gram-negative bacteria, the molecular determinants of these processes are unknown for MspA. In this study, we show that mutation and deletion of methionine 183 in the highly conserved C-terminus of MspA and mutation of the conserved tryptophan 40 lead to a complete loss of protein in heat extracts of M. smegmatis. Swapping these residues partially restores the heat stability of MspA indicating that methionine 183 and tryptophan 40 form a conserved sulfur-π electron interaction, which stabilizes the MspA monomer. Flow cytometry showed that all MspA mutants are surface-accessible demonstrating that oligomerization and membrane integration in M. smegmatis are not affected. Thus, the conserved C-terminus of MspA is essential for its thermal stability, but it is not required for protein assembly in its native membrane, indicating that this process is mediated by a mechanism distinct from that in Gram-negative bacteria. These findings will benefit the rational design of MspA-like pores to tailor their properties in current and future applications.

Frontiers of stem cell engineering for nanotechnology-mediated drug delivery systems.

Cell biology approaches have gained a successful integration, development and application of nanotechnology with stem cell engineering and have led to the emergence of a new interdisciplinary field known as stem cell nanotechnology (SCN). Recent studies showed the potential and the advancement of developments for SCN applications in drug delivery systems. Cancer, neurodegenerative, muscle and blood diseases, cell and gene therapies, and tissue engineering and regenerative medicine applications are the important targets of SCN. In this overview, we searched the literature using the common online websites for research and read the open access, full-text available articles since 2013. The studies vary according to the type of disease they targeted and the strategies they proposed, whether diagnostic or therapeutic. In addition to the use of stem cells, the utilisation of their membranes, secretomes, exosomes and extracellular vesicles with an appropriate nanotechnology strategy is also an aspect of the research. This brief overview of stem cell nanotechnology over the last ten years aims to provide insight into the frontiers of stem cell engineering for nanotechnology-mediated drug delivery systems.

Design and Fabrication of Environmentally Responsive Nanoparticles for the Diagnosis and Treatment of Atherosclerosis.

Cardiovascular disease poses a significant threat to human health in today's society. A major contributor to cardiovascular disease is atherosclerosis (AS). The development of plaque in the affected areas involves a complex pathological environment, and the disease progresses rapidly. Nanotechnology, combined with emerging diagnostic and treatment methods, offers the potential for the management of this condition. This paper presents the latest advancements in environment-intelligent responsive controlled-release nanoparticles designed specifically for the pathological environment of AS, which includes characteristics such as low pH, high reactive oxygen species levels, high shear stress, and multienzymes. Additionally, the paper summarizes the applications and features of nanotechnology in interventional therapy for AS, including percutaneous transluminal coronary angioplasty and drug-eluting stents. Furthermore, the application of nanotechnology in the diagnosis of AS shows promising real-time, accurate, and continuous effects. Lastly, the paper explores the future prospects of nanotechnology, highlighting the tremendous potential in the diagnosis and treatment of atherosclerotic diseases, especially with the ongoing development in nano gas, quantum dots, and Metal-Organic Frameworks materials.

Exposure to zinc oxide nanoparticles induced reproductive toxicities in male Sprague Dawley rats

BackgroundThis research delves into the reproductive toxicology of zinc oxide nanoparticles (ZnO-NPs) in male Sprague Dawley rats. It specifically examines the repercussions of Zn accumulation in the testes, alterations in testosterone levels, and histopathological changes in the gonadal tissues. AimsThe primary objective of this study is to elucidate the extent of reproductive toxicity induced by ZnO-NPs in male Sprague Dawley rats. The investigation aims to contribute to a deeper understanding of the potential endocrine and reproductive disruptions caused by ZnO-NPs exposure. MethodsCharacterization techniques including SEM-EDX and XRD affirmed the characteristic nature of ZnO-NPs. Twenty-five healthy post weaning rats (200–250 g) were intraperitoneally exposed to different concentrations of ZnO-NPs @ 10 or 20 or 30 mg/kg BW for 28 days on alternate days. ResultsResults showed significant dose dependent decline in the body weight and testicular somatic index of rats. It also showed significant dose dependent accumulation of Zn in testis with increasing dose of ZnO-NPs. Conversely, serum testosterone level and sperm count were reduced with increasing dose of ZnO-NPs. Histological results showed dose dependent abnormalities i.e., vacuolization, edema, hemorrhage, destruction of seminiferous tubules, loss of germ cells and necrosis in rat testis. ConclusionThe findings of this study clearly indicate that high doses of zinc oxide nanoparticles (ZnO-NPs) can adversely affect the structural integrity and functional efficacy of the male reproductive system. Given these results, it becomes crucial to implement stringent precautionary measures in the utilization of ZnO-NPs, particularly in cosmetics and other relevant sectors. Such measures are imperative to mitigate the toxicological impact of ZnO-NPs on the male reproductive system and potentially on other related physiological functions. This study underscores the need for regulatory vigilance and safety assessments in the application of nanotechnology to safeguard human health.

Probing the interface of choline chloride-based deep eutectic solvent ethaline with gold surfaces: A molecular dynamics simulation study

Technologies involving a solvent|surface interface, such as nanotechnology, electrochemistry, and energy storage applications, are actively pursuing ecologically responsible and sustainable development practices. In response to this pressing need, deep eutectic solvents have emerged as a promising solution to bridge the gap between technological requirements and environmental concerns. In this work, we present the results of a molecular dynamics simulation study of the interface between a monocrystalline gold surface and the deep eutectic solvent ethaline, where a molar ratio of 1:2 choline chloride:ethylene glycol was used for ethaline. The simulations covered a range of temperatures from 313 K to 343 K and applied charge values ranging from 0 to ± 24 μC cm‒2. Several key interfacial properties were thoroughly analyzed, including among others, charge density profiles, radial distribution functions, hydrogen bond close contacts, and molecular orientation. Additionally, we examined how the differential capacitance varied upon the applied potential. Our findings reveal that, at neutral surfaces, all components of the solvent are present in the innermost layer, with ethylene glycol molecules being the most prevalent, followed by choline cations and a residual amount of chloride anions. For lower applied charges, this mixed composition at the boundary layer persists, despite the growing accumulation of ionic species with charges opposite to that of the electrode. As surface polarization increases, unique innermost boundary layers composed exclusively of one of the ionic species and the hydrogen bond donor molecules are observed, forming a multilayer structure, with subsequent layers enriched of paired counterions. Interestingly, even at higher applied charges, choline cations and ethylene glycol molecules tended to orient themselves in a parallel fashion toward the electrodes. Differential capacitance curves exhibited a camel-shaped behavior, suggesting a complex interplay of electrochemical processes at the DES|Au(100) interface. In summary, our study provides valuable insights into the interfacial properties of deep eutectic solvents on gold surfaces and their response to changes in temperature and potential, which are crucial for understanding and optimizing deep eutectic solvent-based electrochemical systems.
